We are responding to a Letter to the Editor addressing the Method section of our paper “Different measures of ‘genome-wide’ DNA methylation exhibit unique properties in placental and somatic tissues.” The letter raised concerns that the protocol for Epigentek’s MethylFlash kit was followed incorrectly based on the wording of an online publication of our article. We admittedly made an error in the language used to describe the MethylFlash protocol in our initial submission and thus this was corrected as soon as it was brought to our attention. However, the error was only in language and not procedure. We are confident that the protocol was followed as stated in the insert provided with the MethylFlashTM Methylated DNA Quantification kit (Colorimetric).  相似文献

In one of our previous Editor’s pages, we announced that our Netherlands Heart Journal (NHJ) received a 2009 impact factor of 1.4, which is a crucial step for our journal. This achievement was a consequence of another important step for our journal, being the recognition by PubMed as of January 2007. From that moment on, the Chief Editorial Board and our Publisher observed a considerable increase in submitted articles. In 2007 we received a total of 62 submissions.  相似文献

From 2003 through 2009 we treated 35 patients who suffered from an isolated capitellum humerus fracture whom we treated with osteosynthesis. Patients who presented with concomitant fractures were not included. Thirty-four patients were categorized as Type I (Hahn-Steinthal) while one patient was Type IV (McKee). We describe the mechanism of injury and compared our results with works available in the literature. The average age of our patients was 38.6 years which was much younger than many articles about this type of injury found in the literature. The ratio of women to men in our study was 20:15. The surgical treatment was performed with various methods including: Kirschner wires, AO screws, Herbert screws and TwinFix screws. We discuss type of injury, days after injury operative treatment is performed, type of osteosynthesis used, the surgical approaches used for our treatment of capitellum humeri fractures, possible complications and our postoperative treatment. Results at the conclusion of treatment were excellent. Range of motion, shown in detail for each patient, was measured preoperatively, 1 month and 3 months postoperatively. We concluded that the major factors in successful treatment are how quickly the surgical treatment is performed after injury and early postoperative rehabilitation.  相似文献

在荧光定量PCR基础上建立一种简单有效并且高度灵敏的TB-ARMSkras基因突变检测方法,并对其检测性能进行评估,探讨其临床应用价值。针对kras基因8种常见的点突变类型,通过设计并优化突变特异性引物、野生型特异性封闭引物并综合应用突变富集扩增反应条件等多种手段,提高点突变检测的灵敏度和特异性,采用已知野生型基因组样品和构建的突变质粒作为标准品,进行方法学评价;通过对临床样本的检测及与现有商品化试剂盒的比较进行性能验证;通过对术前血浆和配对组织样品的对比检测,评估方法是否适用于血液样本的检测。建立了TB-ARMS kras突变检测的新方法,能检测的最低突变率可达到0.01%。通过综合采用野生型特异性封闭引物和突变富集扩增条件等方法证明了其0.01%的突变检测灵敏度。检测准确性优于现有商品化试剂盒,血浆DNA TB-ARMS qPCR检测结果与配对组织DNA测序结果相符合。因此,TB-ARMS kras基因突变检测方法具有广泛的临床应用价值,既适用于临床组织样品的检测,也可应用于液体活检。  相似文献

To upscale DSC size to commercial size, our group was involved in developing a commercial DSC panel, which could show the industrial way and prospect. The repeatable DSC module with the size of 150 mm × 200 mm and photoelectric conversion efficiency around 6% was reproduced in our laboratory. The DSC panel up to the size of 450 mm × 800 mm was fabricated. This is the high efficiency and industrial production design of a DSC panel, a primary power station with 500 W was installed on our roof for charging the battery. The study on the stability and performance of the DSC module, panel and future production are ongoing in our laboratory. In this article, we will report a systematic study in the test and the design from the single cell, the modules, to the panels, and our design of a 500 W primary DSC power station.  相似文献

Several pathogenic mutations in the LRRK2 gene have been implicated in familial and sporadic cases of Parkinson's disease (PD). We screened 103 sporadic French PD patients for the presence of the LRRK2 R1441G and G2019S mutations. The R1441G mutation was absent in our PD sporadic cases, but the G2019S mutation was present in 2 of them (1.9%). Clinical features in our 2 patients were not different from classic PD. One of our patients was of Berberian (North Africa) origin. Our 2 patients displayed genetic profiles consistent with the same ancestral haplotype as previously reported for carriers of the LRRK2 G2019S mutation.  相似文献

