To clarify the role of the fungal nitrate assimilation pathway in nitrate reduction by mycorrhizal plants, nitrate reductase (NR)-deficient (NR) mutants of the ectomycorrhizal basidiomycete Hebeloma cylindrosporum Romagnesi have been selected. These mutants were produced by u.v. mutagenesis on protoplasts originating from homokaryotic mycelia belonging to complementary mating types of this heterothallic tetrapolar species. Chlorate-resistant mutants were first selected in the presence of different nitrogen (N) sources in the culture medium. Among 1495 chlorate resistant mycelia, 30 failed to grow on nitrate and lacked a detectable NR activity. Growth tests on different N sources suggested that the NR activity of all the different mutants is specifically impaired as a result of mutations in either the gene coding for NR apoprotein or genes controlling the synthesis of the molybdenum cofactor. Furthermore, restoration of NR activity in some of the dikaryons obtained after crosses between the different mutant mycelia suggested that not all the selected mutations mapped in the same gene. Utilization of N on a NH415NO3 medium was studied for two mutant strains and their corresponding wild-type homokaryons. None of the mutants could use nitrate whereas 15N enrichment values indicated that 13–27% of N present in 13-d-old wild-type mycelia originated from nitrate. Apparently, the mutant mycelia do not compensate their inability to use nitrate by a more efficient use of ammonium. These different NR mutants still form mycorrhizas with the habitual host plant, Pinus pinaster (Ait.), making them suitable for study of the contribution of the fungal nitrate assimilation pathway to nitrate assimilation by mycorrhizal plants.  相似文献

Previous work in the Colorado alpine ecosystem has shown that amino acids are a potentially important N source for the sedge, Kobresia myosuroides . This plant is the only known sedge to harbour associations with ectomycorrhizal fungi. The aim of the present work was to test the hypothesis that these ectomycorrhizas transfer N from amino acids in the soil solution to the host plant, and thereby have an important role in the N nutrition of this species. We used a two-chamber system (rhizoboxes) in which K. myosuroides plants were separated from a soil chamber by nylon mesh that allowed fungal hyphae, but not plant roots, to cross it. Injections of [15N, 2-13C]glycine were made into the soil chamber. The hyphal crossings on half of the rhizoboxes were regularly disrupted to control for leakage of label across the barrier. Plants in the intact rhizoboxes showed significantly higher 15N enrichment than those in controls, and mycorrhizal root tips were significantly more enriched than bulk roots. The mycorrhizas transferred an average of 1.3% of the added 15N label to plants, a figure comparable to those obtained in previous studies in which plant roots were directly exposed to label. We conclude that fungal associations have an important role in the N nutrition of K. myosuroides by transferring N from amino acids to their hosts.  相似文献

Arbuscular mycorrhizal fungi can enhance nutrient acquisition by a plant via their extraradical hyphae. This is particularly true for phosphorus, but the case for nitrogen (N) has been less clear. In our growth systems there was a small air-gap between root and hyphal compartments, which eliminated diffusion of nutrients between compartments. Moreover, our methods allowed us to distinguish between nitrate and ammonium. We found that N transfer to Zea maize L. depends on the sources fed to the hyphae of Glomus aggregatum Schenck & Smith. In experiment 1, despite the fact that plant demand for N was already met, plants received 10 times as much 15N from ammonium than from nitrate. In experiment 2, 74% of shoot-N was derived from the slow-release urea added to the hyphal compartment while only 2.9% was derived from the nitrate-N. Intraradical hyphae isolated from roots contained a considerable amount of 15N in the cell wall even when 15N-nitrate was the source. We conclude that the mycorrhizal fungus can rapidly deliver ammonium-N to the plants, and that while the fungus can absorb nitrate, it apparently lacks the capacity to transfer it to the plant.  相似文献

