Gas exchanges, chlorophyll (Chl) a fluorescence and carboxylation activities of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) and phosphoenolpyruvate carboxylase (PEPC) were determined in tomato (Lycopersicon esculentum Mill.) fruits picked at different developmental stages (immature, red-turning, mature, and over-ripe). The fruits did not show signs of CO2 fixation. However, photochemical activity was detectable and an effective electron transport was observed, the values of Chl fluorescence parameters in green fruits being similar to those determined in the leaves. The RuBPCO activity, which was similar to those recorded in the leaves at the immature stage of the fruit, decreased as the fruit ripened. PEPC activity was always higher than RuBPCO activity.  相似文献

In spring and winter cultivars of oilseed rape (Brassica napus var. oleifera), acclimation of photosynthetic apparatus to cold was connected with the increase in activities of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) and sucrose-phosphate synthase (SPS). Conversely, cold de-acclimation entailed the decline of RuBPCO and SPS activities. The rate of this photosynthetic de-acclimation might depend on day temperature. On the other hand, temperature rise during de-acclimation (identical during the day and night) resulted in the improvement of photosynthetic activity measured by means of chlorophyll fluorescence. An increase in SPS activity (and even transitory increase in RuBPCO activity) was observed when the elongation growth rate (EGR) accelerated during de-acclimation. Throughout re-acclimation, plants with high EGR were unable to maintain or recover higher photosynthetic capacity, despite the fact that SPS activity remained high or even increased during re-acclimation.  相似文献

The alterations of organization of leaf tissues and cell ultrastructure as a consequence of salt stress (75 and 150 mM NaCl) were studied in two tomato (Lycopersicum esculentum Mill.) cultivars showing different salinity tolerance. The salinity brought changes in cell shape, volume of intercellular spaces and chloroplast number, shape and size. These characteristics were specific in each cultivar. The ultrastructural changes were also different in the two tomato cultivars studied and the most important ones were in the number and size of starch granules in chloroplasts, the number of electron-dense corpuscules in the cytoplasm, the structure of mitochondria, and number of plastoglobuli. Activation state of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) is an important parameter determining the rate of net photosynthesis (P N) in situ for which no information is available with reference to altitude. We analyzed activation state along with P N in three plant species and their cultivars grown at low (LA, 1 300 m) and high (HA, 4 200 m) altitudes. No significant change in P N and the initial activity of RuBPCO was obtained with reference to altitude. However, activation state of RuBPCO was reduced significantly in the HA plants as compared to the LA ones. Hence low partial pressure of CO2 prevailing at HA might be responsible for the lower activation state of RuBPCO.  相似文献

以欧美107杨(Populus×euramericana ‘Neva',Wt)和转拟南芥液泡膜Na~+/H~+逆向转运蛋白基因AtNHX1的欧美107杨新品系(Tr) 幼苗为材料,研究了高低度盐胁迫对两品系幼苗光合色素含量、光合参数和叶绿体超微结构的影响,以阐明转AtNHX1基因杨树的耐盐性与其光合作用及叶绿体结构之间的关系.结果表明:(1)盐处理后,两品系叶片叶绿素含量、类胡萝卜素含量、净光合速率、蒸腾速率和气孔导度均下降,且高盐度处理下降幅度更大;同等盐度处理下,Tr品系叶片叶绿素含量、净光合速率和气孔导度的下降幅度显著低于Wt品系,且在高盐度处理间差异更大;两品系杨树叶片P_n下降的原因在低盐处理时以气孔限制为主,而在高盐下则是气孔限制和非气孔限制共同作用的结果.(2)盐胁迫对T_r 品系叶片叶绿体超微结构的影响较轻,其在高盐下仍保持了较好的内部结构;盐胁迫Wt品系叶绿体则缩皱成球形,内部结构趋向简单,以至解体,脂质球显著增多.可见,盐胁迫导致杨树叶绿体结构破坏而引起叶绿体色素含量下降,最终降低其光合作用效率;同等盐度胁迫下,转AtNHX1基因品系叶片保持了较完整的叶绿体超微结构、更高的叶绿素含量,能维持较好的光合状态,从而表现出较高的耐盐能力.  相似文献

