In this study, the soap stock as a sole carbon source was used for growing a carotenoid producing yeast (Rhodotorula rubra). The application of soap stock resulted in increase of carotenoids yield up to 5.36 folds when compared with the grown cultures on glucose. On the best Monod equation fitted on the specific growth rate (μ) data, the maximum specific growth rate (μm) and half-saturation concentration (KS) were respectively determined at 0.064 h−1 and 3.26 g L−1 for total fatty acids presented in soap stock. Further tests on the carotenogenesis process were carried out in a cell-immobilized airlift photobioreactor where the natural loofa sponge was used for immobilization of the cells. The performance of the bioreactor was statistically studied by the response surface methodology (RSM) where aeration rate of 0.11 vvm and light irradiation intensity of 2517 Lx provided an optimum condition for producing β-carotene with a specific production rate of 22.65 mg gcell−1 day−1.  相似文献

A kinetic model has been developed to estimate the specific growth rate of Phaeodactylum tricornutum in batch cultures. The cultures were carried out in a laboratory scale photobioreactor. Some factors like pH, temperature and irradiance were studied. In the first case, an optimum pH of 7.8 and a specific growth rate of 0.064 h−1 were achieved for certain nitrate conditions and illumination. The temperature influence has been modelled by a modified Sinclair model. The optimum temperature was achieved at 20.4 °C in aerated cultures and at 22.3 °C in non-aerated cultures. Better adaptation to low temperatures than high ones has been obtained. The experiments carried out with different irradiances drive to a simple Monod's equation for the irradiance influence on growth, with semi-saturation irradiance of 10.2 μEinstein−2 s−1 in aerated cultures and of 6.8 μEinstein m−2 s−1 in non-aerated cultures.  相似文献

Effects of irradiance and water flow on formation and growth of filamentous and spongy thalli of Codium fragile (Suringar) Hariot growing on vinylon threads were investigated at the laboratory culture. They showed clear differences in their irradiance and water flow requirements for their formation and growth. Spongy thalli were formed from the cultured filamentous thalli only at the high water flow velocity (10 cm s−1). Number of the spongy thalli remarkably increased with increasing irradiance because those at 10, 50 and 100 μmol m−2 s−1 reached 0, 2 and 76 thalli m−1, respectively, by 10 weeks of culture. In contrast, filamentous thalli were formed from the cultured spongy thalli at 0 and 3 cm s−1, and difference in irradiance had no effect on their formation. Growth of the spongy thalli greatly accelerated under the combination of the high irradiance and high water velocity (200 μmol m−2 s−1 and 10 cm s−1) because their relative growth rate in wet weight under the condition was two–four times higher than those at the other examined irradiances and water velocities. On the other hand, difference in water velocity had no effect on growth of the filamentous thalli under flowing water, and their growth decelerated at the high irradiance (200 μmol m−2 s−1). This demonstrates that water flow is a major factor controlling the formation of the spongy and filamentous thalli. The formation and growth of the spongy thalli surely occur under the combination of the high irradiance and fast flowing water. In contrast, the formation of the filamentous thalli occurs in the calm water, and their growth is inhibited under the high irradiance.  相似文献

Meristematic growth and loss of distal tissue from blades of two ecologically important species in the south-east Pacific, Lessonia nigrescens and Lessonia trabeculata, was evaluated during 1 year. Comparative growth was determined by a hole-punch method, loss of distal tissue from the blades was determined by subtracting final blade length (with loss) from expected blade lengths (without loss); growth and tissue loss were transformed to fresh biomass units for calculation of inter-algae differences. The results showed that blade elongation rate increased at the beginning of spring, and declined towards the end of summer, with mean values between 0.40 and 0.08 cm day−1 for L. nigrescens, and 0.65–0.17 cm day−1 for L. trabeculata. Loss of distal tissue varied seasonally when examined as length units for both species; with mean values between 0.24 and 0.10 cm day−1 for L. nigrescens, and 0.51–0.25 cm day−1 for L. trabeculata. Variations in fresh biomass units were only observed in Lessonia trabeculata, increasing in spring, with mean values to 0.13 g (fresh weight) day−1. Annual growth and loss of distal tissue were higher in L. trabeculata (0.41 and 0.39 cm day−1, respectively) than in L nigrescens (0.19 and 0.15 cm day−1). When growth and tissue loss were considered as fresh biomass, monthly gains significantly outweighed loss of distal tissue in both species, but parallel results based on length data followed a different trend. L. trabeculata released about 50% of its growth biomass as particulate organic matter, while the comparative value for L. nigrescens was about 20%.  相似文献

