In a beech (Fagus sylvatica L.) stand in north-west Germany vegetation of two transects (25m:1m and 20m:1m) was mapped and contents of macronutrients (Ca, Mg and K), micronutrients (Fe, Mn, Zn and Cu), and potentially phytotoxic metals (Pb, Cd, Ni and Al) were measured in different soil compartments and in roots, rhizomes, stems and leaves of two forest floor plant species (Mercurialis perennis L. and Polygonatum multiflorum L.). NH4Cl extractable cation contents, pH and other soil variables were also determined.The highest macronutrient contents could be found in the leaves of M. perennis and P. multiflorum. Heavy metals and Al accumulated in the roots. Correlation analysis suggests a considerable translocation of Zn and Cd between below- and above-ground organs of both investigated forest floor plants. No significant correlation was found between the contents of the other elements in the below- and above-ground parts.Available data indicate a considerable uptake by the plants not only of nutrients, but also of heavy metals from the upper mineral soil. Amounts of heavy metals and Al solubilized in the presence of NH4Cl increased with decreasing pH, whereas levels of soluble Ca and Mg were maximal at high pH-values of the extracts. It can be concluded that element uptake in the investigated plants is indirectly controlled by the pH of the upper mineral soil.  相似文献

Phytoactivity and allelopathic studies are heavily dependent on germination bioassays of water solutions of allelochemical(s), which necessarily imply that pH and osmotic pressure vary among treatments and between treatments and controls and are therefore a confounding factor in the assessment of seed germination responses to allelochemical(s). When the contribution of pH and osmotic pressure to seed germination responses is considered in experimental designs their effects are almost without exceptions examined separately being assumed, without any evidences, that pH and osmotic pressure act independently on seed germination responses. The objectives of this work were to examine experimentally such assumption using wheat, lettuce, and subterranean clover cultivars to evaluate and model the combined effects on germination of pH and osmotic pressure in the range between 3.0–6.0 and 0–100 mOsmol kg?1, respectively. Empirical equations are fitted, discussed, and the need to consider the simultaneous effects of pH and osmotic pressure firmly established. Finally, the use of the equations fitted and its impact on conclusions is exemplified in a dose-response bioassay of water extracts of Cistus ladanifer on seed germination using subterranean clover as target species where hormesis was found before allelochemical effects were corrected for pH and osmotic pressure values of control and extracts.  相似文献

A simple and rapid bioassay was implemented to detect the germination activity of extracts from soils in pre/post-burn conditions. Soil samples taken from burnt, unburnt and adjacent plots at depths of 0–2, 2–4, 4–6 and 6–8 cm before and after burning mesic grassland in South Africa were analysed for germination activity over an eight-week period. Soil samples were extracted using dichloromethane and bioassayed using Grand Rapids lettuce (Lactuca sativa L.) achenes (seeds). The Grand Rapids lettuce seeds exhibited greater germination percentages when treated with extracts from burnt soil compared to the other plots. The magnitude of the germination activity declined with time since the burn. The Grand Rapids lettuce seeds also exhibited significantly higher germination when treated with unburnt soil extracts compared to the control (distilled water) which indicates the existence of other factors controlling germination in unburnt soil. Germination activity in the adjacent plots decreased with time. These findings indicate that the germination activity of the smoke derived from burning plant-material diffuses into the soil and its persistence declines with time. Considering that the soil seed bank contains viable seeds, at a moderate depth, and that they are initially unaffected by the heat of the fire, then smoke residues following a fire can influence the germination and recruitment of plant species that are responsive to smoke-derived compounds and are represented in the germinable soil seed bank.  相似文献

