1. The nucleic acid metabolism in the pyridoxine-deficient rat has been investigated through studies on the incorporation of radioactivity from various isotopically labelled compounds into liver and spleen DNA and RNA. 2. In pyridoxine deficiency, the incorporation of radioactivity from sodium [14C]formate was apparently increased. The magnitude of this effect on incorporation into liver RNA and DNA and spleen RNA was approximately the same. The incorporation into spleen DNA was enhanced to a much greater degree. Administration of pyridoxine 24hr. before the rats were killed reversed the changes in incorporation of radioactivity from [14C]formate. 3. In pyridoxine deficiency, the incorporation of radioactivity from dl-[3-14C]serine, [8-14C]adenine, [Me-3H]thymidine and [2-14C]deoxyuridine was decreased. The incorporation of radioactivity from l-[Me-14C]methionine was not affected. No noteworthy differences in the effect of pyridoxine deficiency on the incorporation of radioactivity from dl-[3-14C]serine into DNA and RNA were observed, whereas the effect of the deficiency on the incorporation of radioactivity from [8-14C]adenine into spleen DNA was somewhat greater than that into spleen RNA. Administration of pyridoxine 24hr. before the rats were killed reversed the changes in incorporation of radioactivity from [3-14C]serine and [8-14C]adenine. 4. The adverse effects of pyridoxine deficiency on the biosynthesis of nucleic acids and cell multiplication are discussed in relation to the role of pyridoxal phosphate in the production of C1 units via the serine-hydroxymethylase reaction.  相似文献

The formation of polysomes in relation to RNA synthesis was investigated in tobacco (Nicotiana tabacum L.) pollen cultivated submersely for a period of 12 h. The percentage of polysomes was estimated by determining the number of ribosomes carrying nascent polypeptides using RNase and 0.8 M KC1 treatment of the ribosomal preparation. This approach showed that the proportion of ribosomes “active” in protein synthesis amounts to about 12 % in dry pollen rising to 46 % within 10 min of imbibition and to 66 % during the period 1–4 h of cultivation. The latter increase is accompanied by a rapid incorporation of uridine-14C into polysome-as-sociated RNA and is sensitive to actinomycin D. The rapidly labelled RNA isolated from the ribosomal preparation sedirnented in the range from about 5S to 30S. Longer labelling periods led to a gradual shift of the major peak of this heterogeneous RNA from about 11.5S and 14S to about 7.5S and the development of radioactivity peaks in the position of 18S and 28S RNA. Uridine incorporated both into the active ribosomes and into the subunits of inactive ribosomes at a more or less constant rate during the whole 12-h period of cultivation. These results present evidence that in addition to the initial combination of the existing ribosomes with the stored mRNA following imbibition, an activation of pollen tube genome and its implication in directing protein synthesis take place during the early phases of pollen tube growth. From the results on kinetics of labelling of ribosomes it appears that in pollen tubes new synthesized ribosomal subunits enter polysomes directly, the entry of 40S subunits being more rapid than that of 60S subunits.  相似文献

The RNA formed in pollen tubes during 4 hours of growthin vitro was resolved by chromatography on methylated albumine on kieselguhr (MAK) into three principal fractions. Acoording to the labelling from uracil-14C about 11% was eluted with tRNA and 5 S RNA (low molecular weight RNA), 76% just after rRNA (D-RNA) and nearly 14% was recovered from the column by SDS at 35 °C (TB-RNA). In the presence of actinomycin D at concentration of 30 μg ml-1 the synthesis of the three classes of RNA was inhibited by 71%, 97% and 70% respectively. On sucrose density gradient the radioactive low molecular weight RNA sedimented at 4 S-5 S which suggests that one or both of these RNA species are synthesized in pollen tubes. The D-RNA eluted from the MAK column is polydisperse in size exhibiting a wide range of sedimentation values up to about 35 S with a large peak at 9 S-10 S and two smaller peaks at 14 S-15 S and at about 23 S. The rapid labelling and the polydisperse rather low molecular weight character suggest that the D-RNA is a heterogeneous population of mRNA. The sedimentation profile of TB-RNA was similar to that of D-RNA. The RNA synthesized in the presence of32PBO3-4 or uracil-14C exhibited no radioactivity peaks corresponding to sedimentation peaks of rRNA.  相似文献

