Spraying a zeatin solution on the mature leaf of an intact sunflowerplant (Helianthus annuus L. cv. Large Russian) caused a two-folddecrease in leaf temperature during the 20 min after treatment.Scanning electron microscope photographs taken 2 min after cytokinintreatment, however, showed a small but distinct stimulationof the opening of the central pore of the stomata, indicatingthat the latent period of cytokinin-induced stomatal openingis only 2 min. Five minutes after treatment with zeatin, thestomata at both the central pore and the outer ledge had openedwidely. On the other hand, zeatin riboside treatment did notshow a distinct effect on the stomatal opening within theseperiods. The stomata of a water-sprayed leaf closed within 1min after the treatment. Flooding the soil greatly reduced stomatal opening and leafgrowth rate of pea (Pisum sativum L cv. Feltham First) seedlings.This was despite the fact that leaf water potential and turgorwere not significantly affected by the treatment. Potassiumuptake and transport to the leaves was reduced by flooding.Stomata of flooded plants could be reopened by incubating leavesin solutions containing KC1. These observations raise the possibilitythat nutrient deficiency may limit stomatal opening and growthin flooded plants. We also consider the possibility that potassiumdeficiency may interact with a modification in the balance ofgrowth regulators in the leaves to modify stomatal behaviourand growth. The diuranl rhythm of the stomatal aperture size was investigatedusing isolated epidermal strips from a CAM plant, Notonia grandiflora.Maximal stomatal opening occurred at pH 6.5. The nocturnal stomatalopening was stimulated by KCl which produced stomatal closurein light. The stimulatory effect of KCl on stomatal openingin drakness was suppressed by ABA. The apparent cuticular component of transpiration of stomatabearing leaf epidermis was estimated by restricting stomataldiffusion by mass flow of air in the opposite direction. Thiswas achieved by applying an air pressure gradient across theamphistomatous leaf. Some assumptions of the previously suggestedmethod (antrcek and Slav?k, 1990) were experimentally verifiedusing maize leaves. The technique makes possible a quantitativeestimation of cuticular water loss including that of the externalperistomatal (i.e. vapour not passing through the pores) andthe respective conductance when the stomata are partially open. In addition to the fact that the cuticular portion of the totalleaf vapour loss (i.e. relative cuticular transpiration) dependson stomatal opening, even the absolute value of apparent cuticulartranspiration was (1) increased by lower vapour pressure deficitand (2) decreased with closing stomata. These changes, inducedby variations in a vapour pressure deficit of 2.45?0.35 kPa,ranged between 0.66?0.14µg cm –2 s–1. Theabsolute value of apparent cuticular transpiration changed onaverage by a factor of 2.3 due to stomata opening or closingwhich was induced by turning the light on or by exogenous ABAapplication. Possible interference by residual vapour diffusingthrough the stomatal pore was evaluated by the model application.An attempt was also made to assess the cuticular component ofCO2-uptake rate. The effects of three morphactins, chlorflurenol, flurenol andEMD 7301 W, were examined on the stomatal aperture using isolatedepidermal strips of Commelina benghalensis. Morphactins produced,a striking decrease in the stomatal opening in light but hadno effect on stomatal closure in darkness. Various catalystsand inhibitors of photophosphorylation had no influence on themorphactin-induced stomatal closure. The stimulatory effectsof ATP, pyruvate and KC1 on stomatal opening were suppressedby the morphactins. The cytokinin, benzyladenine stimulatedthe stomatal opening even in the presence of a morphactin. The effects of elevated atmospheric CO2, alone or in combinationwith water stress, on stomatal frequency in groundnut (Arachishypogaea (L.) cv. Kadiri-3) were investigated. CO2 exerted significanteffects on stomatal frequency only in irrigated plants. Theeffects of drought on leaf development out weighed the smallereffects of CO2 concentration, although reductions in stomatalfrequency induced by elevated atmo-spheric CO2 were still observed.When stands of groundnut were grown under irrigated conditionswith unrestricted root systems, an increase in atmospheric CO2from 375 to 700 ppmv decreased stomatal frequency on both leafsurfaces by up to 16% in droughted plants, stomatal frequencywas reduced by 8% on the adaxial leaf surface only. Elevatedatmospheric CO2 promoted larger reductions in leaf conductancethan the changes in stomatal frequency, indicating partial stomatalclosure. As a result, the groundnut stands grown at elevatedCO2 utilized the available soil moisture more slowly than thosegrown under ambient CO2, there by extending the growing period.Despite the large variations in cell frequencies induced bydrought, there was no treatment effect on either stomatal indexor the adaxial/abaxial stomatalfrequency ratio. The data suggestthat the effects of future increases in atmospheric CO2 concentrationon stomatal frequency in groundnut are likely to be small, especiallyunder conditions of water stress, but that the combination ofassociated reductions in leaf con-ductance and enhanced assimilationat elevated CO2 will be important in semi-arid regions Key words: Arachis hypogaea L, Leguminosae, groundnu, stomatal frequency, CO2, drought  相似文献