The relationship between light intensity, nitrogen availability and pigmentation was investigated in mixotrophic and heterotrophic cultures of the unicellular red alga Galdieria sulphuraria 074G, a potential host for production of the blue pigment, phycocyanin (PC). During the exponential growth phase of batch cultures, G. sulphuraria 074G contained 2–4 mg phycocyanin per g dry weight. In carbon-limited and nitrogen-sufficient batch cultures grown in darkness, this value increased to 8–12 mg g−1 dry weight during the stationary phase, whereas the phycocyanin content in nitrogen-deficient cells decreased to values below 1 mg g−1 dry weight during stationary phase. Light intensities between 0 and 100 μmol photons m−2 s−1 had no influence on phycocyanin accumulation in mixotrophic cultures grown on glucose or fructose, while light stimulated phycocyanin synthesis in cultures grown on glycerol, in which the phycocyanin content in stationary phase was increased from 10 mg g−1 dry weight in darkness to 20 mg g−1 dry weight at a light intensity of 80 μmol photons m−2 s−1. At higher light intensities, less phycocyanin accumulated than at lower intensities, irrespective of the carbon substrate used. In carbon-limited continuous flow cultures grown on glucose or glycerol at a dilution rate of 0.63 day−1, corresponding to 50% of the maximum specific growth rate, the highest steady-state phycocyanin content of 15–28 mg g−1 dry weight was found at 65 μmol photons m−2 s−1. In contrast to the apparent glucose repression of light-induced PC synthesis observed in batch cultures, no glucose repression of the light stimulation was observed in continuous flow cultures because the glucose concentration in the culture supernatant always remained at limiting levels. Despite the fact that G. sulphuraria 074G contains less phycocyanin than some other microalgae and cyanobacteria, the ability of G. sulphuraria 074G to grow and synthesize phycocyanin in heterotrophic or mixotrophic cultures makes it an interesting alternative to the cyanobacterium, Spirulina platensis presently used for synthesis of phycocyanin.  相似文献

Chara hispida forms dense beds (0.78–0.95 kg DW m−2) in Colgada Lake. The ability of Chara meadows to act as a nitrogen source or sink was evaluated by the following methods: (1) investigating Chara growth, (2) nitrogen incorporation and decomposition laboratory experiments and (3) relating experimental results to field conditions. Sediment oospores were germinated in large aquaria and observed growth rates were 0.001 m day−1 (shoot length) and 0.0002 g day−1 (dry weight). Nitrogen uptake rates were determined by addition of K15NO3 during two different periods of Chara growth and the rates were 1.21 and 3.86 μM g DW−1 h−1 when charophytes were 166 days old (not sexually mature) and 323 days old (sexually mature), respectively. After the uptake experiments, the same charophytes were allowed to decompose within two types of litter bags (3 mm-pore litter bags and entire, non-porous plastic litter bags). Decomposition rates of Ch. hispida were 0.016 and 0.009 day−1 in perforated and non-perforated bags, respectively, and fit a negative exponential model. The nitrogen release rate, calculated as the disappearance of N content from Chara tissues, was 0.012 day−1 and there were no statistically significant differences between the values from the two different bag types. The dissolved organic nitrogen concentrations in aquarium and non-perforated litter bags waters increased linearly with time due to the leaching of soluble compounds from Chara. The rate of N loss from Chara tissues, total nitrogen and dissolved organic nitrogen release rates and the decrease in initial dry weight rate were all lower than the daily rate of Chara N uptake. By extrapolating laboratory data to field situations, we determined that approximately 38% of the N taken up by charophytes in Colgada Lake during the growth period is retained. Given the high charophyte biomass in the lake, its ability to incorporate nitrogen, its low decomposition rate and its ability to over-winter, we conclude that Chara beds could be acting as nitrogen sinks in this ecosystem.  相似文献