Summary The labelling of nucleic acids of growing cells of the blue-green algae Anacystis nidulans and Synechocystis aquatilis by radioactive precursors has been studies. A. nidulans cells most actively incorporate radioactivity from [2-14C]uracil into both RNA and DNA, while S. aquatilis cells incorporate most effectively [2-14C]uracil and [2-14C]thymine.Deoxyadenosine does not affect incorporation of label from [2-14C]thymidine into DNA, but weakly inhibits [2-14C]thymine incorporation into both nucleic acids and significantly suppresses the incorporation of [2-14C]uracil.The radioactivity from [2-14C]uracil and [2-14C]thymine is found in RNA uracil and cytosine and DNA thymine and cytosine. The radioactivity of [2-14C]thymidine is incorporated into DNA thymine and cytosine. These results and data of comparative studies of nucleic acid labelling by [2-14C]thymine and [5-methyl-14C]thymine suggest that the incorporation of thymine and thymidine into nucleic acids of A. nidulans and S. aquatilis is accompanied by demethylation of these precursors. In this respect blue-green algae resemble fungi and certain green algae.  相似文献

  1. The incorporation of mevalonate-2-14C into ipomeamarone in sweet potato root tissue infected by Ceratocystis fimbriata was demonstrated, but the rate was low when compared with acetate-2-14C. No dilution effect of mevalonate was noted during the incorporation of acetate-2-14C into ipomeamarone. This is very likely to result from the passive transfer of mevalonate into the cells.

  2. No dilution effect of acetate during the incorporation of mevalonate-2-14C into ipomeamarone was noted. This indicates that mevalonate is not incorporated into ipomeamarone after its conversion to acetate.

  3. Evidence for incorporation of acetate-2-14C into mevalonate was shown by the fact that the specific radioactivity of mevalonic acid benzhydrylamide was not lowered throughout repetitive crystallizations. These data also support the participation of mevalonate in ipomeamarone synthesis as an intermediate.

1. Male rats were injected intraperitoneally with l-[35S]methionine, [32P]-phosphate and [2-14C]acetate. The animals were killed at various times up to 72hr. after injection, and liver mitochondria were prepared and fractionated into soluble protein, insoluble protein and lipid for assay of the radioactivity of each fraction. 2. The maximal specific radioactivity of total mitochondrial phospholipid with respect to both 32P and 14C was attained after approx. 6hr. 3. 32P was incorporated most rapidly into phosphatidylethanolamine, maximal incorporation being attained after approx. 6hr.; maximal incorporation into lecithin occurred after 6–12hr. The specific radioactivity of cardiolipin was still slowly increasing at the end of the experiment (72hr.). 4. There were no major differences between the rates of incorporation of 14C into the lecithin, phosphatidylethanolamine and cardiolipin fractions of mitochondrial phospholipid, maximal incorporation in each case occurring after approx. 6hr. 5. Maximal incorporation of 35S into both soluble and insoluble protein fractions was attained less than 12hr. after injection, the maximal specific radioactivity of soluble protein being higher than that of insoluble protein.  相似文献

In the present paper RNA from apple-tree callus tissue labelled with 6-benzyl-minopurine14) was studied. The RNA isolated from this tissue was prepared as sample for electron microscopic studies and was also used as biochemical control. The electron microscopic autoradiograms obtained showed the labelled structure of RNA, sRNA and rRNA. The incorporation of labelled purine rings was confirmed in all three RNA types by the radioactivity, which was also proved in nucleotides after hydrolysation of RNA fractions. The results were compared with RNA isolated from tissue cultivated on a non-radiactive medium.  相似文献

Lipogenesis in the fatty liver of rat which was induced by feeding an amino acid-irnbalanced diet containing 8% casein supplemented with 0.3% dl-methionine has been investigated by measuring the incorporation of acetate-1-14C into various lipid fractions during in vitro incubation of liver slices.

In the incorporation of acetate-1-14C into the total lipid per one g of the slices, no significant difference for the imbalance group was observed. However, the total radioactivity of liver lipid per 100 g of the body and the incorporation into triglyceride in the lipids were significantly higher in the imbalance group than in the control group. Conversion of acetate-1-14C to CO2 was not impaired in the imbalance group.

It is evident from these results that the induction of this type of fatty liver is due mainly to the synthesis of triglyceride.  相似文献

The incorporation of radioactivity from intracisternally injected (6−14C) orotic acid into cerebral RNA and precursor pool was markedly elevated in 25 day-old hypothyroid rats compared to controls. Similar results were achieved with (2−14C) uridine as the isotopic precursor. The increase in specific radioactivity of cerebral ribosomal RNA and the purified nucleotide precursor pool was essentially restored to normal by a single injection of L-thyroxine at 22 days of age (72 hours prior to sacrifice). The results are compatible with an increased intracellular uptake (transport) of RNA precursors and reflect the participation of thyroid hormones in the regulation of events at the membrane during brain development.  相似文献

Lipogenesis in the fatty liver of rat, which was induced by feeding an amino acid unbalanced diet containing 8% casein supplemented with 0.3% dl-methionine, has been studied by measuring the incorporation of glycerol-1-14C, palmitate-1-14C, citrate-1,5-14C, pyruvate-1-14C and pyruvate-2-14C into various lipid fractions and 14CO2 during in vitro incubation of liver slices.

