野菊与菊花杂交中花粉活力和柱头可授性及胚胎发育研究   总被引:4,自引:1,他引:3  
应用石蜡制片、活体压片、光学显微镜及扫描电子显微镜观察等方法,研究了四倍体河南云台山野菊(Dendranthema indicum)与栽培菊花'钟山金山'(D.grandiflorum 'Zhongshanjinshan')种间杂交中父本花粉活力、花粉在柱头萌发、花粉管生长及胚胎发育情况等.结果发现,父本云台山野菊的花粉活力在授粉时为12%左右.人工授粉后的不同时间,在柱头上都观察到正常萌发的花粉粒,且花粉管都能进入柱头,其中,在授粉后0.5 h时,平均每柱头有5.9粒花粉萌发;12 h时,为59.9粒;而24和48 h时,则分别降为47.1和35.7粒.此外,在授粉后8、10、12和15 d时,分别在49.1%、40.8%、39.7%和38.5%子房内观察到正常发育的胚胎,最终杂交结实率为44.8%,而母本自然开放结实率为52.3%.研究表明,授粉前其多数母本雌蕊发育良好、授粉后多数花粉能在柱头正常萌发和花粉管正常生长,在受精后大部分胚胎发育正常是野菊与栽培菊种间杂交较高结实率的重要保证,而授粉前父本较低的花粉活力对杂交结实率影响不大.  相似文献

The cross B. campestris ssp. japonica x B. oleracea var. botrytis fails due to incompatibility barrier at the stigma. To realize this cross, irradiated compatible pollen (mentor pollen) was used before the incompatible pollination. The presence of mentor pollen stimulated the incompatible pollen to germinate and effect fertilization and seed set. One hybrid was thus obtained. Most of the seeds were inviable. Of the 5 plants raised one was a hybrid and 4 resembled the female parent.  相似文献

Based on the biology peculiarity of Liriodendron L., the non-bagged pollination technique was used widely in cross breeding of the genus. Theoretical analysis and indirect detection indicated that the pollen contamination rate of this pollination technique was no higher than 1%. But there is no evidence from direct detection by now. The pollen contamination rate produced in this pollination technique, for the first time, was detected by means of RAPD markers in this study. The results revealed that all the 93 progenies detected, derived from the non-bagged pollination technique, were progenies of the purpose male tree of the cross. No pollen contamination was found. Because the frequency of the 4 paternal marker fragments selected for the detection were relatively low in the population, the reliability of the pollen contamination rate detected was very high (>99%). Therefore, the non-bagged pollination technique is viable in Liriodendron . The authors also considered that RAPD markers had higher potential of use in other fields of botanic study such as pollen competition, pollen flow, pollen contamination of forest seed orchard, etc.  相似文献

根据鹅掌楸属 (LiriodendronL .)的生物学特点 ,实践上常采用不套袋杂交授粉技术 ,理论分析和间接检测结果证明这种杂交授粉方式的花粉污染率低于 1% ,但到目前为止仍缺乏直接的检测证据。本试验首次采用RAPD标记对鹅掌楸属种间不套袋杂交授粉的花粉污染率进行了检测 ,结果证明 93个受检不套袋杂交授粉子代均来自目的父本 ,而非花粉污染所致。通过对所用 4个父本标记谱带在试验群体中出现频率的测定和分析 ,证明这种检测结果的可靠性大于 99% ,从而认为对鹅掌楸属进行不套袋杂交授粉是可行的。并就这一方法在植物学研究领域的应用潜力进行了讨论  相似文献

The primary hypothesis of this study was that in the Galápagos Islands, fruit and seed set via nocturnal pollination would exceed that of diurnal pollination because of the greater insect activity at night typical of hot, arid regions. Clerodendrum molle, a widespread member of the Galápagos flora was submitted to pollination experiments, visitor observations, nectar sampling, pollen transfer studies, pollen?Covule ratio studies, and pollen measurements. Flowers set fruit and seed via open pollination, autonomous autogamy, facilitated autogamy, facilitated cross-pollination, diurnal pollination, and nocturnal pollination. Results indicate no significant difference in fruit set. Cross-pollinated flowers showed a significant increase in seed set over all treatments except facilitated autogamy. Nocturnal and diurnal fruit and seed set did not differ significantly. Nocturnal visitors included ants, spiders, hawk moths, and roaches whereas diurnal visitors included carpenter bees and ants. Galápagos members of C. molle exhibit incomplete protandry and set fruit via autonomous autogamy as a result of natural selection in an environment with few faithful pollinators. Overall low fruit and seed set are likely because of a combination of pollen and resource limitation, the latter exacerbated by nectar robbing. Studies of island angiosperms are crucial for complete understanding of the population dynamics of indigenous plants, which can assist conservation officials in protecting these species.  相似文献

