Extracts obtained from hemolymph of sexually mature males of the Caribbean fruit fly Anastrepha suspensa contained four biologically important terpenoid components of the sex pheromone. The four components were identified as farnesene, bisabolene, anastrephin, and epianastrepin based on their relative retention indexes from capillary gas chromatography analysis, using both apolar and polar phase columns and their chemical ionization (isobutane) mass spectra. The ratio of the components in extracts of hemolymph was the same as the ratio present in the volatile blend of pheromone released by sexually mature males during the reproductive period. Studies conducted to determine the effect of age on amounts of these components in hemolymph indicated that they increased from undetectable levels on the day of adult emergence to maximum levels on day eight. The increases in amounts of the components present in hemolymph with increasing age were correlated with increases in amounts of volatile pheromone released by males. Time of day studies showed that the amounts of these components in hemolymph followed the daily pattern of release of volatile pheromone components. Other components of the sex pheromone including ocimene, (Z)-3-nonen-1-ol, (Z,Z)-3,6-nonadien-1-ol and suspensolide were not found in extracts of hemolymph. Sex pheromonal components of the tephritid fruit fly Bactrocera dorsalis (Hendel), 2-allyl-4,5-dimethoxyphenol and (E)-coniferyl alcohol, are biosynthesized from a highly potent male attractant, methyl eugenol, then sequestered and stored in the rectal gland prior to their release during courtship at dusk. These sex pheromonal components have been detected in the haemolymph and crop organ. Hence, attempts were made to separate and identify the haemolymph fractions which contained the sex pheromonal components. Identification of these bioactive fractions in methyl eugenol-fed male flies using gel filtration column chromatography and biodetection using live male flies showed two fractions as highly attractive to conspecific males. These fractions show a significant increase in protein absorbance in the elution profile of haemolymph from methyl eugenol-fed males compared with that from methyl eugenol-deprived males. The molecular mass of these bioactive fractions as determined by using gel filtration was in the peptide range of 3.3 to 5.5 kDa. Subsequent gas chromatography-mass spectrometry analyses further confirmed the presence of the pheromonal components in the bioactive fractions. The presence of these methyl eugenol-derived sex pheromonal components in specific haemolymph fractions suggests the involvement of a sex pheromone binding complex.  相似文献

In tephritid fruit flies of the genus Bactrocera Macquart, a group of plant derived compounds (sensu amplo ‘male lures’) enhance the mating success of males that have consumed them. For flies responding to the male lure methyl eugenol, this is due to the accumulation of chemicals derived from the male lure in the male rectal gland (site of pheromone synthesis) and the subsequent release of an attractive pheromone. Cuelure, raspberry ketone and zingerone are a second, related group of male lures to which many Bactrocera species respond. Raspberry ketone and cuelure are both known to accumulate in the rectal gland of males as raspberry ketone, but it is not known if the emitted male pheromone is subsequently altered in complexity or is more attractive to females. Using Bactrocera tryoni as our test insect, and cuelure and zingerone as our test chemicals, we assess: (i) lure accumulation in the rectal gland; (ii) if the lures are released exclusively in association with the male pheromone; and (iii) if the pheromone of lure-fed males is more attractive to females than the pheromone of lure-unfed males. As previously documented, we found cuelure was stored in its hydroxyl form of raspberry ketone, while zingerone was stored largely in an unaltered state. Small but consistent amounts of raspberry ketone and β-(4-hydroxy-3-methoxyphenyl)-propionic acid were also detected in zingerone-fed flies. Males released the ingested lures or their analogues, along with endogenous pheromone chemicals, only during the dusk courtship period. More females responded to squashed rectal glands extracted from flies fed on cuelure than to glands from control flies, while more females responded to the pheromone of calling cuelure-fed males than to control males. The response to zingerone treatments in both cases was not different from the control. The results show that male B. tryoni release ingested lures as part of their pheromone blend and, at least for cuelure, this attracts more females.  相似文献

