Telomeres are short tandem repeated sequences of DNA found at the ends of eukaryotic chromosomes that function in stabilizing chromosomal end integrity. In vivo studies of somatic tissue of mammals and birds have shown a correlation between telomere length and organismal age within species, and correlations between telomere shortening rate and lifespan among species. This result presents the tantalizing possibility that telomere length could be used to provide much needed information on age, ageing and survival in natural populations where longitudinal studies are lacking. Here we review methods available for measuring telomere length and discuss the potential uses and limitations of telomeres as age and ageing estimators in the fields of vertebrate ecology, evolution and conservation. 相似文献

A Sex-linked Microsatellite Locus Isolated from the Y Chromosome of Lake Charr, Salvelinus Namaycush

A small-insert library was constructed after microdissection of the short arm of the Y chromosome of lake charr, Salvelinus namaycush. Clones from this library were sequenced and two dinucleotide CA-repeat microsatellite sequences were recovered. Oligonucleotide primers for one locus were designed and optimized for amplification by PCR. This locus (designated Yp136) was tested for sex linkage in twelve lake charr families and found to be a distance of 37 centimorgans (cM) from the sex-determining locus. This microsatellite locus was also examined in three informative, gynogenetic/diploid, lake charr crosses for linkage to the centromere on the X chromosome. Results from these families show that this locus is 19cM from the centromere. The combination of linkage data and microdissection information places the sex-determining locus near the telomere of the Y chromosome in lake charr. This is consistent with studies of several other fish species including some salmonids that place the sex locus near the telomere. 相似文献

Telomere dynamics link molecular and cellular mechanisms with organismal processes and therefore may explain variation in a number of important life-history traits. Telomere length has been used to estimate age in free-living populations of animals. Such estimation is a potentially powerful tool in the context of population dynamics and management, as well as the study of life-history trade-offs. The number of studies utilizing telomere restriction fragment assays in the fields of ecology and evolution is steadily growing. However, the field lacks methodological and analytical standardization resulting in considerable variation in telomere length and therefore in the usefulness of these techniques. Here, we illustrate new laboratory and analytical methods to reliably measure telomere length from blood erythrocytes and accurately assess the relationship between telomeres and age. We demonstrate the importance of analysing those telomeres most relevant to age-related studies: the shortest telomeres. We present a reliable method to quickly identify an analysis window (the telomere optimal estimate, TOE) which approaches the optimal window for age estimation. Because the TOE focuses on the shortest telomeres - those telomeres which signal cellular senescence and ageing - TOE can also be used to compare telomeres in age-matched individuals. We also compare constant- and pulsed-field gel electrophoresis to show how each can influence telomere measurement. The use of TOE should provide powerful telomere-based age estimation and enable organismal biologists to readily uncover individual and longitudinal differences with regard to telomere dynamics. 相似文献

Telomere length and dynamics are increasingly scrutinized as ultimate determinants of performance, including age-dependent mortality and fecundity. Few studies have investigated longevity in relation to telomere length (TL) in the wild and none has analysed longevity in relation to TL soon after hatching, despite the fact that telomere shortening may mostly occur early in life. We show that TL in nestling barn swallows (Hirundo rustica) in the wild does not predict longevity. However, TL positively covaries with body size, suggesting that individuals with large TL can afford to grow larger without paying the cost of reduced TL, and/or that benign rearing conditions ensure both large body size and low rates of telomere shortening. Overall, our study hints at a role of TL in developmental processes, but also indicates a need for further analyses to assess the expectation that TL in young individuals predicts longevity in the wild. 相似文献

Telomere length (TL) limits somatic cell replication. However, the shortest among the telomeres in each nucleus, not mean TL, is thought to induce replicative senescence. Researchers have relied on Southern blotting (SB), and techniques calibrated by SB, for precise measurements of TL in epidemiological studies. However, SB provides little information on the shortest telomeres among the 92 telomeres in the nucleus of human somatic cells. Therefore, little is known about the accumulation of short telomeres with age, or whether it limits the human lifespan. To fill this knowledge void, we used the Telomere-Shortest-Length-Assay (TeSLA), a method that tallies and measures single telomeres of all chromosomes. We charted the age-dependent buildup of short telomeres (<3 kb) in human hematopoietic cells from 334 individuals (birth-89 years) from the general population, and 18 patients with dyskeratosis congenita-telomere biology disorders (DC/TBDs), whose hematopoietic cells have presumably reached or are close to their replicative limit. For comparison, we also measured TL with SB. We found that in hematopoietic cells, the buildup of short telomeres occurs in parallel with the shortening with age of mean TL. However, the proportion of short telomeres was lower in octogenarians from the general population than in patients with DC/TBDs. At any age, mean TL was longer and the proportion of short telomeres lower in females than in males. We conclude that though converging to the TL-mediated replicative limit, hematopoietic cell telomeres are unlikely to reach this limit during the lifespan of most contemporary humans. 相似文献

