The capacity of mitochondria from callus and virus tumour tissuecultures of Rumex acetosa L. to oxidize Krebs-cycle acids wasinvestigated. The rate of oxygen uptake of callus mitochondriawas considerably greater. The rate of substrate oxidation bymitochondria from both tissues decreased with age of the culture.Callus cells were found to contain considerably greater quantitiesof mitochondria than tumour cells. The oxidative activitiesof mitochondria and growth-rates of tissues were affected bythe composition of the culture medium. There was no direct relationshipbetween growth-rate and oxidative activity.  相似文献

ABSTRACT: BACKGROUND: Patterns of genetic divergence between populations of facultative metallophytes have been investigated extensively. However, most previous investigations have focused on a single plant species making it unclear if genetic divergence shows common patterns or, conversely, is species-specific. The herbs Rumex acetosa L. and Commelina communis L. are two pseudo-metallophytes thriving in both normal and cupriferous soils along the middle and lower reaches of the Yangtze River in China. Their non-metallicolous and metallicolous populations are often sympatric thus providing an ideal opportunity for comparative estimation of genetic structures and divergence under the selective pressure derived from copper toxicity. RESULTS: In the present study, patterns of genetic divergence of R. acetosa and C. communis, including metal tolerance, genetic structure and genetic relationships between populations, were investigated and compared using hydroponic experiments, AFLP, ISSR and chloroplast genetic markers. Our results show a significant reduction in genetic diversity in metallicolous populations of C. communis but not in R. acetosa. Moreover, genetic differentiation is less in R. acetosa than in C. communis, the latter species also shows a clustering of its metallicolous populations. CONCLUSIONS: We propose that the genetic divergences apparent in R. acetosa and C. communis, and the contrasting responses of the two species to copper contamination, might be attributed to the differences in their intrinsic physiological and ecological properties. No simple and generalised conclusions on genetic divergence in pseudo-metallophytes can thus be drawn.  相似文献

Infections by influenza A viruses (IAV) are a major health burden to mankind. The current antiviral arsenal against IAV is limited and novel drugs are urgently required. Medicinal plants are known as an abundant source for bioactive compounds, including antiviral agents. The aim of the present study was to characterize the anti-IAV potential of a proanthocyanidin-enriched extract derived from the aerial parts of Rumex acetosa (RA), and to identify active compounds of RA, their mode of action, and structural features conferring anti-IAV activity. In a modified MTT (MTTIAV) assay, RA was shown to inhibit growth of the IAV strain PR8 (H1N1) and a clinical isolate of IAV(H1N1)pdm09 with a half-maximal inhibitory concentration (IC50) of 2.5 µg/mL and 2.2 µg/mL, and a selectivity index (SI) (half-maximal cytotoxic concentration (CC50)/IC50)) of 32 and 36, respectively. At RA concentrations>1 µg/mL plaque formation of IAV(H1N1)pdm09 was abrogated. RA was also active against an oseltamivir-resistant isolate of IAV(H1N1)pdm09. TNF-α and EGF-induced signal transduction in A549 cells was not affected by RA. The dimeric proanthocyanidin epicatechin-3-O-gallate-(4β→8)-epicatechin-3′-O-gallate (procyanidin B2-di-gallate) was identified as the main active principle of RA (IC50 approx. 15 µM, SI≥13). RA and procyanidin B2-di-gallate blocked attachment of IAV and interfered with viral penetration at higher concentrations. Galloylation of the procyanidin core structure was shown to be a prerequisite for anti-IAV activity; o-trihydroxylation in the B-ring increased the anti-IAV activity. In silico docking studies indicated that procyanidin B2-di-gallate is able to interact with the receptor binding site of IAV(H1N1)pdm09 hemagglutinin (HA). In conclusion, the proanthocyanidin-enriched extract RA and its main active constituent procyanidin B2-di-gallate protect cells from IAV infection by inhibiting viral entry into the host cell. RA and procyanidin B2-di-gallate appear to be a promising expansion of the currently available anti-influenza agents.  相似文献

Callus cultures were established from dioecious plant species Rumex acetosella and R. acetosa, using cotyledons, hypocotyls and stem tips of aseptically germinated seedlings as primary explants. Cultures were also established from male and female R. acetosella adult plants, starting from vegetative lateral buds. Cell division was induced using a high 2,4-D concentration, while bud induction and multiplication were stimulated on a medium with high BAP/IAA ratio. Cotyledon fragments of both species produced only rhizogenic calli. Hypocotyl-derived calli of R. acetosella produced buds, while those of R. acetosa showed no bud forming response under these conditions. Bud multiplication occurred in stem tip cultures of both species and in lateral bud cultures of R. acetosella. Calli derived from male plants produced more buds than those from female. Shoots were easily rooted using IBA, and plantlets were effectively transferred to soil. Flowering was not induced in culture. The sex of regenerated male and female plants was not altered by the culture conditions.  相似文献

