The diarrhea observed in patients which cholera is known to be related to secretion of water and electrolytes into the intestinal lumen. However, the exact mechanisms involved in these secretory processes have remained unclear. Although it is clear that purified toxin acts on epithelial cell metabolism, its activity on Na+ transport across intestinal mucosa is equivocal: reported either to prevent net Na+ absorption or to cause net secretion of Na+ from serosa to mucosa. Since total transmural Na+ fluxes across “leaky” epithelia involve very significant movement via a paracellular shunt pathway, we studied the effects of cholera toxin on the cellular and paracellular pathways of Na+ movement. Unidirectional Na+ fluxes were examined as functions of applied potential in control tissues and in tissues from the same animal treated with purified cholera toxin. Treatment of rabbit ileum in vitro with toxin stimulated the cellular component of serosa-to-mucosa Na+ flux (from 2.41 ± 0.49 μequiv./h per cm2 under control conditions to 4.71 ± 0.43 μequiv./h per cm2 after treatment with toxin, P < 0.01). The effect of cholera toxin on Na+ movement through the cells from mucosa to serosa appeared to be insignificant. Finally, a marked decrease in the Na+ permeability (P < 0.01) and no detectable significant changes in transference number for Na+ of the paracellular shunt pathway were observed following treatment with cholera toxin. These results provide direct evidence for the hypothesis that purified cholera toxin stimulates active sodium secretion but has minimal effect on sodium absorption.  相似文献

Serotonin has previously been implicated as the cause of the diarrhea associated with carcinoid syndrome and the amine has been shown by others to be an intestinal secretagogue in preparations of intestinal loops invivo. In the present paper the action of serotonin on isolated segments of rabbit ileal mucosa stripped of muscle layers was studied invitro. Serotonin (10?4M) caused an abrupt significant rise in short-circuit current (Isc) across the mucosal epithelial cell layer but this effect was transient. No change was observed in tissue conductance. In this preparation, serotonin did not alter 22Na, 36Cl or residual ion fluxes across the mucosa. High blood serotonin levels for a period of several days also did not alter ion fluxes or Isc in isolated rabbit ileum. Therefore, it is concluded that serotonin must cause its secretory activity observed invivo by some mechanism other than a direct action on epithelial cell transport mechanisms.  相似文献

A photoaffinity probe for the vitamin D-dependent chick intestinal calcium binding protein (CaBP) has been prepared by conjugation of methyl-4-azidobenzoimidate (MABI) to lactoperoxidase-125I-iodinated CaBP to yield 125I-CaBP-MABI: [3 moles MABI per mole CaBP]. After incubation invitro of 125I-CaBP-MABI (28,000 daltons) in model systems with bovine intestinal alkaline phosphatase (AP) (67,000 daltons), a UV light-dependent crosslinking occurred to yield a conjugate with a molecular weight of 95,000 (by SDS-gel electrophoresis); no crosslinking occurred with E.coli alkaline phosphatase. The formation of the 125I-CaBP-MABI-AP was found to occur only in the presence of calcium.  相似文献

Diabetes is a life threatening disease and its onset is linked with both environmental and genetic factors. Zinc metabolism gets altered during diabetes and results in many complications. The present study was designed to elucidate the effects of zinc supplementation on the biokinetics of 65Zn in whole body, liver and its biodistribution in diabetic rats. The animals were divided into four groups viz; normal control; diabetic (single intraperitoneal injection of alloxan 150 mg/kg body weight); zinc treated (227 mg/l in drinking water); and diabetic + zinc treated. To carry out biokinetics study, each rat was injected intraperitoneally with 0.74 MBq radioactivity of 65Zn following 4 weeks of different treatments and the radioactivity was determined by using a suitably shielded scintillation counter. Alloxan induced diabetic rats showed a significant decrease in both the fast (Tb1) and slow (Tb2) components of biological half-life of 65Zn which, however, were normalized in whole body (P > 0.05) following zinc supplementation. In case of liver, Tb2 component was brought back to the normal but Tb1 component was not increased significantly. The present study indicates that the paucity of zinc in the tissues of the diabetic animals was due to decreased retention of tissue zinc as evidenced by increased serum Zn, hyperzincuria and increased rate of uptake of 65Zn by the liver. Zinc supplementation caused a significant improvement in the retention of zinc in the tissues and is therefore likely to be of benefit in the treatment of diabetes.  相似文献

