The effect of inactivation of the PPX1 and PPN1 genes encoding the yeast exopolyphosphatases on the activities of these enzymes and polyphosphate content in the cytosol of Saccharomyces cerevisiae was studied under Pi deficit and Pi excess in the cultivation medium. Under Pi deficit, exopolyphosphatase activity in strain CRN (with inactivated PPN1 gene) and in the parent strain CRY increased 3- and 1.5-fold, respectively. In the strain CRX (with inactivated PPX1 gene), exopolyphosphatase activity did not change under Pi deficit. Transfer from Pi-deficient to Pi-rich medium was accompanied by an ~1.7-fold increase of exopolyphosphatase activities in the cytosol preparations of strains CRY, CRX, and CRN. In the cytosol of the double mutant, exopolyphosphatase activity was practically absent under all of the above cultivation conditions. The content of polyphosphates in the cytosol preparations of all strains under study substantially decreased under Pi deficit. Transfer from Pi-deficient to Pi-rich medium was accompanied by polyphosphate over-accumulation only in the cytosol preparations of stains CRX and CNX, where their levels increased ~1.3 and 3.5-fold, respectively. No over-accumulation was observed in the parent strain CRY and in the PPN1-deficient strain CRN. These data suggest that the exopolyphosphatases encoded by the PPX1 and PPN1 genes are not involved in polyphosphate synthesis.  相似文献

We have studied the effects of a low-intensity electromagnetic field (EMF) with a frequency of 30 MHz in continuous-wave and amplitude-modulation modes on Daphnia magna in a laboratory culture. The exposure range is from 10 to 10000 s. We examine the parental generation (irradiated) and three subsequent (F1–F3) generations (nonirradiated). It is found that irradiation does not affect the total fertility of any specimen in four generations (P–F3), except for continuous EMF exposure for 10000 s, which increases the total fertility in generations F2 and F3 by 23 and 43%, respectively. Electromagnetic radiation has a significant impact on offspring quality and causes fetal abnormalities in the offspring of all generations.  相似文献

β-Lapachone (β-Lap), morphine (Mor), and electromagnetic field (EMF) generate reactive oxygen species. The goal of the present study was to examine the effects of Mor and EMF, in combination with β-Lap on the cell growth inhibition and expression of several antioxidant genes. The 0.50 mT intensity of 50 Hz EMF and two exposure conditions (“15 min field-on/15 min field-off” and “30 min field-on continuously”) on SH-SY5Y cells were used. The effects of Mor and EMF, in combination with β-Lap on cell growth inhibition and the expression levels of several antioxidant genes (NQO1, NQO2, SOD1, SOD2, CAT, GSTO1, GSTM2, GSTM3, GSTP1, MGST1, MGST3) in SH-SY5Y cells were measured. The relative mRNA levels were calculated according to the \({{\text{2}}^ - }^{{\Delta \Delta {{\text{C}}_{\text{t}}}}}\). Whereas NQO1 mRNA level decreased in the “15 min field-on/15 min field-off” condition, the expression level of NQO2 was increased. Both NQO1 and NQO2 expressions increased in Mor treated cells. IC50 values of β-Lap in combination with Mor, EMF, and “Mor?+?EMF” were higher than cells treated only with β-Lap. The NQO1 expression level in the cells treated with β-Lap was higher than the other treatments, indicating that β-Lap induces the expression of NQO1. Moreover, multiple linear regression analysis indicated that NQO1 mRNA levels were associated positively with β-Lap and negatively with EMF. At least in part, the mRNA levels of NQO1 were associated with IC50 values of β-Lap in designed treatments. There is a negative association between mRNA levels of NQO1 and IC50 values of β-Lap but not NQO2.  相似文献

Mobile phones are widely used in the modern world. However, biological effects of electromagnetic radiation produced by mobile phones are largely unknown. In this report, we show biological effects of the mobile phone 835 MHz electromagnetic field (EMF) in the Drosophila model system. When flies were exposed to the specific absorption rate (SAR) 1.6 W/kg, which is the proposed exposure limit by the American National Standards Institute (ANSI), more than 90% of the flies were viable even after the 30 h exposure. However, in the SAR 4.0 W/kg strong EMF exposure, viability dropped from the 12 h exposure. These EMF exposures triggered stress response and increased the production of reactive oxygen species. The EMF exposures also activated extracellular signal regulated kinase (ERK) and c-Jun N-terminal kinase (JNK) signaling, but not p38 kinase signaling. Interestingly, SAR 1.6 W/kg activated mainly ERK signaling and expression of an anti-apoptotic gene, whereas SAR 4.0 W/kg strongly activated JNK signaling and expression of apoptotic genes. In addition, SAR 4.0 W/kg amplified the number of apoptotic cells in the fly brain. These findings demonstrate that the exposure limit on electromagnetic radiation proposed by ANSI triggered ERK-survival signaling but the strong electromagnetic radiation activated JNK-apoptotic signaling in Drosophila.  相似文献

