F?/Cl? selectivity in CLCF-type F?/H+ antiporters

Many bacterial species protect themselves against environmental F⁻ toxicity by exporting this anion from the cytoplasm via CLC^F F⁻/H⁺ antiporters, a subclass of CLC superfamily anion transporters. Strong F⁻ over Cl⁻ selectivity is biologically essential for these membrane proteins because Cl⁻ is orders of magnitude more abundant in the biosphere than F⁻. Sequence comparisons reveal differences between CLC^Fs and canonical Cl⁻-transporting CLCs within regions that, in the canonical CLCs, coordinate Cl⁻ ion and govern anion transport. A phylogenetic split within the CLC^F clade, manifested in sequence divergence in the vicinity of this ion-binding center, raises the possibility that these two CLC^F subclades might exhibit differences in anion selectivity. Several CLC^F homologues from each subclade were examined for F⁻/Cl⁻ selectivity of anion transport and equilibrium binding. Differences in both of these anion-selectivity metrics correlate with sequence divergence among CLC^Fs. Chimeric constructs identify two residues in this region that largely account for the subclade differences in selectivity. In addition, these experiments serendipitously uncovered an unusually steep, Cl⁻-specific voltage dependence of transport that greatly enhances F⁻ selectivity at low voltage.

Conclusions Malgré ses enclaves de savance et la prairie, riches en espèces tropicales xérophiles, le territoire étudié appartient incontestablement au domaine forestier guineo-équatorial. Plusieurs climax, tous forestiers, appartenant, suivant les conditions locales, aux ordres des Triplochitetalia, Lophiretalia et Parinarietalia, y sont représentés. En fait, cette végétation primitive est profondément dégradée, aux basses altitudes par les défrichements, et sur les crêtes par les feux. C'est dire tout l'intérêt que présente, dans ce territoire, la protection de la Nature; la mise en réserve naturelle intégrale du massif du Nimba, promulguée en juillet 1944 sur l'initiative de M. le Professeur Heim et de M. le Professeur Monod, Directeur de l'Institut Français d'Afrique Noire, préservera de la destruction les richesses botaniques de cette région montagneuse.Reçue par la rédaction le 10-XI-1950.

Neuropeptide F: a ubiquitous invertebrate neuromediator?

Using specific antisera, neuropeptide F (NPF)-related peptides have been identified immunocytochemically as widespread and abundant in the nervous systems of all invertebrate taxa examined so far. To date, four NPFs have been isolated and sequenced: from the cestode, Moniezia expansa and the turbellarian, Artioposthia triangulata, and from the molluscs, Helix aspersa and Aplysia californica; a related nonapeptide has been sequenced also from Loligo vulgaris. These peptides all display structural characteristics of the vertebrate NPY superfamily of peptides and appear, therefore, to represent invertebrate members of this superfamily. In this respect, invertebrate NPFs most likely represent the precursors of the vertebrate NPY superfamily. Homologies between the gene structure of human NPY and molluscan NPF (A. californica) support the view that the NPY/NPF gene is of ancient lineage. Although NPF (A. californica) has been found to inhibit the activity of the abdominal ganglia in Aplysia, its widespread expression in this mollusc would suggest multiple functions; the physiological role(s) of NPFs in other invertebrates awaits examination. The abundance and apparent ubiquitous nature of NPF-related peptides establishes them as evolutionarily-ancient molecules that likely serve important physiological functions in invertebrate neurobiology.
