Pools of sera from patients with Graves' disease or Hashimoto's thyroiditis highly inhibit the binding to human thyroid membranes of one of 19 monoclonal antibodies raised against preparations of human thyroid membranes. This monoclonal antibody reacts with human and bovine thyroid peroxidase and bovine lactoperoxidase but not with human hemoglobin, cytochrome c and other related molecules. These results indicate that the thyroid peroxidase and the microsomal antigen are antigenically related. These data taken together with those from other groups, highly suggest that thyroid peroxidase is the microsomal antigen involved in autoimmune thyroid diseases.  相似文献

Iodothyronines induced catalatic (H2O2-decomposing) activity of thyroid peroxidase and lactoperoxidase, the effect increasing in the order of thyroxine (T4) greater than triiodothyronine (T3) greater than diiodothyronine (T2). The iodothyronines served as electron donors in the peroxidase reactions, and during the reactions the catalytic intermediate of thyroid peroxidase was confirmed to be Compound II for T4 and Compound I for T3 and T2 and from the Soret absorption spectra obtained by stopped-flow measurements. Rate constants for the reactions between T4 and Compound II, T3 and Compound I, and T2 and Compound I were estimated at 1.9 x 10(5), 1.3 x 10(6), and 7.1 x 10(5) M-1.s-1, respectively. Unlike the case of thyroid peroxidase, the catalytic intermediate of lactoperoxidase observed during the oxidation of iodothyronines was invariably Compound II. From these and other data it was concluded that thyroid peroxidase catalyzed one-electron oxidation of T4 and two-electron oxidations of T2 and T3 while lactoperoxidase catalyzed exclusively one-electron oxidation of the iodothyronines. Iodide was released during the enzymatic oxidation of iodothyronines, irrespective of the mechanism of one-electron and two-electron oxidations. The amount of released iodide increased in the order of T4 greater than T3 greater than T2. The iodothyronines-induced catalatic activity of these peroxidases was ascribable to the release of iodide, but it was also found that the iodide-enhanced catalatic activity was stimulated by iodothyronines. In this case the effect of iodothyronines was greater in the order of T2 greater than T3 greater than T4, which was consistent with the order of iodothyronine activation for the iodinium cation transfer from enzyme to acceptor.  相似文献

Thyroid peroxidase (TPO) and lactoperoxidase (LPO) display significant catalatic activity at pH 7.0 in the presence of low concentrations of iodide, based both on measurements of H2O2 disappearance and O2 evolution. In the absence of iodide only minor catalatic activity was detected. The stimulatory effect of iodide could not be explained by protection of the enzymes against inactivation by H2O2. A mechanism is suggested involving an enzyme-hypoiodite complex as an intermediate.  相似文献

Summary The fine structural localization of the endogeneous peroxidase activity in the thyroid of the young frog was studied. The reaction product for peroxidase was observed over the peripheral luminal colloid and apical region of the follicular epithelial cell. Most apical small granules and some parts of Golgi lamellae and a few Golgi vesicles were specifically stained. The cisternae of rough endoplasmic reticulum and the nuclear cisternae did not demonstrate any positive reaction for peroxidase activity with difference from that of various cells of mammalia. In this study, only mature peroxidase seems to be positive for its reaction and the enzyme in the rough endoplasmic reticulum is considered to be too immature to react for DAB method in the frog thyroid cell. The relationship between the localization of peroxidase reaction and the site of the iodination of thyroglobulin was discussed.  相似文献

In this study initially a precancerous condition, leukoplakia, was develop at 6 weeks treatment of DMBA whereas in the animals treated both DMBA + Vit. A, leukoplakia was seen at 10 weeks followed by papilloma or nodules at 12 weeks. Tumours induced by DMBA were more in number than DMBA + Vit. A treated tumours. The histological and ultrastructural changes were enhanced and prominent in DMBA treated animals at 12 weeks, where as these changes were considerably less in animals treated with DMBA + vit. Summary Chronic indirect stimulation of fast twitch rabbit muscle (tibialis anterior and extensor digitorum longus) with a frequency of 10 Hz induced a progressive transformation of the sarcoplasmic reticulum (SR). Ultrastructural changes as studied by electron microscopy of freeze-fractured vesicles consisted in a decrease of intramembranous particles of the concave (A) face and an increase of particles in the convex (B) face. The asymmetry of the membrane proved to be lowered. Changes in the particle density of theA face were mainly confined to the 7–9 nm particles. Electrophoretic analyses revealed a decrease in the 115,000-M r Ca2+ transport ATPase. The reduced density of the 7–9 nm particles correlated well with decreased activities in Ca2+-dependent ATPase as well as with decreases in initial and maximum Ca2+ uptake.  相似文献

