Williams Syndrome provides a striking test case for discourses on disability, because the characteristics associated with Williams Syndrome involve a combination of “abilities” and “disabilities”. For example, Williams Syndrome is associated with disabilities in mathematics and spatial cognition. However, Williams Syndrome individuals also tend to “have a unique strength in their expressive language skills”, and are socially outgoing and unselfconscious when meeting new people. Children with Williams are said to be “typically unafraid of strangers and show a greater interest in contact with adults than with their peers.” This apparently keen social knowledge is a counterexample to the discussion of disability among academic philosophers, especially philosophers in the early modern period. Locke infamously used the example of disability to claim that Descartes’ arguments in favor of innate ideas were incorrect. On the contrary, Williams Syndrome may stand as an example of innate social knowledge; something that could benefit current discourse in philosophy, disability theory, and medical ethics.

Universal HIV testing and immediate antiretroviral therapy for infected individuals has been proposed as a way of reducing the transmission of HIV and thereby bringing the HIV epidemic under control. It is unclear whether transmission during early HIV infection—before individuals are likely to have been diagnosed with HIV and started on antiretroviral therapy—will compromise the effectiveness of treatment as prevention. In quantitative biology, observed data are fitted to a model that captures the essence of the system under investigation in order to obtain estimates of the parameters of the model, as well as their standard errors and interactions. The fitting is best done by the method of maximum likelihood, though least-squares fits are often used as an approximation because the calculations are perceived to be simpler. Here Brian Williams and Chris Dye argue that the method of maximum likelihood is generally preferable to least squares giving the best estimates of the parameters for data with any given error distribution, and the calculations are no more difficult than for least-squares fitting. They offer a relatively simple explanation of the methods and describe its implementation using examples from leishmaniasis epidemiology.  相似文献

It was previously reported that three soybean (Glycine max [L.] Merr.) nodulation mutants (NOD1-3, NOD2-4, and NOD3-7) were partially tolerant to nitrate when nitrate was supplied simultaneously with inoculation at the time of transplanting. The current study evaluated the effect of short-term nitrate treatment on nitrogenase activity (C2H2 reduction per plant and per nodule weight) and on relative abundance of ureides when nitrate application was delayed until plants were 3 weeks old and nodules were fully developed. Nitrogenase activity of the mutants was similar to that of Williams after an initial 3-week growth period, prior to nitrate treatment. Application of 5 millimolar nitrate resulted in greater inhibition of nitrogenase activity in Williams than in the three mutants. NOD1-3 was most tolerant of nitrate among the mutants tested and showed the highest relative abundance of ureides. Although C2H2 reduction activity per plant for NOD1-3 was higher than for Williams in the presence of nitrate, C2H2 reduction activity per gram of nodules was lower for NOD1-3 than for Williams in the presence and absence of nitrate. Compared to Williams, NOD1-3 had higher nodule ureide concentration and had similar glutamine synthetase activity in nodule tissue, indicating its nodules have normal nitrogen assimilation pathways. Nitrate application resulted in ureide accumulation in nodule tissue as well as in all plant parts assayed. Unexpectedly, nitrate treatment also increased the rate of ureide degradative capacity of leaves in both NOD1-3 and Williams. The data confirmed that nitrogenase activity of the selected nodulation mutants was more, but still only partially, tolerant of nitrate compared with the Williams parent.  相似文献

The laboratory performance of sinusoidal tide models is discussed in relation to their construction and use. Constant-flow sinusoidal models are discussed in detail and a formula is derived to described their maximal rate of change of water-level, which is shown to occur at mid-tide. This maximum rate of change is directly proportional to the tidal amplitude and inversely proportional to the tidal period. It is shown that the maximal rate of change of water-level is limiting in the construction of tide models because it governs the rates of inflow and outflow of sea water. The inflow rate must be sufficiently great to keep up with changes in the level of the outflow pipe as it rotate. The outlet pipe, however, must be sufficiently large to remove water at an adequate rate during the ebb of the tide.  相似文献

