(NniPrCr)2222-N2) (1, NniPr = bis(2,6-diisopropylphenyl)pentane-2,4-ketiminato), formally a chromium(I) complex, was exposed to a variety of small molecules possessing hetero- or homo-atomic single, double, or triple bonds. Reaction of 1 with adamantyl azide yielded complexes in various higher oxidation states, such as (NniPrCrII)22-NAd) (2), NniPrCrIII2-N4Ad2) (3), or NniPrCrV(NAd)2 (4). Compound 1 was found to reductively couple 3-pentanone to form NniPrCr(κ2-O2C2Et4) (5) and reductively couple benzylidene aniline to form both NniPrCrIII(cis2-C28H22N2) (6a) an NniPrCrIII(trans2-C28H22N2) (6b). Reaction with a stochiometric amount of benzylidene aniline yielded the imine complex NniPrCr(η2-NPhCHPh) (7). Exposure of 1 to a bulky isocyanide formed the octahedral NniPrCrI[CN(C6H4(Me)2]4 (9). Complex 1 was also found to break the O-O, NO, and S-S bonds in various small molecules to form NniPrCrII(OCMe3) (11), NniPrCrV(O)(NPh) (8), and [NniPrCrII(μ-SPh)]2 (10).  相似文献

Nelumbo nucifera (Nn.) fiber is the isolated secondary wall of the Nn. leafstalk Tracheary elements which has a unique shape. As the shape of the fiber may strongly affect the industrial uses especially for textile usage, the morphology and structure of Nn. fiber at different growth stages were investigated by several techniques in the present work. The Nn. fiber has spriral morphology with cellulose I structure. The diameter of mature fiber is about 4 μm and the cross-section shows elliptical or slightly oval shape without lumen. These findings aim to deeply understand the structure of Nn. fiber which is expected to be helpful to bring Nn. fiber into industrial use.  相似文献

The phenomenon of magnetic flux breakthrough into a wire array during its implosion was studied experimentally at the Angara-5-1 facility. It is shown that breakthroughs develop in the final stage of plasma production from the wire material and occur near the initial wire position. The spatial distributions of the azimuthal magnetic field within tungsten, molybdenum, copper, and aluminum wire arrays were studied using magnetic probes. The distributions of the azimuthal magnetic field B φ(z, t) along the array height in different stages of implosion were measured, and the characteristic dimensions of regions with a nonuniform magnetic field that appear during magnetic flux breakthroughs at the outer boundary of the wire array plasma were determined. The dimensions of these regions are compared with those of the regions with depressed plasma radiation observed in frame and time-integrated X-ray images. The dynamics of the distribution B φ(z, t) in regions with a nonuniform magnetic field during breakthroughs of the azimuthal magnetic flux is compared with that of the spatial distribution of pinch radiation in the frame X-ray images in different stages of implosion. The experimental data on the characteristics of spatially nonuniform breakthroughs of the magnetic flux into the wire array are analyzed using the plasma rainstorm model proposed by V.V. Aleksandrov et al. (JETP 97, 745 (2003)). The plasma density in the region of magnetic flux breakthrough is estimated.  相似文献

The experiments presented here were based on the conclusions of our previous results. In order to avoid introduction of expression plasmid and to balance the NADH/NAD ratio, the NADH biosynthetic enzyme, i.e., NAD-dependent glyceraldehyde-3-phosphate dehydrogenase (GADPH), was replaced by NADP-dependent GADPH, which was used to biosynthesize NADPH rather than NADH. The results indicated that the NADH/NAD ratio significantly decreased, and glucose consumption and l-lysine production drastically improved. Moreover, increasing the flux through l-lysine biosynthetic pathway and disruption of ilvN and hom, which involve in the branched amino acid and l-methionine biosynthesis, further improved l-lysine production by Corynebacterium glutamicum. Compared to the original strain C. glutamicum Lys5, the l-lysine production and glucose conversion efficiency (α) were enhanced to 81.0 ± 6.59 mM and 36.45 % by the resulting strain C. glutamicum Lys5-8 in shake flask. In addition, the by-products (i.e., l-threonine, l-methionine and l-valine) were significantly decreased as results of genetic modification in homoserine dehydrogenase (HSD) and acetohydroxyacid synthase (AHAS). In fed-batch fermentation, C. glutamicum Lys5-8 began to produce l-lysine at post-exponential growth phase and continuously increased over 36 h to a final titer of 896 ± 33.41 mM. The l-lysine productivity was 2.73 g l?1 h?1 and the α was 47.06 % after 48 h. However, the attenuation of MurE was not beneficial to increase the l-lysine production because of decreasing the cell growth. Based on the above-mentioned results, we get the following conclusions: cofactor NADPH, precursor, the flux through l-lysine biosynthetic pathway and DCW are beneficial to improve l-lysine production in C. glutamicum.  相似文献

