A mycological survey was carried out for the first time, on red flint corn samples from the northwestern Andinian region of Argentina in the 1999 and 2000 harvest seasons. Species of the genus Fusarium were the most prevalent component of the flint corn mycoflora present in all provinces.F. verticillioides was the predominant Fusarium isolated in the 1999 harvest season in the the region, and was found at higher incidence level than those observed on commercial semident corn hybrids harvested in the main corn production area in Argentina (Pampean region). During the 2000 harvest season, Fusarium graminearum was most commonly isolated species in Salta province.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

Argentina is the first popcorn exporting country worldwide. In 1997-1998 harvest season, 40,000 ha were sown with a production of 125,000 tons; 120,000 tons of this production were exported to more than 40 countries. The objectives of this study were to isolate and to identify the fungi present in this cereal and to assess the occurrence of mycotoxins in freshly harvested popcorn in Buenos Aires province, Argentina, in 1999. All popcorn samples showed fungal contamination. A total of 4,211 isolates were recovered from popcorn kernel samples. The prevalent species isolated wasFusarium verticillioides followed byPenicillium funiculosum, F. graminearum andAlternaria alternata. No aflatoxins, zearalenone, deoxynivalenol or ochratoxin A were detected in the 42 popcorn simples. All samples but one were contaminated with fumonisins (FB) in a range in ug/kg of (not detected-529) for FB1; (not detected-216) for FB2 and (not detected-103) for FB3. Fumonisin contamination levels in Argentinean popcorn were lower than observed in flint corn. No significant differences in fungal and fumonisins contamination levels were observed in the different tested hybrids.  相似文献

Sorghum (Sorghum bicolor (L.) Moench) is an important cereal produced in and exported from Argentina. The risk of contamination by mycotoxins is related to the mycoflora associated with the sorghum grain. This paper reports on the identification of internal mycoflora of sorghum grain harvested in Argentina in 1991, 1992 and 1993, years with different total rainfall levels. A mycological survey was carried out on sorghum samples, from a location at the humid Argentinian pampa, using a Fusarium/dematiaceous fungi selective medium. The relative density of the prevalent fungal genera were statistically compared. Genus Fusarium was the most prevalent component of the internal seedborne mycoflora in the three harvest seasons. Genera Alternaria, Phoma, Penicillium and Aspergillus were also isolated. The predominant Fusarium was F. moniliforme and the most frequently isolated species of Alternaria, Phoma, Penicillium and Aspergillus were Alt. alternata, Pho. sorghima, Pen. funiculosum and Asp. flavus, respectively. This is the first report of the isolation of Fusarium napiforme in sorghum grain in Argentina. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

Although the corn leafhopper Dalbulus maidis (DeLong and Wolcott) is the most important vector of maize pathogens in Latin America, little is known about how and where it overwinters (passes the dry season), particularly in Mexico. The objectives of this study were (1) to monitor the abundance of D. maidis adults throughout the dry season in maize and maize-free habitats and (2) to determine where and how D. maidis adults, exposed or nonexposed to the maize pathogen Spiroplasma kunkelii Whitcomb, overwinter in a maize-free habitat. Work for the first objective was done during the two consecutive dry seasons of 1999-2000 and 2000-2001; the second objective was done during the dry seasons of 2003-2004 and 2005-2006. During the dry winter seasons, D. maidis was prevalent as long as maize was present in irrigated areas. The leafhopper was found in 52 of the 58 irrigated maize fields sampled in Mexico at the end of the dry seasons of 1999-2000 and 2000-2001. However, leafhopper adults were not found in nonirrigated maize-free habitats at high elevation during the dry winter season (February, March, and April), although leafhopper adults were prevalent on perennial wild grasses in January after maize harvest. Additional experiments revealed, however, that corn leafhopper adults, although few in number, survived the entire dry season in these nonirrigated maize-free fields. Also, no detectable difference in survival existed between leafhoppers exposed and those not exposed to S. kunkelli during the two dry seasons in the maize-free habitat.  相似文献

