Summary N-acetyltransferase (NAT) activity in pineal glands exhibits a circadian rhythm with peak activity occurring in the dark-time. We previously showed that inGallus domesticus chicks pretreated with LD12:12, NAT activity was increased by dark exposure (peak dark sensitivity occurred during the expected dark-time) or decreased by light at night (peak light sensitivity occurred early in the night during the time of dark sensitivity). In this study we mapped dark sensitivity vs time (for NAT activity increase in response to 2 h dark pulses), and light sensitivity vs time (for NAT activity decrease in response to 10 min or 30 min light pulses) over a cycle for 3-week old chicks,Gallus domesticus, pretreated with long (LD16:8) or short photoperiod (LD8:16). Sensitivity to light was increased in the second 8 h after L/D by LD8:16. Attraction of maleAgrotis segetum Dennis & Schiffermüller (Lepidoptera: Noctuidae) to sex pheromone traps in fields, which were treated with one or three pheromone components was investigated. Small plots of 1/4ha size were treated with synthetic pheromone, released by 25 evenly dispersed latex rubber tube dispensers. The dispensers were loaded with either 500 μg Z5-10:OAc (50 mg/ha), or 1000 μg Z7-12:OAc (100 mg/ha), or a 3-component mixture consisting of 500 μg Z5-10:OAc+1000 μg Z7-12:OAc+1000 μg Z9-14:OAc. Pheromone traps were placed both within and outside of the treated area in a cross design, with an intertrap spacing of 15 m. Release rates from disruption dispensers were measured in the laboratory after being exposed in the field. The release rates of the components were estimated to be 0.44, 0.11, and 0.06 μg/h/dispenser for Z5-10:OAc, Z7-12:OAc and Z9-14:OAc, respectively. The highest effect of disruption was achieved by the three-component blend, resulting in a significant suppression of trap catches extending 5 m outside of the treated area. The Z5-10:OAc treatment resulted in reduced trap catches inside the treated area, but the effect did not extend outside. Z7-12:OAc alone did not result in any significant reduction in trap catch. The results indicate that different mechanisms may explain the disruptive effect of the treatments and that the single pheromone components are not as effective as the three-component blend.  相似文献

The locomotor activity of the millipede Glyphiulus cavernicolus (Spirostreptida), which occupies the deeper recesses of a cave, was monitored in light-dark (LD) cycles (12h light and 12h darkness), constant darkness (DD), and constant light (LL) conditions. These millipedes live inside the cave and are apparently never exposed to any periodic factors of the environment such as light-dark, temperature, and humidity cycles. The activity of a considerable fraction of these millipedes was found to show circadian rhythm, which entrained to a 12:12 LD cycle with maximum activity during the dark phase of the LD cycle. Under constant darkness (DD), 56.5% of the millipedes (n = 23) showed circadian rhythms, with average free-running period of 25.7h ± 3.3h (mean ± SD, range 22.3h to 35.0h). The remaining 43.5% of the millipedes, however, did not show any clear-cut rhythm. Under DD conditions following an exposure to LD cycles, 66.7% (n = 9) showed faint circadian rhythm, with average free-running period of 24.0h ± 0.8h (mean ± SD, range 22.9h to 25.2h). Under constant light (LL) conditions, only 2 millipedes of 11 showed free-running rhythms, with average period length of 33.3h ± 1.3h. The results suggest that these cave-dwelling millipedes still possess the capacity to measure time and respond to light and dark situations. Firing and resetting of traps in aquatic Utricularia species are associated with water flow and trap volume changes. In this study, trap thickness was used as a measure of water flow and was monitored automatically using an electronic position sensor. The basic characteristics of mechanically stimulated firing and resetting were measured in isolated traps from 13 aquatic Utricularia species and in two trap size categories in Utricularia reflexa. This allowed to study the relationship between these trap functions and trap thickness and length (criteria of trap size). Additionally, the characteristics of spontaneous firings (without any mechanical stimulation) were compared for U. reflexa traps fed or denied prey during a 1-day period. On the absolute scale, the 13 Utricularia species differed considerably in their firing and resetting rates. Significant interspecific differences were also found in the magnitude of firing (in total 3.7–4.2 times) and resetting rates (10–24 times) per unit trap thickness or length. Overall, traps of Utricularia australis, Utricularia stellaris and Utricularia inflata showed the greatest firing and resetting rates. The relative magnitude of firing per unit trap thickness or length showed a highly significant negative correlation with both trap thickness and length and the same also held for the relative resetting rates. Smaller and narrower traps are thus relatively more effective at trap firing and resetting than larger traps. Neither firing nor resetting characteristics were significantly different between unfed and prey-fed traps of U. reflexa and this was also true for the occurrence of spontaneous firings. A strict linear resetting rate, without any lag-period, was found during the first 3 min after trap firing in U. reflexa. This suggests that water is pumped out of the trap continuously and probably recirculates. Given the concepts of spontaneous firing and water recirculation, an ecological model based on the literature data has been devised to quantify the daily N and P gain from the ambient water by Utricularia traps devoid of animal prey. The model shows that the total N and P content estimated in the trap fluid is too high to be accumulated from only the ambient water. This implies that the prey-free traps do not take up N or P from the trap fluid but rather exude a quantity of these nutrients to the fluid to support the microbial community. Therefore, the trap microorganisms behave more as parasites than commensals and represent an additional ecological cost for trap maintenance.  相似文献