Uniculm wheat (Triticum aestivum L.) was grown to maturity at four concentrations of nitrogen corresponding to 3 (N1), 6 (N2), 9 (N3) and 12 (N4) g m–2. Penultimate and flag leaves were examined throughout the ontogeny. Sub-optimal concentrations of N resulted in sharp decline in both area and dry mass of the leaves. Decline in leaf area was due to fewer mesophyll cells. Net photosynthetic rate (PN) increased up to full expansion, remained constant for about a week and then declined. PN, nitrogen, ribulose-1,5-bis-phosphate carboxylase/oxygenase (RuBPCO) amount and activity, chlorophyll and soluble protein contents were similar at all the N concentrations. Both amount and activity of RuBPCO in the flag leaf were about two fold higher as compared to penultimate leaf, but PN was similar. This indicates the presence of an excess amount of RuBPCO in the flag leaf.  相似文献

Littleseed canarygrass (Phalaris minor Retz.) is a major weed in wheat fields, and has developed resistance to the commonly used herbicide isoproturon. This study explores the potential use of isoxaflutole, a pre-emergence herbicide, to control littleseed canarygrass. Greenhouse studies were carried out to determine the phytotoxicity of isoxaflutole in relation to shoot height, fresh shoot biomass and leaf chlorophyll concentration of wheat and littleseed canarygrass. Electron microscopy was used to examine any damage to leaf chloroplast at ultrastructural level. Results indicate that isoxaflutole (0.5 mg/L) significantly reduced the shoot height of littleseed canarygrass (39.6%), but no significant reduction in the shoot height of wheat was observed (9.6%) when compared to control. None of the concentrations (0.05, 0.1, 0.5 and 1 mg/L) of isoxaflutole altered soil chemistry in relation to pH, organic matter, macro or micro inorganic ions. While untreated littleseed canarygrass leaves had elongated chloroplast, starch grains and small number of plastoglobuli; treated littleseed canarygrass leaves had swollen chloroplast, large number of plastoglobuli, and a lack of starch grains. We conclude that isoxaflutole can be an effective herbicide for controlling littleseed canarygrass.  相似文献

Leaf samples of Mn-deficient and Mn-sufficient (control) ‘Navelate’ orange plants grown in a greenhouse were taken to investigate the effects of Mn deficiency in leaf structure and chloroplast ultrastructure. Total leaf chlorophyll concentration was significantly lower in Mn-deficient plants than in control ones. Entire lamina thickness was not altered due to Mn deficiency. However, Mn deficiency resulted in disorganization of mesophyll cells, mainly of palisade parenchyma cells. The number of mesophyll chloroplasts per cellular area and their length were both affected negatively. The membranous system of chloroplasts was also disorganized. The percentages of starch grains and plastoglobuli per chloroplast of Mn-deficient leaves were significantly greater than those of control leaves.  相似文献

A series of experiments were conducted to characterize the water stress-induced changes in the activities of RuBP carboxylase (RuBPCO) and sucrose phosphate synthase (SPS), photosystem 2 activity, and contents of chlorophylls, carotenoids, starch, sucrose, amino acids, free proline, proteins and nucleic acids in mulberry (Morus alba L. cv. K-2) leaves. Water stress progressively reduced the activities of RuBPCO and SPS in the leaf extracts, the chlorophyll content, and PS2 activity in isolated chloroplasts. Plants exposed to drought showed lower content of starch and sucrose but higher total sugar content than control plants. While the soluble protein content decreased under water stress, the amino acid content increased. Proline accumulation (2.5-fold) was noticed in stressed leaves. A reduction in the contents of DNA and RNA was observed. Reduced nitrogen content was associated with the reduction in nitrate reductase activity. SDS-PAGE protein profile showed few additional proteins (78 and 92 kDa) in the water stressed plants compared to control plants.  相似文献