Toxins produced as secondary metabolites can play important roles in phytoplankton communities and contribute to the ecological success of harmful algal bloom (HAB) taxa. Toxin composition and content in phytoplankton are affected by a suite of environmental factors, including nutrient availability. Changes in nutrient availability can increase or decrease toxin content and alter toxin composition, depending on toxin stoichiometry and the mechanisms by which nutrient limitation affects toxin production. The studies that have assessed the effects of nutrient availability on brevetoxin content of the HAB species Karenia brevis have reported contradictory results, although there is growing support that nutrient limitation increases brevetoxin content. In this study, we assessed the effects of decreased nitrogen (N) and phosphorus (P) availability on brevetoxin content and composition of K. brevis grown in chemostats at steady state by altering the nutrient supply ratios of incoming media from the Redfield Ratio. Overall, brevetoxin content was greatest in cultures grown at the lowest rate, regardless of the nutrient supply ratio (i.e., under both Redfield and N-limiting supply ratios). Compared to cultures grown at 0.2 d−1, cultures grown at 0.1 d−1 exhibited 5-fold increases in intracellular toxin content. In contrast, at constant growth rates, N-limiting supply ratios decreased intracellular brevetoxin content by approximately one-third, although this result was significant only in cultures growing at the fastest rate of 0.23 d−1. P-limiting supply ratios had no effect on brevetoxin content or composition. In addition, when cultures grown at rates of 0.2 d−1 were supplied with balanced/Redfield N:P supply ratios, but different absolute nutrient concentrations, toxin content was greater under greater nutrient concentrations. These findings suggest that when growth rate is not nutrient limited, there is a positive relationship between nutrient availability and brevetoxin content. This work contributes to previous studies by demonstrating strong growth rates effects on brevetoxin content and that growth rate and nutrient availability can independently or together affect toxin content of K. brevis. Moreover, our work underscores the value of the chemostat as a tool to elucidate the mechanisms by which nutrient availability and growth rate affect toxin production and content of HAB species.  相似文献

The filamentous fungus Paecilomyces lilacinus was grown on n-hexadecane in submerged (SmC) and solid-state (SSC) cultures. The maximum CO2 production rate in SmC (Vmax = 11.7 mg CO2 Lg−1 day−1) was three times lower than in SSC (Vmax = 40.4 mg CO2 Lg−1 day−1). The P. lilacinus hydrophobin (PLHYD) yield from the SSC was 1.3 mg PLHYD g protein−1, but in SmC, this protein was not detected. The PLHYD showed a critical micelle concentration of 0.45 mg mL−1. In addition, the PLHYD modified the hydrophobicity of Teflon from 130.1 ± 2° to 47 ± 2°, forming porous structures with some filaments <1 μm and globular aggregates <0.25 μm diameter. The interfacial studies of this PLHYD could be the basis for the use of the protein to modify surfaces and to stabilize compounds in emulsions.  相似文献

Bisexual populations of the charophyte Chara canescens (Desv. et Loisel. in Loisel., 1810) containing male and female individuals are rarely found. Two experiments were carried out to study whether male and female algae from the same site exhibit different physiological capacities, especially with respect to light acclimation.Algae from two different shore levels and from laboratory cultures acclimated to six irradiance conditions (35–500 μmol photons m−2 s−1) were compared. Field measurements showed that both female and male algae of C. canescens are able to acclimate to daily changes in solar irradiance. The quantum yield of Photosystem II (PSII) decreased with increasing irradiance in the morning and increased with decreasing irradiance in the afternoon. Growth experiments showed increasing growth rates from 35 μmol photons m−2 s−1 (∼7 mg FW) up to 500 μmol photons m−2 s−1 (∼27 mg FW) in female and male C. canescens. The irradiance saturation point for photosynthesis (Ek) was about 140 μmol m−2 s−1 for both sexes within the whole range of acclimation irradiances. The maximum photosynthesis rate at saturating irradiances (Pmax) of male algae was highest at Ek, whereas Pmax of female algae was highest at 500 μmol photons m−2 s−1. The photosynthetic efficiency in the light-limited range (α) increased in female C. canescens and decreased in male C. canescens. The ratio of the non-photochemical quenching parameter (NPQ) to the relative electron transport rates rETR(MT) increased in both sexes with irradiance, but showed a steeper increase in male than in female algae. Pigment analysis showed similar acclimation pattern for male and female C. canescens. Chl a/Chl b ratios of both sexes were constant over the whole range of Eg, whereas Chl a/carotenoid ratios in male and female C. canescens decreased from 70 μmol photons m−2 s−1 upwards. Pigment analysis pointed out that the carotenes α-, β- and γ-carotene were more prominent in male than in female algae.Our results indicate that female C. canescens are more efficient in light acclimation than male algae from the same site. Nevertheless, further investigations of bisexual C. canescens populations resolving CO2-uptake mechanisms and/or genetic differences are needed.  相似文献