To develop further the methods for estimation of NOx absorption by plants supplied with 15N-labelled fertilizer, we proposed a new calculation method, total N fixed method (TNF), and compared with the 15N dilution method and the classical mass balance method (MB).
Hydroponically grown soybean plants were supplied with 15N-labelled nitrate and exposed to 200–250 nl l−1 NO2 for 7 d. The proportions of the N derived from NO2 to total N in exposed plants were estimated by the three methods.
The reported rates of NO2 absorption by several plant species, estimated by the 15N dilution method, were recalculated using the TNF method. The results of the two methods were compared and showed that: (1) The 15N dilution method overestimated the content of NO2-N in exposed plants compared with the MB method whilst the TNF method produced estimations of NO2-N closer to those by the MB method when the plants were supplied with 5 m M nitrate. (2) The differences in estimations between the MB method and either the 15N dilution method or the TNF method increased with decreasing supply of 15N-labelled nitrate to roots.  相似文献

Most work on root proliferation to a localized nutrient supply has ignored the possible role of mycorrhizal fungi, despite their key role in nutrient acquisition. Interactions between roots of Plantago lanceolata , an added arbuscular mycorrhiza (AM) inoculum and nitrogen capture from an organic patch ( Lolium perenne shoot material) dual-labelled with 15N and 13C were investigated, to determine whether root proliferation and nitrogen (N) capture was affected by the presence of AM fungi. Decomposition of the organic patch in the presence and absence of roots peaked in all treatments at day 3, as shown by the amounts of 13CO2 detected in the soil atmosphere. Plant N concentrations were higher in the treatments with added inoculum 10 d after patch addition, but thereafter did not differ among treatments. Plant phosphorus concentrations at the end of the experiment were depressed by the addition of the organic residue in the absence of mycorrhizal inoculum. Although uninoculated plants were also colonized by mycorrhizal fungi, colonization was enhanced at all times by the added inoculum. Addition of the AM inoculum increased root production, observed in situ by the use of minirhizotron tubes, most pronouncedly within the organic patch zone. Patch N capture by the end of the experiment was c . 7.5% and was not significantly different as a result of adding an AM inoculum. Furthermore, no 13C enrichments were detected in the plant material in any of the treatments showing that intact organic compounds were not taken up. Thus, although the added AM fungal inoculum benefited P. lanceolata seedlings in terms of P concentrations of tissues it did not increase total N capture or affect the form in which N was captured by P. lanceolata roots.  相似文献

Ratios of nitrogen (N) isotopes in leaves could elucidate underlying patterns of N cycling across ecological gradients. To better understand global-scale patterns of N cycling, we compiled data on foliar N isotope ratios (δ15N), foliar N concentrations, mycorrhizal type and climate for over 11 000 plants worldwide. Arbuscular mycorrhizal, ectomycorrhizal, and ericoid mycorrhizal plants were depleted in foliar δ15N by 2‰, 3.2‰, 5.9‰, respectively, relative to nonmycorrhizal plants. Foliar δ15N increased with decreasing mean annual precipitation and with increasing mean annual temperature (MAT) across sites with MAT ≥ −0.5°C, but was invariant with MAT across sites with MAT < −0.5°C. In independent landscape-level to regional-level studies, foliar δ15N increased with increasing N availability; at the global scale, foliar δ15N increased with increasing foliar N concentrations and decreasing foliar phosphorus (P) concentrations. Together, these results suggest that warm, dry ecosystems have the highest N availability, while plants with high N concentrations, on average, occupy sites with higher N availability than plants with low N concentrations. Global-scale comparisons of other components of the N cycle are still required for better mechanistic understanding of the determinants of variation in foliar δ15N and ultimately global patterns in N cycling.  相似文献

