Mechanism of soybean nodule adaptation to different oxygen pressures   总被引:7,自引:2,他引:7  
Abstract. Soybean nodules showed the ability to adapt to oxygen pressures above and below ambient levels and this adaptation involved a decrease in cortical intercellular air-spaces with increasing oxygen pressure. Nodules were grown in oxygen pressures from 4.7 to 75 kPa and the decrease in number and size of cortical intercellular spaces with increasing oxygen pressure was the result of a change in cell structure and the deposition of an electron dense material within intercellular spaces. Exposure to a saturating pressure of acetylene caused a similar inhibition of respiration and nitrogenase activity in nodules developed in oxygen pressures from 4.7 to 47 kPa, suggesting that putative acetylene-induced changes in oxygen diffusion resistance occur by a different mechanism than that involved in long-term adaptation to oxygen. However, in nodules grown at 75 kPa oxygen, the initial specific activities were lower and did not show an acetylene induced decline. The results are discussed in terms of the current theories of regulation of nitrogenase activity by oxygen availability.  相似文献

The effect of excision on O2 diffusion and metabolism in soybean nodules   总被引:2,自引:0,他引:2  
Nitrogen-fixing nodules of soybean [Glycine max (L.) Merr. cv. Maple Arrow inoculated with Bradyrhizobium japonicum USDA 16] were studied before and after excision from the root to determine the role the O2 regulation plays in the inhibition of nodule activity and the potential for using excised nodules nodules in studies of nodule metabolism. Relative nitrogenase (EC 1.7.99.2) activity (H2 evolution in N2:O2) and nodule respiration (CO2 evolution) were monitored first in intact nodulated roots and then in freshly excised nodules of the same plant to determine the time course of the decline in nodule metabolism. Folowing excision, nitrogenase activity and respiration declined rapidly in the first minute and then more gradually. After 40 min the rate of H2 evolution was only 14–28% of that in the intact plant. In some nodules activity declined steadily, and in others there was a partial recovery in activity ca 10 min after detachment. Infected cell O2 concentration (Oi), measured by a spectro-photometric technique, also declined after nodule detachment with a time course similar to the declines in nitrogenase activity and respiration. Following excision, Oi levels declined rapidly from ca 21 nM in attached nodules to 8–12 nM at 4–10 min after excision and then more gradually to 2–3 nM O2 at 30–40 min after excision. These results show that the nodules' permeability to gas diffusion continued to be regulated for up to 40 min after detachement. At 40 min after detachment, when excised nodules displayed steady-state rates of gas exchange, linear increases in pO2 from 20 to 100% at 4% min?1 resulted in recoveries of H2 and CO2 evolution, indicating that Oi limited nitrogenase activity durig this period, and that energy reserves were greatly in excess of the O2 available for respiration. When detached nodules were equilibrated for 12 h at 20, 30 and 50% O2, Oi values measured at supra-ambient pO2 were greater than those at 20% O2 and were linked with a more rapid decline in nitrogenase activity. Also, increases in external pO2 (Oc) failed to stimulate nodule metabolism, suggesting that the nodules' energy reserves were no longer greatly in excess of their respiratory demands. It was concluded that soybean nodules could provide useful material for steady-state studies of nodule metabolism between 40 and 240 min after detachment, but to attain metabolic rates equivalent to in vivo rates the nodules must be exposed to above-ambient pO2.  相似文献

A spin-echo pulse sequence was used to obtain 1H nuclear magnetic resonance microimages of soybean nodules, with resolution to 40 mm and 3D presentation. At an acquisition time (TR) of c. 270 ms and a short echo time (TE) of 8.3 ms, a high proton intensity was detected in infected tissue and vascular strands, relative to the cortex. A longer TE of 15.5 ms was associated with a relatively low proton intensity in infected tissue. Thus protons in infected tissue were characterized by a short T2. Following detopping of the plant, or treatment of the root system with Ar:O2, proton intensity (TE 5.9, TR 120 ms) decreased exponentially in a zone corresponding to the inner cortex-outer infected region. This result is interpreted as being caused by a loss of water mobility (i.e. increase in viscosity) or a filling of intercellular air spaces with fluid (i.e. loss of air-water interfaces). The membrane impermeant tracer lucifer yellow was observed to infiltrate from the rhizosphere through the nodule cortex into the infected region in intact nodules of detopped and Ar:O2-treated, but not control, plants. This result is consistent with the filling of air spaces with fluid following the imposed treatments, allowing infiltration of aqueous tracers. Variation in intercellular air space volume within the inner cortex has been suggested as a mechanism to allow a variable rate of diffusion of O2 into soybean nodules.  相似文献

