Mesenchymal control of branching pattern in the fetal mouse lung 总被引:1,自引:0,他引:1
The effect of mesenchyme on specialization of respiratory epithelium in the fetal mouse was tested in organ cultures. Heterologous combinations were made between respiratory and non-respiratory lung epithelia and the corresponding mesenchymes. Isolated terminal respiratory buds of fetal mouse lungs were recombined with mesenchyme from chick lung parabronchi, mouse trachea or from the avascular, non-respiratory air sacs of chick lungs. Isolated non-branching chick air sacs were combined with mouse terminal bud mesenchyme or mesenchyme from the respiratory branches of chick lungs. Air sac epithelia branched in a pattern characteristic of the chick lung when combined with chick respiratory mesenchyme and in a pattern characteristic of mouse lung when combined with mouse terminal bud mesenchyme. Mouse terminal bud epithelia did not branch with either mouse tracheal mesenchyme or chick air sac mesenchyme but branched in a chick pattern with chick parabronchial mesenchyme. Electron microscopic examination of the cultures showed that all chick air sac epithelial cultures failed to produce surfactant (lamellar bodies) even when they branched. Control cultures of mouse terminal buds contained large numbers of lamellar bodies; mesenchyme which suppressed branching reduced the number of lamellar bodies to only a few in a small proportion of the cells. Culture medium supplemented with growth factors and hormones increased the number of lamellar bodies in heterologous mouse combinations but did not bring the number to control levels. Supplemented medium had no effect on lamellar body production by chick air sac epithelium. The results indicate that branching pattern is determined by the mesenchyme surrounding the epithelial primordium. However, the capacity to synthesize surfactant is determined by the source of the epithelium; mesenchyme may control the degree of expression but not the absolute presence or absence of the differentiated condition. 相似文献

In alveolar type II cells, the release of surfactant is considerably delayed after the formation of exocytotic fusion pores, suggesting that content dispersal may be limited by fusion pore diameter and subject to regulation at a postfusion level. To address this issue, we used confocal FRAP and N-(3-triethylammoniumpropyl)-4-(4-[dibutylamino]styryl) pyridinium dibromide (FM 1-43), a dye yielding intense localized fluorescence of surfactant when entering the vesicle lumen through the fusion pore (Haller, T., J. Ortmayr, F. Friedrich, H. Volkl, and P. Dietl. 1998. Proc. Natl. Acad. Sci. USA. 95:1579-1584). Thus, we have been able to monitor the dynamics of individual fusion pores up to hours in intact cells, and to calculate pore diameters using a diffusion model derived from Fick's law. After formation, fusion pores were arrested in a state impeding the release of vesicle contents, and expanded at irregular times thereafter. The expansion rate of initial pores and the probability of late expansions were increased by elevation of the cytoplasmic Ca2+ concentration. Consistently, content release correlated with the occurrence of Ca2+ oscillations in ATP-treated cells, and expanded fusion pores were detectable by EM. This study supports a new concept in exocytosis, implicating fusion pores in the regulation of content release for extended periods after initial formation. 相似文献

Summary We developed methodology to isolate and culture rat alveolar Type II cells under conditions that preserved their proliferative
capacity, and applied lipofection to introduce an immortalizing gene into the cells. Briefly, the alveolar Type II cells were
isolated from male F344 rats using airway perfusion with a pronase solution followed by incubation for 30 min at 37° C. Cells
obtained by pronase digestion were predominantly epithelial in morphology and were positive for Papanicolaou and alkaline
phosphatase staining. These cells could be maintained on an extracellular matrix of fibronectin and Type IV collagen in a
low serum, insulin-supplemented Ham’s F12 growth medium for four to five passages. Rat alveolar epithelial cells obtained
by this method were transformed with the SV40-T antigen gene and two immortalized cell lines (RLE-6T and RLE-6TN) were obtained.
The RLE-6T line exhibits positive nuclear immunostaining for the SV40-T antigen and the RLE-6TN line does not. PCR analysis
of genomic DNA from the RLE-6T and RLE-6TN cells demonstrated the T-antigen gene was present only in the RLE-6T line indicating
the RLE-6TN line is likely derived from a spontaneous transformant. After more than 50 population doublings, the RLE-6T cells
stained positive for cytokeratin, possessed alkaline phosphatase activity, and contained lipid-containing inclusion bodies
(phosphine 3R staining); all characteristics of alveolar Type II cells. The RLE-6TN cells exhibited similar characteristics
except they did not express alkaline phosphatase activity. Early passage RLE-6T and 6TN cells showed a near diploid chromosome
number. However, at later passages the 6T cells became polyploid, while the 6TN genotype remained stable. The RLE-6T and 6TN
cells were not tumorigenic in nude mice. The cell isolation methods reported and the novel cell lines produced represent potentially
useful tools to study the role of pulmonary epithelial cells in neoplastic and nonneoplastic lung disease. 相似文献