Abstract. The 15N isotope was used to compare the uptake and the assimilation of NH4+ and NO3 nitrogen in ryegrass ( Lolium perenne L.) during regrowth after cutting. Uptake of nitrate-N, expressed per plant, was at all times greater than ammonium-N uptake and assimilation decreased in roots and stubble while its assimilation was maintained at a high level in leaves. It has been suggested that ammonium assimilation is directly related to the availability of carbohydrates in the sink organ (leaves) resulting from their remobilization from the source organs (roots and stubble). Nitrate reduction decreased in all organs, while the uptake of NO3 was still high. After this first period of regrowth, nitrogen assimilation both from nitrate and ammonium increased in all the plants. Nitrate reduction capacity (expressed in μg NO3-N reduced per g D.W. per d) is 7.5 and 22.5 times greater in leaves than in stubble and roots, respectively. Therefore, nitrogen assimilation in stubble and particularly in roots was mainly dependent on ammonium nitrogen.  相似文献

In order to establish the relationship of nitrogen nutrition to root formation in vitro, efficiency of various forms of nitrogen supply was investigated in micropropagated shoots of Pinus pinaster. Nitrate (3.3 m M as N) containing medium supplemented with glutamine (2 m M as N) resulted in optimal rooting (control medium) whereas the rooting response was inhibited by the presence of glutamate (2 m M as N) in the medium. Study of 15NO3 behaviour in shoots cultured on the control medium showed a marked increase of 15N incorporation during the second half of a period of auxin treatment (total duration 16 days). The majority of the nitrogen incorporated from the medium was accumulated into protein in the rooting zone. Autoradiography of [3H]-thymidine in this reacting zone showed strong DNA synthesis at this time. Addition of glutamate both prevented the root formation process and resulted in a marked slowdown of nitrate uptake and decrease of protein synthesis.  相似文献

Red clover, Trifolium pratense L., is the dominant forage legume in Sweden and is usually harvested twice per year, once in June and once in August. Two 15N-based methods –15N isotopic dilution (ID) and 15N natural abundance (NA) – were used to study N2 fixation from spring until first harvest in late June, from first to second harvest in late August, and from second harvest until first frost in autumn in Umeå, Sweden. The material studied comprized three neighbouring fields carrying a first year ley, a second year ley and a third year ley. For the 15N ID method, small amounts of highly enriched 15N-nitrate were added to experimental plots. The non-legumes in the plots, essentially Phleum pratense L. together with Festuca pratensis L., served as reference plants for both the ID and 15N NA measurements. Dry matter, N and 15N were separately analysed in leaves (laminae), stems (including petioles), stubble and roots. The proportion of N derived from air (pNdfa) was then calculated for each plant part and for whole plants. Estimates of the proportion of N derived from N2 fixation (pNdfa) were always very high, usually ≥0.8. Generally, estimates of pNdfa obtained by the ID and NA methods were similar, but the ID method gave higher estimates of pNdfa than the NA method when the highest N2 fixation levels were recorded, at the August harvest. Regression analyses suggest that estimates of pNdfa in leaves could provide useful indications of pNdfa in shoots and whole T. pratense plants, thus avoiding the need for time-consuming root analyses.  相似文献

To investigate nitrogen assimilation and translocation in Zea mays L. colonized by the vesicular-arbuscular mycorrhizal (VAM) fungus Glomus fasciculatum (Thax. sensu Gerd.), we measured key enzyme activities, 15N incorporation into free amino acids, and 15N translocation from roots to shoots. Glutamine synthetase and nitrate reductase activities were increased in both roots and shoots compared with control plants, and glutamate dehydrogenase activity increased in roots only. In the presence of [15N]ammonium, glutamine amide was the most heavily labeled product. More label was incorporated into amino acids in VAM plants. The kinetics of 15N labeling and effects of methionine sulfoximine on distribution of 15N-labeled products were entirely consistent with the operation of the glutamate synthase cycle. No evidence was found for ammonium assimilation via glutamate dehydrogenase. 15N translocation from roots to shoots through the xylem was higher in VAM plants compared with control plants. These results establish that, in maize, VAM fungi increase ammonium assimilation, glutamine production, and xylem nitrogen translocation. Unlike some ectomycorrhizal fungi, VAM fungi do not appear to alter the pathway of ammonium assimilation in roots of their hosts.  相似文献

