The photosynthetic response of the purple sulfur bacterium Chromatium vinosum DSM 185 to different degrees of illumination was analyzed. The microorganism was grown in continuous culture, and samples were taken from the effluent of the culture and incubated at different irradiances to determine the specific rate of sulfur oxidation as a measure of the photosynthetic activity of the organism. The activities obtained were plotted as a function of the specific rate of light uptake, and for each set of data a photosynthesis equation was fitted, which allowed the estimation of Pmax (photosynthetic capacity), qk (the threshold irradiance for light limitation), and m (maintenance coefficient). The results indicated that cells grown under light limitation are able to achieve higher photosynthetic activities than cells grown under light saturation. The photosynthetic capacity (Pmax) remained constant under all the conditions of illumination tested, while the maintenance expenses (m) were higher under light limitation. The effects of iron limitation on photoacclimation to dynamic irradiance were studied in Phaeocystis antarctica G. Karst. and Fragilariopsis cylindrus (Grunow) W. Krieg. in terms of growth rate, photosynthetic parameters, pigment composition, and fluorescence characteristics. Under dynamic light conditions mimicking vertical mixing below the euphotic zone, P. antarctica displayed higher growth rates than F. cylindrus both under iron (Fe)–replete and Fe‐limiting conditions. Both species showed xanthophyll de‐epoxidation that was accompanied by low levels of nonphotochemical quenching (NPQ) during the irradiance maximum of the light cycle. The potential for NPQ at light levels corresponding to full sunlight was substantial in both species and increased under Fe limitation in F. cylindrus. Although the decline in Fv/Fm under Fe limitation was similar in both species, the accompanying decrease in the maximum rate of photosynthesis and growth rate was much stronger in F. cylindrus. Analysis of the electron transport rates through PSII and on to carbon (C) fixation revealed a large potential for photoprotective cyclic electron transport (CET) in F. cylindrus, particularly under Fe limitation. Probably, CET aided the photoprotection in F. cylindrus, but it also reduced photosynthetic efficiency at higher light intensities. P. antarctica, on the other hand, was able to efficiently use electrons flowing through PSII for C fixation at all light levels, particularly under Fe limitation. Thus, Fe limitation enhanced the photophysiological differences between P. antarctica and diatoms, supporting field observations where P. antarctica is found to dominate deeply mixed water columns, whereas diatoms dominate shallower mixed layers.  相似文献

Plantago lanceolata is a gynodioecious species: In natural populations male steriles (MS) coexist with hermaphrodites (H). Since male steriles have a reproductive disadvantage, without any compensation for their loss in male function by an increase in female function, they are expected to disappear from the population. In this study we investigated the possibility that differences in ecologically important photosynthetic characteristics, between MS and H lines of P. lanceolata. play a role in maintaining gynodioecy. One MS line and two H lines were grown under conditions of high N and light availability, as well as under either N limitation or light limitation, to investigate whether the sex types respond differently to environmental constraints. Photosynthetic light-response and CO2-response curves were made, together with leaf organic N and chlorophyll determinations. There were only few small differences between the lines and since the MS line did not differ in any of the determined photosynthetic characteristics from either H line, it is unlikely that these differences are involved in maintaining male sterility in populations of P. lanceolata. The low-light-grown plants showed a high degree of acclimation as shown by a two-fold higher leaf area to leaf weight ratio (SLA), a two-fold higher investment of N in light harvesting, and higher net photosynthetic rates under low-light conditions, as compared to the high-light-grown plants. The low-N-grown plants used their organic N more efficiently in photosynthesis compared to plants grown at an optimal N supply. This was mainly due to the N-limited plants having leaves with a lower organic N content and thus lower photosynthetic capacities. To a lesser extent it was due to the higher value for the curvature factor of the light-response curves of the N-limited plants, to their decreased rates of photorespiration and possibly to their relatively higher allocation of organic N to photosynthetic functions.  相似文献