Specimens of mullet (Mugil cephalus), a marine fish, were given single doses of 3-methylcholanthrene intraperitoneally and the activity of the microsomal mixed-function oxygenase system in the liver was measured by the metabolism of benzo(a)pyrene. The enzyme system was found to be inducible with concomitant ultrastructural changes in the hepatocytes. The specific activity and the metabolic profile approximate those of the rat.  相似文献

A sensitive fluorimetric method with guaiacol as the hydrogen donor for peroxidase activity is described. The gist of this method is measurement of the fluorescence (excitation, 300 nm; emission, 340 nm) in cyclohexane of mainly a guaiacol dimer which forms in the early phase of the color formation. Optimum conditions of the reaction were compared for horseradish peroxidase and rat thyroid peroxidase preparations. The fluorescence intensity obtained by this method using an enzyme preparation from 120 of a rat thyroid gland correlated well with the color development by the guaiacol method using the same preparation from whole glands of a rat.  相似文献

Human anti-thyroid peroxidase (TPO) autoantibodies (aAb) are generated during autoimmune thyroid diseases (AITD). Within recent years, increasing knowledge of the TPO-specific aAb repertoire, gained mainly by the use of combinatorial library methodology, has led to the cloning and sequencing of around 180 human anti-TPO aAb. Analysis of the immunoglobulin (Ig) variable (V) genes encoding the TPO aAb in the ImMunoGeneTics database (IMGT) (http://imgt.cines.fr) reveals major features of the TPO-directed aAb repertoire during AITD. Heavy chain VH domains of TPO-specific aAb from Graves' disease patients preferentially use D proximal IGHV1 genes, whereas those from Hashimoto's thyroiditis are characterized more frequently by IGHV3 genes, mainly located in the middle of the IGH locus. A large proportion of the anti-TPO heavy chain VH domains is obtained following a VDJ recombination process that uses inverted D genes. J distal IGKV1 and IGLV1 genes are predominantly used in TPO aAb. In contrast to the numerous somatic hypermutations in the TPO-specific heavy chains, there is only limited amino acid replacement in most of the TPO-specific light chains, particularly in those encoded by J proximal IGLV or IGKV genes, suggesting that a defect in receptor editing can occur during aAb generation in AITD. Among the predominant IGHV1 or IGKV1 TPO aAb, conserved somatic mutations are the hallmark of the TPO aAb repertoire. The aim of this review is to provide new insights into aAb generation against TPO, a major autoantigen involved in AITD.  相似文献

A distribution of immunoglobulin G (IgG) subclass of anti-thyroid peroxidase (TPO) autoantibodies was studied to know whether anti-TPO autoantibodies are closely implicated in the pathogenesis of human autoimmune thyroid diseases. As a result of analyzing 14 patients' sera, 7 with Graves' disease and 7 with Hashimoto's thyroiditis, anti-TPO autoantibodies were found to consist of mainly IgG1 subclass. Percentages of both IgG1 and IgG2 subclasses in IgG class of autoantibodies corresponded to those in the normal serum composition, whereas IgG3 subclass was scarcely contained in anti-TPO autoantibodies and IgG4 subclass markedly increased. It was thought that anti-TPO autoantibodies had a capability to lyse thyroid follicular cells by the mechanism of antibody-dependent complement-mediated cytolysis, because IgG1 and IgG2 subclasses of antibodies can fix complement and TPO locates in apical membrane surface of thyroid follicular cells. Comparing Graves' disease with Hashimoto's thyroiditis, mean percentages of both IgG1 and IgG2 subclasses of 2 groups were statistically different. Namely, sera of patients with Graves' disease had higher and lower mean percentages of IgG1 and IgG2 subclasses of autoantibodies, respectively, than those with Hashimoto's thyroiditis, though no plausible explanation for these differences can be offered at the present time.  相似文献