Interactive effects of elevated atmospheric CO2 and arbuscular mycorrhizal (AM) fungi on biomass production and N2 fixation were investigated using black locust ( Robinia pseudoacacia ). Seedlings were grown in growth chambers maintained at either 350 μmol mol−1 or 710 μmol mol−1 CO2. Seedlings were inoculated with Rhizobium spp. and were grown with or without AM fungi. The 15N isotope dilution method was used to determine N source partitioning between N2 fixation and inorganic fertilizer uptake. Elevated atmospheric CO2 significantly increased the percentage of fine roots that were colonized by AM fungi. Mycorrhizal seedlings grown under elevated CO2 had the greatest overall plant biomass production, nodulation, N and P content, and root N absorption. Additionally, elevated CO2 levels enhanced nodule and root mass production, as well as N2 fixation rates, of non- mycorrhizal seedlings. However, the relative response of biomass production to CO2 enrichment was greater in non-mycorrhizal seedlings than in mycorrhizal seedlings. This study provides strong evidence that arbuscular mycorrhizal fungi play an important role in the extent to which plant nutrition of symbiotic N2-fixing tree species is affected by enriched atmospheric CO2.  相似文献

The natural 15N abundance of amide-exporting nodules was compared to that of shoots in 12 plant species. Nodules were statistically less abundant in 15N than shoots in one of three cultivars of Pisum sativum L., in Vicia faba L. and in Medicago sativa L., but the 15N depletion of nodules was very samall. Nodules were statistically more abundant in 15N than shoots in Trifolium pratense L., depending on time during the growing season, Cyamopsis tetragonaloba L. Taub. and 7 Lupinus species, but the enrichment was small except for C. tetragonalova and 6 Lupinus species. Nodules of 3 Lupinus species infected with Rhizobium lupini isolated from Lupinus subcarnosa Hook, were only slightly enriched in 15N, but nodules of two of these species were substantially enriched in 15N when infected with a mix of other Rhizobium lupini strains. The third species, L. texensis Hook., was not infected by this mix of strains. Differences in 15N abundance between nodules and other tissues of amide-exporting and ureide-exporting nodules from several studies are tabulated. All ureide-exporting nodules in this tabulation are enriched in 15N. Amide-exporting nodules are considerably more variable in this regard. These results confirm that events associated with ureide synthesis or transport cannot be the sole cause of the substantial 15N enrichment seen in nodules.  相似文献

Nitrate uptake, reduction and translocation were examined in 5-week-old Austrian pines ( Pinus nigra Arnold var. nigricans Host.) during exposure to 5 m M NaNO3. The rate of nitrate uptake was linear during the 7 h light period. 15N-NO3 was detected in all parts of the pine except in the needles. By the 7th hour, 43% of the absorbed nitrate had been reduced, and this increased to 64% by the 24th hour. The major part of the total reduction occurred in the roots at this growth stage. Accumulation of 15N in reduced soluble and insoluble fractions was more prevalent in roots than in shoots. In the needles, the translocated nitrogen was mainly incorporated into the insoluble fraction. It is likely that most of the nitrogen from nitrate was transported from the roots to the aerial organs as organic nitrogen; however part of the upward nitrogen flux took place as nitrate ions.
An experiment in which an exposure for 24 h to 5 m M Na15NO3 was followed by 13 days exposure to Na14NO3 (pulse chase experiment) revealed a half time of about 1 day for depletion of root nitrate. A large part of this depletion was due to the loss of 15N-NO3 to the nutrient solution. The remaining pool of 15N-nitrate was partitioned between a metabolically inactive and an active pool. During the chase period, the simultaneous decrease of 15N-incorporation in the soluble N fraction and increase in the insoluble N fraction in different pine parts, particularly in the needles, suggested that protein synthesis occurred mostly in young tissues of the shoot and was the major sink of the newly absorbed 15N-NO3.  相似文献

