The analysis of growth and movements of seedling organs of kidney bean (Phaseolus vulgaris L.) provides a pattern of periodic phases of activity and relaxation. The existence of a central organ which would control the phase relationships, is not anticipated in the integrity of the plant. The cyclic activity of individual organs shows itself by growth associated with oscillation movements. One and the same organ may simultaneously accomplish oscillatory movements with a diurnal and ultradiurnal frequency. These rhythms originate during the organ development; the first pair of kidney bean leaves at first executes oscillation movements with a diurnal frequency and only after it is fully developed it exhibits a diurnal cycle with the photophil phase upwards and the scotophil downwards, the oscillations with an ultradiurnal oycle being maintained. The movements of the two leaves are synchronous, but there occur short sections with a desynchronous cycle. Simultaneously with these oscillations, in which the leaf petiole takes part, the adult leaf performs oscillatory movements perpendicular to the longitudinal leaf axis, the so-called side swings, controlled by periodical changes of the joint attaching the leaf blade. Their frequency is practically identical with that of the ultradiurnal cycle. Thus the periodic growth activity of the kidney bean results in growth oscillations passing in the diurnal cycle with a frequency of 0.043 rev.h-1, their ascending and descending phases consisting of periodical ultradiurnal oscillations in cycles of 0.73–0.59 rev.h-1. The epicotyl growth shows a similar pattern: into the basic diurnal nutation cycle with a frequency of 0.042 rev.h-1 ultradiurnal oscillation cycles are incorporated having a similar frequency to that revealed in leaves (0.69–0.64 rev.h-1). The diurnal oscillatory cycles belong to a system established on the basis of periodicity of day and night and other geophysical cycles. The ultradiurnal rhythmic oscillations are presumed to be an expression of the geocontrol system of root and shoot growth direction and orientation of the organ in space. The shape of their trajectories in bean leaves is contradictory to this; they are not spatial helices, as the kybernetic model would presuppose, but have a vertical, upwards and downwards course in one plane. Since these oscillatory movements with an ultradiurnal cycle cease after petiole excision from the stem and after shoot apex amputation, one may presume that they are coupled with the low-frequency oscillatory system of the epicotyl.  相似文献

Portions of the shoot system from young light-grown pea (Pisum sativum L.) seedlings were excised and circumnutation studied using time-lapse cinematography. Removal of the youngest leaf or shoot tip as well as ringing the stem with 20 mM triiodobenzoic acid severely restricted circumnutation. Treating the stump of the excised leaf with lanolin containing 10?4 M indole-3-acetic acid or replacing the leaf with an artificial aluminum leaf both partially restored circumnutation. When the leaf was replaced with both auxin and an artificial leaf circumnutation continued at approximately the rate of the intact plant. This graphically shows the involvement of both auxin and gravitropism in circumnutation.  相似文献

The analysis of diurnal oscillatory movements of tobacco leaves was used in the diagnosis of viral infection of plants. The oscillatory helices circumscribed by a growing leaf of a healthy plant were regular, but some deviations, particularly in the transition points, were recorded. In order to make clear the cause of these irregularities of trajectory, the course of elongation of leaf petiole and blade in relation to localization and shift of zones of elongation during ontogenesis was analysed. The present analysis is similar to that described by the author's earlier experiments with pea roots. Oscillatory curves circumscribed by petiole, projected on a horizontal plane, were compared with curves circumscribed by the blade tip. The analysis of the leaves of different ages enabled us to study this process in dependence on growth rate. It was confirmed that oscillations are a result of elongation; the extent of oscillations is quantitatively dependent on the growth rate. An analysis of the zones of growth showed that in petiole the active meristems are localized near to its base while in the leaf lamina they move gradually during the ontogenesis from the apical to the basal part of the leaf blade. Active meristems of young rapidly growing leaves are localized approximately in the middle of the blade while those of old leaves were found in close proximity to the base of the lamina. The growth rate of petiole can be expressed in hundreds of mm per hour (4.8×10?2 mm h?1); half of this value was recorded for its apical part. The growth rate of leaf blade was found approximately ten times higher (3.2×10?1 mm h?1). The oscillatory movements of growing leaf consists of two integrate components: of oscillations originating in the base of the petiole and of oscillations of leaf blade the centrum of which is localized in the basal third of the blade. The arrangement of the experiments did not enable us to determine to what extent the phototropic response of leaf blade participates in leaf movements. The movements of leaves of an intact plant are evidently affected by rhythmic stem oscillations. Stem is an integral part of a system which participates in the transfer of information in plants.  相似文献

