Quantifying and optimizing the polyphenol content of Phyllanthus maderaspatensis was accomplished using a single-solvent HPTLC system. Analyzing hydroalcoholic extracts for kaempferol, rutin, ellagic acid, quercetin, catechin, and gallic acid, we simultaneously quantified and optimized their concentration. In the experiment, the methanol to water ratio (%), temperature (°C), and time of extraction (min) were all optimized using a Box-Behnken statistical design. Kaempferol, rutin, ellagic acid, quercetin, catechin, and gallic acid were among the dependent variables analyzed. In the HPTLC separation, silica gel 60F254 plates were used, and toluene, ethyl acetate, and formic acid (5:4:1) made up the mobile phase. For kaempferol, rutin, ellagic acid, quercetin, catechin, and gallic acid, densitometric measurements were carried out using the absorbance mode at 254 nm. Hydroalcoholic extract of P. maderaspatensis contains rutin (0.344), catechin (2.62), gallic acid (0.93), ellagic acid (0.172), quercetin (0.0108) and kaempferol (0.06). Further, it may be affected by more than one factor at a time, resulting in a varying degree of reaction. A negative correlation was found between X1 (extraction time (min)) and X2 (temperature), as well as X1 and X3 (solvent ratios). Taking these characteristics into consideration, the method outlined here is a validated HPTLC method for measuring kaempferol, rutin, ellagic acid, quercetin, catechin, and gallic acid.  相似文献

为了解贵州金刺梨(Rosa sterilis D.Shi)果实和叶片中的活性成分及其抗氧化活性,以贵州普定县金刺梨种植基地的果实和叶片为试材,测定其活性成分含量及其抗氧化活性,并对各项指标进行相关性分析。结果显示:没食子酸、芦丁、槲皮素、儿茶素、鞣花酸、绿原酸、阿魏酸是供试金刺梨果实和叶片的主要酚类成分,金刺梨果实和叶片中活性组分差异显著(P<0.05),果实中p-香豆酸、总黄酮和抗坏血酸的含量相对较高,而叶片中没食子酸、儿茶素、绿原酸、表儿茶素、阿魏酸、鞣花酸、芦丁、槲皮素和总酚含量均高于果实;金刺梨果实抗氧化活性值均显著高于叶片(P<0.05);相关性分析发现:总黄酮对总还原力(TRPA)值的贡献极强,抗坏血酸对Fe3+还原抗氧化能力(FRAP)值贡献最强,槲皮素对ABTS值的贡献最强,说明金刺梨果实和叶片是一种具有较高开发价值的药食同源资源。  相似文献

Five traditional medicinal food from the Tibetan plateau including Nitraria tangutorum Bobrov (NT), Hippophae rhamnoides L. (HR), Lycium ruthenicum Murray (LR), Lycium barbarum L. (LB) and Rubus corchorifolius L.f. (RC) are rich in phenolic compounds. However, the detailed studies about the phenolic compounds remain scarce. Therefore, we established a rapid method for the simultaneous identification and quantification of the phenolic compounds from berries via Ultra Performance Liquid Chromatography‐Quadruple‐Orbitrap MS system (UPLC‐Q‐Orbitrap MS). This method was verified from many aspects including detection limit, quantification limit, precision, repeatability, stability, average recovery rate and recovery range, and then was used to analyze the phenolic compounds in these five species of berries. Finally, a total of 21 phenolic compounds were directly identified by comparing the retention time and exact mass, of which 14 compounds were identified by us for the first time in berries from the Tibetan plateau, including one flavonoid aglycone (myricetin), 11 phenolic acids (gallic acid, protocatechuate, chlorogenic acid, vanillic acid, caffeic acid, syringic acid, p‐coumaric acid, ferulic acid, 2‐hydroxybenzeneacetic acid and ellagic acid), one flavanol (catechin) and one dihydrochalcone flavonoid (phloretin). Quantitative results showed that rutin, myricetin, quercetin and kaempferol were the main flavonoids. Moreover, a variety of phenolic acid compounds were also detected in most of the berries from the Tibetan plateau. Among these compounds, the contents of protocatechuate and chlorogenic acid were high, and high levels of catechin and phloretin were also detected in these plateau berries.  相似文献