Purple photosynthetic bacteria produce H2 from organic compounds by an anaerobic light-dependent electron transfer process in which nitrogenase functions as the terminal catalyst. It has been established that the H2-evolving function of nitrogenase is inhibited by N2 and ammonium salts, and is maximally expressed in cells growing photoheterotrophically with certain amino acids as sources of nitrogen. In the present studies with Rhodopseudomonas capsulata, nutritional factors affecting the rate and magnitude of H2 photoproduction in cultures growing with amino acid nitrogen sources were examined. The highest H2 yields and rates of formation were observed with the organic acids: lactate, pyruvate, malate, and succinate in media containing glutamate as the N source; under optimal conditions with excess lactate, H2 was produced at rates of ca. 130 ml/h per g(dry weight) of cells. Hydrogen production is significantly influenced by the N/C ratio in the growth substrates; when this ratio exceeds a critical value, free ammonia appears in the medium and H2 is not evolved. In the "standard" lactate + glutamate system, both H2 production and growth are "saturated" at a light intesity of ca. 600 ft-c (6,500 lux). Evolution of H2, however, occurs during growth at lithe intensities as low as 50 to 100 ft-c (540 to 1,080 lux), i.e., under conditions of energy limitation. In circumstances in which energy conversion rate and supplies of reducing power exceed the capacity of the biosynthetic machinery, energy-dependent H2 production presumably represents a regulatory device that facilitates "energy-idling." It appears that even when light intensity (energy) is limiting, a significant fraction of the available reducing power and adenosine 5'-triphosphate is diverted to nitrogenase, resulting in H2 formation and a bioenergetic burden to the cell.  相似文献

1. Three common species of freshwater phytoplankton, the diatom Nitzschia sp., green alga Sphaerocystis schroeteri and cyanobacterium Phormidium luridum, were grown under contrasting daylengths [18 : 6 h light : dark cycles (LD) versus 6 : 18 h LD] and phosphorus (P) regimes (P‐sufficient versus 1 μm P). The rates of growth and photosynthesis, as well as growth efficiencies and pigment concentrations, were compared among treatments. 2. The growth and photosynthetic parameters of the three species depended on both P status and daylength in a species‐specific way. The responses to P limitation depended on daylength and, conversely, the responses to daylength depended on P status. 3. Growth rates and the maximum rates of photosynthesis (Pmax) of all species decreased under P limitation under both light regimes. However, the decrease of Pmax because of P limitation was greater under long daylength. The Pmax of the green alga S. schroeteri decreased the most (ca. sixfold) under P limitation compared with the other two species. The photosynthesis saturation parameter Ik also decreased under P limitation; the decline was significant in Nitzschia and Sphaerocystis. P‐limitation significantly increased photoinhibition (β) in Nitzschia and Sphaerocystis, but not in Phormidium. The excess photochemical capacity (the ratio of the maximum photosynthesis rate to the photosynthesis rate at the growth irradiance), characterising the ability to utilise fluctuating light, was significantly lower under P limitation. 4. The growth efficiency (growth rate normalised to daylength) declined with increasing daylength in all species. Under short daylength the cyanobacterium Phormidium had the lowest growth efficiency of the three species. 5. The cellular chlorophyll a concentration in both Nitzschia and Sphaerocystis was significantly higher under short daylength, but only under P‐sufficient conditions. In Nitzschia, under short daylength, P‐limitation significantly decreased cellular chlorophyll concentration. In contrast, P‐limitation increased cellular chlorophyll concentration in Sphaerocystis, but under long daylength only. The ratio of chlorophyll a to b in the green alga also declined under short daylength and under P‐limited conditions.  相似文献

The amounts of extracellular dissolved organic phosphorus (DOP) produced by 8 species of marine planktonic algae under various conditions of light, salinity, and nutrition were compared. Large amounts, more than 20% of the total P in the system, were excreted by Cyclotella cryptica, Thalassiosira fluviatilis, Dunaliella tertiolecta, and Synechococcus under 1 or more of the experimental conditions. Excretion of DOP was proportional to light intensity in Dunaliella, Rhodomonas, Chlorella sp., and Coccolithus huxleyi. Phosphorus limitation reduced DOP production by Cyclotella and Thalassiosira, nitrogen limitation reduced DOP production by Phaeodactylum, Dunaliella, and Rhodomonas, and lack of iron reduced DOP levels in Cyclotella cultures. Salinity affected growth, but no clear relationship to DOP excretion was evident. The DOP eliminated during growth was reassimilated by the species that produced it and by other species, but lack of alkaline phosphatase reduced the amount of DOP that was available to certain algae.  相似文献