The common brown centipede, Lithobius jorJcatus (L.), releases a sex-specific pheromone which emanates from distinctive cuticular openings on the coxae of the hind legs-the coxal pores (Littlewood & Blower, 1987). The pheromone can be isolated from tissue extracts of the coxal organs (surrounding the coxal pores) and from substrata which have been 'marked by male or female L. forjcatus and presented to centipedes in the form of a behavioural bio-assay.
In this paper, further evidence is presented for a centipede pheromone, the chemical components of male and female secretions are separated using Gas Layer Chromatography (GLC), a qualitative chemical analysis is described and the nature of the pheromone is discussed.  相似文献

After being acclimatized to feeding on fruit flies, the Asian house gecko, Hemidactylus frenatus Duméril & Bibron (Gekkonidae:Squamata), consumed fewer methyl eugenol (ME) fed male fruit flies, Bactrocera papayae Drew & Hancock (Tephritidae: Diptera) than when offered ME-deprived males. After one-day exposure to only ME-fed males, the geckos avoided feeding on female flies when in the presence of ME-fed males. When mechanically disturbed, the ME-fed males spontaneously ejaculated a rectal secretion which contains phenyl propanoids that deter the predator.The ME-fed males also competed significantly better than normal (ME-deprived) males for virgin females. Male B. papayae converts ME to three other phenyl propanoids which act in concert as a sex pheromone to attract females during courtship and as an allomone to the gecko.  相似文献

The search for effective female attractants emanating from the host or body of fruit flies has been an area of intensive research for over three decades. In the present study, bodies of male Mediterranean fruit flies, Ceratitis capitata (Wiedemann) (Diptera: Tephritidae), were extracted with diethyl ether or methanol and subjected to gas chromatography–mass spectrometry. Analysis revealed substantial qualitative and quantitative differences between males from a laboratory culture and wild males captured alive in an orchard. Most notably, the hydrocarbon sesquiterpene (±)‐α‐copaene, which is known to be involved in the sexual behaviour of the species, was found in substantial amounts in wild males, but was not detected in laboratory males. In laboratory tests, 15 laboratory or wild male equivalents of diethyl ether extracts or combined diethyl ether and methanol extracts, or, to a lesser extent, methanol extracts alone, were found to attract virgin females. In a citrus orchard, traps baited with combined diethyl ether and methanol extracts of wild males attracted significantly more virgin females than traps baited with various doses of pyranone or blends of other compounds identified in the extracts or reported in the literature, such as ethyl acetate, ethyl‐(E)‐3‐octenoate, and 1‐pyrroline. Traps baited with blends of compounds, however, displayed substantial attractiveness compared to control (non‐baited) traps. These results are important for better understanding the mating system of C. capitata as well as for further improving existing monitoring and control systems.  相似文献

To restore male mating competitiveness of Mexican fruit flies, Anastrepha ludens (Loew) (Diptera: Tephritidae), reared for sterile insect releases by the Mexican Fruit fly Eradication Campaign, two strain replacement techniques were evaluated. Field cage male competitiveness tests revealed that laboratory males of the Metapa strain mated 3 times less often with wild females than field-collected wild males. A strain developed from the cross of wild males and laboratory females (hybrid strain) was similar to a strain developed from the cross of laboratory males and females (laboratory strain) in that its females produced similar amounts of eggs and the eggs displayed similar levels of hatch and egg-to-pupa transformation in artificial diet. By contrast, a strain developed from the cross of wild males and females (wild strain), forced into artificial rearing, experienced a series of bottlenecks involving reduced egg laying and extremely poor development in diet. The male F1 progeny of the hybrid strain and field-collected wild males outcompeted Fl laboratory males in field cage tests for matings with field-collected wild females. In conclusion, we found that strains developed from the cross of wild males and laboratory females allowed us to restore male mating competitiveness of F1 Mexican fruit flies without compromising mass-rearing production.  相似文献

