The complexation of VO2+ ion with the high molecular mass components of the blood serum, human serum transferrin (hTf) and albumin (HSA), has been re-examined using EPR spectroscopy. In the case of transferrin, the results confirm those previously obtained, showing that VO2+ ion occupies three different binding sites, A, B1 and B2, distinguishable in the X-band anisotropic spectrum recorded in D2O. With albumin the results show that a dinuclear complex (VO)2dHSA is formed in equimolar aqueous solutions or with an excess of protein; in the presence of an excess of VO2+, the multinuclear complex (VO)xmHSA is the prevalent species, where x = 5-6 indicates the equivalents of metal ion coordinated by HSA. The structure of the dinuclear species is discussed and the donor atoms involved in the metal coordination are proposed on the basis of the measured EPR parameters. Two different binding modes of albumin can be distinguished varying the pH, with only one species being present at the physiological value. The results show that the previously named “strong” site is not the N-terminal copper binding site, and some hypothesis on the metal coordination is discussed, with the 51V Az values for the proposed donor sets obtained by DFT (density functional theory) calculations. Finally, preliminary results obtained in the ternary system VO2+/hTf/HSA are shown in order to determine the different binding strength of the two proteins. Due to the low VO2+ concentration used, the recording of the EPR spectra through the repeated acquisition of the weak signals is essential to obtain a good signal to noise ratio in these systems.  相似文献

We obtained a regressing-tumor antiserum specific for the unique sequence of the transforming protein P140 of Fujinami sarcoma virus by injecting Fischer rats with syngeneic embryo cells transformed with Fujinami sarcoma virus. This serum is capable of immunoprecipitating a protein of 98,000 daltons from cell extracts of normal, uninfected chicken bone marrow cells. This normal cellular protein (NCP98) was shown to be structurally related to P140, sharing the majority of 35S-methionine-labeled tryptic peptides with the viral gene product P140. NCP98 is a phosphoprotein in vivo, with an associated in vitro protein kinase activity, capable of phosphorylating specifically at tyrosine residues of NCP98 itself and a-casein, an externally added substrate. This kinase activity is biochemically indistinguishable from the kinase activity associated with P140 by all criteria tested. Moreover, in vitro-phosphorylated NCP98 and P140 shared the same phosphopeptides. The expression of NCP98 is tissue-specific. It is readily detectable in bone marrow cells and detectable to a lesser extent in liver and lung cells from 6–18 day old chickens.  相似文献

An acetone-formol fixation technique with subsequent paraffin-embedding suitable for immunofluorescent study of different antigens, including serum proteins, is described. This technique was used for detection of albumin, transferrin, and alpha-fetoprotein distribution in the normal and regenerating liver of mice. Albumin and transferrin were always found together in the same hepatocytes, both under normal conditions and in regeneration. In the regenerating liver alpha-fetoprotein was encountered independently of the two other proteins, although it was revealed in the same zones. Only in the perinecrotic zone did each alpha-fetoprotein-positive hepatocyte contain albumin and transferrin.  相似文献

The capacity of continuous cell of XXIIa mouse hepatoma (strain MHXXIIa) to synthesize alpha-fetoprotein, albumin and transferrin was studied by immunoautoradiography. Albumin and transferrin were detected in the polyethylene glycol concentrated growth medium of hepatoma cells on the 5th year (the 55th month) of their cultivation. alpha-fetoprotein was not found. Only transferrin was revealed in the growth medium of hepa toma cells of the 8th year (the 92d month) of cultivation. Two clonal cultures obtained on the 8th year of hepatoma cell cultivation were also characterized by the ability to synthesize transferrin. The continuous mouse hepatoma cells retained their malignancy. The agar micro-precipitation reaction showed the presence of alpha-fetofetoprotein in lyfogel concentrated serum of mice with tumors formed after inoculation of the hepatoma cells of the 5th year of cultivation. However, alpha-fetoprotein was not detected in the serum of mice with tumors induced by inoculation of the hepatoma cells of the 8th year of cultivation.  相似文献