A sex pheromone-baited delta trap was found to be inefficient at eliciting landing and entering of the trap by maleCtenopseustis obliquana. The inefficiency of the delta trap related to turbulence altering the pheromone plume and the concomitant effect on the flight manoeuvres of male moths. In the wind tunnel, high proportions of males flew upwind and landed on the sides, outside, of the trap, but only a relatively small proportion of these males entered the trap and contacted the sticky surface. When males approached the delta trap, they tended to fly in wide zigzags (i.e., large inter-track reversal distances) and at an altitude near the top of the trap, where the trap was relatively narrow in width (compared to the bottom). Thus, these flight manoeuvres largely precluded males from entering the trap. Greater numbers of male moths entered the trap when: (i) the front barriers of the delta trap were removed, (ii) pheromone dosage was increased to 300 μg, and (iii) the trap design was changed to a rectangular one. The first two changes appeared to influence the flight manoeuvres of males (who appeared to fly with narrower inter-track reversal distances), while the third change apparently did not affect the glight manoeuvres of males, but rather allowed more males to enter the trap because of the greater area of the entrance. The low trap catches of maleC. obliquana in the field are also a consequence of the glue which is currently used. After contact with this glue most males are able to escape, flying off the sticky surface and losing contact with the pheromone. A field trial found that delta traps with another glue caught more than three times the number of males of the related tortricid moth,Epiphyas postvittana, than delta traps with the currently used glue.  相似文献

The relationship between the degree of chilling resistance and phase shifting caused by low-temperature pulses was examined in two circadian rhythms in cotton (Gossypium hirsutum L. cv. Deltapine 50) seedlings grown under light-dark cycles of 1212 h at 33° C. The seedlings showed a circadian rhythm of chilling resistance and of cotyledon movement. A pulse of 19° C for 12 h during the chilling-sensitive phase (light period) caused a phase delay of 6 h, while a similar temperature pulse during the chilling-resistant phase (dark period) did not cause any phase shift. Exposure to 19° C, 85% RH (relative humidity) for 12 h during the dark period induced chilling resistance in the following otherwise chilling-sensitive light period. In this light period a 12-h 19° C pulse did not cause a phase shift of chilling resistance. Pulses of low temperatures (5–19° C) were more effective in causing phase delays in the rhythm of cotyledon movement when given during the chilling-sensitive phase than when given during the chilling-resistant phase. A 12-h pulse of 5° C, 100% RH during the light period caused a phase delay of cotyledon movement of 12 h. However, when that pulse had been preceded by a chill-acclimating exposure to 19° C, 85% RH for 12 h during the dark period the phase delay was shortened to 6 h. The correlation between higher degree of chilling resistance and the prevention or shortening of the phase delay caused by low temperatures indicates that the mechanism that increases chilling resistance directly or indirectly confers greater ability for prevention of phase shifting by low temperatures in circadian rhythms.Abbreviations CT circadian time - LDC light-dark cycle of 24 h - RH relative humidity  相似文献