The MdSGHV is a double-stranded DNA virus that replicates in the salivary glands of infected adult house flies. Transmission of this non-occluded, enveloped virus is believed to be mediated orally via deposition and consumption of oral secretions composed of salivary gland secretions and crop contents. In this study, transmission electron micrographs of crops from infected flies showed numerous enveloped virions in the crop lumen adjacent to the cuticular intima, as well as on the hemocoel side in close vicinity to muscle cells. Oral treatments of newly emerged flies with viremic salivary gland homogenates, crop homogenates, or gradient-purified virus resulted in an average 44% infection. Virus released via oral secretion was infectious when ingested by newly emerged adult flies, resulting in an average 66% infection. Using quantitative real-time PCR, MdSGHV DNA was quantified in oral secretions and excreta obtained from viremic flies. Between 2 and 4 days post-infection (dpi), viral copy numbers in oral secretions increased exponentially and from 5 to 21 dpi each infected fly released an average 106 MdSGHV copies per feeding event. Excreta samples collected overnight from individual infected flies at 5 dpi contained an average 6.5 × 105 viral copies. Low but detectable infection rates were produced when newly emerged flies were challenged with excreta samples. In summary, evaluation of the quantity and infectivity of MdSGHV released by individual infected house flies clearly showed that deposition of oral secretions and excreta onto a shared food substrate is the main route of natural MdSGHV transmission among adult house flies.  相似文献

Ecdysteroid titres in whole flies and different tissues of adult male and female Drosophila were determined at various times after eclosion using a radioimmunoassay. The ecdysteroid titre decreased as the flies matured after eclosion. The differences in titre between males and females can be accounted for by their difference in body weight. The ecdysteroids were found to be distributed throughout several tissues. At eclosion not all of the ecdysteroid complement present could be accounted for by that found localised in tissues. After maturation of the flies the ecdysteroids in various tissues can account for the majority of that detected in whole-fly extracts. Ecdysteroids were produced during in vitro culture of various tissues, but the quantities detected were low by comparison with ring glands of wandering 3rd-instar larvae. Neither the ovaries nor the abdominal body walls (fat body) seem to be a major source of hormone, and they are only able to convert minute quantities of ecdysone to the biologically active form, 20-hydroxyecdysone, in vitro. The amounts of 20-hydroxyecdysone present were measured using high performance liquid chromatography and radioimmunoassay. We tentatively suggest that the differential experession of the yolk-protein-genes in the fat bodies of males and females does not result from differences in hormone titres between them.  相似文献

Volatile compounds from Drosophila melanogaster males and females dramatically affect male courtship behaviour. These substances, which have been extracted from flies of different ages and genotypes, have been analysed by gas chromatography (GC) and in behavioural assays. Extracts from virgin females and males have different gas chromatographic profiles, which may reflect the fact that extract from virgin females stimulates high levels of courtship between males over short distances, while extract from mature wild-type males does not affect sexual behaviour. However, volatile compounds from very young males or males expressing the fruitless (fru) mutation do stimulate courtship between males, and chromatographic profiles of young male and fru male extracts differ from the GC profile of extracts from mature wild-type males.  相似文献

Knowledge about Phortica variegata (Drosophilidae, Steganinae), the intermediate host of the eyeworm Thelazia callipaeda (Spirurida, Thelaziidae), is confined to experimental studies. To investigate the role P. variegata plays in the transmission of T. callipaeda under natural conditions, the population dynamics of these flies in the natural environment and their feeding preferences (on vegetables and/or animal lachrymal secretions) were examined. From April to November 2005, a total number of 969 (557 males and 412 females) P. variegata flies were collected weekly in a region of southern Italy with a history of canine thelaziosis. The flies were identified and dissected or subjected to a PCR assay specific for a region within the ribosomal ITS-1 DNA of T. callipaeda. The zoophilic preferences of P. variegata were assessed by collecting flies around the eyes of a person or around a fruit bait. Seven hundred and twenty flies (398 males and 322 females) were dissected under a stereomicroscope; 249 flies (158 males and 91 females) that died prior to the dissection were subjected to molecular investigation. Only P. variegata males were infected with larval T. callipaeda both at dissection (six, 0.83%) and with the specific PCR (seven, 2.81%), representing a total percentage of 1.34% flies infected. Interestingly, only males were collected around the eyes, compared with a male/female ratio of 1:4 around the fruit. This survey indicated that P. variegata males act as intermediate hosts of T. callipaeda under natural conditions in Europe. Both the zoophilic behaviour of P. variegata males on lachrymal secretions and their role as vector of T. callipaeda have been discussed as they represent a peculiarity in medical and veterinary entomology. The synchrony between the fly population dynamics and the biology of the nematode in the definitive host provides an interesting model for exploring the co-evolution of Thelazia spp. with their hosts.  相似文献

