In this research, micro and nanoparticles of Spirulina platensis dead biomass were obtained, characterized and employed to removal FD&C red no. 40 and acid blue 9 synthetic dyes from aqueous solutions. The effects of particle size (micro and nano) and biosorbent dosage (from 50 to 750 mg) were studied. Pseudo-first order, pseudo-second order and Elovich models were used to evaluate the biosorption kinetics. The biosorption nature was verified using energy dispersive X-ray spectroscopy (EDS). The best results for both dyes were found using 250 mg of nanoparticles, in these conditions, the biosorption capacities were 295 mg g?1 and 1450 mg g?1, and the percentages of dye removal were 15.0 and 72.5% for the FD&C red no. 40 and acid blue 9, respectively. Pseudo-first order model was the more adequate to represent the biosorption of both dyes onto microparticles, and Elovich model was more appropriate to the biosorption onto nanoparticles. The EDS results suggested that the dyes biosorption onto microparticles occurred mainly by physical interactions, and for the nanoparticles, chemisorption was dominant.  相似文献

The removal of Remazol Blue and Reactive Black B by the immobilized thermophilic cyanobacterial strain Phormidium sp. was investigated under thermophilic conditions in a batch system, in order to determine the optimal conditions required for the highest dye removal. In the experiments, performed at pH 8.5, with different initial dye concentrations between 9.1 mg l−1 and 82.1 mg l−1 and at 45 °C, calcium alginate immobilized Phormidium sp. showed high dye decolorization, with maximum uptake yields ranging from 50% to 88% at all dye concentrations tested. When the effects of high dye concentrations on dye removal were investigated, the highest uptake yield in the beads was 50.3% for 82.1 mg l−1 Remazol Blue and 60.0% for 79.5 mg l−1 Reactive Black B. The highest color removal was detected at 45 °C and 50 °C incubation temperatures for all dye concentrations. As the temperature decreased, the removal yield of immobilized Phormidium sp. also decreased. At about 75 mg l−1 initial dye concentrations, the highest specific dye uptake measured was 41.29–41.17 mg g−1 for Remazol Blue and 47.69–43.82 mg g−1 for Reactive Black B at 45 °C and 50 °C incubation temperatures, respectively, after 8 days incubation.  相似文献

This research highlights the possibility of employing a fermentation industry waste (Corynebacterium glutamicum) for the removal of nickel(II) ions from aqueous solution. Furthermore, it necessitates the importance of detailed examinations on the possible differences in the biosorption performance, even for the same biomass, but from different origins. Two types of C. glutamicum, obtained from different industrial sources, were used in this study. With respect to nickel speciation and biosorption performance, pH 6 was identified as an optimal condition. Of the two types of C. glutamicum used, the biomass with excess negatively charged groups performed well in the binding of Ni2+ ions. To enhance the feasibility of using the biomass in column mode, as well as its reuse for multiple cycles, C. glutamicum was immobilized in a polysulfone matrix. Both the free and immobilized biomasses performed relatively well, with maximum experimental uptakes of 111.4 and 102.4 mg g−1, respectively. An up-flow packed column loaded with immobilized biomass was employed for the removal of Ni2+ ions. The column performed well in the biosorption of nickel(II), and exhibited a delayed and favorable breakthrough curve, with Ni2+ uptake and percentage removal of 48.1 mg g−1 biomass and 60.4%, respectively.  相似文献

Carbon fixation and allocation were studied using 13C incubation and leaf marking techniques in mature monospecific stands of Enhalus acoroides L.f. Royle in August 1998 and January 1999 in Banten Bay, Indonesia. The highest rate of 13C uptake (>0.008 g 13C g C−1 d−1) was found in the middle to distal parts of leaves of E. acoroides. Young and senescing leaves numbers had lower 13C incorporation compared to mature leaves. The incorporation of 13C by epiphytes on the leaves was higher than that of the leaves themselves (>0.01 g 13C g C−1 d−1). The results showed that turbidity of the water influenced the leaf growth, productivity and Relative Growth Rate of E. acoroides, which were lower at Kepuh Island, the more turbid site. However, at Kepuh Island, where the water column was turbid, the plant could still harvest sufficient light for an uptake rate of 13C, higher than the uptake rates at Kubur and Panjang Islands, stations with a much more transparent water column (on average 0.0047 g 13C g C−1 d−1 at Kepuh Island, versus 0.0045 g 13C g C−1 d−1 at Panjang Island and 0.0034 g 13C g C−1 d−1 at Kubur Island). There was evidence that 13C was exported from the incubated shoots to the roots and rhizomes and to neighboring shoots of E. acoroides in clear water, but not in turbid water. We suggest that single shoots of E. acoroides are able to grow in turbid water under low light conditions. They assimilate sufficient carbon for their own maintenance but are not able to export to neighboring plant parts.  相似文献

