Summary 1. Iron which is shown to be present as a contaminant on leaf surfaces of tomato can be removed completely by washing the leaves for 30 sec in 0.3% teepol and water followed by a 30 sec rinsing in distilled water.2. By using radioactive iron it is shown that there was no loss of the metal from within the plants during leaf washing. Agreement between chemical and radioassay data for iron showed that the metal was completely removed by the wash procedure.3. In tomato grown in the glasshouse consistent differences between the iron contents of normal and chlorotic leaves were obtained after leaf washing. When the plants were grown in a dustproof chamber differences in iron status of green and chlorotic leaves were obtained without washing.4. Added radioactive and carrier iron was fully recovered from acid digests of a variety of plant material, specially chosen to represent a range of silica contents.  相似文献

Summary 32P-labelled monocalcium phosphate solution was supplied to the root system of individual wheat plants within a field crop two weeks after emergence. Three levels of carrier P, equivalent to 2.5, 5, and 10 kgP ha–1 were used. The distribution of32P between shoots and the soil inorganic and organic P fractions was measured after a further six weeks growth.There was no evidence for the incorporation of32P-orthophosphate into soil organic P fractions in the wheat rhizosphere in the period between germination and mid-tillering.The suitability of the single plant technique to measure plant available P in field soils was assessed by calculating A values from plants grown in soil witth different fertilizer P histories. There was a significant linear relationship between A values and the amount of soil P extractable with 0.5M NaHCO3 from the different fertilizer treatments. Although the technique is unlikely to given an absolute value for plant available nutrient, it can provide quantitative data for comparative trials with different forms of fertilizer, methods of fertilizer application or amounts of available soil water. The values obtained should be termed comparative A values or Ac values.  相似文献

Effects of soil compaction on the microbial populations of melon and maize rhizoplane were investigated in quantity and quality. The numbers of culturable bacteria and fluorescent pseudomonads on the rhizoplane were higher when plants were grown in more compacted soil and the relative increase was larger in fluorescent pseudomonads. Total bacterial counts, however, did not appear to be affected by soil compaction, resulting in the increase in the culturable bacteria among total counts in more compacted soil. The determination of extracellular enzymatic properties (pectinase, -glucosidase, -glucosidase and -galactosidase) of each 100 isolates from bulk soil and root samples suggested that the microbial populations on the rhizoplane, especially when plants were grown in highly, compacted soil, were composed of high ratios of bacteria with abilities to utilize root exudates efficiently. The microbial community structure estimated from the colony forming curves of bulk soil and root samples suggested that the microbial populations on the rhizoplane, especially when plants were grown in compacted soil, were likely to be composed of more r-strategists which were defined as those who formed colonies within 2 days.  相似文献

Summary When soils are suspended in solutions of labelled calcium chloride isotopic exchange with the labile soil calcium occurs rapidly. This may be followed by a slow secondary exchange reaction, but its magnitude is not great and equilibrium is nearly, if not completely, attained within 7 days.When, however, plants are grown in soil throughout which carrier-free calcium-45 has been thoroughly mixed, it is found that the calcium-45 absorbed by the plants has equilibrated with a quantity of soil calcium larger than that which undergoes isotopic exchange when soils are suspended in solutions of labelled calcium chloride. The analysis of plants grown for varying periods shows that equilibration can continue for several weeks, and that the quantity of soil calcium with which the calcium-45 is associated can be increased both by the addition of electrolytes to the soil and by growing plants under exhaustion conditions. In 5 soils the extra calcium which equilibrated with calcium-45 in this way never exceeded 3.5 per cent of the total soil calcium, and was usually considerably lower.The continued equilibration of calcium-45 with soil calcium causes the specific activity (Ca45/stable Ca) of the calcium entering plants to decrease. Because the calcium in plant roots has, on average, been absorbed more recently than that in shoots, the latter show higher specific activities.The causes of these effects are discussed and consideration is given to their significance in the interpretation of results of experiments involving the use of calcium-45 as a tracer in soils.  相似文献