Antioxidant potential of Rosa L. leaves methanolic extracts was evaluated in vitro using a spectrophotometric method based on measuring the radical scavenging effect on 2,2-di-phenyl-1-picrylhydrazyl (DPPH) radicals. The contents of ellagic acid, quercetin and kaempferol in the extracts from leaves of seventeen rose species were determined using SPE-RP-HPLC methods. Additionally, total phenolic content was determined spectrophotometrically according to the Folin-Ciocalteu procedure and calculated as gallic acid equivalents (GAE). Remarkable high antioxidant activity and high total phenolic content (5.7% < GAE < 15.2%), large ellagic acid (EA) content from 9.37 to 19.42 mg/g of dry weight, a quercetin content ranging from 3.68 to 15.81 mg/g of dry weight and kaempferol content from 1.25 to 9.41mg/g of dry weight were found in rose leaves. Significant correlation between EA (r(2) = 0.6131), quercetin (r(2) = 0.5158), total phenolic content (r(2)= 0.8485) and antioxidant activity was observed. Basing on the studies conducted one may assume that the extracts of rose leaves are a rich source of natural antioxidants and could be used to prevent free-radical-induced deleterious effects.  相似文献

Of numerous species belonging to the medicinally important genus Hypericum, only H. perforatum L. and H. maculatum Crantz grow widely in Estonia. A comparative biochemical systematics study of hypericins, hyperforins and other phenolics within Hypericum spp. growing in Estonia was performed. For comprehensive metabolomic investigation, 42 samples of H. perforatum and 16 samples of aerial parts of H. maculatum were collected in two consecutive years from various locations; methanolic extracts were prepared from airdried leaves and flowers. The concentrations of a quinic acid derivative, caffeic acid glucoside, vanillic acid glucoside, neochlorogenic acid, chlorogenic acid, catechin, epicatechin, myricetin glucoside, hyperoside, isoquercitrin, rutin, quercetin pentoside, quercitrin, kaempferol glucoside, kaempferol rutinoside, quercetin, hyperforin, adhyperforin, protopseudohypericin, pseudohypericin, and hypericin were determined by LC-DAD-MS/MS. All the aforementioned compounds were detected in both species, although some at very different levels – H. maculatum contained rutin and hyperforins only in trace amounts and overall tended to contain more phenolic compounds. The level of total hypericins was the same for both species. These results constitute a further contribution to the systematic knowledge about the Hypericum spp. Results of principal component analysis (PCA) demonstrated distinct between-years and between-species diversity in the chemical composition of the plants studied. Between-years diversity in Hypericum spp. has not been addressed before.  相似文献

Stable free radical scavenging and antiperoxidative activities of resveratrol, a component of grapes and red wine, were evaluated and compared with some other known bioflavonoids (quercetin, catechin, kaempferol, myricetin, fisetin, ellagic acid and naringenin) widely present in the plant kingdom. Free radical scavenging activity was measured in an in vitro chemical system (DPPH assay), while for antiperoxidative activity, biological system comprising of hepatic and pulmonary homogenates was employed. Antiradical activity assay showed quercetin and myricetin to be stronger antiradical agents than resveratrol. Structure-activity study revealed that O-dihydroxy group on ring B of flavonoid plays a crucial role. A double bond at 2-3 position conjugated with a 4-oxo function and hydroxy groups at positions 3 and 5 also contribute towards antiradical activity of flavonoids. Resveratrol exhibited stronger antiradical activity than kaempferol and naringenin and was also more efficient than alpha-tocopherol, a known strong endogenous non-flavonoid antioxidant, used for comparison. In vitro antiperoxidative assay showed fisetin as the strongest and kaempferol as the weakest antioxidant. Resveratrol was found to be stronger antioxidant than catechin, myricetin, kaempferol and naringenin, but was weaker than quercetin, fisetin and alpha-tocopherol. Antiradical and antiperoxidative activities of resveratrol may explain its beneficial effects in disease states. Assays exhibited no direct correlation between antiradical and antiperoxidative activities of the phenolics.  相似文献

