Transfer of the catfish, Heteropneustes fossilis , to 10% sea water (101 mosmol l−1) or to 0·4% NaCl (140 mosmol l−1) does not evoke any change in plasma osmolarity from the normal freshwater values. There is, however, a reduction in urine flow rate (UFR) and increase in urine osmolarity without any change in the rate of osmolar clearance. In isosmotic (25% sea water or 0·7% NaCl) and in hyperosmotic (30% sea water or 1·1% NaCl) media there is a significant increase in plasma osmolarity accompanied by marked reduction in glomerular filtration rate (GFR), UFR and free water clearance. The results suggest that the catfish cannot effectively osmoregulate in isosmotic or hyperosmotic media and that the inability of the renal tubules to increase reabsorption of water and to reduce free water clearance may account for the restricted range of salinity tolerance of this catfish. Also, in the hyperosmotic media, plasma levels of cortisol are lowered while in the proximal pars distalis the corticotrophs appear active, suggesting increased utilization and clearance of cortisol. Prolactin-secreting cells, however, are degranulated and chromophobic in catfish maintained in hyperosmotic environment.  相似文献

In spontaneously breathing, chloralose narcotized and bilaterally vagotomized cats under mannite saline diuresis, the isolated carotid sinus were perfused for 1 hr with arterial and then for 1 hr with venous blood, and subsequently again with arterial blood for 1 hr. Either measure d or calculated were the parameters of arterial acid-base status, the arterial systemic bl-od pressure, the p-aminohippuric acid (PAH), inulin (IN), and osmolar clearance, the filtration fraction, the tubular reabsorption of oxmotically free water, the urinary time volume, and fractional excretions of the kidneys. Stimulation of the carotic chemoreceptors increased respiration, slightly enhanced the CPAH, and caused a minor fall of CIN with little altered filtration fraction. The renal resistance increased. The absolute and fractional excretions of urine, sodium, potassium, and of osmotically active particles rose significantly. Tubular reabsorption of osmotically free water showed a singificant increase. Urinary osmolarity remained constant in general. The changes of water and electrolyte excretion did not correlate with those of renal hemodynamics. The results argue against natriuresis being caused on chemoreceptor stimulation by increased filtrate volume of the whole kidney, or by decreased osmolarity of the renal medulla. The results are discussed with regard to the renal function in arterial oxygen deficiency.  相似文献

The effect of hypophysectomy on the hypothalamic neurosecretory system of the catfish, H. fossilis, was studied. Hypophysectomy resulted initially in an accumulation of NSM at the distal ends of the cut axons. The axons had grown and were reorganized into a neurohypophysis-like structure 3 weeks after hypophysectomy, and this persisted even more than 3 years post-hypophysectomy.  相似文献

The relationship between coelomic injections of mercuric chloride doses and osmoregulatory responses was measured. Response parameters were weight increases and blood osmolarity decreases 72 hr after dose administration. Massive edema and large decreases in blood osmolarity could be completely prevented by subcutaneous injections of equimolar sodium selenite. Mercury induced damage did not involve alterations of either selenium-dependent or non-selenium-dependent glutathione peroxidase activities.  相似文献

1. The enzymes glucose 6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, phosphoglucomutase, UDP-glucose pyrophosphorylase, phosphofructokinase, ATP-citrate lyase and acetyl-CoA carboxylase have been assayed in rat mammary glands in various stages of involution after hypophysectomy and weaning. 2. After hypophysectomy all seven enzymes decline in activity over a 12–16hr. period but the extent of the decline varies, with acetyl-CoA carboxylase becoming almost totally inactive, ATP-citrate lyase and phosphofructokinase showing a large decrease, and the remaining enzymes a less marked decline. 3. Within 24hr. of removing the litter a change in the pattern of enzyme activity is found very similar to that after hypophysectomy. 4. The significance of these results is discussed in relation to the endocrine control of mammary gland metabolism and the mechanisms of involution.  相似文献

Circadian water and sodium excretion in patients with renal arterial stenosis is inverted, but if the fluid intake is reduced it can return to normal. The findings also furnish indirect evidence that changes in circadian water and sodium excretion in patients with renal arterial stenosis are related to the degree of hydration of the organism. When evaluating circadian water and sodium excretion in patients with renal arterial stenosis, the osmolarity (specific gravity) of the urine should thus be taken into account.  相似文献