Soil respiration is the main form of carbon flux from soil to atmosphere in the global carbon cycle. The effect of temperature on soil respiration rate is important in evaluating the potential feedback of soil organic carbon to global warming. We incubated soils from the alpine meadow zone and upper rocky zone along an altitudinal gradient (4400–5500 m a.s.l.) on the Tibetan Plateau under various temperature and soil moisture conditions. We evaluated the potential effects of temperature and soil moisture on soil respiration and its variation across altitudes. Soil respiration rates increased as the temperature increased. At 60% of soil water content, they averaged 0.21–5.33 μmol g soil−1 day−1 in the alpine meadow zone and 0.11–0.50 μmol g soil−1 day−1 in the rocky zone over the experimental temperature range. Soil respiration rates in the rocky zone did not increase between 25 and 35 °C, probably because of heat stress. Rates of decomposition of organic matter were high in the rocky zone, where the CN ratio was smaller than in the middle altitudes. Soil respiration rates also increased with increasing soil water content from 10% to 80% at 15 °C, averaging 0.04–2.00 μmol g soil−1 day−1 in the alpine meadow zone and 0.03–0.35 μmol g soil−1 day−1 in the rocky zone. Maximum respiration rates were obtained in the middle part of the alpine slope in any case of experimental temperature and soil moisture. The change patterns in soil respiration rate along altitude showed similar change pattern in soil carbon content. Although the altitude is a variable including various environmental factors, it might be used as a surrogate parameter of soil carbon content in alpine zone. Results suggest that temperature, soil moisture and altitude are used as appropriate environmental indicators for estimating the spatial distribution of potential soil respiration in alpine zone.  相似文献

The influence of nitrogen and phosphorus pulses on Chaetomorpha linum (Muller) Kutzing growth and photosynthesis was studied in laboratory experiments. Photosynthesis and growth of C. linum from Tancada lagoon seems limited by both nitrogen and phosphorus, as indicated by the high rate (4.7–11.6 mg O2 g−1 dry weight h−1) of light-saturated photosynthesis (Pm) and growth rates observed under nitrogen plus phosphorus enrichment in relation to enrichment by nitrogen alone (2.9–7.6 mg O2 g−1 dry weight h−1). Significant increase in nitrogen and phosphorus content as percentage of dry weight was observed in C. linum fertilized with a single nutrient or with nitrogen plus phosphorus. In Tancada lagoon, when availability of nitrogen to primary producers is by pulses, an increase of nitrate concentration in the water column (from 6 to 100 μM) has a greater effect on growth of C. linum (growth rate: 0.13 day−1) than an increase in ammonium concentration (from 20 to 100 μM and growth rate: 0.11 day−1). For a given thallus nitrogen content (0.6–1.4% N), both Pm and the photosynthetic efficiency (α) normalized to dry weight were correlated (r2 = 0.73, p < 0.005) indicating that variations in electron transport were coupled to variations in C-fixation capacity. Optimizing both α and Pm may be a general characteristic of thin-structured opportunistic algae in more variable estuarine environments.  相似文献