Hyphal fusion during initial stages of trap formation by Arthrobotrys oligospora was studied by video-enhanced contrast and electron microscopy. Trap initials grew perpendicularly to the parent hypha, then curved around and anastomosed with a peg that developed on the hypha. Trap initials usually developed 40–140 m apart while the anastomosis occurred 20–25 m from the initial. Vigorous cytoplasmic movements in trap initials and developed traps corresponded to intense staining with fluorescein diacetate (FDA) of these cells. In addition, bundles of microfilaments were seen in developing loops of traps. On fusion organelle migration took place from the tip cell of the trap into the peg. Later on a septum was formed at the site of fusion.  相似文献

Sticky rectangle traps of various yellow colours and fluorescence made of cardboard were field tested against western cherry fruit fly, Rhagoletis indifferens Curran, in paired trap preference experiments in Washington state. In a first experiment that involved comparisons of Alpha Scents (proprietary paint), Fluorescent Yellow (aerosol paint), Saffron Thread and Neon Light (semi‐gloss enamel paints) traps at two sites, the best performing trap was the Saffron Thread trap. In a second experiment comparing Citrus Splash (semi‐gloss enamel paint) with Alpha Scents and with Macaw and Lemon Zest (both semi‐gloss enamel paints) traps at a third site, the Citrus Splash trap outperformed the Alpha Scents trap. The Citrus Splash trap did not differ statistically from Saffron Thread and Lemon Zest traps, even though it caught 51% more flies than the Lemon Zest trap. The Alpha Scents trap caught significantly more non‐target Diptera than Fluorescent Yellow, Neon Light and Citrus Splash traps at two of three trap sites where non‐target Diptera populations were relatively high and overall it appeared less selective than the Citrus Splash trap. Results suggest that sticky rectangle traps painted semi‐gloss enamel Saffron Thread or Citrus Splash with low fluorescence could be highly useful for detecting R. indifferens.  相似文献

The effect of temperature and relative humidity (RH) on the time course of the rhythmic endogenous changes of chilling resistance was studied in cotton (Gossypium hirsutum L. cv. Deltapine 50) seedlings grown under light-dark cycles of 12:12 h. The resistant phase to 5° C, 85% RH lasted during most of the dark period while to 5° C, 100% RH it was longer and extended into the last half of the light period because a transient phase advance occurred when chilling started at the middle of the light period. Seedlings acclimated by low temperature were resistant throughout the light-dark cycle. A treatment with 100% RH before chilling to acclimated seedlings introduced a sensitive phase that corresponded to that of non-acclimated seedlings. In non-acclimated seedlings, this treatment decreased the resistance but the basic pattern of the rhythm was sustained. We examined the effects of pinealectomy and blinding (bilateral ocular enucleation) on the circadian locomotor activity rhythm in the Japanese newt, Cynops pyrrhogaster. The pinealectomized newts were entrained to a light-dark cycle of 12 h light and 12 h darkness. After transfer to constant darkness they showed residual rhythmicity for at least several days which was gradually disrupted in prolonged constant darkness. Blinded newts were also entrained to a 12 h light/12 h dark cycle. In subsequent constant darkness they showed free-running rhythms of locomotor activity. However, the freerunning periods noticeably increased compared with those observed in the previous period of constant darkness before blinding. In blinded newts entrained to the light/dark cycle the activity rhythms were gradually disrupted after pinealectomy even in the presence of the light/dark cycle. The spatial distribution of catches of male European pine sawfly,Neodiprion sertifer (Geoffr.) (Hymenoptera, Diprionidae), was studied using different pheromone trap arrays. In hexagonal trap groups, trap interference was evident by reduced catch in the central of seven traps spaced 10 to 40 m apart. When the trap spacings were either 5 or 80 m no significant reduction could be shown. The interaction was more pronounced for strong (100 μg) pheromone ([2S, 3S, 7S]-3,7-dimethyl-2-pentadecanyl acetate) lures than for weak ones (10 μg). Similarly, the inner traps caught less than the outer traps in grids of 6×6 traps spaced 20 or 50 m apart. Mark-release-recapture experiments in the grid array confirmed that a majority of the males originated from outside the trap group and were caught in the first trap they encountered. There were no differences between catches in downwind, crosswind and upwind traps. Thus, no ‘overshooting’ was evident during average conditions, i.e. the sawflies did not divert from the trap initially attracting them by flying to the upwind trap. However, the proportion caught in the upwind traps increased with increasing wind velocity, suggesting more overlapping pheromone plumes at higher wind speeds. Also, the recapture rate of released males increased with increasing wind velocity up to a daily average of 3.1 m/s at 2m, indicating that sawflies more easily find the pheromone source, probably due to a more stable pheromone puff trajectory during higher wind velocities. The application of pheromone traps in e.g. monitoring studies is discussed and a distance of at least 50 m between the traps with strong lures used in the present study is recommended to avoid trap interaction.  相似文献