Mated Redback Spider Females Re-Advertise Receptivity Months after Mating   总被引:1,自引:0,他引:1  
In many species, selection acts on males to recognize female reproductive status at a distance using pheromones. Unmated females may actively seek to attract males; however, mated females may become cryptic to avoid attracting additional males if multiple matings are costly. Although females of many species cease pheromone production after mating, it is often unclear whether this is a strategic part of a female reproductive strategy, or whether this is because of chemical manipulation by males. If variation in pheromone production is part of the female’s strategy, then we predicted mated females should eventually re‐advertise receptivity if the benefits of multiple mating increase with time since copulation (e.g. because of sperm depletion). Here, we tested this prediction in Australian redback spiders (Latrodectus hasselti). First, we replicated earlier results by showing that virgin males discriminate female maturity and mating status based exclusively on web‐borne chemicals. Our results show this difference must arise from a change in chemical deposition in the web as we controlled for web volume differences between mated and virgin females. Male activity on extracts from webs of virgin females exceeded activity on a solvent control and on extracts of webs of just‐mated females—confirming that female redbacks cease pheromone production immediately after mating. Second, we tested a new prediction that mated females might re‐advertise receptivity near the end of their normal breeding season to replenish diminished sperm stores prior to overwintering. Consistent with the prediction of strategic advertisement, we show that male activity on extracts from females’ webs increased significantly 3 mo after the female first mated (typical length of the breeding season). Thus, these females had begun to add pheromone to their web again. At this time, 26% of these females re‐mated with a second male. If females re‐advertise receptivity to ensure adequate sperm stores, then we predicted a positive relationship between female reproductive output during the 3‐mo interval after copulation and the subsequent intensity of male response to web extracts. However, differences in male activity time were not related to the total number of spiderlings or the number of egg sacs a female had produced during the 3‐mo interval after the first copulation. This result could arise if male chemical manipulation of female receptivity decreases with time after copulation, or if the testing interval used in our study was too long to reveal variation in sperm depletion in females. Thus, although our results are consistent with the idea that females strategically alter pheromonal advertisement, we cannot distinguish this from the hypothesis that female receptivity arises from chemical manipulation by males.  相似文献

Wings and legs of the gregarious desert locust, Schistocerca gregaria have been shown to be release sites of phenylacetonitrile (PAN), the major adult male-produced pheromone. However, there is limited information on the distribution of PAN within the locust. Here we show, using gas chromatography-mass spectrometry (GC-MS), that PAN occurs in nearly all body parts of both adult males and females of the locust in varying amounts. PAN was 20-fold more concentrated in males than in females. In females, PAN was concentrated more in the tarsal segments. The greatest amounts of PAN were in 2- and 3-week old female and male body parts, respectively. No trace of PAN was found in similar ages and sexes of the solitarious phase desert locust. Our results show that PAN is distributed in the body matrix of both sexes of gregarious phase locusts and suggest that no specific tissue is responsible for biosynthesis of the pheromone.  相似文献

Although visually transmitted social signals are well documented in many diurnal iguanians, including collared lizards, secretory femoral glands also suggest a role of chemical signals in intraspecific communication. We conducted laboratory trials to test the extent to which male and female collared lizards responded by tongue‐flicking femoral gland secretions, neutral (water), and odoriferous (cologne) control substances, males distinguished self‐secretions from those produced by unfamiliar rival males, and females distinguish secretions from unfamiliar vs. familiar males. Both males and females spent similar amounts of time in four arena quadrants each with a Petri dish treated with one of the four test compounds. Males gave more tongue flicks/trial to secretions produced by unfamiliar rivals and cologne than they tongue‐flicked their own secretions and water. By contrast, the number of tongue flicks by females on control substances and familiar and unfamiliar males was similar. Results support the hypothesis that femoral gland secretions function in intrasexual signaling among male collared lizards, perhaps allowing them to distinguish unfamiliar rivals. Females tongue‐flicked secretions from familiar and unfamiliar males with similar frequency that was high relative to that of males, suggesting a possible role of secretions in assessment of males. Ours is the first evidence of a signaling role of femoral gland secretions in collared lizards and adds to a growing body of evidence that chemical signaling has evolved in diurnal lizards that also have highly developed visual‐based signaling.  相似文献