Nitrogen fixation as well as structural and functional properties of the photosynthetic apparatus were studied with phototrophically grown chemostat cultures of Rhodobacter capsulatus strain 37b4. Illumination was varied between 3,000 and 30,000 lx at a constant dilution rate of D=0.075 h-1. Steady state parameters of growth revealed two forms of limitation, i.e. energy limitation in the range of 3,000 to about 10,000 lx and nitrogen limitation at higher illuminations. Over the entire range of illumination, the specific bacteriochlorophyll content and the amount of total bacteriochlorophyll per photochemical reaction center remained essentially constant. Photophosphorylation activity remained constant up to 20,000 lx but was slightly increased at 30,000 lx. Hydrogen evolution and acetylene reduction activities of cellular nitrogenase were assayed under saturating light conditions with samples taken from cultures growing under steady state conditions. In spite of the apparent constancy of the composition and activity of the photosynthetic apparatus under energy limitation, maximal specific acetylene reduction and hydrogen evolution activities increased by factors of 3 and 8, respectively, when illumination of the culture was raised from 3,000 to about 15,000 lx. Above 15,000 lx, both activities of nitrogenase approached constancy.We, therefore, conclude that neither under energy limitation nor under nitrogen limitation the function of nitrogenase depended on the photosynthetic activities. Moreover, it is suggested that light did not influence nitrogenase activity under conditions of nitrogen limitation, while under conditions of energy limitation light seemed to influence nitrogenase activities indirectly via glutamate consumption of the cells.  相似文献

Phaeodactylum tricomutum Bohlin (strain TFX-1) was grown under light-, nitrogen-, and phosphorus-limited conditions in continuous or semicontinuous cultures under a 12L-12D light regime. The C, N, and P contents were determined at each steady state, as was the partitioning of cellular organic carbon into protein, lipids, polysaccharides, and metabolic intermediates. All determinations were made at the beginning and again at the end of the light period. The rates of nutrient assimilation and of synthesis of biochemical constituents during the light and dark periods were calculated from the above data, and the periodicities of these processes characterized. The elemental composition of the cells was different under each limitation. In particular, phosphorus limitation severely restricted the ability of the cell to store nitrogen in non-protein forms. Biochemical composition and the diel periodicity of cellular processes also differed between limitations. Nutrient uptake was most strongly periodic under light limitation. Protein synthesis showed increased periodicity under nitrogen limitation, relative to the other limitations, while the periodicity of lipid synthesis was reduced under phosphate limitation. Polysaccharide was synthesized at high rates during the light period and consumed in the dark under all limitations.  相似文献

A natural assemblage of microalgae from a facultative lagoon system treating municipal wastewater was enriched for growth in the effluents of an anaerobic digester processing dairy waste. A green microalga with close resemblance to Chlorella sp. was found to be dominant after multiple cycles of sub‐culturing. Subsequently, the strain (designated as LLAI) was isolated and cultivated in 20× diluted digester effluents under various incident light intensities (255–1,100 µmoles m?2 s?1) to systematically assess growth and nutrient utilization. Our results showed that LLAI production increased with increasing incident light and a maximum productivity of 0.34 g L?1 d?1 was attained when the incident irradiance was 1,100 µmoles m?2 s?1. Lack of growth in the absence of light indicated that the cultures did not grow heterotrophically on the organic compounds present in the medium. However, the cultures were able to uptake organic N and P under phototrophic conditions and our calculations suggest that the carbon associated with these organic nutrients contributed significantly to the production of biomass. Overall, under high light conditions, LLAI cultures utilized half of the soluble organic nitrogen and >90% of the ammonium, orthophosphate, and dissolved organic phosphorus present in the diluted waste. Strain LLAI was also found to accumulate triacylglycerides (TAG) even before the onset of nutrient limitation and a lipid productivity of 37 mg‐TAG L?1 d?1 was measured in cultures incubated at an incident irradiance of 1,100 µmoles m?2 s?1. The results of this study suggest that microalgae isolates from natural environments are well‐suited for nutrient remediation and biomass production from wastewater containing diverse inorganic and organic nutrient species. © 2016 American Institute of Chemical Engineers Biotechnol. A defined medium (MAM) simulating acid mine drainage waters was developed which supported reproducible growth rates of three axenic strains of Euglena mutabilis Schmitz. Growth responses to various pHs and carbon sources were examined under defined culture conditions. A lab strain and two 5eld isolates, tested over pH range 1.5-9.0, grew best under acidic conditions (pH < 5.5) with highest growth rates at pH 3-4. Photoauxotrophic growth rates of all strains at pH 3 were improved significantly over unstirred batch controls by bubbling with air and even more by enrichment with 5% CO2 in air. These results confirmed inorganic carbon limitation in batch culture. Organic carbon substrates were tested as possible carbon supplements in batch culture at pH 3. None of the strains survived in the dark on any of the twenty organic sources added. In the light, the lab strain exhibited some photoheterotrophic growth potential on glucose, sucrose, ethanol, and amino acids but growth was inhibited by acetate. Field strains showed little or no growth improvement with any organic substrate addition. Under simultaneous enrichment with acetate and 5% CO2 acetate continued to be inhibitory. Simultaneous enrichment with glucose and 5% CO2 gave higher yields of the lab strain than with CO2 alone but did not enhance growth of the field strain. We conclude that E. mutabilis is an acidophilic photoauxotroph which appears unable to use organic carbon supplements for growth even under conditions of carbon limitation.  相似文献