Alloxan diabetes in rats was found to decrease the level of phospholipids in the heart. Measurement of specific phosphatides showed that the decrease was restricted only to phosphatidylethanolamine and lysophosphatidylcholine. Study of invivo incorporation of 32Pi indicated an impairment of phosphatidylethanolamine synthesis and conversion of phosphatidylcholine into lysophosphatidyl choline in the heart of diabetic rats. Treatment of diabetic rats with insulin restored the levels of phosphatidylethanolamine and lysophosphatidylcholine and incorporation of 32Pi into these phosphatides to almost normal.  相似文献

The effect of chronic streptozotocin-induced diabetes was studied on intestinal microvillous membrane surface carbohydrate groups. After 7 weeks of diabetes, purified microvillous membranes were prepared from rat small intestine and surface galactoproteins identified by labeling with galactose oxidase/sodium boro[3H]hydride. Membrane surface sialic acid residues were labeled using the sodium metaperiodate/sodium boro[3H]hydride technique. Membranes were solubilized in SDS and protein labeling analyzed by acrylamide electrophoresis. Membranes from diabetic rats showed an 81% increase in galactoprotein labeling (P< 0.02) while labeling of sialic acid residues was unchanged. The greatest increase in galactoprotein labeling occurred in protein monomers of Mr 116 000–200 000, where there was a 155% increase in labeling (P< 0.005). These results indicate that intestinal microvillous membrane protein glycosylation is altered in chronic diabetes. This increase in surface membrane carbohydrates could explain the decreased rates of proteolytic degradation previously described for at least one microvillous protein. An increase in membrane galactose groups has also been noted in hepatocyte and kidney glomerular basement membranes, which suggests the presence of a systematic change in membrane protein glycosylation occurring as a result of the diabetic state.  相似文献

Tissue cultures grown from stem explants of three Actinidia species and a hybrid species rapidly converted N6-isopentenyladenine (i6Ade) to zeatin (io6Ade), a potent hydroxylated cytokinin. Within 24 h on 50 uM i6Ade, callus tissues of A.chinensis × arguta accumulated 83 ± 6 nmol/g io6Ade which was purified using HPLC and identified by its characteristic UV and mass spectra. Activity converting i6Ade to io6Ade was also demonstrated in stem segments from intact plants where it was low in the tip (3 cm), highest in the region corresponding to rapid leaf growth and very low in the mature stem. Root segments converted i6Ade to io6Ade almost as rapidly as the most active region of the stem while leaf petioles produced little io6Ade. Fruits of A.arguta and A.chinensis produced little or no io6Ade, respectively.  相似文献

An intestinal zinc-binding protein, induced by parenteral zinc administration, has been isolated and characterized. Based upon its elution behavior in two chromatographic systems, Zn2+/protein ratio of 5.0–5.6 gram atoms/mole, a Zn2+/SH ratio of about 3.0, paucity of both aromatic amino acids and absorbance at 280 nm, abundance of cysteic acid residues (28–31%), and low molecular weight (6,000–7,000 daltons), the protein meets the criteria for classification as a metallothionein and is more properly named zinc-thionein. Orally administered 65Zn was found to bind to intestinal zinc-thionein and thus this intracellular protein may function as a component of the mechanism responsible for mammalian zinc homeostasis at the level of intestinal absorption.  相似文献

