Formalisms are developed for calculating the absorption wavelengths, dipole strengths and rotational strengths for dimers of bacteriochlorophyll and related molecules. The expressions explicitly consider the mixing of bacteriochlorophyll's four main excited states (Qy, Qx, Bx and By) in the ground and excited states of the dimer. This mixing must be considered in order to account for the hyperchromism and nonconservative circular dichroism found experimentally in oligomers of bacteriochlorophyll and bacteriopheophytin. The spectroscopic properties of the eight absorption bands of a bacteriopheophytin dimer are calculated as functions of the geometry of the dimer. The importance of the mixing of nondegenerate excited states, and of the mixing of doubly-excited states into the dimer's ground state, is evaluated by comparisons with calculations in which these phenomena are neglected. Structures for bacteriopheophytin dimers are found for which most of the calculated spectroscopic properties are consistent with the properties seen experimentally. Possible explanations are considered for the remaining discrepancies between the calculated and observed properties.  相似文献

Undoped and Er-doped NaY(WO4)2 disordered single crystals have been grown by the Czochralski technique. The specific heat and thermal conductivity (κ) of these crystals have been characterized from T = 4 K to 700 K and 360 K, respectively. It is shown that κ exhibits anisotropy characteristic of single crystals as well as a κ(T) behavior observed in glasses, with a saturation mean free phonon path of 3.6 Å and 4.5 Å for propagation along a and c crystal axes, respectively. The relative energy positions and irreducible representations of Stark Er3+ levels up to 4G7/2 multiplet have been determined by the combination of experimental low (<10 K) temperature optical absorption and photoluminescence measurements and simulations with a single-electron Hamiltonian including both free-ion and crystal field interactions. Absorption, emission and gain cross sections of the 4I13/24I15/2 laser related transition have been determined at 77 K. The 4I13/2 Er3+ lifetime (τ) was measured in the temperature range of 77–300 K, and was found to change from τ (77K) ≈ 4.5 ms to τ (300K) ≈ 3.5 ms. Laser operation is demonstrated at 77 K and 300 K by resonantly pumping the 4I13/2 multiplet at λ≈1500 nm with a broadband (FWHM≈20 nm) diode laser source perfectly matching the 77 K crystal 4I15/24I13/2 absorption profile. At 77 K as much as 5.5 W of output power were obtained in π-polarized configuration with a slope efficiency versus absorbed pump power of 57%, the free running laser wavelength in air was λ≈1611 nm with the laser output bandwidth of 3.5 nm. The laser emission was tunable over 30.7 nm, from 1590.7 nm to 1621.4 nm, for the same π-polarized configuration.  相似文献

A photochemical reaction-center preparation has been made from a second bacteriochlorophyll b-containing organism, Thiocapsa pfennigii. The reaction-center unit is thought to be composed of one P-960, four bacteriochlorophyll, two bacteriopheophytin, one carotenoid molecules and polypeptides of Mr 40000, 37000, 34000, 27000 and 26000 probably plus quinones and metal atoms. The preparation also contains a low-potential cytochrome c-555 and a high-potential cytochrome c-557 bound to the reaction center in a 3–4:2–3:1 molar ratio with respect to P-960. The 40 kDa subunit is associated with the cytochromes, while the 37, 34 and 27 + 26 kDa subunits are proposed to be equivalent to the H, M and L polypeptides of bacteriochlorophyll a-containing reaction centers. The cytochromes are oxidized by P-960+. The three near-infrared absorption bands at 788, 840 and 968 nm are assigned to bacteriopheophytin, bacteriochlorophyll and the primary donor (P-960), respectively. The 778 nm peak resolves into two at 77 K; no further resolution of the other two peaks occurs. Illumination of the sodium dithionite-reduced reaction centers at 77 K by 960 nm-light results in P-960, transferring one electron from cytochrome c-555 mainly to a bacteriopheophytin molecule, absorbing at 781 nm. A similar treatment at room temperatures reduces most of the two bacteriopheophytin molecules. It is argued that both bacteriopheophytin molecules, possibly with some contribution from bacteriochlorophyll, form an intermediary electron-carrier complex between P-960 and a quinone in T. pfennigii. We could not substantiate that a bacteriochlorophyll molecule precedes the bacteriopheophytins in the electron transfer sequence. Although the biochemical characteristics of the reaction center are very similar to those of the other known bacterioclorophyll b-containing reaction center, that from Rhodopseudomonas viridis, their spectral characteristics are not. This has helped elucidate more about the function of each spectral form and led us to conclude that the 850 nm form in Rps. viridis is not the higher energy transition of the special pair of bacteriochlorophyll molecules forming P-960. Laser-flash-in-duced absorbance changes in T. pfennigii reaction-center preparation should now lead to a more complete understanding of the mechanism of the primary photochemical event.  相似文献

