The genetic transformation of plants is an important biotechnological tool used for crop improvement for many decades. The present study was focussed to investigate various factors affecting genetic transformation of potato cultivar ‘Kufri Chipsona 1’. It was observed that explants pre-cultured for 2 days on MS2 medium (MS medium containing 10 µM silver nitrate, 10 µM BA, 15 µM GA3), injured with a surgical blade and co-cultivated with Agrobacterium tumefaciens strain EHA105 [O.D600 (0.6)] for 2 days results in maximum transient β-glucuronidase (GUS) expression. The addition of 100 µM acetosyringone in MS2 medium also increased rate of transient GUS expression in both the explants. Clumps of putative transgenic shoots were regenerated using the optimised culture conditions from leaf and internodal explants. The stable integration of T-DNA was established using histochemical staining for GUS and amplification of DNA fragment specific to nptII and uidA genes. Within the clumps, around 67.85% of shoots showed uniform GUS expression in all the tissues and about 32.15% shoots show intermittent GUS expression establishing chimeric nature. Uniform GUS staining of the tissue was used as initial marker of non-chimeric transgenic shoots. Quantitative expression of nptII transgene was found to be directly proportional to uniformity of GUS staining in transgenic shoots. The present investigation indicated that manipulation of culture conditions and the medium composition may help to get transgenic shoots with uniform expression of transgene in all the tissues of potato cultivar ‘Kufri Chipsona 1’.  相似文献

The goal of this research is to enhance our knowledge of thecontributions of doliolids to the planktonic community as consumersand secondary producers. The objectives are to quantify feedingand growth rates of Dolioletta gegenbauri gonozooids at fourfood concentrations and four temperatures in order to determinetheir impact as grazers throughout the water column. Althoughdoliolids are abundant in numerous regions of the coastal ocean,and are considered to be major planktonic grazers, data on ratesof feeding and growth are scarce. Laboratory experiments wereconducted at 16.5, 20, 23.5 and 26.5°C to quantify removalof a 50:50 volumetric concentration of Thalassiosira weissflogiiand Rhodomonas sp. at four different food concentrations of20, 60, 160 and 390 µg C l–1. Results from theseexperiments suggest that clearance rates are similar at concentrationsfrom 20 to 60 µg C l –1, and decrease as the foodconcentrations increase to 160 and 390 µg C l –1.The ingestion rates increase over a range of phytoplankton concentrationsfrom 20 to 160 µg C l –1, then decrease when abnormallyhigh concentrations of 390 µg C l –1 are offered.Clearance and ingestion rates increase as temperature increasesfrom 16.5 to 26.5°C. The exponential growth rates rangefrom k = 0.2–0.7, with the lowest rates occurring at thehighest food concentration. Growth rates increase with increasingtemperature from K = 0.1–0.3 at 16.5°C to 0.45–0.7at 26.5°C. In each case, the small- and medium-sized zooidshad higher growth rates than the larger gonozooids. These resultssuggest that doliolid feeding and growth rates are a functionof environmental food concentrations and temperatures, and implythat they can be important consumers in a changing neritic environment.  相似文献

Optimal decision-analytic designs are deterministic. Such designsare appropriately criticized in the context of clinical trialsbecause they are subject to assignment bias. On the other hand,balanced randomized designs may assign an excessive number ofpatients to a treatment arm that is performing relatively poorly.We propose a compromise between these two extremes, one thatachieves some of the good characteristics of both. We introducea constrained optimal adaptive design for a fully sequentialrandomized clinical trial with k arms and n patients. An r-designis one for which, at each allocation, each arm has probabilityat least r of being chosen, 0 r 1/k. An optimal design amongall r-designs is called r-optimal. An r1-design is also an r2-designif r1 r2. A design without constraint is the special case r = 0and a balanced randomized design is the special case r = 1/k.The optimization criterion is to maximize the expected overallutility in a Bayesian decision-analytic approach, where utilityis the sum over the utilities for individual patients over a‘patient horizon’ N. We prove analytically thatthere exists an r-optimal design such that each patient is assignedto a particular one of the arms with probability 1 – (k – 1)r,and to the remaining arms with probability r. We also show thatthe balanced design is asymptotically r-optimal for any givenr, 0 r < 1/k, as N/n  . This implies that everyr-optimal design is asymptotically optimal without constraint.Numerical computations using backward induction for k = 2arms show that, in general, this asymptotic optimality featurefor r-optimal designs can be accomplished with moderate trialsize n if the patient horizon N is large relative to n. We alsoshow that, in a trial with an r-optimal design, r < 1/2,fewer patients are assigned to an inferior arm than when followinga balanced design, even for r-optimal designs having the samestatistical power as a balanced design. Microsphere and morphometric techniques were used to investigate any circulatory changes that accompany secretion by the salt glands of hatchling Chelonia mydas. Salt glands were activated by a salt load of 27.0 mmol NaCl kg body mass (BM)−1, resulting in a mean sodium secretion rate of 4.14 ± 0.11 mmol Na kg BM−1 h−1 for a single gland. Microsphere entrapment was approximately 160–180 times greater in the active salt gland than the inactive gland, inferring a similar change in blood flow through salt gland capillaries. The concentration of microspheres trapped in the salt gland was significantly correlated with the rate of tear production (ml kg BM−1 h−1) and the total rate of sodium secretion (mmol Na kg BM−1 h−1) but not with tear sodium concentration (mmol Na l−1). Adrenaline (500 μg kg BM−1) inhibited tear production within 2 min and reduced microsphere entrapment by approximately 95% compared with active glands. The volume of filled blood vessels increased from 0.03 ± 0.01% of secretory lobe volume in inactive salt gland sections to 0.70 ± 0.11% in active gland sections. The results demonstrate that capillary blood flow in the salt gland of C. mydas can regulate the activity of the gland as a whole. Accepted: 12 July 2000  相似文献