Oscillations of the number of cases around an average endemic level are common in several infectious diseases. In this paper we study simple deterministic models, where the oscillations arise either solely from periodically varying contact rates or from the combined effect of large initial perturbation, small periodic variation of the contact rate, and the destabilizing nature of infectious and latent periods when described as time delays. The main results are: (a) For a model with a periodically varying contact rate and a recovery rate, a threshold amplitude of variation is found by numerical and analytic methods at which 2-year subharmonic resonance appears. (b) Approximate analytic relationships are derived for the amplitude and phase of the forced 1-year oscillations below this threshold and for the 2-year oscillations above it—in terms of the reproduction rate of the infection. (c) Similar calculations are performed when the recovery rate is replaced by a fixed infectious period represented by a pure time delay. The threshold amplitude of variation in the contact rate is found here to be smaller than in the recovery rate model. (d) A model with a fixed infectious period and a constant contact rate is considered. The nontrivial steady state is shown to be locally stable for the parameter range of interest. However, the ratio of the imaginary to real parts of the eigenvalues in the characteristic equation is increased as compared to the corresponding model with a recovery rate. (e) For the model with a fixed infectious period and a constant contact rate an approximation method indicates consistency in a certain range of contact rates with the existence of an unstable periodic solution about the locally stable steady state. The actual existence of such a solution is not verified. The interpretation is that the destabilizing effect of the introduction of a pure delay into the model becomes more significant as the distance in the variables space from the endemic steady state is increased. (f) For a fixed infectious period and very small subthreshold variation in the contact rate, two different types of solutions are found numerically: yearly small-amplitude oscillations about an endemic average and large-amplitude oscillations of a subharmonic period. The pattern seen depends on the initial conditions. For a sufficiently large initial deviation from the endemic level even very small seasonal variations lead to regular recurrent outbreaks of the disease. The effect of latent periods and of changing the form of the interaction are also considered.  相似文献

Soybean (Glycine max) is a self-pollinating species that has relatively low nucleotide polymorphism rates compared with other crop species. Despite the low rate of nucleotide polymorphisms, a wide range of heritable phenotypic variation exists. There is even evidence for heritable phenotypic variation among individuals within some cultivars. Williams 82, the soybean cultivar used to produce the reference genome sequence, was derived from backcrossing a Phytophthora root rot resistance locus from the donor parent Kingwa into the recurrent parent Williams. To explore the genetic basis of intracultivar variation, we investigated the nucleotide, structural, and gene content variation of different Williams 82 individuals. Williams 82 individuals exhibited variation in the number and size of introgressed Kingwa loci. In these regions of genomic heterogeneity, the reference Williams 82 genome sequence consists of a mosaic of Williams and Kingwa haplotypes. Genomic structural variation between Williams and Kingwa was maintained between the Williams 82 individuals within the regions of heterogeneity. Additionally, the regions of heterogeneity exhibited gene content differences between Williams 82 individuals. These findings show that genetic heterogeneity in Williams 82 primarily originated from the differential segregation of polymorphic chromosomal regions following the backcross and single-seed descent generations of the breeding process. We conclude that soybean haplotypes can possess a high rate of structural and gene content variation, and the impact of intracultivar genetic heterogeneity may be significant. This detailed characterization will be useful for interpreting soybean genomic data sets and highlights important considerations for research communities that are developing or utilizing a reference genome sequence.  相似文献

Rates of photosynthesis, sucrose synthesis, starch accumulation and degradation were measured in sugar beet (Beta vulgaris L.) and bean (Phaseolus vulgaris L.) plants under a square-wave light regime and under a sinusoidal regime that simulated the natural daylight period. Photosynthesis rate increased in a measured manner in direct proportion to the increasing light level. In contrast to this close correspondence between photosynthesis and light, a lag in photosynthesis rate was seen during the initial hour under square-wave illumination. The leaf appeared to be responding to limits set by carbon metabolism rather than by gas exchange or light reactions. Under the sinusoidal regime starch degradation occurred during the first and last 2 hours of the photoperiod, likely in response to photosynthesis rate rather than directly to light level. Starch broke down when photosynthesis was below a threshold rate and accumulated above this rate. Under square-wave illumination, accumulation of starch did not begin until irradiance was at full level for an hour or more and photosynthesis was at or near its maximum. Under a sinusoidal light regime, sucrose synthesis rate comprised carbon that was newly fixed throughout the day plus that from starch degradation at the beginning and end of the day. Synthesis of sucrose from recently fixed carbon increased with increasing net carbon fixation rate while its formation from degradation of starch decreased correspondingly. The complementary sources of carbon maintained a relatively steady rate of sucrose synthesis under the changing daytime irradiance.  相似文献