The primary processes of the photochemical cycle of light-adapted bacteriorhodopsin (BR) were studied by various experimental techniques with a time resolution of 5 × 10-13 s. The following results were obtained. (a) After optical excitation the first excited singlet state S1 of bacteriorhodopsin is observed via its fluorescence and absorption properties. The population of the excited singlet state decays with a lifetime τ1 of ~0.7 ps (430 ± 50 fs) (52). (b) With the same time constant the first ground-state intermediate J builds up. Its absorption spectrum is red-shifted relative to the spectrum of BR by ~30 nm. (c) The second photoproduct K, which appears with a time constant of τ2 = 5 ps shows a red-shift of 20 nm, relative to the peak of BR. Its absorption remains constant for the observation time of 300 ps. (d) Upon suspending bacteriorhodopsin in D2O and deuterating the retinal Schiff base at its nitrogen (lysine 216), the same photoproducts J and K are observed. The relaxation time constants τ1 and τ2 remain unchanged upon deuteration within the experimental accuracy of 20%.  相似文献

To the revolution-ellipsoidal and spherical membranous shell (cell mitochondrion) are introduced the equations for the calculation of both the modulus of elasticity (Young's modulus) and the stresses, which exist at the membrane. The existing pressure difference between the inner and outer surface of the membrane is calculated in the dilution of seawater media in the osmotic steady state. The experimental results are obtained by using egg cells of the sea urchin, Strongylocentrotus purpuratus. Up to the specific volume of the egg cell (VE ≈ 35·10-8 cm3) Boyle-van't Hoff's law is valid (defined as the subelastic range) beyond that the elastic stresses exist (elastic range). For the maximum value of the stresses existing at the cell wall one obtains σ ≈ 5.5·106 dyne/cm2 and for the modulus of elasticity E = 1.0·107 dyne/cm2, which is constant when the value of relative strain εν > 15%. The breaking limit by an approximate calculation is σU ≈ 11·106 dyne/cm2. The membrane is assumed to be convoluted and its hypothetical degree of folding was calculated [unk]a = 34%. The results are compared with the values existing in the literature and other types of cells are found to have values of elasticity in the same range as values of the membrane of S. purpuratus. Both compression and cell elastometer methods are criticized and in certain cases results of these methods are considered to belong to the subelastic domain.  相似文献