Arthritis was induced by injecting cationic amidated bovine serum albumin (aBSA) (pI approximately 9.2) into the knee joint of immunized guinea pigs and the mechanisms of articular cartilage destruction were studied morphologically and biochemically. Marked synovitis associated with polymorphonuclear leukocyte (PML) infiltration occurred within 1 day of the challenge. Articular cartilage infiltrated by PMLs was almost completely destroyed after 2 weeks. During the initial destructive process, proteoglycans were depleted from the cartilage and later collagen fibers disappeared. Granulation tissue growing in the inflamed synovium and bone marrow replaced the destroyed cartilage and joint cavity and formed fibrous scar tissue (fibrous ankylosis) by 8 weeks. Subsequently, the knee joints developed cartilagenous ankylosis by 12 weeks and finally bony ankylosis at 28 weeks. Autoradiography using 125I-aBSA and immunofluorescence studies for immunoglobulin (IgG) and complement (C3) demonstrated that the antigen is trapped in all zones of the articular cartilage and serves as a trigger for immune complex formation. Significantly increased neutral proteinase activities against substrates of proteoglycan subunits, [3H]carboxymethylated transferrin and L-pyroglutamyl-L-prolyl-L-valine-paranitroanilide were detected in homogenates of the synovium and cartilage from arthritic knee joints 1 and 2 weeks after induction. Inhibitor studies and pH curves suggested that the proteinase is leukocyte elastase. Measurable amounts of gelatinolytic activity, detected by activation with 4-aminophenylmercuric acetate and inhibited with EDTA, were also present in the same samples, but there was no detectable collagenase activity. The data on SDS-gelatin substrate gel showed that the proteinase is gelatinase derived from PMLs. These results suggest that in aBSA-induced arthritis, elastase and gelatinase from PMLs invading articular cartilage may play important roles in cartilage destruction.  相似文献

Alpha-fetoprotein and fetal serum albumin have been simultaneously purified from fetal bovine serum by mild procedures utilizing ammonium sulfate, hydrophobic interaction, immobilized metal (nickel) affinity chromatography, and isoelectric focusing. The lipidic extract from each protein was analyzed by gas chromatography and the peak appearing just after the arachidonic acid was identified as squalene by gas chromatography-mass spectrometry. This isoprenoid was not detected formerly in these proteins from human, rat, bovine, and pig. Until recently, in the analysis of the fatty acid composition of the alpha-fetoprotein and serum albumin from mammals, a peak has been assigned in the last part of the chromatographic profile, after arachidonic acid, to docosahexaenoic acid. In the present work, it was found that the peak corresponds to squalene instead of docosahexaenoic acid. Furthermore, we conclude that bovine alpha-fetoprotein and fetal serum albumin carry squalene, but not docosahexaenoic acid. These results agree with others obtained analyzing the same proteins from chick embryo.  相似文献

Using a computer program designed to detect evolutionary relationships between proteins, I find that residues 72-110 of the mature sequence of human tissue-type plasminogen activator (t-PA) and 39 residues at the carboxy terminus of human albumin have a comparison score that is 8.8 standard deviation units higher than that obtained with a comparison of randomized sequences of these proteins. The probability (p) of getting this score by chance is approximately 10(-18), indicating that part of t-PA and albumin are derived from a common ancestor. I also find that alpha-fetoprotein, a relative of albumin is related to t-PA. Part of this region on t-PA has been previously shown to be related to epidermal growth factor. t-PA, albumin, alpha-fetoprotein, and epidermal growth factor have diverse biological activities. The finding that these proteins are related suggests some new approaches for studying their functions.  相似文献

