Abstract .1. To benefit from the putative genetic advantages of multiple mating with multiple partners, female insects would be expected to select against mating with the same male twice when another potential partner is present.
2. This paper examined whether female Gryllus bimaculatus (Gryllidae) preferred to mate with a novel partner over a partner with which they had mated previously.
3. Females presented with a choice preferred significantly to mate with novel males over previously mated males, and preferred to do so even when the potentially confounding influence of male–male competition was controlled for.
4. The potential advantages of such a mate choice pattern and possible ways in which the choice is mediated are discussed.  相似文献

Transgenic insects have been artificially produced to study functions of interesting developmental genes, using insect transposons such as piggyBac. In the case of the cricket, however, transgenic animals have not yet been successfully artificially produced. In the present study, we examined whether the piggyBac transposon functions as a tool for gene delivery in embryos of Gryllus bimaculatus. We used either a piggyBac helper plasmid or a helper RNA synthesized in vitro as a transposase source. An excision assay revealed that the helper RNA was more effective in early Gryllus eggs to transpose a marker gene of eGFP than the helper plasmid containing the piggyBac transposase gene driven by the G. bimaculatus actin3/4 promoter. Further, only when the helper RNA was used, somatic transformation of the embryo with the eGFP gene was observed. These results suggest that the piggyBac system with the helper RNA may be effective for making transgenic crickets.  相似文献

  • 1 The adaptive significance of multiple mating by female Gryllus bimaculatus (De Geer) was investigated.
  • 2 Multiple mating prevented the depletion of sperm stores and, therefore, maintained high hatching success. This may not, however, explain the high frequency of remating in this species.
  • 3 Male-derived egg stimulants known to be passed with sperm at mating increased the number of eggs produced only when females mated throughout their lifespans.
  • 4 Spermatophore consumption appeared to provide nutrients which, while they did not increase the quantity of eggs, increased egg quality as indicated by weight. Females who consumed spermatophores had a greater hatching success.
  • 5 While females may derive non-genetic benefits from mating, these are apparently long-term benefits; females must mate throughout their lives in order to accrue them.
  • 6 Since the benefits of mating may not be derived from individual males, the spermatophores and their contents in this species are best considered as mating effort.

We have developed a new set of 27 polymorphic markers for each of two cricket species, Gryllus bimaculatus and Gryllus campestris. Initially, 14 published G. bimaculatus loci were tested in G. campestris; however, only five loci were polymorphic. Therefore, we isolated an additional 50 new microsatellite loci from G. bimaculatus and tested these in both species. In a minimum of 20 individuals, 27 of the new loci were polymorphic in G. bimaculatus and 25 in G. campestris.  相似文献

ABSTRACT. The corpora cardiaca of the cricket, Gryllus bimaculatus De Geer, contain peptidic material which elicits hyperlipaemia in migratory locusts and in crickets. Appreciable amounts of this adipokinetic factor are found only in the corpora cardiaca of the cricket, and not in other parts of the nervous system. The factor is heat-stable and retains its activity after incubation with the exopeptidases leucine aminopeptidase and carboxypeptidase A. Fractionation of a methanolic extract of corpora cardiaca from Gryllus bimaculatus by RP-HPLC shows that the adipokinetic activity is confined to a single absorbance peak. The amino acid composition of the purified adipokinetic neuropeptide was determined after acid hydrolysis with HCI and with methanesulfonic acid. The factor is an octapeptide containing the following amino acid residues in equimolar amounts: Asx, Thr, Ser, Glx, Gly, Val, Phe and Trp. This composition closely resembles the AKHI II molecule from Schistocerca species , and the Gryllus peptide can be assigned to the AKH/RPCH family of neuropeptides.  相似文献