Allelopathy has been regarded as a mechanism for successful exotic plant invasion. However, it is not clear if and what effects of allelopathic substances may exert on soil nutrient. The exotic plant Mikania micrantha H.B.K. (M. micrantha) has invaded many forests in south China, and recent studies have suggested it has allelopathic potential for other plants and soil microbial community. Thus, we hypothesized that M. micrantha could influence soil nutrients and N transformation through allelopathy. We measured total C and N, NO3 , NH4 + and pH of the soil beneath M. micrantha and the adjacent open soil, and then measured the above soil properties after treating soil with 3 concentrations of aqueous extracts of M. micrantha (T1: 0.005 g ml−1; T2: 0.025 g ml−1; T3: 0.100 g ml−1). In addition, a bioassay was conducted to determine the allelopathic potential of the soil beneath M. micrantha. The results showed that M. micrantha significantly affected soil nutrients and N transformation. Soil beneath M. micrantha had inhibitory effects on seed germination and seedling growth of test plant, and had significantly higher C, N, ammonia, net nitrification rate than those of open soil. The plant extracts decreased soil pH, and T1 decreased it the most, and it increased soil C and N, and T1 represented the greatest increase in both C and N. The extracts also increased both NO3 and NH4 + in soil, whereas no significant difference existed among the 3 extract treatments. Compared to the water control, the soil net mineralization rate was higher under T1, while lower under T2 and T3. However, the extracts increased the soil nitrification rates under all the treatments (T1, T2 and T3). Our results suggest that the water soluble allelochemicals of M. micrantha improve soil nutrient availability, through which the invasive plant M. micrantha may successfully invade and establish in new habitats.  相似文献

Summary Phosphorus in soils from plantation of red pine (Pinus resinosa Ait.) was determined using six extractants: 0.002N H2SO4 (pH 3.0); 0.025N HCl+ +0.03N NH4F; 0.5N NaHCO3 (pH 8.5);N NH4OAc (pH 4.8); anion exchange resin (Dower –2, Cl-form); H2O. Correlations of extractable P with Al- and Al-+Fe-P indicated that these fractions are the dominant forms of inorganic P in most of the soils.Uptake of P by corn and Monterey pine seedlings grown in greenhouse culture was correlated with soil P extracted by the different methods. The most successful of the extractants for predicting P uptake was resin extractable P; the simple correlation coefficients were 0.811 and 0.609 for pine and corn respectively. P uptake by pine correlated significantly with 0.002N H2SO4 P (r=0.679),N NH4OAc P (r=0.443), H2O P (r=0.549) and Al-+Fe-P (r=0.532) while P uptake by corn correlated with 0.002N H2SO4 P (r=0.579), H2O P (r=0.477) and organic P (r=0.460). Per cent P in pine seedling tops correlated significantly with 0.002N H2SO4, resin andN NH4 OAc extractable P. 研究了不同pH值土壤以及重度盐碱土和非盐碱土浸提液对羊草(Leymus chinensis)种子萌发及幼苗生长的影响。不同pH值土壤由配土和调酸两种方式取得, 前者由重度盐碱土(pH = 10.24)与非盐碱土(pH = 7.49)按不同比例配制, 后者是由重度苏打盐碱土经硫酸调酸处理得到。配土发芽的实验结果表明, 当pH值在7.49-9.14时, 种子的发芽率均在50%以上, 且幼苗能够正常生长; pH = 9.53时, 羊草种子的发芽率低于50%, 仅部分幼苗个体能够成活; 当pH > 9.86时, 幼苗在萌发后50天左右全部枯死。说明羊草种子个体萌发期最大耐受pH值在9.14-9.53之间。重度苏打盐碱土调酸发芽实验结果表明, 当调酸后 的土壤pH值降至7.0-10.0时, 均能显著促进羊草种子的萌发, 且幼苗生长正常。确定了羊草种子萌发的最适pH值为8.0-8.5。土/水比为1: 1的非盐碱土浸提液能够显著提高羊草种子发芽率, 而重度苏打盐碱土浸提液对羊草种子萌发没有产生显著抑制。  相似文献