Domoic acid (DA) poisoning in the southern part of the California Current System has been associated typically with blooms of Pseudo-nitzschia australis. The environmental variables that promote growth and DA production in the Mexican part of this system have not been identified. The present study investigated the effect of temperature and two nutrient ratios on the growth characteristics and DA content of two (BTS-1, BTS-2) P. australis strains isolated from the Pacific coast of northern Baja California peninsula, México. Of the different temperatures assayed (10, 12, 14, 15, 18 and 20 °C), the maximum cell abundance was detected at 12 °C for BTS-2 and 14 °C for BTS-1. The highest maximum specific growth rate (1.69 day−1) was measured at 15 °C for BTS-2. With the exception of cells maintained at 15 °C, growth characteristics were similar in P. australis cultured in a high Si:NO3 (2.5) or low Si:NO3 (0.5) ratio at each temperature. Dissolved (dDA) and cellular (cDA) DA content measured at the stationary phase of growth was similar in cells cultivated at the different temperatures. No difference in cDA (between 0.11 and 1.87 pg DA cell−1) was observed in cells cultivated at the two nutrient ratios. To evaluate if P. australis accumulates DA (cDA + dDA) at different stages of the culture and not only during the stationary phase of growth, the BTS-1 strain was cultivated at 14 °C and the content of this toxin was measured during culture development. The cultures were maintained at high (HL; 200 μmol quanta m−2 s−1) and low light (LL; 30 μmol quanta m−2 s−1) and in the two nutrient ratios to evaluate the effect of these variables on DA content. The photosynthetic performance and pigment concentration were measured as indicators of the physiological condition of the cells. cDA was detected in all culture conditions and during the different stages of growth. The highest DA content was measured during the lag phase of growth and it was present mainly in the medium (dDA = 70.83 pg DA cell−1). Cells cultivated at HL produced more DA than LL cultured cells. P. australis cultured in HL presented lower photosynthetic rates than LL cells and had similar concentrations of photoprotective pigments and the highest maximum photosynthetic rates were detected during the lag phase of growth in all culture conditions. The results demonstrate that P. australis from northern Baja California peninsula presents a narrow temperature range for optimal growth under batch culture conditions. P. australis produce DA at different stages of growth, and DA content was related to the light intensity at which the cells were cultivated.  相似文献

The responses of decomposition to nitrogen (N) and phosphorus (P) supply were investigated in three leaf species: Eichhornia crassipes, Vallisneria natans, and Potamogeton maackianus. Decomposition was fastest in E. crassipes (0.047–0.099 day−1), intermediate in V. natans (about 0.030 day−1), and slowest in P. maackianus (about 0.010 day−1). Increase in P-availability increased the decomposition rate of E. crassipes by 68–87%, whereas the impact of N-availability alone was insignificant. Both N- and P-availability in waters had no significant impact on the decomposition rates of V. natans and P. maackianus (P > 0.05). The effects of P-availability on the N and P content levels of the three species were significant (P < 0.01), except for the impact on N content of V. natans (P = 0.526). In contrast, environment N-availability was insignificant. These results indicate that the responses of decomposition to nutrient availability depend on plant species and nutrient type. P-availability has stronger effects on litter nutrient dynamics than N-availability.  相似文献

The mixed cultures has been isolated from industrial saline wastewater contaminated with chromium(VI), using enrichment in the presence of 50 mg l−1 chromium(VI) and 4% (w/v) NaCl at pH 8. In this study, the molasses (M) medium was selected a suitable medium for the effective chromium bioaccumulation by the mixed cultures. Eleven pure isolates obtained from mixed cultures and some of them showed high bioaccumulation in the M media containing about 100 mg l−1 chromium(VI) and 4% NaCl. The strain 8 (99.3%) and 10 (99.1%) were able to bioaccumulate more efficient than the mixed culture (98.9%) in this media. But the highest specific Cr uptake was obtained by the mixed cultures followed by strain 8 and 10 with 56.71, 33.14 and 21.7 mg g−1, respectively. Bioaccumulation of chromium(VI) ions by the strain 8 growing in the media with chromium(VI) and NaCl was studied in a batch system as a function of initial chromium(VI) (86.6–547.6 mg l−1) and NaCl (0, 2, 4, 6% w/v) concentrations. During all the experiments, the uptake yield of the strain 8 was highly affected from NaCl concentrations in the medium at high initial chromium(VI) concentrations. But at low chromium(VI) concentration, strain 8 was not affected from NaCl concentrations in the medium. The maximum uptake yield were obtained in the M media with 2% NaCl as 98.8% for 110.0 mg l−1, 98.6% for 217.1 mg l−1, 98.6% for 381.7 mg l−1 and 98.2% for 547.6 mg l−1 initial chromium(VI) concentrations. The strain 8 tolerated a 6% (w/v) NaCl concentration was able to bioaccumulate more than 95% of the applied chromium(VI) at the 97.6–224.4 mg l−1 initial chromium(VI) concentrations. The results presented in this paper was shown that these pure and mixed cultures might be of use for the bioaccumulation of chromium(VI) from saline wastewater.  相似文献

