The paper disk method of screening several plant growth regulators was evaluated. Leaf explants ofVigna unguiculata (L) Walp. were placed on solidified Murashige and Skoog's minimal organics medium containing 0.5 mg/l nicotinic acid. Hormones were tested, singly and in combinations, on paper disks in large Petri plates (150×20 mm). Hormones tested were 2,4-D (2,4-dichlorophenoxyacetic acid), 2,4,5-T (2,4,5-trichlorophenoxyacetic acid), IAA (indole-3-acetic acid), IBA (indole-3-butyric acid), picloram (4-amino-3,5,6-trichloropicolinic acid), dicamba (3,6-dichloro-2-methoxybenzoic acid), BA (6-benzyladenine), 2iP (2-isopentenyl adenine), and kinetin [6-(furfurylamino)-purine]. Root formation was stimulated by IAA and IBA; dicamba, picloram, 2,4-D, and 2,4,5-T stimulated callus formation. All cytokinins tested suppressed root formation. Dicamba in combination with either 2iP or kinetin induced the greatest callus formation. Root formation was optimal with kinetin and either IAA or IBA. The disk method provided a rapid, nonquantitative evaluation of callus and root formation from leaf disks.  相似文献

Within 3 weeks of culture, excised cotyledon expiants of Camellia sinensis (L.) O. Kuntze produced somatic embryos without intermediate callus when cultured in Murashige and Skoog's basal medium with 30 g–1 sucrose. In medium without plant growth regulators, up to 60% of the cultures developed somatic embryos. Embryogenic competence was reduced by increasing concentrations of plant growth regulators tested (i.e. kinetin, 6-benzylaminopurine, and indole butyric acid). The somatic embryos developed, grew to maturity without being subcultured within 6–8 weeks. Secondary embryogenesis was not observed. Germination of isolated mature somatic embryos was low in medium without plant growth regulators. Up to 53% and 60% germination occurred when medium impregnated with kinetin at 1.8 mgl–1 or 1.0 mgl–1 6-benzylaminopurine were used respectively. Callus was also routinely produced when cotyledons were cultured in MS basal medium with auxins (2,4-dichlorophenoxyacetic acid and indole acetic acid). Callus induction was however, also achieved in plant growth regulator free medium. The interrelationship between ethylene and growth regulators in the senescence of romaine lettuce (Lactuca sativa L.) leaves was studied. Gibberellic acid (GA3), kinetin, and 3-indoleacetic acid (IAA) retarded chlorophyll loss from leaf discs which were floated on hormone solutions. Abscisic acid (ABA) and ethephon enhanced chlorophyll loss and antagonized the senescence-retarding effect of GA3 and kinetin. A high concentration of IAA (10–4 M) caused accelerated chlorophyll loss, whereas a similar concentration of kinetin neither retarded nor promoted chlorophyll loss. The ineffectiveness of IAA and kinetin at their supraoptimal concentrations in retarding leaf senescence was related to increased production of ethylene induced in the treated leaf discs. GA3 was the most effective in retarding chlorophyll loss and did not stimulate ethylene production at all. The senescence-enhancing effect of ABA was not mediated by ethylene. However, the moderately increased production of ethylene, induced by relatively high concentrations of ABA, could act synergistically with the latter to accelerate chlorophyll loss. A simple and rapid HPLC-based method was developed for simultaneous determination of major classes of plant growth regulators (PGRs) in Monostroma and different species of Ulva. The plant growth regulators determined included gibberellic acid (GA3), indole-3-acetic acid (IAA), abscisic acid (ABA), indole-3-butyric acid (IBA), salicylic acid and kinetin riboside (KR) and their respective elution time was 2.75, 3.3, 3.91, 4.95, 5.39 and 6.59 min. The parameters optimized for distinct separation of PGRs were mobile phase (60:40 methanol and 0.6% acetic acid in water), column temperature (35 °C) and flow rate (1 ml/min). This method presented an excellent linearity (0.2–100 μg/ml) with limit of detection (LOD) as 0.2 μg/ml for ABA, 0.5 μg/ml for KR and salicylic acid, and 1 μg/ml for IAA, IBA and GA3. The precision and accuracy of the method was evaluated after inter and intra day analysis in triplicates. The effect of plant matrix was compensated after spiking and the resultant recoveries estimated were in the range of 80–120%. Each PGR thereby detected were further characterized by ESI-MS analysis. The method optimized in this study determined IBA along with IAA for the first time in the seaweed species investigated except Ulva linza where the former was not detected. In all the species studied, ABA level was detected to be the highest while kinetin riboside was the lowest. In comparison to earlier methods of PGR analysis, sample preparation and analysis time were substantially reduced while allowing determination of more classes of PGRs simultaneously.  相似文献