Weed assemblages from late medieval cornfields have been studied for the first time in northern Switzerland. Eleven samples from at least two different grain stores were investigated. The samples were collected from the carbonised remains of six wooden houses built in the late 13th century A.D. and which burnt down in the middle of the 15th century. The weed floras found in the spelt (Triticum spelta) and oats (Avena sativa) indicate a high botanical diversity in the cornfields at harvest time. Although oats are normally a summer crop and spelt a winter crop, both summer and winter crop weeds (as well as many different present-day grassland taxa) were found in each type of grain. Most of the weeds found were perennial plants, which was interpreted as an indication of both extensive tillage of the arable land and application of the three-field rotation system (Dreifelderwirtschaft). The spectra of the two palaeophytocoenoses (assemblages of ancient plant remains) studied suggest that the phytosociological method may not be reliable for classification of the late medieval remains into summer and winter crop weed communities. These findings should provide a better understanding of the development of anthropogenic plant communities, and in particular, the development of crop weed communities.  相似文献

In 1986 and 1987 surveys were conducted of 34 (1986) and 35 (1987) peanut (Arachis hypogaea L.) fields in which the plants showed various degrees of chlorosis. In the areas concerned, plant appearance was classified according to a chlorotic index and corresponding soil samples were taken and analysed for CaCO3, pH, NO3–N and DTPA-extractable Fe in 1986 and for CaCO3, NO3–N and active lime in 1987.Regression analyses showed that CaCO3, active lime and NO3–N were positively correlated, while DTPA-extractable Fe was negatively correlated, with the chlorosis problem. The critical levels above which plants were chlorotic were 20 to 25% CaCO3 and 10% active lime. Plants began to be chlorotic when DTPA-extractable Fe was below 2.5 mg·kg–1. The soil factors examined explained about 60% of the variability in plant chlorosis.  相似文献

To study the influence of soil moisture on phosphorus (P) depletion in the rhizosphere, maize (Zea mays cv. Trak) was pre-grown in vermiculite filled-PVC tubes for 9 days and then the plants with the tubes were transplanted into soil columns maintained at two soil moisture levels () of 0.14 and 0.20 cm3 cm–3 for 10 days. The soil columns were separated at 1 cm depth by a nylon screen of 53 m inner mesh size, into 1 cm soil layer above and 3 cm soil column below screen. A root mat developed over the screen, but root hairs only could penetrate it. Regardless of the soil moisture level in the columns, and adequate and equal water and nutrients supply was maintained via wicks from an external nutrient solution to the plant roots in vermiculite. After 10 days, the soil columns were separated from the root mats, quickly frozen in liquid nitrogen and sliced into thin layers (0.2mm) using a refrigerated microtome to give soil samples at defined distances from the root mats for analyses. Lower soil moisture (=0.14) resulted in narrower and steeper depletion profile of 0.5 M NaHCO3 extractable P (NaHCO3-Pi) as compared to higher soil moisture (=0.20). Depletion of P in soil solution in the immediate vicinity of root mats did not differ much but the extension of the depletion zones was 0.10 cm at =0.14 and 0.20 cm at =0.20. The depletion up to 0.05cm with =0.14 and up to 0.07 cm with =0.20 was uniform, and may be attributed to the depletion in the root hair zone. Beyond the root hair zones, the theory of diffusion and mass flow was able to explain the observed differences in shape and extent of the P depletion profiles at the two soil moisture levels.  相似文献

Several lines of evidence suggest that nitrogen in most tropical forests is relatively more available than N in most temperate forests, and even that it may function as an excess nutrient in many tropical forests. If this is correct, tropical forests should have more open N cycles than temperate forests, with both inputs and outputs of N large relative to N cycling within systems. Consequent differences in both the magnitude and the pathways of N loss imply that tropical forests should in general be more15N enriched than are most temperate forests. In order to test this hypothesis, we compared the nitrogen stable isotopic composition of tree leaves and soils from a variety of tropical and temperate forests. Foliar 15N values from tropical forests averaged 6.5 higher than from temperate forests. Within the tropics, ecosystems with relatively low N availability (montane forests, forests on sandy soils) were significantly more depleted in15N than other tropical forests. The average 15N values for tropical forest soils, either for surface or for depth samples, were almost 8 higher than temperate forest soils. These results provide another line of evidence that N is relatively abundant in many tropical forest ecosystems.  相似文献

Gibberellic acid (29 or 290 M) injected into drip irrigation lines significantly stimulated internode elongation of dwarf peas, and the 290-M soil treatment produced significantly taller plants than did the 29-M treatment. GA3 uptake may limit GA-induced internode elongation when GA3 is applied to soil, in contrast to results obtained for hydroponically grown plants, where uptake initially appeared to exceed the rate of hormone metabolism (andersonet al.). It is likely that biodegradation or chemical inactivation limited the plant-availability of GA3 in the soil. Degradation of moderate GA3 concentrations in a moist, aerobic loamy fine sand was nearly complete within five days, indicating that the inefficiency of soil applications may outweight the benefits provided by reducing labor costs associated with foliar-spray applications.  相似文献

