A special centre for prosthetic hip replacement set up 10 years ago has been successful in its primary objectives, producing high quality work at low cost and stimulating research. The project has not yet been successful in achieving a permanent staffing structure, but this is the effect of certain British ideologies in surgical training, with the result that craftsmanship and technology cannot be reconciled with pure intellectual attainment.The success of the project suggests the importance of isolating elective surgery from emergency surgery in the interests of cost-effectiveness. Problems of isolation from teaching hospitals are not insurmountable, but isolation to some degree is essential to achieve the minimum size necessary for efficiency and to ensure unbroken professional team-work among all levels of staff. While maximum efficiency in this field can be achieved only in special centres, too many primary operations will be required every year to insist that total hip replacement should be done only in special centres. A special feature of this field is the need for a permanent staffing structure to hand on technical expertise as an unbroken tradition.  相似文献

Endotoxin recognition: in fish or not in fish?   总被引:1,自引:0,他引:1  
The interaction between pathogens and their multicellular hosts is initiated by activation of pathogen recognition receptors (PRRs). These receptors, that include most notably members of the toll-like receptor (TLR) family, recognize specific pathogen-associated molecular patterns (PAMPs). TLR4 is a central part of the receptor complex that is involved in the activation of the immune system by lipopolysaccharide (LPS) through the specific recognition of its endotoxic moiety (Lipid A). This is a critical event that is essential for the immune response to Gram-negative bacteria as well as the etiology of endotoxic shock. Interestingly, compared to mammals, fish are resistant to endotoxic shock. This in vivo resistance concurs with in vitro studies demonstrating significantly lowered sensitivity of fish leukocytes to LPS activation. Further, our in vitro analyses demonstrate that in trout mononuclear phagocytes, LPS fails to induce antiviral genes, an event that occurs downstream of TLR4 and is required for the development of endotoxic shock. Finally, an in silico approach that includes mining of different piscine genomic and EST databases, reveals the presence in fish of all of the major TLR signaling elements except for the molecules specifically involved in TLR4-mediated endotoxin recognition and signaling in mammals. Collectively, our analysis questions the existence of TLR4-mediated cellular responses to LPS in fish. We further speculate that other receptors, in particular beta-2 integrins, may play a primary role in the activation of piscine leukocytes by LPS.  相似文献

Toads (Rhinella arenarum) received training with a novel incentive procedure involving access to solutions of different NaCl concentrations. In Experiment 1, instrumental behavior and weight variation data confirmed that such solutions yield incentive values ranging from appetitive (deionized water, DW, leading to weight gain), to neutral (300 mM slightly hypertonic solution, leading to no net weight gain or loss), and aversive (800 mM highly hypertonic solution leading to weight loss). In Experiment 2, a downshift from DW to a 300 mM solution or an upshift from a 300 mM solution to DW led to a gradual adjustment in instrumental behavior. In Experiment 3, extinction was similar after acquisition with access to only DW or with a random mixture of DW and 300 mM. In Experiment 4, a downshift from DW to 225, 212, or 200 mM solutions led again to gradual adjustments. These findings add to a growing body of comparative evidence suggesting that amphibians adjust to incentive shifts on the basis of habit formation and reorganization.  相似文献

Chara corallina Klein ex Willd. wm. R.D.W. (= C. australis R. Br.), a fresh water alga, maintains a constant internal osmotic pressure when external osmotic pressure is increased. This results in a decrease in turgor pressure. Chara osmoregulates effectively in the presence of high CaCl2 and raffinose, but is less efficient in response to increased NaCl. Decreasing external pH from 7 to 5 results in a decrease in turgor, but increasing it to values as high as 9 has no effect. Increasing the daily amount of light from 0.5 to 24 hours has no effect on turgor.  相似文献

We evaluate the arguments and evidence for a terrestrial vs an aquatic origin for the Insecta. The evidence falls into three categories: (1) evidence that does not support one view more than the other; (2) speculative evidence, which should carry little weight; and (3) evidence that does support one view more than the other. Category 1 includes evidence from locomotory and osmoregulatory systems; plausible functions have been proposed for 'protowings' in both aquatic and terrestrial environments, while locomotory and osmoregulatory mechanisms of insects shed little light on their origins. Fossils, phylogenetic speculation, gill structure and life histories fall into category 2, in which, although speculative, the evidence favours a terrestrial origin. The earliest fossil hexapods were apparently terrestrial and unequivocally aquatic hexapods do not appear until 60–70 million years later, while sister-group relationships point to a terrestrial life style from at least the Hexapoda-Myriapoda stem group. The great variation in gill structure, even within orders, suggests convergence, and the more or less completely aquatic life histories are better interpreted as steps towards independence from land, rather than signs of an aquatic origin. Category 3 includes evidence from the tracheal system. In order to have evolved in water, a tracheal system must have first invaginated, then connected with the body wall for gas exchange with the water, and thirdly connected with the internal organs. It is difficult to envisage functions for the first two stages; on the other hand, the system could have readily evolved on land by invagination of respiratory surfaces, and then have been modified to effect gas exchange in water via gills.  相似文献