The main photosynthesis and respiration parameters (dark respiration rate, light saturated production rate, saturation irradiance, photosynthetic efficiency) were measured on a total of 23 macrophytes of the Thau lagoon (2 Phanerogams, 5 Chlorophyceae, 10 Rhodophyceae and 6 Phaeophyceae). Those measurements were performed in vitro under controlled conditions, close to the natural ones, and at several seasons. Concomitantly, measurements of pigment concentrations, carbon, phosphorous and nitrogen contents in tissues were performed. Seasonal intra-specific variability of photosynthetic parameters was found very high, enlightening an important acclimatation capacity. The highest photosynthetic capacities were found for Chlorophyceae (e.g. Monostroma obscurum thalli at 17 °C, 982 μmol O2 g−1 dw h−1 and 9.1 μmol O2 g−1 dw h−1/μmol photons m−2 s−1, respectively for light saturated net production rate and photosynthetic efficiency) and Phanerogams (e.g. Nanozostera noltii leaves at 25 °C, 583 μmol O2 g−1 dw h−1 and 2.6 μmol O2 g−1 dw h−1/μmol photons m−2 s−1 respectively for light saturated net production rate and photosynthetic efficiency). As expected, species with a high surface/volume ratio were found to be more productive than coarsely branched thalli and thick blades shaped species. Contrary to Rd (ranging 6.7–794 μmol O2 g−1 dw h−1, respectively for Rytiphlaea tinctoria at 7 °C and for Dasya sessilis at 25 °C) for which a positive relationship with water temperature was found whatever the species studied, the evolution of P/I curves with temperature exhibited different responses amongst the species. The results allowed to show summer nitrogen limitation for some species (Gracilaria bursa-pastoris and Ulva spp.) and to propose temperature preferences based on the photosynthetic parameters for some others (N. noltii, Zostera marina, Chaetomorpha linum).  相似文献

The ectoine-excreting bacterial strain of Halomonas salina was employed in the co-production of poly-β-hydroxybutyrate (PHB) and ectoine (Ect) during a fermentation process (PHB/Ect co-production). An efficient PHB/Ect co-production process was carried out at low NaCl concentration (30 g L−1). It was established using 1H Nuclear Magnetic Resonance spectroscopy that H. salina produces PHB. The effects of the NaCl concentration, the initial C/N ratio, the phosphate concentration and mixed carbon sources were investigated with respect to PHB/Ect co-production. The PHB/Ect co-production system comprised growing and non-growing cell phases and was developed with NaCl concentration of 30 g L−1. The optimal conditions for PHB/Ect co-production by the ectoine-excreting strain of H. salina were 30 g L−1 NaCl, with an initial C/N ratio of 15, an initial phosphate concentration of 12 g L−1 and mixed carbon sources of 55 g L−1 glucose and 25 g L−1 monosodium glutamate. Using a PHB/Ect co-production system with growing and non-growing cell phases prevents the inhibition of PHB synthesis by high concentration of NaCl and significantly reduces ectoine degradation. PHB and ectoine concentrations as high as 35.3 g L−1 and 8.6 g L−1, respectively, were achieved. The efficient co-production of PHB and ectoine at a low NaCl concentration has been realised.  相似文献

 Enzymatic hydrolysis of corncob and ethanol fermentation from cellulosic hydrolysate were investigated. After corncob was pretreated by 1% H2SO4 at 108 °C for 3 h, the cellulosic residue was hydrolyzed by cellulase from Trichoderma reesei ZU-02 and the hydrolysis yield was 67.5%. Poor cellobiase activity in T. reesei cellulase restricted the conversion of cellobiose to glucose, and the accumulation of cellobiose caused severe feedback inhibition to the activities of β-1,4-endoglucanase and β-1,4-exoglucanase in cellulase system. Supplementing cellobiase from Aspergillus niger ZU-07 greatly reduced the inhibitory effect caused by cellobiose, and the hydrolysis yield was improved to 83.9% with enhanced cellobiase activity of 6.5 CBU g−1 substrate. Fed-batch hydrolysis process was started with a batch hydrolysis containing 100 g l−1 substrate, with cellulosic residue added at 6 and 12 h twice to get a final substrate concentration of 200 g l−1. After 60 h of reaction, the reducing sugar concentration reached 116.3 g l−1 with a hydrolysis yield of 79.5%. Further fermentation of cellulosic hydrolysate containing 95.3 g l−1 glucose was performed using Saccharomyces cerevisiae 316, and 45.7 g l−1 ethanol was obtained within 18 h. The research results are meaningful in fuel ethanol production from agricultural residue instead of grain starch.  相似文献