Summary A comparison was made of several laboratory methods for estimating the yield of phosphorus in plants grown in greenhouse cultures on samples of 22 soils from different parts of the United States. Summary The gene for -CGTase from an alkalophilic bacterium, Bacillus sp. #1011, was cloned in an Escherichia coli phage D69 and recloned in an E. coli plasmid pBR322 and a B. subtilis plasmid pUB110. An E. coli recombinant plasmid pTUE202 and a B. subtilis plasmid pTUB703 were selected from ten plasmids, because the transformants by each of the two plasmids produced the highest amount of extracellular -CGTase in each strain. The plasmids were stably maintained and expressed in each bacterial strain. A common DNA region of approximately 2.5 kb was defined in the ten plasmids, and the enzymatic activity was lost when a part of the common region was deleted. Summary I investigated the extent and adaptive importance of genetically-based variation in plant water relations in two populations of the annual plant Cleome serrulata found growing along relatively short (<30 m) and mild soil moisture gradients. Field measurements of predawn plant water potentials showed that plants at the dry end of the moisture gradients had consistently lower Plant in May and June of 1984; differences up to 0.9 MPa were seen along the gradients. Seeds were collected from maternal plants growing along the moisture gradients and then grown under well-watered conditions in the greenhouse. Pressure-volume curves were constructed for a total of 92 seedlings from 25 maternal plants when the seedlings were four weeks old.Considerable genetic variation in the four highly correlated water potential components was seen in both populations, suggesting relatively high heritabilities (h20.5). A partial correlation analysis revealed that cell wall elasticity was higher in seedlings from maternal plants which grew in the dry portions of each site. This suggested that natural selection had acted on this character during one or more previous generations. It appears that slight variations in the physiological genotype can significantly affect overall fitness in C. serulata.  相似文献

Nitrogen (N) resorption from senescing tissues enables plants to conserve and reuse this important nutrient. As such, it is expected that plant species adapted to infertile soils could have a higher N-resorption efficiency (percentage reduction of nitrogen between green and senescing tissues) and/or higher N-resorption proficiency (absolute reduction of nitrogen in senescing tissues) than those adapted to fertile soils. To test this hypothesis, we investigated the relationships among soil characteristics (total N, nitrate-N, ammonium-N, pH and moisture) and N resorption in Stipa kryloviiRoshev., a species occurred widely in natural grasslands of northern China. N contents in green and senescing tissues were 6.7±0.1 and 3.3±0.1 mg g–1, respectively. The mean value of N-resorption efficiency was found to be 72.1%. The N-resorption efficiency in S. kryloviiwas independent of soil characteristics. The N-resorption proficiency in S. kryloviiwas dependent on soil nitrate- and ammonium-N, but it was relatively independent of soil total N. The N-resorption proficiency was negatively correlated with soil pH and moisture. There was a positive correlation between N concentration in green tissues and resorption efficiency. However, N-resorption efficiency was not correlated significantly with N concentration in senescing tissues. These results indicate that the intraspecific variation in N resorption of Stipa kryloviiRoshev. is associated with soil regimes and that higher N resorption on N-poor soils is an adaptive strategy for S. kryloviito maximize N use under conditions of limited N supply.  相似文献

Aims

A commonly accepted challenge when visualising plant roots in X-ray micro Computed Tomography (μCT) images is the similar X-ray attenuation of plant roots and soil phases. Soil moisture content remains a recognised, yet currently uncharacterised source of segmentation error. This work sought to quantify the effect of soil moisture content on the ability to segment roots from soil in μCT images.

Methods

Rice (Oryza sativa) plants grown in contrasting soils (loamy sand and clay loam) were μCT scanned daily for nine days whilst drying from saturation. Root volumes were segmented from μCT images and compared with volumes derived by root washing.

Results

At saturation the overlapping attenuation values of root material, water-filled soil pores and soil organic matter significantly hindered segmentation. However, in dry soil (ca. six days of drying post-saturation) the air-filled pores increased image noise adjacent to roots and impeded accurate visualisation of root material. The root volume was most accurately segmented at field capacity.