The electromagnetic field (EMF) is newly considered as an exogenous environmental stimulus that is closely related to ion transportation on the cellular membrane, maintaining the internal ionic homeostasis. Cation transports of Ca2+ and other metal ions, Cd2+, Zn2+, and Mn2+were studied in terms of the external Ca2+ stress, [Ca2+]ext, and exposure to the physical EMF. A specific yeast strain K667 was used for controlling CAX5 (cation/H+ exchanger) expression. Culture samples were exposed to 60 Hz, 0.1 mT sinusoidal or square magnetics waves, and intracellular cations of each sample were measured and analyzed. AtCAX5 transformant yeast grew normally under the metallic stress. However, the growth of the control group was significantly inhibited under the same cation concentration; 60 Hz and 0.1 mT magnetic field enhanced intracellular cation concentrations significantly as exposure time increased both in the AtCAX5 transformed yeast and in the control group. However, the AtCAX5-transformed yeast showed higher concentration of the intracellular cations than the control group under the same exposure EMF. AtCAX5-transformed yeasts displayed an increment in [Ca2+]int, [K+]int, [Na+]int, and [Zn2+]int concentration under the presence of both sinusoidal and square-waved EMF stresses compared to the control group, which shows that AtCAX5 expressed in the vacuole play an important role in maintaining the homeostasis of intracellular cations. These findings could be utilized in the cultivation of the crops which were resistant to excessive exogenous ions or in the production of biomass containing a large proportion of ions for nutritional food or in the bioremediation process in metal-polluted environments.  相似文献

The current study aimed to develop a prolonged-release pramipexole (PPX) transdermal patch for the treatment of Parkinson’s disease. Permeation parameters of PPX were investigated using human cadaver skin. Pramipexole patches were prepared using DURO-TAK® pressure-sensitive-adhesive (PSA) and evaluated for drug stability, drug loading, in vitro drug release, and in vitro permeation through mouse skin. The results indicated that blends of DURO-TAK® 87-2852 and DURO-TAK® 87-2510 were suitable for creating a prolonged-release PPX patch due to their advantages in drug release, drug loading, and stability. The final formulation consisted of 87-2852/87-2510 (70:30), 10% PG, and 15% PPX and showed a cumulative permeation amount of 1497.19?±?102.90 μg/cm2 with a continuous flux over 6.0 μg/(cm2·h) across human cadaver skin for 7 days. In vivo studies in rats indicated that PPX patch produced a significantly longer (p?<?0.001) half-life (t 1/2, 75.16?±?17.37 h) and mean residence time (MRT, 135.89?±?24.12 h) relative to oral tablets (Sifrol®) and had a relative bioavailability of 51.64?±?21.32%. Therefore, this study demonstrated the feasibility of developing a prolonged-release PPX patch, which proposed the potential to serve as an alternate to conventional oral tablets and may therefore improve patient compliance.  相似文献

It is found that for Enterococcus hirae ATCC9790 bacteria grown in anaerobic conditions, one-hour exposure to low-intensity (radiant power of 0.06 mW/cm2) coherent extremely high frequency electromagnetic radiation (from 45 to 53 GHz), or millimeter electromagnetic radiation, leads to an appreciable increase in latent growth time and to a decrease in specific growth rate; herein, the effects intensify as the frequency increases from 49 to 53 GHz. The result is enhanced at an increase in the radiation duration from 30 min to 1 h; however, a further increase in the exposure time up to 2 h does not lead to intensification of the effect. It is shown that the effect of extremely high frequency electromagnetic radiation on Enterococcus hirae does not depend on pH of the medium (pH 6.0 or 8.0). It may be expected that these bacteria have protective or reparation mechanisms that compensate long-term action of this radiation; it is not improbable that various mechanisms of pH regulation are present as well.  相似文献