A new bacteriochlorophyll from brown-colored chlorobiaceae   总被引:4,自引:0,他引:4  
A new bacteriochlorophyll has been isolated by thin layer chromatography from all strains of the brown-colored Chlorobiaceae Chlorobium phaeobacteroides and Chlorobium phaeovibrioides. The new bacteriochlorophyll e —like the bacteriochlorophylls c and d—represents the major amount of bacteriochlorophyll in the cells in addition to small amounts of bacteriochlorophyll a. Bacteriochlorophyll e can be differentiated from the bacteriochlorophylls c and d by its absorption maxima in aceton and its different R f -value in the thin layer chromatogram. The structure of the new bacteriochlorophyll e has been elucidated on the basis of mass spectra, 1H- and 13C-NMR-spectra, the UV/VIS-spectrum as well as IR-, ORD-, and CD-spectra. The new bacteriochlorophyll has the same relationship to bacteriochlorophyll c as chlorophyll b from green plants to chlorophyll a; therefore, bacteriochlorophyll e represents the first formyl-substituted chlorophyll from bacteria. The tetrapyrrole electron donors and acceptors (bacteriochlorophyll, BCh; bacteriopheophytin, BPh) within the bacterial photosynthetic reaction center (RC) are arranged with a specific geometry that permits rapid (picosecond time scale) electron tunneling to occur between them. Here we have measured the angle between the molecular planes of the bacteriochlorophyll dimer (primary donor), B2, and the acceptor bacteriopheophytin, H, by analyzing the dichroism of the absorption change associated with H reduction, formed by photoselection with RCs of Rhodopseudomonas viridis. This angle between molecular planes is found to be 60° ± 2. This means that the ultrafast electron tunneling must occur between donors and acceptors that are fixed by the protein to have a noncoplanar alignment. Nearly perpendicular alignments have been determined for other electron tunneling complexes involving RCs. These geometries can be contrasted with models proposed for heme-heme electron transfer complexes, which have emphasized that mutually parallel orientations should permit the most kinetically facile transfers.  相似文献

Bacteriochlorophyll c in vivo is a mixture of at least 5 homologs, all of which form aggregates in CH2Cl2, CHCl3 and CCl4. Three homologs exist mainly in the 2-R-(1-hydroxyethyl) configuration, whereas the other two homologs, 4-isobutyl-5-ethyl and 4-isobutyl-5-methyl farnesyl bacteriochlorophyll c, exist mainly in the 2-S-(1-hydroxyethyl) configuration (Smith KM, Craig GW, Kehres LA and Pfennig N (1983) J. Chromatograph. 281: 209–223). In CCl4 the S-homologs form an aggregate of 2–3 molecules whose absorption (747 nm maximum) and circular dichroism spectra resemble those of the chlorosome. In CH2Cl2, CHCl3 and CCl4 the 4-n-propyl homolog (R-configuration) forms dimers absorbing at ca. 680 nm and higher aggregates absorbing at 705–710 nm. In CCl4 the dimerization constant is approx. 10 µM–1 (1000 times that for chlorophyll a). Pigment analysis was performed by means of normal phase HPLC on a number of bacteriochlorophyll a and b containing species of purple bacteria that contain a core antenna only. At least 99% of the bacteriochlorophyll in Rhodobacter sphaeroides R26, Rhodopseudomonas viridis and Thiocapsa pfennigii was esterified with phytol (BChl a p and BChl b p, respectively). Rhodospirillum rubrum contained only BChl a esterified with geranyl-geraniol (BChl a GG). Rhodospirillum sodomense and Rhodopseudomonas marina contained, in addition to BChl a p, small amounts of BChl a GG, and presumably also of BChl a esterified with dihydro and tetrahydro geranyl-geraniol (2,10,14-phytatrienol and probably 2,14-phytadienol). In all species bacteriopheophytin (BPhe) esterified with phytol was present. The BChl/BPhe ratio indicated that in these species a constant number of 25 ± 3 antenna BChls is present per reaction centre. This number supports a model in which the core antenna consists of 12 - heterodimers surrounding the reaction centre. Determination of the in vivo extinction coefficient of BChl in the core-reaction centre complex yielded a value of ca. 140 mM–1 cm–1 for BChl a containing species and of 130 mM–1 cm–1 for Rhodopseudomonas viridis.Abbreviations BChl bacteriochlorophyll - BPhe bacteriopheophytin - GG geranyl-geraniol - LHI and LHII core and peripheral antenna complexes - P phytol - RC reaction centre Dedicated to the memory of Professor D.I. The reaction of mixed valence state cytochrome oxidase (CuA2+a3+ · CuB+a32+) with O2 at 173 K has been investigated in purified potato mitochondria by low temperature flash photolysis and rad wavelength scanning optical spectrometry in the visible region. The kinetics of the reaction have been analyzed simultaneously at six wavelength pairs (586-630, 590-630, 594-630, 604-630, 607-630, and 610-630 nanometers) by nonlinear optimization techniques, and found to proceed by a two-species sequential mechanism. The “pure” difference spectra of the two species, IM and IIM, relative to unliganded mixed valence state cytochrome oxidase have been obtained. The difference spectrum of species IM is characterized by a peak at 591 nanometers, with a shoulder at 584 nanometers and a trough at 602 nanometers, and that of species IIM by an α band split into a prominent peak at 607 nanometers and a small side peak at 594 nanometers. Evidence is presented to suggest that these two bands arise from O2 → CuB2+ and O2a32+ charge transfer transitions which would imply that O2 forms a bridging ligand between CuB and the iron atom of cytochrome a3 in species IIM. The kinetics of the reaction and the spectral characteristics of species IM and IIM obtained with the potato mitochondrial system are compared and contrasted with data in the literature on the beef heart mitochondrial system.  相似文献