A mycological survey was carried out, for the first time, on black bean samples from the northwestern Argentinean province of Salta in the 1999 harvest season. Ten varieties of black beans were evaluated at three locations. Species of the genus Alternaria were the most prevalent component of the black bean mycoflora. Species of Fusarium, Sclerotinia, Rhizoctonia and Acremonium were also recorded. The predominant species of the genera isolated were Alternaria alternata, Sclerotinia sclerotiorum, Rhizoctonia solani, Fusarium semitectum and Acremonium strictum. An analysis of variance was applied to determine possible differences between black bean varieties. Variety FT88/519 was the most susceptible to Sclerotinia sclerotinia infection, while the variety DOR 604 was the least susceptible. As toxigenic species were recovered, Alternaria toxins, zearalenone and trichothecenes may pose a contamination risk for black bean.  相似文献

The relative density and isolation frequency of the prevalent fungi were statistically compared between locations and harvest seasons. The genusAlternaria was the most prevalent component of the internal seedborne mycoflora in the two harvest seasons. Fungi belonging to the generaPhoma, Fusarium, Microdochium, Penicillium andAspergillus were also isolated. In 1998–2000 a monitoring of the spectrum of Fusarium species on winter wheat was carried out in the Rhineland. The epidemic spread ofFusarium spp. on wheat plants during growing season was investigated as well as the grain contamination after harvest.F avenaceum was the Fusarium species isolated most frequently followed byF culmorum, F poae andF graminearum. Microdochium nivale occurred considerably only in 1998. Both, susceptibility and plant height of the cultivars were correlated to the incidence of Fusarium species /M nivale on harvested kernels; interactions with cropping intensities were detected. The incidence ofF poae seemed to be independent of the cultivar-specific Fusarium susceptibility. Despite the lack of disease symptoms, between growth stages 45–85 mycelium ofFusarium spp. was detectable in the leaves as well as conidia on the leaf surfaces.  相似文献

The effects of no-tillage (NT), conventional tillage (CT), and crop rotation on soybean yield and population dynamics of Heterodera glycines were compared during a 7-year study in a silty clay loam soil with 6% organic matter. Either H. glycines-resistant ''Linford'' soybean or susceptible ''Williams 82'' soybean was rotated with corn and grown on 76-cm-wide rows in both tillage systems. Soybean was planted in 1994, 1996, 1998, 1999, and 2000. Yield of Linford was significantly greater than Williams 82 in all years. Soybean yield was affected by tillage in 1999 and 2000. No-tillage production tended to support more reproduction (R = number of eggs at harvest/number of eggs at planting) on both cultivars. The largest R for Williams 82 were in 1998: 58.35 for NT plots and 11.78 for CT plots. For Linford, the largest R were 12.09 for NT plots in 1996, and 3.71 for CT in 1999. When corn was planted, R decreased more in NT. When soybean was planted in years subsequent to 1994, numbers of eggs at harvest (Pf) were greater for Williams 82 NT than for Williams 82 CT or Linford in both tillage systems; however, crop rotation with corn negated these population increases. The soil became suppressive to H. glycines in 1999 and was suppressive in 2000. After the 3 years of continuous soybean, Pf per 250 cm[sup3] soil were 2,870 for Williams 82 NT, 791 for Williams 82 CT, 544 for Linford NT, and 990 for Linford CT in 2000, compared with Pf of 13,100 for Williams 82 NT, 15,000 for Williams CT, 2,360 for Linford NT, and 2,050 for Linford CT in 1994. Describing population dynamics solely on the basis of R was not adequate, but also required independent examination of initial populations following overwintering and Pf after the growing season. Planting soybean either NT or CT in rotation with corn did not result in long-term increases in numbers of H. glycines eggs.  相似文献

The mean numbers of colony-forming units of molds present on vegetables at the time of harvest were in the range of 4.2 X 10(3) to 6.7 X 10(3)/g for all vegetables except cucurbits and asparagus. The numbers were not influenced by cultivars within species, duration of the growing season, distant separation of the growing fields, or elevation above ground. Numbers increased with rainfall during either of 3 days before harvest and decreased when the mean daily temperature exceeded 24 degrees C. The most frequently isolated fungi were Aureobasidium pullulans, Fusarium species, Alternaria tenuis, Epicoccum nigrum, Mucor species, Chaetomium fimeti, Rhizopus nigricans, and Phoma species, in decreasing order. All other molds comprised less than 2% of the isolates. Aspergilli and penicillia occurred sporadically and in low numbers. Fusarium species were dominant on cucurbits.  相似文献