Two arsenic (As) hyperaccumulators (Pteris multifida and Pteris vittata) and a non-hyperaccumulator (Pteris semipinnata) were exposed to different As concentrations under hydroponic conditions. Five flavonoids in these fern species were determined by high-performance liquid chromatography (HPLC). Flavonoid production in P. multifida and P. semipinnata was also studied in 0 and 20 mg As L−1 treatments at different cultivation times. No significant differences were observed regarding the contents of quercetin, isoquercitrin and kaempferol in the fronds. The contents of rutin, quercetin, kaempferol and total flavonoids were also not significantly different in the roots of the three fern species under the same As treatment. However, significant differences were observed in contents of rutin, quercetin, hyperin, kaempferol and total flavonoids over time in the 20 mg As L−1 treatment. In general, the changes in flavonoid contents in the As hyperaccumulators were not directly related to As accumulation.  相似文献

Late leaf spot (LLS) caused by fungi Passalora personata is generally more destructive and difficult to control than early leaf spot. The aim of this study was to decipher biochemical defense mechanism in groundnut genotypes against P. personata by identifying resistance specific biomarkers and metabolic pathways induced during host–pathogen interaction. Metabolomics of non-infected and infected leaves of moderately resistant (GPBD4 and ICGV86590), resistant (KDG128 and RHRG06083) and susceptible (GG20, JL24 and TMV2) genotypes was carried out at 5 days after infection (65 days after sowing). Non-targeted metabolite analysis using GC–MS revealed total 77 metabolites including carbohydrates, sugar alcohols, amino acids, fatty acids, polyamines, phenolics, terpenes and sterols. Variable importance in projection (VIP) measure of partial least squares-discriminant analysis (PLS-DA) showed that resistant and moderately resistant genotypes possessed higher intensities of ribonic acid, cinnamic acid, malic acid, squalene, xylulose, galactose, fructose, glucose, β-amyrin and hydroquinone while susceptible genotypes had higher amount of gluconic acid 2-methoxime, ribo-hexose-3-ulose and gluconic acid. Heat map analysis showed that resistant genotypes had higher intensities of β-amyrin, hydroquinone in non-infected and malic acid, squalene, putrescine and 2,3,4-trihydroxybutyric acid in infected leaves. Dendrogram analysis further separated resistant genotypes in the same cluster along with infected moderately resistant genotypes. The most significant pathways identified are: linoleic acid metabolism, flavone and flavonol biosynthesis, cutin, suberin and wax biosynthesis, pentose and glucuronate interconversions, starch and sucrose metabolism, stilbenoid biosynthesis and ascorbate and aldarate metabolism. Targeted metabolite analysis further confirmed that resistant genotypes possessed higher content of primary metabolites sucrose, glucose, fructose, malic acid and citric acid. Moreover, resistant genotypes possessed higher content of salicylic, coumaric, ferulic, cinnamic, gallic acid (phenolic acids) and kaempferol, quercetin and catechin (flavonols). Thus metabolites having higher accumulation in resistant genotypes can be used as biomarkers for screening of LSS resistant germplasm. These results unravel that higher amount of primary metabolites leads to stimulate the accumulation of more amounts of secondary metabolites such as phenolic acid, flavanols, stilbenes and terpenoids (squalene and β-amyrin) biosynthesis which are ultimately involved in defense mechanism against LLS pathogen.Supplementary InformationThe online version contains supplementary material available at 10.1007/s12298-021-00985-5.  相似文献

Background

Excessive production of free radicals causes direct damage to biological molecules such as DNA, proteins, lipids, carbohydrates leading to tumor development and progression. Natural antioxidant molecules from phytochemicals of plant origin may directly inhibit either their production or limit their propagation or destroy them to protect the system. In the present study, Monodora myristica a non-timber forest product consumed in Cameroon as spice was screened for its free radical scavenging properties, antioxidant and enzymes protective activities. Its phenolic compound profile was also realized by HPLC.

