Summary There is no significant change in Ca++ efflux rate from plasmodia during chemotactic responses to several sugars, whereas substantially increased Ca++ efflux caused by EDTA does not significantly affect movement. Synopsis A new device is described for improving the accuracy of measuring the thickness of cryostat sections by the focusing technique in the light microscope. The necessity of such measurements is demonstrated by the great variation (range 2.55 m–11.93 m) in the thickness of serial cross-sections of frozen muscle biopsies from 12 healthy men. The final dehydration of the sections was found to reduce the thickness of fresh sections by 47%. However, dehydration caused the cross-sectional area to be reduced by only 2.8%. 相似文献

In barley (Hordeum vulgare L.) leaves, differential ion accumulation commonly results in inorganic phosphate (Pi) being confined to the mesophyll and Ca(2+) to the epidermis, with preferential epidermal accumulation of Cl(-), Na(+), and some other ions. The pattern was confirmed in this study for major inorganic anions and cations by analysis of barley leaf protoplasts. The work focused on the extent to which differences in plasma membrane ion transport processes underlie these observations. Ion transport across the plasma membrane of barley epidermal and mesophyll protoplasts was investigated electrophysiologically (by microelectrode impalement and patch clamping) and radiometrically. Data from both approaches suggested that similar types of ion-selective channels and membrane transporters, which catalyze the transport of Ca(2+), K(+), Na(+), and Pi, exist in the plasma membrane of the two cell types. In general, the simple presence or absence of ion transporters could not explain cell-type-specific differences in ion accumulation. However, patch-clamp data suggested that differential regulation of instantaneously activating ion channels in the plasma membrane could explain the preferential accumulation of Na(+) in the epidermis. 相似文献

In this work we explored the potential of quantum dots for fluorescent detection of lymphoid surface antigens. To optimize detection with quantum dots, we upgraded a fluorescent microscope that allowed us obtaining multiple images from different quantum dots on a single section. Specimens stained with quantum dots remained stable over two weeks and practically did not bleach under the mercury lamp during scores of minutes. Double staining of frozen sections with direct conjugates of quantum dots with primary mouse monoclonal antibodies demonstrated direct conjugate high specificity and sensitivity. High stability of quantum dots’ fluorescence allows their use in diagnostics to analyze antigen coexpression on lymphoid tissue sections. “Spillover” of fluorescent signals from quantum dots into adjacent fluorescent channels maximally separated by 40 nm did not exceed 8%, which renders spectral compensation unnecessary. 相似文献

Despite the large amount of geomorphological, geodynamic and geophysical data obtained from Mars missions, much is still unknown about Martian mineralogy and paragenetic assemblages, which is fundamental to an understanding of its entire geological history. Minerals are not only indicators of the physical–chemical settings of the different environments and their later changes, but also they could (and do) play a crucial astrobiological role related with the possibility of existence of extinct or extant Martian life. This paper aims: (1) to present a synoptic review of the main water-related Martian minerals (mainly jarosite and other sulfates) discovered up to the present time; (2) to emphasize their significance as environmental geomarkers, on the basis of their geological settings and mineral parageneses on earth (in particular in the context of some selected terrestrial analogues), and (3) to show that their differential UV shielding properties, against the hostile environmental conditions of the Martian surface, are of a great importance for the search for extraterrestrial life. 相似文献

Effects of diethylstilbestrol (DES) on ion fluxes in oat roots (Avena sativa L.) were investigated by measuring K+ and Cl− absorption and K+ efflux. DES rapidly decreased the absorption of K+ (86Rb) and 36Cl− by excised roots; 10−4 molar DES inhibited Cl− absorption in 1 minute and K+ absorption in 1 to 2 minutes. With a 10-minute incubation period, K+ and Cl− absorption were inhibited 50% by 1.1×10−5 molar and 8.4×10−6 molar DES, respectively. Treatment for 3 minutes with 10−4 molar DES caused irreversible inhibition of K+ absorption. Increasing concentrations of KCl in the absorption media decreased the DES inhibition. Experiments with the DES analogs, DES dipropionate, dienestrol and hexestrol, showed that the steric configuration and the hydroxyl group of the DES molecule are important in determining the inhibitory capacity of the compound. 相似文献

