`Black Mexican Sweet' (BMS) maize (Zea mays L.) tissue cultures were selected for tolerance to sethoxydim. Sethoxydim, a cyclohexanedione, and haloxyfop, an aryloxyphenoxypropionate, exert herbicidal activity on most monocots including maize by inhibiting acetyl-coenzyme A carboxylase (ACCase). Selected line B10S grew on medium containing 10 micromolar sethoxydim. Lines B50S and B100S were subsequent selections from B10S that grew on medium containing 50 and 100 micromolar sethoxydim, respectively. Growth rates of BMS, B10S, B50S, and B100S were similar in the absence of herbicide. Herbicide concentrations reducing growth by 50% were 0.6, 4.5, 35, and 26 micromolar sethoxydim and 0.06, 0.5, 5.4, and 1.8 micromolar haloxyfop for BMS, B10S, B50S, and B100S, respectively. Sethoxydim and haloxyfop concentrations that inhibited ACCase by 50% were similar for BMS, B10S, B50S, and B100S. However, ACCase activities were 6.01, 10.7, 16.1, and 11.4 nmol HCO3− incorporated per milligram of protein per minute in extracts of BMS, B10S, B50S, and B100S, respectively, suggesting that increased wild-type ACCase activity conferred herbicide tolerance. Incorporation of [14C]acetate into the nonpolar lipid fraction was higher for B50S than for BMS in the absence of sethoxydim providing further evidence for an increase in ACCase activity in the selected line. In the presence of 5 micromolar sethoxydim, [14C]acetate incorporation by B50S was similar to that for untreated BMS. The levels of a biotin-containing polypeptide (about 220,000 molecular weight), presumably the ACCase subunit, were increased in the tissue cultures that exhibited elevated ACCase activity indicating overproduction of the ACCase enzyme. 相似文献

超积累植物由于其对重金属具有地上部超积累以及超耐受等特性,不仅是研究植物离子转运及毒性耐受的理想模式,而且在植物修复的发展和应用中具有不可替代的作用。伴矿景天是近年在我国境内发现的一种景天科镉(Cd)/锌(Zn)超积累植物。为鉴定其富集和耐受Cd的关键基因,笔者构建了其酵母表达cDNA文库,利用酵母的遗传互补系统筛选到一个极大提高了酵母抗Cd能力的基因SpMT2。SpMT2属于富含半胱氨酸(Cys)的金属硫蛋白(Metallothionein)家族。亚细胞定位表明SpMT2表达于酵母细胞质中,并特异地提高酵母对Cd的抗性。进一步研究发现SpMT2的表达显著降低了酵母液泡中Cd含量,但酵母吸收的总Cd含量无显著变化。推测抗性增加是由于SpMT2在酵母细胞质中通过螯合Cd从而降低Cd对酵母的毒害。qRT-PCR分析表明SpMT2在伴矿景天的根和地上部都高丰度表达,且不受Cd诱导变化。鉴于SpMT2也定位于植物细胞质中,结合上述结果,推测SpMT2可能在伴矿景天细胞质中螯合Cd,在降低Cd毒害的同时可能还保持Cd在细胞质中的流动性,从而在Cd长途转运过程中也发挥重要作用。 相似文献

Cultured carrot (Daucus carota L.) cells were adapted to growing in 25 millimolar glyphosate by transfer into progressively higher concentrations of the herbicide. Tolerance was increased 52-fold, and the adaptation was stable in the absence of glyphosate. The uptake of glyphosate was similar for adapted and nonadapted cells. Activity of the enzyme 5-enolpyruvylshikimic acid-3-phosphate synthase was 12-fold higher in the adapted line compared to nonadapted cells, while activities of shikimate dehydrogenase and anthranilate synthase were similar in the two cell types. The adapted cells had higher levels of free amino acids—especially threonine, methionine, tyrosine, phenylalanine, tryptophan, histidine, and arginine—than did nonadapted cells. Glyphosate treatment caused decreases of 50 to 65% in the levels of serine, glycine, methionine, tyrosine, phenylalanine, and tryptophan in nonadapted cells, but caused little change in free amino acid levels in adapted cells.
The adaptation reported here supports the growing body of evidence linking tolerance to glyphosate with increased levels of the enzyme 5-enolpyruvylshikimic acid-3-phosphate synthase. The elevated levels of aromatic amino acids, which may confer resistance in adapted cells, suggest that control of the shikimate pathway may be altered in these cells.