1. The isolated microsome fraction of regenerating rat liver was incubated with cell sap, a source of energy and [35S]methionine, [14C]isoleucine or [14C]leucine for different periods of time, and microsomal albumin isolated. 2. The distribution of these isotopes in albumin was determined by separation of tryptic peptides from the protein. Radioactivity was measured in peptides either qualitatively by radioautography or quantitatively by labelling with both 3H and 14C. 3. A gradient of radioactivity existed at all times in albumin isolated after incubating microsomes. 4. The shorter the incubation time the fewer the peptides labelled in albumin, but the peptides with highest specific activity after short incubation times corresponded to those with highest specific activities after long incubation times. 5. Leucine released from the C-terminus of albumin had a higher specific activity than the mean specific activity of the remaining leucine residues in albumin. 6. The peptide with the highest specific activity in albumin is probably derived from the C-terminus of the protein. 7. [14C]Glutamic acid is incorporated into the N-terminus of albumin after incubating the microsome fraction with this isotopically labelled amino acid, cell sap and a source of energy. The specific activity of the N-terminal glutamic acid under these conditions is less than the mean specific activity of the remaining glutamic acid and glutamine residues in albumin. 8. The results are interpreted as reflecting a sequential synthesis of serum albumin in the isolated microsome fraction of rat liver. The direction of synthesis of albumin is from the N-terminus towards the C-terminus. 9. The bulk of incorporation of radioactive amino acid into albumin in the isolated microsome fraction is due to completion of partially completed, pre-existing peptide and polypeptide chains. A limited synthesis of new chains of albumin does, however, occur.  相似文献

Binding affinities of ten polycyclic aromatic hydrocarbons to albumin were determined: anthracene, its eight oxy-derivatives: anthraquinone, 9-anthracenemethanol, 9-anthraldehyde, 9-anthracenecarboxylic acid, 1,4-dihydroxyanthraquinone, 1,5-dihydroxyanthraquinone, 1,8-dihydroxyanthraquinone, 2,6-dihydroxyanthraquinone and benzo[a]pyrene. The quenching of albumin fluorescence was used to measure the PAH - protein interaction. The theoretical curve of calculated fluorescence was fitted to experimental data after necessary corrections regarding PAHs fluorescence and inner filter effect. From the numerical fitting the final association constants were calculated. Anthracene and anthraquinone failed to quench the albumin fluorescence. 9-anthracenecarboxylic acid showed the highest, while 9-anthracenemethanol the weakest albumin binding affinity. The affinity constants determined for 9-anthraldehyde and benzo[a]pyrene were of the same magnitude and indicated low-affinity binding to albumin. The constants obtained for the four dihydroxyanthraquinones were higher, but dissimilar, which suggests that the position of the functional group in anthracene molecule influences the binding constant. Moreover, this study suggests that the type of substituent plays a significant role in PAH-albumin complex formation. The carboxylic group increases the binding affinity of the anthracene molecule the most rather than the presence of both carbonyl and hydroxyl groups. The lowest affinity constants were obtained for aldehyde, methyl and carbonyl substituents.  相似文献

Summary The Land Dayaks and the Sea Dayaks of Sarawak were surveyed for haptoglobin, transferrin and serum albumin variants. The Hp1 gene frequency was 0.385 in 283 Land Dayaks as well as in 205 Sea Dayaks. The TfDChi gene frequency in 283 Land Dayaks was 0.030 and in 188 Sea Dayaks it was 0.040. Chiral discrimination of racemic carbazole carbonyl (CC)-amino acids with linear alkyl sidechain (C(1)-C(4)) by bovine serum albumin (BSA) was investigated by competitive replacement experiments using dansyl-L-proline and dansyl-D-norvaline as fluorescent probes. It was found that the CC derivatives of the D-forms of alanine (C(1)), amino butyric acid (C(2)), norvaline (C(3)), and norleucine (C(4)) bound to the dansyl-L-proline site much more strongly than their L-forms, whereas the interactions between both enantiomers of these amino acids with dansyl-D-norvaline site were slight.  相似文献

The binding affinity of human serum albumin (HSA) to three antimalarial indolone-N-oxide derivatives, INODs, was investigated under simulated physiological conditions using fluorescence spectroscopy in combination with UV-vis absorption and circular dichroism (CD) spectroscopy. Analysis of fluorescence quenching data of HSA by these compounds at different temperatures using Stern-Volmer and Lineweaver-Burk methods revealed the formation of a ground state indolone-HSA complex with binding affinities of the order 10(4) M(-1). The thermodynamic parameters ΔG, ΔH, and ΔS, calculated at different temperatures, indicated that the binding reaction was endothermic and hydrophobic interactions play a major role in this association. The conformational changes of HSA were investigated qualitatively using synchronous fluorescence and quantitatively using CD. Site marker competitive experiments showed that the binding process took place primarily at site I (subdomain IIA) of HSA. The number of binding sites and the apparent binding constants were also studied in the presence of different ions.  相似文献