Abstract

Uptake of metals by whole Eichhornia crassipes plants and excised roots was studied using a bioassay system. Results indicate that in time-series bioassay at concentration of 10 mg L?1 of either Zn or Cu, rapid uptake by whole plants occurred during the first 4 h, but subsequently levelled off after 48 h. A greater proportion of metals absorbed remain in the root system rather than being translocated to other parts of plant. When treated with different combinations of both Cu and Zn, some synergistic effect of metals appeared to have occurred as indicated by enhanced uptake when bioassays involved combination of metals. Absorption kinetics of monovalent K+ by excised Eichhornia roots after a 24-h treatment, indicated an initial linear trend over the range of 1–10 mg K L?1, but subsequently levelled off at 15 mg K L?1 concentration with Vmax of 7.5 × 10–6 M K g?1 dry tissue h?1 and Km of 1.1 × 10?3 M K. Potentiometric titrations revealed existence of pH-dependent charge densities on root system which have variable affinities for cations and helps explain the ability of Eichhornia roots to absorb and accumulate large amounts of metal ions especially at high pH of media.  相似文献

Deschampsia flexuosa (L.) Trin. is an abundant grass species in the ground flora of acidic beech forests in southern Sweden. Generally, the species is restricted to a limited soil pH range (pH 4–5). The main objective was to study the influence of different soil acidities on germination, initial root development and on the growth of the species. The experiments were carried out under controlled conditions and designed to simulate the physico-chemical conditions present in the field. By using forest soils within the pH range 4.0 to 8.3 and artificial variation in pH (3.2 to 7.6) of soil-water extracts, it was possible to evaluate the influence of soil reaction and the H+ per se. In all experiments seeds have been used. Germination was significantly delayed in the very acid soil (pH 4.0) in comparison to the germination in soils within the pH range (4.4 to 6.4). Soil substances, other than the H+, might be responsible for this delay in germination, whereas development of the radicle was markedly affected by increasing H+ concentrations. Especially the development of root hairs was sensitive to H+ and was significantly reduced at a pH<-3.8. By increasing soil acidity the injury symptoms, including curling and discolouring, became more intense and at the highest acidity (pH 3.2) the radicles appeared brown, stunted and the root hairs were lacking. Most favourable growth was obtained at pH 4.4 and 5.0. Soil pH levels above and below this range limited both shoot and root growth. The results showed very good correspondence with observations made in Beech forest soils in southern Sweden, where the species was growing in soils within the pH range 3.9 to 5.1 with a peak growth at pH 4.3. This study shows that in soils at pH≤3.8, the poor development of the radicle may be crucial in the establishment ofDeschampsia flexuosa. Root hair development was more sensitive to soil acidity than radicle elongation. Germination was delayed in very acid Beech forest soils but other factors than the H-ion per se may be responsible for this delay.  相似文献

The toxicity associated with suspended sediments from the Rhone River (Switzerland-France) was determined with three acute bioassays. Large volume water samples were centrifuged for recovery of suspended solids in November 1989; one sample was taken as a control upstream from Lake Geneva and the 9 remainder downstream from Geneva to the Mediterranean Sea, with a single sample of the major tributary the Saône at Lyon. Heavy metals (Hg, Cd, Cr, Cu, Ni, Pb, Zn) and organic contaminants (OCs, PCBs, PAHs) bound to sediment were analysed and extracted by elutriation with filtered lake water and by organic solvent (dichloromethane). Sediment water elutriates were tested with algal fractionation bioassays (AFB) using Lake Geneva ambient phytoplankton, with Daphnia magna and Microtox® acute toxicity tests, whereas organic extracts were utilized in the latter two bioassays to evaluate the potential sediment toxicity.The bulk analyses of the sediment together with elutriate metal concentrations indicated the highest contamination of sediment downstream of Lyon. Medium contamination appeared for the stations downstream of Geneva, in the Saône River and at the Rhone River mouth. The station upstream of Lyon had low concentrations, comparable to the values in the Upper Rhone. Organic contaminants are mainly observed downstream of Lyon and their concentrations decline onwards to the sea. The bioassays Microtox® on organic extracts and AFB on the elutriates show a similar toxicity trend, but differ in that Microtox was more sensitive to organics whereas the algal test responded predominantly to metals. This difference is believed to be due to the different extraction procedures used, rather than to the tests themselves. Daphnia magna was the least sensitive and appeared to give a broader band response to the observed contaminants in the sediment. The bioassay results when integrated confirm that the biotoxicity trends relate well to the composition of the sediment, a factor which emphasizes the need for battery testing in ecotoxicological assessment.  相似文献