The total radioactivity of liver lipid per 100 g of the body and the incorporation of each substrate into triglyceride in the lipid were significantly higher in the imbalance group than the control group. Conversion of each substrate to 14CO2 was not imparied in the imbalance group.

It is evident from these results that the induction of this type of fatty liver is due mainly to the triglyceride synthesis by both the fatty acid synthesis and the transesterification of fatty acid.

These results are considered to support the previous assumption in which acetate-1-14C was used as a precursor.  相似文献

Synthesis of neutral and polar lipids in pollen of Tradescantia paludosa during germination and tube growth was studied by the incorporation of acetate-[1-14C] into lipids in the presence and absence of inhibitors of RNA and protein synthesis. The proteins required for the synthesis of both neutral lipids and phospholipids are not made de novo during germination but are already present in the mature ungerminated pollen grain and they are functionally stable during the first 2 hr of pollen growth.  相似文献

[1-14C]-Acetate incorporation into total and polar lipids was studied in the growing pollen tubes of Crotalaria juncea. Ungerminated pollen had phosphatidyl inositol, phosphatidyl serine, phosphatidyl choline, phosphatidyl ethanolamine, phosphatidyl glycerol, monogalactosyl diglyceride, digalactosyl diglyceride, sulpholipid and steryl glycosides. In the growing pollen tubes considerable [1-14C]-acetate incorporation was observed into the individual polar lipids. The exogenous carbon source significantly influenced lipid biosynthesis. Boric acid (20mg/l.) promoted both pollen tube growth and acetate incorporation into phospholipids. In comparison to 5′-adenosine monophosphate, cyclic-3′,5′-adenosine monophosphate (cAMP) promoted tube growth and also enhanced phospho-and glycolipid biosynthesis. The regulation of membrane component biosynthesis by cAMP is suggested.  相似文献

The incorporation of the radioactivity from acetate-1-14C, acetate-2-14C, dl-methionine-1-14C, dl-methionine-2-14C, dl-methionine-3,4-14C, dl-homomethionine-2-14C, dl-allyl-glycine-2-14C, and dl-2-amino-5-hydroxyvalerate-2-14C into the aglycones of progoitrin, gluconapin, and glucobrassicanapin of maturing rape plants (Brassica campestris L.) was investigated. Radioactivity from dl-methionine-2-14C, dl-methionine-3,4-14C, dl-homomethionine-2-14C, and acetate-2-14C were incorporated into the 3 major thioglucosides. The other organic compounds were poorly incorporated except for dl-allylglycine-2-14C into glucobrassicanapin. The results obtained suggest that the rape plant can synthesize amino acids by the condensation of acetate (as acetyl CoA) to α-keto acids to yield a homologue of the original amino acid. These newly formed amino acids are then employed to synthesize the 3 major thioglucosides.  相似文献

Concentrations of cycloheximide as low as 3 μg/ml inhibited incorporation of labeled orotic acid or uridine into RNA cytidylic acid of soybean (Glycine max) hypocotyl sections. Even lower concentrations of this well known protein synthesis inhibitor interfered with conversion of labeled cytidine into RNA uridylic acid. Both cycloheximide and puromycin inhibited absorption of 3H-phenylalanine and its incorporation into protein, but puromycin did not significantly affect the labeling patterns of RNA cytidylic and uridylic acids when orotic acid-6-14C was fed. Results give further support to the hypothesis that cycloheximide inhibits the interconversion of uridine and cytidine nucleotides, presumably by acting as a glutamine antagonist in the glutamine-dependent reaction catalyzed by cytidine triphosphate synthetase.  相似文献

The structure and biological activity (the level of the labelled precursor incorporation into RNA) of active and repressed chromatin of the liver and small intestine mucosa were studied in adult (6-8 months) and old (24-26 months) rats. The content of repressed chromatin fraction in both tissues is found to increase with age. In the liver of old rats the level of [14C[ orotic acid incorporation into RNA of chromatin fractions decreases, radioactivity of the acid-soluble fraction being unchanged. In the small intestine mucosa a high leve of [14C] orotic acid incorporation into chromatin RNA with ageing is due to an increase in permeability of the mucosa cells.  相似文献