This study was designed to determine the effect of zinc on the biological half-lives of 65Zn in whole body and liver and on distribution of 65Zn in different organs of rats following nickel toxicity. Sprague-Dawley (SD) rats received either nickel in the form NiSO4·6H2O at a dose of 800 mg/L in drinking water, zinc in the form of ZnSO4·7H2O at a dose of 227 mg/L in drinking water, and nickel plus zinc or drinking water alone for a total duration of 8 wk. All of the rats were injected with a tracer dose of 0.37 MBq 65Zn at the end of the treatment period. The effects of different treatments were studied on biological half-lives of 65Zn in whole body and liver and on the distribution of 65Zn in different organs of rats. In the present study, we have noted that nickel treatment to normal rats caused a significant decrease in the slow component (Tb2) in liver, which improved following zinc supplementation. Nickel administration to normal-diet-fed animals caused significant lowering in the percentage uptake of 65Zn values in the brain, liver, and intestine. However, the administration of zinc to nickel-treated rats improved the status of 65Zn in different organs. The Tb2 in the liver and the percentage uptake of 65Zn values elevated following zinc supplementation to nickel-treated rats.  相似文献

We investigated with an in vivo single pass perfusion technique, the effect of glucocorticoids on net magnesium and calcium absorption from the small and large intestine of suckling and adolescent rats. In control rats, rates of net magnesium and calcium absorption were several fold greater in both small and large intestinal segments of suckling rats compared to corresponding rates in segments of adolescent rats. Methylprednisolone 30 mg/kg body weight daily for three days, suppressed significantly net magnesium and calcium absorption from the small and large intestinal segments of suckling rats only. Methylprednisolone had no effect on either net magnesium or calcium absorption in adolescent rats. The mechanism(s) responsible for the observed decrease in net magnesium and calcium absorption in the suckling period by glucocorticoids are discussed.  相似文献

Vitamin D (calciferol) has been postulated [A.W. Norman, Science 149, 184 1965] to mediate its physiological effects via an inductive process which produces new RNA and proteins. This was tested directly by studying the effect of the biologically active form of calciferol, 1,25-dihydroxycholecalciferol to stimulate in vivo the pulse labeling of intestinal mucosa by 3H uridine. The maximum stimulation of RNA labeling occurs 6 hours after the intracardial administration of 325 pmoles of 1,25-dihydroxy-cholecalciferol: the first effects were apparent within 3–4 hours. Also, actinomycin D was shown to block both 1,25-dihydroxy-cholecalciferol stimulated Ca2+ transport and RNA synthesis. The chronology of events occurring after administration of 1,25-dihydroxy-cholecalciferol suggest that the primary biochemical response of intestinal mucosa to 1,25-dihydroxy-cholecalciferol is probably the initiation of RNA and protein synthesis.  相似文献

Histidine has been reported to affect body zinc status by increasing urinary zinc excretion. The effects of experimental histidinemia on distribution of65Zn in anesthetized rats were studied. Infusion ofl-histidine at a rate sufficient to raise plasma concentrations to approximately 2mm for 6h starting 48 h after a single intraperitoneal65Zn injection did not alter65Zn activities in a variety of tissues when compared with anesthetized uninfused animals. However, plasma65Zn and erythrocyte65Zn were decreased, and liver65Zn was increased. If65Zn was injected intravenously during histidine infusion, net accumulation of zinc by some tissues was increased, but uptake by others was reduced relative to uninfused animals. In all cases, however, uptake expressed relative to plasma65Zn levels was increased when allowance was made for the more rapid fall in plasma65Zn during histidine infusion. Similar infusions ofd-histidine produced quantitatively similar effects. Since enzymatic mechanisms and amino acid carriers would be expected to show stereoselectivity, such processes are unlikely to be involved in the zinc distribution changes described. The possibility of zinc transport by a hitherto unidentified carrier is discussed. These experiments confirm that histidinemia can affect zinc status, but any associated changes in urinary zinc excretion do not seem adequate to account for the tissue changes found.  相似文献

While 2-aminoanthracene and 2-aminofluorene are converted to frameshift mutagens by microsomal preparations from rat livers, the microsomes from the intestinal mucosa of the same animals, under the experimental conditions used herein, either have little such activity or lack it altogether. Cell-free extracts of the colon anaerobe Bacteroidesfragilis may exhibit such activity to varying degrees depending upon the conditions of incubation. However mixtures consisting of cell-free extracts from B. fragilis and microsomes from intestinal mucosa demonstrate significant- more than additive- activity in converting these chemicals to mutagens.  相似文献