Telomeres are dynamic DNA-protein structures that form protective caps at the ends of eukaryotic chromosomes. Although initial telomere length is partly genetically determined, subsequent accelerated telomere shortening has been linked to elevated levels of oxidative stress. Recent studies show that short telomere length alone is insufficient to induce cellular senescence; advanced attrition of these repetitive DNA sequences does, however, reflect ageing processes. Furthermore, telomeres vary widely in length between individuals of the same age, suggesting that individuals differ in their exposure or response to telomere-shortening stress factors. Here, we show that residual telomere length predicts fitness components in two phylogenetically distant bird species: longevity in sand martins, Riparia riparia, and lifetime reproductive success in dunlins, Calidris alpina. Our results therefore imply that individuals with longer than expected telomeres for their age are of higher quality. 相似文献

Estradiol has been shown to increase the level of thioredoxin mRNA in the uterus of the ovariectomized (ovx) rat. In this study the influence of progesterone, androgens, the anti-estrogen ICI 182780 and the anti-androgen Flutamid on thioredoxin expression, has been studied in the rat uterus. Thioredoxin mRNA concentrations were determined by solution hybridization. Ovx rats treated with progesterone alone showed no effect on thioredoxin expression. Combined treatment of ICI 182780 and estradiol attenuated the estradiol-induced increase in thioredoxin mRNA. When ovx rats were treated with a testosterone depot, the amount of thioredoxin mRNA was increased five-fold after 48 h and remained at that level during the rest of the 168 h monitored. A similar increase in thioredoxin mRNA could be seen after 5-dihydrotestosterone treatment, indicating a true androgenic effect. In addition, the anti-androgen Flutamid attenuated the thioredoxin mRNA increase seen after 5-dihydrotestosterone treatment alone.
It is concluded that thioredoxin mRNA is regulated by growth promoting gonadal steroids in the rat uterus. The attenuation of the estrogen and androgen-induced increases of the thioredoxin mRNA with ICI 182780 and Flutamid, indicate that the effect is mediated via the estrogen receptor and androgen receptor respectively. None of these hormones affected the hepatic thioredoxin mRNA level in the same animals. 相似文献

Leukocyte telomere length (LTL) is ostensibly a bio-indicator of human aging. Here we report that African Americans have longer LTL than whites. We studied cross-sectionally 2453 individuals from the National Heart, Lung, and Blood Institute (NHLBI) Family Heart Study (age = 30–93 years) and the Bogalusa Heart Study (age = 19–37 years), comprising 1742 whites and 711 African Americans. We measured LTL by Southern blots of the terminal restriction fragments length. In 234 participants, telomere repeats were also measured by quantitative polymerase chain reaction (qPCR). Adjusted for age and body mass index (BMI), the respective leukocyte telomere lengths (mean ± SEM) were considerably longer in African Americans than in whites both in the Family Heart Study (7.004 ± 0.033 kb vs. 6.735 ± 0.024 kb, p < 0.0001) and the Bogalusa Heart Study (7.923 ± 0.063 kb vs. 7.296 ± 0.039 kb, p < 0.0001). We confirmed the racial effect on LTL by qPCR (3.038 ± 0.565 T/S units for African Americans vs. 2.714 ± 0.487 T/S units for whites, p < 0.001). Cross-sectionally, sex- and BMI-adjusted LTL became shorter with age (range 19–93 years) at a steeper slope in African Americans than in whites (0.029 kb year−1 vs. 0.020 kb year−1 , respectively, p = 0.0001). We suggest that racial difference in LTL arises from a host of interacting biological factors, including replication rates of hematopoietic stem cells. 相似文献

Parasites are a strong selective force that can influence fitness‐related traits. The length of chromosome‐capping telomeres can be used to assess the long‐term costs of parasitism, as telomere loss accelerates in response to environmental stressors and often precedes poorer survival prospects. Here, we explored the sex‐specific effects of ectoparasite removal on morphology and telomere length in nestling tree swallows (Tachycineta bicolor). To do so, we experimentally removed blow fly (Protocalliphora spp.) larvae from nests using Permethrin, a broad‐spectrum insecticide. Compared to water‐treated controls, insecticide treatment of nests had a sex‐biased effect on blood telomere length: ectoparasite removal resulted in significantly longer telomeres in males but not females. While this treatment did not influence nestling body mass, it was associated with reduced feather development regardless of sex. This may reflect a relaxed pressure to fledge quickly in the absence of parasites, or alternatively, could be a negative side effect of permethrin on morphology. Exploring robust sex‐specific telomere dynamics in response to early‐life environmental pressures such as parasitism will shed light on sexual dimorphism in adult life histories and aging. 相似文献