ABSTRACT. Incubation conditions were established for a short-term radiochemical assay of spontaneous juvenile hormone (JH) biosynthesis in vitro by corpora allata from adult female Gryllus bimaculatus. The only JH synthesized was shown by HPLC to be JH III. A further incubation product, predominantly extracted from the corpora allata, was thought to be the JH III precursor, methyl farnesoate. In adult females reared at a constant temperature of 27°C the synthetic activities of the corpora allata-corpora cardiaca complexes in vitro increase from almost zero to a high peak value 4 days after the imaginal moult. Thereafter the activity decreases to varying intermediate levels, but always lower than the first maximum. Two days after the first peak in corpus allatum activity, ovarian fresh weight increases dramatically and the first oviposition occurs 2 days later.
Topical application of JH III to females reared at 20°C, which usually have a low fecundity, causes a dose-dependent stimulation of egg production and oviposition.  相似文献

The cricket Gryllus bimaculatus displayed freezing (thanatosis) after struggling while the femoro-tibial joints of the walking legs were forcibly restrained. Myographic recording indicated that strong contraction of the flexor tibia muscle “leg flexion response” occurred under this restrained condition. During thanatosis, when the femoro-tibial joint was passively displaced and held for several seconds, it maintained its new position (catalepsy). Only discharge of the slow flexor units was mechanically indispensable for maintaining thanatosis and catalepsy. Differing roles of identified neuron subgroups of the femoral chordotonal organ were elucidated using this behavioral substrate. Ablation of the dorsal group neurons in the ventral scoloparium strengthened the leg flexion response and the normal resistance reflex, while ablation of the ventral group weakened both motor outputs. Ablation of the dorsal scoloparium neurons, or other main sensory nerves caused no detectable deficiency in femoro-tibial joint control. These results imply that both modes of flexor muscle activation promoted by the ventral group neurons are normally held under inhibitory control by the dorsal group. It is hypothesized that this antagonistic function causes immobilization of the femoro-tibial joint in a wide range of angles in thanatosis and catalepsy. Accepted: 12 November 1998  相似文献

Adipokinetic hormone (AKH) regulates energy homeostasis in insects by mobilizing lipid and carbohydrate from the fat body. Here, using RNA sequencing data, we identified cDNAs encoding AKH (GbAKH) and its highly homologous hormone AKH/corazonin-related peptide (GbACP) in the corpora cardiaca of the two-spotted cricket, Gryllus bimaculatus. RT-PCR revealed that GbAKH and GbACP are predominantly expressed in the corpora cardiaca and corpora allata, respectively. Phylogenetic analysis confirmed that the identified GbAKH and GbACP belong to the clades containing other AKHs and ACPs, respectively. Injection of synthetic GbAKH and GbACP elevated hemolymph carbohydrate and lipid levels and reduced food intake significantly. In contrast, knockdown of GbAKH and GbACP by RNA interference increased the food intake, although hemolymph lipid level was not altered. Collectively, this study provides evidence that ACP regulates hemolymph carbohydrate and lipid levels in cricket, possibly collaborative contribution with AKH to the maintenance of energy homeostasis.  相似文献

Abstract Allatectomy of young penultimate nymphs of Gryllus bimaculatus De Geer (Gryllidae) resulted in prothetelic creatures which exhibited reproductive competence. The same operation performed on young last instar nymphs resulted in moulting to morphologically normal adults. Allatectomized morphologically normal adult females, as well as prothetelic ones, showed the same level of sexual receptivity as untreated control females. Allatectomized morphologically normal and prothetelic females laid viable eggs, but rate of egg laying and number of eggs produced by these females were much reduced in comparison with the controls. Administration of methoprene (a Juvenile Hormone analogue) to allatectomized females restored egg production to a more or less normal rate. Removal of the spermatophore within 10 min of copulation had no effect on subsequent sexual receptivity of the females, nor on the reduced rate of egg laying by the allatectomized females, but did affect the rate of egg laying by control females.
It is suggested that the corpora allata (CA) and the Juvenile Hormone (JH) play no major role in controlling basic sexual receptivity of G.bimaculatus females, and do not have an all-encompassing control on egg production, though they do exert a marked quantitative effect on the rate of egg production.  相似文献