Plant growth, leaf protein and chlorophyll content, and chloroplastultrastructure as affected by nitrogen (N) were examined infour rice (Oryza sativa L.) cultivars grown in culture solutionunder controlled conditions. Increasing N concentration generallyincreased height and shoot dry weight of all cultivars. Cultivardifferences were significant at normal N level (40 ppm). Amongcultivars, IR8 was most responsive to increasing N, having thesignificantly highest shoot dry weight and protein content.Total chlorophyll and protein contents varied with cultivarand N, but chlorophyll a/b ratio remained constant. At the ultrastructurallevel, chloroplasts had generally well-developed grana and stromalamellae at 40 ppm.N. Chloroplasts at high N had from one tofour times as many grana as the N-deficient chloroplasts. Nitrogendeficiency reduced the size of the chloroplast, grana-stromalamellae and resulted in fewer poorly stacked grana. Increasingthe N level (120 ppm) above the normal level did not significantlyaffect chloroplast size of any cultivar, except for IR8 whichhad the largest chloroplast. A reduction in the number of starchgrains was observed in IR8, but more were present in ER36 underN-deficient conditions. The size of starch grains was not affectedby N and did not differ among cultivars. Plastoglobuli appearedto be larger under N-deficient conditions. Nitrogen had no effecton the number of plastoglobuli but cultivar differences existed.The highly N-responsive IR8 (based on dry weight) had the largestchloroplast which increased with N level. The increase in chloroplastsize accounted for the increase in both chlorophyll and proteincontents and, consequently, dry weight. Increased oxidative stress displayed during dark-senescence of wheat leaves (Triticum aestivum L.) is caused not only by the increased levels of radicals but also by a loss of antioxidant capacity. Mature leaves were incubated in 6-benzylaminopurine (BAP 10−4 M) or water (control) during 6 d in the dark. The senescence-delaying effect of BAP was associated with the retention of the chloroplast structure, 60% of the initial content of chlorophyll (Chl) and 77% of the initial content of protein. BAP reduced the degradation of the light-harvesting chlorophyll a/b binding protein (LHCP-2), and the large (LSU) and small subunits (SSU) of Rubisco. Our results indicated that the presence of the NADPH:protochlorophyllide oxidoreductase (POR, EC.1.6.99.1) was not promoted by the cytokinin, leading to the conclusion that BAP maintains the level of Chl, preventing its degradation, rather than inducing Chl biosynthesis. The internal structure of chloroplasts was maintained in BAP-treated leaves for up to 6 d, with well-organized grana thylakoids and small plastoglobuli; in contrast, chloroplasts of control leaves deteriorated rapidly from day 4 with disorganized internal membranes, and more and larger plastoglobuli. BAP increased the activities of catalase (CAT, EC 1.11.1.6) and ascorbate peroxidase (APX, EC 1.11.1.11) and reduced the level of H2O2 in the delayed-senescence tissue. The present research indicates that BAP reduces levels of reactive oxygen species (ROS), and enhances the activity of antioxidant enzymes (CAT, APX). Our results suggest that BAP protects the cell membranes and the photosynthetic machinery from oxidative damage during delay of senescence in the dark.  相似文献

