Legionella pneumophila (Legionnaires disease bacterium) of serogroup 1 was isolated from an algal-bacterial mat community growing at 45 degrees C in a man-made thermal effluent. This isolate was grown in mineral salts medium at 45 degrees C in association with the blue-green alga (cyanobacterium) Fischerella sp. over a pH range of 6.9 to 7.6. L. pneumophila was apparently using algal extracellular products as its carbon and energy sources. These observations indicate that the temperature, pH, and nutritional requirements of L. pneumophila are not as stringent as those previously observed when cultured on complex media. This association between L. pneumophila and certain blue-green algae suggests an explanation for the apparent widespread distribution of the bacterium in nature.  相似文献

Detection ofLegionella pneumophila by serogroup-specific fluorescent antibodies was combined with a tetrazolium dye (INT) to measure electron transport activity. The biological uptake and reduction of the INT dye was studied in pure cultures and in natural water samples with respect to temperature. Uptake was complete within 60 min. Controls inhibited with formaldehyde demonstrated little activity. Both the in vitro and in situ determinations suggested that the electron transport system ofLegionella was active over a temperature range of 25 to 60°C.  相似文献

The effects of fluctuations in the irradiance onScenedesmus quadricauda, Chlorella vulgaris andSynechococcus elongatus were studied in dilute cultures using arrays of red light emitting diodes. The growth rate and the rate of photoinhibition were compared using intermittent and equivalent continuous light regimes in small-size (30 ml) bioreactors. The CO2 dependent photosynthetic oxygen evolution rates in the intermittent and continuous light regimes were compared for different light/dark ratios and different mean irradiances. The kinetics of the electron transfer reactions were investigated using a double-modulation fluorometer. The rates of photosynthetic oxygen evolution normalized to equal mean irradiance were lower or equal in the intermittent light compared to the maximum rate found in the equivalent optimal continuous light regime. In contrast, the growth rates in the intermittent light can be higher than the growth rate in the equivalent continuous light. Photoinhibition is presented as an example of a physiological process affecting the growth rate that occurs at different rates in the intermittent and equivalent continuous lights. The difference in the dynamics of the redox state of the plastoquinone pool is proposed to be responsible for the low photoinhibition rates observed in the intermittent light.  相似文献

Two serotype 1 strains ofLegionella pneumophila, Phildelphia 2 and Bellingham, were tested for their ability to metabolize five common substrates by measuring14CO2 released and14C-carbon incorporated into macromolecules. No major differences were noted between the two strains or preparations grown in the yolk sac of chick embryos or agar-broth diphasic medium, following 2 or 14 pasaages on agar. Glutamate was the most actively metabolized substrate, followed by glutamine. Acetate, glucose, and succinate were utilized at much more moderate rates. Changes in cell density and substrate concentration altered the channeling of glutamate and glucose into CO2 and macromolecules. Specific CO2 felease from glutamate was greatest at low cell density and high substrate concentration, while carbon incorporation was increased at high substrate concentration. A reciprocal relationship was noted with glucose: the proportion of carbon incorporation was enhanced at low substrate concentration, but CO2 release paralleled increases in substrate concentration. The pH optimum for glutamate carbon incorporation and CO2 release was 5.5 and 6.1, respectively, but 25% of both activities were retained at pH 3.1. CO2 release from glucose was maximal at pH 7.5 with negligible activity at pH 3.1. Pathways of glucose metabolism were explored by employing glucose, glucose-1-phosphate, and glucose-6-phosphate labeled in various carbon positions. The glycolytic pathway appeared to play a lesser role than the pentose phosphate and/or Entner-Doudoroff pathways. Glucose-1-phosphate was metabolized at a much higher rate than glucose or glucose-6-phosphate. We conclude that glutamate is utilized primarily as an energy source while glucose may serve as an important metabolite for the nutrition ofL. pneumophila.  相似文献

Survival ofLegionella pneumophila SG 1 in seawater and river water was assessed using plate counts on buffered charcoal yeast extract agar amended with α-ketoglutarate (BCYEα) and [3H]thymidine-labeling. The [3H]thymidine-labeling method for assessing survival ofL. pneumophila in aquatic environments was compared with viable counts, direct fluorescent microscopy (DFA), and acridine orange direct counts (AODC). Protozoa were isolated from the samples employed in the study and identified by characteristic trophozite and cyst morphology. Selective filtration employing 2.0 μm Nucleopore filters was used to determine the effect of grazing on survival ofL. pneumophila in seawater and river water.Legionella viability as measured by plate counts (CFU/ml), declined to a greater extent than cell lysis, assessed by thymidine, DFA, and AODC counts, suggesting thatL. pneumophila survives in aquatic habitats to a greater extent than revealed through culturable counts.  相似文献