Abstract. A glycoprotein which occludes intercellular spaces in the inner cortex of legume nodules may be involved in controlling oxygen diffusion into rhizobial-infected cells. Here we investigated this possibility by localizing the glycoprotein using monoclonal antibodies and immunogold labelling in nodulated roots of soybean cv. Clarke inoculated with Bradyrhizobium japonicum strain RCR3442 exposed to atmospheres with either 10, 21 (control) or 40% oxygen for 28d. Infected cells showed evidence of premature senescence when grown in above or below ambient pO2 particularly at 10% oxygen, although cortical cells appeared to be little altered by oxygen treatment. In the inner cortical cells, more glycoprotein was seen to be occluding intercellular spaces of those nodules subjected to 40% oxygen and less in those nodules exposed to 10% oxygen, when compared to controls. This observation, made at the light microscope level (using silver enhancement) was confirmed under the TEM using immunogold labelling. Therefore, it is suggested that intercellular space glycoprotein is one of the structural components of the diffusion resistance in the cortex of legume nodules.  相似文献

After a 2 h exposure of intact soybean nodules to high concentrations of NaCl (100mol m?3) or oxygen (8OkPa O2), morphometric computations carried out using an image analysis technique on semi-thin sections showed that both treatments induced a decrease in the area of the inner-cortex cells, which were then characterized by a tangential elongation. In contrast, no significant change in area occurred in the middle-cortex cells although their elongation decreased. Electron microscopic observations showed that in the inner-cortex cells changes included the presence of wall infoldings, an enlarged periplasmic space and a lobate nucleus whose chromatin distribution differed from that of the control. Structural changes also occurred in the endoplasmic reticulum, microbodies, mitochondria and plastids. From several of these changes, which are similar to those noted in osmocontractil cells in response to external stimuli, it can be hypothesized that the inner cortex may provide a potential mechanism for the control of oxygen diffusion through the nodules.  相似文献

The effects of increasing rhizosphere pO2on nitrogenase activity and nodule resistance to O2diffusion were investigated in soybean plants [Glycine max (L.) Merr. cv. Harosoy 63] in which nitrogenase (EC 1.7.99.2) activities were inhibited by (a) removal of the phloem tissue at the base of the stem (stem girdling), (b) exposure of roots to 10 mM NO3over 5 days (NO3-treated), or (c) partial inactivation of nitrogenase activity by an exposure of nodulated roots to 100 kPa O2(O2-inhibitcd). In control plants and in plants which had been treated with 100 kPa O2, increasing rhizosphere O2concentrations in 10 kPa increments from 20 to 70 kPa did not alter the steady-state nitrogenase activity. In contrast, in plants in which nitrogenase activities were depressed by stem girdling or by exposure to NO3, increasing rhizosphere pO2resulted in a recovery of 57 or 67%, respectively, of the initial, depressed rates of nitrogenase activity. This suggests that the nitrogenase activity of stem-girdled and NO3-treated soybeans was O2-limited. For each treatment, theoretical resistance values for O2diffusion into nodules were estimated from measured rates of CO2exchange, assuming a respiratory quotient of 1.1 and 0 kPa of O2in the infected cells. At an external partial pressure of 20 kPa O2, the stem-girdled and NO3--treated plants displayed resistance values which were 4 to 8.6 times higher than those in the nodules of the control plants. In control and O2-inhibited plants, increases in pO2from 20 to 70 kPa in 10 kPa increments resulted in a 2.5- to 3.9-fold increase in diffusion resistance to O2, and had little effect on either respiration or nitrogenase activity. In contrast, in stem-girdled and NO3--treated plants, increases in external pO2had little effect on diffusion resistance to O2, but resulted in a 2.3- to 3.2-fold increase in nodule respiration and nitrogenase activity. These results are consistent with stem-girdling and NO3--inhibition treatments limiting phloem supply to nodules causing an increase in diffusion resistance to O2at 20 kPa and an apparent insensitivity of diffusion resistance to increases in external pO2.  相似文献

