Summary Soybean (Glycine max L. Merr. cv. Amsoy 71) plants were grown in a greenhouse in a soil very low in plant-available P, and plants were harvested 5 times over a 21-week growth period. Soybeans were inoculated with one of two species of VAM fungi or received daily one of three nutrient solutions of different P concentrations (0.0, 0.2, or 1.0mMP). Until week 9, the dry weights, leaf areas and developmental stage of soybeans inoculated withG. fasciculatum orG. mosseae were similar to the 1.0 or 0.2mMP-treated plants, respectively. Phosphorus concentrations were significantly lower in VAM plants at weeks 6 and 9 as compared to non-VAM soybeans given 1.0mMP, suggesting P input in VAM plants was immediately used for new growth. Total P input for VAM plants was linear over 21 weeks, and the average rate of P uptake for these plants was 0.19mg P d−1. Estimated specific P uptake rates (SPUR) for the mycorrhizae (VAM roots) were twice that of the control (0.0mMP) roots. The calculated SPURs forG. fasciculatum andG. mosseae hyphae were 95 and 120μg P g−1 VAM d−1 respectively, a 4 to 5 fold increase over non-inoculated roots, indicating more attention must be paid to P assimilation by VAM fungi in P-fixing substrates. Summary Brassica nigra and selected species of Leucaena and Sesbania were used as indicator hosts in a greenhouse experiment designed to establish distinct categories of mycorrhizal dependence. The plants were grown in an oxisol with different concentrations of established soil solution P in the presence or absence of the vesicular-arbuscular mycorrhizal (VAM) fungus Glomus aggregatum. The extent to which the plant species depended on the fungus for dry matter production diminished with increased concentrations of soil solution P, but the magnitude of this decrease varied from species to species. Five distinct mycorrhizal categories are proposed based on the differences observed, ranging from non-dependent to very highly dependent. The critical soil solution P concentrations that were useful for separating host species into distinct VAM-dependency groups were 0.02 and 0.2 mg/l. Species differing in their mycorrhizal dependency differed with respect to the soil solution P concentration required for the expression of maximum VAM effectiveness, the degree to which increasing concentrations of P depressed VAM infection and the pattern of immobile nutrient accumulation.Contribution from the Hawaii Institute of Tropical Agriculture and Human Resources Journal Series No. 3547  相似文献

Summary Soybean plants were grown in a soil very low in available P. Seedlings were inoculated with two vesicular-arbuscular mycorrhizal (VAM) fungi or were left non-inoculated and fertilized with P. Assimilation and allocation of micronutrients (Fe, Mn, Zn, and Cu) were determined during host development, and the uptake of trace elements in VAM plants was compared to P-fertilized, non-VAM plants of similar weight, growth stage, and P status. Copper and zinc concentrations were always higher in VAM plants, while iron and manganese concentrations were lower than in the equivalent P-fertilized soybeans. Differences in the micronutrient content of fully-mature soybean pods reflected differences in the leaves and roots. Thus, for trace elements, seed quality can be altered by VAM colonization in a fashion not duplicated by P fertilizer. The residual effect of the fungicide chlorothalonil on the vesicular-arbuscular mycorrhizal (VAM) symbiosis was evaluated in a greenhouse experiment. The soil used was an oxisol (Tropeptic Eutrustox) treated with P to obtain target levels near-optimal for VAM activity or sufficient for nonmycorrhizal host growth. In the uninoculated soil treated with the former P level, the fungicide reduced VAM colonization of roots and completely suppressed symbiotic effectiveness measured in terms of pinnule P content. When this soil was inoculated with Glomus aggregatum, symbiotic effectiveness was significantly reduced but not eliminated by 50 mg of the fungicide kg−1. At higher chlorothalonil levels, VAM effectiveness but not VAM colonization was completely suppressed in the inoculated soil. The pattern with which chlorothalonil influenced tissue P content and dry matter yield at the time of harvest closely paralleled its effect on VAM effectiveness. In the soil treated with P level sufficient for nonmycorrhizal host growth, the adverse effect of the fungicide on the above variables was appreciably milder than when the host relied on VAM fungi for its P supply. The toxic effect of the fungicide, therefore, was partly offset by P fertilization, suggesting that VAM fungi were more sensitive to chlorothalonil than the host. Our results demonstrate that although the toxic effect of chlorothalonil declined as a function of time, a significant level of toxicity persisted 12.5 weeks after the chemical was applied to soil. Low-input agricultural systems that do not rely on fertilizers may be more dependent on vesicular-arbuscular mycorrhizal [VAM] fungi than conventionally managed systems. We studied populations of spores of VAM fungi, mycorrhiza formation and nutrient utilization of maize (Zea mays L.) grown in moldboard plowed, chisel-disked or no-tilled soil under conventional and low-input agricultural systems. Maize shoots and roots were collected at four growth stages. Soils under low-input management had higher VAM fungus spore populations than soils under conventional management. Spore populations and colonization of maize roots by VAM fungi were higher in no-tilled than in moldboard plowed or chisel-disked soil. The inoculum potential of soil collected in the autumn was greater for no-till and chisel-disked soils than for moldboard plowed soils and greater for low-input than conventionally farmed soil. The effects of tillage and farming system on N uptake and utilization varied with growth stage of the maize plants. The effect of farming system on P use efficiency was significant at the vegetative stages only, with higher efficiencies in plants under low-input management. The effect of tillage was consistent through all growth stages, with higher P use efficiencies in plants under moldboard plow and chisel-disk than under no-till. Plants grown in no-tilled soils had the highest shoot P concentrations throughout the experiment. This benefit of enhanced VAM fungus colonization, particularly in the low-input system in the absence of effective weed control and with likely lower soil temperatures, did not translate into enhanced growth and yield.  相似文献