After a short review of our method, we compared the results of the radiographs of 18 cases of CdLS to the results of our normal material. All of our normal statistical data was obtained from AP and lateral view radiographs at a distance of 1 m, in the prevailing conditions of everyday practice in a radiological department. We studied quantitative changes such as microcephaly and brachycephaly, and for the first time we introduced qualitative changes such as occipital rotation and the study of obliquity of the foramen magnum (line BAO). We found microcephaly in all our cases of CdLS and brachycephaly in only 72%. Furthermore, we found a lack of positive occipital rotation in all our cases of CdLS and the line BAO turned posteriorly with O higher than BA. The same conditions were observed on our normal material only in an early postnatal stage.  相似文献

After the publication of [1], we were alerted to an error in our data. The error was an one-off miscalculation in the extraction of position information for our set of true negatives. Our data set should have used randomly selected non-edited cytosines (C) as true negatives, but the data generation phase resulted in a set of nucleotides that were each one nucleotide downstream of known, unedited cytosines. The consequences of this error are reflected in changes to our results, although the general conclusions presented in our original publication remain largely unchanged.  相似文献

Chemical ligation methods for the assembly of functional proteins continue to advance our basic understanding of protein structure and function. In this work, we report on our progress towards the full synthesis of HIV-1 Tat utilizing our newly developed ligation method; side-chain assisted ligation. The HIV-1 Tat was assembled from three fragments wherein the two thioester peptides were synthesized efficiently using the side-chain anchoring strategy following Fmoc-SPPS. The side-chain assisted ligation step was efficient and provided the ligation product in good yield. Following this step, native chemical ligation was used to fully assemble the HIV-1 Tat protein. Although the removal of the auxiliary in small peptides was straightforward, in the case of HIV-1 Tat this step was inefficient thus hampering the completion of the synthesis.  相似文献

Background

Cross-cultural adaptation is a necessary process to effectively use existing instruments in other cultural and language settings. The process of cross-culturally adapting, including translation, of existing instruments is considered a critical set to establishing a meaningful instrument for use in another setting. Using a multi-step approach is considered best practice in achieving cultural and semantic equivalence of the adapted version. We aimed to ensure the content validity of our instruments in the cultural context of KwaZulu-Natal, South Africa.

Methods

The Iowa Infant Feeding Attitudes Scale, Breastfeeding Self-Efficacy Scale-Short Form and additional items comprise our consolidated instrument, which was cross-culturally adapted utilizing a multi-step approach during August 2012. Cross-cultural adaptation was achieved through steps to maintain content validity and attain semantic equivalence in the target version. Specifically, Lynn’s recommendation to apply an item-level content validity index score was followed. The revised instrument was translated and back-translated. To ensure semantic equivalence, Brislin’s back-translation approach was utilized followed by the committee review to address any discrepancies that emerged from translation.

Results

Our consolidated instrument was adapted to be culturally relevant and translated to yield more reliable and valid results for use in our larger research study to measure infant feeding determinants effectively in our target cultural context.

Conclusions

Undertaking rigorous steps to effectively ensure cross-cultural adaptation increases our confidence that the conclusions we make based on our self-report instrument(s) will be stronger. In this way, our aim to achieve strong cross-cultural adaptation of our consolidated instruments was achieved while also providing a clear framework for other researchers choosing to utilize existing instruments for work in other cultural, geographic and population settings.