The limited seed production of insect-depended plant, Liriodendron chinense was once considered to be pollen-limited and insufficient cross pollination. In this study, we counted pollen grains deposited on stigmas in three populations in Guizhou, Hunan and Zhejiang provinces of China respectively. Over 61.9% stigmas were pollinated. From 1994 to 1996, the mean number of pollen grains on each stigma ranged from 4.4 to 42.6, much more than ovules(2) in each pistil. Based on observations of three years, both the pollination rate and pollen quantity on stigmas significantly affected seed set. When flowers opened without stamens dehiscencing at the early stage of anthesis, stigmas received considerable quantity of pollen grains. Pollen grains from different sources were able to germinate and pollen tube growth rates were not greatly variable. It is very likely that pollen grains arrived first would fertilize eggs. Since only several pollen tubes went through the stylar canal, the potential pollen competition may exist. In this case, there would be strong selection on floral syndrome which benefit early insect visits. Pollen grains from the early visits would have a greater chance to fertilize ovules than those from later visits, which implies that cross pollination is the predominant breeding system of this plant. The conclusion was also confirmed by following four artificial experiments. Three treatments, including flowers bagged, netted or with the perianth removed, all reduced seed set clearly, but flowers with the stamens removed (emasculation)had a higher seed production by open pollination. As the rates of deposited stigmas in three populations were 6~8 times more than full seed set, we considerthat lower seed production in this plant may not mainly be due to pollen limitation.  相似文献

Self-incompatibility and synchronous protandrous dichogamy have previously been reported in Ziziphus species. In this work, we conducted a comparative analysis of fluorescence microscopy observations of pollen tube growth following controlled cross pollinations of emasculated flowers and self-pollinations of non-emasculated flowers in three Ziziphus species, Z. jujuba, Z. mauritiana and Z. spina-christi, with the aim to determine the type of the self-incompatibility system of each species. In addition, to test whether autonomous self-pollination, parthenocarpy or agamospermy occurs, flowers were emasculated (or not) and covered. Fruit set and seed viability were monitored. The presence of binucleate pollen grains and the cessation of pollen tube growth in the style suggest that the self-incompatibility system operating in the studied Ziziphus species is gametophytically controlled. Controlled self-pollination in Z. mauritiana resulted in fruits that dropped off before maturation, whereas in Z. spina-christi the flowers dropped off one or two days after pollination. Following controlled self-pollination, small fruits lacking viable seeds were obtained in Z. jujuba, probably due to the stimulus provided by pollination (stimulative parthenocarpy). In the cultivar Tamar of Z. jujuba, the relatively high percentage of seedless fruits obtained in emasculated bagged flowers without hand pollination suggests that this cultivar can set seedless fruits without any pollination stimulus.  相似文献

The effect of self pollen on the success of cross pollinations was studied in Campsis radicans (L.) Seem., a species previously considered self-sterile. The application of self pollen to stigmas in combination with or preceding the application of cross pollen markedly reduced the likelihood of fruit production. This effect was not due to the dilution of cross pollen nor to physical blocking of the stigmatic surface, since mixtures of talc and cross pollen were as effective as pure cross pollen in causing fruit production. Pollen tubes produced by self pollen grew at rates similar to those from cross pollen, and penetrated the ovary. The interference caused by self pollen appears to take place in the ovary, although it cannot be stated to what degree it is prezygotic or postzygotic. The marked protandry occurring in C. radicans is presumed to be due at least partly to selection for avoidance of these negative effects on female reproductive success. Up to 33% of viable seeds from mixed self + cross pollinations were sired by self pollen. The term “cryptic self-fertility” is coined to describe this phenomenon where pollination with loads of pure self pollen rarely or never yields fruit, but pollination using mixtures of self and cross pollen yields fruit containing considerable numbers of selfed seed. Estimates of selfing frequency that are based on pollination using loads of purely self pollen will be in error for species possessing cryptic self-fertility.  相似文献

Background and Aims

Animal pollination is typically an uncertain process that interacts with self-incompatibility status to determine reproductive success. Seed set is often pollen-limited, but species with late-acting self-incompatibility (SI) may be particularly vulnerable, if self-pollen deposition results in ovule discounting. Pollination is examined and the occurrence of late-acting SI and ovule discounting assessed in Cyrtanthus breviflorus.