In plants, an electrical potential and circumnutation disturbances are a part of a response to environmental and internal stimuli. Precise relations between electrical potential changes and circumnutation mechanisms are unclear. We have found recently that glutamate (Glu) injection into Helianthus annuus stem induced a series of action potentials (APs) and a transient decrease in circumnutation activity. A theoretical explanation for this finding is discussed here taking into considerations data about the ion mechanism of AP and circumnutation as well as about the metabolic and signaling pathways of glutamate and their possible interactions.Key words: action potential, circumnutation, elongation, glutamate, Helianthus annuus, plant movement  相似文献

We determined leaf and stem strength for Arundo donax from plants grown in different geographic areas and at different times within their growing cycle. Mean leaf strength for plants collected within California was 1.72 Newtons (N) and ranged from 0.36 to 6.32 N, based on 1170 individual determinations. For leaves collected from 30 plants within four states between July 11 and 20, 2007, mean leaf strength was 1.58 N based on 936 determinations. Values ranged from 0.24 to 4.90 N. Overall, leaf strength showed statistically significant changes depending on the sampling date, sampling location, type of leaf sampled, and position within the leaf where the measurement was taken. In general leaf strength was greater near the base of the leaf and decreased with distance away from the base; leaf strength changed as the growing season progressed; and first year leaves had leaf strength values about 25% greater than leaves produced on stems >1-year old. This represents two of the three age categories of leaves which may be present on giant reed at any one time. Stem strength and stem wall thickness were strongly correlated (Kendall's Tau b = 0.92, P < 0.0001, N = 26). Linear regression indicated that mean stem strength decreased by approximately 6.8% (95% confidence limits 5.8-7.7%) from one node to the successive node progressing from the stem base to the shoot tip. These results imply that the ability of biological control agents to damage A. donax leaves may not be the same across the locations this plant occurs or at all times during the growing season within a given location.  相似文献

The growth and circumnutation of the stem of three-week old Helianthus annuus in the 16:8 h light:dark photoperiod were monitored using an angular position-sensing transducer and a time lapse photography system. It was found that the rate of growth and circumnutation reached a high level in the dark stage; in the light stage, however, only the growth rate reached the same high level, whereas the circumnutations were weak. These results showed that in the light stage the stem circumnutation was downregulated more strongly than the growth. Short-term stem responses to darkening and illumination were a further display of the relation between growth and circumnutations. Switching off the light caused an increase in the growth and circumnutation rate. In some cases it was accompanied by changes in the rotation direction. On the other hand, switching the light on caused an immediate transient (several-minute long) decrease in the growth rate resulting in stem contraction, and this was accompanied by an almost complete pause of circumnutation. Additionally, under light, there occurred a subsequent decrease in the magnitude, disturbance of circumnutation trajectory and, in some cases, changes in the direction of rotation. The observed stem contraction and disturbance of circumnutation imply the occurrence of turgor changes in sunflower stem, which may be caused by a non-wounding, darkening or illumination stimulus. Our experiments indicate that the disturbances of the growth rate are accompanied by changes in circumnutation parameters but we have also seen that there is no simple quantitative relation between growth rate and circumnutation rate.Key words: Helianthus annuus, plant movement, circumnutation, elongation, growth, stem contraction  相似文献