This study deals with combining the biologi cal removal of organic halogens with the removal of nitrogen from bleached kraft pulp mill wastewater in fluidized-bed reactors under nitrifying and denitrifying conditions. Untreated and biotreated bleached kraft pulp mill wastewaters had no detrimental effect on nitrification or denitrification. The nitrifying biofilm reactor, pregrown on synthetic inorganic feed with ammonia, removed without a lag phase adsorbable organic halogens [7.2 mg Cl (g biomass volatile solids)−1day−1] from bleached kraft pulp mill wastewater and selected chlorophenols from synthetic wastewater. Electron microscopical examination of the biofilm showed that bacteria, morphologically similar to the nitrifying species Nitrosomonas or Nitrobacter, and Nitrosospira were dominant. The denitrifying fluidized-bed reactor, pregrown on nitrate and methanol, denitrified without a lag phase bleached kraft pulp mill wastewater. Under denitrifying conditions, 35% of the total organic carbon content of untreated bleached kraft pulp mill waste water was removed. The reducing power delivered by untreated bleached kraft pulp mill wastewater for denitrification was 2 mmol electrons/mmol carbon mineralized. In natural herbaceous vegetation plants are exposed to a vertical light gradient. In experiments, however, morphogenetic responses of stoloniferous plants to shade have nearly always been tested under homogeneous shade conditions. In this study we simulated a vertical light gradient and found that the response of Hydrocotyle vulgaris in this gradient differed considerably from the responses to homogenous shade. Petioles grew longer while at the same time the specific weight of petioles increased. The elongated petioles raised leaf-blades into better-lit places resulting in higher biomass. Though leaves in the light gradient started their growth under low-light conditions, the size of the leaf-blade was the same as in high light. Internodes were longer than in homogeneous shade conditions but specific weight decreased, probably due to increased allocation to the fast-growing petioles. Phototrophic growth of Rhodobacter capsulatus (formerly Rhodopseudomonas capsulata) under anaerobic conditions with either butyrate or propionate as carbonsource was dependent on the presence of either CO2 or an auxiliary oxidant. NO - 3 , N2O, trimethylamine-N-oxide (TMAO) or dimethylsulphoxide (DMSO) were effective provided the appropriate anaerobic respiratory pathway was present. NO - 3 was reduced extensively to NO - 3 , TMAO to trimethylamine and DMSO to dimethylsulphide under these conditions. Analysis of culture fluids by nuclear magnetic resonance showed that two moles of TMAO or DMSO were reduced per mole of butyrate utilized and one mole of either oxidant was reduced per mole of propionate consumed. The growth rate of Rb. capsulatus on succinate or malate as carbon source was enhanced by TMAO in cultures at low light intensity but not at high light intensities. A new function for anaerobic respiration during photosynthesis is proposed: it permits reducing equivalents from reduced substrates to pass to auxiliary oxidants present in the medium. The use of CO2 or auxiliary oxidants under phototrophic conditions may be influence by the availability of energy from light. The yeast Yarrowia lipolytica is one of the most intensively studied “non-conventional” yeast species. Its ability to secrete various organic acids, like pyruvic (PA), citric, isocitric, and alpha-ketoglutaric (KGA) acid, in large amounts is of interest for biotechnological applications. We have studied the effect of the alpha-ketoglutarate dehydrogenase (KGDH) complex on the production process of KGA. Being well studied in Saccharomyces cerevisiae this enzyme complex consists of three subunits: alpha-ketoglutarate dehydrogenase, dihydrolipoyl transsuccinylase, and lipoamide dehydrogenase. Here we report the effect of overexpression of these subunits encoding genes and resulting increase of specific KGDH activity on organic acid production under several conditions of growth limitation and an excess of carbon source in Y. lipolytica. The constructed strain containing multiple copies of all three KGDH genes showed a reduced production of KGA and an elevated production of PA under conditions of KGA production. However, an increased activity of the KGDH complex had no influence on organic acid production under citric acid production conditions.  相似文献