Fusarium species are worldwide causal agents of ear rot in cereals. Their toxigenic potential is a health risk for both humans and animals. In Argentina, most identification of these fungi has been based on morphological and cross-fertility criteria which are time consuming and require considerable expertise in Fusarium taxonomy and physiology. DNA based approaches have been reported as rapid, sensitive and specific alternatives to identify the main fumonisin and trichothecene-producing Fusarium species. In this work, we used PCR assays and the partial sequence of TEF1-alpha gene (Translation Elongation Factor-1 alpha) to identify the fumonisin and trichothecene-producing species in Fusarium isolates from diverse regions of Argentina. The relative efficiency and reliability of those methods to improve mycotoxin risk prediction in this country were also assessed. Species-specific PCR assays were targeted toward multicopy IGS (Intergenic Spacer of rDNA units) and on the toxin biosynthetic genes FUM1 (fumonisins) and TRI13 and TRI7 genes (trichothecenes). PCR assays based on FUM1 gene and IGS sequences allowed detection and discrimination of the fumonisin producers Fusarium proliferatum and Fusarium verticillioides. Molecular identification of nonfumonisin producers from Gibberella fujikuroi species complex was possible after determination of TEF1-alplha gene sequences, which indicated the presence of Fusarium subglutinans, Fusarium andiyazi and Fusarium thapsinum. TEF-1 alpha gene sequences also allowed discrimination of the different species of the Fusarium graminearum complex (F. graminearum sensu lato) as F. graminearum sensu stricto, Fusarium meridionale and Fusarium boothii. The last two species belonged to NIV chemotype and were detected for the first time in the subtropical region of Argentina while F. graminearum sensu stricto was DON producer only, which was also confirmed by specific PCR assays based on TRI137/TRI7 genes. Our results indicated that the PCR assays evaluated in this work are reliable diagnostic tools to detect the main toxigenic Fusarium species associated to cereal grains in Argentina. An extensive epidemiological survey based on the approach presented in this work is currently in progress to know the mycotoxigenic hazard of Fusarium species in cereal grains from the subtropical region of Argentina.  相似文献

Corn samples were collected in 1999 from three departments of Entre Réos province, Argentina, and were surveyed for mould contamination and natural occurrence ofFusarium mycotoxins, ochratoxin A and aflatoxins.Fusarium verticillioides was the most prevalent fungal species recorded at all departments. Zearalenone, deoxynivalenol and ochratoxin A were not found in any samples. Only one of the 52 corn samples analysed was contaminated with aflatoxin B1 (17 μg/kg). Fumonisin B1 was found in 58 % of samples (range of positive samples: 47– 3,347 μg/kg), fumonisin B2 in 33.0 % (range of positive samples: 23–537 μg/kg) and fumonisin B3 in 25.0 % (range of positive samples: 24–287 μg/kg) of them. This is the first report on the natural occurrence of mycotoxins in corn from Entre Ríos province, Argentina. Levels of fumonisins were lower than detected in other Argentinian provinces.  相似文献

The objectives of this study were the examination of genetic similarities in a diverse group of maize inbreds and an investigation of the incidence of shared haplotypes within closely related groups. Size polymorphisms from 218 mapped simple-sequence repeats (SSR) for 57 entries were detected with the ABI377 Prism system and scored with Genotyper software. The standard error for the estimated size of identical PCR products was 0.13 base pairs. Size estimates were used to examine genetic relationships among the Iodent, flint, corn belt dent, sweet corn and popcorn groups in Zea mays L. inbreds. Cluster analysis of SSR distance data from 57 entries showed similarity between the European flints (F2, F7 and EP1), CO109 and the su1 sweet corns developed in the United States. The inbred F64 from Argentina was distinct from all other entries. Close examination of two sources of B37 revealed that the Purdue University version of B37 contains a set of alleles characteristic of B73. Five groups (Iodents, European flints, the B73 group of corn belt dents, su1 sweet corn, popcorn) show persistent within-group haplotypes. Received: 17 July 2000 / Accepted: 5 January 2001  相似文献