Decay-associated fluorescence spectra of the green alga Scenedesmus obliquus have been measured by single-photon timing with picosecond resolution in various states of light adaptation. The data have been analyzed by applying a global data analysis procedure. The amplitudes of the decay-associated spectra allow a determination of the relative antenna sizes of the photosystems. We arrive at the following conclusions: (a) The fluorescence kinetics of algal cells with open PS II centers (F0 level) have to be described by a sum of three exponential components. These decay components are attributed to photosystem (PS) I (τ ≈ 85 ps, λmaxem ≈ 695-700 nm), open PS II α-centers (τ ≈ 300 ps, λmaxem = 685 nm), and open PS II β-centers (τ ≈ 600 ps, λmaxem = 685 nm). A fourth component of very low amplitude (τ ≈ 2.2-2.3 ns, λmaxem = 685 nm) derives from dead chlorophyll. (b) At the Fmax level of fluorescence there are also three decay components. They originate from PS I with properties identical to those at the F0 level, from closed PS II α-centers (τ ≈ 2.2 ns, λmaxem = 685 nm) and from closed PS β-centers (τ ≈ 1.2 ns, λmaxem = 685 nm). (c) The major effect of light-induced state transitions on the fluorescence kinetics involves a change in the relative antenna size of α- and β-units brought about by the reversible migration of light-harvesting complexes between α-centers and β-centers. (d) A transition to state II does not measurably increase the direct absorption cross-section (antenna size) of PS I. Our data can be rationalized in terms of a model of the antenna organization that relates the effects of state transitions and light-harvesting complex phosphorylation with the concepts of PS II α,β-heterogeneity. We discuss why our results are in disagreement with those of a recent lifetime study of Chlorella by M. Hodges and I. Moya (1986, Biochim. Biophys. Acta., 849:193-202).  相似文献

1. The absorption spectra of deutero- and proto-ferrihaem in aqueous solution at 25°C show marked changes with concentration and pH in the Soret band region. Quantitative studies of these phenomena imply that they are associated with ferrihaem dimerization and with protolytic equilibria involving monomeric (M) and dimeric (D) ferrihaem species according to the scheme: [Formula: see text] 2. For deuteroferrihaem we obtain K=1.9×10−2, pKa(M)=7.1, pKa(D)=7.4. Protoferrihaem has a much higher dimerization constant, K=4.5 and pKa(D)=7.5 (pKa(M) is not accessible). 3. Possible structural relationships between monomeric and dimeric ferrihaem species in solution are discussed in relation to recent work on the oxo-bridged nature of crystalline ferrihaem dimers.  相似文献