Growth, nodulation and N2 fixation inGlycine max L. Merr., cv. Biison as affected by the relative humidity of air (RH) during the dark period (95 or 50 – 65 %) and day/night root temperature (Tr) (28/28, 25/25, 18/18, 22/28, 22/18 °C) were studied. The growth parameters (plant fresh and dry mass, yield), nodulation (nodule number and fresh mass) and N2 fixation abilities (total nitrogen content, nitrogenase activity) increased significantly with the increasing Tr. In addition, at the same Tr during the day all studied parameters were increased at the higher Tr during the dark period. Growth, nodulation and N2 fixation were significantly enhanced at low RH. The findings indicate that all studied parameters could be regulated by environmental factors during the dark period.  相似文献

BACKGROUND AND AIMS: Soybean (Glycine max) is among the many legumes that are well known for 'hardseededness'. This feature can be beneficial for long-term seed survival, but is undesirable for the food processing industry. There is substantial disagreement concerning the mechanisms and related structures that control the permeability properties of soybean seed coats. In this work, the structural component that controls water entry into the seed is identified. METHODS: Six soybean cultivars were tested for their seed coat permeabilities to water. To identify the structural feature(s) that may contribute to the determination of these permeabilities, fluorescent tracer dyes, and light and electron microscopic techniques were used. KEY RESULTS: The cultivar 'Tachanagaha' has the most permeable seed coat, 'OX 951' the least permeable seed coat, and the permeabilities of the rest ('Harovinton', 'Williams', 'Clark L 67-3469', and 'Harosoy 63') are intermediate. All seeds have surface deposits, depressions, a light line, and a cuticle about 0.2 microm thick overlaying the palisade layer. In permeable cultivars the cuticle tends to break, whereas in impermeable seeds of 'OX 951' it remains intact. In the case of permeable seed coats, the majority of the cracks are from 1 to 5 micro m wide and from 20 to 200 micro m long, and occur more frequently on the dorsal side than in other regions of the seed coat, a position that correlates with the site of initial water uptake. CONCLUSIONS: The cuticle of the palisade layer is the key factor that determines the permeability property of a soybean seed coat. The cuticle of a permeable seed coat is mechanically weak and develops small cracks through which water can pass. The cuticle of an impermeable seed coat is mechanically strong and does not crack under normal circumstances.  相似文献

Soybean is an Important cash crop and its productivity is significantly hampered by salt stress. High salt Imposes negative impacts on growth, nodulation, agronomy traits, seed quality and quantity, and thus reduces the yield of soybean. To cope with salt stress, soybean has developed several tolerance mechanisms, including: (I) maintenance of ion homeostasis; (ii) adjustment in response to osmotic stress; (iii) restoration of osmotic balance; and (iv) other metabolic and structural adaptations. The regulatory network for abiotic stress responses in higher plants has been studied extensively in model plants such as Arabidopsis thaliana. Some homologous components involved in salt stress responses have been identified in soybean. In this review, we tried to integrate the relevant works on soybean and proposes a working model to descdbe Its salt stress responses at the molecular level.  相似文献

Effects of drought on nitrogen fixation in soybean root nodules   总被引:3,自引:0,他引:3  
Soybean plants [Glycine max (L.) Merr.] were grown in silica sand and were drought stressed for a 4 week period during reproductive development and without any mineral N supply in order to maximize demand for fixed nitrogen. A strain of Bradyrhizobium japonicum that forms large quantities of polysaccharide in nodules was used to determine whether or not the supply of reduced carbon to bacteroids limits nitrogenase activity. A depression of 30–40% in nitrogen content in leaves and pods of stressed plants indicated a marked decline in nitrogen fixation activity during the drought period. A 50% increase in the accumulation of bacterial polysaccharide in nodules accompanied this major decrease in nitrogen fixation activity and this result indicates that the negative impact of drought on nodules was not due to a depression of carbon supply to bacteroids. The drought treatment resulted in a statistically significant increase in N concentration in leaves and pods. Because N concentration and chlorophyll concentration in leaves were not depressed, there was no evidence of nitrogen deficiency in drought‐stressed plants, and this result indicates that the negative impact of drought on nodule function was not the cause of the depression of shoot growth. At the end of the drought period, the concentration of carbohydrates, amino nitrogen, and ureides was significantly increased in nodules on drought‐stressed plants. The overall results support the view that, under drought conditions, nitrogen fixation activity in nodules was depressed because demand for fixed N to support growth was lower.  相似文献