Melatonin stimulates the expansion of etiolated lupin cotyledons   总被引:1,自引:0,他引:1  
Melatonin (N-acetyl-5-methoxytryptamine) is an indoleamine which is structurally related to tryptophan, serotonin and indole-3-acetic acid (IAA), among other important substances. Many studies have clearly demonstrated its presence in different plant organs, including roots, stems, leaves, flowers, fruits and seeds. Since it discovery in plants in 1995, authors have postulated many physiological roles for melatonin, although research into this molecule in plants is still in its infancy. The data presented in this study demonstrate that melatonin stimulates the expansion of etiolated cotyledons of lupin (Lupinus albus L.) to a similar extent to that observed for IAA but less than in the case of kinetin. Endogenous melatonin in imbibed cotyledons has been quantified using a liquid chromatography method with fluorescence detection and capacity of cotyledons to absorb melatonin has been determined. The observed effect of melatonin on lupin cotyledon expansion can be added to the other effects demonstrated by our group such as its role as growth promoter and rooting promotor in adventitious and lateral roots.  相似文献

The physiological and morphological factors necessary for efficient accumulation of sucrose in sugar beet (Beta vulgaris L.) are considered in relation to potential uses of plant growth regulators to modify the anatomy of storage roots so as to increase sucrose content and yield. The percentage of sucrose in root fresh and dry matter is closely related to root structure. Sugar beet, mangold and chard are three sub-species of Beta vulgaris that differ considerably in their anatomy, assimilate partitioning, sucrose concentration and root dry matter yield. The concentrations of indole-3-acetic acid (IAA), abscisic acid (ABA) and cytokinins were measured during the growth of the storage root in each of these cultivars. Correlations were found between the phytohormone levels and the formation of secondary cambia and their subsequent cell division and expansion activity.  相似文献

Benzyladenine (BA), zeatin and kinetin caused the unfolding of cucumber (Cucumis sativus L. Var. Jing-ye No. 4) cotyledons. But the other plant hormones e.g. gibberellic acid (GA3), indoleacetic acid(IAA), ethylene (ethrel), abscisic acid (ABA) and other related compound e.g. KCl, adenine were not so. This response was caused by even lower concentration (0.01 ppm). Therefore, it could be used as a bioassay for cytokinins. KCl affaeted synergistically on cytokinin-induced unfolding. Basally applied BA (10 ppm) stimulated the opening of the intact hypocotylar hook. The light affacted synergistically on BA-induced opening. However, basally applied BA prevented the opening of decapitated hooks, especially in the light. The cotyledons were found to play a part of effects in maintaining the hook.  相似文献

Different plant explants of Persian buttercup (Ranunculus asiaticus L.) were screened for callus induction and adventitious shoot regeneration on different media to establish totipotent cultures. Murashige & Skoog (MS) medium was used, supplemented with different concentrations of the following growth regulators: kinetin, benzyladenine (BA), naphthaleneacetic acid (NAA) and indoleacetic acid (IAA). Callus was induced and adventitious buds regenerated only from cotyledonary explants after 4–5 weeks. Subculture of the regenerated buds on the same basal medium in presence of gibberellic acid (GA3) and BA produced well-organized shoots. Rooting was obtained by transferring shoots to growth regulator-free MS medium. A high rate of shoot multiplication has been achieved on medium with high concentration of kinetin and long-day photoperiod. Finally the plants were successfully transferred to soil and grown in a greenhouse.  相似文献

The effects of indole-3-acetic acid (IAA), abscisic acid (ABA), gibberellic acid (GA3) and kinetin on the hydrolytic activity of proton pumps (adenosine triphosphatase, H+-ATPase, pyrophosphatase, H+-PPase) of tonoplasts isolated from stored red beet (Beta vulgaris L. cv. Bordo) roots were studied. Results suggest that the phytohormones can regulate the hydrolytic activities of H+-ATPase and H+-PPase of the vacuolar membrane. Each of the proton pumps of the tonoplast has its own regulators in spite of similar localization and functions. IAA and kinetin seem to be regulators of the hydrolytic activity for H+-PPase whereas for H+-ATPase it may be GA3. 采用丝氨酸内肽酶抑制剂和植物生长调节剂处理离体黄瓜叶片,研究了黄瓜叶片暗诱导衰老过程中丝氨酸内肽酶的作用。结果表明,6-BA50μmol/L与丝氨酸内肽酶抑制剂AEBSF能抑制叶片内肽酶活性的升高,延缓蛋白质降解,而ABA50μmol/L则促进了内肽酶活性的升高:其作用效果与AEBSF相反。活性电泳结果显示,黄瓜叶片中检测到6条内肽酶同工酶,其中4条(CEP2、3、4、6)为丝氨酸类型内肽酶,而ABA使丝氨酸内肽酶CEP2、3、4、6的活性明显增强,提示了丝氨酸类型内肽酶在黄瓜叶片衰老过程中具有重要作用。  相似文献

