The microbial production, by the genus Rhizopus, of a gaseous saturated-hydrocarbon mixture was studied under aerobic conditions. Rhizopus strains, comprising 13 strains of 9 species, were tested as to their ability to produce a gaseous hydrocarbon mixture. Except for one strain, all the strains tested produced more than two kinds of gaseous hydrocarbons simultaneously when grown in nutrient broth containing glucose. Rhizopus japonicus IFO 4758 was selected as being typical of these producers of mixed gaseous hydrocarbons. When this organism was cultivated in a synthetic medium supplemented with l-cysteine under aerobic conditions, the maximum production of the total gaseous hydrocarbon mixture reached ca. 10 nl/ml culture broth/hr. The gaseous hydrocarbon mixture produced was composed mainly of paraffin hydrocarbons, i.e., ca. 74% pentane, ca. 16% propane and a trace amount of methane. The ratios of saturated to unsaturated, and even to odd number hydrocarbons produced by this fungus were 95 : 5 and 90: 10, respectively. The biosynthetic pathways for the production of these gases are discussed in comparison with the biosynthetic pathways for ethylene and isobutene in microorganisms.  相似文献

The iturinic antibiotics, which contain long chain β-amino acids, are produced by Bacillus subtilis. Screening these strains for the presence of a possible precursor of the iturinic antibiotics, we isolated a lipopeptide containing β-hydroxy fatty acids. The structure of this compound was studied and it appears to be identical or structurally very similar to surfactin. The carbon chain of its β-hydroxy fatty acids was n C16, iso C16, iso C15 or anteiso C15. The percentages of each β-hydroxy fatty acids varied according to the strain producing iturinic antibiotics and were influenced by addition of branched-chain α-amino acids to the culture medium. These results demonstrate for the first time that iso C14 β-hydroxy fatty acid is a constituent present in such a surfactin like lipopeptide. Besides, the presence of radioactive β-hydroxy fatty acids in the phospholipids when the strains were grown in the presence of sodium [14C]acetate seems also characterize the different strains producing iturinic antibiotics.  相似文献

One species of hydrocarbon utilizing bacteria was isolated from soil. This strain was named as Achromobacter petrophilum No. 4017. This bacterial species utilizes normal hydrocarbons with carbon chains of nC10 to nC18, but does not utilize glucose or other carbohydrates. Achromobacter petrophilum forms small amounts of green-yellow, green-blue and violet fluorescent compounds in the medium containing n-hexadecane (nC16) as a carbon source. The mutant strain, No. 4510, which requires hypoxanthine and thiamine for growth, was obtained from Achromobacter petrophilum No. 4017 by ultraviolet irradiation and formed considerable amounts of green-yellow fluorescent compound by the addition of guanine to the n-hexadecane medium. This fluorescent compound was crystallized from culture broth.  相似文献

When cultivating different yeast genera on the mixture of C11−C23 n-alkanes with predominant C16 and C17 content C16 and C17 fatty acids predominated in the synthetized cell fat. Besides these C11∶0, C15∶0 and C18∶1 fatty acids were also present in higher quantities in all strains tested. This conclusion is in agreement with results of the residualn-alkanes estimation where during the fermentation most of the C16 and C17 hydrocarbons decreased from the original hydrocarbon mixture. From these experiments it can be provisionally concluded that the qualitative composition of the cell fat is not dependent on the yeast strain investigated, the amount of fat on the dry weight basis depends on the strain.  相似文献

The carbon source markedly influenced the qualitative and quantitative composition of cellular hydrocarbons in Cladosporium resinae. Total lipid and hydrocarbon content was greater in cells grown on n-alkanes than in cells grown on glucose or glutamic acid. Glucose-grown cells contained a spectrum of aliphatic hydrocarbons from C7 to C36; pristane and n-hexadecane comprised 98% of the total. Cells grown on glutamic acid contained C7 to C23 hydrocarbons; n-tridecane, n-tetradecane, n-hexadecane, and pristane made up 74% of the total. n-Decane-grown cells yielded C8 to C32 compounds, and n-hexadecane (96%) was the major hydrocarbon. Cells grown on individual n-alkanes from C11 to C15 all contained C11 to C28 hydrocarbons, and cells grown on n-hexadecane contained C11 to C32 hydrocarbons. In n-undecane-grown cells, n-hexadecane and pristane made up 92% of the total, but in cells grown on C12 to C16 n-alkanes the major cellular hydrocarbon was the one on which the cells were grown. This suggests that cells cultured on n-alkanes of C12 or longer accumulate n-alkanes prior to oxidizing them.  相似文献