Fluorescence Detected Magnetic Resonance (FDMR) spectra have been measured for whole cells and isolated chlorosomal fractions for the green photosyntheic bacteria Chlorobium phaeobacteroides (containing bacteriochlorophyll e, and isorenieratene as major carotenoid) and Chlorobium limicola (containing bacteriochlorophyll c, and chlorobactene as major carotenoid). The observed transition at 237 MHz (identical in both bacteria) and > 1100 MHz can be assigned, by analogy with published data on other carotenoids, to the 2E and D + E transitions, respectively, of Chlorobium carotenoids. Their zero field splitting (ZFS) parameters are estimated to be: |D|=0.0332 cm–1 and |E|=0.0039 cm–1 (chlorobactene), and |D|=0.0355 cm–1 and |E|=0.0039 cm–1 (isorenieratene). In the intermediate frequency range 300–1000 MHz the observed transitions can be assigned to chlorosomal bacteriochlorophylls c and e, and to bacteriochlorophyll a located in the chlorosome envelope and water-soluble protein. We have measured the extent of flash-induced electron transfer from the bacteriochlorophyll dimer, P, to the bacteriopheophytin in the M-subunit, HM, in reaction centers of Rhodopseudomonas viridis. This has been done by measuring the transient states produced by excitation of reaction centers trapped in the PHL HM state at 90 K. Under these conditions the normal forward electron transfer to the bacteriopheophytin in the L-subunit, HL, is blocked and the yield of transient P+HM can be estimated with respect to the lifetime of P*. Under these conditions flash induced absorbance decreases of the bacteriochlorophyll dimer 990 nm band suggest that a transient P+ state is formed with a quantum yield of 0.09±0.06 compared to that formed during normal photochemistry. These transient measurements provide an upper limited on the yield of a transient P+ HM state. An estimate of 0.09 as the yield of the P+ HM state is consistent with all current observations. This estimate and the lifetime of P* suggest that the electron transfer rate from P* to HM, kM, is about 5 × 109 sec–1 (M = 200ps). These measurements suggest that the a branching ratio kL/kM is on the order of 200. The large value of the branching ratio is remarkable in view of the structural symmetry of the reaction center. This measurement should be useful for electron transfer calculations based upon the reaction center structure.  相似文献

Air dried films of purple membranes (PM) from Halobacterium halobium containing the photochromic protein bacteriorhodopsin (BR) were prepared and the BR-photocycle of this material analyzed. The absorption maxima of the initial state Bmax = 570 nm) and the photochemical intermediate Mmax = 412 nm), which is the longest living intermediate in suspension (τ ≈ 10 ms), were spectrally well separated. Light-induced population gratings between B and M were used for reversible holographic recording in these dry PM films. The resolution (>5,000 lines/mm) of PM films was comparable to the corresponding values of conventional photochromic recording materials. The longterm stability toward photochemical degradation of PM films is excellent (> 100.000 recording cycles). The spectral bandwidth (400-680 nm) of such films covers nearly the whole visible spectrum. Both the photochemical transition from BM with wavelengths in the green-red range and from MB with blue light were utilized for holographic recording. The latter possibility (MB) seems to be advantageous for several applications because the holographic grating is only formed during reconstruction. Higher reading intensities lead to higher population of the M-state and result in an increase of the fringe contrast instead of decreasing it. New possibilities for the further development of holographic media based on bacteriorhodopsin are raised by the availability of PM variants with modified optical properties. By the use of the variant BR-326, which differs from the wildtype PM by a single amino acid exchange (aspartate-96 → asparagine), the sensitivity of PM films is increased by ~50% from 12 cm2/J to 19 cm2/J for recording with 568 nm. The sensitivity for recording with 413 nm (33 cm2/J) is not influenced by the amino acid exchange. The observed diffraction efficiency η of PM films with BR-326 is twice that of BR-wildtype (BR-WT) films and is in the range of conventional organic photochromics (≈ 1%). In dried films of both BR-WT and BR-326 the M-decay was shown to be at least biexponential.  相似文献

