The viability of seeds is associated with ageing and storageconditions. A loss of viability is accompanied by slow germination,reduced growth, and a decline in protein and poly(A)+RNA synthesis.This paper reports on the activity of poly(A) polymerase indry and germinating embryos of Triticum durum Desf. cv. Cappellicaryopses of different ages and viability. The enzyme was presentas a single form during ageing and germination. The poly(A)polymerase was active at decreasing levels in all aged dry embryos,in parallel with loss of viability. Its activity strongly increasedduring the germination only in viable embryos. The observedincrease was due to de novo synthesis of the enzyme. Poly(A)polymerase synthesis was low during germination of less viableembryos and absent in older ones. Reduced poly(A) polymeraseactivity in dry or germinated wheat embryos may cause a shorteningof poly(A) chains in vitro and a decline in poly(A)+RNA synthesis.Copyright1995, 1999 Academic Press Triticum durum Desf. cv. Nutrient absorption in wheat and sugarbeet was studied duringpre-emergence growth by adding 0, 7, 10.5 or 14 mol m-3nitrogen(N) to the growth medium. Seedling growth and carbon, N and15Ncontents of the seedling parts were measured. Differences betweenthe natural abundance of15N in seeds and in nutrient solutionwere used to determine the proportion of N in the organs originatingfrom seed reserves and from absorption. Absorption began laterfor wheat than for sugarbeet and had less effect on seedlinggrowth. The absorbed N was found mainly in roots. Compared towheat, sugarbeet seedling N content was greatly altered andthe hypocotyl showed increased elongation when nutrients wereadded. Most of the absorbed N was found in the radicle and hypocotylwith less in the cotyledons. The relationship between coleoptile elongation and survivalvs. alcoholic fermentation of rice under anoxia is examinedusing eight cultivars differing in submergence tolerance. Anoxiawas imposed on either 1 or 4 d aerated seeds either by N2 flushingsubmerged tissues or by incubating tissues in stagnant deoxygenatedagar at 0·1% w/v; the latter simulated the stagnant conditionsof waterlogged soil. Two cultivars that were most tolerant tosubmergence also had the greatest tolerance to anoxia, whilea submergence intolerant cultivar was also intolerant to anoxia. Coleoptile growth under anoxia was related to rates of ethanolsynthesis (RE), however differences between growth during anoxiaand survival after anoxia indicated that post-anoxic injurymay also be important in rice seeds exposed to anoxia. The correlationbetween coleoptile growth and RE measured on a tissue basisusing intact seeds was r2 = 0·67 among six varietiesover 0-3 d anoxia. This correlation improved to about r2 = 0·85using RE of (embryos plus coleoptiles) over 0-3 d, or coleoptilesat 3 d after anoxia. Coleoptile growth of individual seeds wasusually poorly correlated to RE in these cultivars at 2-3 dafter anoxia. When coleoptiles of similar lengths were obtainedfrom different cultivars using 4 d aerated seeds, there weredifferences in RE and coleoptile growth which were related tocoleoptile growth during 3 or 5 d anoxia, either on a tissue(r2 = 0·85) or a fresh weight basis (r2 = 0·70-0·97respectively). To elucidate the mechanism of the phosphorylation of myo-inositolin the process of phytate formation, feeding experiments oforthophosphate-32P and myo-inositol-3H in the ripening grainsof rice and wheat were performed. It was found that 32P and3H were incorporated into myo-inositol mono- and hexa-phosphates.The same results were obtained when a mixture of "cold" myo-inositolpolyphosphates was administered to the grains before feedingphosphate-32P. Based on these results it is concluded that phosphorylationof free myo-myo-inositol in the formation of phytate does nottake place in a stepwise fashion but may proceed through anunknown myo-inositol derivative. Inorganic cation concentrations were measured in shoots of hexaploidbread wheat (Triticum aestivum L.) and its presumed ancestorsgrown at 100 mol m–3 external NaCl. Aegilops squarrosaand T. aestivum had high K/Na ratios while T. dicoccoides andAe. speltoides had low K/Na ratios. T. monococcum although havinga high K/Na ratio, had the highest total salt load of the fivespecies tested. The effect of the D genome (from Ae. squarrosa)was further investigated in seedlings of synthetic hexaploidwheats, and was again found to improve cation selectivity. Differentresponses were obtained from root and shoot tissue in this experiment.One synthetic hexaploid and its constituent parents were grownto maturity at 100 mol m-3 NaCl and the yields recorded. Despitecomplications due to increased tillering in the stressed hexaploid,it was possible to show