Summary Centrifugal elutriation (counterflow centrifugation) was used to develop a reproducible method for obtaining a nearly pure
population of isolated alveolar type II cells. Lung was dissociated into individual cells with recrystallized trypsin, and
the type II cells were partially purified by centrifugation on a discontinuous density gradient. The alveolar type II cells
were finally purified by centrifugal elutriation. Cells were collected from the elutriator rotor by stepwise increases in
flow rates. Cells obtained at flow rates of 7 and 14 ml per min were lymphocytes, other small cells, a few type II cells and
cell debris; cells collected at flow rates of 18 and 22 ml per min were mainly type II cells; and cells collected at flow
rates of 28, 34 and 43 ml per min were macrophages, some type II cells, other lung cells and cell aggregates. At flow rates
of 18 and 22 ml per min, 1.9±1.0×106 cells per rat lung (mean±S.D.,n=30) were recovered of which 86±6% were type II cells. At these flow rates, 94% of the cells excluded the vital dye erythrosin
B from their cytoplasm. They consumed oxygen at a rate of 101±21 nmol per hr·106 cells (mean±S.D.,n=4), and their oxygen consumption increased only 10% after 10mm sodium succinate was added. The cells incorporated [14C]leucine into protein and lipid for 4 hr. Electron micrographs of the cells collected at flow rates of 18 and 22 ml per min
show a high percentage of morphologically intact alveolar type II cells. We conclude that centrifugal elutriation is a reproducible
method for obtaining nearly pure, metabolically active alveolar type II cells.

Summary A serum-free culture medium (defined medium = DM) was elaborated by adding to Eagle’s minimum essential medium (MEM), non-essential
amino acids, transferrin, putrescine, tripeptide glycyl-histidyl-lysine, somatostatin, sodium selenite, ethanolamine, phosphoethanolamine,
sodium pyruvate, and metal trace elements. This medium was tested for its ability to support sustained surfactant biosynthesis
in fetal alveolar epithelial type II cells. For up to 8 days, ultrastructure was maintained with persistance of lamellar inclusion
bodies. Thymidine incorporation into DNA was enhanced about 50% in DM as compared with MEM, whereas it was enhanced 300% in
10% fetal bovine serum. With DM, the incorporation of tritiated choline into phosphatidylcholine (PC) of isolated surfactant
material was about twice that with MEM. Deletion experiments evidenced the prominent role of pyruvate, transferrin, and selenium
in the stimulation of surfactant PC biosynthesis. The addition of biotin to DM enhanced surfactant PC biosynthesis slightly
and nonsurfactant PC biosynthesis markedly. The presence of nucleosides seemed unfavorable to the synthesis of surfactant
PC. Type II cells responded to the addition of epidermal growth factor and insulinlike growth factor-I both by increased thymidine
incorporation into DNA and choline incorporation into PC. It is concluded that DM represents a useful tool for cultivating
type II cells without loss of their specialized properties and for studying the regulation of cell proliferation and surfactant