Pseudomonas luteola was immobilized by entrapment in alginate–silicate sol–gel beads for decolorization of the azo dye, Reactive Red 22. The influences of biomass loading and operating conditions on specific decolorization rate and dye removal efficiency were studied in details. The immobilized cells were found to be less sensitive to changes in agitation rates (dissolved oxygen levels) and pH values. Michaelis–Menten kinetics could be used to describe the decolorization kinetics with the kinetic parameters being 36.5 mg g−1 h−1, 300.1 mg l−1 and 18.2 mg g−1 h−1, 449.8 mg l−1 for free and immobilized cells, respectively. After five repeated batch cycles, the decolorization rate of the free cells decreased by nearly 54%, while immobilized cells still retained 82% of their original activity. The immobilized cells exhibited better thermal stability during storage and reaction when compared with free cells. From SEM observation, a dense silicate gel layer was found to surround the macroporous alginate–silicate core, which resulted in much improved mechanical stability over that of alginate beads when tested under shaking conditions. Alginate–silicate matrices appeared to be the best matrix for immobilization of P. luteola in decolorization of Reactive Red 22 when compared with previous results using synthetic or natural polymer matrices.  相似文献

Pure glycerol and glycerol-rich product (GRP) obtained from the biodiesel industries were used as carbon source for the production of a new extracellular polysaccharide (EPS) by Pseudomonas oleovorans NRRL B-14682. The influence of temperature (20–40 °C) and pH (6.0–8.0) was studied. A temperature of 30 °C and pH control at 6.8 gave the maximum cell growth and EPS production. The culture attained a maximum cell dry weight (CDW) of 9.55 g l−1 and an EPS concentration of 11.82 g l−1 when cultivated with pure glycerol. GRP was a suitable carbon source, as shown by the slightly higher EPS concentration (12.18 g l−1). The EPS productivity obtained with GRP (3.85 g l−1 d−1) was almost twice that obtained with pure glycerol (2.00 g l−1 d−1). Also, the yield on glycerol was higher for the cultivation with GRP (0.36 g g−1) than for pure glycerol (0.28 g g−1). The EPS was a high molecular weight heteropolysaccharide, composed by neutral sugars (37–80 wt% galactose, 2–30 wt% glucose, 0.5–25 wt% mannose and 0.5–20 wt% rhamnose) and containing acyl group substituents (pyruvil, acetyl and succinyl were identified). The EPS forms highly viscous aqueous dispersions with many potential commercial applications.  相似文献

Members of the Chenopodiaceae are well adapted to both salt and drought stress and can serve as model species to understand the mechanisms of tolerance in plants. We grew Atriplex hortensis (ATHO), A. canescens (ATCA), and A. lentiformis (ATLE) along a NaCL salinity gradient under non-water-limited conditions and in drying soils in greenhouse experiments. The species differed in photosynthetic carbon fixation pathway, capacity for sodium uptake, and habitat preferences. Under non-water-limited conditions, ATLE (C4) maintained high growth rates up to 30 g L−1 NaCl. ATHO (C3) had lower growth than ATLE at high salinities, while ATCA (C4) grew more slowly than either ATLE or ATHO and showed no net growth above 20 g L−1 NaCl. ATHO and ATLE accumulated twice as much sodium in their shoots as ATCA, but all three species had increasing sodium levels at higher salinities. Potassium, magnesium and calcium levels were relatively constant over the salinity gradient. All three species showed marked accumulation of chloride across the salinity gradient, whereas nitrate, phosphorous and sulfate decreased with salinity. The effect of drought was simulated by growing plants in sealed pots with an initial charge of water plus NaCl, and allowing them to grow to the end point at which they no longer were able to extract water from the soil solution. Drought and salinity were not additive stress factors for Atriplex spp. in this experiment. NaCl increased their ability to extract water from the soil solution compared to fresh water controls. ATLE showed increased shoot dry matter production and increased water use efficiency (WUE) as initial salinity levels increased from 0 to 30 g L−1 NaCl, whereas dry matter production and WUE peaked at 5 g L−1 for ATHO and ATCA. Final soil moisture salinities tolerated by species were 85 g L−1, 55 g L−1 and 160 g L−1 NaCl for ATHO, ATCA and ATLE, respectively. C4 photosynthesis and sodium accumulation in shoots were associated with high drought and salt tolerance.  相似文献