Methods

The pollination system was characterized by observing floral visitors and assessing nectar production and spectral reflectance of flowers. To assess late-acting SI and ovule discounting, growth of self- and cross-pollen tubes, and seed set following open pollination or hand pollination with varying proportions of self- and cross-pollen, were examined.

Key Results

Native honeybees Apis mellifera scutellata pollinated flowers as they actively collected pollen. Most flowers (≥70 %) did not contain nectar, while the rest produced minute volumes of dilute nectar. The flowers which are yellow to humans are visually conspicuous to bees with a strong contrast between UV-reflecting tepals and UV-absorbing anthers and pollen. Plants were self-incompatible, but self-rejection was late-acting and both self- and cross-pollen tubes penetrated ovules. Seed set of open-pollinated flowers was pollen-limited, despite pollen deposition exceeding ovule number by 6-fold. Open-pollinated seed set was similar to that of the cross + self-pollen treatment, but was less than that of the cross-pollen-only treatment.

Conclusions

Flowers of C. breviflorus are pollinated primarily by pollen-collecting bees and possess a late-acting SI system, previously unknown in this clade of the Amaryllidaceae. Pollinators of C. breviflorus deposit mixtures of cross- and self-pollen and, because SI is late-acting, self-pollen disables ovules, reducing female fertility. This study thus contributes to growing evidence that seed production in plants with late-acting SI systems is frequently limited by pollen quality, even when pollinators are abundant.  相似文献

Two apple crosses and one pear cross were carried out using three different pollen densities. In one of the apple crosses, both fruit and seed set decreased when a mixture of pollen and powder (diluted pollen treatment) was used for pollination. Also, seedlings from the undiluted treatment were more vigorous than those originating from pollination with diluted pollen. In the other apple cross, however, no effect on fruit set, seed set or growth in the first year was found. Mean diameter after the second year differed among treatments, but did not seem to be related to pollen density. For pear, the number of good seeds decreased and the number of deaf, probably unfertilized, seeds increased with decreasing pollen density. However, there was no apparent significant effect of pollen density on seedling growth. With respect to scab resistance and mildew attack in both apple crosses, no differences among treatments were found. The results indicate that gametophytic competition may occur in apple crosses, but that it is probably weaker in pear.  相似文献

Amphicarpy is a fascinating reproductive strategy, defined as fruit produced both below the soil surface and as aerial fruit on the same plant. Trifolium polymorphum is a grassland species subject to herbivory that combines amphicarpy with vegetative reproduction through stolons. Underground flowers have been described as obligate autogamous and aerial ones as self‐compatible allogamous, with aerial floral traits favouring cross‐pollination. In the present work we performed different pollination treatments on aerial flowers to analyse rates of pollen tube development and offspring fitness, measured as fruit set, seed production and germination percentage. This last variable was compared to that of seeds produced underground. No significant differences were found between fruit set in self‐ and cross‐pollinations. Seed production was higher in self‐pollinations, which is consistent with the higher rate of pollen tube development observed in self‐crosses. Spontaneous self‐pollination is limited in aerial flowers; thus pollen transfer by means of a vector is required even within the same flower. Germination tests showed that aerial seeds produced after self‐ and cross‐pollination did not differ in fitness, but underground seeds had higher germination percentage than aerial ones. Thus, we conclude that T. polymorphum has a mixed mating system. In grasslands with heavy grazing pressure, clonal propagation and underground seed production ensure persistence in the field. An intermediate level of selfing in aerial flowers ensures offspring, but morphological (herkogamy) and functional (dicogamy) floral traits maintain a window to incorporate genetic variability, allowing the species to tolerate temporal and spatial pressures.  相似文献

Nouelia insignis Franch. (Asteraceae) is a short, narrow endemic and endangered tree, growing with a natural population in the dry and hot valley of the Jinsha River in the southwest area of China. In this work, flowering phenology (time and duration), floral biology, visit frequency and behavior of pollinators, and pollination characteristics were studied based on investigation in the field and analysis in the laboratory with the help of a stereomicroscope, and the relationship between seed setting rate and reproductive traits, as well as the relationship between flowering time and rainfall before flowering, was tested using the method of general linear regression model. The results showed that natural population of N. insignis exhibited high flowering synchrony with relatively stable flowering duration, and the flowering time fluctuated greatly depending on the rainfall 5 months before flowering. The pollination of N. insignis required pollinators, and insect activities played a very important role in the pollination process. However, lack of the pollinators was not a limitation for reproductive fitness in N. insignis, although the number of pollinators was small and the frequency of visits was low. In addition, no pollen limitation was found during pollination. The average seed setting rate of N. insignis in the natural condition was only 1.52%–3.73%, and it was generally affected by changes in flowering phenology between years and had a higher seed set in early flowering year. The annual variation of seed set might be related to the annual variations of stamen and pistil functions, such as changes of pollen viability and stigma receptivity, which were closely related to flowering time. The results of this study are of value for further conservation actions on natural population of this threatened endemic plant.  相似文献