Summary To investigate the effect of indigenous VAM fungi and of increasing the amount of natural inoculum barley was grown in containers buried in the field with uninoculated and inoculated irradiated soil and with uninoculated and inoculated untreated soil from two locations, one low and one high in available P. The experiment was set up with 3 P fertilizer applications (0, 15, 30 kg P/ha). Growth and uptake of P was measured. The inocula were prepared from natural VAM populations. VAM fungal infection was established in the irradiated soil at a lower level than in the untreated soil. VAM fungal infection was decreased by increasing P fertilizer application. In the soil low in available P VAM increased concentration of P and total uptake of P. VAM did not cause an increased growth. The reason for this may be the low establishment of VAM in the irradiated soil and/or because the indigenous VAM species were not efficient. It is also possible that a pronounced growth increase due to irradiation the soil may have masked a smaller effect of the indigenous VAM fungi. Increasing the amount of natural inoculum in the untreated soil influenced neither VAM frequency nor growth.  相似文献

Cassava (Manihot esculenta Crantz) was grown in the greenhouse and in the field at different levels of phosphorus applied, with or without inoculation with VA mycorrhiza in sterilized or unsterilized soil. When grown in a sterilized soil to which eight levels of P had been applied the non-inoculated plants required the application of 3200 kg P ha−1 to reach near-maximum yield of plant dry matter (DM) at 3 months. Inoculated plants, however, showed only a minor response to applied P. Mycorrhizal inoculation in the P check increased top growth over 80 fold and total P uptake over 100 fold. Relating dry matter produced to the available P concentration in the soil (Bray II), a critical level of 15 ppm P was obtained for mycorrhizal and 190 ppm P for non-mycorrhizal plants. This indicates that the determination of critical levels of P in the soil is highly dependent on the degree of mycorrhizal infection of the root system. In a second greenhouse trial with two sterilized and non-sterilized soils it was found that in both sterilized soils, inoculation was most effective at intermediate levels of applied P resulting in a 15–30 fold increase in DM at 100 kg P ha−1. In the unsterilized soil inoculation had no significant effect in the quilichao soil, but increased DM over 3 fold in the Carimagua soil, indicating that the latter had a native mycorrhizal population less effective than the former. When cassava was grown in the field in plots with 11 levels of P applied, uninoculated plants grown in sterilized soil remained extremely P deficient for 4–5 months after which they recuperated through mycorrhizal infection from unsterilized borders or subsoil. Still, after 11 months inoculation had increased root yields by 40%. In the non-sterilized soil inoculation had no significant effect as the introduced strain was equally as effective as the native mycorrhizal population. These trials indicate that cassava is extremely dependent on an effective mycorrhizal association for normal growth in low-P soils, but that in most natural soils this association is rapidly established and inoculation of cassava in the field can only be effective in soils with a low quantity and quality of native mycorrhiza. To test the hypothesis that high levels of soluble phosphate applied in combination with VAM fungi, to citrus plants, can cause growth depression even in the absence of other limiting factors, and also to test if rock phosphate, under these conditions, may be a satisfactory P source, a greenhouse experiment was conducted using sterilized soil with four levels of phosphate (0, 50, 100 and 200 ppm P) supplied either as soluble P or as rock phosphate. Citrus seedlings were either inoculated with the vesicular-arbuscular mycorrhizal (VAM) fungus Glomus etunicatum or left uninoculated. Six months after the start of the experiment, the plants were harvested and shoot dry weight, P and K uptake, root colonization and the number of spores in 50 cm3 of soil were determined. Significant increases were found in dry matter yields and in P and K contents, due to VAM fungus inoculation, at the zero and 50 ppm soluble P levels and at all rock phosphate levels. At 100 ppm soluble P, the development of VAM plants was equilvalent to that of non-VAM plants, and at 200 ppm, growth was significantly less than that of non-VAM