A cyprinid fish,Pseudogobio esocinus showed gradual bradycardia at oxygen saturation (%) of less than 29.7±4.6 (1.89±0.29 ml/l of oxygen concentration), surfacing at 14.7±1.3 (0.94±0.09ml/l), drastic decrease of oxygen consumption at less than 14.2±0.8 (0.91 ±0.06ml/l) and asphyxia at 9.7±1.4 (0.62±0.09ml/l). The fish avoided water having low oxygen saturation of less than 54.0± 5.4 (3.38±0.30ml/l), and markedly at less than 26.2±3.4 (1.62±0.16 ml/l).  相似文献

High concentrations of acetate, the main by-product of Escherichia coli (E. coli) high cell density culture, inhibit bacterial growth and l-threonine production. Since metabolic overflux causes acetate accumulation, we attempted to reduce acetate production by redirecting glycolysis flux to the pentose phosphate pathway by deleting the genes encoding phosphofructokinase (pfk) and/or pyruvate kinase (pyk) in an l-threonine-producing strain of E. coli, THRD. pykF, pykA, pfkA, and pfkB deletion mutants produced less acetate (9.44 ± 0.83, 3.86 ± 0.88, 0.30 ± 0.25, and 6.99 ± 0.85 g/l, respectively) than wild-type THRD cultures (19.75 ± 0.93 g/l). THRDΔpykF and THRDΔpykA produced 11.05 and 5.35 % more l-threonine, and achieved a 10.91 and 5.60 % higher yield on glucose, respectively. While THRDΔpfkA grew more slowly and produced less l-threonine than THRD, THRDΔpfkB produced levels of l-threonine (102.28 ± 2.80 g/l) and a yield on glucose (0.34 g/g) similar to that of THRD. The dual deletion mutant THRDΔpfkBΔpykF also achieved low acetate (7.42 ± 0.81 g/l) and high l-threonine yields (111.37 ± 2.71 g/l). The level of NADPH in THRDΔpfkA cultures was depressed, whereas all other mutants produced more NADPH than THRD did. These results demonstrated that modification of glycolysis in E. coli THRD reduced acetate production and increased accumulation of l-threonine.  相似文献

Results of experimental studies of the implosion of quasi-spherical wire (or metalized fiber) arrays are presented. The goal of the experiments was to achieve synchronous three-dimensional compression of the plasma produced in different regions of a quasi-spherical array into its geometrical center. To search for optimal synchronization conditions, quasi-spherical arrays with different initial profiles of the linear mass were used. The following dependences of the linear mass on the poloidal angle were used: m l (θ) ∝ sin–1θ and m l (θ) ∝ sin–2θ. The compression dynamics of such arrays was compared with that of quasi-spherical arrays without linear mass profiling, m l (θ) = const. To verify the experimental data, the spatiotemporal dynamics of plasma compression in quasi-spherical arrays was studied using various diagnostics. The experiments on three-dimensional implosion of quasi-spherical arrays made it possible to study how the frozen-in magnetic field of the discharge current penetrates into the array. By measuring the magnetic field in the plasma of a quasi-spherical array, information is obtained on the processes of plasma production and formation of plasma flows from the wire/fiber regions with and without an additionally deposited mass. It is found that penetration of the magnetic flux depends on the initial linear mass profile m l (θ) of the quasi-spherical array. From space-resolved spectral measurements and frame imaging of plasma X-ray emission, information is obtained on the dimensions and shape of the X-ray source formed during the implosion of a quasi-spherical array. The intensity of this source is estimated and compared with that of the Z-pinch formed during the implosion of a cylindrical array.  相似文献