Results

This study demonstrated that M. myristica has scavenging properties against DPPH, OH, NO, and ABTS radicals which vary in a dose depending manner. It also showed an antioxidant potential that was comparable with that of Butylated Hydroxytoluene (BHT) and vitamin C used as standard. The aqueous ethanol extract of M. myristica barks (AEH); showed a significantly higher content in polyphenolic compounds (21.44 ± 0.24 mg caffeic acid/g dried extract) and flavonoid (5.69 ± 0.07 quercetin equivalent mg/g of dried weight) as compared to the other studied extracts. The HPLC analysis of the barks and leaves revealed the presence of several polyphenols. The acids (3,4-OH-benzoic, caffeic, gallic, O- and P- coumaric, syringic, vanillic), alcohols (tyrosol and OH-tyrosol), theobromine, quercetin, rutin, catechine and apigenin were the identified and quantified polyphenols. All the tested extracts demonstrated a high protective potential on the superoxide dismutase (SOD), catalase and peroxidase activities.

Conclusion

Finally, the different extracts from M. myristica and specifically the aqueous ethanol extract reveal several properties such as higher free radical scavenging properties, significant antioxidant capacities and protective potential effects on liver enzymes.  相似文献

The European dewberry (Rubus caesius) is a perennial shrub that is widely distributed in Europe but can also be found in North America. In folk medicine, the European dewberry is used to treat hyperglycaemia, diarrhoea and inflammation. LC-MS analysis of the European dewberry confirmed the presence of 35 compounds, mostly flavonoids, phenolic acids and derivatives of ellagic acid. Phytochemical analysis of R. caesius leaves led to the isolation of nine phenolics, namely: quercetin 3-O-β-D-rutinoside (1), kaempferol 3-O-β-D-glucuronide (2), quercetin 3-O-β-D-glucuronide (3), methyl brevifolincarboxylate (4), kaempferol 3-O-β-D-(6″-O-(E)-p-coumaroyl)-glucoside (5), kaempferol (6), quercetin (7), pedunculagin (8), and ellagic acid (9). Compounds 18 were isolated from this species for the first time. The chemophenetic significance was discussed.  相似文献

The effect of various flavonoids, lectins and phenyl βD‐glucoside on larval survival, weights and the activities of digestive (total serine protease and trypsin) and detoxifying (esterase and glutathione‐S‐transferase) enzymes of Spodoptera litura larvae at 7 days after treatment was studied through diet incorporation assay. Flavonoids (rutin, chlorogenic acid, quinic acid, caffeic acid, naringenin, quercitin, kaempferol, myricetin, catechin, and ferulic acid) were incorporated in artificial diet at 100, 500 and 1000 ppm, lectins: groundnut leaf lectin (GLL), concavalin A (ConA) and phenyl βD‐glucoside at 1, 2 and 5 μg/mL. Flavonoids such as rutin, quercitin and kaempferol at 1000 ppm were more toxic to S. litura larvae than quinic acid, caffeic acid, naringenin, myricetin, catechin, and ferulic acid. Larval growth and development were significantly reduced in S. litura larvae fed on a diet with GLL and ConA at 5 μg/mL compared to the larvae fed at 2 and 1 μg/mL concentrations. The larvae fed on flavonoid‐treated diets showed significant reduction in serine protease, trypsin and esterase activities. The flavonoids such as rutin, chlorogenic acid, quinic acid, naringenin, quercitin, kaempferol and myricetin, and lectins, GLL and ConA can be utilized in insect control programs.  相似文献