Fetal hepatocytes were cultured alone or in association with primitive biliary cells (RLEC) in the presence or absence of dexamethasone. Cell-cell contacts were established 3 h or five days after hepatocyte seeding and their effects on hepatocyte growth and functional activities were evaluated in the presence or absence of dexamethasone. Establishment of cellular interactions with RLEC in coculture decreased hepatocyte growth, while it stimulated production of alpha-fetoprotein, albumin and transferrin. Addition of dexamethasone to coculture inhibited alpha-fetoprotein secretion and maintained the synthesis rate of albumin and transferrin together with an additional inhibition of DNA synthesis. The levels of mRNAs corresponding to the three proteins were also measured. We observed that the levels of alpha-fetoprotein, albumin and transferrin secretion in cocultures maintained in the presence or absence of dexamethasone were well correlated with the relative amounts of their corresponding mRNAs. Consequently, it may be assumed that the primitive mechanism involved in the increased functional activity of fetal hepatocytes in coculture is of pretranslational origin. Furthermore, the present data provide evidence that heterotypic interactions and dexamethasone act as distinct modulators of growth and maturation of fetal rat hepatocytes.  相似文献

We have shown that the platelet tropomyosin binding protein described in the accompanying paper (Gerhard, M. D., DiGirolamo, P. M., and Hitchcock-DeGregori, S. E. (1985) J. Biol. Chem. 260, 3221-3227) is identical with human serum albumin. The immunological determinants are completely shared; the tryptic peptide maps are the same; the proteins comigrate on two-dimensional gels; and the amino acid sequences of the first 33 amino acids are the same. Although human serum albumin in plasma or commercially prepared protein will not bind tropomyosin-Affi-Gel 15, it will bind following purification from plasma by chromatography on hydroxylapatite.  相似文献

血清白蛋白是必不可少的生命物质,在生命运动和发展延续中起着重要的作用。它不仅能维持正常的血浆渗透压,最重要的是能够储存和运输众多的内源性和外源性物质。本文利用生物信息学的方法分析了几种不同物种的血清白蛋白的结构信息和疏水性特点,研究表明人、牛、猴、兔、狼、猫的血清白蛋白序列均属于亲水性蛋白质,在100 bp以内的疏水性值差别比较明显。通过对血清白蛋白进行多序列比对分析,发现兔血清白蛋白的氨基酸突变的数目是最多的。在这几种血清白蛋白序列中,氨基酸突变更容易发生在结构相似、极性相似和能量比较接近的氨基酸之间,如D和L、E和D。对于人血清白蛋白来说,从疏水性的丙氨酸A到酸性的谷氨酸E的突变比较多,使得人血清白蛋白在进化过程中的亲水性增强,是个很好的储存和运输小分子的载体。这些基于生物信息学方面的血清白蛋白的突变及其进化关系的研究,为进一步研究药物与血清白蛋白的相互作用在其他物种中表现和特点提供了良好的基础。  相似文献

1. Pig alpha-fetoprotein (AFP) and albumin were isolated from fetal serum by DEAE-Sephadex ion exchange chromatography combined with Cibacron Blue-Sepharose and trypsin-Sepharose adsorptions. 2. AFP, fetal albumin and adult albumin carried 2.6, 2.4, and 1.9 moles of fatty acids per mole of protein, respectively. 3. Most of fatty acids bound to AFP were polyunsaturated: mainly arachidonic (20:4, n-6) and docosahexaenoic (22:6, n-3) acids, which accounted respectively for 21.7 and 18.8% of the total fatty acids. 4. By contrast, the fatty acids found in the albumins (fetal and adult) were preferentially saturated and monounsaturated. 5. Arachidonic acid was a minor component in both albumins, and no docosahexaenoic acid was detected.  相似文献