On the basis of a two-component (two-fluid) hydrodynamic model, it is shown that the probable phenomenon of solar core rotation with a velocity higher than the average velocity of global rotation of the Sun, discovered by the SOHO mission, can be related to fast solid-body rotation of the light hydrogen component of the solar plasma, which is caused by thermonuclear fusion of hydrogen into helium inside the hot dense solar core. Thermonuclear fusion of four protons into a helium nucleus (α-particle) creates a large free specific volume per unit particle due to the large difference between the densities of the solar plasma and nuclear matter. As a result, an efficient volumetric sink of one of the components of the solar substance—hydrogen—forms inside the solar core. Therefore, a steady-state radial proton flux converging to the center should exist inside the Sun, which maintains a constant concentration of hydrogen as it burns out in the solar core. It is demonstrated that such a converging flux of hydrogen plasma with the radial velocity v r (r) = ?βr creates a convective, v r ?v φ/?r, and a local Coriolis, v r v φ/r,φ nonlinear hydrodynamic forces in the solar plasma, rotating with the azimuthal velocity v φ. In the absence of dissipation, these forces should cause an exponential growth of the solid-body rotation velocity of the hydrogen component inside the solar core. However, friction between the hydrogen and helium components of the solar plasma due to Coulomb collisions of protons with α-particles results in a steady-state regime of rotation of the hydrogen component in the solar core with an angular velocity substantially exceeding the global rotational velocity of the Sun. It is suggested that the observed differential (liquid-like) rotation of the visible surface of the Sun (photosphere) with the maximum angular velocity at the equator is caused by sold-body rotation of the solar plasma in the radiation zone and strong turbulence in the tachocline layer, where the turbulent viscosity reaches its maximum value at the equator. There, the tachocline layer exerts the most efficient drag on the less dense outer layers of the solar plasma, which are slowed down due to the interaction with the ambient space plasma (solar wind).  相似文献

Nodal explants of in vivo plants and in vitro seedlings of Wattakaka volubilis were cultured on Murashige and Skoog medium fortified with various concentrations of cytokinins — BA (0.5–5 mg l?1), KN (0.5–10 mg l?1),TDZ (0.05–1 mg l?1) either singly or in combination with NAA (0.1 mg l?1). KN proved best for inducing healthy shoots in both in vitro and in vivo derived explants. Maximum number of shoots (14.1±0.84) with 80% regeneration frequency was obtained from nodal explants of seedlings cultured on 5 mg 1?1 KN + 0.1 mg l?1 NAA. In vivo nodal explants produced a maximum of 4.2 shoots on MS medium fortified with 2 mg l?1 BA+0.1 mg l?1 NAA. The differentiated shoots from both could be rooted with 85% frequency on 1/2 strength MS medium (1% sucrose) with 0.6% agar + 1 mg l?1 IBA + 0.2 mg l?1 KN. Rooted shoots were transplanted to vermiculite-soil (3:1) mixture in polyethylene covered pots with 45% transplantation success. Peroxidase isozymes (native PAGE) analysis helped to verify the variation in regenerated plants.  相似文献