Three types of transgenic plants of Solanum tuberosum cvs. Kamyk and Oreb, and Nicotiana tabacum cvs. Maryland Mammoth and Trapezond were selected according to intensity of introduced ipt gene expression and resulting amount of synthesised cytokinins (CKs). In comparison with controls, original transgenic regenerants grown in vitro showed a massive increase of CK contents, in tobacco by 379 % and in potato by 159 % (MAS). Potato grown in soil from tubers of transgenic plants demonstrated a moderate increase (44 %) of CK contents (MOD). Transgenic tobacco grown from seeds in vitro did not show any significant change in CK contents (NOT). Initial (RuBPCi and RuBPOi) and total (RuBPCt) activities of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO), and the activity of phosphoenolpyruvate carboxylase (PEPC) were not significantly affected by the transformation in the NOT plants. In the MOD plants, the RuBPCO activities were stimulated by up to 34 % whereas the PEPC activity was decreased by 17 %. On the other hand, all the measured enzyme activities were 32 – 91 % lower in the MAS. Leaf area, fresh and dry masses, and chlorophyll and soluble protein contents also went down with increasing CK amounts in the transformants. Dependence of RuBPCi/RuBPOi and RuBPCt/PEPC ratios on the relative CK amounts in transgenic plants revealed that the individual enzyme activities were not affected uniformly. Endogenous CK contents in the MAS thus apparently exceeded an optimum needed for positive effects on many physiological traits and became a stress factor for such plants.  相似文献

Abstract. Peas were grown in controlled environments (12h white fluorescent light. ∼47 μmol photons m-2 s 1/12 dark, 25 °C), using (1) 15-min far-red illumination at the end of each photoperiod (brief FR) to simulate the increase in the far-red/red ratio near the end of the day, and (2) high levels of supplementary far-red light (red:far-red ratio=0.04) during the entire photoperiod (long-term FR) to simulate extreme shade conditions under a plant canopy. Brief FR illumination led to marked morphological effects attributable to phytochrome regulation, namely, an increase in internodal length, but a decrease in leaflet area, chloroplast size and chlorophyll content per chloroplast compared with the control. Significantly, brief FR illumination had little or no effect on the amounts of the major chloroplast components (ribulose 1.5-biphosphate carboxylase, adenosine triphosphate synthase, cytochrome b/f complex and Photosystem II) relative to chlorophyll or Photosystem I, and the leaf photosynthetic capacities per unit chlorophyll were similar. In contrast, supplementing high levels of far-red light during the entire photoperiod not only led to the phytochrome effects above, but there was also a marked increase in leaf photosynthetic capacity per unit chlorophyll. due to increased amounts of the major chloroplast components relative to chlorophyll or Photosystem I. We hypothesize that supplementary far-red light, absorbed by Photosystem I, induced an increase in the major chloroplast components by a photosynthetic feedback mechanism. In fully greened leaves, we propose that the two photosystems themselves, rather than phytochrome, may be the predominent sensors of light quantity in triggering modulations of the stoichiometries of chloroplast components, which in turn lead to varying photosynthetic capacities.  相似文献

Effects of plant hormones indole-3-yl-acetic acid (IAA), gibberellic acid (GA), benzylaminopurine (BAP), abscisic acid (ABA) and ethrel (ETH) in 5 M concentration on gas exchange, ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO, EC 4.1.1.39) activity, pigment content and yield in cotton (Gossypium hirsutum L. cv. H-777) under drought were studied. At reproductive stage (55 – 60 d after sowing) these hormones were sprayed on shoots one day prior to stress imposition by withholding irrigation. The soil moisture of control plants was kept at field capacity. Net photosynthetic rate (PN), stomatal conductance (gs), transpiration rate (E), carboxylation efficiency (CE), water use efficiency (WUE), RuBPCO activity, boll number per plant, seed number per plant and lint mass per plant significantly decreased at drought while chlorophyll (Chl) b content and flower number per plant increased. ABA and ETH significantly reduced gas exchange parameters, Chl a and Chl b content. Detrimental drought effect on PN, gs, E, CE, RuBPCO and lint mass per plant was significantly alleviated by BAP and also its effect on seed number and lint mass per plant was significantly alleviated with the ABA treatment.  相似文献