Summary The relative efficiency of seven extractants for estimating available Cu in sierozem soils of Haryana was studied. Bulk samples of 15 soils ranging in neutral normal amm. acetate extractable Cu from 0.12 to 0.20 ppm were subjected to screen-house study. The quantities of Cu extracted with neutral normal amm. acetate, Morgan’s reagent (pH 4.8)N ammonium acetate (pH 4.8), 0.1N HCl, 0.02M EDTA, critrate-EDTA and DTPA from soils were examined for their correlation with responses of maize in terms of Bray’s percentage yield and percentage Cu uptake. The highest coefficient of correlation was obtained between Cu extractable with neutral 1N NH4OAc and Bray’s per cent yield and per cent Cu uptake. All other methods showed lower values of correlation. The critical level of available Cu estimated with use of neutral normal NH4OAc was 0.16 ppm. Below this value, responses to applied Cu can be expected.  相似文献

Abstract

Remediation of toxic metals by bacteria offers a relatively inexpensive and efficient way for the decontamination of soil and associated environments. The present study was carried out to investigate the surface characteristics, adsorption, and remobilization of Cd and Cu on bacteria and their composites with soil colloidal components, which are the most active constituents in soils. The bacterial strain NTG-01 (Enterobacter aerogenes), which was both Cd- and Cu-resistant, was isolated from a heavily Cu-contaminated soil of the mining area in Daye suburb of Hubei Province, China. Batch laboratory experiments with NTG-01 and soil colloids were performed to quantify adsorption of Cu and Cd. The surface area of kaolinite and the soil colloids from an Alfisol and Ultisol increased by 3.0–8.8% after the introduction of the bacteria. In the presence of bacterial cells, the negative charges of soil colloid systems increased and the positive charges decreased, shifting pH from 4.0 to 6.5. Our results demonstrate that bacteria promote the adsorption of Cd and Cu by kaolinite and soil colloid systems. However, the heavy metals bound by the bacterial composites could also be easily released by NH4NO3 and EDTA. Caution should be taken when using such bacterial strains in bioremediation of heavy metal-contaminated soils.  相似文献

Heavy metal pollution of the soils around an abandoned Pb-Zn mine site located in the Alcudia Valley (South Central Spain) have been characterized by analysis of extractable and total metal concentrations in 60 samples of arable, pasture, and mine lands. The samples showed a broad range of size-particle distribution, cation exchange capacity, and pH values as well as high levels of total metal concentrations (up to 98510 mg kg?1 of Pb, up to 20912 mg kg?1 of Zn, and up to 61 mg kg?1 of Cd). In order to assess the potential availability of metals the metal partitioning in two different soil size fractions (<2 mm and <63 μm) was determined using EDTA and CaCl2 as sequestering reagents. The average contents of Pb, Zn, and Cd in the <63 μm particle size fraction for both extractions were higher than those of the <2 mm fraction due to the high metal adsorption capacity of the fine soil particles. Concentrations of heavy metals extracted by CaCl2 were up to three orders of magnitude lower than those extracted by EDTA, because CaCl2 only extracts the easily mobile fraction. Metal concentrations extracted by both procedures in the two granulometric fractions increased with total metal concentrations, thus increasing the potential environmental risk associated to heavy metal pollution.  相似文献