The influence of arachidonic acid (AA) on the zinc flux rates of jejunal segments, isolated from streptozotocin-induced diabetic rats injected with saline or with insulin, was investigated using an Ussing chamber technique. Although the zinc flux rates from mucosa-to-serosa (Jms) of normal rats were inhibited by addition of 5 microM AA to the jejunal segment bathing medium (46.4 +/- 5.0 vs 32.6 +/- 4.3 nmol/hr/cm2), AA had no effect on the Jms of diabetic rats either with or without insulin treatment. Induction of diabetes also significantly reduced Jms (46.4 +/- 5.0 vs 22.1 +/- 4.9 nmol/hr/cm2), but 3 day insulin treatment (NPH 8 U/Kg/day subcutaneously) did not reverse this effect (29.2 +/- 5.1 nmol/hr/cm2). Addition of AA to the serosal side did not significantly alter the zinc flux rate from serosa-to-mucosa (Jsm) in either control, diabetic or diabetic rats treated with insulin. The net zinc absorption rate (Jnet) of jejunal segments was decreased in diabetic rats compared to controls (13.2 +/- 3.0 vs -0.7 +/- 2.1 nmol/hr/cm2), but normalization of blood glucose with 3 day insulin treatment did not increase Jnet. Addition of AA was associated with a tendency to increase zinc uptake capacity. This change reached statistical significance in insulin treated diabetic rats. Short-circuit current (Isc) for diabetic rats was increased compared to controls but addition of AA to the mucosal side bathing medium decreased Isc in all groups. The results indicate that the zinc flux rate in the small intestine of streptozotocin-induced diabetic rats is decreased, that zinc uptake capacity of the small intestine does not directly reflect the zinc flux rate across the small intestine, and that AA or one of its metabolites may play a significant role in the control of the zinc flux across the intestinal epithelium.  相似文献

Relationships between perinatal mortality, disrupted uteroplacental function and prostaglandin metabolism have been studied in Zn-deficient rats. Uterine contractility in vitro, placental blood flow in viro, and uterine and placental prostaglandin synthesis from [1?14C] arachidonic acid in vitro were investigated at day 22 of pregnancy. High amplitude uterine contractions were almost completely eliminated and utero-placental blood flow was decreased by 85% by Zn deficiency. Synthesis of [1?14C]-prostaglandin E2, F and 6-keto-F from [1?14C] arachidonic acid decreased significantly in uterine tissue but increased in placentae. These possibly inter-related effects may contribute to the high perinatal mortality observed in Zn deficiency.  相似文献

The proximal colon of the new-born pig maintains a stable short-circuit current which is partly dependent upon the presence of methionine. This interaction between methionine and short circuit current shows Michaelis- Menten knetics with a Km of 0.24 mM and a V of 27 μA·cm?2. The net flux of methionine to the serosal surface of proximal colons also shows a hyperbolic relation to the external concentration of methionine (Km 0.38 mM; V 10.4 nmol·cm?2·min?1). The proximal colon concentrates methionine within its epithelium giving a mucosal to medium ratio of 11.2 ± 1.9 (90 min incubation in 1 mM methionine).The ability of the colon to transport methionine across and concentrate methionine within its mucosa is maintained for at least 24 h after birth. Colonic transport of amino acids could be physiologically important in the pig, where the immediate post-natal transfer of immune globulins has been shown to cause a temporary inhibition of normal intestinal function.  相似文献

Nuclear binding of corticosteroids in fish tissues was investigated in two target organs of the trout Salmo gairdnerii irideus: the liver and the intestinal mucosa. Incubation of intact nuclei with [3H]-cortisol or [3H]-dexamethasone failed to demonstrate high-affinity binding of these steroids to proteins. Exchange assay of [3H]-cortisol in high-salt nuclear extracts indicated an association constant Ka = 1.9 × 104 M?1 for intestinal mucosa and 2.1 × 104 M?1 for liver. In sea water-adapted trout, the association constant remained the same as in fresh water.These results extend previous observations obtained on the cytosol which showed that no high-affinity receptors could be disclosed in fish tissues using these two corticosteroids.  相似文献