Profiles of shortwave radiation, net radiation and temperaturewere measured in swards of three grasses of contrasting structureLolium perenne cv. S24, L. perenne cv. Reveille and Festucaarundinacea cv. S170. Measurements were also made of the reflectionof shortwave radiation, leaf water potential and stomatal resistance.Differences in canopy structure influenced the absorption andreflection of radiation by the varieties. The absorption ofnet radiation and its influence on air temperature inside thecanopy was shown to vary with canopy structure. Calculationsshowed that diurnal changes in the reflection and transmissionof light (400–700 nm) would have little effect on canopyphotosynthesis. No clear relationship between leaf extensionrate, temperature and leaf water potential could be established,although decreases in water potential did appear to reduce thepotential response of leaf extension rate to temperature.  相似文献

Despite the observation first made by von Mohl in 1856, thatepidermal cells greatly influence stomatal aperture, subsequentstudies have failed to pay adequate attention to epidermal cellviability or to quantify the degree of its influence on aperturein epidermal strips and leaf sections. Using Vicia faba stripsand leaf sections we found the following: (i) a non-linear relationshipbetween aperture and guard cell contact with live epidermalcells; (ii) epidermal cell viability on isolated strips hada threshold at about 25 °C; (iii) epidermal strips withdead epidermal cells had wider apertures and lower variabilitythan strips with live cells or intact leaf sections; (iv) afterepidermal cell viability was accounted for, stomatal aperturesshowed no significant differences between isolated strips orstrips removed from leaf sections treated in the same manner;(v) highly variable apertures appeared to be the normal conditionof the intact leaf. Caution should therefore be used in interpretingstomatal behaviour from epidermal strips without first takinginto account mechanical interactions between the guard and surroundingepidermal cells. A comparison was made of stomatal behaviour, and related phenomena,between leaves of garden pea (Pisum sativum cv. Feltham First)inoculated with powdery mildew fungus (Erysiphe pisi) and uninfectedleaves on healthy plants. Twenty four hours after inoculation,stomata opened more widely in the light in infected leaves thanin healthy leaves. Thereafter, stomatal opening was progressivelyreduced by infection and stomata failed to close completelyin the dark until, 7 d after inoculation, all movements ceasedand stomata remained partly open. Transpiration in the lightfollowed closely the pattem of stomatal opening and, after anearly increase compared with healthy controls, was progressivelyreduced by infection. Evidence is presented that transpirationfrom the fungus was less than the reduction in transpiraationfrom the leaf which was caused when development of the myceliumincreased the boundary layer resistance of the leaf. Seven daysafter inoculation, transpiration in the dark was greater frominfected leaves than from healthy leaves because of partly openstomata in the dark. Net photosynthesis in infected leaves was reduced within 24h of inoculation to a level below that found in healthy leavesand thereafter it declined progressively. The initial reductionwas due to a transient increase in photorespiration, for whenthe glycolate pathway was inhibited by a 2% O2 concentrationthere was no difference between the (gross) photosynthetic ratesof healthy and infected leaves. Changes in photorespirationrate were confirmed from the interpretation of the CO2 burston darkening. Reduced stomatal opening was a contributory causeof the reduction in net photosynthesis in the later stages ofinfection. Since the rate of gross photosynthesis, but not therate of photorespiration, of infected plants fell below thatof healthy plants, and infected plants had a higher rate ofrelease of CO2 in the dark than healthy plants from the thirdday after inoculation onwards, infected plants consume an increasinglygreater proportion of their photosynthate in respiratory processesthan do healthy plants. Stomatal sensitivity to water stress was investigated in pearlmillet [Pennisetum americanum (L.) Leeke] in relation to stageof plant development, leaf water status and ABA content by samplingplants at midday. For the same leaf water potential (), droughtedplants with emerged panicles were found to have a greater leafconductance (gL), indicative of greater stomatal opening, thanplants sampled prior to panicle emergence. The difference betweensuch flowering (F) and non-flowering (NF) plants in at stomatalclosure was estimated to be at least 0.6 MPa. This differencewas considered unlikely to be the result of differential bulkleaf osmotic adjustment, and for most samples from both F andNF plants, bulk leaf turgor potential (p) was estimated to bezero. Stomatal closure in NF plants was associated in two genotypes(BJ 104 and line 112) with higher leaf ABA levels. Differencesin ABA levels between F and NF plants were, however, smalleror absent in genotypes Serere 39 and B282. These genotypes wereat lower than BJ 104 and line 112 when sampled and showed smallerdifferences between F and NF plants in conductance. Lower ABA levels in F plants are ascribed either to effectsof leaf ageing or to effects of flowering on ABA content ofthe leaf. Significant differences in gL in the absence of