that the addition of the D genome enhancedyield characteristics in the hexaploid wheat. An experimentwith synthetic hexaploids derived from the tetraploid wheatvariety "Langdon" and several Ae. squarrosa accessions revealeddifferences in vegetative growth rates between the differentsynthetic hexaploids in the presence or absence of 150 or 200mol m–3 external NaCl. The possibility of transferringsalt tolerance genes from Ae. squarrosa to hexaploid wheat usingsynthetic hexaploids as bridging species is discussed. Biochemical properties, i.e. endogenous abscisic acid, proline,sugars, respiration, adenosine phosphates and adenylate energycharge, and growth and moisture content were measured duringthe development of seeds of Machilus thunbergii. As dry matteraccumulated in the embryo during development, moisture content,ABA, proline, respiration and sugars all declined. At maturity,the dry mass of the seeds failed to attain a plateau beforethe period of natural seed shedding; the axis and cotyledonsreached moisture contents of 58 and 45%, respectively. Dryingof immature seeds at 73% relative humidity and 25 °C for30 d resulted in a complete loss of viability at all developmentalstages tested with the exception of mature seeds that were ableto tolerate a 5% decrease in moisture content before germinationdeclined. ABA was detected in all embryos tested, with a maximum value16.·16 µg g-1 d. wt about midway through development.Although the presence of ABA induced no tolerance to desiccationof mature seeds, it did coincide with decreased content of waterin the developing seeds and decreased respiration. Desiccationdamage of M. thunbergii seeds occurred when moisture contentwas still high (45%) and this damage was not related to theabsence of oligosaccharides in the mature seeds. We concludethat developing embryos and mature seeds of M. thunbergii haveproperties common to many recalcitrant seeds, with seeds beingsensitive to desiccation at all stages, having a prominent ABApeak, little proline, lacking oligosaccharides, and specifically,little dormancy and a moderate rate of respiration of matureseeds (0·9 µmol O2 min-1 g-1 f. wt). Adenosinetriphosphate content and energy charge decreased from stagefour to stage eight of seed development, then increased againto 103 nmol g-1 d. wt and 0·73, respectively, in matureseeds. The effects of different concentrations of L--aminooxy-ß-phenyIpropionicacid (AOPP), an analog of L-phenylalanine, on the activity ofphenylalanine ammonia-lyase (PAL, EC 4.3.1.5 [EC] ) and the growthof radicles in 24 h old germinating lettuce (Lactuca salivaL.) seeds were investigated. AOPP causes a significant inhibitionof PAL activity in the seeds (85% inhibition at 104 M). It alsocauses a stimulation of radicle growth at that concentration.The results show that the inhibition of PAL by AOPP may be dueto an irreversible binding of the inhibitor to the enzyme leadingto its inactivation. AOPP also inhibits ethylene biosynthesisin germinating lettuce seeds which could probably explain thestimulation of radicle growth in these seeds. The enzyme shows typical Michaelis-Menten kinetics. The Km forL-phenylalanine is 4.2 x 105 M. The enzyme does not show anytyrosine ammonia-lyase activity. Various substrate analogs suchas D-phenylalanine, p-fluorophenylalanine, ß-phenyllacticacid, tryptophan and the product of the enzyme reaction, trans-cinnamicacid, inhibit the enzyme competitively. The relationship between coleoptile elongation and alcoholicfermentation of rice under anoxia is examined using seeds either:(a) N2 flushed during submergence, (b) incubated in stagnantdeoxygenated agar at 0·1% w/v to simulate the stagnantconditions of waterlogged soil, or (c) incubated in waterloggedsoil. Coleoptile elongation growth was greater for N2 flushing> stagnant agar > soil; seed survival was also greatestin this order over 1-5 d. Ethanol concentrations in coleoptiles and intact seeds (cv.IR42) were approximately 300 and 100 mol m-3 respectively whenseeds were grown 3 d in stagnant agar, however 92% of the ethanolin seeds diffused into the external medium when solutions weremixed for 5-10 s. Coleoptile growth under anoxia was relatedto rates of ethanol synthesis (RE) in different treatments;there was greater coleoptile growth and RE for seeds in N2 flushedsolutions than in stagnant deoxygenated agar. Coleoptile growthof individual seeds was also related to the RE of each seedat 2-3 d after anoxia (r2 = 0·46). Analysis of different tissues was important in evaluating growthand metabolism of coleoptiles. Although the coleoptile onlyaccounted for 0·7% of seed dry weight at 3 d after anoxia,it contained 21% of the ethanol produced by rice seeds. Therewere also three-fold higher rates of RE on a fresh weight basisin