A simple bioassay based on the inhibition by abscisic acid (ABA) of cucumber (Cucumis sativus L., cv. National Pickling) hypocotyl elongation was developed. Sections of 3-day-old dark-grown cucumber hypocotyl taken from 0–5 mm immediately below the cotyledon were used for the assay. A dark incubation period of 20 h was followed by an exposure to light for 24 h. Under these conditions, the inhibition of hypocotyl elongation is proportional to the abscisic acid applied. The minimum detectable level of abscisic acid was 10–9 M, and the range of linear response to abscisic acid was between 10–7 and 10–3 M. This assay is 10 times more sensitive than the cucumber cotyledon greening bioassay for abscisic acid.  相似文献

When intact etiolated 2-day cucumber (Cucumis sativus) embryos were treated with indoleacetic acid (IAA), gibberellin A7 (GA7), or kinetin, chromatin derived from the embryonic axes exhibited an increased capacity to support RNA synthesis in either the presence or the absence of bacterial RNA polymerase. An IAA effect on cucumber RNA polymerase activity was evident after 4 hours of hormone treatment; the IAA effect on DNA template activity (bacterial RNA polymerase added) occurred after longer treatments (12 hours). GA7 also promoted template activity, but again only after a prior stimulation of endogenous chromatin activity. After 12 hours of kinetin treatment, both endogenous chromatin and DNA template activities were substantially above control values, but longer kinetin treatments caused these activities to decline in magnitude. When chromatin was prepared from hypocotyl segments that were floated on a GA7 solution, a GA-induced increase in endogenous chromatin activity occurred, but only if cotyledon tissue was left attached to the segments during the period of hormone treatment.  相似文献

Tomato leaf disks were inoculated with tobacco mosaic virus (TMV) and floated for 7 days on solutions of kinetin and benzyladenine in the range 20-0-002 mg/1. Virus content was reduced at the higher and increased at the lower concentrations. Benlate and benomyl showed a peak of cytokinin activity in the Amaranthus betacyanin bioassay equivalent to c. 0–002 fig/l kinetin. At concentrations above 25 and 100 mg a.i./l for Benlate and benomyl respectively, both compounds increased the TMV content of tomato leaf disks. Cucumber mosaic virus (CMV) content in cucumber cotyledon disks was increased by Benlate and benomyl treatment (50–100 mg/1). Applied as a soil drench (50–500 mg a.i./l) when the plants were inoculated, Benlate increased the CMV content of infected seedlings. The number of starch-iodide lesions (a measure of susceptibility) was unaltered in cotyledons treated with Benlate 7 days before or immediately after inoculation. Infectivity of crude infective cucumber sap was unaffected by benomyl incorporation, whereas Benlate reduced infectivity at higher concentrations (1000–5000 mg/1). Under the experimental conditions described, Benlate, benomyl, benzyladenine and kinetin had no effect on the chlorophyll content of tomato leaf disks, and intact seedlings.  相似文献

This study was conducted to establish a plant cell culture system for the production of medically important secondary metabolites fromXanthium strumarium. The effects of plant growth regulators including NAA, 2,4-D, kinetin, and ABA were examined in terms of callus induction, maintenance of callus and suspension cultures. It was shown that callus was induced upon treatment with NAA while embryo was induced after treatment with 2,4-D. Callus formation was further improved by treatment with ABA and NAA. The level of callusing increased by 17–29% for the seed case, cotyledon, leaf, and hypocotyl and by 96% in the case of the root. Suspension cell lines were established using calli produced from cotyledon, hypocotyl and root and cultured at 25°C under light conditions. The cells grew up to 15 g/L with NAA 2 ppm, BA 2 ppm, and ABA 1 ppm treatment. Supernatants of suspension cultures of cell lines derived from coyledon and hypocotyl produced some distinctive secondary metabolites, one of which was identified as 8-epi-tomentosin, which belongs to the xanthanolides. The amounts of 8-epi-tomentosin produced by the cotyledon-and hypocotylderived cell lines were 13.4 mg/L and 11.0 mg/L, respectively.  相似文献

