Two nitrate reductase (NR) mutants were selected for low nitrate reductase (LNR) activity by in vivo NR microassays of M2 seedlings derived from nitrosomethylurea-mutagenized soybean (Glycine max [L.] Merr. cv Williams) seeds. The mutants (LNR-5 and LNR-6) appeared to have normal nitrate-inducible NR activity. Both mutants, however, showed decreased NR activity in vivo and in vitro compared with the wild-type. In vitro FMNH2-dependent nitrate reduction and Cyt c reductase activity of nitrate-grown plants, and nitrogenous gas evolution during in vivo NR assays of urea-grown plants, were also decreased in the mutants. The latter observation was due to insufficient generation of nitrite substrate, rather than some inherent difference in enzyme between mutant and wild-type plants. When grown on urea, crude extracts of LNR-5 and LNR-6 lines had similar NADPH:NR activities to that of the wild type, but both mutants had very little NADH:NR activity, relative to the wild type. Blue Sepharose columns loaded with NR extract of urea-grown mutants and sequentially eluted with NADPH and NADH yielded a NADPH:NR peak only, while the wild-type yielded both NADPH: and NADH:NR peaks. Activity profiles confirmed the lack of constitutive NADH:NR in the mutants throughout development. The results provide additional support to our claim that wild-type soybean contains three NR isozymes, namely, constitutive NADPH:NR (c1NR), constitutive NADH:NR (c2NR), and nitrate-inducible NR (iNR).  相似文献

NADH:nitrate reductase (EC 1.6.6.1) and NAD(P)H:nitrate reductase (EC 1.6.6.2) were purified from wild-type soybean (Glycine max [L.] Merr., cv Williams) and nr1-mutant soybean plants. Purification included Blue Sepharose- and hydroxylapatite-column chromatography using acetone powders from fully expanded unifoliolate leaves as the enzyme source.

Two forms of constitutive nitrate reductase were sequentially eluted with NADPH and NADH from Blue Sepharose loaded with extract from wild-type plants grown on urea as sole nitrogen source. The form eluted with NADPH was designated c1NR, and the form eluted with NADH was designated c2NR. Nitrate-grown nr1 mutant soybean plants yielded a NADH:nitrate reductase (designated iNR) when Blue Sepharose columns were eluted with NADH; NADPH failed to elute any NR form from Blue Sepharose loaded with this extract. Both c1NR and c2NR had similar pH optima of 6.5, sedimentation behavior (s20,w of 5.5-6.0), and electrophoretic mobility. However, c1NR was more active with NADPH than with NADH, while c2NR preferred NADH as electron donor. Apparent Michaelis constants for nitrate were 5 millimolar (c1NR) and 0.19 millimolar (c2NR). The iNR from the mutant had a pH optimum of 7.5, s20,w of 7.6, and was less mobile on polyacrylamide gels than c1NR and c2NR. The iNR preferred NADH over NADPH and had an apparent Michaelis constant of 0.13 millimolar for nitrate.

Thus, wild-type soybean contains two forms of constitutive nitrate reductase, both differing in their physical properties from nitrate reductases common in higher plants. The inducible nitrate reductase form present in soybeans, however, appears to be similar to most substrateinduced nitrate reductases found in higher plants.

The effect of tungsten on the development of endogenous and nitrate-induced NADH- and FMNH2-linked nitrate reductase activities in primary leaves of 10-day-old soybean (Glycine max [L.] Merr.) seedlings was studied. The seedlings were grown with or without exogenous nitrate. High levels of endogenous nitrate reductase activities developed in leaves of seedlings grown without nitrate. However, no endogenous nitrite reductase activity was detected in such seedlings. The FMNH2-linked nitrate reductase activity was about 40% of NADH-linked activity. Tungsten had little or no effect on the development of endogenous NADH- and FMNH2-linked nitrate reductase activities, respectively. By contrast, in nitrate-grown seedlings, tungsten only inhibited the nitrate-induced portion of NADH-linked nitrate reductase activity, whereas the FMNH2-linked activity was inhibited completely. Tungsten had no effect on the development of nitrate-induced nitrite reductase activity. The complete inhibition of FMNH2-linked nitrate reductase activity by tungsten in nitrate-grown plants was apparently an artifact caused by the reduction of nitrite by nitrite reductase in the assay system. The results suggest that in soybean leaves either the endogenous nitrate reductase does not require molybdenum or the molybdenum present in the seed is preferentially utilized by the enzyme complex as compared to nitrate-induced nitrate reductase.  相似文献