The cytochrome composition of membrane vesicles ofLegionella pneumophila has been examined by low temperature (77°K) and room temperature difference spectroscopy, and cytochromes of thec, b, a, andd types have been detected. The presence ofc-type cytochrome was verified by formation of the pyridine ferrohemochromogen. A carbon monoxide-bindingc-type cytochrome was detected in CO-reduced minus reduced difference spectra and may also function in cytochromec reductase activity. Respiratory activities were determined for membrane vesicles, and reduced nicotinamide adenine dinucleotide (NADH) was the most rapidly oxidized substrate (199 nmol per min per mg protein), followed by succinate and malate. Cytochrome oxidase activity was demonstrated using ascorbate andN,N,N,N-tetramethyl-p-phenylenediamine (TMPD) (39 nmol per min per mg of protein). High levels of cyanide (K i =10 mM) inhibited NADH oxidation, while low levels of 2-heptyl-4-hydroxyquinoline-N-oxide (HQNO, 18 and 37 M) inhibited NADH oxidation by nearly 90%. The respiratory chain appeared to be complex and terminated by at least three terminal oxidases. Superoxide dismutase activity, but not catalase activity, was detected in cellular extracts.  相似文献

The numbers of presumptiveLegionella pneumophila cells in waters and sediments of nine different cooling towers located on the same site in the northeastern United States were determined at approximately monthly intervals for 18 months. All systems received makeup water from the same source and received the same chemical treatments. PresumptiveL. pneumophila were found in both water and sediment samples from all systems on all sampling dates. An important result of this study was the finding that tower sediments represent large reservoirs ofL. pneumophila. The only correlation between levels of presumptiveL. pneumophila and any of the physical, chemical, or operating characteristics evaluated was with winter shutdown and drainage followed by a nonoperational period. These systems showed a definite seasonal response with the highest levels of presumptiveL. pneumophila found in the summer and fall. Systems operated year round showed relatively constant numbers ofL. pneumophila in both water and sediments.  相似文献

Biodegradation of dyes by some green algae and cyanobacteria   总被引:1,自引:0,他引:1  
The ability of Chlorella vulgaris, Lyngbya lagerlerimi, Nostoc lincki, Oscillatoria rubescens, Elkatothrix viridis and Volvox aureus to decolorize and remove methyl red, orange II, G-Red (FN-3G), basic cationic, and basic fuchsin was investigated. These algae showed different efficiency for colour removal; varied from 4 to 95% according to the algal species, its growth state and the dye molecular structure. Basic cationic and basic fuchsin were the most susceptible dyes for decolorisation and removal by all algae being tested, and up to 82% of methyl red was also removed by N. lincki and O. rubescens. However, the algal activity to decolorize orange II and G-Red was markedly fluctuated and lower. C. vulgaris displayed activity to remove 43.7 and 59.12% while as V. aureus removed 5.02 and 3.25% of the added dyes respectively. The results also showed that treatment of either C. vulgaris or N. Linckia with G-Red or methyl red, respectively, induced the algal azo dye reductase enzyme by 72 and 71% at the same order.  相似文献

Codon usage in higher plants, green algae, and cyanobacteria   总被引:3,自引:1,他引:2  
Codon usage is the selective and nonrandom use of synonymous codons by an organism to encode the amino acids in the genes for its proteins. During the last few years, a large number of plant genes have been cloned and sequenced, which now permits a meaningful comparison of codon usage in higher plants, algae, and cyanobacteria. For the nuclear and organellar genes of these organisms, a small set of preferred codons are used for encoding proteins. Codon usage is different for each genome type with the variation mainly occurring in choices between codons ending in cytidine (C) or guanosine (G) versus those ending in adenosine (A) or uridine (U). For organellar genomes, chloroplastic and mitochrondrial proteins are encoded mainly with codons ending in A or U. In most cyanobacteria and the nuclei of green algae, proteins are encoded preferentially with codons ending in C or G. Although only a few nuclear genes of higher plants have been sequenced, a clear distinction between Magnoliopsida (dicot) and Liliopsida (monocot) codon usage is evident. Dicot genes use a set of 44 preferred codons with a slight preference for codons ending in A or U. Monocot codon usage is more restricted with an average of 38 codons preferred, which are predominantly those ending in C or G. But two classes of genes can be recognized in monocots. One set of monocot genes uses codons similar to those in dicots, while the other genes are highly biased toward codons ending in C or G with a pattern similar to nuclear genes of green algae. Codon usage is discussed in relation to evolution of plants and prospects for intergenic transfer of particular genes.  相似文献