Anaerobic oxidation of alkanes by newly isolated denitrifying bacteria   总被引:11,自引:0,他引:11  
The capacity of denitrifying bacteria for anaerobic utilization of saturated hydrocarbons (alkanes) was investigated with n-alkanes of various chain lengths and with crude oil in enrichment cultures containing nitrate as electron acceptor. Three distinct types of denitrifying bacteria were isolated in pure culture. A strain (HxN1) with oval-shaped, nonmotile cells originated from a denitrifying enrichment culture with crude oil and was isolated with n-hexane (C6H14). Another strain (OcN1) with slender, rod-shaped, motile cells was isolated from an enrichment culture with n-octane (C8H18). A third strain (HdN1) with oval, somewhat pleomorphic, partly motile cells originated from an enrichment culture with aliphatic mineral oil and was isolated with n-hexadecane (C16H34). Cells of hexane-utilizing strain HxN1 grew homogeneously in the growth medium and did not adhere to the alkane phase, in contrast to the two other strains. Quantification of substrate consumption and cell growth revealed the capacity for complete oxidation of alkanes under strictly anoxic conditions, with nitrate being reduced to dinitrogen. Four new strains of Botryococcus braunii were isolated from Japanese waters and cultured under defined conditions. Their hydrocarbon content and composition were analyzed and compared with those of the Darwin and Berkeley strains. The Yamanaka strain produced only alkadienes characteristic of the A race, whereas the others, the Yayoi, Kawaguchi-1 and -2 strains as well as the Darwin and Berkeley strains, produced botryococcenes peculiar to the B race. The hydrocarbon content of the Yamanaka strain was 16.1 % dry weight and that of the B race strains ranged from 9.7 to 37.9%. Botryococcene composition of the Japanese strains differed from each other as well as from the Darwin and Berkeley strains. More than 50% of the hydrocarbons in the Yayoi, Darwin, and Berkeley strains were composed of C34H58, but the main components were different from one another as isomers. The Kawaguchi-1 and -2 strains did not have a high level of C34 botryococcenes, C32 ones being the main components. In these strains significant amounts of squalene-related compounds were detected.  相似文献

Production of biosurfactants by acidophilic mycobacteria was demonstrated in the course of aerobic degradation of hydrocarbons (n-tridecane, n-tricosane, n-hexacosane, model mixtures of С14–С17, С1219, and С9–С21n-alkanes, 2,2,4,4,6,8,8-heptamethylnonane, squalane, and butylcyclohexane) and their complex mixtures (hydrocarbon gas condensate, kerosene, black oil, and paraffin oil) under extremely acidic conditions (pH 2.5). When grown on hydrocarbons, the studied bacterial culture AGS10 caused a decrease in the surface and interfacial tension of the solutions (to the lowest observed values of 26.0 and 1.3 mN/m, respectively) compared to the bacteria-free control. The rheological characteristics of the culture changed only when mycobacteria were grown on hydrocarbons. Neither the medium nor the cell-free culture liquid had the surfactant activity, which indicated formation of an endotype biosurfactant by mycobacteria. Biodegradation of n-alkanes was accompanied by an increase in cell numbers, surfactant production, and changes in the hydrophobicity of bacterial cell surface and in associated phenomena of adsorption and desorption to the hydrocarbon phase. Research on AGS10 culture liquids containing the raw biosurfactant demonstrated the preservation of its activity within a broad range of pH, temperature, and salinity.  相似文献

Optimum conditions for the growth ofPseudomonas arvilla, a hydrocarbon utiliser, have been studied. The microorganism produced economic cell yield at pH 5.7 and 4% kerosene concentration. C10-C16 hydrocarbons were utilised by the strain. The growth was maximum on n-decane. Supplementation of the hydrocarbon medium with 0.5% glucose stimulated the growth. Glutamic acid 16.0 mg; leucine 9.0 mg; valine 10.0 mg; methionine 2.5 mg; arginine 2.5 mg; histidine 1.0 mg were present in 100 ml of the broth. Cell protein contained leucine 13.69%, isoleucine 4.9%, histidine 4.37%, tryptophan 2.33%, methionine 1.8% and arginine 2.70%.  相似文献