Conclusions

Root volumes can be accurately segmented from μCT images of undisturbed soil without compromising the growth requirements of the plant providing soil moisture content is kept at field capacity. We propose all future studies in this area should consider the error associated with scanning at different soil moisture contents.  相似文献

Summary A simple formula is derived to calculate the nucleocytoplasmic RNA-Efflux per nuclear Pore complex per min (REP-rate) which is generally applicable both for growing and stationary eukaryotic cells. In actively growing cells this REP-rate is mainly dependent on the cytoplasmic RNA-pool, the number of RNA-transporting pores, and the growth constant of RNA. These parameters are determined in logarithmicTetrahymena pyriformis GL. In this organism, 45 molecules both of the larger ribosomal RNA (25s rRNA) and of the smaller (17s rRNA) are transported per pore per min from nucleus to cytoplasm. Pulse-label experiments with3H-uridine indicate that the 25s rRNA is obviously transferred more slowly to the cytoplasm than the 17s rRNA. We postulate a gating hypothesis on the regulation of the nucleocytoplasmic RNA-efflux by nuclear pore complexes. This gating hypothesis suggests that nucleopores are controlling points of secondary importance in the sequence of gene expression, and do not directly control the cytoplasmic protein synthesis in eukaryotic cells.  相似文献

The relation between decomposition rates and soil salinity and moisture conditions in tidal marshes of the Westerschelde estuary was investigated. In the first part of the study, these soil factors were experimentally manipulated in field plots which were either screened from rainwater or which received an additional weekly supply of freshwater from April to September 1989. These treatments had no clear effect on soil salinities and moisture conditions in a low marsh site. Decomposition rates of Spartina anglica leaves (kept in litterbags in the plots) also did not differ between treatments. In screened plots of a middle marsh site, decomposition rate of Elymus pycnanthus leaves decreased significantly. The effect of the experimental treatments on soil moisture content was variable, but comparatively high soil salinity values (up to 61.3) were consistently found in these plots. It is suggested that the elevated salinity levels induced the decrease in decomposition rate.In the second part of the study, cellulolytic decomposition, measured by loss of tensile strength of strips of cotton test cloth, was investigated in relation to a non-manipulated range of soil salinities (3.8–24.2), by exposing the strips in a series of tidal marshes along the salt gradient of the Westerschelde estuary. No correlation between decomposition rate and soil salinity was found. In addition, no relation was found between decomposition rate and soil water content. The results of both parts of this study lead us to the hypothesis that rate limitation of decomposition in estuarine tidal marsh soils is found at high soil salinities only.  相似文献

The triazine dyes: Cibacron Blue 3GA, Reactive Red 120, Reactive Yellow 86, Reactive Green 19, Reactive Blue 4, Reactive Brown 10 inhibited the activity of a purified preparation of 1,6fucosyltransferase (GDP-L-fucose:N-acetyl -glucosaminide 6--L-fucosyltransferase, EC 2.4.1.68) from human blood platelets. Cibacron Blue 3GA and Reactive Red 120 were examined for the nature of the inhibition and both were found to be competitive inhibitors of the enzyme, with Ki=11[emsp4 ]M and 2[emsp4 ]M, respectively. The two dyes inhibited also serum glycosyltransferases: 1,2fucosyltransferase (GDP-L-fucose: -D-galactosyl-R2--L-fucosyltransferase, EC 2.4.1.69), 1,4galactosyltransferase (UDP-galactose: N-acetyl-D-glucosamine 4--D-galactosyltransferase, EC 2.4.1.90) and 1,3N-acetylglucosaminyltransferase (UDP-GlcNAc: 4--D-galactosyl-D-glucose). Cibacron Blue 3GA was a more effective inhibitor of the glycosyltransferases that use UDP-linked sugar donors than Reactive Red 120 while the latter was a stronger inhibitor of the fucosyltransferases that use GDP-linked donor. All four glycosyltransferases could be affinity purified on Cibacron Blue 3GA-Agarose columns. The order of elution of glycosyltransferases from the columns with solutions of 0.25–1.0[emsp4 ]M potassium iodide also depended upon the structure of nucleotide sugar donor, i.e. whether it contained UDP or GDP. Thus, triazine dyes should interact with the sugar donor binding sites of glycosyltransferases. The main advantages of the use of triazine dyes as affinity ligands for isolation of glycosyltransferases are their universal applicability regardless of enzyme specificity, low cost, and insensitivity to high concentration of other proteins present in the solution.  相似文献