Legionella pneumophila is the causative agent of Legionellosis in man and considered an opportunistic intracellular Gram-negative bacterium that preferentially infects macrophages. The presence of a plasmid in these organisms was determined in cultures of the bacteria grown in vitro. A correlation was observed between the growth of virulent strains of theLegionella in murine macrophages and growth on standard buffered charcoal yeast extract agar supplemented with 0.1% -ketoglutarate (BCYE) agar medium rich in cysteine and widely used for growth of the bacteria in vitro. In contrast, the avirulent isolates of these strains grew well on supplemented Mueller-Hinton (SMH) agar utilized for differentiating virulent from avirulentLegionella. However, one virulent strain ofLegionella (the Iowa strain) was found to grow moderately well on the SMH agar. In addition, test strains ofLegionella that infect in vitro human monocytes were found to grow moderately well on the BCYE- agar, but did not grow on the SMH agar. Examination of these strains for plasmid DNA expression showed that extra chromosomal DNA-containing molecules were present in theL. pneumophila strains characterized as virulent for in vitro growth in macrophages. However, the avirulent strains that replicated in the human monocytes readily but only poorly in the permissive murine macrophages did not show evidence of similar plasmid DNA expression.  相似文献

The inhibitory effect of serum on the growth and survival ofLegionella pneumophila Bloomington 2 was investigated. When incubated in the presence of 20%–50% normal human serum for 10 h, viability was decreased by >99%. Heat-inactivated or <40% normal serum supplemented with 50 M iron was not inhibitory. The addition of guinea pig complement to heat-inactivated serum resulted in killing of approximately 98% of the cells. Growth in buffered yeast extract broth was inhibited by the addition of ferric iron-binding compounds. Minimum bactericidal concentrations at 37°C were 10 M apotransferrin, 35 M 1,10-phenanthroline, and 50 M deferoxamine. Addition of iron chelators to normal serum did not accelerate killing. Egg yolk-passaged virulent strains and agar-grown avirulent strains exhibited similar serum sensitivity. Results of this study indicate that complement and serum transferrin are antagonistic to the growth ofLegionella in serum.  相似文献

Forty-nine strains of filamentous, mat-forming cyanobacteria isolated from the Arctic, subarctic and Antarctic environments were screened for their potential use in outdoor waste-water treatment systems designed for cold north-temperate climates. The most promising isolate (strain E18, Phormidium sp. from a high Arctic lake) grew well at low temperatures and formed aggregates (flocs) that could be readily harvested by sedimentation. We evaluated the growth and nutrient uptake abilities of E18 relative to a community of green algae (a Chlorococcalean assemblage, denoted Vc) sampled from a tertiary treatment system in Valcartier, Canada. E18 had superior growth rates below 15°C Canada. (μ = 0.20 d-1 at 10°C under continuous irradiance of 225 μmol photon m-2 s-1) and higher phosphate uptake rates below 10°C (k = 0.050 d-1 at 5°C) relative to Vc (μ=0.087 d-1 at 10°C and k = 0.020 d-1 at 5°C, respectively). The green algal assemblage generally performed better than E18 at high temperatures (at 25°C, μ = 0.39 d-1 and k = 0.34 d-1 for Vc; μ = 0.28 d-1 and k = 0.33 d-1 for E18). However, E18 removed nitrate more efficiently than Vc at most temperatures including 25°C. Polar cyanobacteria such as strain E18 are appropriate species for waste-water treatment in cold climates during spring and autumn. Under warmer summer conditions, fast-growing green algae such as the Vc assemblage are likely to colonize and dominate, but warm-water Phormidium isolates could be used at that time. Six herbicides were tested for their effects on the population growth of a range of green algae and cyanobacteria by an easily replicated low-volume liquid culture technique using Repli-dishes. Diuron, propanil and atrazine were most inhibitory, chlorpropham was intermediate and MCPA and glyphosate were least inhibitory. Chlorpropham was more inhibitory to green algae than to cyanobacteria. The effects of chlorpropham and 3-chloroaniline, a metabolite, on populations of the cyanobacterium Anacystis nidulans and the alga Chlamydomonas reinhardii were monitored in larger scale batch cultures. Both compounds reduced the growth rate although in some cases there was partial recovery. 3-Chloroaniline was less inhibitory than the parent herbicide chlorpropham.  相似文献