expanding tissues in the base of the coleoptile relativeto the elongated tissues at the apex. Immature Glycine max (L.) Merrill seeds, initially between 50and 450 mg f. wt, were grown and matured successfully in vitro.Excised seeds were floated in a liquid medium containing 5 percent sucrose, minerals and glutamine in flasks incubated at25 °C under 300 to 350 µE m–2 s–1 fluorescentlight. During 16 to 21 d in culture, seeds grew to a matured. wt of 100 to 600 mg per seed at an average rate of 5 to 25mg d. wt per seed d–1 depending on initial size. Growthrates were maximal during the first 8 to 10 d in vitro but declinedwith loss of green colour in the cotyledons. Seed coats rupturedwith rapid cotyledon expansion during the first 2 d in culture.Embryos were tolerant to desiccation and 80 to 90 per cent germinatedif removed from culture before complete loss of green colour.The growth of excised seeds in vitro exceeded the growth ofseeds in detached pods, but when windows were cut in pods topermit direct exposure of seeds to the medium, seed growth wascomparable. We studied the course of early leaf area expansion and specificleaf area (SLA) in potato (Solanum tuberosum L.) and wheat (Triticumaestivum L.) genotypes and tested whether air temperature explainsdifferences in these courses within different environments.Such knowledge can be used to improve crop growth modelling.The relative rate of leaf area expansion (RL) of potato andwheat decreased with thermal time, but was nearly linear upto a leaf area index (L) of 1.0. TheRL (L < 1; mean: 17.9x 10-3°C-1 d-1) of potato showed an interaction betweengenotype and environment, and varied with year. TheRL (L <1; mean: 7.1 x 10-3°C-1 d-1) of winter wheat was lower thanthat of spring wheat (mean: 10.9 x 10-3°C-1 d-1), and bothvaried with year. SLAof potato increased nearly linearly withthermal time from 5 to 15 m2 kg-1at 50% emergence, to 20 to25 m2 kg-1at 155°Cd, and then decreased slightly. The SLAofboth winter and spring wheat began at 16 to 23 m2 kg-1and inmost cases increased slightly with thermal time. In potato,regression parameters of SLAwith thermal time were affectedby environment (management conditions and year) and genotype;in wheat they were affected by environment (year and site).Treatment effects on RLof potato were not correlated with thoseon SLA , and were only partly correlated for wheat. Thereforewe conclude that the early foliar expansion of potato is associatedwith a strong increase in SLA , and not so for wheat. Incorporation of labelled mevalonate into abscisic acid (ABA)has been demonstrated in the cotyledons of mature avocado seeds,embryos and endosperms of developing wheat seeds, and avocadostems. The increase in ABA concentration on wilting parallelsthe increased incorporation of [2–14C)mevalonate intoABA in avocado leaves and stems, suggesting that the increasein ABA content occurs by synthesis rather than by release froma stored precursor. Incorporation of [2–14C]mevalonateby avocado mesocarp segments is unaffected by an 18 per centwater loss. The ABA content of roots was hardly affected bya 30 per cent water loss, indicating that the wilt-activatedmechanism is not fully operative in these tissues. Submerged Ceratophyllum plants and submerged parts of Callitricheshoots show a twofold increase in ABA content on wilting whereasthe aerial rosettes of the latter plant show a sixfold increase.This suggests that the occurrence of the wilt-induced mechanismis affected by previous growth conditions as well as by themorphology of the tissue.  相似文献

During in vitro growth and maturation of soybean seeds, cessationof embryo growth and dry weight accumulation occurred in thepresence of abundant C and N nutrients. Axis followed by cotyledontissues changed from green to yellow, and post-harvest germinationpotential declined if cultured after yellowing of axis tissues.A tissue specific accumulat;on of methanol occurred during thein vitro culture of immature seeds (i.e. initially 50 to 70mg fresh weight) to maturity in liquid medium. Methanol accumulatedto 3.0 g m–3 or 50 µg seed–1 in the medium,while methanol decreased from 37 to about 3.0 µg g–1fresh weight in cotyledons. By contrast, axis tissues increased20-fold in methanol concentration to 90 µg g–1 during20 d in culture. Ethanol was present only in trace amounts inaxis tissues and medium. Addition of exogenous methanol vapourto in situ grown seeds during precocious maturation decreasedsubsequent seedling vigour and germination with increasing levelsof exposure. Methanol accumulation in axis tissues during thegermination phase was not correlated with high temperature andtissue water content treatments which simulated pre-harvestdeterioration of seeds. However, the accumulation of methanolduring in