A mixed fermentation strategy based on exponentially fed-batch cultures (EFBC) and nutrient pulses with sucrose and yeast extract was developed to achieve a high concentration of PHB by Azotobacter vinelandii OPNA, which carries a mutation on the regulatory systems PTSNtr and RsmA-RsmZ/Y, that negatively regulate the synthesis of PHB. Culture of the OPNA strain in shake flaks containing PY-sucrose medium significantly improved growth and PHB production with respect to the results obtained from the cultures with the parental strain (OP). When the OPNA strain was cultured in a batch fermentation keeping constant the DOT at 4%, the maximal growth rate (0.16 h−1) and PHB yield (0.30 gPHB gSuc−1) were reached. Later, in EFBC, the OPNA strain increased three fold the biomass and 2.2 fold the PHB concentration in relation to the values obtained from the batch cultures. Finally, using a strategy of exponential feeding coupled with nutrient pulses (with sucrose and yeast extract) the production of PHB increased 7-fold to reach a maximal PHB concentration of 27.3 ± 3.2 g L−1 at 60 h of fermentation. Overall, the use of the mutant of A. vinelandii OPNA, impaired in the PHB regulatory systems, in combination with a mixed fermentation strategy could be a feasible strategy to optimize the PHB production at industrial level.  相似文献

Microbial electrolysis cells (MECs) with autotrophic biocathode are a promising technology for removal of pollutants in wastewater. The aim of this study was to investigate the effect of initial acidity of wastewater on performance of sulfate-reducing biocathodes. MECs with biocathodes were operated with initial pH values of catholyte ranged from 3.0 to 7.0. The optimum initial pH value was 6.0 with a maximum sulfate reductive rate and biomass of 57 mg L−1 d−1 and 2.1 ± 0.4 mg g−1, respectively. With initial pH 7.0, the pH value of catholyte increased to 9.8 ± 0.2 after an operation cycle, which resulted in low performance of the biocathode. A considerable sulfate reductive rate of 31 ± 0.85 mg L−1 d−1 was achieved with initial pH 3.0. Desulfovibrio sp. grew dominantly with abundance of 46%–66% in the cathode biofilm with initial pH values from 3.0 to 6.0 and contributed to the sulfate reduction. Clostridium and Parapedobacter also had high abundance in pH 6.0 cathode, indicated that interspecies electron transfer between electrochemical active and sulfate-reducing bacteria could play an important role in sulfate removal. The results suggest that acidity of catholyte is an important factor to be considered to utilize autotrophic biocathode MECs for wastewater treatment.  相似文献

The photosynthetic and repiratory metabolism of Zostera marina and maerl communities was compared, in the same area of the Bay of Brest in March–April, using benthic chambers. PE curves for both oxygen and carbon were established for bottom irradiances between 0 and 525 μmol m−2 s−1. An exponential function was fitted to calculate daily production. Community metabolic quotients did not differ for maerl and seagrass beds. Community photosynthetic quotients were significantly higher (1.19) whereas community respiratory quotients were lower (0.70) than 1. Maerl and seagrass bed PE curves mainly differed by the minimum saturating irradiance (Ek). Net community production was estimated to 26.8 mmol C m−2 d−1 for Z. marina meadows and 8.6 mmol C m−2 d−1 for maerl beds. The two communities can, therefore, be considered as autotrophic during the March–April period. Community respiration did not differ between Z. marina meadows and maerl beds, with an average value of 53.8 mmol C m−2 d−1 during a day. In this study, a protocol for initiation of callus and shoot cultures from leaves and shoot tips explants of different silybium genotypes collected from different locations in Egypt was established. Callus cultures were initiated from leaves explants and exposed to different concentrations of the precursor (coniferyl alcohol). Shoot cultures were initiated from shoot tips explants. Moreover, the produced plants of the different Silybium shoots as well as intact plants were subjected to protein screening using SDS–PAGE analysis.Results obtained revealed that the optimum medium for growth and maintenance of friable callus was MS medium supplemented with 0.25 mg L−1 2,4-Dichlorophenoxy acetic acid (2,4-D) + 0.25 mg L−1 Kinetin (Kin). The best medium for proliferation of high number of shoots was MS-medium with 0.25 mg L−1 each of Benzyl Adinine (BA) and Naphthalene Acetic Acid (NAA). Coniferyl alcohol in concentration of 30 μM caused an increase in accumulation of silymarin contents in most callus cultures. SDS–PAGE of different Silybium shoots revealed that the protein profiles of 100% of in vitro produced plantlets similar to their control.  相似文献