A split root system for nitrogen uptake, in which one part of the root system was exposed to nitrogen-free nutrient and the other to circulated buffered ammonium, was used to investigate the effects of ammonium per se on the enzyme pathway for its assimilation in nodules and roots of leguminous plants. Plants of Trifolium repens L. cv. Grasslands Huia grown in the system showed similar growth and similar free amino acid content in the NH+4-fed roots and in nodulated plants. Studies of ammonium assimilation using [13N]-NH+4, applied to Glycine max [L.] Merr. cv. Amsoy plants, showed the label to be assimilated into amino acids in the NH+4-fed roots and to be transported to the tops before subsequently appearing in the minus-N side of the split root system. Analysis of the xylem sap showed [13N]-asparagine to be the principal labelled amino acid component. In these plants, levels of both allantoate and the nodule-specific isoenzyme aspartate aminotransferase-P2 were at least 10 times higher in the NH+4-fed roots than in the minus-N side of the split root system. These studies strongly suggest that a nodule-type of ammonium assimilation was occurring in the NH+4-fed side of the split root, and that this part of the root was transporting assimilatory products to the tops of the plants in a fashion analogous to that of a nitrogen-fixing nodule. These data implicate the involvement of NH+4 in the induction of its own assimilatory pathway.  相似文献

Assimilation of N by heterotrophic soil microbial biomass is associated with decomposition of organic matter in the soil. The form of N assimilated can be either low molecular weight organic N released from the breakdown of organic matter (direct assimilation), or NH+4 and NO3 from the soil inorganic N pool, into which mineralized organic N is released (mineralization immobilization turnover). The kinetics of C and N turnover in soil is quantifiable by means of computer simulation models. NCSOIL was constructed to represent the two assimilation schemes. The rate of N assimilation depends on the rate of C assimilation and microbial C/N ratio, thereby rendering it independent of the assimilation scheme. However, if any of the N forms is labeled, a different amount of labeled N assimilation will be simulated by the different schemes. Experimental data on inorganic N and 15N and on organic 15N dynamics in soils incubated with 15N added as NH+4 or organic N were compared with data simulated by different model schemes. Direct assimilation could not account for the amount of 15N assimilated in any of the experimental treatments. The best fit of the model to experimental data was obtained for the mineralization immobilization turnover scheme when both NH+4 and NO3 were assimilated, in proportion to their concentration in the soil.  相似文献

Symbiotic dinitrogen fixation by legume trees represents a substantial N input in agroforestry systems, which may benefit the associated crops. Applying 15N labelling, we studied N transfer via common mycelial networks (CMN) and root exudation from the legume tree Gliricidia sepium to the associated fodder grass Dichantium aristatum . The plants were grown in greenhouse in shared pots in full interaction (treatment FI) or with their root systems separated with a fine mesh that allowed N transfer via CMN only (treatment MY). Tree root exudation was measured separately with hydroponics. Nitrogen transfer estimates were based on the isotopic signature of N ( δ 15N) transferred from the donor. We obtained a range for estimates by calculating transfer with δ 15N of tree roots and exudates. Nitrogen transfer was 3.7–14.0 and 0.7–2.5% of grass total N in treatments FI and MY, respectively. Root δ 15N gave the lower and exudate δ 15N the higher estimates. Transfer in FI probably occurred mainly via root exudation. Transfer in MY correlated negatively with grass root N concentration, implying that it was driven by source-sink relationships between the plants. The range of transfer estimates, depending on source δ 15N applied, indicates the need of understanding the transfer mechanisms as a basis for reliable estimates.  相似文献