A two-alternative, forced-choice visual duration discrimination task was used to examine the effect of an intermittent, 50 Hz, 100 μT magnetic field on accuracy at two different times of the day. A total of 59 female and 40 male subjects with an age range of 18 to 46 years were studied under both field-exposed and sham-exposed conditions. The subject's task was to decide which of two sequentially presented light flashes had the longer duration, percentage correct being the measure of performance. The data were gathered under double-blind conditions with sham and real exposure counterbalanced. Exposure to the magnetic field produced a small improvement in accuracy but only at the most difficult level of the task, with female subjects showing a larger improvement than males. The time of day at which the study was run had no effect on performance. Despite the relatively large number of subjects used and a relaxed alpha level (P = .3), the statistical power of the test to detect the observed effect was still only 0.71. We analyzed experimentally the relative contribution of phytoplankton and periphyton in two shallow lakes from the Pampa Plain (Argentina) that represent opposite scenarios according to the alternative states hypothesis for shallow lakes: a clear lake with submerged macrophytes, and a turbid lake with high phytoplankton biomass. To study the temporal changes of both microalgal communities under such contrasting conditions, we placed enclosures in the littoral zone of each lake, including natural phytoplankton and artificial substrata, half previously colonized by periphyton until a mature stage and half clean to analyze periphyton colonization. In the clear vegetated shallow lake, periphyton chlorophyll a concentrations were 3–6 times higher than those of the phytoplankton community. In contrast, phytoplankton chlorophyll a concentrations were 76–1,325 times higher than those of periphyton in the turbid lake. Here, under light limitation conditions, the colonization of the periphyton was significantly lower than in the clear lake. Our results indicate that in turbid shallow lakes, the light limitation caused by phytoplankton determines a low periphyton biomass dominated by heterotrophic components. In clear vegetated shallow lakes, where nitrogen limitation probably occurs, periphyton may develop higher biomass, most likely due to their higher efficiency in nutrient recycling.  相似文献

Ascorbate is oxidized into the radical monodehydroascorbate (MDHA) through ascorbate oxidase or peroxidase activity or non‐enzymatically by reactive oxygen species. Regeneration of ascorbate from MDHA is ensured by the enzyme MDHA reductase (MDHAR). Previous work has shown that growth processes and yield can be altered by modifying the activity of enzymes that recycle ascorbate; therefore, we have studied similar processes in cherry tomato (Solanum lycopersium L.) under‐ or overexpressing MDHAR. Physiological and metabolic characterization of these lines was carried out under different light conditions or by manipulating the source–sink ratio. Independently of the light regime, slower early growth of all organs was observed in MDHAR silenced lines, decreasing final fruit yield. Photosynthesis was altered as was the accumulation of hexoses and sucrose in a light‐dependent manner in plantlets. Sucrose accumulation was also repressed in young fruits and final yield of MDHAR silenced lines showed a stronger decrease under carbon limitation, and the phenotype was partially restored by reducing fruit load. Ascorbate and MDHA appear to be involved in control of growth and sugar metabolism in cherry tomato and the associated enzymes could be potential targets for yield improvement.  相似文献