plants. Root colonization and sporulation were reduced at higher P levels. The absolute growth depression of VAM plants at the higher P level was likely due to P toxicity. In addition, high leaf P and K concentrations may have interfered with carbohydrate distribution and utilization in these symbioses. In a greenhouse experiment involving an acid soil teff [Eragrostis tef (Zucc.) Trotter] plants failed to grow unless the soil was limed or inoculated with either of two vesicular-arbuscular-mycorrhizal (VAM) fungi,Glomus mosseae orGlomus macrocarpum. Plant growth increased by liming and to a lesser extent by VAM fungal inoculation. Liming also enhanced root colonization by VAM fungi. Five tropical soils were either not inoculated or inoculated with the vesicular-arbuscular mycorrhizal (VAM) fungus Glomus aggregatum. The degree to which VAM effectiveness was expressed in the soils was evaluated prior and after solution P status was adjusted for optimal VAM activity. VAM effectiveness determined by monitoring P concentrations of pinnules of Leucaena leucocephala leaves as a function of time and as dry matter yield determined at the time of harvest, indicated that in three of the soils VAM effectiveness was either very restricted or altogether unexpressed irrespective of vesicular-arbuscular mycorrhizal fungal (VAMF) inoculation if soil solution P was not optimized for VAM effectiveness. After P optimization, effectiveness was significantly increased by VAMF inoculation although in four of the soils, densities of indigenous VAMF propagules greatly exceeded that attained by the inoculum after it was mixed with soil. Mycorrhizal fungal inoculation effects varied from soil to soil, depending on the extent to which the effectiveness of indigenous and introduced endophytes was enhanced by P optimization and the similarity of inherent soil solution P concentrations to the range known to be optimum for VAM effectiveness. In a pot experiment, wheat was grown for 50 days in two heat-sterilized low-phosphorus (P) soils supplied with organic P as Na-phytate. Seed inoculation with the phosphatase-producing fungus (PPF) Aspergillus fumigatus or soil inoculation with the vesicular-arbuscular mycorrhizal (VAM) fungus Glomus mosseae increased shoot and root dry weight and root length, phosphatase activity in the rhizosphere and shoot concentrations of P and to a lesser extent of K and Mg. As a rule, the greatest effects on those parameters were most in the combined inoculation treatment (PPF + VAM). Shoot concentrations of Cu and Zn were only enhanced by VAM, not by PPF. At harvest, depletion of organic P in the rhizosphere soil increased in the order of: sterilized soil < PPF < VAM < PPF + VAM which corresponded with the enhanced P concentrations in the plants. The results demonstrate that organic P in form of Na-Phytate is efficiently used by VAM and that use of organic P can be increased by simultaneous inoculation with phosphatase-producing fungi.  相似文献

Maize (Zea mays L. cv. Alize) plants were grown in a calcareous soil in pots divided by 30-m nylon nets into three compartments, the central one for root growth and the outer ones for hyphal growth. Sterle soil was inoculated with either (1) rhizosphere microorganisms other than vesicular-arbuscular mycorrhizal (VAM) fungi, (2) rhizosphere microorganisms together with a VAM fungus [Glomus mosseae (Nicol. and Gerd.) Gerdemann and Trappel], or (3) with a gamma-irradiated inoculum as control. Plants were grown under controlled-climate conditions and harvested after 3 or 6 weeks. VAM plants had higher shootroot ratios than non-VAM plants. After 6 weeks, the concentrations of P, Zn and Cu in roots and shoots had significantly increased with VAM colonization, whereas Mn concentrations had significantly decreased. Root exudates were collected on agar sheets placed on the interface between root and hyphal compartments. Six-week-old VAM and non-VAM plants had similar root exudate compositions of 72–73% reducing sugars, 17–18% phenolics, 7% organic acids and 3% amino acids. In another experiment in which root exudates were collected on agar sheets with or without antibiotics, the amounts of amino acids and carbohydrates recovered were similar in VAM and non-VAM plants. However, threeto sixfold higher amounts of carbohydrates, amino acids and phenolics were recovered when antibiotics were added to the agar sheets. Thus, the high microbial activity in the rhizosphere and on the rhizoplane limits the exudates recovered from roots.  相似文献