Nitrate reductase (NR) activity and nitrite reductase (NiR) mRNA levels were monitored in Black Mexican Sweet maize (Zea mays L.) suspension cultures after the addition of nitrate. Maximal induction occurred with 20 millimolar nitrate and within 2 hours. Both NR and NiR mRNA were transiently induced with levels decreasing after the 2 hours despite the continued presence of nitrate in the medium. Neither ammonia nor chlorate prevented the induction of NR. Furthermore, removal of nitrate, followed by its readdition 22 to 48 hours later, did not result in reinduction of activity or message. NR was synthesized de novo, since cycloheximide completely blocked its induction. Cycloheximide had no effect on the induction of NiR mRNA or on the transient nature of its induction. These results are similar to those reported previously for maize seedlings.  相似文献

Nitrogen assimilation in three nitrate reductase (NR) mutants of soybean (Glycine max L. Merr. cv Williams) was studied in the growth chamber and in the field. These mutants, LNR-2, LNR-3, and LNR-4, lack the non-NO3-inducible or constitutive fraction of leaf NR activity found in wild-type plants, but this had no effect on the concentration of nitrogen accumulated when grown on NO3 in the growth chamber. Dry weight accumulation of two of the mutants (LNR-3 and LNR-4) was decreased relative to LNR-2 and wild type. In the field, LNR-2 had dry weights and nitrogen concentrations similar to the wild type at 34 and 61 days after planting, and at maturity. Acetylene reduction activities were also similar at 61 days.  相似文献

Native PAGE of Triton x-100-solubilized membranes from Bradyrhizobium japonicum strain PJ17 grown microaerobically (2% O2, v/v) in defined nitrate-containing medium resolved two catalytically active nitrate reductase (NR) species with apparent molecular masses of 160 kDa (NRI) and 200 kDa (NRII). NRI and NRII were also found in membranes from cells of strain PJ17 that were first grown in defined medium with glutamate and further incubated microaerobically in the presence of 5 mmol/l KNO3. However, only NRI was detected in cell membranes of strain PJ17 when nitrate was omitted from the microaerobic incubation medium. Four mutants unable to grow at low O2 tension in the presence of nitrate were isolated after transposon Tn5 mutagenesis. Membranes from mutants GRF110 and GRF116 showed mainly NRI, while the other two mutants, GRF3 and GRF4, expressed mostly NRII. These results indicate that the ability of B. japonicum PJ17 to grow under microaerobic conditions depends upon the presence of two membrane-bound NR enzymes whose synthesis seem to be independently induced by microaerobiosis (NRI) or by both microaerobiosis and nitrate (NRII).Abbreviations NR Nitrate reductase - M r Relative molecular mass - PMSF Phenylmethylsulfonyl fluoride  相似文献

Plasmid DNA carrying either the nitrate reductase (NR) gene or the argininosuccinate lyase gene as selectable markers and the correspondingChlamydomonas reinhardtii mutants as recipient strains have been used to isolate regulatory mutants for nitrate assimilation by insertional mutagenesis. Identification of putative regulatory mutants was based on their chlorate sensitivity in the presence of ammonium. Among 8975 transformants, two mutants, N1 and T1, were obtained. Genetic characterization of these mutants indicated that they carry recessive mutations at two different loci, namedNrg1 andNrg2. The mutation in N1 was shown to be linked to the plasmid insertion. Two copies of the nitrate reductase plasmid, one of them truncated, were inserted in the N1 genome in inverse orientation. In addition to the chlorate sensitivity phenotype in the presence of ammonium, these mutants expressed NR, nitrite reductase and nitrate transport activities in ammonium-nitrate media. Kinetic constants for ammonium (14C-methylammonium) transport, as well as enzymatic activities related to the ammonium-regulated metabolic pathway for xanthine utilization, were not affected in these strains. The data strongly suggest thatNrg1 andNrg2 are regulatory genes which specifically mediate the negative control exerted by ammonium on the nitrate assimilation pathway inC. reinhardtii.  相似文献