Male field crickets produce calling songs that are used for attracting conspecific females for mating. Acoustic communication was studied in the trilling field cricket, Gryllus rubens.A trilled calling song consists of a continuous train of sound pulses, each of which was repeated at a stereotyped rate. Singing males were recorded at different temperatures. The pulse period of the calling song decreased with increasing temperature; there was less effect of temperature on pulse duration. Female phonotaxis was studied on a noncompensating, spherical treadmill. In two-stimulus (choice) playback experiments, females preferred the conspecific trilled song over the chirped calling song of a sympatric species, Gryllus fultoni.This preference persisted even when the song of G. fultoniwas 6 dB louder. Females also discriminated between synthetic trills having different pulse periods; females chose trills with the conspecific pulse period over trills having lower and higher pulse periods.  相似文献

Although female mating preferences are a focus of current controversy,little detailed information exists on female preferences withinnatural populations. In the field cricket Gryllus integer, malecalls attract sexually receptive females, and females preferentiallymove toward male calls with longer calling bouts (periods ofcalling containing no pause greater than 0.10 s in real time).This study investigated female preferences for other variablesof the male song, including syllable period, chirp pause, andnumber of syllables per chirp. Male song was measured in thefield to determine mean values for each variable in nature.Female preferences were determined using a locomotor-compensatordevice, on which females ran in response to sequential playbacksof synthesized male song. Mean female preferences correspondedroughly to mean male song variables. Nonetheless, females variedgreatly in their responses to synthesized calls differing insyllable period, syllable number, and chirp pause. Moreover,individual females who were more selective for any one variablealso tended to be more selective for others. These results showthat females may differ from one another in their mating preferencesand degrees of selectivity, even within a single population.  相似文献

The influence of operational sex ratio on the mating behavior of female field crickets,Gryllus pennsylvanicus, was investigated. Females were predicted to be more discriminating under conditions of high mate availability and show less selectivity when males were rare. Such selectivity was indicated in this study with the proportion of courtships leading to a mating changing with sex ratio. Females accepted almost 70% of all courtships at the female-biased sex ratio, but only about half of all courtships were successful at even or male-biased sex ratios. Females moved least at the female-biased sex ratio. There was also a trend for females to be guarded more under male-biased conditions. Female weight did not influence any of the behaviors examined.  相似文献

The allatostatin (AST) type A gene of the cricket Gryllus bimaculatus encodes a hormone precursor including at least 14 putative peptides with a common C-terminus Y/FXFGL/Iamide. By RT-PCR we have analyzed the expression of the allatostatin precursor in various tissues of 0-21 days old adult virgin and mated females. In 3-day-old virgin females, the gene is strongly expressed in the brain (oesophageal ganglion), the suboesophageal ganglion and the caecum, but to a lower extent in other parts of the digestive tract (ileum, midgut, colon), and in various other tissues such as the fat body, ovaries and female accessory reproductive glands. In the brain and ovaries of virgin females, the AST expression is rather constant throughout adult life, whereas in brains of mated animals, expression is low until day 7, but increases sharply from day 8 onwards to reach values triple those before day 7. In ovaries of mated animals AST gene expression is also age-dependent, with high expression rates during the first 4 days after imaginable moult, a second but smaller peak from day 15 to 21, and very low values in between. In the fat body of virgin crickets allatostatin expression is high during the first 9 days after ecdysis and declines thereafter, whereas in mated animals two peak values, day 1 and day 6, are observed, and a third peak in older animals.  相似文献

Abstract. Female preferences for conspicuous male calls have been documented in many groups. However, relatively few studies have examined the metabolic costs associated with the production of call types preferred by females. We measured the oxygen consumption of calling male Gryllus lineaticeps Stål crickets using closed chamber respirometry. Calling song was recorded concurrently. The average increase in mass-specific oxygen consumption during calling was 2.7 times basal rates of oxygen consumption, and calling males consumed approximately 1.2ml O2g-lh-1. Oxygen consumption increased with increasing chirp rate and pulse duration, but not with increasing chirp duration. Females of this species prefer higher chirp rates, thus some call types that increase the male's attractiveness to females require more metabolic energy to produce.  相似文献