Circumnutation is a helical organ movement widespread among plants. It is variable due to a different magnitude of trajectory (amplitude) outlined by the organ tip, duration of one cycle (period), circular, elliptical, pendulum-like or irregular shape and clock- and counterclockwise direction of rotation. Some of those movement parameters are regulated by circadian clock and show daily and infradian rhythms. Circumnutation is influenced by light, temperature, chemicals and can depend on organ morphology. The diversity of this phenomenon is easier to see now that the digital time-lapse video method is developing fast. Whether circumnutation is an endogenous action, a reaction to exogenous stimuli or has a combined character has been discussed for a long time. Similarly, the relationship between growth and circumnutation is still unclear. In the mechanism of circumnutation, epidermal and endodermal cells as well as plasmodesmata, plasma membrane, ions (Ca2+, K+ and Cl), ion channels and the proton pump (H+ATPase) are engaged. Based on these data, the hypothetical electrophysiological model of the circumnutation mechanism has been proposed here. Circumnutation is a plant growth movement in which the tips of axial organs draw a circular orbit. Although it has been studied since the nineteenth century, its mechanism and significance are still unclear. Greened adzuki bean (Vigna angularis) epicotyls exhibited a clockwise circumnutation in the top view with a constant period of 60 min under continuous white light. The bending zone of circumnutation on the epicotyls was always located in the region 1–3 cm below the tip, and its basal end was almost identical to the apical end of the region where the epicotyl had completely elongated. Therefore, epidermal cells that construct the bending zone are constantly turning over with their elongation growth. Since exogenously applied auxin transport inhibitors and indole-3-acetic acid (IAA) impaired circumnutation without any effect on the elongation rate of epicotyls, we attempted to identify the distribution pattern of endogenous auxin. Taking advantage of its large size, we separated the bending zone of epicotyls into two halves along the longitudinal axis, either convex/concave pairs in the plane of curvature of circumnutation or pre-convex/pre-concave pairs perpendicular to the plane. By liquid chromatography–mass spectrometry, we found, for the first time, that IAA and gibberellin A1 were asymmetrically distributed in the pre-convex part in the region 1–2 cm below the tip. This region of epicotyl sections exhibited the highest responsiveness to exogenously applied hormones, and the latent period between the hormone application and the detection of a significant enhancement in elongation was 15 min. Our results suggest that circumnutation in adzuki bean epicotyls with a 60 min period is maintained by differential growth in the bending zone, which reflects the hormonal status 15 min before and which is shifting sequentially in a circumferential direction. Cortical microtubules do not seem to be involved in this regulation.  相似文献

In compound leaves, leaflet primordia are initiated directionally along the lateral sides. Our understanding of the molecular basis of leaflet initiation has improved, but the regulatory mechanisms underlying spatio-temporal patterns remain unclear. In this study, we investigated the mechanisms of acropetal (from the base to the tip) progression of leaflet initiation in Eschscholzia californica. We established an ultraviolet-laser ablation system to manipulate compound-leaf development. Local ablation at the leaflet incipient site generated leaves with asymmetric morphology. In the majority of cases, leaflets that were initiated on the ablated sides shifted apically. Finite time-course observation revealed that the timing of leaflet initiation was delayed, but the distance from the leaf tip did not decrease. These results were suggestive of the local spacing mechanism in leaflet initiation, whereby the distance from the leaf tip and adjacent pre-existing leaflet determines the position of leaflet initiation. To understand how such a local patterning mechanism generates a global pattern of successive leaflet initiation, we assessed the growth rate gradient along the apical–basal axis. Our time-course analysis revealed differential growth rates along the apical–basal axis of the leaf, which can explain the acropetal progression of leaflet initiation. We propose that a leaflet is initiated at a site where the distances from pre-existing leaflets and the leaf tip are sufficient. Furthermore, the differential growth rate may be a developmental factor underlying the directionality of leaflet initiation.  相似文献

The principal objective of the research reported here was to determine whether a plant's periodic growth oscillations, called circumnutations, would persist in the absence of a significant gravitational or inertial force. The definitive experiment was made possible by access to the condition of protracted near weightlessness in an earth satellite. The experiment, performed during the first flight of Spacelab on the National Aeronautics and Space Administration shuttle, Columbia, in November and December, 1983, tested a biophysical model, proposed in 1967, that might account for circumnutation as a gravity-dependent growth response. However, circumnutations were observed in microgravity. They continued for many hours without stimulation by a significant g-force. Therefore, neither a gravitational nor an inertial g-force was an absolute requirement for initation or continuation of circumnutation. On average, circumnutation was significantly more vigorous in satellite orbit than on earth-based clinostats. Therefore, at least for sunflower (Helianthus annuus L.) circumnutation, clinostatting is not the functional equivalent of weightlessness.  相似文献

Three types of rhythmic movements of Phaseolus vulgaris L. (pole beans) were examined collectively and their characteristics compared. Although the ultradian rhythms of shoot circumnutation and leaf movement, as well as the circadian rhythm of leaf movement, occurred simultaneously, each rhythm could be expressed independently of the other two. Shoot circumnutation and ultradian leaf movements displayed the same period (80 min at 25°C and Q10⋍2), while the period of the circadian leaf movements was not temperature dependent (Q10⋍1). Interaction into the plant between two ultradian rhythms (shoot circumnutation and ultradian leaf movement) with the same period and coexistence in the pulvinus of an ultradian with a circadian rhythm are discussed.  相似文献