Heavy trap formation by Arthrobotrys oligospora in liquid culture   总被引:1,自引:0,他引:1  
Abstract The nematode-trapping fungus Arthrobotrys oligospora was grown in liquid culture in modified separatory funnels with vigorous air bubbling. The growth medium was a dilute soya peptone broth supplemented with a trap-inducing substance. The dipeptide l -phenylalanyl- l -valine or its constituents phenylalanine and valine were used as inducers of trap formation. Trap formation started within 2 days after inoculation and the traps increased in number and size during a 7-day period. The traps formed in liquid culture were fully functional in trapping nematodes. At the ultrastructural level they were characterized by the presence of many electron-dense microbodies similar to those in trap cells grown on solid media. Biomass increase, amounting to 6–7 mg dry weight · ml−1, and trap formation were highly synchronized. The mycelial mass was homogeneously dispersed in the growth vessel during the entire growth period. Heavy trap formation in liquid culture by this fungus has not been reported previously.  相似文献

We have studied the fate of electron dense microbodies in nematode-trapping organs (traps) of the fungus A. oligospora during the initial hours following nematode capture. The interaction studies were performed with isolated traps which had captured a nematode under conditions where the fungal cells had no access to external energy sources. Video enhanced contrast microscopy showed that under these conditions the number of dense bodies present in the trap cell that formed the penetration tube, rapidly decreased. During subsequent penetration and development of the infection bulb this decrease continued while at this time common cell organelles such as mitochondria and vacuoles were formed. This was confirmed by electron microscopy which also revealed that the dense bodies were degraded by means of an autophagic process. The organelles were degraded individually and finally turned into compartments which, based on ultrastructural criteria, were considered vacuoles. Fusion of such vacuoles into larger organelles frequently occurred. The degradation process was initiated early in the interaction since initial stages were already evident within 15 min after capture. Generally it took 1–2 h before the infection bulb had fully developed and trophic hyphae formation started. During this time the original trap cell, characterized by numerous dense bodies, was transformed into an active vegetative hyphal cell containing typical cell organelles such as nuclei, mitochondria, a strongly proliferated endoplasmic reticulum, vacuoles and normal microbodies but lacked dense bodies. This disappearance of dense bodies was confined to the cell that penetrated the nematode and—less frequently—its two neighbouring cells in the hyphal loop. In the other cells, constituting the trap, the dense bodies remained unaffected. As will be discussed, the present results support our current view that traps of A. oligospora contribute to the survival of the organism in its natural environment.  相似文献

Summary Locomotor activity of the river lamprey, Lampetra japonica, was investigated under a light-dark (LD 1212) cycle and under continuous dark conditions. Intact lampreys were entrained to the light:dark cycle. They were active mainly in the early half of the dark period and inactive in light period. The light:dark entrainment continued in 72.7% of lampreys after the removal of bilateral eyes, but additional pinealectomy made the entrainment disappear in all lampreys. When lampreys were pinealectomized with their eyes intact, light: dark entrainment was abolished in most cases. The results indicate that the pineal organ of the lamprey is a photoreceptive organ responsible for synchronizing locomotor activity to LD cycle. Under continuous dark conditions, the locomotor activity began to free-run with a period of 21.3 ± 0.9 h (mean ± SD, n = 53). This circadian rhythmicity was not affected by the removal of lateral eyes but was abolished by pinealectomy. Summary The ability of social stimuli to act as entraining agents of circadian rhythms was investigated in golden hamsters (Mesocricetus auratus). In a first experiment, pairs of male hamsters (one of them enucleated and the other intact) were maintained under a light-dark (LD) cycle with a period of 23.3 h. Running-wheel activity was recorded to determine the effect of social interaction on the free-running circadian rhythm of activity. In several pairs, general activity and body temperature were also recorded. In all pairs the intact animals entrained to the LD cycle, whereas the activity rhythms of the enucleated animals free-ran with periods of approximately 24 h and showed no apparent sign of synchronization or relative coordination with the other member of the pair. In a second experiment, male hamsters maintained in constant darkness received pulses of social interaction, which have been reported to induce phase shifts of the activity rhythm. Consistent phase shifts in the running-wheel activity rhythm were not induced by the social pulses in our experiment. In contrast to ad hoc methods of developing traps for pest monitoring systems, a systematic approach using direct observation of animals allowed a greater understanding of the reasons why trap catch was significantly different in traps of basically similar design. The information gained using this approach could then be used to guide the further development of the trap. The same feature on two related designs of cockroach trap was varied: the slope of the ramp leading into the trap was either 60°, 30°, or 0°. The 30° ramp version of both traps caught significantly more Blattella germanica (L.) (Dictyoptera: Blattellidae). The 60° and 0° ramp versions both caught equal, lower, numbers but observation showed that these net catches were achieved by quite different means; few insects entered over the 60° ramps but none escaped, whereas all entered over the 0° ramps but half escaped. Similar approaches could be applied to other insect-trap systems.  相似文献