Microorganisms and their hosts communicate through an array of signals. Many physiological processes regulated in quorum sensing (QS) are dependent on auto-inducers, like N-acyl-homoserine lactones (AHLs) as in numerous groups of both gram-positive and gram-negative bacteria. In vitro grown seven-day old chickpea seedlings treated with plant growth promoting bacteria (PGPRs) were used to screen the AHL mimicking and for phytochemical substances like phytohormones and secondary metabolites such as phenolics and flavonoids. Potential anti-quorum sensing (anti-QS) activity surrounding the roots on semi-solid agar lawn of Chromobacterium violaceum (ATCC12742) was observed. Crude protein (4.46–8.30 μg/mL) and methanolic extracts (100 μg/mL) of seedling gave moderate anti-QS activity against CV12742 anti QS bioassay, respectively. Crude protein and methanolic extract of Bacillus amyloliquefaciens (34.00 ± 2.23; 34.00 ± 4.33 mm) and B. subtilis A (27.00 ± 2.10; 3.29 ± 2.16 mm) treated samples showed higher zone of inhibition due to anti-QS activity. Phytohormone analysis using LC–MS for zeatin, auxin and methyl jasmonate (MeJA) indicated that phytohormones were significantly upregulated by 1909.80 ng/g FW, 669.67 ng/g FW and 244.55 ng/g FW, respectively in Pseudomonas brassicacearum treated seedlings compared to control. UHPLC of PGPR treated seedlings showed overly expressed gallic acid, protocatechuic acid, catechin, p-hydroxybenzoic acid, caffeic acid, catechol, vanillin, and ferulic acid in B. amyloliquefaciens treated seedlings compared to others. Enrichment analysis identified significant pathways related to metabolism, biosynthesis of secondary metabolites. Previous studies have demonstrated that certain flavonoids can have an inhibitory effect on angiotensin-converting enzyme (ACE) activity, which plays a key role in the regulation of arterial blood pressure. In the present study, 17 flavonoids belonging to five structural subtypes were evaluated in vitro for their ability to inhibit ACE in order to establish the structural basis of their bioactivity. The ACE inhibitory (ACEI) activity of these 17 flavonoids was determined by fluorimetric method at two concentrations (500 µM and 100 µM). Their inhibitory potencies ranged from 17 to 95% at 500 µM and from 0 to 57% at 100 µM. In both cases, the highest ACEI activity was obtained for luteolin. Following the determination of ACEI activity, the flavonoids with higher ACEI activity (i.e., ACEI >60% at 500 µM) were selected for further IC50 determination. The IC50 values for luteolin, quercetin, rutin, kaempferol, rhoifolin and apigenin K were 23, 43, 64, 178, 183 and 196 µM, respectively. Our results suggest that flavonoids are an excellent source of functional antihypertensive products. Furthermore, our structure-activity relationship studies show that the combination of sub-structures on the flavonoid skeleton that increase ACEI activity is made up of the following elements: (a) the catechol group in the B-ring, (b) the double bond between C2 and C3 at the C-ring, and (c) the cetone group in C4 at the C-ring. Protein-ligand docking studies are used to understand the molecular basis for these results.  相似文献

An anaerobic, quercetin-degrading bacterium was isolated from human feces and identified as Clostridium orbiscindens by comparative 16S rRNA gene sequence analysis. The organism was tested for its ability to transform several flavonoids. The isolated C. orbiscindens strain converted quercetin and taxifolin to 3,4-dihydroxyphenylacetic acid; luteolin and eriodictyol to 3-(3,4-dihydroxyphenyl)propionic acid; and apigenin, naringenin, and phloretin to 3-(4-hydroxyphenyl)propionic acid, respectively. Genistein and daidzein were not utilized. The glycosidic bonds of luteolin-3-glucoside, luteolin-5-glucoside, naringenin-7-neohesperidoside (naringin), quercetin-3-glucoside, quercetin-3-rutinoside (rutin), and phloretin-2′-glucoside were not cleaved. Based on the intermediates and products detected, pathways for the degradation of the flavonol quercetin and the flavones apigenin and luteolin are proposed. To investigate the numerical importance of C. orbiscindens in the human intestinal tract, a species-specific oligonucleotide probe was designed and tested for its specificity. Application of the probe to fecal samples from 10 human subjects proved the presence of C. orbiscindens in 8 out of the 10 samples tested. The numbers ranged from 1.87 × 108 to 2.50 × 109 cells g of fecal dry mass−1, corresponding to a mean count of 4.40 × 108 cells g of dry feces−1.  相似文献