Members of a social group have to make collective decisions in order to synchronise their activities. In a shared consensus decision, all group members can take part in the decision whereas in an unshared consensus decision, one individual, usually a dominant member of the group, takes the decision for the rest of the group. It has been suggested that the type of decision-making of a species could be influenced by its social style. To investigate this further, we studied collective movements in two species with opposed social systems, the Tonkean macaque (Macaca tonkeana) and the rhesus macaque (Macaca mulatta). From our results, it appears that the decision to move is the result of the choices and actions of several individuals in both groups. However, this consensus decision involved nearly all group members in Tonkean macaques whereas dominant and old individuals took a prominent role in rhesus macaques. Thus, we suggest that Tonkean macaques display equally shared consensus decisions to move, whereas in the same context rhesus macaque exhibit partially shared consensus decisions. Such a difference in making a collective decision might be linked to the different social systems of the two studied species.  相似文献

MicroRNAs are small non-coding RNAs that negatively regulate gene expression through mRNA degradation or translational repression. It is becoming increasingly recognized that miRNAs play central roles in almost all cellular processes, and especially during development. The function of miRNAs in hematopoiesis, including erythropoiesis, is beginning to be elucidated. Birds are anosmic or at best microsmatic… This misbelief persisted until very recently and has strongly influenced the outcome of communication studies in birds, with olfaction remaining neglected as compared to acoustic and visual channels. However, there is now clear empirical evidence showing that olfaction is perfectly functional in birds and birds use olfactory information in a variety of ethological contexts. Although the existence of pheromones has never been formally demonstrated in this vertebrate class, different groups of birds, such as petrels, auklets and ducks have been shown to produce specific scents that could play a significant role in within-species social interactions. Behavioral experiments have indeed demonstrated that these odors influence the behavior of conspecifics. Additionally, in quail, deprivation of olfactory inputs decreases neuronal activation induced by sexual interactions with a female. It seems therefore well established that birds enjoy a functional sense of smell and a fast growing body of experimental evidence suggests that they use this channel of olfactory communication to control their social life. The unequivocal identification of an avian pheromone is, however, still ahead of us but there are now many exciting opportunities to unravel the behavioral and physiological particularities of chemical communication in birds.  相似文献

Abstract Borstlap (1983) has alleged that (a) there are no abrupt changes in curves for the concentration dependence of solute uptake in plants, and (b) many such uptake isotherms may be described by the sum of two Michaelis-Menten terms and a linear term. These claims are considered in detail in connection with the recent finding (Nandi, Pant & Nissen, 1987) that phosphate uptake by corn roots increased more rapidly within the higher phases, i.e. at high external phosphate concentrations, but also levelled off faster than predicted from Michaelis-Menten kinetics. Similar deviations are, in retrospect, also found for uptake of other solutes and result in fewer phases at high external solute concentrations. The simplified and strikingly similar multiphasic patterns in the present paper show that (a) the abrupt changes in published isotherms are not due to error in the data, and (b) uptake isotherms cannot be adequately represented by the sum of two Michaelis-Menten terms and a linear term, or by similar continuous functions, if sufficiently detailed and precise data are used. These findings are not consistent with the existence of multiple uptake mechanisms, including free diffusion, in the plasmalemma. Uptake occurs instead by a single, multiphasic mechanism for each solute or group of related solutes. The similarities in the multiphasic patterns indicate, furthermore, that influx of the various solutes may be coupled.  相似文献

Epigenetic silencing is a pervasive mode of gene regulation in multicellular eukaryotes: stable differentiation of somatic cell types requires the maintenance of subsets of genes in an active or silent state. The variety of molecules involved, and the requirement for active maintenance of epigenetic states, creates the potential for errors on a large scale. When epigenetic errors - or epimutations - activate or inactivate a critical gene, they may cause disease. An epimutation that occurs in the germline or early embryo can affect all, or most, of the soma and phenocopy genetic disease. But the stochastic and reversible nature of epigenetic phenomena predicts that epimutations are likely to be mosaic and inherited in a nonmendelian manner; epigenetic diseases will thus rarely behave in the comfortably predictable manner of genetic diseases but will display variable expressivity and complex patterns of inheritance. Much phenotypic variation and common disease might be explained by epigenetic variation and aberration. The known examples of true epigenetic disease are at present limited, but this may reflect only the difficulty in distinguishing causal epigenetic aberrations from those that are merely consequences of disease, a challenge further extended by the impact of environmental agents on epigenetic mechanisms. The rapidly developing molecular characterization of epigenomes, and the new ability to survey epigenetic marks on whole genomes, may answer many questions about the causal role of epigenetics in disease; these answers have the potential to transform our understanding of human disease.  相似文献