We examined nine pairs of near-isogenic hybrids of Bacillus thuringiensis (Bt) and non-Bt corn, Zea mays L., at two locations in 1999 and three locations in 2000 to compare the effects of Bt toxins on damage caused by Helicoverpa zea (Boddie) to whorl stage field corn, and ear damage at harvest, as well as yield. We found that whorl damage was less in all Bt hybrids compared with their non-Bt counterparts each year and at each location. Differences in ear damage between Bt and non-Bt hybrids, however, differed in 1999 and 2000. In 1999, only one Bt hybrid, NC+5788Bt, had less ear damage than its non-Bt counterpart at the dryland site, whereas four Bt hybrids, C8120Bt, P31B13Bt, P33VO8Bt, and NC+5788Bt, had less damage at the irrigated site. In 2000, most Bt hybrids had less ear damage than their non-Bt counterparts at each location. Differences in whorl damage did not translate into yield differences. However, variations in ear damage were partially reflected in yield differences. In 1999, P31B13Bt and P33V08Bt had higher yields than their non-Bt counterparts at both sites, whereas in 2000 all Bt hybrids had higher yields. Also, although whorl damage was not correlated with yield, ear damage was negatively correlated with yield; increasing ear damage by H. zea decreased yield for Bt and non-Bt hybrids alike. Overall, depending on location and year, each centimeter of H. zea ear damage reduced yield by between 2 and 13%.  相似文献

A 2-yr field and laboratory study investigated insect resistance of the maize, Zea mays L., inbred Tex6, which has previously demonstrated resistance to Aspergillus ear rot and aflatoxin production, relative to susceptible inbred B73. Field studies indicated significantly greater resistance to insect feeding of V4-V8 growth stage Tex6 plants compared with B73 plants in both years, primarily to flea beetles (Chaetonema spp.). Field studies of natural (1999) and artificial (2000) infestations of corn earworms, Helicoverpa zea (Boddie), indicated much lower levels of kernel damage at milk stage (approximately three-fold) and smaller surviving larvae (approximately three-fold) in Tex6 compared with B73 ears. At harvest similar trends in reduction of numbers of damaged kernels per ear, as well as incidence and numbers of kernels per ear symptomatically infected by Fusarium spp. were noted. Laboratory studies indicated little difference in mortality or survivor weight of caterpillars or sap beetle adults caged with milk stage kernels of the two inbreds. However, assays with silks indicated significantly greater mortality of H. zea in both 1999 and 2000, and European corn borer, Ostrinia nubilalis (Hübner) in 1999 (only year tested) when fed Tex6 silks compared with B73 silks. Pollinated Tex6 silks were generally darker colored and more toxic than unpollinated silks. Thus, it is possible that commercially usable inbreds with resistance to insects, which also contribute to the mycotoxin problem through vectoring and damage, could be produced using Tex6 as a source.  相似文献

During the 1989 corn harvest season, numerous reports of equine leukoencephalomalacia (ELEM) outbreaks and a pulmonary edema (PPE) syndrome in swine from several regions of the United States were received by the National Veterinary Services Laboratories (NVSL), Ames, Iowa. Previous and concurrent research linked Fusarium moniliforme and fumonisin-contaminated feeds to both diseases. Chemical and mycological investigations revealed fumonisin B1 (FB1) concentrations of 20 to 360 ppm in suspect swine feeds and 8 to 117 ppm in suspect equine feeds. Nonproblem feeds contained concentrations below 8 ppm. Fusarium moniliforme and Fusarium proliferatum were isolated from both problem and nonproblem equine and swine feeds. When cultured on autoclaved corn, the F. moniliforme and F. proliferatum isolates produced respective FB1 and fumonisin B2 (FB2) that range from less than 5 to more than 2450 ppm and less than 5 to more than 1000 ppm, respectively. Isolates from both problem and nonproblem feeds produced high levels (greater than 500 ppm) in culture. Reported here is a review of chemical and mycological data resulting from the study of several cases of PPE and ELEM.  相似文献