Protocorm cultures of Dendrobium candidum were established in balloon type bubble bioreactors using Murashige and Skoog (MS) medium with 0.5 mg l−1 α-naphthaleneacetic acid (NAA), 2.5% (w/v) sucrose, 5:25 mM NH4:NO3 and 1% (v/v) banana homogenate for the production of biomass and bioactive compounds. In 3 l bioreactor containing 2 l medium, a maximum protocorm biomass (21.0 g l−1 dry biomass) and also optimum quantities of total polysaccharides (389.3 mg g−1 DW), coumarins (18.0 mg g−1 DW), polyphenolics (11.9 mg g−1 DW), and flavonoids (4.5 mg g−1 DW) were achieved after 7 weeks of culture. Based on these studies, 5 and 10 l bioreactor cultures were established to harvest 80 g and 160 g dry biomass. In 10 l bioreactors, the protocorms grown were accumulated with optimal levels of polysaccharides (424.1 mg g−1 DW), coumarins (15.8 mg g−1 DW), polyphenols (9.03 mg g−1 DW) and flavonoids (4.7 mg g−1 DW). The bioreactor technology developed here will be useful for the production of important bioactive compounds from D. candidum.  相似文献

The inhibitory effect of ammonium sulfate on a commercial mixed culture, used in biological waste-water treatment was studied under aerobic batch conditions. Several mathematical models of enzyme and growth kinetics including a death factor were analyzed through nonlinear regression to find the best fit to corresponding data of inhibition. The best fit model was found to be the generalized Monod type with a death factor having the biokinetic parameters; μmax 0.681 h−1, Ks 0.224 g dm−3, Ki 56240 g dm−3, K 0.055 g dm−3 and kd 0.052 h−1 to represent the experimental data accurately. The low saturation coefficient value along with high maximum specific growth rate and inhibition coefficient denotes the competitive characteristics of commercial mixed cultures in the biological treatment of high ammonium polluted waste waters.  相似文献

Selective purification still poses a challenge in the downstream processing of biomolecules such as proteins and especially enzymes. In this study a polyethylene glycol 3000 (PEG 3000)–phosphate aqueous two-phase system at 25 °C and pH 7 was successfully used for laccase purification and separation. Initially, the effect of phase forming components on enzyme activities in homogenous systems was studied. In the course of the extraction experiments tie lines, enzyme source, initial enzyme activities, phase ratio and sodium chloride concentrations were varied and their influence on the activity partitioning was determined. Partitioning results were validated using clear-native-PAGE and isoelectric focusing. Based on these results, the separation of laccases from Trametes versicolor and Pleurotus sapidus was investigated using the principle of superposition. Sodium chloride was used to adjust laccase partitioning in the applied aqueous two-phase system (ATPS). Finally, two modes of operation are proposed depending on the aim of the purification task. One mode with 0.133 g g−1 of PEG3000, 0.063 g g−1 of phosphate and without sodium chloride separates P. sapidus laccases from T. versicolor laccases with clearance factors of 5.23 and 6.45, respectively. The other mode of operation with 0.124 g g−1 of PEG3000, 0.063 g g−1 of phosphate and 0.013 g g−1 of sodium chloride enables a partitioning of both laccases into the bottom phase of the ATPS resulting in a purification factor of 2.74 and 96% activity recovery.  相似文献

Conidiation and lytic enzyme production by Trichoderma viride at different solids concentration of pre-treated municipal wastewater sludge was examined in a 15-L fermenter. The maximum conidia concentration (5.94 × 107 CFU mL−1 at 96 h) was obtained at 30 g L−1 suspended solids. The maximum lytic enzyme activities were achieved around 12–30 h of fermentation. Bioassay against a fungal phytopathogen, Fusarium sp. showed maximum activity in the sample drawn around 96 h of fermentation at 30 g L−1 suspended solids concentration. Entomotoxicity against spruce budworm larvae showed maximum value ≈17290 SBU μL−1 at 30 g L−1 suspended solids concentration at the end of fermentation (96 h). Plant bioassay showed dual action of T. viride, i.e., disease prevention and growth promotion. The rheological analyses of fermentation sludges showed the pseudoplastic behaviour. In order to maintain required dissolved oxygen concentration ≥30%, the agitation and aeration requirements significantly increased at 35 g L−1 compared to 30 and 25 g L−1. The oxygen uptake rate and volumetric oxygen mass transfer coefficient, kLa at 35 g L−1 did not increase in comparison to 30 g L−1 due to rheological complexity of the broth during fermentation. Thus, the successful fermentation operation of the biocontrol fungus T. viride is a rational indication of its potential for mass-scale production for agriculture and forest sector as a biocontrol agent.  相似文献