differencesin ABA content are taken to imply changes in stomatal sensitivityto the hormone or in its access to the stomatal complex. Kinetin retarded the decrease in chlorophyll content in leafdiscs from 5 species of plants with amphistomatous leaves, wherethe upper surface was exposed to air, but not in Rumex acetosera.When leaf discs were floated so that the lower surface was exposed,the effect of kinetin was less evident. Kinetin also stimulatedtranspiration in leaf discs from Nicotiana tabacum (amphistomatous),but not in leaf discs from Paederia chinensis (hypostomatous).Nor kinetin did retard chlorophyll breakdown in this specieswhen leaf discs were floated so that the stomatal surface wasin contact with the solution. The ineffectiveness of cytokininsin chlorophyll retention in leaf discs from hypostomatous leaveswas not due to reduced uptake of benzylaminopurine-14C. Chlorophyll retention was severely inhibited by coating theleaf surface with vaseline either in the presence or absenceof kinetin. Leaf discs floated on a solution exposed to CO2-lessair retained more chlorophyll than those in normal air. Thereis thus a close relationship between stomatal opening (as measuredby stimulation of transpiration) and chlorophyll retention,as influenced by cytokinins. The technical device for continuous microscopic observationsof stomatal movements in a gas exchange chamber using digitalimage analysis, earlier described by Kappenet al. (1994), wastechnically improved. By electronic remote control, it is nowpossible repeatedly to record over a period of several daysidentical stomata on a 25 50 mm leaf area. The responses ofindividual stomata to various light-dark sequences in the lightand in the dark phase were investigated on attached leaves ofViciafaba (L.). The amplitude of stomatal oscillations caused byalternatinglight was highest on the first day of an experimental seriesand decreased with repeated application, indicating a long-termadjustment to changing light conditions by decreasing sensitivityto intermittent darkening. In the usual dark phase light-darksequences had little effect on the stomatal aperture, whichremained small. These results were widely convergent with thestomatal conductance calculated from the gas exchange measurements.Very prominent is the role of an endogenous rhythmicity of thestomatal response. It could be demonstrated (1) by autonomousstomatal opening before the light phase started; (2) by a suppressionof dark response in the early morning hours; (3) by a decreasinglight stimulation in the afternoon; (4) by further increaseof aperture for several hours if no light was provided in thelight phase. Closely adjacent stomata could show divergent dark-openingmovements pointing to an autonomous control mechanism locatedin the guard cells. The endogenously controlled morning openingprovides full assimilation capacity in the usually humid morninghours when transpiratory water loss associated with C02-uptakeis comparatively small. To evaluate whether abscisic acid (ABA) in the xylem sap playsan important role in controlling stomatal aperture of field-grownPrunus persica trees under drought conditions, stomatal conductance(g) and xylem ABA concentrations were monitored both in irrigatedand non-irrigated trees, on two consecutive summer days (threetimes a day). Stomata1 conductance of non-irrigated trees hada morning maximum and declined afterwards. The changes in gduring the day, rather than resulting from variations in theconcentrations of ABA in the xylem sap or the delivery rateof this compound to the leaves, were associated with changesin the relationship between g and xylem ABA. The stomata ofwater-stressed trees opened during the first hours of the day,despite the occurrence of a high concentration of ABA in thexylem sap. However, stomatal responsiveness to ABA in the xylemwas enhanced throughout the day. As a result, a tight inverserelationship between g and the logarithm of xylem ABA concentrationwas found both at midday and in the afternoon. A similar relationshipbetween g and ABA was found when exogenous ABA was fed to leavesdetached from well-watered trees. These results indicate thatABA derived from the xylem may account for the differences ing observed between field-grown peach trees growing with differentsoil water availabilities. Several possible explanations forthe apparent low stomatal sensitivity to xylem ABA in the morning,are discussed, such as high leaf water potential, low temperatureand high cytokinin activity. Measurements of electrical potential difference (PD) and potassiumactivity were carried out on the intact leaf of Tradescantiavirginiana. PD gradients across the stomatal complex were observedwith both open and closed stomata. The guard cell PD appearedto be linearly related to stomatal aperture. With the stomataopen a gradient of potassium activity across the stomatal complexwas observed which became reversed on stomatal closure. Calculationof the driving forces on potassium suggested that it was distributedpassively between the vacuoles of the cells of the stomatalcomplex. The electrophysiological data obtained from this investigationenabled potassium activity in the apoplast to be calculated.The results showed that on stomatal closure there was a massiveincrease in the potassium activity in the guard cell wall. Epidermal strips and leaf fragments of Commelina and leaf fragmentsof maize were incubated on solutions containing naturally-occurringor synthetic