No significant changes in plasma cortisol and plasma osmolarity (the indicators of primary and secondary response respectively) were observed when the blood samples were obtained from unanaesthetized, anaesthetized and stressed catfish, H. fossilis. The results suggest that the catfish is fairly hardy and not easily susceptible to stress by routine laboratory handling. The sustained plasma glucose levels and decreased liver and muscle glycogen concentrations during cessation of feeding of the catfish suggest that during period of food deprivation, it draws its energy through glycogenolysis. Hence, in any study dealing with carbohydrate metabolism, the catfish needs to be fed during acclimation and experimental periods.  相似文献

Potassium transport in microvessels isolated from rat brain by a technique involving density gradient centrifugation was studied in HEPES buffer solutions of varying osmolarity from 200 to 420 mosmols, containing different concentration of sodium chloride, choline chloride, or sodium nitrate. The flux of 86Rb (as a tracer for K) into and out of the endothelial cells was estimated. Potassium influx was very sensitive to the osmolarity of the medium. Ouabain-insensitive K-component was reduced in hypotonic medium and was increased in medium made hypertonic with sodium chloride or mannitol. Choline chloride replacement caused a large reduction in K influx. Potassium influx was significant decrease when nitrate is substituted for chloride ion in isotonic and hypertonic media, whereas a slight decrease was found in hypotonic medium. The decrease of K influx in the ion-replacement medium is due to a decrement of the ouabain-insensitive component. Potassium efflux was unchanged in hypotonic medium but was somewhat reduced in hypertonic medium. The marked effect of medium osmolarity on K fluxes suggests that these fluxes may be responsible for the volume regulatory K movements. The possible mechanism of changes of K flux under anisotonic media is also discussed.  相似文献

  • 2.2. Following transfer to fresh water sodium uptake follows Michaelis-Menton kinetics. Fmax = 2.40 mmol Na/1 ECF/hr, Km = 0.26 mmol Na/1. The uptake system is fully activated immediately following transfer to fresh water.

  • 6.6. At pH 5 and 20 μmol Al/1 there is little further effect on sodium uptake but after 6 hr in aluminium the inhibition of sodium uptake continues after return to neutral aluminium fresh water and uptake is only 50% of normal 24 hr later.

Normal and hypophysectomized animals, fed ad lib., both with and without growth hormone treatment, were injected with 4 microcuries of P32/100 g. body weight, and then autopsied at varying time intervals following the injection.The plasma of hypophysectomized animals possesses a greater P32 activity than normal plasma at all intervals from 0.5 to 24 hr. after P32 injection. Maintenance of hypophysectomized animals with growth hormone results in a plasma P32-activity level much smaller than that found in the plasma of hypophysectomized controls, yet greater than that found in the plasma of normal animals.The normal liver uptake of P32 shows a maximum between 0.5 and 1 hr. after intraperitoneal injection. Kidney shows a maximum in less than 0.5 hour, and thymus attains a maximum activity level in 2 hr. Hypophysectomy results in an increased liver, kidney, and thymus P32 uptake when compared to the normal. After hypophysectomy the time of maximal P32 activity in liver is shifted to 4 hr. after injection. Growth hormone therapy in the hypophysectomized animal lowers the P32-uptake levels of kidney and liver toward normal, and brings down the P32 level in thymus to normal.Hypophysectomy with or without growth hormone therapy, results in a muscle P32 accumulation which is less than normal. Growth hormone administration to the normal animal causes an increased P32 accumulation by muscle.The P32 uptake by the tibia of the hypophysectomized rat is less than normal. Maintenance of the hypophysectomized rat with growth hormone prevents the decrease in bone P32 uptake due to hypophysectomy.  相似文献

Prolactin (PRL) cell activity was investigated in eels kept in fresh water (FW), deionized water (DW) supplemented or not with Ca (2 mM), in Ca-enriched FW (10 mM), in normal (Ca 3.4 mM) or Ca-free 1/3 sea water (SW), and in SW (Ca 10.2 mM) or Ca-free SW (Ca 0.15 mM). Light-microscopic studies, including measurement of the nuclear area and cell height, showed that PRL cell activity, reduced in DW, is not affected by Ca supplementation. Activity is reduced in Ca-enriched FW, in 1/3 SW and in SW, conditions inducing an increase in the plasma sodium level. The lack of calcium in saline environments partly suppresses the nuclear atrophy occurring in SW. There is no significant correlation between external or total plasma calcium concentration and PRL cell activity. In artificial Ca-free SW, eels show a rapid increase in plasma osmolarity and sodium levels; there is a significant negative correlation between these two plasma values and the nuclear area or cell height of PRL cells. As in some other teleosts, plasma osmolarity and plasma sodium seem to play a more important role than external or internal calcium in controlling PRL secretion. This correlation is not apparent in eels kept in SW, having unstimulated PRL cells but active calcium-sensitive (Ca-s) cells in the pars intermedia.  相似文献