vitro seed development and maturation in liquid culturemay contribute to reduced post-harvest germination performance. Intact wild-type tomato (Lycopersicon esculentum cv. Moneymaker)seeds do not complete germination to the same percentage orat the same speed as intact ABA-deficient sitiens (sitw) mutantseeds when seeds of both genotypes are imbibed on polyethyleneglycol (PEG) solutions of –0.3 to –1.5 MPa osmoticpotential. However, if the thicker testas of wild-type seedsare removed (stripped) from the micropyle without damaging theendosperm, both the percentage and speed of germination at lowexternal water potential are similar to that of sitw mutantseeds. Removing the micropylar end of the testa from sitw seedsdid not enhance either the speed or percentage of germinationon PEG solution. Despite similar germination percentage and speed between strippedwild-type seeds and either stripped or intact sitw seeds underosmotic stress, some differences in seed metabolism are evidentbetween genotypes. Yield stress threshold (Y) and volumetric extensibility () arethe rheological properties that appear to control root growth.In this study they were measured in wheat roots by means ofparallel measurement of the growth rate (r) of intact wheatroots and of the turgor pressures (P) of individual cells withinthe expansion zone. Growth and turgor pressure were manipulatedby immersion in graded osmoticum (mannitol) solutions. Turgorwas measured with a pressure probe and growth rate by visualobservation. The influence of various growth conditions on Yand was investigated; (a) At 27 °C.In 0.5 mol m–3 CaCl2 r, P, Y and were20.7±4.6 µm min–1, 0.77±0.05 MPa,0.07±0.03 MPa and 26±1.9 µm min–1MPa–1 (expressed as increase in length), respectively.Following 24 h growth in 10 mol m–3 KC1 these parametersbecame 12.3±3.5 µm min–1, 0.72±0.04MPa, 0.13±0.01 MPa and 21±0.7 µm min–1MPa–1. After 24 h osmotic adjustment in 150 mol m–3mannitol/0.5 mol m–3 CaCl2 r= 19.6±4.2 µmmin–1, P = 0.68±0.05 MPa and Y and were 0.07±0.04MPa and 30±0.2 µm min–1 MPa–01, respectively.After 24 h growth in 350 mol m–3 mannitol/0.5 mol m–3CaCl2 r= 13.3±4.1 µm min–1, P= 0.58±0.07MPa, Y=0.12±0.01 MPa and ø 32±0.2 tim min–1MPa–1. During osmotic adjustment in 200 mol m–3mannitol/0.5 mol m–3 CaCl2, with or without KCl, the recoveryof growth rate corresponded to turgor pressure recovery (t1/2approximately 3 h). (b) At 15 °C. Lowered temperature dramatically influencedthe growth parameters which became r= 8.3±2.8 um min–1,P=0.78 MPa, r=<0.2 MPa and =15±0.1 µm min–1MPa–1. Therefore, Y and are influenced by 10 mol m–3 K+ ionsand low temperature. In each case the effective pressure forgrowth (P-Y) was large indicating that small fluctuations ofsoil water potential will not stop root elongation. Effluxes of K+ and Ca2+ from root segments of both wheat, Triticunaestivum L. cv. Capelle and mung bean, Vigna radiata (L.) Wilczek,were measured in the presence or absence of 20 mol m–3para-fluorophenylalanine (p-FPA). The results were used to estimatethe compartment contents and transmembrane K+ and Ca2+ fluxesin root cortex cells. Using the Ussing-Teorell flux equationas the criterion, it was concluded that entry of K+ from theoutside solution to the cytoplasm, and from the cytoplasm tothe vacuole were active in both wheat and mung bean. Also, inboth species, Ca2+ entered the cytoplasm passively across theplasmalemma and was actively pumped back to the external solution.However, interpretation of the direction of active transportacross the tonoplast depends upon an assumption about Ca2+ activityin the cytoplasm. The only qualitative effect of p-FPA was to alter the drivingforce for K+ influx, across the plasmalemma in wheat, from anactive to a passive one. Quantitative effects of the analoguewere seen for K+ fluxes in both wheat and mung bean and forCa2+ fluxes in wheat. The p-FPA reduced transport of K+ in bothspecies, while transport of Ca2+ was unaffected. The implicationsof these results for the ‘two pump hypothesis’ arediscussed. The application of GA3in aqueous solution to leaves or flowersof hermaphrodite cultivars of strawberry, Redgauntlet and Rabunda,prevented growth of the receptacle despite hand pollination.This inhibitory effect occurred only when GA3 was applied priorto anthesis. Although viable pollen was produced and germinatedto grow down the styles of treated plants, no seeds were formed.Receptacle growth failed underneath the unfertilized carpels,but the basal region devoid of carpels enlarged and ripened.The effect of GA3 was the same in vivo and for flowers grownin vitro. ABA and BAP also inhibited growth of pollinated flowersin vitro, but neither substance stimulated growth of the baseof the receptacle. 