 The localization of osteopontin (OP) was examined in Meckel’s cartilage cells that bipotentially expressed cartilage and bone phenotypes during cellular transformation in vitro. Cultured cells were analyzed by in situ hybridization, immunostaining followed by light and electron microscopy, electron microscopy, and electron probe microanalysis. The combination of ultrastructural analysis and immunoperoxidase staining indicated that OP-synthesizing cells were cells that were autonomously undergoing a change from chondrocytes to bone-forming cells at the top of nodules. Double immunofluorescence staining of 2-week-old cultures revealed that OP was first synthesized by chondrocytic cells at the top of nodules. After further time in culture, the distribution of OP expanded from the central toward the peripheral regions of the nodules. Electron probe microanalysis revealed that the localization of OP was associated with matrices of calcified cartilage and osteoid nodules that contained calcium and phosphorus. Immunoperoxidase electron microscopy revealed that, in addition to the intracellular immunoreactivity in chondrocytes and small round cells that were undergoing transformation, matrix foci of calcospherites and matrix vesicles, in particular, included growing crystals that were immunopositive for OP. An intense signal due to mRNA for OP in 3-week-old cultures was detected in nodule-forming round cells, while fibroblastic cells, spreading in a monolayer over the periphery of nodules, were only weakly labeled. These findings indicate that OP might be expressed sequentially by chondrocytes and by cells that are transdifferentiating further and exhibit an osteocytic phenotype, and moreover, that expression of OP is closely associated with calcifying foci in the extracellular matrix. Summary Induced interferon- (IFN-) mRNA was localized in human FS-4 fibroblasts by in situ hybridization using biotinylated probes. The hybridization sites were detected by incubation with a nick-translated genomic DNA probe (1.8 kb) via streptavidin-colloidal gold followed by silver contrast enhancement. The positive signals were observed by reflection-contrast light microscopy. IFN- mRNA was transiently induced by poly r(I):r(C) in fibroblasts 2–4 h after induction. Induction in the presence of cycloheximide and actinomycin D (superinduction conditions) exhibited an enhanced level of IFN- mRNA with a maximum at 4–8 h. The kinetics of the IFN- mRNA expression in the cytoplasm as revealed by in situ hybridization proved to be compatible with the results of Northern biotting experiments of total cellular RNA.  相似文献

Summary Turtle's retinae were incubated in isotonic Ringer and then studied with the electron microscope in order to determine if in these conditions an extracellular space appeared. In the incubated retinae, as in the normal ones no extracellular space was observed. The distance between adjacent membranes is of 120–250 Å and is filled with a cementing material.The Müller cell of the turtle's retina is described. The existence, at the choroidal end of this cell, of villous projections filled with vesicles and the accumulation of mitochondria under these projections would suggest that a process of micropinocytosis takes place in this site, and that it may be metabolically more active. After incubation, Müller cells showed no appreciable changes, in contrast with what happens to astrocytes in incubated slices of CNS (Gerschenfeld et al. 1959).The addition of sodium-l-glutamate to the incubation medium determined a marked swelling of the nervous endings in the inner synaptic layer and a similar though less pronounced change in the ganglion cells. This would indicate that the surcharge of water, sodium and chloride produced by glutamate (Ames 1956) is localized in this layer.The selective action of glutamate on the inner synaptic layer, in contrast with the excellent preservation of the outer synaptic layer, is probably linked to differences in biochemistry and physiology of synapses in these two strata.Work supported by a research grant from the Air Force Office of Scientific Research (5–60), U.S.A.  相似文献

In order to understand the functional significance of Na,K-ATPase subunits as well as their isoenzymes, a precise subcellular localization of these in the myocyte is a crucial prerequisite. Cytochemical, immunofluorescence, preembedding immunogold and horse radish peroxidasediaminobenzidine methods, demonstrated 1 isoenzyme immunoreactivity on the sarcolemma, T-tubules and the subsarcolemmal cisterns of the adult cardiac myocytes. Cytochemically, ouabain resistant Na,K-ATPase precipitate was localized only in the subsarcolemmal cisterns and junctional sarcoplasmic reticulum. For 2 isoenzyme, immunoreactivity was demonstrated on the sarcolemma as well as in all areas of the myocytes in particularly a close proximation to the sarcoplasmic reticulum and microsomes. For 3 isoenzyme, only a weak insignificant signal was noted on the sarcolemma, intercalated disc and sarcoplasm. It is suggested that cytochemical ouabain resistant precipitate present in subsarcolemmal cisterns and junctional sarcoplasmic reticulum represent 1 isoenzyme of Na,K-ATPase. A differential as well as unique localization of subunit isoenzymes of Na,K-ATPase in specific structures of cardiac myocytes may suggest importance in physiological function at these sites.  相似文献