Effects of jasmonic acid, methyl jasmonate and ABA on cytokinin-induced soybean ( Glycine max Merrill cv. Kingen No. 1) callus growth and radish ( Raphanus sativus L. cv. Comet) cotyledon growth were studied. Jasmonic acid and methyl jasmonate were powerful inhibitors of kinetin- or N-phenyl-N'-(2-chloro-4-pyridyl)urea-induced soybean callus growth, the former being more effective than the latter, especially at low concentrations (0.45 to 4.5 μ M ). These compounds could completely eliminate kinetin- or N-phenyl-N'-(2-chloro-4-pyridyl)urea-induced soybean callus growth at 45 μ M . At low concentrations ABA had no effect but at 450 μ M it completely eliminated callus growth induced by kinetin. Of the compounds tested ABA was the most powerful inhibitor of radish cotyledon growth in the presence or absence of kinetin. Jasmonic acid and methyl jasmonate were much less effective than ABA in this assay system.  相似文献

The Effect of Potassium on Cotyledon Expansion Induced by Cytokinins   总被引:3,自引:0,他引:3  
Potassium has been found to enhance greatly the expansion response of cucumber cotyledons to cytokinins. A reduction of the response to kinetin is obtained with increasing age of the cotyledons. The lesser response is associated with lower levels of potassium remaining in the cotyledon. A high level of KCI in the incubation medium offsets the lower potassium content of the tissue and enables a much larger response to the cytokinins. At 40 mM KCI the response to kinetin is 4.2 times greater than in the absence of KCI. Calcium increases the effect of potassium on the response to kinetin. When incubated in 40 mM KCI and 10 mM CaCI2 with 10 mg/I 6-benzylamino-purine, the final weight of the cotyledons is 6.8 times the initial weight after just 4 days. This KCI-CaCI2 combination is also found to promote chlorophyll synthesis in the usual cucumber cotyledon bioassay.  相似文献

The plant-growth regulators, indole-3-carboxylic acids, were introduced into N-acyl ethanolamines, and a series of N-acyl O-indolylalkyl ethanolamines were prepared. Their biological activities to regulate rape hypocotyl elongation, cucumber cotyledon expansion and common wheat coleoptile growth were tested. The results indicate that the title compounds inhibited rape hypocotyl elongation, especially the indole-3-propionic acid derivatives, whose bioactivity was better than that of indole-3-acetic acid.  相似文献

The cucumber cotyledon greening bioassay is frequently used for detecting cytokinins. Beneficial modifications of the original technique included using 5-day-old cucumber (Cucumus sativus L., cv. National Pickling) cotyledons treated with combinations of 40 millimolar KCl and various concentrations of cytokinins. A dark incubation period of 20 hours was followed by an exposure to light for 3.5 hours. Under these conditions, extremely low (0.0001 milligram per liter) concentrations of N6-benzyladenine, zeatin, kinetin, or zeatin riboside can be detected. Of the four cytokinins tested, kinetin appeared to be the least active. With these improvements, the assay is 10 times more sensitive than is the previously described cucumber cotyledon greening bioassay for cytokinins.  相似文献

Salicylic acid (SA) is one of the most important signaling molecules in plant growth and defense responses to biotic and abiotic stresses. Here, the effect of exogenous SA on the stomatal movements was investigated in cotyledons of cucumber (Cucumis sativus L.) seedlings. Application of different SA concentrations could induce the reduction in stomatal aperture and conductance, especially at a concentration of 0.5 mM. Using the isolated epidermal strips, stomata were found to close notably in response to exogenous SA, even at a concentration as low as 0.001 mM. Further study showed that a SA-induced decrease in the stomatal aperture was intensified by the higher SA concentrations, longer exposure, and lower pH of the medium. In addition, to understand the relationship between stomatal closure and endogenous hormone contents, the levels of ABA, IAA, and gibberellin (GA3) were assayed under SA treatment. SA significantly increased endogenous ABA but not IAA and GA3 content. A significant negative correlation (p ≤ 0.01) was observed between stomatal conductance and the ratio of ABA to (GA3 + IAA) during SA application. It was suggested that exogenous SA could change the balance of endogenous hormones and thereby induce stomatal closure in cotyledons of cucumber seedlings.  相似文献

Changes in the IAA and ABA contents in cucumber (Cucumis sativus L.) leaves during adaptation to drought were studied. An increase in the water-retaining capacity and heat resistance of leaves indicating the onset of adaptation occurred when the leaf growth has been already suppressed. There was a transient increase in the ABA content during the initial stage of adaptation. An increased IAA content was maintained for a longer period, throughout about two-third of the adaptation period. A second increase in the ABA content was observed before the onset of leaf permanent wilting, when IAA content already decreased. Our data suggest that not only ABA, but also IAA are involved in the development of defense responses during the adaptation to drought.  相似文献