The osmotic role of nitrate during aftermath growth of Lolium perenne L. cv. Réveille was investigated. Plants were grown from seed in a controlled environment using a liquid medium with 1.0 m M NH4NO3 as nitrogen source.
Eight-week-old plants were cut 4.0 cm above the root system and then harvested over a 14-day period of regrowth on the same initial nutrient solution, except that nitrate was 15N labelled. Throughout the experimental period, nitrate storage and reduction in roots were low. In stubble and especially in leaves, nitrate accumulated during the first 6 days of regrowth whereas nitrate reduction mainly occurred after this period. Analyses of carbohydrate, chloride and potassium contents in stubble and leaves showed that the accumulation of nitrate osmotically compensated for the decrease in soluble sugars during the first 6 days of regrowth.
The cumulative osmotic potential of sugars, chloride and nitrate in differently treated plants was studied in stubble and leaves. Compared with uncut plants, the lower carbohydrate concentrations found in cut plants regrowing on 1.0 m M NH4NO3 were compensated for by an accumulation of nitrate. During aftermath growth on low nitrogen nutrition (0.2 m M NH4NO3), chloride replaced nitrate, supporting the proposed osmotic function of nitrate.
It is concluded that nitrate is involved in the osmotic adjustment of ryegrass during regrowth after cutting.  相似文献

The influence of salinity on the activity of nitrate reductase (NR, EC 1.6.6.1) and the level of the molybdenum cofactor (MoCo) as affected by the source and concentration of nitrogen was studied in annual ryegrass ( Lolium multiflorum cv. Westerwoldicum). Plants grown in sand were irrigated with nutrient solution with an electrical conductivity of 2 or 11.2 dS m−1, containing nitrogen (0.5 or 4.5 m M ) in the form of NH4NO3 or NaNO3 Salinity-treated (11.2 dS m−1) plants produced less biomass and more organic nitrogen while accumulating more NO3 than control plants. Increased nitrogen concentration in the irrigation solutions enhanced biomass and organic nitrogen production as well as NO3 accumulation irrespective of the electrical conductivity. Salinity inhibited shoot growth and increased shoot NR activity of plants receiving 4.5 m M NH4NO3 or NaNO3. Similar effects were observed in roots of plants grown in 4.5 m M NaNO3. Nitrate added to a complementation medium containing ryegrass MoCo and the NR apoprotein of Neurospora crassa mutant nit-1 stimulated the activity of the reconstituted NR (NADPH-nitrate reductase, EC 1.6.6.3). Increased salinity and nitrogen in the nutrient solutions caused an increase of MoCo content in roots and shoots. Similar results were observed for NR activity in the shoots. The increase of MoCo in response to salinity was more pronounced than that of NR, especially in the roots. We conclude that the pool size of MoCo in ryegrass is not constant, but varies in response to nutritional and environmental factors.  相似文献

Mosses have been used as biomonitors of atmospheric pollution for some years, but few studies have been carried out on the effect of NOx emissions from traffic on moss tissue N. Eight species of moss (102 samples) growing on walls or roofs next to roads exposed to different traffic densities were collected from urban and rural sites in the UK. The shoots were sampled for total N, their stable isotope 15N/14N content (δ15N) and heavy metal content (Pb, Zn). There was a lack of correlation between tissue total N and traffic exposure, but a very good correlation between traffic exposure and tissue δ15N. Plants collected near motorways or busy urban roads had δ15N values ranging between +6 and −1‰, while in rural areas with hardly any traffic these ranged from −2 to −12‰. In a separate survey of mosses, the average δ15N of shoots from busy roadsides in London was +3.66‰, whereas from samples collected from farm buildings near poultry or cattle pens it was −7.8‰. This indicates that the two main atmospheric N sources, NOx and NHx, have different δ15N signatures, the former tending to be positive and the latter negative. Tissue concentrations of both Pb and Zn show a strong positive correlation with traffic exposure, with Zn in particular being greater than Pb. The results are discussed with regard to the use of moss tissue Zn as a means for monitoring or mapping pollution from vehicles, and of δ15N as an aid to distinguish between urban (NOx) and rural (NHx) forms of N pollution.  相似文献