Abstract. In soybean, stores of carbon within the leaf have been demonstrated to support nodule metabolism under both photosynthetic and non-photosynthetic conditions. Indeed, a net depletion of nodule starch is observed only under conditions of suboptimal rates of nodule metabolism. Therefore, maximal rates of nodule metabolism are associated with a continual supply of phloem sap to the nodule, delivering water, carbon and other solutes. A restriction of phloem supply to the nodule may result in changes in turgor between the apoplast of the export pathway and the symplast of the nodule. This change may cause the observed decrease in the permeability to gases and to the rate of product export from nodules deprived of a phloem supply. It is suggested that nodule metabolism is homeostatically regulated in terms of internal O2 levels by the delivery of phloem water and solutes.  相似文献

The possibility that respiration limits oxygen access to nitrogenase was tested by artificially upsetting the balance between oxygen consumption (respiration) and oxygen influx (diffusion). Argon treatment of the nodulated root system on intact plants stopped in vivo nitrogenase activity almost completely. Upon return to air, nitrogenase activity was very low and recovered gradually to full activity after about 5 h. In vitro measurements on nodule homogenates indicated that active nitrogenase was lost upon the shift from low (argon) to normal (air) oxygen. Maintenance of nodulated root systems at low temperature (2°C) inhibited both respiration and in vivo nitrogenase activity. Upon return to normal temperature (22°C), oxygen uptake recovered very rapidly, but nitrogenase activity recovered only gradually to full activity after about 5 to 6 h. Again, loss of active nitrogenase could, at least partly, explain the reduced in vivo nitrogenase activity. The effects from a temporarily impaired balance between oxygen consumption and oxygen influx thus point to the importance of respiration for limiting oxygen access to nitrogenase.  相似文献

Analysis of apoplastic solutes in the cortex of soybean nodules   总被引:3,自引:0,他引:3  
Various techniques were used to extract solutes from the free space of intact soybean [ Glycine max (L.) Merr.] nodules. A variety of solutes (carbohydrates, amino acids, organic acids, ions) was found, but the major solute obtained with all methods was allantoic acid. Most work was done with a technique involving vacuum infiltration of intact detached nodules with water. This approach provided rapid sampling of the apoplastic solutes, and the results indicated that solutes were not derived from the xylem and phloem of ruptured vascular bundles. Infiltration of intact nodules with Fast Green showed dye penetration only to the barrier in the inner cortex, indicating that infected tissues did not contribute to solute composition. Although allantoic acid was the only ureide which could be detected in solute samples, no evidence was obtained for the presence of allantoinase in the cortical apoplast. The results suggest the transport of allantoic acid by an apoplastic route in nodules or the release of allantoic acid to the cortical apoplast in response to treatments which disrupt ureide export. Calculated values for solute concentrations in the cortical apoplast were in the hundred millimolar range, suggesting that apoplastic solutes may represent a significant osmotic component in the nodule cortex.  相似文献

The vascular anatomy of soybean nodules [Glycine max (L.) Merr.] suggests that export of solutes in the xylem should be dependent on influx of water in the phloem. However, after severing of stem xylem and phloem by shoot decapitation, export of ureides from nodules continued at an approximately linear rate for 5h. This result was obtained with decapitated roots remaining in the sand medium, but when roots were disturbed by removal from the rooting medium prior to shoot decapitation, export of ureides from nodules was greatly reduced. Stem exudate could not be collected from disturbed roots, indicating that flow in the root xylem had ceased. Thus, ureide export from nodules appeared to be dependent on a continuation of flow in the root xylem. When seedlings were fed a mixture of 3H2O and 14C-inulin for periods of 14–21 min, nodules had higher 3H/14C ratios than roots from which they were detached. The combined results are not consistent with the proposal that export of nitrogenous compounds from nodules is dependent on import of water via the phloem. The results do support the view that a portion of the water required for xylem export from soybean nodules is supplied via a symplastic route from root cortex to nodule cortex to the nodule vascular apoplast.  相似文献