Functional zooplankton bioassays based on ingestion, reproduction and respiration are described, with methods for a new ingestion bioassay included. All bioassays are compared using three indices, including the variability of controls, the range of experimental responses, and a listing of contaminants causing inhibition/stimulation of response. The ingestion bioassay showed the greatest range of response, and was sensitive to pesticides, PCBs and heavy metals. It was also commonly characterized by a hormesis response. The reproduction bioassay showed the lowest variability, illustrated a reduced range of response, and was sensitive to nutrients and heavy metals. In one study, the respiration bioassay was sensitive only to PCBs.  相似文献

【目的】研究入侵植物薇甘菊提取物对土壤氮素矿化的影响及化感利己作用,为薇甘菊通过释放化感物质促进自身生长并排斥本地植物生长提供实验证据,为揭示薇甘菊的入侵机制提供理论依据。【方法】采用经典的化感生测实验、化感物质添加实验以及盆栽控制实验,对比研究薇甘菊及其本地伴生植物水提液和粗提物的化感生测效应、水提液和粗提物添加对土壤氮素的影响以及水提液添加对薇甘菊生长的影响。【结果】与本地伴生种相比,薇甘菊水提液的化感抑制作用较强且促进了土壤硝态氮的生成;而薇甘菊粗提物的化感作用较弱且抑制了土壤铵态氮和硝态氮的生成。水提液盆栽控制实验结果表明,与本地伴生种火炭母相比,薇甘菊水提液具有显著的化感利己作用,这与薇甘菊水提液处理后薇甘菊生长土壤中的蛋白酶活性增强和有效氮含量增加相一致。【结论】薇甘菊可通过水溶性化感物质促进有效氮的生成来实现化感利己作用。  相似文献

Summary Freshly precipitated ferric and aluminium oxides were added separately to soil and their effects on the availability of both native and added molybdenum were determined using subterranean clover (Trifolium subterraneum L.) as the experimental plant. Both oxides lowered the molybdenum content of the clover and, although it had a smaller specific surface area, ferric oxide had a greater effect than aluminium oxide. In a parallel study with similarly treated soil, the amounts of molybdenum extracted by 0.01M CaCl2, 0.82M NaHCO3 andM NH4OAc were also lowered by both oxides, and ferric oxide again had the greater effect. Extraction with 0.275M NaHC2O4, as proposed elsewhere for assessing available molybdenum, did not show these differences.  相似文献

A field growth chamber study was conducted to determine the effects of ozone and simulated acid rain (SAR) on soil heavy metals. Loblolly pine (Pinus taeda L.), grown in open-top chambers, was exposed to three concentrations of ozone (charcoal filtered air with 0.026 µL O3 L-1, and two non-filtered treatments in which ozone concentrations were 0.074 µL L-1 and 0.147 µL L-1, respectively) and two levels of SAR (pH 3.5 and 5.2). Ozone was applied for 12 h d-1 for 9 months and acid rain deposition was 125 mm event-1. After 9 months exposure, soil pH, organic matter and DTPA-extractable heavy metals (Cd, Pb, Zn, Mn, Fe, Cu) were determined on soil samples collected from exposed chambers at two depths (0–15 cm and 15–30 cm). Simulated acid rain decreased the original soil pH. The concentrations of Cd, Pb and Mn at SAR pH 3.5 were significantly higher than at SAR pH 5.2. Ozone did not affect Zn, Fe and Cu, but a significant interaction between pH and O3 on Mn, Pb and Cd was observed. Due to the poor drainage capacity of this soil, leaching of heavy metals was not observed.  相似文献