We have investigated the role of carnitine palmitoyltransferase (EC 2.3.1.21) in pulmonar type II pneumocyte, a lung cell responsible for the synthesis of surface active lipids. Adult type II pneumocytes were isolated from rat lung and purified by differential adherence. When these lung cells were incubated with radioactive palmitate, the percentage of radioactivity recovered into dipalmitoylphosphatidylcholine (DPPC), a major surface active lipid, was almost 60% with respect to total phosphatidylcholine (PC) molecular species. Cellular lysates from type II pneumocytes contained detectable amount of carnitine palmitoyltransferase (CPT) activity (1 nmol/min/mg). Most of the CPT activity found in these cells could be inhibited by incubating them for 60 min with 5 M tetradecylglycidic acid (TDGA), a specific and irreversible CPT inhibitor of the malonyl-CoA sensitive CPT isoform (CPT I). TDGA treatment of adult type II pneumocytes caused a significant reduction in the incorporation of radioactive palmitate into PC, though this effect did not seem to be specific for DPPC. TDGA affected the incorporation of radioactive palmitate at the sn2 rather than the sn1 position of the glycerol backbone of PC. The incorporation of radioactive palmitate into DPPC was also observed when these lung cells were incubated with palmitate-labeled palmitoyl-L-carnitine. Our data suggest that type II pneumocyte CPT may play an important role in remodelling PC fatty acid composition and hence DPPC synthesis. 相似文献

Summary The role of epithelial-mesenchymal interactions in the stimulation of lung development by estrogen is now investigated using
organ cultures of lung from male and female fetal rats taken from Days 17 to 21 of gestation. Estradiol at 1 μg/ml was found
to reduce cell proliferation in explants taken during a rapid growth phase (Day 18) and to stimulate surfactant synthesis
in both males and females only in Day 20 explants when cell division is much slower. At this time more epithelial cells from
estrogen-treated explants contained lamellar bodies, which were also secreted to fill the air sacs. These cultures also showed
a significant increase in the frequency of cell-to-cell contacts between epithelial cells and fibroblasts. Uptake of tritiated
estradiol by explants increased from Day 18 onward, and by autoradiography, labeling was located predominantly over fibroblasts.
Using pure cultures of fetal and adult cells, uptake of labeled estradiol was significantly higher in fibroblasts than in
corresponding epithelial cells, and estradiol did not directly enhance palmitate incorporation into epithelial cells. The
results suggest that the earlier maturation and increased surfactant synthesis in female fetal lung is related at least in
part to enhanced binding of estrogen by the fibroblast with subsequent transfer of a maturation factor to the fetal epithelium.

The phospholipase A2 (PLA2) activity of peroxiredoxin (Prdx)6 has important physiological roles in the synthesis of lung surfactant and in the repair of peroxidized cell membranes. These functions require the activity of a lysophospholipid acyl transferase as a critical component of the phospholipid remodeling pathway. We now describe a lysophosphatidylcholine acyl transferase (LPCAT) activity for Prdx6 that showed a strong preference for lysophosphatidylcholine (LPC) as the head group and for palmitoyl CoA in the acylation reaction. The calculated kinetic constants for acylation were Km 18 μM and Vmax 30 nmol/min/mg protein; the Vmax was increased 25-fold by phosphorylation of the protein while Km was unchanged. Study of recombinant protein in vitro and in mouse pulmonary microvascular endothelial cells infected with a lentiviral vector construct indicated that amino acid D31 is crucial for LPCAT activity. A linear incorporation of labeled fatty acyl CoA into dipalmitoyl phosphatidylcholine (PC) indicated that LPC generated by Prdx6 PLA2 activity remained bound to the enzyme for the reacylation reaction. Prdx6 is the first LPCAT enzyme with demonstrated cytoplasmic localization. Thus, Prdx6 is a complete enzyme comprising both PLA2 and LPCAT activities for the remodeling pathway of PC synthesis or for repair of membrane lipid peroxidation. 相似文献