Female crickets lacking experience with phonotaxis to conspecific calling song respond to trains of continuously repeated sound pulses (trill), whereas experienced females do not. In the present study such inexperienced crickets were tested for their responsiveness to trills of pulse repetition periods from 30 to 70 ms on a Y- shaped maze. Stimulation with a repetition period of 30 ms led to unexpectedly low phonotactic and exploratory activity. Initial stimulation with trills of 30- ms repetition period drastically reduced the responsiveness of inexperienced animals to conspecific calling song and other attractive stimuli. Effects of visual stimulation on the phonotactic behavior of female crickets are demonstrated. Threatening visual stimuli changed the behavior of experienced animals to a state that resembles that of inexperienced animals. The relevance of these observations is discussed with respect to the development of the auditory pattern recognition mechanism in crickets.  相似文献

The cricket Gryllus bimaculatus is a typical hemimetabolous intermediate germ insect, in which the processes of segmentation and appendage formation differ from those in Drosophila, a holometabolous long germ insect. In order to compare their developmental mechanisms, we have focused on Gryllus orthologs of the Drosophila developmental regulatory genes and studied their functions. Here, we report a functional analysis of the Gryllus ortholog of extradenticle (Gbexd) using embryonic and parental RNA interference (RNAi) techniques. We found the following: (1) RNAi suppression of Gb′exd results in the deletion or fusion of body segments. Especially the head was often very severely affected. This gap-like phenotype may be related to reduced expression of the gap genes hunchback and Krüppel in early RNAi germbands. (2) In the appendages, several segments (podomeres) were fused. (3) Head appendages including the antenna were transformed to a leg-like structure consisting of at least one proximal podomere as well as several tarsomeres. The defects in appendages are reminiscent of the phenotype caused by large exd clones in Drosophila antennal discs. These findings led us to the conclusion that (1) Gb′exd is required for segment patterning in the gnathal to abdominal region, acting in a gap gene-like manner in the anterior region. (2) Gb′exd plays important roles in formation of the appendages and the determination of their identities, acting as a regulatory switch that chooses between the fates of head appendages versus the appendage ground state. Although functions of Gb′exd in appendage patterning appear fundamentally conserved between Gryllus and Drosophila, its role in body segmentation may differ from that of Drosophila exd.  相似文献

Fighting is often composed of discrete agonistic displays. Few studies have partitioned fighting behavior into its component agonistic displays and evaluated the relationships between the frequency of the displays and the potential benefits of fighting, particularly mating success. In this study, we quantified the frequency of male field cricket, Gryllus bimaculatus, agonistic displays. The displays were quantified under three social environments which varied in the potential benefits of fighting: males with other males only, males with other males and female scents, and males with other males and females. We found that (1) the presence of females elicited an increase in agonistic displays characteristic of intermediate levels of escalation, (2) female scents did not produce a similar increase in the frequency of agonistic displays, and (3) in the presence of females, the frequency of agonistic displays was positively correlated with mating success. Aggressive stridulation, an energetically low-cost display, was more strongly associated with mating success than were more costly displays. The results are discussed in the context of the evolutionary theory of aggression and in the context of cricket mating systems.  相似文献

The detailed developmental morphology of the spermatophore of a cricket (Gryllus bimaculatus, De Geer) is described for the first time. A specific behavioural reference point is identified from which time developmental processes proceed in a more or less robotic manner. The structures previously identified as forming part of the fully formed spermatophore, that is, the ampulla, the sperm tube and the attachment plate are all identified in detail during their development. Interestingly the attachment plate material is extruded as an amorphous mass from the ejaculatory duct prior to its insertion into the dorsal pouch, the sperm do not enter the sperm sac until the end of spermatophore formation, and the sperm tube itself is formed from a flattened layer of material which rolls up into a tube and involves the use of a die mechanism.  相似文献