Samples of the green colonial alga Botryococcus braunii, collected from various localities, were grown in the laboratory and examined for their hydrocarbon content and morphology. Although few differences appeared between the ultrastructures of the samples, the nature of their hydrocarbons, which remains unchanged at any stage of growth, allows the distinction of two physiological races viz algae producing odd-numbered unbranched alkadienes and trienes (C25C31) (the A race) and those producing polymethylated triterpenes CnH2n-10 (C30C37), the botryococcenes (the B race). In laboratory culture, the hydrocarbon content of these new strains is very high, from 30 to 60% of the dry biomass. For the two races the greatest hydrocarbon productivity takes place during the active growth phase. The important variability observed in botryococcene distribution could originate both from genetic and environmental factors.  相似文献

Streptomyces strains isolated from the Kuwait Burgan oil field were defined as S. griseoflavus, S. parvus, and S. plicatus utilised n-hexadecane, n-octadecane (purified fractions of mineral oil), kerosene, and crude oil as sole carbon and energy sources. The strains were incubated with n-alkanes and increase of the fatty acid content with chain length equivalent to the employed n-alkanes was observed. Signal transducing GTP-binding proteins (GBPs) play an important role in n-alkane uptake in streptomycetes. Specific activators of GBPs increased the uptake of hydrocarbons. Using the hydrophobic fluorescent dye diphenylhexatrien (DPH) as a probe, it was found that the microviscosity of the hydrophobic inner region of the cellular membrane is significantly lower in hydrocarbon utilisers than in non-utilisers. This difference probably reflects differences in the fatty acid composition of the strains. When cultures were grown in n-alkane containing media, electron microscopy revealed that the hydrocarbon utilisers showed less-electron dense areas as inclusions in the cytoplasm. Soil samples inoculated with Streptomyces strains eliminated hydrocarbons much faster than those not containing these strains, serving as control. When inorganic medium was supplied with n-hexadecane-1-14C as sole carbon and energy source, radioactive CO2 was detected. Since streptomycetes have not been used until now for oil elimination, though they are known as abundant soil bacteria tolerating extreme conditions, their possible use for bioremediation of hydrocarbon contaminated soils is discussed.  相似文献

The production of microbial cell substances from hydrocarbons has been attracting attention of people for many years. Production of bacterial cell from hydrocarbons is disadvantageous because of the difficulty in separating cell from the broth.

We have tested hydrocarbon-utilizing yeasts isolated from garden soil for cell production. The effect of medium composition on yeast growth and the utilization of individual hydrocarbon by yeast, strain Y-3, were investigated.

As a nitrogen source, urea was more effective than ammonium nitrate. When a very smal! amount of corn steep liquor was added, yeast growth was very improved. Aliphatic series of hydrocarbon lower than C9 were not or very slightly assimilated by this yeast.

Generally speaking, series of even-number hydrocarbons were more effective than those of odd-number hydrocarbons.

We found that the yeast Y-3 strain reported in the previous paper1) has a diterminal oxidation system of hydrocarbon.

This yeast capable of growing in mineral-salts solution with hydrocarbons as sole source of carbon produced a series of dioic acid from n-undecane. These acids are 1,11-undecane dioic acid, 1,9-nonane dioic acid (azelaic acid), 1,7-heptane dioic acid (pimelic acid) and 1,5-pentane dioic acid (glutaric acid). 1,10-Decane dioic acid (sebacic acid) was also isolated from n-decane cultures.

Azelaic acid was partially transformed into pimelic acid and glutaric acid by treating it with resting cells of this yeast.

1,11-Undecane dioic was also transformed into azelaic acid pimelic acid, and glutaric acid by the same treatment as described above.  相似文献

The production of microbial cell substances from hydrocarbons has become the object of commercial attention. We have tested hydrocarbon-utilizing bacterial strains for cell production, and found out one strain of high cell yield, which is very similar to Pseudomonas aeruginosa. The effect of medium composition on cell yield and the utilization of individual hydrocarbons by this strain were investigated. Sixty percent of the added kerosene was converted into cell materials in the following medium of optimum composition: kerosene 2.5%, urea 0.13%, dipotassium phosphate 0.25%, magnesium sulfate 7 aq. 0.1% and tap water. Aliphatic series lower than C10, aromatic and naphthenic hydrocarbons were not or very slightly assimilated. However, aliphatic members higher than C12 were utilized with increasing case. Especially, n-docosane and octadecene-1 were utilized very effectively, and 70% of them were converted into cell substances.  相似文献