A plant’s growth and fitness are influenced by species interactions, including those belowground. In primary successional systems, belowground organisms are known to have particularly important control over plant growth. Exotic plant invasions in these and other habitats may in part be explained by altered associations with belowground organisms compared to native plants. We investigated the growth responses of two foundation grasses on Great Lakes sand dunes, the native grass Ammophila breviligulata and the exotic grass Leymus arenarius, to two groups of soil organisms with important roles in dune succession: arbuscular mycorrhizal fungi (AMF) and plant-parasitic nematodes (PPN). We manipulated the presence/absence of two generalist belowground species known to occur in Great Lakes dunes, Rhizophagus intraradices (AMF) and Pratylenchus penetrans (PPN) in a factorial greenhouse experiment and assessed the biomass production and root architectural traits of the plants. There were clear differences in growth and above- and belowground architecture between Ammophila and Leymus, with Leymus plants being bigger, taller, and having longer roots than Ammophila. Inoculation with Rhizophagus increased above- and belowground biomass production by ~32% for both plant species. Inoculation with Pratylenchus decreased aboveground biomass production by ~36% for both plant species. However belowground, the exotic Leymus was significantly more resistant to PPN than the native Ammophila, and gained more benefits from AMF in belowground tri-trophic interactions than Ammophila. Overall, our results indicate that differences in plant architecture coupled with altered belowground interactions with AMF and PPN have the potential to promote exotic plant invasion.  相似文献

Forest ecosystems of the Pacific Northwest of the USA are changing as a result of climate change. Specifically, rise of global temperatures, decline of winter precipitation, earlier loss of snowpack, and increased summer drought are altering the range of Pinus contorta. Simultaneously, flux in environmental conditions within the historic P. contorta range may facilitate the encroachment of P. ponderosa into P. contorta territory. Furthermore, successful pine species migration may be constrained by the distribution or co-migration of ectomycorrhizal fungi (EMF). Knowledge of the linkages among soil fungal diversity, community structure, and environmental factors is critical to understanding the organization and stability of pine ecosystems. The objectives of this study were to establish a foundational knowledge of the EMF communities of P. ponderosa and P. contorta in the Deschutes National Forest, OR, USA, and to examine soil characteristics associated with community composition. We examined EMF root tips of P. ponderosa and P. contorta in soil cores and conducted soil chemistry analysis for P. ponderosa cores. Results indicate that Cenococcum geophilum, Rhizopogon salebrosus, and Inocybe flocculosa were dominant in both P. contorta and P. ponderosa soil cores. Rhizopogon spp. were ubiquitous in P. ponderosa cores. There was no significant difference in the species composition of EMF communities of P. ponderosa and P. contorta. Ordination analysis of P. ponderosa soils suggested that soil pH, plant-available phosphorus (Bray), total phosphorus (P), carbon (C), mineralizable nitrogen (N), ammonium (NH4), and nitrate (NO3) are driving EMF community composition in P. ponderosa stands. We found a significant linear relationship between EMF species richness and mineralizable N. In conclusion, P. ponderosa and P. contorta, within the Deschutes National Forest, share the same dominant EMF species, which implies that P. ponderosa may be able to successfully establish within the historic P. contorta range and dominant EMF assemblages may be conserved.  相似文献

Withania somnifera, also known as Indian ginseng is known to contain valuable bioactive compounds, called withanolides that structurally resemble ginsenosides of Panax ginseng. These compounds provide the basis of pharmacological relevance in traditional systems of medicine. In the present study, 150 hairy root lines of W. somnifera were induced of which nine fast growing lines were analysed for their growth and withanolide content. Hairy root line W9 was selected due to its high specific growth rate (0.196 ± 0.005 d?1) and high withanolide content. The response to different concentrations of elicitors (methyl jasmonate and P. indica cell homogenate) and various exposure durations was assessed in the W9 hairy root line. The withanolide content as well as the pattern of gene expression from MVA, MEP and sterol pathway, was evaluated using qPCR. Though gene expression and withanolide content were found to be elevated in almost all MeJ and CHP treatments, the exposure of hairy roots to 15 μM MeJ for 4 h gave the maximum withanolide yield. The results suggest that the elicitation potential of methyl jasmonate was higher than that of P. indica cell homogenate for increasing withanolide levels in hairy roots of W. somnifera.  相似文献

Physiological stress can bring major molecular and cellular change to a living cell which further decide its survival or tolerance to the stress exposure. Cyanobacteria like Anabaena has been shown to tolerate high levels of different stresses like oxidative, desiccation, UV, and gamma radiation. They are able to withstand and recover remarkably without any lethal mutation when exposed to high doses of gamma radiation or prolonged duration of desiccation. In the present work, the modifications in protein profiles of Anabaena 7120 cells after exposure to 6 kGy of 60Co γ-rays and 6 days of desiccation, and the proteome dynamics during post stress recovery were investigated. Differentially expressed proteins during stress and recovery were identified by MALDI-ToF or LC-MS, which generated a partial proteome map of Anabaena 7120. Anabaena cells went through protein recycling—phase of protein degradation following by their resynthesis, which helped them to recover remarkably. The data suggests an overlap in proteome changes during recovery against radiation and desiccation stress.  相似文献