Selenastrum minutum (Naeg.) Collins was grown over a wide range of growth rates under phosphate or nitrate limitation with non-limiting nutrients added to great excess. This resulted in saturated luxury consumption. The relationships between growth rate and cell quota for the limiting nutrients were well described by the Droop relationship. The observed variability in N cell quota under N limitation as reflected in kQ·Qmax?1*, was similar in magnitude to previously reported values but kQ·Qmax?1* for P under P limitation was greater than previously reported for other species. These results were evaluated in light of the optimum ratio hypothesis. Our findings support previous work suggesting that the use of a single optimum ratio (kQi·KQj?1) is inappropriate for dealing with a species growing under steady-state nutrient limitation. Under these conditions the optimum ratio should be viewed as a growth rate dependent variable. Two approaches for testing the growth rate dependency of optimum ratios are proposed. The capacity for luxury consumption differed between nutrients and was growth rate dependent. At low growth rates, the coefficient of luxury consumption (Rsat) for P was ca. four times that for N. The set of all possible relationships between N and P cell quota under these conditions was reported and these values were then used to establish the cellular N:P niche boundaries for S. minutum. Cell quotas of non-limiting nutrients were not described by the Droop equation. Analysis showed that as the cellular N:P ratio deviates from the optimum ratio, the ability of the Droop equation to describe the relationship between growth rate and non-limiting cell quotas decreases. When non-limiting nutrient cell quotas are saturated, the Droop equation appears to be invalid. Previously reported patterns of non-limiting nutrient utilization are summarized in support of this conclusion. The physiological and ecological consequences of luxury consumption and growth rate dependent optimum ratios are considered.  相似文献

Summary Kraft pine and straw lignins were fractionated into aqueous soluble and organic soluble-ether insoluble parts. Chemical analysis, UV characteristics, and gel permeation chromatograms of crude and fractionated lignins were studied. Using pure and mixed, N-limited and non N-limited standing cultures of several fungal species, the biodegradability of curde and fractionated lignins was compared. Straw lignins, especially the aqueous fraction were degraded by most of the fungi studied. Except for Sporotrichum pulverulentum, nitrogen limitation did not seem to favour degradation. The best fungi for degradation under conditions of N-limitation were S. pulverulentum, Humicola fuscoatra, and Aspergillus wentii, under sufficient nitrogen: A. wentii, Chaetomium cellulolyticum and H. fuscoatra. The greatest percentage degradation, 55%, was obtained with S. pulverulentum under nitrogen limited conditions from 1 gl–1 organic soluble-ether insoluble kraft lignin. Gel chromatography showed that the degradation was over the complete molecular size range.  相似文献

The biochemical basis for variations in the critical nitrogen‐to‐phosphorus (N:P) ratio, which defines the transition between N‐ and P‐limitation of growth rate, is currently not well understood. To assess this issue, we cultured the cryptophyte Rhinomonas reticulata NOVARINO in chemostats with inflow nitrate‐to‐phosphate ratios ranging from 5 to 60 mol N·(mol P)?1 at two light intensities. The nitrate‐to‐phosphate ratio marking the transition between N‐ and P‐limitation was independent of light intensity and was between 30 and 45 mol N/mol P. In N‐limited cells, the particulate N:P ratio was stable at around 23 mol N/mol P over a range of inflow nitrate‐to‐phosphate from 5 to 30, whereas in P‐limited cells this ratio was around 90 mol N/mol P at inflow nitrate‐to‐phosphate ratios of 45 and 60. Cell phosphorus decreased with increasing nitrate‐to‐phosphate ratio up to the critical nitrate‐to‐phosphate ratio for each light intensity, above which they remained stable. The C:P of R. reticulata cells increased with increasing inflow nitrate‐to‐phosphate from around the Redfield value (106 mol C/mol P) to around 700. There was a significant effect of light on C:P in the N‐ limited cells, with higher C:P under high light conditions that was not observed in the P‐limited chemostats. Cellular RNA was not influenced by light but was greatly influenced by the type of nutrient limitation. In contrast, chl a, C, N, and protein were not influenced by the nitrate‐to‐phosphate in the inflow medium. Total protein per RNA was independent of light intensity but exhibited a maximum at inflow nitrate‐to‐phosphate of 30. Our results suggest a strong “two‐level” homeostatic mechanism of cellular N and P content in R. reticulata with two distinct states that are determined by the type of nutrient limitation and not by light.  相似文献