We have developed a Culture system for guinea pig alveolar type II cells using an epithelium-denuded human amnion membrane as a substratum. The differentiated morphology was maintained for 3 wk by both air-interface feeding and immersion feeding when type II cells were cultured on the basement membrane side of the amnion with fibroblasts on the opposite side (coculture). Functionally high levels of surfactant protein B (SP-B) and C (SP-C) messenger ribonucleic acids (mRNAs) were expressed even after the 3-wk cultivation and surfactant protein A mRNA was detected on day 10 of the culture. The differentiation was also maintained when fibroblasts were cultured on lower chambers of the culture plates (separate culture). In contrast, culture of type II cells without fibroblasts (monoculture) could not preserve the mature morphology. When the monoculture was supplemented with keratinocyte growth factor or hepatocyte growth factor, a monolayer of rather cuboidal type II cells with apical microvilli was maintained. However, the percent area of lamellar bodies in these cells was significantly less than that in freshly isolated type II cells, and mRNA expressions of SP-B and SP-C were also considerably suppressed. These findings suggest that other growth factors or combinations of these factors are necessary for the maintenance of the differentiated phenotype. As substratum, a permeable collagen membrane or a thin gel layer of Engelbreth-Holm-Swarm mouse sarcoma extracts did not preserve the mature characteristics. This culture system using an acellular human amnion membrane may provide novel models for research in type II cells. 相似文献

Summary Single-cell-cloned cell lines have been established from primary cultures of neonatal rat mammary glands. A representative
cuboidal cell line, Rama 704, shows the presence of intermediate filamental proteins keratin and vimentin, and occasional
cells express milk fat globule membrane antigens on their apical surfaces. Rama 704 cells grow as a cuboidal pavement in culture
and produce hemispherical blisters or domes when confluent. Noteworthy ultrastructural features are the presence of junctional
complexes, desmosomes, and apical microvilli typical of epithelia. Cells seeded within floating collagen gels with form a
variety of multicellular outgrowths, some of which are ductlike in morphology and are composed of polarized cells surrounding
a central lumen. The cuboidal cells produce elongated cells under conditions of high cell density and also when cells float
off collagen gels and reattach to the plastic substrate. The former elongated cells have been cloned and three cel lines established:
Rama 710, 711, and 712; the latter uncloned elongated cells are termed Rama 704E. The cloned elongated cells show an increase
in the amounts of basement membrane proteins deposited, a lack of junctional complexes and microvilli, and an increase in
the amount of rough endoplasmic reticulum compared with their parental cells. Rama 704E cells show an enhanced deposition
of basement membrane proteins and increased amounts of actin in the cytoplasm over the elongated cell lines and contain microfilaments
and pincocytotic vesicles similar to those seen in myoepithelial cells. All the elongated cells and lines fail to form ductlike
structures within collagen gels. None of the cell lines form tumors in syngeneic rats although they all produce some tumors
in nude mice, which are composed of cords of epithelioid cells and spindle cells in varying proportions. In addition, some
of the Rama 704 tumors contain rhabdomyoblastic elements that penetrate the host fat pad. This is the first report of the
isolation and characterization of a stable cuboidal cell line from a neonatal rat mammary gland. The Rama 704 cell line shows
morphological and biochemical features of mammary epithelial cells and converts at high cell density to elongated cells that

The membrane protein carcinoembryonic antigen cell adhesion molecule (CEACAM6) is expressed in the epithelium of various tissues, participating in innate immune defense, cell proliferation and differentiation, with overexpression in gastrointestinal tract, pancreatic and lung tumors. It is developmentally and hormonally regulated in fetal human lung, with an apparent increased production in preterm infants with respiratory failure. To further examine the expression and cell localization of CEACAM6, we performed immunohistochemical and biochemical studies in lung specimens from infants with and without chronic lung disease. CEACAM6 protein and mRNA were increased ~4-fold in lungs from infants with chronic lung disease as compared with controls. By immunostaining, CEACAM6 expression was markedly increased in the lung parenchyma of infants and children with a variety of chronic lung disorders, localizing to hyperplastic epithelial cells with a ~7-fold elevated proliferative rate by PCNA staining. Some of these cells also co-expressed membrane markers of both type I and type II cells, which is not observed in normal postnatal lung, suggesting they are transitional epithelial cells. We suggest that CEACAM6 is both a marker of lung epithelial progenitor cells and a contributor to the proliferative response after injury due to its anti-apoptotic and cell adhesive properties. 相似文献