The differential contribution of body contact and feeding to the formation of the ultradian rhythm of heart rate was evaluated by applying two different schedules of two- and three-hour periods for feeding with a single schedule of three hours for body contact during an observation period of seven days. A chi-square periodogram was used to calculate the period of ultradian rhythm. As a result, a three-hour ultradian rhythm of heart rates was detected in all subjects, which seems to correspond to either schedule of body contact or of feeding. However, no clear changes in the ultradia n rhythm of heart rate were observed corresponding to changes in feeding schedules. Remote biopsy darting of polar bears (Ursus maritimus) is less invasive and time intensive than physical capture and is therefore useful when capture is challenging or unsafe. We worked with two manufacturers to develop a combination biopsy and marking dart for use on polar bears. We had an 80% success rate of collecting a tissue sample with a single biopsy dart and collected tissue samples from 143 polar bears on land, in water, and on sea ice. Dye marks ensured that 96% of the bears were not resampled during the same sampling period, and we recovered 96% of the darts fired. Biopsy heads with 5 mm diameters collected an average of 0.12 g of fur, tissue, and subcutaneous adipose tissue, while biopsy heads with 7 mm diameters collected an average of 0.32 g. Tissue samples were 99.3% successful (142 of 143 samples) in providing a genetic and sex identification of individuals. We had a 64% success rate collecting adipose tissue and we successfully examined fatty acid signatures in all adipose samples. Adipose lipid content values were lower compared to values from immobilized or harvested polar bears, indicating that our method was not suitable for quantifying adipose lipid content.  相似文献

This paper investigates the dynamic, distributed pressure response of the human fingerpad in vivo when it first makes contact with an object. A flat probe was indented against the fingerpad at a 20 to 40 degree angle. Ramp-and-hold and sinusoidal displacement trajectories were applied to the fingerpad within a force range of 0-2 N. The dynamic spatial distribution of the pressure response was measured using a tactile array sensor. Both the local pressure variation and the total force exhibited nonlinear stiffness (exponential with displacement) and significant temporal relaxation. The shape of the contact pressure distribution could plausibly be described by an inverted paraboloid. A model based on the contact of a rigid plane (the object) and a linear viscoelastic sphere (the fingerpad), modified to include a nonlinear modulus of elasticity, can account for the principal features of the distributed pressure response.  相似文献

The theoretical orientation, encapsulated in the semi‐serious concept of ‘tooglies'of F.E. Williams, government anthropologist for the Territory of Papua in the inter‐war period, is considered. His ideas about culture change are contrasted with those of Bronislaw Malinowski, who acted as his mentor at one time. His treatment of the social organisational anomaly of Sogeri Koiari ‘sex affiliation’, often cited as a case of parallel bilineal descent, is compared with Margaret Mead's analysis of the ‘Mundugumor ropes’, which is classed as the opposite, cross‐sex bilineal descent. It is shown that Williams was able to get a clearer insight into this anomalous data than Mead and Fortune, and, on the whole, worked with an understanding of culture and Papuan social organisation that presages the relational approach of today.  相似文献

Comparisons were made of photosynthesis in three light limited cyclostat cultures (LD = 8:16, dilution rate 0.7 d–1) of Chlorella pyrenoidosa, differing only in the dynamics of irradiance supply: as a constant rate, i.e. a block culture; as a sine function of the light period, i.e. a sinusoidal culture; as an 8 h sine function superimposed by an 1 h sine function, i.e. an oscillating culture. The sinusoidal culture had a constant minimum quantum requirement for oxygen evolution (QR) of 10.8 over the photoperiod. The OR of the oscillating culture increased from 24 to 37 during the photoperiod. From changes in and P max we suggest that: (1) photosynthetic units (PSU) of the block and sinusoidal sulture increased in number; (2) increasingly fewer chlorophyll molecules participated in oxygenic photosynthesis with a decreasing turnover time of the PSU's during an oscillating photoperiod. Values of I k decreased slightly in the block culture, increased slightly in the sinusoidal culture and showed a twofold increase in the oscillating culture. From the ratio of in situ oxygen production (qO2) and P max we infer a balanced equilibrium between photosystem activity and electron transport capacity for the block and sinusoidal culture. We hypothesize that the qO2 values of the oscillating culture underestimated true oxygen production rates due to a nonlinear response at peak light intensities. The results show that a dynamical photoperiod provoked significantly different photosynthetic responses, even though the overall growth rate was unaffected.  相似文献