BACKGROUND AND AIMS: Ozone effects on peatland vegetation are poorly understood. Since stress responses are often first visible in cell ultrastructure, electron microscopy was used to assess the sensitivity of common peatland plants to elevated ozone concentrations. METHODS: Three moss species (Sphagnum angustifolium, S. magellanicum and S. papillosum), a graminoid (Eriophorum vaginatum) and two dwarf shrubs (Vaccinium oxycoccus and Andromeda polifolia), all growing within an intact canopy on peat monoliths, were exposed to a concentration of 0, 50, 100 or 150 ppb ozone in two separate growth chamber experiments simulating either summer or autumn conditions in central Finland. After a 4- or 5-week-long exposure, samples were photographed in a transmission electron microscope and analysed quantitatively using image processing software. KEY RESULTS: In the chlorophyllose cells of the Sphagnum moss leaves from the capitulum, ozone exposure led to a decrease in chloroplast area and in granum stack thickness and various changes in plastoglobuli and cell wall thickness, depending on the species and the experiment. In E. vaginatum, ozone exposure significantly reduced chloroplast cross-sectional areas and the amount of starch, whereas there were no clear changes in the plastoglobuli. In the dwarf shrubs, ozone induced thickening of the cell wall and an increase in the size of plastoglobuli under summer conditions. In contrast, under autumn conditions the cell wall thickness remained unchanged but ozone exposure led to a transient increase in the chloroplast and starch areas, and in the number and size of plastoglobuli. CONCLUSIONS: Ozone responses in the Sphagnum mosses were comparable to typical ozone stress symptoms of higher plants, and indicated sensitivity especially in S. angustifolium. The responses in the dwarf shrubs suggest stimulation of photosynthesis by low ozone concentrations and ozone sensitivity only under cool autumn conditions.  相似文献

The effect of dark-chilling and subsequent photoactivation on chloroplast structure and arrangements of chlorophyll–protein complexes in thylakoid membranes was studied in chilling-tolerant (CT) pea and in chilling-sensitive (CS) tomato. Dark-chilling did not influence chlorophyll content and Chl a/b ratio in thylakoids of both species. A decline of Chl a fluorescence intensity and an increase of the ratio of fluorescence intensities of PSI and PSII at 120 K was observed after dark-chilling in thylakoids isolated from tomato, but not from pea leaves. Chilling of pea leaves induced an increase of the relative contribution of LHCII and PSII fluorescence. A substantial decrease of the LHCII/PSII fluorescence accompanied by an increase of that from LHCI/PSI was observed in thylakoids from chilled tomato leaves; both were attenuated by photoactivation. Chlorophyll fluorescence of bright grana discs in chloroplasts from dark-chilled leaves, detected by confocal laser scanning microscopy, was more condensed in pea but significantly dispersed in tomato, compared with control samples. The chloroplast images from transmission-electron microscopy revealed that dark-chilling induced an increase of the degree of grana stacking only in pea chloroplasts. Analyses of O-J-D-I-P fluorescence induction curves in leaves of CS tomato before and after recovery from chilling indicate changes in electron transport rates at acceptor- and donor side of PS II and an increase in antenna size. In CT pea leaves these effects were absent, except for a small but irreversible effect on PSII activity and antenna size. Thus, the differences in chloroplast structure between CS and CT plants, induced by dark-chilling are a consequence of different thylakoid supercomplexes rearrangements. A field experiment was conducted to investigate the changes in chlorophyll (Chl) and nitrogen (N) contents, ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) and phosphoenolpyruvate carboxylase (PEPC) contents and PEPC activity, and the photon-saturated net photosynthetic rate (P Nsat), and their relationships with leaf senescence in two maize hybrids with different senescent appearance. One stay-green (cv. P3845) and one earlier senescent (cv. Hokkou 55) hybrid were used in this study, and we found that Chl and N contents and the P Nsat in individual leaves of P3845 were greater than those in corresponding leaves of Hokkou 55 at the successive growth stages. In addition, larger contents of RuBPCO and PEPC, and a greater activity of PEPC were observed in P3845. Due to the lower rates of decrease of Chl, RuBPCO, and PEPC amounts per unit of N, and the lower net C translocation rate per unit of N in the stay-green hybrid, leaf senescence was delayed in comparison to the earlier senescent hybrid.  相似文献