Abstract Membrane potentials (pd's) of epidermal bladder cells and green leaf cells of Mesembryanthemum crystallinum L. are rather low (between ?10 and ?40 mV). During growth on 400 mM NaCl membrane potentials decrease further. As shown previously, plants grown on 400 mM NaCl show the diurnal changes of malate levels typical for plants having crassulacean acid metabolism. Therefore, in this study membrane potentials were measured at different times of the day, but no diurnal variations of pd were correlated with diurnal oscillations of malate levels. Resting potentials are similar in bladder cells and in green leaf cells and are similar in continuous light and darkness. Both bladder and leaf cells display light-triggered photosynthesis-dependent oscillations of the membrane potential although the bladder cells do not appear to be photosynthetically very active. This suggests electrical coupling between the bladder cells and the underlying green cells. Over a larger distance (2 mm) in the leaves, however, there is no direct evidence for electrical coupling. Cl?, Na+, and K+ concentrations are similar in bladder cells and in the photosynthetically active tissue of leaves and stems. Bladder cells appear to contain high concentrations of free oxalate. The present findings corroborate earlier conclusions that the epidermal bladder cells of M. crystallinum function as peripheral water reservoirs providing protection from short term water stress.  相似文献

Quantitative histochemical methods were used to measure glyceraldehyde-3-phosphate dehydrogenase-NADP activity in developing and mature leaves of Dianthus chinensis L. The activity values were compared on the basis of illumination of developing leaves, the maturity of stomata, and the activity of enzymes concerned with carbohydrate oxidation. Activity on a kilogram dry weight per hour basis increased from the youngest primordia (50 mmol) to fully expanded leaves (1900 mmol) and then declined in older, senescing leaves. Enzymic activity increased from leaf base (400 mmol) to tip (1000 mmol) in leaf pair four, the tip of which was exposed to light, while activity of enzymes of carbohydrate oxidation decline. We propose that this leaf is beginning the transition to autotrophy, but does not fix significant amounts of atmospheric carbon because the stomata are not functional until leaf pair six.  相似文献

Cottonwood (Populus deltoides, Marsh.) leaves are amphistomatous and have an adaptation in their petiole which allows them to oscillate in wind. A possible function of these oscillations in enhancing gas exchange was studied.

Cottonwood leaves were found to oscillate in the presence of wind velocities frequently encountered in nature. A pressure differential across the leaf was shown to result in bulk flow of air through that leaf. Oscillating a cottonwood leaf at frequencies found to occur in nature was found to increase the rate of O2 flux through the leaf. The measured changes in boundary layer resistances during oscillations were found to be insufficient to account for the increased O2 flux. Thus, the bulk flow of air through an oscillating cottonwood leaf results in a decreased total resistance which is typically 25% less than that of a still leaf.

The diurnal variations in volume and in specific weight were determined for green stems and leaves of Crassulacen acid metabolism (CAM) plants. Volume changes were measured by a water displacement method. Diurnal variations occurred in the volume of green CAM tissues. Their volume increased early in the light period reaching a maximum about mid-day, then the volume decreased to a minimum near midnight. The maximum volume increase each day was about 2.7% of the total volume. Control leaves of C3 and C4 plants exhibited reverse diurnal volume changes of 0.2 to 0.4%. The hypothesis is presented and supported that green CAM tissues should exhibit a diurnal increase in volume due to the increase of internal gas pressure from CO2 and O2 when their stomata are closed. Conversely, the volume should decrease when the gas pressure is decreased.

The second hypothesis presented and supported was that the specific weight (milligrams of dry weight per square centimeter of green surface area) of green CAM tissues should increase at night due to the net fixation of CO2. Green CAM tissues increased their specific weight at night in contrast to control C3 and C4 leaves which decreased their specific weight at night. With Kalanchoë daigremontiana leaves, the calculated increase in specific leaf weight at night based on estimates of carbohydrate available for net CO2 fixation was near 6% and the measured increase in specific leaf weight was 6%.

Diurnal measurements of CAM tissue water content were neither coincident nor reciprocal with their diurnal patterns of either volume or specific weight changes.