Cell suspension cultures of tomato (Lycopersicon esculentum) adapted to growing continuously in the presence of 0.1 mM CdCl2 and accumulated phytochelatins (PCs, poly(-Glu-Cys)n-Gly). The highest level of PCs was measured 4 days after inoculation and coincided with the peak of cellular cadmium concentration. At this time there was an 8-fold molar excess of PC (-Glu-Cys) over Cd. PCs could not be detected after 12 days when the cellular concentration of cadmium was 0.2 mM. These results indicate that PCs are produced in excess of that required to bind the cellular cadmium in the early stage of the culture period followed by degradation of PCs during the stationary phase. Adaptation to 0.1 mM CdCl2 did not increase tolerance to higher concentrations of cadmium when compared with control cells, but did significantly enhance tolerance to both anaerobiosis and heat shock. Exposure of tomato cells to 0.1 mM CdCl2 resulted in several changes in proteins synthesized. 相似文献

Since NO3− availability in the rooting medium seriously limits symbiotic N2 fixation by soybean (Glycine max [L.] Merr.), studies were initiated to select nodulation mutants which were more tolerant to NO3− and were adapted to the Midwest area of the United States. Three independent mutants were selected in the M2 generation from ethyl methanesulfonate or N-nitroso-N-methylurea mutagenized Williams seed. All three mutants (designated NOD1-3, NOD2-4, and NOD3-7) were more extensively nodulated (427 to 770 nodules plant−1) than the Williams parent (187 nodules plant−1) under zero-N growth conditions. This provided evidence that the mutational event(s) affected autoregulatory control of nodulation. Moreover, all three mutants were partially tolerant to NO3−; each retained greater acetylene reduction activity when grown hydroponically with 15 millimolar NO3− than did Williams at 1.5 millimolar NO3−. The NO3− tolerance did not appear to be related to an altered ability to take up or metabolize NO3−, based on solution NO3− depletion and on in vivo nitrate reductase assays. Enhanced nodulation appeared to be controlled by the host plant, being consistent across four Bradyrhizobium japonicum strains tested. In general, the mutant lines produced less dry weight than the control, with root dry weights being more affected than shoot dry weights. The nodulation trait has been stable through the M5 generation in all three mutants. 相似文献

Aspergillus carbonarius and a strain of Penicillium, a cadmium tolerant fungi, are able to metabolize cadmium chloride up to 2% (w/v). Their amino acids analysis on cadmium free and cadmium chloride containing media indicated certain disorders in their metabolic activities. Cystathionine was only detected in both fungi in the presence of cadmium chloride. However, cadmium was incorporated into several types of low and high molecular weight proteins. The amino acids hydrolyzates of a cadmium containing protein are characterized by the presence of high levels of sulfur amino acids; cysteine and methionine. It has been postulated that vacuolar compartmentation might play an important role in naturally selected cadmium tolerance in Silene vulgaris (Moench.) Garcke (Bladder campion). Additionally, a tendency of heavy metals to accumulate in the epidermis has been reported. Since these factors would affect the distribution of cadmium in leaves, we determined the distribution of cadmium in leaves of cadmium tolerant and sensitive ecotypes of Silene vulgaris at different levels of exposure and at different time intervals. Cadmium concentrations were higher in leaves of sensitive plants than in those of cadmium tolerant ones after identical exposure to cadmium for a period of 8 days. The highest cadmium concentrations were found in the lower epidermis of plants of both ecotypes. The amount of cadmium located at the lower epidermis was highest for sensitive plants, although the stomatal density was lower in the sensitive ecotype than in the tolerant one. A possible explanation for this phenomenon is the weak relationship between transpiration (water flow) and element allocation. Our results support the hypothesis that vacuolar storage of cadmium plays an important role in the mechanism of cadmium tolerance in Silene vulgaris . 相似文献

Thirty-day-old seedlings of tomato (Lycopersicon esculentum cv. Kwangsoo) were treated with various cadmium (Cd) concentrations (0, 10, 50, 100, and 500 μM) for up to 20 days, and the
detailed distribution of absorbed Cd and its phytotoxicity in different plant parts (root, stem, and leaves) were investigated.
The accumulation of Cd in plants increased with external Cd concentrations and Cd was strongly retained by roots, with less
than 30% of the absorbed Cd being transported to shoots. Among the leaves, the lower positioned older leaves accumulated more
Cd than the younger leaves. Furthermore, Cd-exposure not only reduced the dry weight and length of both shoot and root, chlorophyll
levels in leaves, and levels of photosynthesis, but also enhanced the concentration of malondialdehyde (a lipid peroxidation
product) in all plant parts. Our results indicate that the physiological impairment of tomato seedlings exposed to toxic levels
of Cd may be related to the internal distribution of absorbed Cd, prolonged exposure, and oxidative stress in different plant