A sub‐acute electromagnetic field (EMF) biological effect study was carried out on rats exposed in the Transverse ElectroMagnetic exposure chamber at 171 MHz Continuous Wave (CW). The experiments involved three exposure levels (15, 25, and 35 V/m) for 15 days with triplicate parallel sham‐exposed controls in each series. All exposure conditions were simulated for the evaluation of the electromagnetic energy distribution and specific absorption rate (SAR) in the rat phantoms. Studies have shown a biphasic biological response depending on time and absorbed electromagnetic energy. Under low SAR, approximately 0.006 W/kg, EMF exposure leads to the stimulation of adrenal gland activity. This process is accompanied by an initial increase of daily excretion of corticosterone and Na+, which is seen as a higher Na+/K+ ratio, followed by a decrease of these parameters over time. It is possible that EMF exposure causes a stress response in animals, which is seen as an increased adrenal activity. The interaction of 1.07-μm laser radiation with plasma of a continuous optical discharge (COD) in xenon and argon at a pressure of p = 3–25 bar and temperature of T = 15 kK has been studied. The threshold power required to sustain COD is found to decrease with increasing gas pressure to P t < 30 W in xenon at p > 20 bar and to P t < 350 W in argon at p > 15 bar. This effect is explained by an increase in the coefficient of laser radiation absorption to 20?25 cm–1 in Xe and 1?2 cm–1 in Ar due to electronic transitions between the broadened excited atomic levels. The COD characteristics also depend on the laser beam refraction in plasma. This effect can be partially compensated by a tighter focusing of the laser beam. COD is applied as a broadband light source with a high spectral brightness.  相似文献

Abstract

The growing spread of mobile phone use is raising concerns about the effect on human health of the electromagnetic field (EMF) these devices emit. The purpose of this study was to investigate the effects on rat pup heart tissue of prenatal exposure to a 900 megahertz (MHz) EMF. For this purpose, pregnant rats were divided into experimental and control groups. Experimental group rats were exposed to a 900?MHz EMF (1?h/d) on days 13–21 of pregnancy. Measurements were performed with rats inside the exposure box in order to determine the distribution of EMF intensity. Our measurements showed that pregnant experimental group rats were exposed to a mean electrical field intensity of 13.77?V/m inside the box (0.50?W/m2). This study continued with male rat pups obtained from both groups. Pups were sacrificed on postnatal day 21, and the heart tissues were extracted. Malondialdehyde, superoxide dismutase and catalase values were significantly higher in the experimental group rats, while glutathione values were lower. Light microscopy revealed irregularities in heart muscle fibers and apoptotic changes in the experimental group. Electron microscopy revealed crista loss and swelling in the mitochondria, degeneration in myofibrils and structural impairments in Z bands. Our study results suggest that exposure to EMF in the prenatal period causes oxidative stress and histopathological changes in male rat pup heart tissue.  相似文献

During exposure to the cell phone electromagnetic field (EMF), some neurons in the brain at areas of peak specific absorption rate (SAR) absorb more electromagnetic energy than is permitted by existing guidelines. The goal of the present work was to investigate the influence of cell phone-like EMF signal on excitability and memory processes in single neurons. A Transverse Electromagnetic Cell (TEM Cell) was used to expose single neurons of mollusk to the EMF.

Finite-Difference Time-Domain (FDTD) method was used for modeling the TEM Cell and the EMF interactions with living nerve ganglion and neurons. Neuron electrophysiology was investigated using standard microelectrode technique. SAR deposited into the single neuron was calculated to be 8.2 W/kg with a temperature increment of 1.21°C. After acute exposure, the threshold of firing of action potentials (AP) was significantly decreased (p ≈ 0.001). Time of habituation to stimulation with the intracellular current injection was increased (p ≈ 0.003). These results indicate that acute exposure to EMF at high SARs impairs the ability of neurons to store information.  相似文献