2-NOA stimulated receptacle growth of pollinatedflowers but did not overcome the inhibitory effect of GA3. Removal of fertile carpels 9 days after pollination preventedfurther receptacle growth. GA3 treatment of the bare receptaclere-started growth but was less effective than 2-NOA. No growth substance treatment induced parthenocarpic developmentin these cultivars when unopened buds were emasculated and culturedwithout pollination, although GA3 induced some swelling of thereceptacle base. The abscisic acid (ABA) content of wrinkled (rr) pea seed tissueshas been quantified during development using multiple-ion-monitoringcombined gas chromatography-mass spectrometry and a deuteratedinternal standard. The level of ABA in the embryo generallyincreased with increasing cotyledon fresh weight while thatin the testa showed a distinct maximum at the time of maximumendosperm volume and the slowing in the growth of the testa.Pods contained relatively little ABA on a fresh weight basis.The total seed ABA content showed a biphasic distribution, thefirst maximum following the maximum growth rate of the testaand the second that of the embryo. The biphasic distributionof ABA in the pea seed was confirmed using a second pea genotype,near-isogenic to the first except for the r locus, and by theanalysis of individual seeds using a radioimmunoassay for ABA.The first maximum was composed mainly of a testa component andthe second mainly of an embryo component. When plants were grownin different environments, wrinkled seeds were found to containslightly more ABA than round (RR) but this was only significantlate in development. Immature seeds were capable of metabolizing17'-deoxy ABA to ABA, as determined by incorporation of either3H or 2H, and the metabolite was present mainly in the testa.The production of ABA in pea seeds is discussed in relationto the development of the different seed tissues. Triticum aestivumxZea mayscrosses are now widely used in theproduction of wheat doubled haploids to produce homozygous lines.Seasonal effects are known to influence the number of haploidembryos produced through wheatxmaize crosses, but the effectsof temperature and light have not been quantified. This studyinvestigated the effect of temperature and light intensity onhaploid embryo production. New Zealand wheat cultivars weregrown in a glasshouse until booting when they were transferredto growth cabinets at three temperatures (day/night; 17/12,22/17 or 27/22 °C at an irradiance of 250 µmol m-2s-1PAR).In another experiment, wheat lines were transferred to a growthcabinet at one of three light intensities (300, 500 or 1000µmol m-2s-1PAR at 22/17 °C day/night, with a photoperiodof 16 h). The temperature and light intensity at which pollinationswere made and subsequent fertilisation and embryo developmentoccurred, significantly (P<0.01) influenced the frequencyof haploid embryo production. The optimal temperature for embryorecovery was 22/17 °C. The greatest number of embryos wasproduced at a light intensity of 1000 µmol m-2s-1. Components of seed yield of cv. Ultra (Lupinus albus L.) andcv. Unicrop (L. angustifolius L.) were measured when grown atthree densities. The low density (10 plants m–2) Unicropyield (34 g seed per plant) was 1.8 times that of Ultra as ithad more branches, pods and seeds per pod. Ultra seeds (310mg per seed) were heavier than Unicrop seeds (180 mg). The branchingpattern of Ultra was less dependent on plant density, henceat 93 plants m–2 it gave a higher per plant yield (7.4vs 6.4 g) than Unicrop at lower densities (83 plants m–2).Density had most influence on pod formation and only small effectson seeds per pod and seed weight. Yield components on the main-steminflorescence were influenced less by density than componentson branch inflorescences. Later formed, higher order generationsof inflorescences were most affected by increased inter- andintra-plant competition. Pod numbers on the main-stem were similarfor both species. Pods formed at higher flower nodes in Unicrop,but the lower flower nodes were less fertile than those in Ultra.Node position of flowers had no influence on seed set in main-stemUnicrop pods, but pods from higher nodes in Ultra formed fewerseeds. In vivo net CO2 exchange characteristics of attached Brassicapods were studied during the entire period of their growth anddevelopment after anthesis. 14CO2 was fed both from the externalatmosphere and internally through the pod cavity, and the anatomyof the pod-wall was examined microscopically. Stomata were observedin the outer epidermal layer of the pod wall. Net in vivo CO2fixation by the pods was observed throughout the period of theirdevelopment and was maximum on day 42 after anthesis (DAA).Compared to the internal feeding experiments, 14CO2 fixationfrom the external environment was very high.