The Rumex acetosa L. in China possesses sex-chromosomes 2n=12A+XX in female plant and 2n=12A+XY1Y2 in male plant. The sex-chromosomes of male plant exhibit a chain of trivalent (Y1-X-Y2) in diakinesis of PMC meiosis. At metaphase Ⅰ, they make a V-shaped or a ring configuration. Some of them may form a XY-bivalent and a Y-univalent. Finally two kinds of reale gametes e.g. 6A+X and .6A+YY, are produced. Monoecious plant may be found in R. acetosa and has 22 chromosomes (22=18A+ XX+YY). This is a triplont with sterility. Cells with different chromosome number in the same plant have also seen in R. acetosa, and such plant never blooms in its life.  相似文献

The dioecious plant Rumex acetosa shows intraspecific karyotype variation, caused by supernumerary heterochromatic segments or DAPI (4',6-diamidino-2 phenylindole)-bands at the ends of the short arms of three pairs of autosomes. A DNA sequence (RAE730) specific to the supernumerary heterochromatic segments was cloned and sequenced. RAE730 was about 730 bp and AT-rich (71% AT-content). Using fluorescence in situ hybridization (FISH), RAE730 was localized in the supernumerary DAPI-positive heterochromatic segments on several mitotic chromosomes and chromocenters in interphase nuclei, but not in the DAPI-bands of Y or B chromosomes. RAE730 was tandemly arranged in the genome, and the copy number varied between plants from 40000 to 304000 copies per 2C, corresponding to the relative amount of supernumerary heterochromatic segments per genome. These results indicate that the karyotype variation caused by the supernumerary heterochromatic segment was generated by amplification or reduction of the tandem repeats of RAE730.  相似文献

从 1 0个大豆品种中筛选出两个耐铝性差异显著的品种 ,研究了其耐铝性与有机酸的关系。经铝处理后 ,吴川品种的相对根长为 1 3 3 .5 % ,化州只有 68.9% ,表明吴川相对耐铝 ,化州对铝较敏感。将不同浓度的AlCl3 加入营养液中处理大豆 1 0d,化州较吴川根长受到较大影响 ,进一步证实吴川相对耐铝毒 ,而化州对酸铝敏感。机理研究发现大豆在铝胁迫下根系可分泌两种有机酸 (草酸、柠檬酸 ) ,其中吴川根系草酸分泌速率提高了 74% ,化州几乎没有提高 ,表明耐铝性大豆品种的根系草酸分泌速率明显提高 ,可增强其缓解酸铝毒性的能力。而二者分泌柠檬酸的速率虽然均有显著提高 ,但处理后感抗品种之间差异不大 ,表明柠檬酸在缓解铝毒性中的作用不大。铝处理下大豆根系虽然分泌两种有机酸 ,但草酸在大豆耐酸铝机制中的作用可能更为重要。  相似文献

Phenolic acids are active antimicrobial compounds and root signaling molecules that play important roles in plant defense responses. They are generally present in plants as glycosides or esters. A range of soluble and bound phenolic acids were detected in roots and root nodules of Arachis hypogaea L., among which five were identified by high performance liquid chromatography (HPLC) coupled with UV–Vis diode array detector (DAD), viz., p-coumaric acid (p-com), p-hydroxybenzaldehyde (HBAld), p-hydroxybenzoic acid (HBA), caffeic acid (CA) and protocatechuic acid (PA). Para-coumaric acid was constitutively present in all fractions whereas HBA was present in the soluble form only in young nodules. CA and PA were mostly present in the wall bound fraction. The root nodules contain higher concentration of phenolic acids than non-nodulated roots and presence of peroxidase and polyphenol oxidase indicate the metabolism of phenolic acids in roots and root nodules. These results indicate that phenolic acids (p-com and CA) in bound-glycosidic or ester forms were major components in cell wall fortification which provide protection to the root nodule from pathogen attack.  相似文献

Detailed meiotic analysis in 28 North West Himalayan populations of dioecious plant Rumex acetosa L. was carried out. The species is generally discussed as an important plant having sex chromosomes. Male meiosis in all the studied populations clearly showed the formation of six bivalents and one trivalent during diakinesis and metaphase-I. The sex chromosomes in male plants exhibit a chain of trivalent (Y1–X–Y2). In addition, among the presently investigated populations ring-shaped trivalents were also observed for the first time in the species. Varied frequency of abnormal segregation of sex trivalent was also observed leading to XY:Y segregation instead of normal X:Y1Y2 segregation. A majority of the populations exhibit normal meiosis. Plants of six populations show meiotic abnormalities like cytomixis, laggards, bridges, chromatin stickiness, etc., leading to reduced pollen fertility. Translocation between an autosome and sex chromosomes was also observed in some of the populations. 0–1B chromosomes were noticed in one population. This is the first ever meiotic analysis of the species from India.  相似文献