DETERMINATION of the frequency of chromosome aberrations in cultured blood lymphocytes may provide a means of measuring ionizing radiation doses, at least after whole body exposure. The barley genes Rpg5, RGA1 and Adf3, which provide a strong resistance to many pathotypes of stem rust, were cloned a few years ago, but it was still unclear whether their homologues were represented in wheat and in related species. The paper describes the results of a bioinformatic research to determine the homologues of Rpg5, RGA1 and Adf3 in the genomes of Triticum aestivum and several wild grasses, which breeders usually use as sources of stem rust resistance, and which are available in the genome databases. It was found that the Th. elongatum sequence Q9FEC6 and T. aestivum sequence Q43655 were the highly identical homologues of the Adf3 sequence. T. urartu M8A999 sequence and T. aestivum W5FCU1 sequence were found to be the closest homologues of Rpg5 complete protein sequence, but the identity of their kinase domains was not as clear as that of the other domains. The separate Rpg5 kinase part analysis did not provide the strong evidences that its orthologs were present in our corn species. T. urartu M7ZZX9 sequence and T. aestivum W5FFP0 and W5FI33 sequences were shown to be the homologues of RGA1. The analysis of the predicted active sites allowed finding out the difference between sequences of Rpg5, RGA1, Adf3 protein and their homologues.  相似文献

UDP-glucosyltransferases (UGTs) contribute to Fusarium head blight (FHB) resistance of wheat and barley by glycosylating the deoxynivalenol (DON), which is produced by Fusarium fungus. In this study, seven alleles of barley HvUGT14077 (GenBank No.GU170356.1) were cloned using RT-PCR. Among them, HvUGT-10W1, which was isolated from a FHB resistant barley variety 10W1, was significantly up-regulated in young spikes after F. graminearum (F.g) inoculation. HvUGT-10W1::GFP was subcellularly located in the plasma membrane and cytoplasm of the wheat protoplasts. In vitro antifungal activity assay showed that the HvUGT-10W1 protein exerted obvious inhibition against the growth of F.g. The silencing of the HvUGT-10W1 by virus-induced gene silencing (VIGS) resulted in compromised FHB resistance of 10W1, which was shown by the increased infected colonies on the leaves. These indicated that the barley HvUGT-10W1 may also contribute to F.g resistance in barley and provided a potential candidate gene to develop transgenic barley with enhanced FHB resistance.  相似文献

Sub-arctic birch forests (Betula pubescens Ehrh. ssp. czerepanovii) periodically suffer large-scale defoliation events caused by the caterpillars of the geometrid moths Epirrita autumnata and Operophtera brumata. Despite their obvious influence on ecosystem primary productivity, little is known about how the associated reduction in belowground C allocation affects soil processes. We quantified the soil response following a natural defoliation event in sub-arctic Sweden by measuring soil respiration, nitrogen availability and ectomycorrhizal fungi (EMF) hyphal production and root tip community composition. There was a reduction in soil respiration and an accumulation of soil inorganic N in defoliated plots, symptomatic of a slowdown of soil processes. This coincided with a reduction of EMF hyphal production and a shift in the EMF community to lower autotrophic C-demanding lineages (for example, /russula-lactarius). We show that microbial and nutrient cycling processes shift to a slower, less C-demanding state in response to canopy defoliation. We speculate that, amongst other factors, a reduction in the potential of EMF biomass to immobilise excess mineral nitrogen resulted in its build-up in the soil. These defoliation events are becoming more geographically widespread with climate warming, and could result in a fundamental shift in sub-arctic ecosystem processes and properties. EMF fungi may be important in mediating the response of soil cycles to defoliation and their role merits further investigation.  相似文献

Exposure of the purple bacteria Rhodobacter sphaeroides MDC6522 isolated from Jermuk mineral springs (Armenia) to extremely high-frequency electromagnetic radiation (51.8 and 53.0 GHz) for 15 min resulted in a pronounced increase in the specific growth rate and H2 photoemission. However, a significant decrease in the specific growth rate (1.6–2.0 times) was observed when the duration of irradiation was prolonged to 1 h. The maximum effect was at a frequency of 53.0 GHz. During irradiation for 1 h, absorption maxima typical of carotenoids gradually disappeared, and the level of bacteriochlorophyll а complexes decreased. Prolonged irradiation also inhibited the H2 production during bacterial growth for 72 h, although it was restored after 96 h of growth. The activity of N,N'-dicyclohexylcarbodiimide-sensitive proton F0F1- ATPase also decreased in Rh. sphaeroides. These results indicate that the membrane-bound F0F1-ATPase may be the main target of action of extremely-high-frequency electromagnetic radiation. The data we obtained can be used in biotechnology for control of growth and hydrogen metabolism of phototrophic bacteria.  相似文献