Black root rot of tobacco, caused by Thielaviopsis basicola, is generally severe at soil pH values >5.6 and suppressed under more acidic conditions (pH < 5.2). Soil acidifying fertilizers containing NH4–N are generally recommended for burley tobacco production in North Carolina, but the effects of N form and application rate on development of black root rot and on the population dynamics of T. basicola have not been determined. Greenhouse and laboratory studies were conducted to evaluate the effects of N form (NH4 + or NO3 ) and rate on pathogen and disease parameters at several initial soil pH levels. A moderately-conducive field soil, initial pH 4.7, was adjusted to a pH of 5.5 or 6.5 by the addition of CaOH2, then amended with the desired nitrogen form and rate. Pathogen populations were determined over time. In addition, spore production in extracts of roots from plants grown in the various nitrogen and pH treatments was determined. Finally, because tobacco responds to acidic soil conditions and exposure to NH4–N by accumulating high concentrations of the polyamine putrescine, the toxicity of putrescine on vegetative growth and reproduction of T. basicola was investigated. Low soil pH and high levels of NH4–N suppressed reproduction of T. basicola in soil and in root extract, while use of NO3–N and depletion of NH4–N resulted in rapid increases in populations of T. basicola. At 20 mM, putrescine inhibited hyphal growth by 60% and aleuriospore production by 98%. Fertilizers that reduced soil pH also reduced reproduction by T. basicola, and thus have potential for management of black root rot by suppressing populations of T. basicola over multiple years of crop production. The suppression of T. basicola and black root rot observed with NH4–N amendments may partially be due to development of an inhibitory environment in the root and not solely to changes in rhizosphere pH.  相似文献

The amount of phosphorus available to algae in the sediments of four lakes in the western part of the Netherlands has been assessed by means of chemical extraction and bioassay techniques. In addition to direct chemical sediment analyses, extractions were carried out with an NTA column method and a stepwise NH4 Cl-NaOH-HCI shaking method, the latter supposedly separating the weakly bound, the Fe- and Al-bound and the Ca-bound phosphates in the sediments. Bioassays, with sediment as the sole source of P, were made withScenedesmus quadricauda in modified Skulberg's 28 medium to determine the amount of phosphates available to algae.The average total P concentration of the sediments varied from 0.8 to 3.6 mg P g–1 dry wt and correlated well with the net external P loading of the lakes. Uptake of P by algae in the bioassays varied from 0.4 to 36% — while NTA extracted 36–69% of the total P. The ratio NH4Cl extracted/ NaOH extracted/ HCI extracted phosphates is different from lake to lake, although in all lakes the highest extractions (27–62% of total P) are found in the NaOH fraction. However, in the peaty sediments of these lakes, the NaOH step extracted not only the Fe- and Al-bound phosphates but, also, large amounts of humus compounds. Hence, this fraction also contains non-available organic P.The results are related to soil type and chemical characteristics of the sediments, and compared with data from other authors. A positive correlation was found between phosphate available to algae and NTA- and NaOH-extractable P, but the correlation with total phosphorus was higher. Moreover, algal-extractable P proved to be positively correlated with total iron and clay content and negatively with the amount of organic matter.It is concluded that the sediments in the investigated lakes show great variability and that the chemical extraction techniques cannot replace the bioassays to assess the amount of phosphorus available to algae.  相似文献

  • This study evaluated the impacts of Canada goldenrod (Solidago canadensis L.) leaf extracts at concentrations of 0, 10 or 20 gl 1 on the germination of lettuce under different forms of heavy metal pollution (Cu2+, Pb2+ or a combination of Cu2+ and Pb2+; 35 mgl 1) during incubation in Petri dishes for 10 days.
  • Goldenrod leaf extracts (high concentration) reduced growth of aboveground and belowground parts of lettuce as well as competition for light and soil nutrients. However, low concentrations of goldenrod leaf extracts dramatically improved growth of lettuce roots, competition for light, soil nutrient availability, leaf photosynthetic area and growth competitiveness. The combination of goldenrod leaf extracts and heavy metal pollution was synergistic on most lettuce germination parameters, probably because high concentrations of goldenrod leaf extracts together with heavy metal pollution had a synergistic negative impact on lettuce germination.
  • Consequently, increased levels of heavy metal pollution may favour invasion of invasive alien species while largely suppressing germination of indigenous species.