Isolated traps from three aquatic Utricularia species were monitored over the course of 1-2 days to verify spontaneous firings (without any mechanical stimulation) and describe their basic characteristics. Isolated traps of three Utricularia species were initially fired by mechanical stimulation and allowed to naturally reset within a period of 24-48 h. Within this resting period, spontaneous firings were found in the traps of all species and in two trap age categories of U. vulgaris. The timing of spontaneous firings was found to be irregular. Spontaneous firings ranged between 0.29 and 2.4 during the 24-h resting period and the mean time between two spontaneous firings was highly variable within each species (319-891 min). There was no quantitative difference between spontaneous and mechanically stimulated firings of the traps. Spontaneous firings could explain how phytoplankton or detritus enters traps even when no prey species are present.  相似文献

Young adult male rats, maintained either in an LD 12: 12 or in continuous illumination (LL) for one week, were given a single injection of 25 μg melatonin/100 g body wt or ethanolic-saline (control) at 17.00 h. Animals from each group were sacrificed at 11.00 h on the following day. The activity of two important steroidogenic enzymes, 17β-hydroxysteroid dehydrogenase (17β-HSD) and Δ5-3β-hydroxysteroid dehydrogenase (Δ5-3β-HSD), and serum concentrations of testosterone, were measured following highly specific and sensitive spectrophotometric techniques and RIA, respectively. A significant decrease in the activity of both the steroidogenic enzymes was noted in the testes of melatonin-treated rats maintained under normal light-dark schedules, but this response was found to be lacking in the LL rats. However, no significant changes in the level of serum testosterone were noted in either group of melatonin-treated rats from the values in respective groups of ethanolic saline-administered LD and LL rats. Exposure of ethanolic saline-injected rats to continuous light also did not cause any change in the steroidogenic activity of the testis from those in LD rats. The study indicates that continuous light as such does not affect the endocrine function of testis but abolishes suppressive effects of melatonin on the steroidogenic activity of the testis in rat.  相似文献

A potent male attractant of sweetpotato weevil helps in monitoring and control of sweetpotato weevil in many production areas around the world. At present, it has not been used in Malaysia. Cost of the components of a trapping system is a major constraint in the adoption of male lure‐baited trapping by growers in Malaysia. Seven field trapping experiments were conducted from February 2013 to November 2015 as part of an effort to develop a simple, easy to construct, cost‐effective and efficient sex pheromone‐baited trap acceptable for use by farmers in Malaysia for monitoring and control of sweetpotato weevil (Cylas formicarius Fabricius). Overall, sweetpotato weevil trap catch was significantly affected by the number of windows in the trap, the killing agent used in the trap and the position of the trap relative to sweetpotato foliage, while trap size and trap colour did not significantly affect trap catch. Trap catch was best in plastic pole traps made from polyethylene terephthalate, with four window opening to facilitate weevil entry, with detergent solution as a killing agent and with the trap positioned from 0 to 40 cm above the crop canopy level. In a comparison study with commercial trap designs, sex pheromone‐baited plastic pole traps caught 60%–78% more weevils than were caught in sex pheromone‐baited delta traps, wing traps or unitraps. Optimization of trap characteristics is important for improving the performance of pheromone‐baited traps for use in population monitoring or mass‐trapping efforts to minimize crop damage by sweetpotato weevil infestation.  相似文献