Among the structurally related flavonoids tested on the bovine kidney low molecular weight protein tyrosine phosphatase (LMrPTP) activity, quercetin activated by about 2.6-fold the p-nitrophenyl-phosphate (p-NPP)-directed reaction, in contrast to morin that acted as a competitive inhibitor, with Ki values of 87, 73 and 50 μM for p-NPP, FMN, and tyrosine-phosphate, respectively. Other related flavonoids, such as rutin, kaempferol, catechin, narigin, phloretin and taxifolin did not significantly affect the LMrPTP activity.

The positions of the hydroxyl groups in the structures of the flavonoids were important for their distinct effects on LMrPTP activity. The hydroxyl groups at C3′ and C4′ and the presence of a double bond at C2 and C3 were essential for the activating effect of quercetin. The absence of the 3′-OH (kaempferol), absence of the double bond (taxifolin) and the presence of the sugar rutinose at the 3-OH (rutin) suppressed the effect of quercetin. The C2′- and C4′-hydroxyl groups, the presence of the double bond, and a C4-ketone group were important requirements for the inhibitory effects of morin.  相似文献

Widely distributed in plants, flavonoids reduce the incidence of cancer and cardiovascular disease. In this study, flavonoid content and composition in members of the Prunus genus were evaluated using liquid chromatography with diode array and electrospray ionization mass spectrometric detection (UPLC-DAD-ESI/QTOF-MS). Flavonoids in plants of the Prunus genus include the basic structures of kaempferol, quercetin, and catechin, and exist as mono-, di-, or tri-glycoside compounds mono-acylated with acetic acid. A total of 23 individual flavonoids were isolated and confirmed, three of which appear to be newly identified compounds: quercetin 3-O-(2″-O-acetyl)neohesperidoside, quercetin 3-O-(4″-O-acetyl)rutinoside, and kaempferol 3-O-(4″-O-acetyl)rutinoside. Japanese apricot and Chinese plum contained the highest amounts of flavonoids in the Prunus genus. During the ripening stage of Japanese apricot, the total flavonol content was reduced, while the catechin content was increased.  相似文献

This study confirmed the ameliorating effect of immature persimmon (Diospyros kaki) ethanolic extract (IPEE) on neuronal cytotoxicity in amyloid beta (Aβ)1–42-induced ICR mice. The administration of IPEE ameliorated the cognitive dysfunction in Aβ1–42-induced mice by improving the spatial working memory, the short-term and long-term memory functions. IPEE protected the cerebral cholinergic system, such as the acetylcholine (ACh) level and acetylcholinesterase (AChE) activity, and antioxidant system, such as the superoxide dismutase (SOD), reduced glutathione (GSH) and malondialdehyde (MDA) contents. In addition, mitochondrial dysfunction against Aβ1–42-induced toxicity was reduced by regulating the reactive oxygen species (ROS), mitochondrial membrane potential and ATP contents. In addition, IPEE regulated the expression levels of tau signaling, such as TNF-α, p-JNK, p-Akt, p-GSK3β, p-tau, p-NF-κB, BAX and caspase 3. Finally, gallic acid, ellagic acid and quercetin 3-O-(6″-acetyl-glucoside) were identified as the physiological compounds of IPEE using ultra-performance liquid chromatography ion mobility separation quadrupole time-of-flight/tandem mass spectrometry (UPLC IMS Q-TOF/MS2).  相似文献