The presence of a pheromone biosynthesis activating neurohormone in the head gandlia, and its effect on the sex phermone biosynthetic pathway, were investigated in the tomato looper, Chrysodeixis chalcites (Esper). Comparison of pheromone components and precursor levels in the presence and absence of the factor was performed using untreated, ligated and ligated and injected virgin females. Pheromone glands of treated and untreated moths were extracted and analyzed by capillary gas chromatography for their most abundant pheromone components, (Z)-7-dodecenyl acetate and (Z)-9-tetradecenyl acetate, and the putative biosynthetic precursors hexadecanoate, (Z)-11-hexadecenoate, (Z)-9-tetradecenoate and (Z)-7-dodecenoate. Comparison of the amounts of the pheromone and precursor components in the three groups of females indicated that a neuroendocrine factor is involved in the regulation of the pheromone biosynthesis in C. chalcites. Lack of such a factor resulted in a marked decrease of the sex pheromone components as well as the three unsaturated putative biosynthetic precursors. However, no decrease was observed in the content of palmitoate, suggesting that the Δ11 desaturation step is affected by the neuroendocrine factor. Injection of head ganglia extracts into ligated females resulted in a recovery of unsaturated precursor and phermone content. Both male and female head ganglia were found to contain a sex pheromone biosynthesis regulatory factor. However, the stimulatory pattern of the factor from the two sexes was different, suggesting that the two factors are quantitatively and/or qualitatively distinct.  相似文献

哺乳动物的包皮腺分泌物对个体间性引诱及繁殖行为的信息交流和种群调节有着重要的作用。本研究以四川短尾鼩(Anourosorex squamipes)为研究对象对劳亚食虫目动物包皮腺挥发性化学成分和化学通讯功能进行报道。采用顶空固相微萃取—气质联用(HS-SPME-GC/MS)的方法,分析四川短尾鼩雄体包皮腺中挥发性物质的化学组成。结果表明:(1)四川短尾鼩雄体包皮腺的挥发性化学成分主要含烷烃类、醇类、酮类、醛类、醚类、酯类、酸类、芳香烃类等45种化合物;(2)成年雄性四川短尾鼩的包皮腺中挥发性成分有39种,幼年雄性含有28种,说明四川短尾鼩成年雄体的包皮腺中挥发性化学成分多于幼年雄体;(3)成年雄体的包皮腺分泌物中含有4种特有的挥发性化学成分,幼年雄体中特有成分仅为1种,表明四川短尾鼩包皮腺中的挥发性化学成分存在年龄差异;(4)四川短尾鼩包皮腺分泌物中不同化学成分相对含量不同,同种化学成分在不同个体间的相对含量存在差异。四川短尾鼩雄性包皮腺挥发性化合物种类丰富,个体、年龄差异明显。推测挥发性化合物中丁酸(Butanoicacid)、乙酸乙酯(Ethyl acetate)、苯酚(Phenol)为四川短尾鼩的信息素。本研究为进一步验证该物种的信息素成分及其传递机制提供了基础数据。  相似文献

The correlation between triacylglycerols containing conjugated diene fatty acyl moieties and pheromone aldehydes in the sex pheromone glands of females of Manduca sexta was investigated. Females decapitated 15 h after adult emergence neither called nor produced pheromone during the natural period of pheromone production on the subsequent two nights. However, these females could be stimulated to produce sex pheromone for prolonged periods by repeated injection of synthetic pheromone biosynthesis activating neuropeptide (PBAN). Gas chromatographic analysis of methanolysis products of lipids extracted from the pheromone glands of decapitated and intact females showed no differences in the amounts of fatty acyl precursors of pheromone. High performance liquid chromatographic analysis of the triacylglycerols containing conjugated diene analogues of the pheromone components (diene TG), obtained 24 and 48 h after decapitation, showed that the total amounts of these components were not affected by decapitation. The amounts of all diene TG peaks declined significantly when decapitated females were stimulated to produce pheromone during a 7 h period by repeated injection of PBAN at 3 h intervals but recovered when pheromone production subsided. These results indicate that PBAN induces liberation of pheromone precursors from the triacylglycerols during pheromone biosynthesis but does not induce replenishment of this storage pool. © 1996 Wiley-Liss, Inc.