In the present study, experiments were performed to investigate the role of nitric oxide (NO) in magnetopriming-induced seed germination and early growth characteristics of soybean (Glycine max) seedlings under salt stress. The NO donor (sodium nitroprusside, SNP), NO scavenger (2-[4-carboxyphenyl]-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide, CPTIO), inhibitors of nitrate reductase (sodium tungstate, ST) or NO synthase (N-nitro-L-Arg-methyl ester, LNAME) and NADPH oxidase inhibitor (diphenylene iodonium, DPI) have been used to measure the role of NO in the alleviation of salinity stress by static magnetic field (SMF of 200 mT, 1 h). Salt stress (50 mM NaCl) significantly reduced germination and early growth of seedlings emerged from non-primed seeds. Pre-treatment of seeds with SMF positively stimulated the germination and consequently promoted the seedling growth. ST, LNAME, CPTIO and DPI significantly decreased the growth of seedling, activities of α-amylase, protease and nitrate reductase (NR), hydrogen peroxide (H2O2), superoxide (O2•−) and NO content in roots of seedlings emerged from non-primed and SMF-primed seeds. However, the extent of reduction was higher with ST in seedlings of SMF-primed seeds under both conditions, whereas SNP promoted all the studied parameters. Moreover, the generation of NO was also confirmed microscopically using a membrane permanent fluorochrome (4-5-diaminofluorescein diacetate [DAF-2 DA]). Further, analysis showed that SMF enhanced the NR activity and triggered the NO production and NR was maximally decreased by ST as compared to LNAME, CPTIO and DPI. Thus, in addition to ROS, NO might be one of the important signaling molecules in magnetopriming-induced salt tolerance in soybean and NR may be responsible for SMF-triggered NO generation in roots of soybean.  相似文献

The kinetics and other characteristics of nitrate reductase (NR, EC 1.6.6.1) in cowpea [Vigna unguiculata (L.) Walp.] seedlings irradiated with biologically effective UV-B radiation (280-320 nm, 3.2 W m-2 s-1) were recorded. The in vivo and in vitro NR activities were inhibited by 34 and 41 % under UV-B treatment, respectively. Both Vmax and Km for the substrate were enhanced by UV-B radiation. The Km for nitrate increased from 1.2 to 1.7 mM after the UV-B irradiation. The change in Km for NADH was from 0.12 to 0.17 mM. The increases in Km indicate that UV-B radiation seriously changes the topology of NR, particularly with respect to the nitrate and NADH binding sites. The rate of NR turnover indicates the extent of damage inflicted by UV-B radiation on the nitrate metabolism. The half-life (t1/2) of NR was reduced from 7 to 4 h in the UV-B treated seedlings. UV-B also inhibited the kinetics of nitrate uptake by plants: its Km increased from 0.08 to 0.12 mM. Growth chamber studies were conducted to assess the relationship between nitrate reductase (NR) activity and development of chlorate (KClO3) toxicity symptoms in leaflets of soybeans [Glycine max (L.) Merr.]. Fourteen day-old soybean seedlings, grown in NO3 - or urea-nutrient solutions, were exposed to various KClO3 concentrations (0 to 2.0 mM) and light levels (100, 67, 33 and 0% of full light which was 750 μE m?2s?1) for 24 h. Visual KClO3 toxicity symptoms were noted and NR activity was measured. Toxicity symptoms (interveinal chlorosis) were evident within 24 h following addition of 0.5 mM KClO3 to the nutrient solution, regardless of N nutrition, and symptom severity increased with increased KClO3 concentration (up to 2.0 mM). Leaflet NR activity was lower following 24 h KClO3 treatments at concentrations of 0.5 mM and higher, indicating that ClO3 - or some reduction product of ClO3 - likely ClO2 - was detrimental to enzyme functionality. The light study supported involvement of NR activity in KClO3 toxicity in that comparison of control and KClO3 treated plants exposed to decreased light levels revealed a decrease in NR activity of control plants parallel to a decrease in severity of KClO3 toxicity symptoms of treated plants. Urea-grown plants, which have an apparent constitutive NR enzyme, were used to verify that the KClO3 toxicity symptoms were not simply N starvation symptoms due to competition of ClO3 - and NO3 - for uptake and reduction. In vivo NR assays also ruled out that ClO3 - was decreasing NR activity through competition with NO3 - for reduction sites. The close relationship between KClO3 toxicity symptoms and NR activity, in response to light treatments, suggested that KClO3 toxicity symptoms were associated with reduction of ClO3 - to ClO2 - by the NR enzyme. However, the possibility that a more direct photochemical reaction occurred in the presence of KClO3 to produce the toxicity symptoms could not be ruled out.  相似文献