Aspidosperma quebracho-blanco (white quebracho) and Schinopsis balansae (red quebracho) are distinctive trees of the South American Park in Argentina. Quebrachos are found in forests that have been exploited very intensively. The object of this work was the identification of biotic and abiotic factors specially fungal pathogen that affect the quality of both species and its relation with germination. Seeds where evaluated through germination test and the percentage of the incidence of fungal agents in two different years of harvest was determined. In S. balansae the germination rate was 77% and of 27% in 2000 and 2001 harvests, respectively. Associations fungi-germination were found in 2001 for Alternaria spp., Curvularia spp., and Fusarium spp., showing an coefficient of correlation = -0.84; -0.85 and -0.73 (p < 0.00004), respectively. A high percentage of vane seeds (55%) was also found in 2001 harvest, due to adverse environmental factors, specifically higher precipitations during flowering. In A. quebracho-blanco seeds, the germination rate was 50% and 90% in 2000 and 2003 respectively, with a 42% of immature seeds in 2000 harvest that was associated to high precipitations and high temperatures during flowering and ripping of fruits. The incidence of pathogens was low and did not have association to germination.  相似文献

Hydrolysis of probe substrates, eight possible monodeoxy and mono-O-methyl analogs of p-nitrophenyl alpha-D-glucopyranoside (pNP alpha-D-Glc), modified at the C-2, C-3, C-4, and C-6 positions, was studied as part of investigations into the glycon specificities of seven alpha-glucosidases (EC 3.2.1.20) isolated from Saccharomyces cerevisiae, Bacillus stearothermophilus, honeybee (two enzymes), sugar beet, flint corn, and Aspergillus niger. The glucosidases from sugar beet, flint corn, and A. niger were found to hydrolyze the 2-deoxy analogs with substantially higher activities than against pNP alpha-D-Glc. Moreover, the flint corn and A. niger enzymes showed hydrolyzing activities, although low, for the 3-deoxy analog. The other four alpha-glucosidases did not exhibit any activities for either the 2- or the 3-deoxy analogs. None of the seven enzymes exhibited any activities toward the 4-deoxy, 6-deoxy, or any of the methoxy analogs. The hydrolysis results, with the deoxy substrate analogs, demonstrated that alpha-glucosidases having remarkably different glycon specificities exist in nature. Further insight into the hydrolysis of deoxyglycosides was obtained by determining the kinetic parameters (k(cat) and K(m)) for the reactions of sugar beet, flint corn, and A. niger enzymes.  相似文献

In 1999 and 2000, yellow sticky cards and sweep net samples were used to document the occurrence of an overwintering adult generation of Chaetocnema pulicaria Melsheimer, corn flea beetle, followed by two distinct populations peaks during the growing season in Iowa Emergence of the overwintering adult generation started in mid-April and continued until early June in both years, with populations as high as 45 +/- 7.9 per 10 sweeps. Periods that ranged from 14 to 32 d were observed in 1999 and 2000 when C. pulicaria was not found following the overwintering generation. The first summer peak of C pulicaria was observed between the end of June into the middle of July, with the highest observed peak at 16.70 +/- 1.42 C. pulicaria per 10 sweeps in cornfields. The second summer peak of C pulicaria was observed between the middle into early September, with populations as high as 27.80 +/- 2.76 C. pulicaria per 10 sweeps. During the growing season, more C. pulicaria were caught on yellow sticky cards originating from soybean borders than from grass borders. There were significantly greater numbers of C. pulicaria on yellow sticky cards located in grass borders adjacent to cornfields at the end of the growing season, compared with yellow sticky cards located within cornfields, indicating the movement of C. pulicaria from the cornfield back into the grass borders at the end of the growing season. In 2000, from August to the end of the corn growing season, significantly more C. pulicaria were found in grass borders than in the cornfields. Based on this new quantitative information, planting time could be altered to avoid the emergence of the overwintering generation of C. pulicaria. In addition, knowledge concerning the seasonalities of the first and second population peaks of C pulicaria during the corn growing season could be used to recommend optimal timing for foliar-applied insecticide applications. This new knowledge concerning the seasonal dynamics of C pulicaria will help to improve management recommendations for Stewart's disease of corn, caused by the bacterium Pantoea stewartii, and that is vectored by C pulicaria.  相似文献

Three mycotoxins were identified in the infected ears: zearalenone, deoxynivalenol, and 15-acetyl-deoxynivalenol. This is the first report of the presence of 15-acetyl-deoxynivalenol on corn ears in the field prior to harvest and in combination with deoxynivalenol and zearalenone. Ninety-nine cultures were selected from colonies growing from kernels on an agar medium; 30% of the cultures were F. graminearum, 23% were F. subglutinans, 20% were F. moniliforme, 14% were F. oxysporum, and 12% were F. proliferatum.  相似文献