In the first of three experiments, eight ovariectomised Greyface ewes primed with exogenous progesterone were used to provide quantitative data on the effects of two contrasting feeding levels (0.3 vs. 1.4 × maintenance) on plasma progesterone concentrations. Over the 9 day study period, mean (± SEM) daily progesterone concentrations were 4.3 ± 0.13 and 3.3 ± 0.17 μg l−1 for the low and high feeding regimens, respectively (P = 0.06), indicating that high feed intake suppressed circulating progesterone levels. The second experiment examined the effect in superovulated Finn-Dorset ewes of a diet supplying either 0.6 (Group L, n = 8) or 2.3 (Group H, n = 8) times their daily energy needs for maintenance, from 1 day before introduction of exogenous progesterone to the time of insemination, on plasma progesterone concentrations and the viability of ova recovered 4 days after insemination. Mean (± SEM) plasma progesterone concentrations were 4.5 ± 0.17 μg l−1 and 2.8 ± 0.16 μg l−1 for L and H ewes, respectively, during the 12 day priming period (P < 0.001). Eight hours after progesterone withdrawal, levels had fallen to 0.9 ± 0.06 μg l−1 and 0.8 ± 0.07 μg l−1, respectively, then rose to 17.8 ± 3.01 μg l−1 and 12.9 ± 2.50 μg l−1 (P > 0.10) at ovum collection. Intervals (mean ± SEM) to oestrous onset (14.5 ± 0.38 h) and the luteinising hormone (LH) surge (27.1 ± 0.98 h) were unaffected by feed intake. Mean (± SEM) ovulation rates (8.1 ± 1.57 vs. 7.8 ± 1.10) and numbers of ova recovered (5.0 ± 1.39 vs. 4.8 ± 1.11) were also similar for each group. However, the proportions of ova considered viable (over 32 cells) at recovery were 0.53 and 0.22 for L and H groups, respectively (P < 0.005). Following 72 h culture (Tissue Culture Medium-199 (M199) + 10% foetal calf serum (FCS)), 0.55 and 0.27, respectively, had developed to blastocysts (P < 0.025). Of ova assessed as viable at recovery, similar proportions (0.86 vs. 0.75) from L and H treatments developed to blastocysts, with corresponding nuclei counts (mean ± SEM) of 55 ± 5.2 and 55 ± 13.2. The third experiment used 12 superovulated Greyface ewes, each offered a different feed level within the range 0.6–2.5 × maintenance, to determine the nature of the relationship between feeding level, pre-ovulatory progesterone concentrations and ovum development at Day 2 following insemination and subsequently during 7 day co-culture (M199 + FCS). Increases in feeding level were accompanied by linear decreases in plasma progesterone (r2 = 0.79, P < 0.001), the interval to oestrous onset (r2 = 0.52, P < 0.01) and timing of the LH surge (r2 = 0.32, P < 0.06). Although undetectable at ovum collection, and somewhat equivocal after 4 day culture, high feeding levels prior to ovulation reduced the proportion of ova (0.16 vs. 0.58) developing to or beyond the expanding blastocyst stage after 7 day culture. Quantitative indices of cell division and protein synthesis confirmed this. In conclusion, excessive feeding during follicular recruitment and oocyte maturation in superovulated ewes imparts a legacy of embryonic loss and developmental retardation.  相似文献

In flowering plants, the process of pollen germination and tube growth is required for successful fertilization. A pollen receptor kinase from tomato (Solanum lycopersicum), LePRK2, has been implicated in signaling during pollen germination and tube growth as well as in mediating pollen (tube)-pistil communication. Here we show that reduced expression of LePRK2 affects four aspects of pollen germination and tube growth. First, the percentage of pollen that germinates is reduced, and the time window for competence to germinate is also shorter. Second, the pollen tube growth rate is reduced both in vitro and in the pistil. Third, tip-localized superoxide production by pollen tubes cannot be increased by exogenous calcium ions. Fourth, pollen tubes have defects in responses to style extract component (STIL), an extracellular growth-promoting signal from the pistil. Pollen tubes transiently overexpressing LePRK2-fluorescent protein fusions had slightly wider tips, whereas pollen tubes coexpressing LePRK2 and its cytoplasmic partner protein KPP (a Rop-GEF) had much wider tips. Together these results show that LePRK2 positively regulates pollen germination and tube growth and is involved in transducing responses to extracellular growth-promoting signals.  相似文献

In the NarthecoceratidaeFlower (1958) there is variation of structures within the tubes of the endosiphuncle which may be empty, or may contain endocones or “bracket diaphragms”,Winstonocera teicherti gen. et. sp. nov. shows numerous diaphragms from which tubes extend a short distance forward. It is suggested that the “bracket diaphragms” are similar structures. It is further suggested that the structures within the tube were originally formed in soft tissue, which was later calcified. This study reports the design, realization, and characterization of a multi-pole magnetic tweezers that enables us to maneuver small magnetic probes inside living cells. So far, magnetic tweezers can be divided into two categories: I), tweezers that allow the exertion of high forces but consist of only one or two poles and therefore are capable of only exerting forces in one direction; and II), tweezers that consist of multiple poles and allow exertion of forces in multiple directions but at very low forces. The magnetic tweezers described here combines both aspects in a single apparatus: high forces in a controllable direction. To this end, micron scale magnetic structures are fabricated using cleanroom technologies. With these tweezers, magnetic flux gradients of nablaB = 8 x 10(3) T m(-1) can be achieved over the dimensions of a single cell. This allows exertion of forces up to 12 pN on paramagnetic probes with a diameter of 350 nm, enabling us to maneuver them through the cytoplasm of a living cell. It is expected that with the current tweezers, picoNewton forces can be exerted on beads as small as 100 nm.  相似文献