Abstract Effects of temperature on the ionic relations and energy metabolism of Chara corallina were investigated. Measurements were made of the ionic content, tracer ion fluxes, and photosynthetic and dark CO2 fixation in isolated cells, and of O2 exchange in photosynthesis and respiration in isolated shoot apices. The total intracellular concentration of K+, Na+ and Cl? was the same in cells held for 5 days in non-growing medium at 15°C (the growth temperature) as in those held at 25°C or 5°C. The tracer influx in the light of all ions tested (Rb+, Na+, CH3NH3+, Cl? and H2PO4?) was lower at 5°C than at 15°C in experiments in which cells were subjected to 5°C for less than 24 h in toto. The influx at 25°C was greater than that at 15°C for H2PO?4, there was no difference between the two temperatures for Na+, while the influx at 25°C was less than that at 15°C for Cl?, Rb+ and CH3NH3+ For Cl? and H2PO?4 similar results were found in later experiments with cells grown at 20—23°C. Photosynthetic CO2 fixation and O2 evolution, and respiratory O2 uptake, are greater at 25°C, and lower at 5°C, than they are at the growth temperature of 15°C. In longer-term pretreatments at the different temperatures, tracer Cl? influx at 15°C and particularly at 25°C were lower than in short-term experiments, while the influx at 5°C was higher. It was concluded from these experiments, and from previous data on H+ free energy differences across the plasmalemma, that (1) the maintenance of internal ion concentrations involves a close balancing of influx and efflux of K+, Na+ and Cl? at all experimental temperatures; (2) the regulation of the tracer fluxes of the ions is kinetic rather than thermodynamic and (3) that the tracer fluxes at low temperatures are not restricted by the rate at which respiration or photosynthesis can supply energy to them. 相似文献

An analytical model, which describes the stationary transformation of light energy to the energy of pigment electronic excitation, has been constructed. A proton pump of the thylakoid membrane has been considered as a two-level conformon. The difference between the energies of the excited and ground states of both the pigment and the protein complex is assumed to be the energy of an absorbed photon. It has been found how the concentration of ions in a lumen and the potential across the thylakoid membrane depend on the concentration of ions in the stroma and the brightness temperature of absorbed radiation. Conditions for the maximum efficiency of the photosynthesis process have been analyzed. This model has been used to determine the electric potential (φ≈6.7 mV) at the chloroplast thylakoid membrane. The calculated value of the electric potential is in good agreement with the experimental data. A limitation on the stoichiometric coefficient of the proton transport through ATP-synthase, m>3, has been found theoretically. 相似文献

A continuous production of fructooligosaccharides from sucrose was investigated by fructosyltransferase immobilized on a high porous resin, Diaion HPA 25. The optimum pH (5.5) and temperature (55°C) of the enzyme for activity was unaltered by immobilization, and the immobilized enzyme became less sensitive to the pH change. The optimal operation conditions of the immobilized enzyme column for maximizing the productivity were as follows: 600 g/L of sucrose feed concentration, flow rate of superficial space velocity 2.7 h?1. When the enzyme column was run at 50°C, about 8% loss of the initial activity of immobilized enzyme was observed after 30 days of continuous operation, during which high productivity of 1174 g/L·h was achieved. The kinds of products obtained using the immobilized enzyme were almost the same as those using soluble enzymes or free cells. 相似文献

We present a simple two-parameter model for surface charge directly associated with ion channels. A spherically symmetric "charged shell" models a distribution of surface charge arrayed about the channel entrance, with a corresponding set of image charges behind the plane of the membrane. The transition between a regime of buffered conductance and a regime of rapidly falling conductance at very low ionic strength is found to depend on the magnitude of the surface charge as well as the separation between the charge and the channel entrance. This resolves an apparent discrepancy between the experimental findings of Naranjo and Latorre (1993. Biophys. J. 64:1038-1050) and previous theoretical computations. The charged-shell model is used in a comparative study of the toad skeletal muscle conductance data of Naranjo and Latorre, the rat skeletal muscle conductances of Ravindran et al. (1992. Biophys. J. 61:494-508), and a second set of rat muscle conductances presented in this paper. 相似文献

Using transgenic Nicotiana plumbaginifolia seedlings in which the calcium reporter aequorin is targeted to the chloroplast stroma, we found that darkness stimulates a considerable flux of Ca(2+) into the stroma. This Ca(2+) flux did not occur immediately after the light-to-dark transition but began approximately 5 min after lights off and increased to a peak at approximately 20 to 30 min after the onset of darkness. Imaging of aequorin emission confirmed that the dark-stimulated luminescence emanated from chloroplast-containing tissues of the seedling. The magnitude of the Ca(2+) flux was proportional to the duration of light exposure (24 to 120 h) before lights off; the longer the duration of light exposure, the larger the dark-stimulated Ca(2+) flux. On the other hand, the magnitude of the dark-stimulated Ca(2+) flux did not appear to vary as a function of circadian time. When seedlings were maintained on a 24-h light/dark cycle, there was a stromal Ca(2+) burst after lights off every day. Moreover, the waveform of the Ca(2+) spike was different during long-day versus short-day light/dark cycles. The dark-stimulated Ca(2+) flux into the chloroplastidic stroma appeared to affect transient changes in cytosolic Ca(2+) levels. DCMU, an inhibitor of photosynthetic electron transport, caused a significant increase in stromal Ca(2+) levels in the light but did not affect the magnitude of the dark-stimulated Ca(2+) flux. This robust Ca(2+) flux likely plays regulatory roles in the sensing of both light/dark transitions and photoperiod. 相似文献