Invasive plants may threaten the reproductive success of native sympatric plants by modifying the pollination process. One potential mechanism takes place through the deposition of invasive pollen onto native stigmas when pollinators are shared among species. We explore how pollen from the invasive plant Brassica nigra influences pre- and post-fertilization stages in the native plant Phacelia parryi, through a series of hand pollination experiments. These two species share pollinators to a high degree. P. parryi flowers were hand-pollinated with either pure conspecific pollen (the control) or with B. nigra pollen applied prior to, simultaneously with, or following conspecific pollen. Application of B. nigra pollen lowered seed set, with the simultaneous application resulting in the highest reduction. Pollen tube growth was also influenced by the presence of invasive pollen, with fewer conspecific pollen tubes reaching the base of P. parryi styles in treatments where B. nigra pollen was applied prior to or simultaneously with conspecific pollen. The deleterious effects of invasive pollen on native seed set in this study are likely not due to loss of stigmatic receptivity since seed set was less affected when heterospecific pollen was applied prior to conspecific pollen, but may instead involve interactions between interspecific pollen grains on the stigma or within the style. Our study highlights the importance of timing of foreign pollen deposition on native stigmas and suggests that interspecific pollen transfer between native and exotic plants may be an important mechanism of competition for pollination in invaded plant communities.  相似文献

The pollination ecology of Gentianella uliginosa , a rare annual in The Netherlands, was studied in three relatively small coastal dune populations. In all populations, the species was fully self-compatible: manual self pollination resulted in a seed set of 92–94%. Autofertility was also high (seed set of caged, unhandled flowers 80–85%). The homogamous flowers were visited by Bombus pascuorum , which collected nectar and transported pollen sternotribically. Differences among populations in bumblebee visitation rate did not affect natural seed set. In one plot, seed set following natural pollination was lower than after manual selñng, and as high as after spontaneous selñng. As flowers here were also visited frequently, this might be due to pollen limitation through heterospecific pollen competition. Support for this hypothesis was derived from another population, where seed set after natural pollination was lower in a plot dominated by co-flowering Mentha aquatica than in a plot with few Mentha. However, differences in water availability might also have caused this. In one population, seed sets of the cross pollination and spontaneous selfing treatments were lower than those of the manual selñng and natural pollination treatments. Whether the low outcrossing success can be attributed to small differences in the treatment of the experimental plants or to outcrossing depression remains unclear. We conclude that this annual species is most likely a predominant selfer. Its high autofertility is probably an adaptation to its wet dune slack habitat, where reproductive assurance may be important to cope with unpredictable fluctuations in water table, summer droughts and pollinator service.  相似文献

In estuarine wetlands, the daily periodic tidal activity has a profound effect on plant growth and reproduction. We studied the effects of tidal action on pollination and reproductive allocation of Sagittaria graminea. Results showed that the species had very different reproductive allocation in tidal and non-tidal habitats. In the tidal area, seed production was only 9.7% of that in non-tidal habitat, however, plants produced more male flowers and nearly twice the corms compared to those in non-tidal habitat. An experiment showed that the time available for effective pollination determined the pollination rate and pollen deposition in the tidal area. A control experiment suggested that low pollen deposition from low visitation frequency is not the main cause of very low seed sets or seed production in this plant in tidal habitat. The negative effects of tides (water) on pollen germination may surpass the influence of low pollen deposition from low visitation frequency. The length of time from pollen deposition to flower being submerged by water affected pollen germination rate on stigmas; more than three hours is necessary to allow pollen germination and complete fertilization to eliminate the risk of pollen grains being washed away by tidal water.  相似文献