After 20 to 50 days in sea water (SW), regressive changes of the kidney occur at the same rate in intact and hypophysectomized eels (Anguilla anguilla). In SW, ovine prolactin (oPRL) increases plasma electrolytes and restores a fresh water (FW) kidney structure; cell height and nuclear area increase in main segments of the nephron: first (P1) and second proximal (P2), distal (D) and initial collecting (C) tubules. A differentiation of new tubules also occurs. This effect is less intense after hypophysectomy and greater in FW. A dose of 10 μg/g body weight/day for 10 days in SW produces some harmful renal effects, barely detected at 6 μg/g/day. The kidney plays a minor role in osmotic adjustment in SW when PRL secretion is reduced (intact eels) or suppressed (hypophysectomized); PRL treatment reverses effects of SW adaptation. An inhibition of gill sodium extrusion may explain the high blood sodium level; however, kidney histological changes suggest a renal participation, perhaps through reduced water permeability as in Platichthys.  相似文献

The total ACh content and AChE activity were determined 1 hr after the i.p. injection of different doses of thiopental sodium (5, 10 and 20 mg/ml/100 g body wt) and barbitone sodium (20, 40 and 80 mg/ml/100 g body wt). The effect of different time intervals (1 min, 10 min, 30 min, 1 hr, 2.5 hr, 5 hr, 8 hr, 12 hr, 24 hr and 48 hr) on the total ACh content and AChE activity was investigated after i.p. injection of 10 mg thiopental sodium and 40 mg barbitone sodium/ml/100 g body wt. Both thiopental sodium and barbitone sodium increased the total ACh content in the brain tissue of Arvicanthis niloticus. Both drugs inhibited the brain AChE activity. It is thought that the increase in the total ACh content in the brain tissue of Arvicanthis niloticus may be due to a decrease in the release of ACh from the neuronal tissue and a decrease in AChE activity.  相似文献

This study describes the effects of desiccation and rehydration on the water content, haemolymph volume (per cent), osmolarity, and concentrations of Na, K, Mg, and Ca in the haemolymph of the lone star tick, Amblyomma americanum.The water content percentages of ‘severely desiccated’, ‘moderately’ and ‘fully hydrated’ ticks were 46·0, 52·8, and 60·3 per cent respectively. The lowest and highest of these were near the minimum and maximum possible.The haemolymph volume (per cent) of ‘severely desiccated’ ticks was regulated near the level of ‘moderately hydrated’ ticks despite significant decreases in total body water content and increases in osmolarity and concentration of sodium. Conversely, the change from ‘severely desiccated’ to ‘moderately hydrated’ ticks can be viewed as causing an increase in total body water, decrease in blood osmolarity and sodium, but little change in haemolymph volume (per cent).Most of the water taken up by ‘moderately hydrated’ ticks (while becoming ‘fully hydrated’) was added to the haemolymph. At the same time, there was little change in the blood osmolarity or haemolymph concentration of sodium. Conversely, the change of ‘fully’ to ‘moderately hydrated’ ticks was marked by a substantial loss of haemolymph volume (per cent) but little change in osmolarity and concentration of sodium.The concentration of potassium was regulated over the full range of desiccating and hydrating conditions. The lone star tick appeared less able to regulate its haemolymph concentrations of Ca and Mg; both fluctuated at the same rate, but inversely as the haemolymph volume (per cent).It appears that a carefully controlled movement of solutes (Na the predominant cation) between haemolymph and non-haemolymph tissue is intimately linked with haemolymph volume regulation and movement of water into the haemolymph during hydration.  相似文献

The data obtained reveal that elevation of extracellular osmolarity with sucrose during reintroduction of Ca-containing medium after 10 minutes of Ca2+ removal prevents loss of haemoglobin in a concentration-dependent mode. Reducing the extracellular osmolarity of the reperfusion medium by means of decreasing the concentration of sodium chloride and calcium chloride exacerbates the loss of haemoglobin from the cardiomyocytes. There is a close correlation between the water contents in tissues and the loss of haemoglobin during the "calcium paradox". The findings suggest dependence of the heart damage during the "calcium paradox" on anionic composition of extracellular space and activity of anionic transporters.  相似文献