Differences in acclimation to elevated growth CO2 (700 μmol mol−1, EC) and elevated temperature (ambient +4 °C, ET) in successive leaves of wheat were investigated in field chambers. At a common measurement CO2, EC increased photosynthesis and the quantum yield of electron transport (Φ) early on in the growth of penultimate leaves, and later decreased them. In contrast, EC did not change photosynthesis, and increased Φ at later growth stages in the flag leaf. Contents of chlorophyll (Chl), ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO), and total soluble protein were initially higher and subsequently lower in penultimate than flag leaves. EC decreased RuBPCO protein content relative to soluble protein and Chl contents throughout the development of penultimate leaves. On the other hand, EC initially increased the RuBPCO:Chl and Chl a/b ratios, but later decreased them in flag leaves. In the flag leaves but not in the penultimate leaves, ET initially decreased initial and specific RuBPCO activities at ambient CO2 (AC) and increased them at EC. Late in leaf growth, ET decreased Chl contents under AC in both kinds of leaves, and had no effect or a positive one under EC. Thus the differences between the two kinds of leaves were due to resource availability, and to EC-increased allocation of resources to photon harvesting in the penultimate leaves, but to increased allocation to carboxylation early on in growth, and to light harvesting subsequently, in the flag leaves.  相似文献

The total activity of aldolase (EC 4.1.2.13) and the activities of cytosol and chloroplast aldolase were determined in seeds, cotyledons, primary leaves and secondary leaves of spinach (Spinacia oleracea L., cv. Monopa) during germination. Total aldolase activity in cotyledons increased from low levels to a low maximum in the dark after one week and to a high maximum in white light after three to four weeks and declined thereafter. The activity in primary and secondary leaves started to rise strongly from the 18th and 26th days, respectively, up to the 42nd day of germination. The levels of aldolase activity paralleled the development of leaf area, chlorophyll content and protein content per leaf except that the leaf area of cotyledons continued to increase steadily up to the 42nd day after the maximum of aldolase activity was reached. Resolution of cytosol- and chloroplast-specific isoenzymes by chromatography on diethylaminoethylcellulose indicated that in the light the cytosol enzyme represented approx. 8% of the total activity in cotyledons, primary and secondary leaves throughout germination, and the chloroplast enzyme represented the remaining 92%. Only in cotyledons of dark-grown seedlings was the cytosol aldolase between 25 and 50% of the total activity. Seeds contained almost exclusively a cytosol aldolase. In cotyledons the increase of total activity in the light was specifically the consequence of an increase in chloroplast aldolase while the cytosol aldolase was little affected by light. The in vitro effect of cadmium on apical segments of Hypnea musciformis was examined. Over a period of 7?days, the segments were cultivated with different concentrations of cadmium, ranging from 50 to 300???M. The samples were processed for microscopic and histochemical analysis of growth rates, content of photosynthetic pigments, and photosynthetic performance. Cadmium treatments increased cell wall thickness and the accumulation of plastoglobuli. Destruction of chloroplast internal organization was observed. Compared to controls, algae exposed to cadmium showed growth rate reduction, depigmentation, and blending in the lateral branches. The content of photosynthetic pigments, including chlorophyll a and phycobiliproteins, decreased after exposure to different concentrations of cadmium. These results agree with the decreased photosynthetic performance and relative electron transport rate observed after exposure of algae to cadmium. Taken together, these findings strongly indicate that cadmium negatively affects the architecture and metabolism of the carragenophyte H. musciformis, thus posing a threat to the economic vitality of this red macroalgae.  相似文献