Lembos websteri Bate and Corophium bonnellii Milne Edwards are tubicolous. The morphology of tubes collected from Laminaria holdfasts is described and contrasted with that of tubes built amongst filamentous algae in a wave-sheltered environment. In both species tube length is a function of animal length. Corophium build tubes with openings raised above the substratum more commonly than Lembos, and will construct mats of tubes away from solid surfaces; Lembos tend to build against solid surfaces. Tube construction appears to depend upon the ‘knitting’ activity of the first two pairs of peraeopods, utilizing a secretion containing acid and sulphated mucopolysaccharides. The process of tube construction on substrata of known granulometry and the morphology of tubes built in the laboratory are described. The burrowing capabilities of both species are compared. On sand, Lembos tend to build on the sediment surface, whilst Corophium tend to burrow into the substratum. Corophium begin tube building after transference to a new situation much sooner than Lembos, with female Lembos starting to build sooner than male Lembos, Corophium can build a covering tube in < 15 min. In the absence of extraneous material, both species line glass tubes with a meshwork of mucoid filaments. In empty dishes, mucoid tubes are rarely built. The ability of Lembos to colonize wave-exposed sites may be related to its preference for more sheltered spaces within kelp holdfasts.  相似文献

We investigated the possibility of interspecific water transfer in an alfalfa (Medicago sativa L.) and maize (Zea mays L.) association. An alfalfa plant was grown through two vertically stacked plastic tubes. A 5 centimeter air gap between tubes was bridged by alfalfa roots. Five-week old maize plants with roots confined to the top tube were not watered, while associated alfalfa roots had free access to water in the bottom tube (the −/+ treatment). Additional treatments included: top and bottom tubes watered (+/+), top and bottom tubes droughted (−/−), and top tube droughted after removal of alfalfa root bridges and routine removal of alfalfa tillers (−*). Predawn leaf water potential of maize in the −/+ treatment fell to −1.5 megapascals 13 days after the start of drought; thereafter, predawn and midday potentials were maintained near −1.9 megapascals. Leaf water potentials of maize in the −/− and −* treatments declined steadily; all plants in these treatments were completely desiccated before day 50. High levels of tritium activity were detected in water extracted from both alfalfa and maize leaves after 3H2O was injected into the bottom −/+ tube at day 70 or later. Maize in the −/+ treatment was able to survive an otherwise lethal period of drought by utilizing water lost by alfalfa roots.  相似文献

Measurements of heat collection rate and heat loss coefficient are crucial for the evaluation of in service water-in-glass evacuated tube solar water heaters. However, conventional measurement requires expensive detection devices and undergoes a series of complicated procedures. To simplify the measurement and reduce the cost, software based on artificial neural networks for measuring heat collection rate and heat loss coefficient of water-in-glass evacuated tube solar water heaters was developed. Using multilayer feed-forward neural networks with back-propagation algorithm, we developed and tested our program on the basis of 915measuredsamples of water-in-glass evacuated tube solar water heaters. This artificial neural networks-based software program automatically obtained accurate heat collection rateand heat loss coefficient using simply "portable test instruments" acquired parameters, including tube length, number of tubes, tube center distance, heat water mass in tank, collector area, angle between tubes and ground and final temperature. Our results show that this software (on both personal computer and Android platforms) is efficient and convenient to predict the heat collection rate and heat loss coefficient due to it slow root mean square errors in prediction. A laboratory culture of an Israeli benthic midge, Chironomus luridus, was exposed to two chemicals: copper and monochloramine. The objective of this study was to determine the protective nature of Chironomus' larval tube. Three experimental conditions were tested: larva with sand tubes, with silt tubes and without tubes. Larvae without tubes were significantly more sensitive to copper and chloramine than larvae that had sand or silt as tube building substrate. The tubes protected the insects against chemicals throughout 14 days of exposure time. Silt tubes had higher protective value than sand tubes, especially when exposed to copper for a short period of time (LC50/ 24 h, with silt, sand, or none: 80.0, 7.0 and 3.4 mg l–1 copper, respectively). C. luridus seemed to be better protected against copper than against chloramine (LC50/ 24 h, with silt, sand, or none: 12.2, 6.4 and 3.7 mg l–1 chloramine, respectively). The acute toxicity of copper to chironomid larvae was investigated using a cytochemical method. Larva in silt tubes had significantly higher non-specific esterase activity in midgut cells than larvae without tubes. We conclude that, in addition to its role in feeding, respiration and anti-predation shelter, the C. luridus tube protects its inhabitant from toxic substances.  相似文献