A toxin from Bacillus thuringiensis kurstaki was lethal to eggs and first- and second-stage larvae of the ruminant nematode Trichostrongylus colubriformis. Sheathed and exsheathed third-stage larvae were also killed by the toxin. However, susceptibility of the ova to the toxin decreased after several hours of development. Heating at 65 C for 1 hr or freezing at 0 C for 3 mo did not affect stability of the toxin. Ovicidal activity of the toxin was not altered by treatment with 13 microbial or mammalian enzymes, but toxicity was reduced by the antibiotics streptomycin or penicillin G and the enzyme inhibitor L-1-tosylamide 2-phenylethylchloromethyl ketone. Cuprous, ferrous, and zinc chlorides also inhibited the ovicidal activity of the toxin. Increased osmolarity of the assay media or solubilization of the toxin from pH 3 to 11 had no effect on toxicity for eggs. The membrane agents sodium vanadate and 4,4'-diisothiocyano-2,2' disulfonic acid stilbene increased (9-fold) and decreased (333-fold) toxicity, respectively. N-acetylneuraminic acid was the only tested sugar that reduced the toxicity of B. t. kurstaki.  相似文献

Dualism in activity has been described in many fish species, including larger individuals of European catfish (Silurus glanis), which are able to switch their activities from nocturnal to diurnal in winter and spring. During the multi‐year telemetry study, seasonal changes in diel activity of 45 juvenile European catfish were investigated. These juveniles exhibited no dualism in movement and were strictly nocturnal and crepuscular with no period of diurnal activity. During winter, the same juvenile European catfish were completely inactive throughout the 24‐hr cycle. Also investigated was the impact of temperature, dissolved oxygen levels and water clarity on fish movement, relocation of resting places and home range size. The only significant correlations were that relocation and home range size increased with rising water temperatures.  相似文献

Tyrode solution containing added glutamine and Leloup's medium 1 has been used as a basic medium for the in vitro culture of the so-called proventriculus of adult Calliphora erythrocephala to elucidate some of the factors controlling the synthesis of peritrophic membranes (PM) in vitro. The formation rate was chosen as a quantitative criterion for the evaluation of the modifications of the incubation media.After systematic variation of osmolarity, pH, and temperature optimal formation rates were obtained in media with an osmolarity of 320 to 360 mOsmol, a pH of 6·8, and an incubation temperature of 27°C. Under these conditions the average rate of formation was in the modified Tyrode solution 3·0±1·1 mm PM/hr, and in Leloup's medium 3·6±0·8 mm PM/hr. In the modified Tyrode solution the formation of PM was complete after 5 to 7 hr, whereas in Leloup's medium it continued up to 24 hr. The addition of β-ecdysone caused an increase of the formation rate of PM to 4·5 to 5·5 mm PM/hr.The results obtained led to the hypothesis that an osmotically regulated enzyme system could be the limiting factor of the formation rate of peritrophic membranes, i.e. a system which could regulate the internal osmolarity of the formative cells by the interconversion of a bulk polymer and its monomer which are needed for the synthesis of PM.  相似文献

Plasma volume (PV) expansion is required for optimal pregnancy outcomes; however, the mechanisms responsible for sodium and water retention in pregnancy remain undefined. This study was designed to test the "arterial underfill hypothesis" of pregnancy which proposes that an enlarged vascular compartment (due to systemic vasodilation and shunting of blood to the placenta) results in renal sodium and water retention and PV expansion. We produced chronic vasodilation by 14 days administration of nifedipine (NIF; 10 mg·kg(-1)·day(-1)) or sodium nitrite (NaNO2; 70 mg·kg(-1)·day(-1)) to normal, nonpregnant female Sprague-Dawley rats. Mean arterial pressure, monitored by telemetry, was reduced by both NIF and NaNO2 but was unchanged in control rats. At day 14, vasodilator treatment lowered hematocrit and increased PV (determined by Evans blue dye dilution). Plasma osmolarity (Posm), sodium (PNa), and total protein concentrations all fell. These responses resemble the responses to normal pregnancy with hemodilution, marked PV expansion, and decreased Posm and PNa. Our previous work indicates a role of increased inner medullary phosphodiesterase-5 (PDE5) in the sodium retention of pregnancy. Here, we found that inner medullary PDE5A mRNA and protein expression were increased by both NIF and NaNO2 treatment vs. control; however, neither renal cortical nor aortic PDE5 expression was changed by vasodilator treatment. We suggest that a primary, persistent vasodilation drives increased inner medullary PDE5 expression which facilitates continual renal Na retention causing "refilling" of the vasculature and volume expansion.  相似文献