Summary For unexplained reasons, nerve growth factor (NGF) exists in very high concentrations in the submandibular gland of the mouse. The NGF in the gland, called 7S-NGF, is a non-covalent complex of three protein subunits, named -, - and -NGF. All the known biological activity resides in the -NGF subunit, and previous studies have shown that -NGF is present in much greater concentrations in the male submandibular gland than in the female gland. The higher concentration in the male is due to the fact that -NGF is synthesized in the granular tubule cells of the submandibular gland. These cells are much more numerous in the male gland.In contrast to -NGF, neither the concentrations of and subunits nor their cellular localization in the mouse submandibular gland have been established. In this study, radioimmunoassays specific for . and subunits determined that both are present in much higher concentrations in the male gland. Immunocytochemical work localized both subunits in the granular tubule cell in the male and female submandibular gland. This indicates that all the components of 7S-NGF exist in a single cell type in the gland and suggests that 7S-NGF can be formed within this cell and secreted as a complex into the saliva.  相似文献

Neuropathic pain may arise following peripheral nerve injury though the molecular mechanisms associated with this are unclear. We used proteomic profiling to examine changes in protein expression associated with the formation of hyper-excitable neuromas derived from rodent saphenous nerves. A two-dimensional difference gel electrophoresis (2D-DIGE) profiling strategy was employed to examine protein expression changes between developing neuromas and normal nerves in whole tissue lysates. We found around 200 proteins which displayed a >1.75-fold change in expression between neuroma and normal nerve and identified 55 of these proteins using mass spectrometry. We also used immunoblotting to examine the expression of low-abundance ion channels Nav1.3, Nav1.8 and calcium channel α2δ-1 subunit in this model, since they have previously been implicated in neuronal hyperexcitability associated with neuropathic pain. Finally, S35methionine in vitro labelling of neuroma and control samples was used to demonstrate local protein synthesis of neuron-specific genes. A number of cytoskeletal proteins, enzymes and proteins associated with oxidative stress were up-regulated in neuromas, whilst overall levels of voltage-gated ion channel proteins were unaffected. We conclude that altered mRNA levels reported in the somata of damaged DRG neurons do not necessarily reflect levels of altered proteins in hyper-excitable damaged nerve endings. An altered repertoire of protein expression, local protein synthesis and topological re-arrangements of ion channels may all play important roles in neuroma hyper-excitability.  相似文献

The immunohistochemical expression of the alpha and beta subunits of S-100 protein in reactive, modified and transformed of myoepithelial cells, salivary pleomorphic was investigated using monoclonal antibodies. With S-100 alpha, normal salivary glands showed strong staining in serous acinar cells and moderate to slight staining in ductal segments, and with S-100 beta staining was slight or negative in acinar cells, but strong in nerve fibres. In pleomorphic salivary adenomas, the immunohistochemical distribution of S-100 alpha and beta proteins indicated great variation in the tumour cells. Some neoplastic cells gave similar staining for both S-100 alpha and beta, others were strongly positive for S-100 alpha and stained only slightly for S-100 beta, or vice versa. Yet other cells were positive for S-100 alpha and negative for S-100 beta, or vice versa. Pleomorphic salivary adenomas were classified both by histopathological criteria and by their staining pattern for S-100 alpha and beta proteins. Great heterogeneity in S-100 alpha and beta protein expression was found in individual tumour cells of both ductal and myoepithelial origin, and no regular pattern was identified. The cellular origin of salivary pleomorphic adenomas is discussed in terms of S-100 alpha and beta protein immunohistochemistry. Pleomorphic adenoma cells may be transformed from reserve cells into tumour cells displaying biologic properties of myoepithelial cells, ductal cells, or a mixture of both.  相似文献

Summary Using anti-human calcitonin serum and a protein !-gold technique, calcitonin was localized at the ultrastructural level in control and calcium gluconate-stimulated thyroid C cells of the rat. Metacercariae of the genus Diplostomum dwelling in the retina of perch Perca fluviatilis and the lens of roach Rutilus rutilus were identified on the basis of adults obtained by feeding various piscivorous birds of the families Laridae, Anatidae and Phasianidae with metacercariae on an experimental basis. Both morphological studies of the metacercariae and adults obtained from the intestine of the infected birds, as well as the suitability in these hosts, indicated that we were dealing with two different species of Diplostomum. The perch form was recognised as D. baeri Dubois, 1937. Although metacercariae of this species matured both in herring gull Larus argentatus and common tern Sterna hirundo, the principal final host seemed to be goosanders Mergus merganser. In contrast, the lens form in roach was identified as D. spathaceum (Rudolphi, 1819) and it seemed to be restricted to birds of the family Laridae. These experimental results agree with parasitological findings from birds infected in the wild.  相似文献