Suspension cultures of Lycopersicon peruvianum were selected for resistance to cadmium by stepwise exposure to increasing concentrations of cadmium sulfate. Resistant cells grow in 1500 micromolar Cd++. This resistance was retained for thirty generations without selection. Both resistant and parental sensitive cultures take up Cd++ at similar rates and to the same final levels. Exposure of sensitive or resistant cultures to Cd++, Cu++, or Zn++ leads to the intracellular accumulation of a low molecular weight, cysteine-rich, cadmium-binding protein. This metallothionein is induced over fifteen fold by 100 M cadmium and builds up to about five fold higher levels in the resistant cultures.Abbreviations Cd++

Sodium chloride tolerant organogenic callus lines of Nicotiana tabacum were developed in vitro on Murashige and Skoog [16] medium supplemented with BA, IAA and different concentration of NaCl. The maximum shoot bud regeneration was achieved from both tolerant and non-tolerant calluses on MS medium supplemented with 1.0 mg/l BA, 0.1 mg/l IAA with or without NaCl within 4 weeks of culture. Standard growth parameters such as fresh weight and dry weight of organogenic callus, growth tolerant index and enzyme activity (peroxidase and catalase) were used as indicators of salt tolerance. The growth tolerance index in the 4-week after the beginning of treatments yielded significant differences among the non-tolerant and tolerant organogenic callus lines. The regenerated shoots were rooted on half-strength MS basal salts supplemented with 2% sucrose but devoid of growth regulator. The regenerated plants from tolerant callus lines were capable of growing in vitro in presence of 175 mM NaCl. SDS-PAGE profile showed that the progenies derived from tolerant sources were tolerant to salt. This investigation may help in the selection and characterization of salt tolerance in plant improvement programme. 相似文献

This paper reports techniques for isolation, selection and long-term passage of bovine aortic endothelium (BAE). A [3H]thymidine-selection technique was developed to limit overgrowth of cultures by contaminating smooth-muscle cells. The resulting cultures could be passaged for a replicative life span of 35 to 40 doublings and maintained a stable, normal karyotpye throughout this period. Despite the fact that these cultures reached a stable monolayer with density-inhibited growth state, postconfluent cells showed focal areas of a second growth pattern called "sprouting." This was seen only when cultures were maintained at high densities for periods of 1 to 2 weeks. Ultrastructural analysis, as well as immunofluorescence studies with markers for endothelial cells (factor VIII) and smooth-muscle cells (actin), indicates that this phenomenon is not due to overgrowth of a residual population of smooth-muscle cells, but may represent a second growth pattern of the endothelial cells themselves. 相似文献

The physiological changes induced by a daily increase of NaCl level, over a period of 4 d, were studied in leaves of the salt-sensitive cultivated tomato species Lycopersicon esculentum and its wild salt-tolerant relative Lycopersicon pennellii. A higher solute contribution to the osmotic adjustment was observed in NaCl-treated leaves of L. pennellii than in those of L. esculentum. This response together with the higher accumulation of inorganic solutes in the wild species and of organic solutes in the cultivated species verified the different salt tolerance mechanisms operating in the two species in the short-term. With regard to the changes induced by salt stress on the free polyamine levels, the putrescine and spermine levels increased with salinity, whereas the spermine levels decreased in both tomato species; nevertheless, the main difference between the two species lays in an earlier and greater accumulation of putrescine induced by salinity in L. pennellii than in L. esculentum. The changes in putrescine levels were associated to changes in amino acids related to its synthesis, and the changes were different in both species. In L. esculentum, the high concentrations of some intermediate compounds (glutamate and arginine) were related to the low accumulation rate of both proline and putrescine. In contrast, in L. pennellii, important reductions in glutamate and arginine levels were found at the end of the salinization period. Moreover, in this last situation, a decline in the putrescine level ran parallel to a high proline accumulation, which suggests that the higher the stress level, the higher the deviation of glutamate to proline occurring in the salt tolerant species. It could be concluded that an early accumulation of the diamine putrescine seems to be associated with salt tolerance in the short-term. 相似文献