The extent, frequency and causes of pollenlimited seed production were examined in partially selffertile populations of Blandfordia grandiflora for 2 years. Percentage seed set of open-pollinated plants (50–57%) did not differ within or between years, and was about 19% less than experimentally cross-pollinated plants (70–75%). Floral visits by honeybees did not differ through the flowering season and the number of pollen grains deposited on stigmas within 12 h of flowers opening exceeded the number of ovules per flower, indicating that the quality rather than the quantity of pollination limited seed set. Pollen limitation was caused by concurrent self- and cross-pollination and the subsequent abortion of some selfed ovules due to inbreeding depression. Natural seed set (55%) was intermediate between selfed (43%) and crossed (75%) flowers and was not increased when flowers that had been available to pollinators for 24 h were hand cross-pollinated, suggesting that ovules were already fertilized. Similarly, experimental pollination with both cross and self pollen within 24 h of flowers opening did not increase seed set relative to natural seed set, indicating that both cross- and self-fertilizations had occurred. In contrast, when selfing followed crossing by 48 h, or vice versa, seed set did not differ from crossed-only or selfed-only flowers, respectively, indicating that ovules were pre-empted by the first pollination. Collectively, these results indicate that under natural conditions self pollen pre-empts ovules, rendering them unavailable for cross-fertilization. This selfing reduces fecundity by 50%, as estimated from the natural production of cross seeds when selfing was prevented. Consequently, selection should favour floral traits, such as increased stigma-anther separation or protandry, that reduce interference between male and female functions that leads to selfing.  相似文献

  • Breeding systems of plants determine their reliance on pollinators and ability to produce seeds following self‐pollination. Self‐sterility, where ovules that are penetrated by self‐pollen tubes that do not develop into seeds, is usually considered to represent either a system of late‐acting self‐incompatibility or strong early inbreeding depression. Importantly, it can lead to impaired female function through ovule or seed discounting when stigmas receive mixtures of self and cross pollen, unless cross pollen is able to reach the ovary ahead of self pollen (‘prepotency’). Self‐sterility associated with ovule penetration by self‐pollen tubes appears to be widespread among the Amaryllidaceae.
  • We tested for self‐sterility in three Cyrtanthus species – C. contractus, C. ventricosus and C. mackenii – by means of controlled hand‐pollination experiments. To determine the growth rates and frequency of ovule penetration by self‐ versus cross‐pollen tubes, we used fluorescence microscopy to examine flowers of C. contractus harvested 24, 48 and 72 h after pollination, in conjunction with a novel method of processing these images digitally. To test the potential for ovule discounting (loss of cross‐fertilisation opportunities when ovules are disabled by self‐pollination), we pollinated flowers of C. contractus and C. mackenii with mixtures of self‐ and cross pollen.
  • We recorded full self‐sterility for C. contractus and C. ventricosus, and partial self‐sterility for C. mackenii. In C. contractus, we found no differences in the growth rates of self‐ and cross‐pollen tubes, nor in the proportions of ovules penetrated by self‐ and cross‐pollen tubes. In this species, seed set was depressed (relative to cross‐pollinated controls) when flowers received a mixture of self and cross pollen, but this was not the case for C. mackenii.
  • These results reveal variation in breeding systems among Cyrtanthus species and highlight the potential for gender conflict in self‐sterile species in which ovules are penetrated and disabled by pollen tubes from self pollen.

Apomixis evolves from a sexual background and usually is linked to polyploidization. Pseudogamous gametophytic apomicts, which require a fertilization to initiate seed development, of various ploidy levels frequently co‐occur with their lower‐ploid sexual ancestors, but the stability of such mixed populations is affected by reproductive interferences mediated by cross‐pollination. Thereby, reproductive success of crosses depends on the difference in ploidy levels of mating partners, that is, on tolerance of deviation from the balanced ratio of maternal versus paternal genomes. Quality of pollen can further affect reproductive success in intercytotype pollinations. Cross‐fertilization, however, can be avoided by selfing which may be induced upon pollination with mixtures of self‐ and cross‐pollen (i.e., mentor effects). We tested for reproductive compatibility of naturally co‐occurring tetraploid sexuals and penta‐ to octoploid apomicts in the rosaceous species Potentilla puberula by means of controlled crosses. We estimated the role of selfing as a crossing barrier and effects of self‐ and cross‐pollen quality as well as maternal: paternal genomic ratios in the endosperm on reproductive success. Cross‐fertilization of sexuals by apomicts was not blocked by selfing, and seed set was reduced in hetero‐ compared to homoploid crosses. Thereby, seed set was negatively related to deviations from balanced parental genomic ratios in the endosperm. In contrast, seed set in the apomictic cytotypes was not reduced in hetero‐ compared to homoploid crosses. Thus, apomictic cytotypes either avoided intercytotype cross‐fertilization through selfing, tolerated intercytotype cross‐fertilizations without negative effects on reproductive success, or even benefitted from higher pollen quality in intercytotype pollinations. Our experiment provides evidence for asymmetric reproductive interference, in favor of the apomicts, with significantly reduced seed set of sexuals in cytologically mixed populations, whereas seed set in apomicts was not affected. Incompleteness of crossing barriers further indicated at least partial losses of a parental genomic endosperm balance requirement.  相似文献