Summary The aim of this study was to develop immortalized cell lines from porcine uterus. Endometrial cells including luminal epithelium
(LE), glandular epithelium (GE), stroma (ST), and myometrium (MYO) were enzymatically isolated from the uterus of a day 12
pregnant gilt. Primary cultures were immortalized by transduction with a retroviral vector containing the E6 and E7 open reading
frames of human papillomavirus type 16 (LXSN-16E6E7) packaged by the amphotropic fibroblast line PA-317. Cells having integrated
the vector were selected by resistance to the neomycin analog G418 (0.4–1.5 mg/ml). Surviving cells were maintained in complete
culture medium containing G418 (0.1 mg/ml) and subcultured for 1 yr. Expression of the E7 protein was confirmed in all cell
lines by Western blotting. Phase contrast microscopy revealed that LE and GE cells exhibited cobblestone morphology, whereas
ST and MYO cells exhibited spindle-shaped morphology. The epithelial origin of LE and GE was confirmed by positive immunostaining
for cytokeratin. Stromal and MYO cells were vimentin-positive, but cytokeratin-negative. The MYO cell lines were positive
for smooth muscle α-actin staining, whereas LE, GE, and ST cell lines were negative for α-actin. Western blotting indicated
that all cell lines expressed both estrogen and progesterone receptors, but only GE cells secreted uteroferrin (UF). Collectively,
these porcine uterine cell lines provide an in vitro model for studying cell type-specific actions of hormones and cytokines,

Summary A culture system utilizing rat esophageal epithelial cells has been developed. Four normal and eightN-nitrosobenzylmethylamine-treated lines were compared with respect to chromosome number, anchorage-independent growth in agarose,
and tumorigenic potential in syngenic rats. All cell lines were aneuploid with nine in the near-tetraploid range and three
in the near-diploid range. No relation between tumorigenic potential and chromosome number or structure was apparent. Similarly,
anchorage-independent growth in agarose did not correlate with tumorigenic potential. Three of the 12 immortalized lines (two
carcinogen-treated and 1 untreated) induced well-differentiated squamous cell carcinomas in syngeneic rats. These tumors had
weak metastatic potentials suggesting that tumorigenic potential and metastatic ability are separately controlled. These cell
lines will be useful for the investigation of factors involved in the conversion of immortalized rat esophageal epithelial
cell lines to lines of high metastatic potential. 相似文献

Specific binding sites for angiotensin II were localized in the developing rat kidney (18th day of pregnancy and immediately before birth) by autoradiography using [125I]-ileu-5-angiotensin II either perfused in vivo through the fetal aorta or added in vitro to frozen sections in an incubation mixture. Specific binding was localized in the walls of the afferent and efferent arterioles, in the intraglomerular cells and in the peritubular arterioles of the subcapsular cortical zone. The immunohistochemical analysis, carried out on receptors saturated with unlabelled angiotensin II perfused through the mother's aorta, confirmed the autoradiographical localization. Antisera against ileu-5-angiotensin II were used in the indirect immunofluorescence technique and in the PAP method. Immunolocalization of angiotensin II was also found in the proximal tubule and in the thick ascending limb of Henle's loop. 相似文献

Summary A high frequency of diploid and near-diploid clones were developed from cell lines derived from adult and newborn rat liver
using micropipettes. There were some differences in morphology, biochemical properties and growth rate between clones. Cloned
cells had low levels of tyrosine transaminase activity, glucose-6-phosphatase activity and albumin content. A diploid clone
and a pseudodiploid clone derived from adult rat liver cell line were positive for α-fetoprotein.