    Neutering of part-fed females eliminated copulatory behavior inDermacentor variabilis (Say) andD. andersoni Stiles males. Extracts from the anterior reproductive tracts of part-fed (7 days) females partly restored the male copulatory behavior in conspecific neutered females. Similar extracts from unfed females did not restore the behavior, suggesting that the pheromone was produced during feeding. Perception of the genital sex pheromone by sensillae on the male cheliceral digits was confirmed by electrophysiological techniques.Males ofD. variabilis andD. andersoni responsed positively to authentic ecdysone and 20-hydroxyccdysone (20HE) in neutered female bioassays. Responses to sterols were significantly lower than to ecdysteroids. Electrophysiological assays suggest a sensitivity of males to high doses of ecdysteroids. The strongest responses were to 20HE in both species. Ecdysteroids, specifically ecdysone and 20HE were shown to be present in the anterior reproductive tracts in excess of amounts that could be explained by mere hemolymph contamination. Ecdysteroids were also found in washings of the vaginal lumen of these two species.Dermacentor andersoni females contained larger amounts of ecdysteroids thanD. variabilis females. 20-hydroxyecdysone and possibly ecdysone appear to be components of the genital sex pheromone ofD. variabilis andD. andersoni.     我们利用二氯甲烷抽提和气质联用(GC-MS)比较分析了正常和阉割布氏田鼠的包皮腺分泌物(PGS)成分。我们检测到33个成分,它们几乎在所有的被测布氏田鼠都存在,其中27个成分以前报道为昆虫的信息素组分。睾丸切除不能使任何一个成分完全消失,但是显著降低了10个首先从GC-MS流失出的小分子成分,即7个饱和与不饱和的乙酸酯,一个饱和六酸酯和两个饱和八酸酯,其中,包括PGS含量最高的成分E,E-法尼醇乙酸酯。因此,可以认为这些受睾丸调节的成分为雄性信息素的候选成分。对雌鼠的嗅觉双项选择测定说明低浓度的PGS和法尼醇乙酸酯水溶液对雌性有吸引作用,而高浓度时都具有趋避作用。这说明PGS具有剂量依赖的性吸引作用,法尼醇乙酸酯是一种剂量依赖的雄性信息素。进一步的数量比较说明,所有检测到成分的百分组成在个体间表现出巨大的个体变异,说明PGS的成分有个体的特异性,可能传递个体的嗅觉信息。  相似文献

    The allium leafminer, Acrolepiopsis sapporensis Matsumura (Lepidoptera: Acrolepiidae), is a pest of Allium species (Liliaceae) in Asia and Hawaii, USA. We identified candidate sex pheromone components in pheromone gland extracts of female moths and field tested the response of male moths to blends with different components and ratios. Gas chromatographic comparison of abdominal tip extracts from both sexes showed three female‐specific components: (Z)‐11‐hexadecenal (Z11‐16:Ald), (Z)‐11‐hexadecenyl acetate (Z11‐16:OAc), and (Z)‐11‐hexadecen‐1‐ol (Z11‐16:OH). These compounds were identified by mass spectral analysis of natural pheromone components and dimethyldisulfide adducts, and retention index comparisons with synthetic standards. The average ratio of three components, Z11‐16:Ald, Z11‐16:OAc, and Z11‐16:OH, in female extract was 33:100:14. Field trapping experiments indicated that all three components were essential for maximal attraction of male moths. Traps baited with a ternary blend mimicking the blend found in the pheromone gland extracts caught significantly more males than traps baited with caged live females.