Lipid metabolism of the subarctic moss Dicranum elongatum was studied by feeding the moss with 2-14C-acetate and, after extraction of the lipids, counting the 14C-content of different lipid fractions immediately after feeding or after chase periods. Translocation of 14C after 14C-feeding was studied with autoradiography. Both low temperature (+6°C) and drought (at +23°C) resulted in increased incorporation of 14C into the neutral lipid (NL) fraction and decreased incorporation of 14C into the glycolipid (GL) fraction of the green shoot part of the moss. The distribution of radioactivity between the NL classes suggests that diacylglycerols (1, 2-DAG) and common triacylglycerols (cTAG) are turned into acetylenic triacylglycerols (aTAG), which are accumulated preferentially. The decrease in the radioactivity of the GL fraction was due to two unknown fractions, whereas 14C incorporation into the chloroplast membrane lipids, monogalactosyl diacylglycerol (MGDG) and digalactosyl diacylglycerol (DGDG), was very low throughout the experiments. The phospho-lipid (PL) fraction accounted for 48–63% of total lipid radioactivity at both low and high temperatures. 2-14C-acetate feeding to the senescent moss part resulted in vigorous 14C incorporation into the lipids, especially into the reserve TAGs. Electron microscopic examination showed the presence of plastids, which explains the capability of the senescent part of the moss for lipid synthesis. The fact that transport of 14C from 2-14C-acetate took place upwards and downwards in the moss shoot, together with the capability for lipid synthesis of the senescent moss part, supports the suggestion that the senescent moss part plays a role as an energy store.  相似文献

—The origin of the acetyl group in acetyl-CoA which is used for the synthesis of ACh in the brain and the relationship of the cholinergic nerve endings to the biochemically defined cerebral compartments of the Krebs cycle intermediates and amino acids were studied by comparing the transfer of radioactivity from intracisternally injected labelled precursors into the acetyl moiety of ACh, glutamate, glutamine, ‘citrate’(= citrate +cis-aconitate + isocitrate), and lipids in the brain of rats. The substrates used for injections were [1-14C]acetate, [2-14C]acetate, [4-14C]acetoacetate, [1-14C]butyrate, [1, 5-14C]citrate, [2-14C]glucose, [5-14C]glutamate, 3-hydroxy[3-14C]butyrate, [2-14C]lactate, [U-14C]leucine, [2-14C]pyruvate and [3H]acetylaspartate. The highest specific radioactivity of the acetyl group of ACh was observed 4 min after the injection of [2-14C]pyruvate. The contribution of pyruvate, lactate and glucose to the biosynthesis of ACh is considerably higher than the contribution of acetoacetate, 3-hydroxybutyrate and acetate; that of citrate and leucine is very low. No incorporation of label from [5-14C]glutamate into ACh was observed. Pyruvate appears to be the most important precursor of the acetyl group of ACh. The incorporation of label from [1, 5-14C]citrate into ACh was very low although citrate did enter the cells, was metabolized rapidly, did not interfere with the metabolism of ACh and the distribution of radioactivity from it in subcellular fractions of the brain was exactly the same as from [2-14C]pyruvate. It appears unlikely that citrate, glutamate or acetate act as transporters of intramitochondrially generated acetyl groups for the biosynthesis of ACh. Carnitine increased the incorporation of label from [1-14C]acetate into brain lipids and lowered its incorporation into ACh. Differences in the degree of labelling which various radioactive precursors produce in brain glutamine as compared to glutamate, previously described after intravenous, intra-arterial, or intraperitoneal administration, were confirmed using direct administration into the cerebrospinal fluid. Specific radioactivities of brain glutamine were higher than those of glutamate after injections of [1-14C]acetate, [2-14C]acetate, [1-14C]butyrate, [1,5-14C]citrate, [3H]acetylaspartate, [U-14C]leucine, and also after [2-14C]pyruvate and [4-14C]acetoacetate. The intracisternal route possibly favours the entry of substrates into the glutamine-synthesizing (‘small’) compartment. Increasing the amount of injected [2-14C]pyruvate lowered the glutamine/glutamate specific radioactivity ratio. The incorporation of 14C from [1-14C]acetate into brain lipids was several times higher than that from other compounds. By the extent of incorporation into brain lipids the substrates formed four groups: acetate > butyrate, acetoacetate, 3-hydroxybutyrate, citrate > pyruvate, lactate, acetylaspartate > glucose, glutamate. The ratios of specific radioactivity of ‘citrate’ over that of ACh and of glutamine over that of ACh were significantly higher after the administration of [1-14C]acetate than after [2-14C]pyruvate. The results indicate that the [1-14C]acetyl-CoA arising from [1-14C]acetate does not enter the same pool as the [1-14C]acetyl-CoA arising from [2-14C]pyruvate, and that the cholinergic nerve endings do not form a part of the acetate-utilizing and glutamine-synthesizing (‘small’) metabolic compartment in the brain. The distribution of radioactivity in subcellular fractions of the brain after the injection of [1-14C]acetate was different from that after [1, 5-14C]citrate. This suggests that [1-14C]acetate and [1, 5-14C]citrate are utilized in different subdivisions of the ‘;small’ compartment.  相似文献