The shoots of cultivated tomato (Lycopersicon esculentum cv. T5) wilt if their roots are exposed to chilling temperatures of around 5 °C. Under the same treatment, a chilling‐tolerant congener (Lycopersicon hirsutum LA 1778) maintains shoot turgor. To determine the physiological basis of this differential response, the effect of chilling on both excised roots and roots of intact plants in pressure chambers were investigated. In excised roots and intact plants, root hydraulic conductance declined with temperature to nearly twice the extent expected from the temperature dependence of the viscosity of water, but the response was similar in both species. The species differed markedly, however, in stomatal behaviour: in L. hirsutum, stomatal conductance declined as root temperatures were lowered, whereas the stomata of L. esculentum remained open until the roots reached 5 °C, and the plants became flaccid and suffered damage. Grafted plants with the shoots of one genotype and roots of another indicated that the differential stomatal behaviour during root chilling has distinct shoot and root components. 相似文献

The effect of cadmium on growth and contents of glutathione (GSH) and phytochelatins (PCs) were investigated in roots and
leaves of tomato plants (Lycopersicon esculentum Mill. cv. 63/5 F1). The accumulation of Cd increased with external Cd concentrations and was considerably higher in roots
than in leaves. Dry mass production decreased under Cd treatment especially in leaves. In both roots and leaves, exposure
to Cd caused an appreciable decline in GSH contents and increase in PCs synthesis proportional to Cd concentrations in the
growth medium. At the same Cd concentration, PCs production was higher in roots than in leaves. The implication of glutathione
in PC synthesis was strongly suggested by the use of buthionine sulfoximine (BSO). The major fraction of Cd accumulated by

+ accumulation seems to be involved in regulating the toxicity of rice seedlings caused by CdCl2. This conclusion was based on the observations that (a) on treatment with CdCl2, NH4
+ content increased rapidly in the leaves of the Cd-sensitive cultivar (cv. Taichung Native 1, TN1) but not in the Cd-tolerant cultivar (cv. Tainumg 67, TNG67), (b) pretreatment with abscisic acid (ABA) enhanced Cd tolerance and reduced Cd-induced NH4
+ accumulation in TN1 seedlings, (c) exogenous application of the ABA biosynthesis inhibitor, fluridone, decreased Cd tolerance and increased NH4
+ content in leaves of TNG67, (d) exogenous application of phosphinothricin, an inhibitor of glutamine synthetase (GS), which resulted in NH4
+ accumulation in the leaves, also induced toxicity similar to Cd in TN1 seedlings. Evidence is presented to show that Cd-induced NH4
+ accumulation in TN1 leaves is attributable to a decrease in GS activity. Since Cd-treated TN1 leaves had higher glutamine and glutamate contents than control leaves, it is unlikely that glutamine (or glutamate) depletion is the mechanism which regulates Cd-induced toxicity. 相似文献

Four cell lines of tomato, Lycopersicon esculentum Mill. cv VFNT-Cherry, were selected for their ability to grow in the presence of up to 6 millimolar CdCl2. The intracellular Cd concentration in these cells was at least 2.3 times higher than in the medium. Growth in media containing higher concentrations of Cd was accompanied by increased production of Cd-binding phytochelatins and a trend toward accumulation of higher molecular weight phytochelatins. At least 90% of the Cd in the most tolerant cells was associated with Cd-phytochelatin complexes. Treatment of tomato seedlings (Lycopersicon esculentum Mill. cv. 63/5 F1) with increasing CdCl2 concentrations in the culture medium resulted in Cd accumulation more important in roots than in leaves. Biomass production
was severely inhibited, even at low Cd concentration. Cd reduced chlorophyll content in leaves and enhanced lipid peroxidation.
An increase in antioxidative enzyme (superoxide dismutase, ascorbate peroxidase, guaiacol peroxidase, glutathione reductase)
activities was more pronounced in leaves than in roots, while catalase activity increased only in roots. In addition, changes

Yeast strains are commonly associated with sugar rich environments. Various fruit samples were selected as source for isolating
yeast cells. The isolated cultures were identified at Genus level by colony morphology, biochemical characteristics and cell
morphological characters. An attempt has been made to check the viability of yeast cells under different concentrations of ethanol.
Ethanol tolerance of each strain was studied by allowing the yeast to grow in liquid YEPD (Yeast Extract Peptone Dextrose)
medium having different concentrations of ethanol. A total of fifteen yeast strains isolated from different samples were used for the
study. Seven strains of Saccharomyces cerevisiae obtained from different fruit sources were screened for ethanol tolerance. The
results obtained in this study show a range of tolerance levels between 7%-12% in all the stains. Further, the cluster analysis based
on 22 RAPD (Random Amplified polymorphic DNA) bands revealed polymorphisms in these seven Saccharomyces strains. 相似文献