This study was designed to characterize the interaction of CH3HgCl or HgCl2 with thyroid peroxidase (TPO). Two types of experiments were performed. First, the thyroids from rats that were given 5.6 mg/kg/day of either CH3HgCl or HgCl2 for 2 weeks by intubation were subjected to histochemical treatment and then to electron microscopy. TPO activities in all cell compartments were inhibited by HgCl2 but not by CH3HgCl. Morphological observation showed that taller epithelia were induced by HgCl2, whereas flattened epithelia forming large follicles were induced by CH3HgCl. The serum thyrotropin level was substantially lowered by CH3HgCl but was unchanged by HgCl2. Second, the guaiacol oxidation by TPO in isolated and ruptured pig thyroid cells was spectrophotometrically monitored in the presence of either CH3HgCl or HgCl2. The TPO was not inhibited by CH3HgCl but was inhibited by HgCl2. These results indicated that CH3HgCl induced a hypothyroid state without affecting TPO, whereas HgCl2 inhibited TPO and induced a hypertropic state owing to compensation for loss of enzyme activity, and that the lack of inhibitory activity of CH3HgCl was not due to the inability to penetrate the cells. Therefore, there appeared to be a differential interaction of organic and inorganic forms of mercurials with the thyroid.  相似文献

Human thyroid peroxidase (TPO) has been purified from thyroid microsomes by immunoaffinity chromatography using a monoclonal antibody (mAb) to TPO. The eluted material had a specific activity of 381 U/mg and exhibited a peak in the Soret region. The ratio of A411 to A280 ranged from 0.20 to 0.25. Upon SDS-polyacrylamide gel electrophoresis, the purified enzyme gave two contiguous bands in the 100 kDa region. Further, it has been demonstrated that sera with anti-microsomal autoantibodies from patients presenting Graves' or Hashimoto's thyroiditis diseases were able to bind to purified TPO and to inhibit in a dose-dependent manner the mAb binding to purified TPO. This suggests that TPO is the thyroid antigen termed to date the microsomal antigen.  相似文献

A good correlation was obtained between the uterotrophic activity of a number of steroids and related compounds and their ability to induce peroxidase in the immature rat uterus in vivo. It is proposed that this biochemical test gives a better indication of an intact receptor-acceptor system in target cells than assays based on the binding to high affinity estrogen receptors.  相似文献

The morphology of the tight junction of rat thyroid epithelium was examined in freeze-fractured material fixed in glutaraldehyde and briefly glycerinated. In normal thyroids the overall appearance of this junctional specialization resembled that of other cell types in many respects. Short-term changes in thyroid activity and hypophysectomy for 3 wk did not obviously affect the appearance of tight junctions. Feeding of the goitrogen, thiouracil, which stimulates secretion of thyroid-stimulating hormone, resulted in the appearance of some very narrow and some very wide, tight junctions or sometimes junctions with both wide and narrow regions within the same cell.  相似文献

We investigated the possible differences between the concentrations of selenium in the whole blood and thyroid tissue of patients with thyroid disease. The study comprises 41 women with nodular goiter, 19 women and 2 men with thyroid cancer, 18 women with Graves’ disease, and 7 women with thyroiditis. The concentration of selenium was determined by the TRXRF method. The lowest mean selenium level was achieved in the whole blood of women with Graves’ disease and the highest in the whole blood of healthy people. In the thyroid cancer tissue, we found the lowest concentration of selenium and the highest in the thyroid gland of women with nodular goiter and Graves’ disease. The low selenium levles in the thyroid tissue may increase thyroid cancer risk.  相似文献

The changes of redox status during culture and their effect on organogenesis were investigated in ginseng callus cultures. Three kinds of Panax ginseng calluses (non-organogenic calluses, root-forming and bud-forming calli) 21 were obtained from each of the three examined auxins (2,4-D, IBA and NAA) at 2 mg l–1 (9.05 M 2,4-D; 9.84 M IBA; 10.74 M NAA) concentration and 0.1 mg l–1 (0.46 M) kinetin over 5 weeks. Peroxidase in the soluble fraction, which indicates oxidative status and shows a correlation with differentiation, was higher in bud-forming calluses cultured in the three types of auxins. On the other hand, reducing capacity (overall reducing capacity of glutathione, ascorbic acid, tocoferol, carotenoids and phenol) which indicates reductive status, was higher when these calluses grew on media containing 2,4-D but specially so in the case of non-organogenic calluses. With the three types of auxin used, the reducing capacity was always higher in non-organogenic calluses and lower in calluses with more organogenic capacity. It seems that organogenesis of calluses is conditioned by a high peroxidase activity in soluble fraction (equal to or more than 0.8 A430/mg protein per min) and therefore a low reducing capacity (less than 0.3 A760/g DW). On the other hand 2,4-D promotes proliferation whereas NAA promotes morphogenesis.  相似文献