Current circumstances in both the health science and health care delivery systems in our country have created serious problems for health professionals, particularly for those of us in the relatively young field of biofeedback. At the same time, recent and emerging developments in the neurosciences and in technologies for biological measurement offer exciting new opportunities for the applied psychophysiology upon which our field is based. Examples of these developments include the elucidation of potential neurophysiological mechanisms that may mediate the psychophysiology of immune system responses, and technical achievements leading to on-line, noninvasive magnetic resonance imaging of brain neurochemistry. The issue of perspective is raised in relation to our response to both the challenge and the opportunities of our times.This was the presidential address delivered to the Association for Applied Psychophysiology and Biofeedback, San Diego, March 19, 1989. Summary The cystic fibrosis (CF) gene deletion F508 was studied in a Belgian population of 74 families and their 83 CF children. The haplotypes for CF and normal chromosomes had previously been determined with several linked DNA probes. In our CF population, the gene deletion F508 was found in 76% of the mutant alleles. Of the deletion F508, 97% segregated with the highest risk haplotype for the CF carrier status. Some 61% of our families were found to be homozygous for this major CF mutation. Each of our three pancreatic sufficiency patients (two of whom were siblings) was heterozygote for the F508 deletion.  相似文献

Both intact and denatured preparations of myosin and actin from frog skeletal muscles produced in rabbits antisera containing antibodies against authentic myosin and actin, respectively, though being contaminated with antibodies against other proteins. Antigenicity of our frog myosin as revealed in agar diffusion tests was indistinguishable from that of cardiac muscle myosin from the same species. Similarly, skeletal muscle myosins from other amphibians shared to a certain extent immunological characteristics with our frog myosin, but those from avian and mammalian materials did not. Similarity in antigenicity was also demonstrated among our skeletal muscle actin, cardiac muscle actin from the same species and skeletal muscle actin from the other anurans studied. However, skeletal muscle actin from an urodele could not clearly be correlated in its immunological properties with our frog actin, and those from avian and mammalian materials were antigenically different from our frog actin. Thus, the degree of antigenic similarity of these muscle proteins seemed to be correlated with the phylogenic relationship of the animals so far studied. The results also indicated that our antisera could only be applied to immuno-cytological and immuno-embryological studies of myosin and actin when the antisera absorbed with the corresponding antigen preparations were used as negative controls.  相似文献

Recent scientific evolutions force us to rethink our profession's position on the scientific map, in relation to our neighboring professions, the ones with which we traditionally have strong collaborative links as well as the newly emerging fields, but also within our own, diverse professional group. We will show that great inspiration can be drawn from our own history, in fact from the early days of the Society. A recent inspiring example has been set by the late Rob Kempton, who died suddenly just months before he was to become President of the International Biometric Society.  相似文献

In our previous publication, a framework for information flow in interaction networks based on random walks with damping was formulated with two fundamental modes: emitting and absorbing. While many other network analysis methods based on random walks or equivalent notions have been developed before and after our earlier work, one can show that they can all be mapped to one of the two modes. In addition to these two fundamental modes, a major strength of our earlier formalism was its accommodation of context-specific directed information flow that yielded plausible and meaningful biological interpretation of protein functions and pathways. However, the directed flow from origins to destinations was induced via a potential function that was heuristic. Here, with a theoretically sound approach called the channel mode, we extend our earlier work for directed information flow. This is achieved by constructing a potential function facilitating a purely probabilistic interpretation of the channel mode. For each network node, the channel mode combines the solutions of emitting and absorbing modes in the same context, producing what we call a channel tensor. The entries of the channel tensor at each node can be interpreted as the amount of flow passing through that node from an origin to a destination. Similarly to our earlier model, the channel mode encompasses damping as a free parameter that controls the locality of information flow. Through examples involving the yeast pheromone response pathway, we illustrate the versatility and stability of our new framework.  相似文献

Our study was a replication and extension with methodological improvements to a previous study on effects of the electromagnetic field (EMF) emitted by a 902 MHz mobile phone on human cognitive functioning. Improvements on the previous study included multicentre testing and a double blind design. A total of 64 subjects (32 men and 32 women) in two independent laboratories performed a battery of 9 cognitive tasks twice: while the EMF was on and while it was off. Reaction times (RTs) and accuracy were recorded. The order of exposure and tasks was counterbalanced across subjects and gender. There were no statistically significant differences in performance between genders or laboratories. Although the RTs and the accuracy of answers were very similar to those of our previous study, our previous results were not replicated. We concluded that EMF had no effect on RTs or on the accuracy of the subjects' answers. Further, our results indicate that our EMF had no immediate effect on human cognitive functioning or that such effects are so small that they are observed on behavior only occasionally.  相似文献