The production of labelled aliphatic hydrocarbons in Anacystis montana and Botryococcus braunii has been studied using Na2CO3 [14C] as a carbon source. The major hydrocarbon produced by A. montana is pentadecane (ca 93%) accompanied by a pentadecene (ca 4%) and other hydrocarbons in the range C13-C17. Long chain (C21-C 33) hydrocarbons could not be detected in this organism. The variety of unsaturated hydrocarbons (C25-C31) previously reported in Botryococcus braunii is confirmed and contrasts with the synthesis of unsaturated C17 hydrocarbons only, in axenic cultures prepared from single cell isolates of this colonial alga.  相似文献

The long-chain saturated and mono-unsaturated hydrocarbon content of the juice sacs of five mandarin cultivars (Mediterranean, Honey, Wilking, Kinnow, King) were examined. Normal homologues accounted for more than 47% of the saturated and more than 75% of the monoene hydrocarbons. In the saturated fraction the major hydrocarbon was n-C25 but in the monoene fraction n-C25 predominated in Kinnow and King while C29 predominated in Mediterranean, Honey and Wilking. All five cultivars could be differentiated from each other and from other citrus species by their hydrocarbon patterns. The noticeably high normal/iso ratios of saturated C23 and C25 hydrocarbons previously shown to be characteristic of mandarin species, Citrus unshiu and C. reticulata, were also found in C. nobilis and C. deliciosa.  相似文献

分子生态学是研究生命系统与环境系统相互作用机理及其分子机制的科学,可以从宏观和微观结合的角度真实反映生态现象的本质。简述产烃布朗葡萄藻形态与化学种等生理生态特征的基础上,综述了近年来国内外布朗葡萄藻分子生态学研究的新进展,主要包括分子系统发育学及其与化学种、基因组、地理来源等之间的关系。经典分类学上,关于布朗葡萄藻属于绿藻门(Chlorophyta)还是黄藻门(Xanthophyta)存在争议,而基于18S核糖体核糖核酸(18S ribosomal ribonucleic acid,18S rRNA)序列的分子系统发育学研究结果将布朗葡萄藻界定为绿藻门、共球藻纲(Trebouxiophyceae)。依据藻株的产烃种类和化学结构特征,可将布朗葡萄藻划分为A、B和L 3个化学种,而布朗葡萄藻的分子系统学进化关系与化学种间高度统一。在基因组大小上,位于同一大亚聚群中的化学种B与L间却存在明显差异,而进化关系较远的化学种B与A间则更相近。不同地理来源布朗葡萄藻的18S rRNA序列和内部转录间隔区(internal transcribed spacer,ITS)多态性较高,提示不同地缘藻株间存有较高的遗传多样性。探讨了布朗葡萄藻分子生态学研究尚待解决的问题,并对今后相关研究做了展望。  相似文献

In this study, the yeast strain P5 isolated from a mangrove system was identified to be a strain of Aureobasidium pullulans var. melanogenum and was found to be able to secrete a large amount of heavy oil into medium. After optimization of the medium for heavy oil production and cell growth by the yeast strain P5, it was found that 120.0 g/l of glucose and 0.1 % corn steep liquor were the most suitable for heavy oil production. During 10-l fermentation, the yeast strain P5 produced 32.5 g/l of heavy oil and cell mass was 23.0 g/l within 168 h. The secreted heavy oils contained 66.15 % of the long-chain n-alkanes and 26.4 % of the fatty acids, whereas the compositions of the fatty acids in the yeast cells were only C16:0 (21.2 %), C16:1(2.8 %), C18:0 (2.9 %), C18:1 (39.8 %), and C18:2 (33.3 %). We think that the secreted heavy oils may be used as a new source of petroleum in marine environments. This is the first report of yeast cells which can secrete the long-chain n-alkanes.  相似文献

The analysis of fatty acids and hydrocarbons in the sporophytes of three Lycopodium species has revealed a characteristic distribution of C16 and C18 acids. The hydrocarbon fraction of the lipids contain a homologous series of monounsaturated alkenes in the C17C30 range with an even to odd preference. Maxima at both C17 and C27 among the n-alkanes reveals similarities both to the distribution of hydrocarbons in other plant groups. The production of spores and their inclusion with one sporophyte does not alter the fatty acid pattern but does decrease the alkene concentration and modifies the alkane distribution, shifting both maxima. The presence of pristane and phytane in all specimens, the dual maxima of alkanes and slight odd to even preference of alkanes is noteworthy in that these characteristics are possessed by geological deposits derived from Lycopodium ancestors.  相似文献