Absence of association with age and/or sex in the case of discrete traits was hypothesized by Berry & Berry (1967). The important implication that skeletal material too fragmentary for sex and age determination may also be used in discrete-trait-studies stimulated us to test this hypothesis for 49 discrete traits in 254 Amsterdam crania of known age and sex. The results were compared with those of other such studies of European skulls. No significant deviation from the hypothesis was found as far as age dependence is concerned while sex difference did occur for some traits. At the same time Corruccini's (1974) conclusion that the hypothesis fails when tested in the Terry collection was found to result largely from the fact that right and left manifestations of lateral traits were treated as being independent. The possibility of including skeletal material too fragmentary for sex and age determination in future discrete-trait-studies is still open for discussion. 相似文献

Sexual patterns of teleosts are extremely diverse and include both gonochorism and hermaphroditism. As a protogynous hermaphroditic fish, all orange-spotted groupers (Epinephelus coioides) develop directly into females, and some individuals change sex to become functional males later in life. This study investigated gonadal restructuring, shifts in sex hormone levels and gene profiles of cultured mature female groupers during the first (main) breeding season of 2019 in Huizhou, China (22° 42′ 02.6″ N, 114° 32′ 10.1″ E). Analysis of gonadal restructuring revealed that females with pre-vitellogenic ovaries underwent vitellogenesis, spawning and regression and then returned to the pre-vitellogenic stage in the late breeding season, at which point some changed sex to become males via the intersex gonad stage. A significant decrease in the level of serum 17β-estradiol (E2) was observed during ovary regression but not during sex change, whereas serum 11-ketotestosterone (11-KT) concentrations increased significantly during sex change with the highest concentration in newly developed males. Consistent with serum hormone changes, a significant decrease in cyp19a1a expression was observed during ovary regression but not during sex change, whereas the expression of cyp11c1 and hsd11b2 increased significantly during sex change. Interestingly, hsd11b2 but not cyp11c1 was significantly upregulated from the pre-vitellogenic ovary stage to the early intersex gonad stage. These results suggest that a decrease in serum E2 concentration and downregulation of cyp19a1a expression are not necessary to trigger the female-to-male transformation, whereas increased 11-KT concentration and upregulation of hsd11b2 expression may be key events for the initiation of sex change in the orange-spotted grouper. 相似文献

The interrelationship between sex skin swelling and the urinary excretion of luteinizing hormone and sex steroids was investigated during ovulatory menstrual cycles in adult female chimpanzees. Estrone was excreted in two peaks, one during the preovulatory and the other during the midluteal phase. Maximum swelling of the sex skin was attained several days before the preovulatory estrone peak. The LH surge preceded or accompanied beginning detumescence of the sex skin which, in turn, was closely correlated with a rising excretion of pregnanediol. Urinary measurements provide integrated estimates of the concentration of fluctuating hormones and, in addition, are safer and easier to make than blood measurements in this species. 相似文献

Visitors at two zoos were observed as they approached the reptile exhibits at the respective institutions. Both age and sex differences were discovered among several variables designed to measure avoidance behavior. At most ages, females more often refused to enter the exhibits than males. Females also spent less time within the exhibits than males. Musculoskeletal markers are frequently used to reconstruct past lifestyles and activity patterns. Yet the reliability of muscle marker measurements has been called into question because they may be confounded by body size. In this study, an aggregate muscle marker variable was calculated using 20 insertion sites (14 femoral, 6 tibial), and I examined their effects on lower limb size (as a proxy for body size), age, and sex. Analyses were made of a sample of 77 (57 males, 20 females) Native British Columbians (3,500-1,500 years BP) and 18th century Quebec prisoners. Muscle markers were measured using two-point observer rating scales; size was measured by standard methods; and age and sex were determined through pelvic, cranial, and dental morphology. Lower limb muscle markers correlated with: age, r=0.61; lower limb size, r=0.52; and sex, r=0.49; P <0.001. Older individuals had higher muscle marker scores, as did larger individuals and males. Based on partial correlations and regression analyses, age was the best overall predictor of lower limb muscle markers. 相似文献
