We developed eight polymorphic microsatellite markers for the endangered salmonid Parahucho perryi from genomic libraries enriched for (GACA)n and (GATA)n repeat sequences. Emphasis was placed on developing highly polymorphic, perfect repeats that could be scored with confidence. Six tetra- and two di-nucleotide loci were screened in 49 individuals from two populations on Sakhalin Island, Russia. Allelic variation was high with eight to 23 alleles per locus and expected heterozygosity ranging from 0.48 to 0.93. These highly variable markers should prove useful in evaluating inbreeding, gene flow and population structure in Sakhalin taimen throughout its range.  相似文献

RFLP analysis of three amplified mtDNA fragments (Cytb/D-loop, ND1/ND2, and ND3/ND4L/ND4) was performed in the following taxa: Parahucho perryi, Hucho taimen, Brachymystax lenok, B. tumensis, Salmo salar, Salvelinus leucomaenis, and S. levanidovi. For mtDNA of P. perryi, a substantial decrease in the haplotype and nucleotide diversity was observed as a result of random genetic drift, caused by a reduction in the effective population size. Nucleotide divergence estimates between the mtDNA haplotypes were determined. Sakhalin taimen P. perryi was found to be approximately equally diverged from S. salar and from the charrs of the genus Salvelinus, by 11.0 and 10.0%, respectively. The divergence between P. perryi and H. taimen constituted 14.6%, between P. perryi and lenoks of the genus Brachymystax, 14.2%, and between H. taimen and Brachymystax, 7.7%. The analysis of possible phylogenetic relationships of the mtDNA from P. perryi among the group of taxa examined confirmed validity of the genus Parahucho. Phylogenetic reconstructions performed showed that robustness of the trees constructed for the complex of phylogenetically informative characters over three mtDNA fragments was considerably higher than that of the trees constructed for individual genes.  相似文献

The behavior of endangered adult Sakhalin taimen Parahucho perryi was tracked during 2008–2010 using acoustic telemetry in the Bekanbeushi River system, which flows through eastern Hokkaido, Japan. Movement distances per unit time of tracked P. perryi were compared between mid- and downstream habitats. Results indicated that movement distances were significantly longer in downstream habitats during all seasons. The movement distances in each stream decreased from spring to autumn. Moreover, tracked P. perryi exhibited crepuscular movement patterns; however, patterns were less pronounced in downstream habitats than in up- and midstream habitats. These findings strongly suggested that adult P. perryi exhibit nearly distinctive movement patterns across stream habitats; thus, fish may adopt different foraging tactics in each stream habitat. Fish moved more frequently in spring, which included the post-wintering and post-spawning season, most likely to search for food and more desirable habitat for recovery.  相似文献

We isolated and characterized eight polymorphic microsatellite markers for Brachymystax lenok (Pallas, 1773) from genomic libraries enriched for (GATA)n, (GACA)n and (ATG)n microsatellites. The number of alleles per locus ranged from two to 17. Heterozygosity ranged from 0.2 to 0.95. In addition, cross‐species amplification was successful for seven loci in Hucho hucho, eight in H. taimen and seven in Parahucho perryi.  相似文献

Spatial and temporal patterns of redd construction by Sakhalin taimen, Hucho perryi , were studied in small streams on northern Hokkaido, Japan. Spawning took place in late April with close relation to discharge patterns. Observed redd size (mean ± 95% C.I.) was 145 ± 11·8 cm in length and 84 ± 5·8 cm in width. The number of egg pockets per redd was either one or two, which is fewer than that of other salmonids reported in the literature. Spawning ground distributions in 1991 and 1992 were similar to each other in terms of the distribution mean and dispersion of the redds. Unlike other salmonids Sakhalin taimen probably constructed multiple redds during one spawning season.  相似文献

Japanese huchen, Hucho perryi, is an endangered fish in Japan. In order to promote a conservation program for this species, it is necessary to evaluate its genetic diversity and population genetics. For this purpose, we designed 15 primers containing dinucleotide microsatellite regions and examined their allelic variation using a total of 23 wild individuals collected from two different localities. Eight loci showed moderate allelic variation ranging from two to four alleles, with expected heterozygosities from 0.043 to 0.548. These markers will be useful for the assessment of genetic variation for this species.  相似文献

The extent of dispersal between populations of Caribbean reef fish remains largely unknown. Resolution of this issue is critical for managing populations and designing marine reserves. As part of a larger study of reef fish dispersal, we developed and characterized 11 polymorphic microsatellite loci for the bicolor damselfish (Stegastes partitus), an abundant reef fish found throughout the Caribbean. These loci will be useful for population genetic and relatedness studies of this species and also appear to be useful in other Caribbean members of the Pomacentridae.  相似文献