The structure and composition of the cutin monomers from the flower petals of Vicia faba were determined by hydrogenolysis (LiAlH4) or deuterolysis (LiAlD4) followed by thin layer chromatography and combined gas-liquid chromatography and mass spectrometry. The major components were 10, 16-dihydroxyhexadecanoic acid (79.8%), 9, 16-dihydroxyhexadecanoic acid (4.2%), 16-hydroxyhexadecanoic acid (4.2%), 18-hydroxyoctadecanoic acid (1.6%), and hexadecanoic acid (2.4%). These results show that flower petal cutin is very similar to leaf cutin of V. faba. Developing petals readily incorporated exogenous [1-14C]palmitic acid into cutin. Direct conversion of the exogeneous acid into 16-hydroxyhexadecanoic acid, 10, 16-dihydroxy-, and 9, 16-dihydroxyhexadecanoic acid was demonstrated by radio gas-liquid chromatography of their chemical degradation products. About 1% of the exogenous [1-14C]palmitic acid was incorporated into C27, C29, and C31n-alkanes, which were identified by combined gas-liquid chromatography and mass spectrometry as the major components of the hydrocarbons of V. faba flowers. The radioactivity distribution among these three alkanes (C27, 15%; C29, 48%; C31, 38%) was similar to the per cent composition of the alkanes (C27, 12%; C29, 43%; C31, 44%). [1-14C]Stearic acid was also incorporated into C27, C29, and C31n-alkanes in good yield (3%). Trichloroacetate, which has been postulated to be an inhibitor of fatty acid elongation, inhibited the conversion of [1-14C]stearic acid to alkanes, and the inhibition was greatest for the longer alkanes. Developing flower petals also incorporated exogenous C28, C30, and C32 acids into alkanes in 0.5% to 5% yields. [G-3H]n-octacosanoic acid (C28) was incorporated into C27, C29, and C31n-alkanes. [G-3H]n-triacontanoic acid (C30) was incorporated mainly into C29 and C31 alkanes, whereas [9, 10, 11-3H]n-dotriacontanoic acid (C32) was converted mainly to C31 alkane. Trichloroacetate inhibited the conversion of the exogenous acids into alkanes with carbon chains longer than the exogenous acid, and at the same time increased the amount of the direct decarboxylation product formed. These results clearly demonstrate direct decarboxylation as well as elongation and decarboxylation of exogenous fatty acids, and thus constitute the most direct evidence thus far obtained for an elongation-decarboxylation mechanism for the biosynthesis of alkanes.  相似文献

Natural relationships, improvement of anaerobic growth on hydrocarbons, and properties that may provide clues to an understanding of oxygen-independent alkane metabolism were studied with two mesophilic sulfate-reducing bacteria, strains Hxd3 and Pnd3. Strain Hxd3 had been formerly isolated from an oil tank; strain Pnd3 was isolated from marine sediment. Strains Hxd3 and Pnd3 grew under strictly anoxic conditions on n-alkanes in the range of C12–C20 and C14–C17, respectively, reducing sulfate to sulfide. Both strains shared 90% 16 S rRNA sequence similarity and clustered with classified species of completely oxidizing, sulfate-reducing bacteria within the δ-subclass of Proteobacteria. Anaerobic growth on alkanes was stimulated by α-cyclodextrin, which served as a non-degradable carrier for the hydrophobic substrate. Cells of strain Hxd3 grown on hydrocarbons and α-cyclodextrin were used to study the composition of cellular fatty acids and in vivo activities. When strain Hxd3 was grown on hexadecane (C16H34), cellular fatty acids with C-odd chains were dominant. Vice versa, cultures grown on heptadecane (C17H36) contained mainly fatty acids with C-even chains. In contrast, during growth on 1-alkenes or fatty acids, a C-even substrate yielded C-even fatty acids, and a C-odd substrate yielded C-odd fatty acids. These results suggest that anaerobic degradation of alkanes by strain Hxd3 does not occur via a desaturation to the corresponding 1-alkenes, a hypothetical reaction formerly discussed in the literature. Rather an alteration of the carbon chain by a C-odd carbon unit is likely to occur during activation; one hypothetical reaction is a terminal addition of a C1 unit. In contrast, fatty acid analyses of strain Pnd3 after growth on alkanes did not indicate an alteration of the carbon chain by a C-odd carbon unit, suggesting that the initial reaction differed from that in strain Hxd3. When hexadecane-grown cells of strain Hxd3 were resuspended in medium with 1-hexadecene, an adaptation period of 2 days was observed.