The Oxygen evolving complex of plant photosystem II is made of a manganese cluster that gives rise to a low temperature EPR multiline signal in the S2 oxidation state. The origin of this EPR signal has been addressed with respect to the question of the magnetic couplings between the electron and nuclear spins of the four possible Mn ions that make up this complex. Considering Mn(III) and Mn(IV) as the only possible oxidation states present in the S2 state, and no large anisotropy of the magnetic tensors, the breadths of the EPR spectra calculated for dimers and trimers with S = ½ have been compared with that of the biological site. It is concluded that neither a dinuclear nor a trinuclear complex made of Mn(III) and Mn(IV) can be responsible for the multiline signal; but that, by contrast, a tetranuclear Mn complex can be the origin of this signal. The general shape of the experimental spectrum, its particular hyperfine pattern, the positions of most of the hyperfine lines and their relative intensities can be fit by a tetramer model described by the following six fitting parameters: g ≈ 1.987, A1 ≈ 122.4 10-4 cm-1, A2 ≈ 87.2 10-4 cm-1, A3 ≈ 81.6 10-4 cm-1, A4 ≈ 19.1 10-4 cm-1 and δH = 24.5 G. A second model described by parameters very close to those given above except for A4 ≈ 77.5 10-4 cm-1 gives an equally good fit. However, no other set of parameters gives an EPR spectrum that reproduces the hyperfine pattern of the S2 multiline signal. This demonstrates that in the S2 state of the oxygen evolving complex, the four manganese ions are organized in a magnetic tetramer.  相似文献

The orientation of pigments and pigment-protein complexes of the green photosynthetic bacterium Prosthecochloris aestuarii was studied by measurement of linear dichroism spectra at 295 and 100 K. Orientation of intact cells and membrane vesicles (Complex I) was obtained by drying on a glass plate. The photochemically active pigment-protein complexes (photosystem-protein complex and reaction center pigment-protein complex) and the antenna bacteriochlorophyll a protein were oriented by pressing a polyacrylamide gel. The data indicate that the near-infrared transitions (Qy) of bacteriochlorophyll c and most bacteriochlorophyll a molecules have a relatively parallel orientation to the membrane, whereas the Qy transitions of the bacteriochlorophyll a in the antenna protein are oriented predominantly perpendicularly to the membrane. Carotenoids and the Qx transitions (590–620 nm) of bacteriochlorophyll a, not belonging to the bacteriochlorophyll a protein, have a relatively perpendicular orientation to the membrane. The absorption and linear dichroism spectra indicate the existence of different pools of bacteriochlorophyll c in the chlorosomes and of carotenoid and bacteriopheophytin c in the cell membrane. The results suggest that the photosystem-protein and reaction center pigment-protein complexes are oriented with their short axes approximately perpendicular to the plane of the membrane. The symmetry axis of the bacteriochlorophyll a protein has an approximately perpendicular orientation.  相似文献

H+ currents in human neutrophils, rat alveolar epithelial cells, and several mammalian phagocyte cell lines were studied using whole-cell and excised-patch tight-seal voltage clamp techniques at temperatures between 6 and 42°C. Effects of temperature on gating kinetics were distinguished from effects on the H+ current amplitude. The activation and deactivation of H+ currents were both highly temperature sensitive, with a Q 10 of 6–9 (activation energy, E a, ≈ 30–38 kcal/mol), greater than for most other ion channels. The similarity of E a for channel opening and closing suggests that the same step may be rate determining. In addition, when the turn-on of H+ currents with depolarization was fitted by a delay and single exponential, both the delay and the time constant (τact) had similarly high Q 10. These results could be explained if H+ channels were composed of several subunits, each of which undergoes a single rate-determining gating transition. H+ current gating in all mammalian cells studied had similarly strong temperature dependences. The H+ conductance increased markedly with temperature, with Q 10 ≥ 2 in whole-cell experiments. In excised patches where depletion would affect the measurement less, the Q 10 was 2.8 at >20°C and 5.3 at <20°C. This temperature sensitivity is much greater than for most other ion channels and for H+ conduction in aqueous solution, but is in the range reported for H+ transport mechanisms other than channels; e.g., carriers and pumps. Evidently, under the conditions employed, the rate-determining step in H+ permeation occurs not in the diffusional approach but during permeation through the channel itself. The large E a of permeation intrinsically limits the conductance of this channel, and appears inconsistent with the channel being a water-filled pore. At physiological temperature, H+ channels provide mammalian cells with an enormous capacity for proton extrusion.  相似文献