In order to evaluate the dispersal pattern of airborne fluoride emissions, from a single source in the city of Ouro Preto, Brazil, the fluoride impact on some herbaceous plant species was studied using the plants as passive bioindicators. Foliar fluoride contents of eight species collected at different distances from an aluminium smelter were analyzed. The plant species were: Baccaharis dracunculifolia, Bidens pilosa, Borreria verticillata, Calopogonium mucunoides, Erigeron bonariensis, Hedychium coronarium, Ipomoea purpurea and Ipomoea cairica. In all species the fluoride accumulation decreased exponentially with the distance from the emission source. There was specific and distinct variation in fluoride accumulation among the species, a group of high-accumulator species (B. dracunculifolia and Bidens pilosa) and a group of low-accumulator species (I. cairica, H. coronarium and Borreria verticillata). C. mucunoides and E. bonariensis occupied an intermediate position. There was a pattern of plant contamination response during the periods analyzed. The plants nearest to the emission source, between 0.4 km northwest and 1.1 km east, showed fluoride contamination traits in leaves reaching values between 100 and 500 μg g−1. Moreover, fluoride contents higher than 1000 μg g−1 were found in these plants. At the most distant stations, situated 2.9 km northwest and 6 km east from the factory, the fluoride content of the dry matter was less than 10 μg g−1 showing that plants at those distances were submitted to minimum contamination. There were different patterns of tolerance among the species analyzed. While B. dracunculifolia accumulated fluoride up to 1500 μg g−1 in dry matter without any signs of injury, Borreria verticillata showed severe necrosis in leaves, but the fluoride content found was not higher than 120 μg g−1.  相似文献

Pseudomonas putida 33 wild strain, subjected to gamma ray mutagenesis and designated as P. putida 300-B mutant was used as microbial rhamnolipid-producer by using distant carbon sources (viz. hydrocarbons, waste frying oils ‘WFOs’, vegetable oil refinery wastes and molasses) in the minimal media under shake flask conditions. The behavior of glucose as co-substrate and growth initiator was examined. The 300-B mutant strain showed its ability to grow on all the substrates tested and produced rhamnolipid surfactants to different extents however; soybean and corn WFOs were observed to be preferred carbon sources followed by kerosene and paraffin oils, respectively. The best cell biomass (3.5 g l−1) and rhamnolipids yield (4.1 g l−1) were obtained with soybean WFO as carbon source and glucose as growth initiator under fed-batch cultivation showing an optimum specific growth rate (μ) of 0.272 h−1, specific product yield (qp) of 0.318 g g−1 h and volumetric productivity (PV) of 0.024 g l−1 h. The critical micelle concentration of its culture supernatant was observed to be 91 mg rhamnolipids l−1 and surface tension as 31.2 mN m−1.  相似文献

Continuous anaerobic fermentations were performed in a novel external-recycle, biofilm reactor using d-glucose and CO2 as carbon substrates. Succinic acid (SA) yields were found to be an increasing function of glucose consumption with the succinic acid to acetic acid ratio increasing from 2.4 g g−1 at a glucose consumption of 10 g L−1, to 5.7 g g−1 at a glucose consumption of 50 g L−1. The formic acid to acetic acid ratio decreased from an equimolar value (0.77 g g−1) at a glucose consumption of 10 g L−1 to a value close to zero at 50 g L−1. The highest SA yield on glucose and highest SA titre obtained were 0.91 g g−1 and 48.5 g L−1 respectively. Metabolic flux analysis based on the established C3 and C4 metabolic pathways of Actinobacillus succinogenes revealed that the increase in the succinate to acetate ratio could not be attributed to the decrease in formic acid and that an additional source of NADH was present. The fraction of unaccounted NADH increased with glucose consumption, suggesting that additional reducing power is present in the medium or is provided by the activation of an alternative metabolic pathway.  相似文献