Summary Field inoculation trials with cassava (Manihot esculenta Crantz) were conducted in Quilichao (typic Dystropept soil) and Carimagua (Haplustox soil). In Quilichao, with a large and effective native VA-mycorrhizal (VAM) population, inoculation withGlomus manihotis did not increase cassava yields significantly, neither when different sources and levels of inoculum material were used, nor with different cassava cultivars, or after stabilizing soil temperature through mulching. Field inoculation did result in a decrease of the coefficient of variation with respect to yield. The high dependency of cassava on an effective VAM association was indicated by a marked decrease in yield after eradication of native VAM by soil sterilization. In Carimagua, with a lower native VAM population, mycorrhizal inoculation withG. manihotis increased yields significantly at intermediate levels of 100 kg/ha of applied P, using either inoculum of cassava orPanicum maximum roots or inoculum of a soil-root mixture of maize or tropical kudzu. Higher or lower levels of P decreased the effect of inoculation on yield. There were no significant differences among P sources, ranging from highly soluble triple superphosphate to low solubility rock phosphates. Inoculation with different VAM isolates had a variable effect on cassava yields, and showed that there may be an interaction between P fertilizer level and isolate efficiency. It is concluded that there may be a potential to increase yields or decrease the fertilizer P requirements of cassava through field inoculation with effective VAM isolates, in the vast areas of acid infertile Oxisols and Ultisols with low native VAM fungal populations, represented by Carimagua.  相似文献

Summary The effects of vesicular-arbuscular mycorrhiza (VAM) on the growth and phosphorus uptake of cocoa seedlings (Theobroma cacao L.) grown for 100 days in polythene bags, were studied at five levels of phosphorus fertilization in both steamed and unsterile Bungor Series soil (a fine clayey, kaolinitic isohyperthermic Typic Paleudult). The cocoa seedlings responded well to phosphorus fertilization and mycorrhizal treatments. Plants inoculated with VAM fungi (Gigaspora spp.) gave the most vigorous growth and higher phosphorus in the leaf tissues in unsterile soil compared to plants grown in steamed soil. However, the mycorrhizal effect was significantly more pronounced (P<0.01) in plants grown in steamed than in unsterile soil. High levels of phosphorus application depressed mycorrhizal development. Phosphorus fertilizer applied at the rates of 250 and 500 ug g−1 soil gave maximum root colonization and spore counts in both soil types used.  相似文献

Nitrogen (N) transfer from N-fixing legumes via vesicular-arbuscular mycorrhizal (VAM) fungi to associated non-fixing plants has been demonstrated in greenhouse experiments. To date, this transfer has been shown only where mineral N is applied shortly before harvest, and hence is readily available. We have yet to demonstrate VAM-mediated N transfer where soil-N is limiting, a condition under which most traditional legume-nonlegume intercrops are grown.In this study, 15N-enriched soil (with 0.28%N) was used to distinguish between the uptake of soil- and atmospherically-derived N in maize grown with beans in the presence or absence of VAM fungi. VAM infection did not result in transfer of fixed N or soil N from bean to maize, despite a VAM-stimulated increase in N fixation in bean. In fact, beans were more competitive for soil N when mycorrhizal. N content in beans increased by 75% with a concomitant 22% decrease in mg N per maize plant. The competitive effect may have resulted from a VAM-mediated shift in carbon allocation in beans (but not maize) from shoots to roots.  相似文献