Summary Patterns of resource allocation in the dioecious Rumex acetosa and R. acetosella were investigated. Males were found to allocate more to reproduction during flower production than females, whereas females invested considerably more in reproduction during seed production. Altogether, females allocated both a higher total amount and a higher proportion of energy to reproduction than did males. By regression analysis, the influence of plant size on reproductive effort was examined separately for males and females. The results indicated that while reproductive effort is sometimes lower for tall plants than for small plants, size-independent effects have a greater influence on reproductive effort than size distribution. An analysis of variance was conducted to investigate the effects of population, season, sex and their interactions on plant size, and an analysis of covariance was used to study differences in resource allocation patterns. Different interaction effects were found to be most important in the two species of Rumex.  相似文献

  • 2 The ovipositional behaviour, larval development, emergence and hibernation of A. curtirostre are described.
  • 3 From dissection of samples of stems of R. acetosa, abundance curves for the stages of A. curtirostre and its chalcid parasitoids were constructed and the between-stage mortalities determined.
  • 4 There is an exponential relationship between R. acetosa stem height and number of A. curtirostre eggs laid.
  • 5 Male stems of R. acetosa appear earlier and are smaller than female stems. Taller stems, at least in female plants, live longer than shorter ones.
  • 6 Apion survival is lower in male than in female stems.
  • 7 Stem sex is the primary determinant of opposition choice with stem thickness a secondary determinant.
  • 8 A. curtirostre eggs introduced experimentally into stems of four Rumex species survived significiantly better in R. acetosa than in any other species. Survival in R. acetosa female plants was twice that in males.

The role of gibberellin (GA) and ethylene in submergence-induced petiole elongation was studied in two species of the genus Rumex. Analysis of endogenous GAs in the flooding-tolerant Rumex palustris Sm. and the intolerant Rumex acetosa L. by gas chromatography-mass spectrometry showed for both species the presence of GA1, GA4, GA9, GA19, GA20 and GA53. Gas chromatography-mass spectrometry analysis of R. palustris petiole tissue of submerged plants showed an increase in levels of 13-OH GAs, especially GA1, compared with drained plants. This effect could be mimicked by application of 5 μL L−1 ethylene. In R. acetosa, no differences between levels of GAs in drained or submerged plants were found. In R. palustris, both submergence and ethylene treatment sensitized petioles to exogenous gibberellic acid (GA3). In R. acetosa the effect was opposite, i.e. submergence and ethylene de-sensitized petioles to GA3. Our results demonstrate the dual effect of ethylene in the submergence response related to flooding tolerance, i.e. in the flooding-tolerant R. palustris ethylene causes an increased concentration of and sensitivity to GA with respect to petiole elongation while in the intolerant R. acetosa ethylene reduces growth independent of GAs. Abstract After the onset of imbibition, the dormant seeds of Rumex obtusifolius and R. crispus are stimulated to germinate by a change from an initial low temperature to a warmer temperature for a relatively brief period: the warmer that temperature the shorter is the optimum period spent at it, and this optimum value is unaffected by the initial temperature. The optimum period is more critical in R. crispus than in R. obtusifolius (about 1 h and 2.5 to 4 h, respectively, for a warmer temperature of 35°C in the dark); in the light the length of the period at the warmer temperature is less critical in both species. The sensitivity of the seeds to the change to the warmer temperature increases with time from the start of imbibition at a rate which is positively related to the initial temperature. In R. obtusifolius maximum sensitivity was typically reached after 3 to 5 d when the initial temperature was 20°C and then remained constant, or declined only slightly, over the period investigated (10 d). At the same initial temperature, however, R. crispus showed a cyclical pattern of sensitivity with peaks occurring at 3–4 d intervals from the start of imbibition.  相似文献

The aluminium cation Al(3+) is toxic to many plants at micromolar concentrations. A range of plant species has evolved mechanisms that enable them to grow on acid soils where toxic concentrations of Al(3+) can limit plant growth. Organic acids play a central role in these aluminium tolerance mechanisms. Some plants detoxify aluminium in the rhizosphere by releasing organic acids that chelate aluminium. In at least two species, wheat and maize, the transport of organic acid anions out of the root cells is mediated by aluminium-activated anion channels in the plasma membrane. Other plants, including species that accumulate aluminium in their leaves, detoxify aluminium internally by forming complexes with organic acids.  相似文献