A pilot-scale vertical submerged membrane bioreactor (VSMBR) with anoxic and oxic zones in one reactor was operated in an attempt to reduce the problems concerning effective removal of organic matter and nutrients from municipal wastewater. Source water with total chemical oxygen demand (TCOD)/total nitrogen (TN) ratio of 5.5 was treated at various temperatures (13–25 °C) over an interval of about 1 year. As a result, total suspended solid (TSS) and TCOD were removed by 100% and higher than 98%, respectively. Moreover, the average removal efficiencies of TN and total phosphorus (TP) were found to be 74% and 78% at 8 h-hydraulic retention time (HRT) and 60-days sludge retention time (SRT). Under these conditions, the specific removal rates (SRR) of TN and TP were found to be 0.093 kg N m−3 day−1 and 0.008 kg P m−3 day−1, and the daily production of excess sludge (DPES), 0.058 kg TSS day−1.  相似文献

In the frame of a feeding experiment with three periods, balance trials were carried out with 18 double-muscled Belgian White–blue bulls, allocated to one of four feeding regimes. The ration consisted of maize silage and concentrate in the ratio of 35:65 on DM basis. Six concentrates were formulated to supply three levels of protein and energy. The three periods corresponded to distinct live weight intervals of 360–460, 460–570 and 570–680 kg. In the first feeding regime, low protein and intermediate energy level were given during the whole trial; in the second regime, protein level decreased at a constant intermediate energy level; in the third regime, the energy level increased at a constant high protein level; in the fourth regime, protein level decreased simultaneously with an increase in the energy level. Total daily intake varied from 6.2 to 9.7 kg for dry matter (DM), from 65 to 106 MJ for metabolisable energy (ME) and from 719 to 1326 g for crude protein (CP) (50 animal observations). At the end of each period, the excretion of the purine derivatives (PD), allantoin and uric acid, was measured after total urine collection during 4 days to estimate microbial nitrogen supply to the duodenum (MNPD). The effect of the intake of DM, organic matter (OM), digestible OM (DOM), digestible carbohydrates (DCHO), total digestible nutrients (TDN), rumen fermentable OM (FOM), metabolisable energy (ME), fermentable ME (FME), CP, digestible CP (DCP) and rumen degradable protein (RDP) on PD and MNPD was examined. Further, the relationship of MNPD to MN, calculated according to different systems, was examined. The amount (mean±SD) of allantoin excreted in urine was 147±23 mmol day−1 and of uric acid 11±3 mmol day−1. The MNPD amounted to 97±21 g day−1, varying from 57 to 154 g day−1, and significantly increased with all measures of nutrient intake. The correlation coefficients, ranging from 0.45 for DCP to 0.57 for DOM and FME, were, however, not significantly different. MNPD showed a larger variation and was on average lower than the potential MN values calculated from the intake of FOM (108±13 g day−1), DCHO (141±16 g day−1) and FME (109±13 g day−1), similar to that calculated from the intake of RDP (99±14 g day−1) and higher than the MN value calculated from the intake of TDN (84±11 g day−1). The correlations of MNPD to the calculated MN values ranged from 0.47 for MNFOM to 0.59 for MNFME, but were not significantly different.  相似文献