Summary Rooted cuttings ofSeverinia buxifolia were inoculated with the vesicular-arbuscular mycorrhizal (VAM) fungusGlomus intraradices or provided an inoculum filtrate (non-VAM plants) and grown in one of seven media combinations of fired montmorillonite clay (FMC) and Canadian peatmoss (CP) at ratios of 100%, 80%, 67%, 50%, 33%, 20%, or 0% FMC. Mycorrhizal infection increased with higher proportions of FMC, but the growth of both VAM and non-VAM plants was reduced with increased FMC amendment. The growth benefit (top and root fresh-dry weights) conferred by mycorrhizal infection was greater at higher levels of FMC in the media. Improved phosphorus uptake by inoculated severinia plants appeared at least partially responsible for increased growth compared to non-VAM plants under conditions of high soluble salts and pH associated with high FMC composition. Nutrient acquisition and growth of citronella Java (Cymbopogon winterianus Jowitt) was studied in a P-deficient sandy soil to determine the effects of mycorrhizal symbiosis and soil compaction. A pasteurized sandy loam soil was inoculated either with rhizosphere microorganisms excluding VAM fungi (non-mycorrhizal) or with the VAM fungus, Glomus intraradices Schenck and Smith (mycorrhizal) and supplied with 0, 50 or 100 mg P kg-1 soil. The soil was compacted to a bulk density of 1.2 and 1.4 Mg m-3 (dry soil basis). G. intraradices substantially increased root and shoot biomass, root length, nutrient (P, Zn and Cu) uptake per unit root length and nutrient concentrations in the plant, compared to inoculation with rhizosphere microorganisms when the soil was at the low bulk density and not amended with P. Little or no plant response to the VAM fungus was observed when the soil was supplied with 50 or 100 mg P kg-1 soil and/or compacted to the highest bulk density. At higher soil compaction and P supply the VAM fungus significantly reduced root length. Non-mycorrhizal plants at higher soil compaction produced relatively thinner roots and had higher concentrations and uptake of P, Zn and Cu than at lower soil compaction, particularly under conditions of P deficiency. The quality of citronella Java oil measured in terms citronellal and d-citronellol concentration did not vary appreciably due to various soil treatments.  相似文献

Summary Lucerne plants were inoculated with the vesicular-arbuscular mycorrhizal (VAM) fungus,Glomus caledonius andRhizobium meliloti and grown in pots in voliére. Treatments were 0,1 and 2 mg P added to 100 g of a soil with low P-fertility. Plants were harvested after 6, 9, 13 and 18 weeks, allowing shoots to regrow between harvests. VAM-infection was determined after 6 and 18 weeks, dry weight, concentration and uptake of P and concentrations of N, K, Ca, Mg, Cu and Zn were determined at all harvests. VAM-inoculation increased growth of shoots, P-concentration and P-uptake at all soil-P levels and the increase was most pronounced at the lowest soil-P level. P-fertilization also increased growth, P-concentration and P-uptake. VAM-inoculation showed tendency to increase N-and K-uptake and decreased Ca-, Mg- and Zn-concentrations in shoots. The application of herbicides has induced symptoms of nutrient deficiencies under some circumstances. This glasshouse study examined the effect of chlorsulfuron on the uptake and utilization of copper (Cu) in four cultivars of wheat plants (Triticum aestivum L. cvs. Kulin, Cranbrook, Gamenya and Bodallin) on a Cu-responsive soil. Application of chlorsulfuron depressed the concentration of Cu in wheat plants receiving either inadequate or adequate Cu. In plants with inadequate Cu supply, chlorsulfuron increased the severity of Cu deficiency. Shoot weight was markedly decreased by chlorsulfuron at all levels of Cu, through decreasing the number of tillers and the elongation of leaves. This decreased growth of shoots occurred prior to the effect on Cu concentration in tissues. The retranslocation of Cu in old tissues over time was unaffected by chlorsulfuron. In all wheat cultivars, the decreased growth of shoots were correlated with the concentration of Cu in the youngest fully emerged leaf blade with critical levels of 1.6−1.7 at day 25 and 0.9−1.0 μg g−1 d. wt. at day 60. The application of chlorsulfuron tended to increase the critical level at day 25 but not at day 60. In addition, Kulin seems to be most, and Cranbrook least, sensitive to chlorsulfuron. This sensitivity was associated with the sensitivity of the cultivars to Cu deficiency. It is suggested that chlorsulfuron application induces Cu deficiency in wheat plants mainly due to effects on the uptake of Cu. Summary A field experiment was carried out to study the influence of vesicular-arbuscular mycorrhiza (VAM) on nutrient uptake and growth of pea at two levels of P fertilizer. Pea was grown in dazomet-fumigated and non-fumigated soil, either with or without addition of inoculum of VAM fungi. Dazomet efficiently eliminated infection by indigenous VAM fungi, whereas roots from untreated soil were extensively infected. Inoculation with VAM fungi increased P uptake by peas in fumigated plots, but had no significant effect on dry matter production in either soil treatment. However, VAM developed less in inoculated fumigated than in untreated plots. Seeds of uninfected plants in fumigated plots contained only 56 and 81%, respectively, of the P and dry matter content of seeds of infected plants in untreated plots. The addition of 60 kg P ha−1 also increased seed yields but to a smaller degree than VAM. The seed yield as a proportion of total shoot yield (harvest index) was higher in VAM infected than in uninfected peas but was unaffected by P fertilizer.