We report a first method for using M13 bacteriophage as a multifunctional scaffold for optically imaging bacterial infections in vivo. We demonstrate that M13 virus conjugated with hundreds of dye molecules (M13‐Dye) can target and distinguish pathogenic infections of F‐ pili expressing and F ‐negative strains of E. coli. Further, in order to tune this M13‐Dye complex suitable for targeting other strains of bacteria, we have used a 1‐step reaction for creating an anti‐bacterial antibody ‐M13‐Dye probe. As an example, we show anti‐S. aureus ‐M13‐Dye able to target and image infections of S. aureus in living hosts, with a 3.7× increase in fluorescence over background. The changes of viscosity, optical reflection and electrical resistivity of blood due to flow are dependent on the orientation and deformation of red cells. From electrical point of view, it can be assumed that blood is suspension of small insulating particles (red cells) in conductive fluid (plasma) when the frequency of supplied voltage is lower than several hundreds KHz. When blood flows, red cells deform and orient in flow direction. Therefore, flowing blood shows anisotropic electrical and optical properties. In steady flow, blood resistivity longitudinal to flow decrease with flow rate, and transverse one increases. Blood flow in living body is not steady but pulsatile. We measured both longitudinal and transverse resistivity changes, optical reflection change and viscosity change of sinusoidally flowing blood in a rectangular conduit. The results are 1) during one period of sinusoidal flow the longitudinal resistivity change is opposite to that of transverse one, 2) the waveform of reflection light change is similar to that of resistance change, and 3) minimum points of both longitudinal resistivity and viscosity changes do not appear at the moment when flow is zero but are delayed. When the amplitude of sinusoidal flow is small and oscillation frequency is high, the phase difference between the zero crossing period of flow and the period of minimum change in resistivity, increases up to 90 degrees. Viscosity of blood decreases with increase of amplitude and frequency of sinusoidal flow.  相似文献

We have modified an electromagnetically braked cycle ergometer to provide a sinusoidally varying work load. This was accomplished by applying a low-frequency sinusoidal voltage at the inputs of the electromagnets using a sinusoidal potentiometer, whose shaft was driven by a variable-speed motor, and a voltage-programmable power supply to amplify the signal. The frequency range is currently 0.0017-0.3 Hz. This technique for sinusoidal work loads allow the period, amplitude, and mean position to be easily changed from a console, even during a test if required.  相似文献

The liquid in the free space of leaf cell walls, the apoplast, is in direct contact with the plasma membrane and its nutrient uptake systems. Therefore, the pH of the apoplast is of utmost interest. We have elaborated a non-destructive method by which excised sunflower leaves ( Helianthus annuus cv. Erika) were perfused with fluorescein isothiocyanate-dextran (FITC-dextran) (4 000 Da) via the transpiration stream. We showed that leaf apoplast pH can be measured by using the fluorescence ratio technique together in conjunction with this dye. Evidence is provided that FITC-dextran does not penetrate the plasma membrane over a period of ca 17 h from the beginning of dye perfusion. Dye enrichment in the leaf apoplast did not cause an 'inner filter effect' and thus the fluorescence ratio was only dependent on pH. In vivo calibration yielded a pKa of 5.92, which was virtually identical to the pKa of 5.93 calculated for dye solutions. Hence, FITC-dextran can be detected in complex environments and covers a pH range prevailing in the leaf apoplast.
Based on this method we developed a microscope image technique visualizing pH gradients between various cell types. The pH in the lumen of the xylem vessel was ca 0.3–0.5 units lower than that of the apoplast of surrounding cells. Nitrate present in the leaf apoplast caused an increase in pH, especially in the dark. Under these conditions, in the intercostal area, the apoplast pH around the stomata was ca 0.5–1.0 units higher than that of the surrounding epidermal cells.  相似文献