It is generally accepted that various physiological, morphological and gene expression phenomena are under the control of a circadian clock, and that this time keeping mechanism is universally present. Although such endogenously regulated phenomena have first been documented in plants more than 250 years ago and much work has been accumulated particularly in the past 70 years, it was not obvious from the literature whether such time keeping mechanisms exist in gymnosperms. Two prominent parameters were investigated in several gymnosperm species which have been demonstrated to be under the control of a circadian clock in many plants: (i) leaf movement and (ii) stomata movement. In young plants of Pinus sylvestris, Picea abies, Taxus baccata, Araucaria angustifolia, Araucaria heterophylla and Ginkgo biloba leaf oscillations could be recorded for about 5 days. However, compared to angiosperm plants, the amplitude was small. The period length under free running conditions (constant temperature and continuous light) was characteristic for the species. Stomatal movement was observed in Ginkgo biloba leaves by electron microscopy. Stomata were open at noon and closed at midnight under normal day/night conditions (LD) as well as under constant light conditions (LL), indicating that stomatal aperture is under circadian control in the gymnosperm Ginkgo biloba. Online recordings of stomata conductance however, exhibited diurnal but not circadian oscillations of net CO2-exchange in G. biloba leaves. Our results show that a circadian clock controls leaf and stomatal movements in gymnosperm species indicating that endogenous time keeping mechanisms are present.  相似文献

Dicot leaf growth is characterized by partly transient tip-to-base gradients of growth processes, structure and function. These gradients develop dynamically and interact with dynamically developing stress conditions like drought. In Ricinus communis plants growing under well-watered and drought conditions growth rates peaked during the late night and minimal values occurred in the late afternoon. During this diurnal course the leaf base always showed much higher rates than the leaf tip. The amplitude of this diurnal course decreased when leaves approached maturity and during drought stress without any significant alteration of the diurnal pattern and it increased during the first days after rewatering. Unique relationships between leaf size and cytological structure were observed. This provided the framework for the analysis of changes in assimilation, transpiration and dark respiration, chlorophyll, protein, carbohydrate, and amino acid concentrations, and of activities of sink-source-related enzymes at the leaf tip and base during leaf development in well-watered and drought-stressed plants. Gas exchange was dominated by physiological rather than by anatomical properties (stomatal density). Tip-to-base gradients in carbohydrate concentrations per dry weight and sink-source-related enzymes were absent, whereas significant gradients were found in amino acid concentrations per dry weight. During drought stress, growing leaves developed source function at smaller leaf size, before specific physiological adaptations to drought occurred. The relevance of the developmental status of individual leaves for the drought-stress response and of the structural changes for the biochemical composition changes is discussed.  相似文献

The physiological control and molecular mechanism of circumnutation (CN) has not yet been fully understood. To gain information on the CN mechanism, the relationship between the changes of electrical potential and movement in the circumnutating sunflower stem and effect of ion channels and proton pump inhibitors on CN parameters were evaluated. Long‐term electrophysiological measurements and injection of solutions of ion channel inhibitors (ICI) into sunflower stem with the simultaneous time‐lapse recording of the movement were made. The oscillations of electrical potential (OEP) – movement relations – consist of cells depolarization on the deflected side of the stem and, at this same time, cells hyperpolarization on the opposite side of the stem. The delay of organ movement in relation to electrical changes of approximately 28 min (22% of the period) may indicate that the ionic fluxes causing the OEP are the primary phenomenon. The biggest decrease of CN period was observed after injection of proton pump (approximately 26%) and cation channel (approximately 25%) inhibitors, while length and amplitude were reduced mainly by calcium channel inhibitors (approximately 67%). Existence of OEP only in circumnutating part of sunflower stem and reduction of CN parameters and OEP amplitude after application of ICI prove that the CN cellular mechanism is associated with transmembrane ion transport.  相似文献

Hexokinase was measured by quantitative histochemical techniques in the apical meristem, primordia, and leaves of Dianthus chinensis L. The structural stages of development in the leaves sampled were determined by light and scanning electron microscopy. The results showed that activity decreased from the youngest primordia (1500 millimoles per kilogram dry weight per hour) to the mature leaves (200 millimoles per kilogram dry weight per hour) and that an intermediate leaf, the fourth youngest, showed the same declining pattern from its base to its tip. Surface views and measurements of these leaves revealed their basipetal maturation as seen by cell size, stomatal development, trichome differentiation, cuticular appearance, and leaf thickness. The intermediate leaf showed features representative of several stages in structural differentiation. It was concluded that the changes in hexokinase activity among the leaves of a shoot and within an individual leaf are similar and correlate with the degree of structural differentiation of the leaves.  相似文献