The objective of this study was to identify factors which limit leaf nitrate reductase (NR) activity as decline occurs during flowering and beginning seed development in soybean (Glycine max [L.] Merr. cv Clark). Level of NR enzyme activity, level of reductant, and availability of NO3 as substrate were evaluated for field-grown soybean from flowering through leaf senescence. Timing of reproductive development was altered within one genotype by (a) exposure of Clark to an artificially short photoperiod to hasten flowering and podfill, and (b) the use of an early flowering isoline. Nitrogen (N) was soil-applied to selected plots at 500 kilograms per hectare as an additional variable. Stem NO3 concentration and in vivo leaf NR activity were significantly correlated (R2 = 0.69 with nitrate in the assay medium and 0.74 without nitrate in the medium at P = 0.001) across six combinations of reproductive and soil N-treatment. The supply of NO3 from the root to the leaf tissue was the primary limitation to leaf NR activity during flowering and podfill. Levels of NR enzyme and reductant were not limiting to leaf NR activity during this period.  相似文献

The plant growth, nitrogen absorption, and assimilation in watermelon (Citrullus lanatus [Thunb.] Mansf.) were investigated in self-grafted and grafted seedlings using the salt-tolerant bottle gourd rootstock Chaofeng Kangshengwang (Lagenaria siceraria Standl.) exposed to 100 mM NaCl for 3 d. The biomass and NO3 uptake rate were significantly increased by rootstock while these values were remarkably decreased by salt stress. However, compared with self-grafted plants, rootstock-grafted plants showed higher salt tolerance with higher biomass and NO3 uptake rate under salt stress. Salinity induced strong accumulation of nitrate, ammonium and protein contents and a significant decrease of nitrogen content and the activities of nitrate reductase (NR), nitrite reductase (NiR), glutamine synthetase (GS), and glutamate synthase (GOGAT) in leaves of self-grafted seedlings. In contrast, salt stress caused a remarkable decrease in nitrate content and the activities of GS and GOGAT, and a significant increase of ammonium, protein, and nitrogen contents and NR activity, in leaves of rootstock-grafted seedlings. Compared with that of self-grafted seedlings, the ammonium content in leaves of rootstock-grafted seedlings was much lower under salt stress. Glutamate dehydrogenase (GDH) activity was notably enhanced in leaves of rootstock-grafted seedlings, whereas it was significantly inhibited in leaves of self-grafted seedlings, under salinity stress. Three GDH isozymes were isolated by native gel electrophoresis and their expressions were greatly enhanced in leaves of rootstock-grafted seedlings than those of self-grafted seedlings under both normal and salt-stress conditions. These results indicated that the salt tolerance of rootstock-grafted seedlings might (be enhanced) owing to the higher nitrogen absorption and the higher activities of enzymes for nitrogen assimilation induced by the rootstock. Furthermore, the detoxification of ammonium by GDH when the GS/GOGAT pathway was inhibited under salt stress might play an important role in the release of salt stress in rootstock-grafted seedlings.  相似文献

The nitrate reductase in the mature root extract of 3-day maize (Zea mays) seedlings was relatively labile in vitro. Insoluble polyvinylpyrrolidone used in the extraction medium produced only a slight increase in the stability of the enzyme. Mixing the mature root extract with that of the root tip promoted the inactivation of nitrate reductase in the latter. The inactivating factor in the mature root was separated from nitrate reductase by (NH4)2SO4 precipitation. Nitrate reductase was found in the 40% (NH4)2SO4 precipitate, while the inactivating factor was largely precipitated by 40 to 55% (NH4)2SO4. The latter fraction of the mature root inactivated the nitrate reductase isolated from the root tip, mature root, and scutellum. The inactivating factor, which has a Q10 15 to 25 C of 2.2, was heat labile, and hence has been designated as a nitrate reductase inactivating enzyme. The reduced flavin mononucleotide nitrate reductase was also inactivated, while an NADH cytochrome c reductase in nitrate-grown seedlings was inactivated but at a slower rate. The inactivating enzyme had no influence on the activity of nitrite reductase, glutamate dehydrogenase, xanthine oxidase, and isocitrate lyase. The activity of the nitrate reductase inactivating enzyme was not influenced by nitrate and was also found in the mature root of minus nitrate-grown seedlings.  相似文献