Abstract The ureide content of soybean (Glycine max (L.) Merr.) nodules was unaffected by variations in the transpirational rate, while whole plant manipulations designed to decrease phloem supply to nodules resulted in lower rates of nitrogenase activity and an increase in the ureide content of the nodules. The rate of ureide export from the nodule was estimated from the exponential rate of decrease in the pool size of ureides in nodules, following exposure to an N2-free atmosphere (Ar:O2). Export was greatly reduced under treatments which reduced phloem supply to the nodule. A water budget for nodules suggested that the delivery of water to the nodule via mass flow in the phloem was comparable to that required for export of ureides from the nodule in the xylem from the nodule. Therefore, we suggest that xylem export from nodules is related to the phloem supply to the nodule rather than to the transpirational flux in the parent root. This suggestion is related to the reported decreases in nodule permeability to gases under conditions of phloem deprivation.  相似文献

Soybean plants require high amounts of nitrogen, which are mainly obtained from biological nitrogen fixation. A field experiment was conducted by soybean (Glycine max) genotypes, growing two varieties (Shohag and BARI Soybean6) and two advanced lines (MTD10 and BGM02026) of soybean with or without Rhizobium sp. BARIRGm901 inoculation. Soybean plants of all genotypes inoculated with Rhizobium sp. BARIRGm901 produced greater nodule numbers, nodule weight, shoot and root biomass, and plant height than non-inoculated plants. Similarly, inoculated plants showed enhanced activity of nitrogenase (NA) enzyme, contributing to higher nitrogen fixation and assimilation, compared to non-inoculated soybean plants in both years. Plants inoculated with Rhizobium sp. BARIRGm901 also showed higher pod, stover, and seed yield than non-inoculated plants. Therefore, Rhizobium sp. BARIRGm901 established an effective symbiotic relationship with a range of soybean genotypes and thus increased the nodulation, growth, and yield of soybean grown in gray terrace soils in Bangladesh.  相似文献

The low gas permeability of a diffusion barrier in the cortex of soybean nodules plays a significant role in the protection of nitrogenase from oxygen inactivation. It may also set an upper limit on nodule respiration and nitrogen fixation rates. Two methods which have been used to quantify the gas permeability of leguminous nodules are reviewed and found to be unreliable. A new assay technique for determining both the nodule activity and gas permeability is developed and tested. This ‘lag-phase’ assay is based on the time nodules require to reach steady-state ethylene production after being exposed to acetylene. The technique is rapid, insensitive to errors in biochemical parameters associated with nitrogenase, and is non-destructive. The method was tested with intact aeroponically grown soybean plants for which the mean nodule gas permeability was found to be 13.3×10−3 mms−1. This corresponds to a layer of cells approximately 35 um thick and is consistent with previously reported values.  相似文献

The permeability (P) of the gaseous diffusion barrier in the nodules of soybean [Glycine max (L.) Merr.] decreases when water deficits are extended over a 7 to 10 d period. The mechanism controlling P changes is unclear, but may result from the release of water to intercellular pathways, and an associated change in the nodule water potential. The purpose of these experiments was to impose water deficit treatments rapidly in order to determine the early sequence of the responses of nodule water potential and nodule gas exchange without the complications that arise from long-term water deficit treatments. A vertical, split-root system was used to separate nodule drying effects from plant water deficits by replacing humidified air that was passed over upper root nodules in well-watered plants with dry air, or by replacing the nutrient solution that surrounded lower roots with -1.0 MPa polyethylene glycol (PEG) solution, or by a combination of the dry air and PEG treatments. The PEG treatment caused large decreases in both the components of nodule water potential and nodule relative water content, but there was no indication that these factors had immediate, direct effects on either nitrogenase activity or P. After 7 h of the PEG treatment a significant decrease in nitrogenase activity was found but no decrease in P was detected. These results indicate that changes in nitrogenase activity in response to water deficits precede decreases in P. Exposure of nodules to dry air in well-watered plants had no significant effect on either nitrogenase activity or P during the 7 h treatment.  相似文献