Mixtures of harmful and harmless algae were grown in discontinuouslydiluted batch cultures under ammonium, nitrate and phosphatelimitation, and at different irradiances (20–500 µjnolquanta m–2 s–1). The species used were Chrysochromulinapolylepis, Emiliania huxleyi type B, Rhodomonas sp., the dinoflagellalesFibrocapsa japonica, Gymnodinium simplex, Gyrodinium aure-olum,Heterocapsa triquetra, Heterosigma carterae, Prorocentrum micansand Alexandrium tamarense, the diatoms Chaetoceros socialis,Cymatosira belgica, Ditylum brightwellii, Laudcria borealis,Odon-telta aunla, Pseudonitzschia pungens, Streptotheca tamesis,and the cyanobacterium Synechococcus. Their growth responsein the mixed algal cultures is discussed in relation to theirabundance in different natural habitats. In comparison withthe other non-diatoms, the mixotrophic C.polylepis grew fastunder all tested nutrient and light limitations.Emiliania huxleyigrew well under nitrogen (N) limitation (with nitrate as N source)and at irradiance levels from 15 up to 500 µmol quantam–2 s–1. No growth of calcifying cells could bedetected under N limitation when ammonium was used as N source.Rhodomonas grew reasonably well under ammonium-N limitationand grew fast at the highest irradi-ance. The dinoflagellateswere poor competitors compared to the Prymnesiophyceae. Theenvironmental fitness of the Prymnesiophyceae appears to beclosely related to the reproductive capacity of the vegetativestage, whereas the natural distribution of dinoflagellates seemsmore closely dependent on the generative reproduction-relatedspecific life cycle characteristics of the individual species.The marine diatoms include a mixture of both types of species.Some marine diatom species clearly have the capability to outcompetenon-diatoms under different types of nutrient and light limitationswhen silicate is in excess. Immuno-compromised patients are particularly susceptible to Legionnaires' Disease. After three cases of the disease occurred in a hospital, a continuous dosing regime using chlorine dioxide was initiated to replace chlorination of the water system. This study identified a number of factors which may have resulted in conditions that would encourage the growth of the water-borne pathogenLegionella pneumophila. The residual chlorination was inadequate for microbial control at the taps furthest from the four storage tanks, of which two were found to be in excess for demand. The temperature of the water in the storage tanks was also found to be above 20° C; a temperature that would encourage microbial growth. A back-up calorifier was present and was found to containL. pneumophila, and linseed oil-based sealants that provide nutrients for microbial growth were also prevalent as jointing compounds in the water circult. Although the shower heads were routinely disinfected, a requirement was identified to also disinfect the shower hoses. NoL. pneumophila were recovered from the water system after the chlorine reduced dioxide disinfection trial. Biofilm was also dramatically reduced after disinfection; however, small microcolonies were identified and proved to be metabolically active when tested with a metabolic indicator. Using light and fluorescence microscopy, the pipe samples removed from the water system were rapidly analysed for biofouling, complementing existing microbiological methods.  相似文献

Blue‐green algae (cyanobacteria) have evolved as the most primitive, oxygenic, plant‐type photosynthetic organisms. Within a single prokaryotic cell, they have uniquely accommodated both oxygenic photosynthesis and aerobic respiration, which are known to produce superoxide and hydrogen peroxide as inevitable byproducts. Two types of superoxide dismutase have been characterized in both N2‐fixing and non‐N2‐fixing cyanobacteria, namely cytosolic iron‐containing superoxide dismutase and thylakoid‐bound manganese‐containing superoxide dismutase. No qualitative differences between various cell types (vegetative cells, heterocysts) were found. In contrast to chloroplasts, most of the cyanobacterial species show catalatic activity. From two species the corresponding enzymes have been characterized as typical prokaryotic (bifunctional) catalase‐peroxidases with homologies to cytochrome c peroxidases and ascorbate peroxidases. In addition to catalatic activity, some strains exhibit ascorbate peroxidase activity, but to date there are no reports detailing purification and characterization.
Cyanobacteria were found to contain low intracellular ascorbate concentrations (30‐100 µ M ) and 2‐5 m M glutathione. Both monodehydroascorbate and glutathione reductase activities were detected in most species examined, whereas dehydroascorbate reductase activity was absent. The question as to whether a glutathione‐ascorbate cycle exists in cyanobacteria cannot be answered at present.  相似文献