The activities of nitrate reductase (NR) and nitrite reductase (NiR) and production of indole-3-acetic acid (IAA) by symblotic nitrate tolerant Tn5 mutant AC-10 of Cicer-Rhizobium strain F-75 and mutants BC-35 and BC-46 of strain G36-84 developed earlier, have been studied under ex planta condition. The rhizobiaI mutants and their parental strains were grown with nitrate (0.0, 0.5, 1, 2 or 4 mM), aerobically and microaerobically. The overall activities of NR were 70–91% lower in aerobically grown and 78–87% lower in microaerobically grown mutant cells compared to their parental strains. Similarly, the overall activities of NiR were 36–55% and 27–37% lower in aerobically and microaerobically grown mutant cells, respectively, compared to their parental strains. On the contrary, the overall production of IAA in the culture medium by aerobically grown mutant cells was significantly higher compared to their parental strains. Based on these results, it has been suggested that impaired NR activity and a favourable NiR/NR ratio preventing nitrite accumulation in the rhizobial mutants, may be responsible for imparting nitrate tolerance to chickpea - Rhizobium symbiotic system.  相似文献

Biomass production, pattern of nodulation, nutrient uptake, net photosynthetic rate (Pn), leaf temperature (Tleaf), leaf nitrate reductase (NR) activity and free proline of Dalbergia sissoo seedlings planted in containers with 120 kg soil were studied under different water stress levels to assess the productive potential of the species in dry areas. Seedlings were irrigated at 20 mm (W1), 14 mm (W2), 10 mm (W3), 8 mm (W4) throughout the experimental period to maintain the respective treatment up to the lowest soil water content of 7.43%, 5.64%, 4.30% and 3.23%, respectively. There was a treatment (W5) in which seedling were irrigated once to −0.03 MPa and left without re-irrigation. Decreased irrigation level resulted in lowering of leaf water potential (LWP), net photosynthetic rate (Pn), total number of root nodules and nodule dry mass and nitrogen uptake in the seedling. Pn, leaf nitrate reductase (NR) activity and seedling biomass were highest in W1 indicating a positive relations of NR activity with CO2 assimilation and biomass production. The decrease in Pn, leaf NR activity and LWP was sharp at W3 onwards. Monthly changes in the values of Pn, Tleaf and NR activity indicate environmental effect on these physiological variables. Proline was detected only in the seedlings of W3, W4 and W5 treatments after February and was highest in the seedlings of W5 treatment. The study suggests that severe water deficit adversely affect physiological and biochemical processes that resulted in reduced growth, nutrient uptake and biomass productivity in D. sissoo seedlings. Re-irrigation above W3 level is recommended for this species.  相似文献

The activity and decay characteristics of nitrate reductase from wheat (Triticum aestivum) were studied in crude, partially-purified and highly-purified preparations. The decay of nitrate reductase activity in crude extracts was due to spontaneous dissociation of the enzyme and to the effects of two decay factors, one present in the 0–30% and the other in the 50–70% saturated (NH4)2SO4 fraction of a crude extract. Low rates of factor-mediated NR decay in vitro were associated with high levels of NR activity in vivo.  相似文献

The objectives of this study were to select and initially characterize mutants of soybean (Glycine max L. Merr. cv Williams) with decreased ability to reduce nitrate. Selection involved a chlorate screen of approximately 12,000 seedlings (progeny of mutagenized seed) and subsequent analyses for low nitrate reductase (LNR) activity. Three lines, designated LNR-2, LNR-3, and LNR-4, were selected by this procedure.