Summary Cultured type II pneumocyte responses to in vitro normoxia (95% air: 5% CO2) or hyperoxia (95% O2:5% CO2) were quantified. Normoxic culture (0 to 96 h) of rabbit type II cells resulted in enhanced cell-monolayer protein and DNA
content. During this same time, cellular activities of superoxide dismutase (SOD), catalase, and glutathione peroxidase (GSH
Px) decreased. Compared to cultures maintained in normoxia, hyperoxic exposure of cultures resulted in decreased cell-associated
protein and DNA content. Exposure to hyperoxia also resulted in cytotoxicity as demonstrated by elevated cellular release
of DNA, lactate dehydrogenase (LDH), and preincorporated 8-[14C]adenine. Cellular catalase and GSH Px activities in hyperoxic cells decreased similarly to normoxic controls. In contrast,
cellular SOD activity in hyperoxic cells decreased less than in normoxic cultures. Cellular SOD activity in hyperoxic cultures,
when normalized for cellular protein, but not DNA, was greater than normoxic values after 24 to 96 h of exposure. Unlike the
decrease in cellular antioxidant enzymes during normoxic and hyperoxic culture, cellular LDH activity increased during both
these exposures. Cellular LDH activity in 24 to 96 h hyperoxia-exposed cells increased to a lesser extent than normoxic controls.
The extent of depression in LDH activity was dependent on whether the activity was normalized for cellular protein or DNA.
Type II pneumocytes, which normally undergo hyperplasia and hypertrophy during hyperoxia in vivo, exhibited oxygen sensitivity
in vitro. Exposure of type II cells to hyperoxia in vitro resulted in alterations in cellular SOD and LDH activities, but
recognition of such changes were dependent on whether enzymatic activities were normalized for cellular DNA or protein.

Summary A number of diploid clones have been isolated from an enzymatic dispersion of normal rat lung. Four of these clones are epithelial
in morphology, the remainder fibroblastic. On the basis of electron microscopic observations, two of the epithelial clones
appear to have originated from type II alveolar pneumonocytes.

Summary Alterations in pulmonary surfactant are partly responsible for the respiratory insufficiency seen under septic shock process. We have used an experimental model of LPS-induced shock in rats to examine the cells responsible for the pulmonary surfactant synthesis and its relationship to lung injury. (14C)Choline incorporation into phosphatidylcholine was significantly reduced in lung homogenates or type II cells obtained from LPS-treated animals. Addition of LPS in vitro fails to increase (14C)choline incorporation in type II cells obtained from LPS-treated animals. We suggest that this depression of pulmonary phosphatidylcholine synthesis may partly explain the occurrence of respiratory failure with septic shock. 相似文献

Summary Two mixed cell lines designated REN-1 and REN-2 were established successfully in continuous in vitro culture from subcutaneously
propagated transplant tissue derived from a nephroblastoma which had occurred spontaneously in an Nb hooded rat. In monolayer
culture the lines consisted of clumps, islands, and cords of densely crowded, small basophilic cells of epithelioid character,
together with mesenchymelike cells occupying the intervening spaces. The proportion of epithelioid cells to mesenchyme could
be enhanced by high seeding densities and the intermittent application ofcishydroxyproline to the medium. A third cell form with the appearance of more mature epithelium was observed as a later development
in one of the monolayer cultures (REN-1). This larger epithelial cell and a homogeneous mesenchymal population were isolated
as cloned cell lines from REN-1 (REN-1-C/2 and REN-1-C/1, respectively), but attempts to clone the dominant basophilic epithelioid
cell were not successful. Light and electron microscopy indicated the small, basophilic epithelioid cells to be morphologically
consistent with the undifferentiated embryonal blast cells of the parent tumor. They were a distinctive population unlike
known malignant cell lines representative of chemically transformed rat kidney mesenchyme and epithelium. The mesenchymelike
cells present in the uncloned cell lines, and cloned in REN-1-C/1, were fibroblasts by ultrastructural criteria and therefore
distinct from the epithelioid moiety. Ultrastructurally the larger epithelium cloned in REN-1-C/2 displayed the features of
differentiated renal epithelium. Subcutaneous transplantation in syngeneic and allogeneic recipients showed the uncloned parent
cell lines containing the basophilic epithelioid population to be highly tumorigenic, producing rapidly growing tumors that
were unequivocal nephoblastomas. The mesenchymal clone, REN-1-C/1, was also tumorigenic but, consistent with its fibroblastic
nature, produced only fibrosarcomas on transplantation. The mature epithelial clone, REN-1-C/2, transplanted as an anaplastic
carcinoma with limited tubule formation. Because of their distinctive morphology and growth behavior, and their ability to
proliferate into nephroblastomas on transplantation, the dominant population of basophilic epithelioid cells in the parent
uncloned cell lines is considered to represent neoplastic kidney cells of undifferentiated embryonal type. The possible host
origin of the mesenchymal population from the supporting stroma of the original transplantation tumor is suggested in discussion.