Summary The localization of S-100 protein was studied in histological sections of retinae from adult rabbits. By use of double-immunolabeling techniques it was shown that most but not all radially oriented vimentin-positive Müller cells were co-labeled by an antiserum to S-100 protein. Glial fibrillary acidic protein-positive astrocytes, which in the rabbit retina are restricted to the medullary rays formed by myelinated optic nerve fibers, consistently showed S-100 protein immunoreactivity. The present report shows that, with respect to S-100 protein staining, Müller cells represent a heterogeneous population of glial elements.  相似文献

J. Neurochem. (2012) 122, 1095-1107. ABSTRACT: Ca(2+) channel β subunits determine the maturation, biophysical properties and cell surface expression of high voltage-activated channels. Thus, we have analysed the expression, regional distribution and subcellular localization of the Ca(v) β subunit family in mice from birth to adulthood. In the hippocampus and cerebellum, Ca(v) β(1) , Ca(v) β(3) and Ca(v) β(4) protein levels increased with age, although there were marked region- and developmental stage-specific differences in their expression. Ca(v) β(1) was predominantly expressed in the strata oriens and radiatum of the hippocampus, and only weakly in the cerebellum. The Ca(v) β(3) subunit was mainly expressed in the strata radiatum and lucidum of the hippocampus and in the molecular layer of the cerebellum. During development, Ca(v) β(3) protein expression in the cerebellum peaked at postnatal days (P) 15 and 21, and had diminished drastically by P60, and in the hippocampus increased with age throughout all subfields. Ca(v) β(4) protein was detected throughout the cerebellum, particularly in the molecular layer, and in contrast to the other subunits, Ca(v) β(4) was mainly detected in the molecular layer and the hilus of the hippocampus. At the subcellular level, Ca(v) β(1) and Ca(v) β(3) were predominantly located post-synaptically in hippocampal pyramidal cells and cerebellar Purkinje cells. Ca(v) β(4) subunits were detected in the pre-synaptic and post-synaptic compartments of both regions, albeit more strongly at post-synaptic sites. These results shed new light on the developmental regulation and subcellular localization of Ca(v) β subunits, and their possible role in pre- and post-synaptic transmission.  相似文献

Abstract

The aim of our study was to determine the distribution of estrogen receptor α (ERα) and progesterone receptor B (PR-B) in the bovine oviduct during the follicular and luteal phases. Bovine oviducts from 23 animals were obtained from a local slaughterhouse. Blood samples from these animals also were taken before death to measure estrogen and progesterone levels. The serum levels of estradiol-17β and progesterone changed during the estrous cycle. Tissue distribution of ERα and PR-B was examined using immunohistochemical techniques and the results showed that ERα and PR-B were stained in nuclei of cells and could be detected in all compartments along the entire oviduct during both the follicular and luteal phases. During the follicular phase, no significant differences were found between ERα and PR-B distribution (p < 0.05), while significant differences were found between ERα and PR-B during the luteal phase (p < 0.05). We results indicated that the frequency and intensity of ERα and PR-β immunoreactivity in the oviduct of bovines varied according to the oviductal cell types and the phases of the sexual cycle.  相似文献

Summary. Although there are a great number of studies concerning the uptake of taurine in several tissues, the regulation of taurine transport has not been studied in the retina after lesioning the optic nerve. In the present study, isolated retinal cells of the goldfish retina were used either immediatly after cell suspension or in culture. The high-affinity transport system of [3H]taurine in these cells was sodium-, temperature- and energy-dependent, and was inhibited by hypotaurine and β-alanine, but not by γ-aminobutyric acid. There was a decrease in the maximal velocity (Vmax) without modifications in the substrate affinity (Km) after optic axotomy. These changes were mantained for up to 15 days after the lesion. The results might be the summation of mechanisms for providing extracellular taurine to be taken up by other retinal cells or eye structures, or regulation by the substrate taurine, which increases after lesioning the optic nerve. The in vivo accumulation of [3H]taurine in the retina after intraocular injection of [3H]taurine was affected by crushing the optic nerve or by axotomy. A progressive retinal decrease in taurine transport was observed after crushing the optic nerve, starting at 7 hours after surgery on the nerve. The uptake of [3H]taurine by the tectum was compensated in the animals that were subjected to crushing of the optic nerve, since the concentration of [3H]taurine was only different from the control value 24 hours after the lesion, indicating an efficient transport by the remaining axons. On the contrary, the low levels of [3H]taurine in the tectum after axotomy might be an index of the non-axonal origin of taurine in the tectum. Axonal transport was illustrated by the differential presence of [3H]taurine in the intact or crushed optic nerve. The uptake of [3H]taurine into retinal cells in culture in the absence or in the presence of taurine might indicate the existence of an adaptive regulation of taurine transport in this tissue, however taurine transport probably differentially occurs in specific populations of retinal cells. The use of a purified preparation of cells might be useful for future studies on the modulation of taurine transport by taurine in the retina and its role during regeneration.