In growth chamber studies, the fully expanded first trifoliolate leaf from NO3-grown LNR-2, LNR-3, and LNR-4 plants had approximately 50% of the wild-type NR activity. Leaves from urea-grown LNR-2, LNR-3, and LNR-4 plants had no NR activity while leaves from comparable wild-type plants had considerable activity; the latter activity does not require the presence of NO3 in the nutrient solution for induction and on this basis is tentatively considered as a constitutive enzyme. Summation of constitutive (urea-grown wild-type plants) and inducible (NO3-grown LNR-2, LNR-3, or LNR-4 plants) leaf NR activities approximated activity in leaves of NO3-grown wild-type plants. Root NR activities were comparable in wild-type and mutant plants grown on NO3, and roots of both plant types lacked constitutive NR activity when grown on urea. In both growth chamber- and field-grown plants, oxides of nitrogen [NO(x)] were evolved from young leaves of wild-type plants, but not from leaves of LNR-2 plants, during in vivo NR assays. Analysis of leaves from different canopy locations showed that constitutive NR activity was confined to the youngest three fully expanded leaves of the wild-type plant and, therefore, on a total plant canopy basis, the NR activity of LNR-2 plants was approximately 75% that of wild-type plants. It is concluded that: (a) the NR activity in leaves of NO3-grown wild-type plants includes both constitutive and inducible activity; (b) the missing NR activity in LNR-2, LNR-3, and LNR-4 leaves is the constitutive component; and (c) the constitutive NR activity is associated with NO(x) evolution and occurs only in physiologically young leaves.

Soybean (Glycine max L. Merr.) leaves contain two forms of nitrate reductase (NR)—NAD(P)H:NR and NADH:NR. Wild-type (cv Williams), nr1 mutant and an unrelated cultivar (Prize) were grown with either no N source or with nitrate. Crude extracts were assayed for NR activities and the enzyme forms were purified on blue Sepharose. Analyses were done by polyacrylamide gel electrophoresis and `Western blotting' using antibodies specific for NR. NAD(P)H:NR was identified as the constitutive NR present in wild-type and Prize, but was absent from the mutant. All three soybean lines contained nitrate-inducible NADH:NR with highest activity at pH 7.5. The results showed that NAD(P)H:NR and constitutive NR were one in the same and confirmed the presence of NADH:NR with pH 7.5 optimum.  相似文献

The objective of this experiment was to elucidate the manner in which N metabolism is influenced by S nutrition. Maize (Zea mays L.) seedlings supplied with Hoagland solution minus SO42− exhibited S deficiency symptoms 12 days after emergence. Prior to development of these symptoms, a decline in leaf blade nitrate reductase (NR, EC 1.6.6.1) activity was observed in S-deprived seedlings compared to normal seedlings. Twelve days after emergence, in vitro NR activity was diminished 50% compared to normal seedlings. Glutamine synthetase (EC 6.3.1.2) and NAD-glutamate dehydrogenase (EC 1.4.1.2) activities were less severely affected (19 and 13%, respectively, at day 12). NADP-glutamate dehydrogenase (EC 1.4.1.4) activity and leaf blade fresh weight were not altered by S deprivation. Concentrations of soluble protein and chlorophyll (a and b) in leaf blades were reduced 18 and 25%, respectively, at day 12. A significantly higher concentration of NO3-N was observed for leaf blade and stem (culms, leaf sheaths, and unfurled leaves) fractions (46 and 31%, respectively) in S-deprived plants. In contrast to the other parameters measured, NR activity in S-deprived seedlings could be readily restored to the normal level by addition of SO42−. The apparent preferential effect of S deprivation on NR activity could be causally related to the observed changes in NO3-N and soluble protein concentration.  相似文献

After x-ray irradiation, 13 mutants of Chlorella sorokiniana incapable of using NO3 as N source were isolated using a pinpoint method. Using immunoprecipitation and Western blot assays, no nitrate reductase was found in five strains while in eight mutants the enzyme was detected. The latter strains contained different patterns of nitrate reductase partial reactions. All isolates were of the nia-type as indicated by the inducibility of purine hydroxylase I and by complementation of nitrate reductase activity in the Neurospora crassa mutant Nit-1. A restoration of NADP-nitrate reductase in Nit-1 was also obtained with NH4+-grown cells indicating that Mo-cofactor is constitutive in Chlorella. Complementation experiments among the Chlorella mutants resulted in restoration of NADH-nitrate reductase activity. The characteristics of some of the Chlorella mutants are discussed in view of an improper orientation of Mo-cofactor in the residual nitrate reductase protein.  相似文献