Roots of a wide range of plant species exude carboxylates, e.g. citrate, into the rhizosphere, to mobilise sparingly available phosphate. We investigated the carboxylates in root exudates of Banksia grandisWilld. (Proteaceae), which occurs on severely phosphate-impoverished soils in Western Australia. Plants were grown in pots with a nutrient-poor quartz sand, with phosphate, at 25 g P g–1, added as either K-phosphate, glycerol phosphate, Fe-phosphate or Al-phosphate.Plants grown on Fe-phosphate or Al-phosphate formed `proteoid' or `cluster' roots, and exuded significant amounts of carboxylates. Plants grown on K-phosphate did not form cluster roots; their leaves were chlorotic, and some of these plants died during the experiment. Plants grown on glycerol phosphate did have cluster roots, but their leaves also became chlorotic, albeit later in the experiment.Tri- and dicarboxylates (citrate, 60%; malate, 25%; trans-aconitate, 14%) were the major carboxylates in root exudates when P was supplied as Al-phosphate. The same tri- and dicarboxylates were also exuded when P was supplied as Fe-phosphate (31, 14 and 12%, respectively). In addition, these plants exuded monocarboxylates (lactate, 30%; acetate, 12%). We analysed the effect of the different carboxylates on the mobilisation of phosphate and Fe in two different types of soils. The ecological significance of the difference in exudate spectrum for the mobilisation of nutrients and for the detoxification of aluminium is discussed.Because the leaves of plants grown with K-phosphate or glycerol-phosphate appeared chlorotic, we analysed the concentrations of P, Fe, Zn, Mn and Cu in these leaves. Only the concentration of total P was considerably higher in leaves of plants grown with K- or glycerol-phosphate than that in leaves of plants grown with Fe- or Al-phosphate. Both the concentration of total Fe and that of reduced Fe was the same in chlorotic leaves as that in leaves of plants grown with Fe- or Al-phosphate, which had a healthy appearance. It is concluded that P-induced chlorosis was not due to a lack of total or reduced Fe; it may have been due to precipitation of Fe by phosphate.  相似文献

Commercial narrow-leafed lupins (Lupinus angustifolius L.) grown on calcareous soils commonly display chlorotic symptoms resembling Fe deficiency. The severity of chlorosis increases with concurrent increases in soil moisture content. Our research has indicated that the rough-seeded lupin species, Lupinus pilosus Murr., has a range of adaptation to calcareous soils, from tolerant to intolerant. A pot experiment was conducted comparing a tolerant, a moderately tolerant and a moderately intolerant genotype of L. pilosus. Plants were grown for 35 days in a calcareous soil (50% CaCO3) at three moisture contents (80%, 100% and 120% of field capacity); the growth was compared with that on a fertile black cracking clay control soil at 70% of field capacity. Visual chlorosis score, chlorophyll meter readings, number of leaves and shoot dry weights were recorded at 14, 21, 28 and 35 days after sowing. Concentrations of chlorophyll, active Fe and nutrients in the youngest fully expanded leaves were also measured. Results showed that increased soil moisture increased the severity of chlorotic symptoms (increased chlorosis score) in all genotypes. The tolerant genotype showed significantly less symptoms than other genotypes at all moisture contents. All genotypes were able to recover from chlorosis symptoms at 80% moisture in the calcareous soil. Chlorosis score negatively correlated with chlorophyll meter readings, chlorophyll concentration and foliar active and total Fe, and Mn concentrations. Visual chlorosis score appeared to be a cost effective, accurate and efficient method enabling classification of the tolerance of genotypes. The chlorotic symptoms were likely to be due to HCO3 - induced nutrient deficiencies or a direct effect of HCO3 - on chlorophyll synthesis. This study indicates that the most probable mechanism of tolerance is related to an ability to prevent uptake of HCO3 - or efficiently sequester it once inside the root which prevents increases in internal pH and transport to the shoots.  相似文献

The biological function of thermostable P450 monooxygenase CYP175A1 from Thermus thermophilus HB27 was studied by functional complementation in Escherichia coli. The gene product of CYP175A1 added hydroxyl groups to both rings of -carotene to form zeaxanthin (,-carotene-3,3-diol) in E. coli, which produces -carotene due to the Erwinia uredovora carotenoid biosynthesis genes. In addition, spectroscopic methods revealed that E. coli carrying CYP175A1 and the cDNA of the Haematococcus pluvialis carotene ketolase was able to synthesise hydroxyechinenone. The predicted amino acid sequence of the enzyme from T. thermophilus does not show substantial similarity with other known -carotene hydroxylases, but 41% with the cytochrome P450 monooxygenase from Bacillus megaterium (CYP102A1, P450 BM3). It is concluded that CYP175 A1 represents a new type of -carotene hydroxylase of the P450 superfamily.  相似文献