Small yellow croaker (Larimichthys polyactis) is an important economic species of marine fishery. We developed and evaluated simple sequence repeat (SSR) markers from expressed sequence tags (ESTs) of Pseudosciaena crocea, Paralichthys olivaceus and Psetta maxima. Characteristics of nine EST–SSR loci were investigated using 46 L. polyactis individuals. The number of alleles per locus ranged from 2 to 6. The observed heterozygosity ranged from 0.0652 to 0.7391, while the expected heterozygosity ranged from 0.0638 to 0.7754. Seven loci departed from Hardy–Weinberg equilibrium (P < 0.01) significantly. These loci and markers will be useful for population genetics and systemic evolution among species of small yellow croaker.  相似文献

Nine dinucelotide microsatellite loci were developed in the Antarctic fur seal Arctocephalus gazella. Each locus possessed between 4 and 9 alleles in a sample of twenty individuals sampled from Bird Island, South Georgia, and expected heterozygosity ranged from 0.52 to 0.84. All but one of the loci conformed to Hardy–Weinberg equilibrium and no evidence was found for genotypic disequilibrium. Additionally, all of the loci successfully cross-amplified in the South American fur seal Arctocephalus australis, the New Zealand fur seal Arctocephalus forsteri and the Steller’s sea lion Eumetopias jubatus, and six also yielded products in the more distantly related harbour seal Phoca vitulina and walrus Odobenus rosmarus. These loci should prove useful for studies of the population genetics of Antarctic fur seals and other important otariid species.  相似文献

Snow leopards (Panthera uncia) are elusive endangered carnivores found in remote mountain regions of Central Asia. New methods for identifying and counting snow leopards are needed for conservation and management efforts. To develop molecular genetic tools for individual identification of hair and faecal samples, we screened 50 microsatellite loci developed for the domestic cat (Felis catus) in 19 captive snow leopards. Forty‐eight loci were polymorphic with numbers of alleles per locus ranging from two to 11. The probability of observing matching genotypes for unrelated individuals (2.1 × 10?11) and siblings (7.5 × 10?5) using the 10 most polymorphic loci was low, suggesting that this panel would easily discriminate among individuals in the wild.  相似文献

Fifteen microsatellite loci were developed for pink ling (Genypterus blacodes), a fish of significant commercial importance to Australasia. Nine loci were examined in samples from five regions of Australia’s South East Fishery. All nine were highly polymorphic; numbers of alleles per locus ranged from 11 to 52 in total samples of 270–306 individuals. The average observed heterozygosity per locus per sample (0.823) was a little lower than the average Hardy–Weinberg expected heterozygosity per sample (0.895), perhaps reflecting the possible presence of null alleles at two loci. There was no significant evidence of genetic stock heterogeneity.  相似文献

We characterized 20 new microsatellite loci isolated from Parnassius apollo nevadensis. The 20 loci displayed 2–12 alleles and expected heterozygosities varied from 0.22 to 0.89; the markers were amplified in nine multiplex reactions. The combined non-exclusion probabilities of this set are 0.001084 and 0.000005 for the first and second parents, respectively. This set of markers will thus enable other studies on reproductive strategies and population genetics in this endangered and previously understudied species. We show, for the first time, evidence of multiple paternity and that females may successfully copulate with more than one male despite the presence of mating plugs. Future studies on this mating behavior and population genetics of the species using the 20 loci here presented will be very important to define its conservation status and management measures.  相似文献

The central stoneroller (Campostoma anomalum) is an abundant, widespread and sexually dimorphic stream minnow that is a useful model for mating system studies as well as a sentinel species for understanding population-level processes for fishes in headwater communities. We developed one genomic library enriched for dinucleotide repeats and isolated 48 putative, novel microsatellite loci. Of those, we present 32 polymorphic and independent microsatellite markers with 3 to 16 alleles per locus and heterozygosity ranging from 0.23 to 0.95. Hence, these markers will be useful for future behavioural, ecological and evolutionary studies using C. anomalum.  相似文献

Eleven microsatellite loci were developed in the swordfish, Xiphias gladius, to examine population structure. Loci were amplified in samples from two geographically distant populations (Ecuador and Mediterranean Sea). Observed heterozygosity ranged from 0.150 to 0.960. Most loci conformed to Hardy–Weinberg expectations with the exception of locus Xg‐59, which had a heterozygote deficit caused probably by null alleles. Locus Xg‐402 did not show significant divergence between the populations. The remaining nine loci showed statistically significant spatial heterogeneity (genic differentiation) between the two populations, making them useful for future studies of population structure in swordfish.  相似文献