cytokinins and/or ABA. The effects of these treatmentson stomatal behaviour were assessed. Cytokinins alone did notpromote stomatal opening in either species but concentrationsof both zeatin and kinetin from 10–3 to 10–1 molm–3 caused some reversal of ABA-stimulated closure ofmaize stomata. The reversal of the ABA effect increased withincreasing cytokinin concentration. Cytokinins had no effecton ABA-stimulated closure of Commelina stomata. When appliedalone, at high concentration (10–1 mol m–3), toCommelina epidermis or leaf pieces both zeatin and kinetin restrictedstomatal opening. It is proposed that a measurement of the peristomatal groove distance (PGD) of guard cells on surface impressions of leaf epidermis can act as a surrogate measure of stomatal aperture. To test this idea, investigations were carried out on two species, one in which it is possible to make direct measurements of pore width with relative ease (Commelina communis L.) and one whose stomata are so small that this is difficult (Phaseolus vulgaris L.). Leaf water vapour conductance measurements were first taken with a porometer, then, without delay, a silicone rubber impression of the leaf was made of the area directly under the porometer cup. From a positive replica of this impression, stomatal aperture, PGD and pore length were measured. The correlations between stomatal aperture and PGD and between PGD and stomatal conductance were positive and highly significant. Because a causal relationship between stomatal aperture and PGD is expected, linear regression was used to obtain equations for converting PGD measurements into estimates of stomatal aperture. These account for 91.7% of the variation of aperture in the case of C. communis and 70.7% in P. vulgaris, suggesting that PGD measurements have potential as an alternative measure of pore width in cases where direct measurements would be both difficult and subject to excessive measurement error or bias.  相似文献

Illuminated leaf discs of Vicia faba were brought into equilibrium with a series of mannitol solutions. The width of stomatal aperture and the osmotic potential of guard cells and epidermal cells were determined. It was found that the maximal aperture was obtained when epidermal cells were at about incipient plasmolysis and that any increase in their turgor pressure brought about a decrease in stomatal aperture. These findings emphasize the importance of epidermal cells in determining the width of the stomatal pore.  相似文献

Mature second leaves of Lolium perenne L. cv. Vigor, were sampledin a spring and summer regrowth period. Effects of CO2enrichmentand increased air temperature on stomatal density, stomatalindex, guard cell length, epidermal cell density, epidermalcell length and mesophyll cell area were examined for differentpositions on the leaf and seasons of growth. Leaf stomatal density was smaller in spring but greater in summerin elevated CO2and higher in both seasons in elevated temperatureand in elevated CO2xtemperature relative to the respective controls.In spring, leaf stomatal index was reduced in elevated CO2butin summer it varied with position on the leaf. In elevated temperature,stomatal index in both seasons was lower at the tip/middle ofthe leaf but slightly higher at the base. In elevated CO2xtemperature,stomatal index varied with position on the leaf and betweenseasons. Leaf epidermal cell density was higher in all treatmentsrelative to controls except in elevated CO2(spring) and elevatedCO2xtemperature (summer), it was reduced at the leaf base. Inall treatments, stomatal density and epidermal cell densitydeclined from leaf tip to base, whilst guard cell length showedan inverse relationship, increasing towards the base. Leaf epidermalcell length and mesophyll cell area increased in elevated CO2inspring and decreased in summer. In elevated CO2xtemperatureleaf epidermal cell length remained unaltered in spring comparedto the control but decreased in summer. Stomatal conductancewas lower in all treatments except in summer in elevated CO2itwas higher than in the ambient CO2. These contrasting responses in anatomy to elevated CO2and temperatureprovide information that might account for differences in seasonalleaf area development observed in L. perenne under the sameconditions. Young seedlings of two cultivars of Phaseolus vulgaris L. (cv.Cacahuate-72 and Michoacan-12A3) were subjected to a soil dryingtreatment. Stomata started to close before any leaf water deficitcould be detected. Soil drying promoted a small degree of ABAaccumulation in roots, but, at the time stomatal closure wasinitiated, at least in one cultivar, xylem ABA and bulk leafABA concentration were not enhanced. The mechanism of the stomatalresponse is discussed in terms of an as-yet unidentified regulatorof stomatal behaviour, redistribution of existing ABA in plants,and a high sensitivity of stomata to small changes in ABA concentration.  相似文献

The effects of H$ and fusicoccin (FC) on stomatal opening inthe dark were investigated using epidermal strips of Commelinacommunis and Vicia faba cv. Ryosai Issun. Citrate-phosphatebuffer induced maximal opening of stomata at pH 3.0 when testedover the range of 2.7 to 5.0. HCl at 1 mM also induced stomatalopening without appreciable accumulation of K$ in the guardcells. After 4 hr treatment with 10 µM FC, stomata openedwith concomitant accumulation of K$ in the guard cells, although1–2 hr treatment caused opening without concomitant K$increase.