We tested whether the thecosome pteropod Limacina helicina from Puget Sound, an urbanized estuary in the northwest continental US, experiences shell dissolution and altered mortality rates when exposed to the high CO2, low aragonite saturation state (Ωa) conditions that occur in Puget Sound and the northeast Pacific Ocean. Five, week-long experiments were conducted in which we incubated pteropods collected from Puget Sound in four carbon chemistry conditions: current summer surface (∼460–500 µatm CO2, Ωa≈1.59), current deep water or surface conditions during upwelling (∼760 and ∼1600–1700 µatm CO2, Ωa≈1.17 and 0.56), and future deep water or surface conditions during upwelling (∼2800–3400 µatm CO2, Ωa≈0.28). We measured shell condition using a scoring regime of five shell characteristics that capture different aspects of shell dissolution. We characterized carbon chemistry conditions in statistical analyses with Ωa, and conducted analyses considering Ωa both as a continuous dataset and as discrete treatments. Shell dissolution increased linearly as aragonite saturation state decreased. Discrete treatment comparisons indicate that shell dissolution was greater in undersaturated treatments compared to oversaturated treatments. Survival increased linearly with aragonite saturation state, though discrete treatment comparisons indicated that survival was similar in all but the lowest saturation state treatment. These results indicate that, under starvation conditions, pteropod survival may not be greatly affected by current and expected near-future aragonite saturation state in the NE Pacific, but shell dissolution may. Given that subsurface waters in Puget Sound’s main basin are undersaturated with respect to aragonite in the winter and can be undersaturated in the summer, the condition and persistence of the species in this estuary warrants further study.  相似文献

Initial reactions involved in the bacterial degradation of polycyclic aromatic hydrocarbons (PAHs) include a ring-dihydroxylation catalyzed by a dioxygenase and a subsequent oxidation of the dihydrodiol products by a dehydrogenase. In this study, the dihydrodiol dehydrogenase from the PAH-degrading Sphingomonas strain CHY-1 has been characterized. The bphB gene encoding PAH dihydrodiol dehydrogenase (PDDH) was cloned and overexpressed as a His-tagged protein. The recombinant protein was purified as a homotetramer with an apparent Mr of 110,000. PDDH oxidized the cis-dihydrodiols derived from biphenyl and eight polycyclic hydrocarbons, including chrysene, benz[a]anthracene, and benzo[a]pyene, to corresponding catechols. Remarkably, the enzyme oxidized pyrene 4,5-dihydrodiol, whereas pyrene is not metabolized by strain CHY-1. The PAH catechols produced by PDDH rapidly auto-oxidized in air but were regenerated upon reaction of the o-quinones formed with NADH. Kinetic analyses performed under anoxic conditions revealed that the enzyme efficiently utilized two- to four-ring dihydrodiols, with Km values in the range of 1.4 to 7.1 μM, and exhibited a much higher Michaelis constant for NAD+ (Km of 160 μM). At pH 7.0, the specificity constant ranged from (1.3 ± 0.1) × 106 M−1 s−1 with benz[a]anthracene 1,2-dihydrodiol to (20.0 ± 0.8) × 106 M−1 s−1 with naphthalene 1,2-dihydrodiol. The catalytic activity of the enzyme was 13-fold higher at pH 9.5. PDDH was subjected to inhibition by NADH and by 3,4-dihydroxyphenanthrene, and the inhibition patterns suggested that the mechanism of the reaction was ordered Bi Bi. The regulation of PDDH activity appears as a means to prevent the accumulation of PAH catechols in bacterial cells.  相似文献