Extracellular radicals produced by Trametes versicolor under quinone redox cycling conditions can degrade a large variety of pollutant compounds, including trichloroethylene (TCE). This study investigated the effect of the agitation speed and the gas–liquid phase volume ratio on TCE degradation using central composite design (CCD) methodology for a future scale-up to a reactor system. The agitation speed ranged from 90 to 200 rpm, and the volume ratio ranged from 0.5 to 4.4. The results demonstrated the important and positive effect of the agitation speed and an interaction between the two factors on TCE degradation. Although the volume ratio did not have a significant effect if the agitation speed value was between 160 and 200 rpm, at lower speed values, the specific pollutant degradation was clearly more extensive at low volume ratios than at high volume ratios. The fitted response surface was validated by performing an experiment using the parameter combination in the model that maximised TCE degradation. The results of the experiments carried out using different biomass concentrations demonstrated that the biomass concentration had a positive effect on pollutant degradation if the amount of biomass present was lower than 1.6 g dry weight l−1. The results show that the maximum TCE degradation was obtained at the highest speed (200 rpm), gas–liquid phase volume ratio (4.4), and a biomass concentration of 1.6 g dry weight l−1.  相似文献

Food concentration and temperature has an effect on phorozooidreproductive longevity, size of gonozooids released, and theamount of carbon released asexually. Doliolid reproduction ratesare a function of environmental food concentrations and temperatures,and the results imply that doliolids can be important secondaryproducers in the neritic environment.  相似文献

To ascertain the feeding habits of benthic juvenile yellowfin goby Acanthogobius flavimanus, the gut contents of 599 specimens (15–41 mm in standard length, SL), collected on a tidal mudflat in the Tama River estuary throughout the diel cycle, were examined. The major prey items changed from harpacticoid copepods to errant and sedentary polychaetes at ca. 20 mm SL. Prey width increased with fish size. Fish of 26–28 mm SL fed mainly from sunset to morning, with highest feeding intensity during twilight hours and/or high tide. Based on the gut evacuation rate estimated from a forced feeding experiment in the laboratory and data for the diel change of mean gut-content volume in the field, the daily ration of juvenile yellowfin goby (26–28 mm SL) was calculated to be 13.8 mm3 fish−1 day−1. This volume is approximately equivalent to 3.9 individuals of the errant polychaete Ceratonereis erythraeensis (9.7 mm in body length, BL) or 8.1 individuals of the sedentary polychaete Prionospio japonica (14.8 mm BL), both species occurring abundantly on the mudflat during the study.  相似文献

Water exchange, temperature tolerance and oxygen consumptionof the snail, Trigonephrus sp., from the southern Namib desertof Namibia were examined and related to activity. At 25°Cand 15% R.H. mean water loss and food and water uptake were5.95 mg. day–1 and 630 mg.day–1, respectively. Bodytemperature tracked sand temperature. Snails tolerated sandtemperatures as high as 45°C. Mean ± S.D. oxygenconsumption rates were 32.0 ± 2.94 µlO2.g totalbody mass–1.h–1 at 15°C, when the snails wereactive, and 11.27 µlO2.g total body mass–1.h–1at 25°C, when the snails were inactive. These values are2-6 times lower than those recorded for the similarly sizedmesic snail, Helix aspersa. Activity experiments indicated thatlow ambient temperatures and high humidities were favoured bythe snails. This, together with the burying behaviour of thesesnails during high temperatures, suggests that they limit stressby restricting activity to physiologically-favourable periods,even though more-extreme conditions may be tolerated.  Stand structure and leaf area distribution of a laurel forest in the Agua García mountains of Tenerife are described. The site is situated at 820 m a.s.l., faces NNE, and has a humid mediterranean climate. Summer droughts are mitigated by relatively high air humidity and clouds. The natural mixed hardwood forest is composed of six major tree species: Laurus azorica (Seub.) Franco, Persea indica (L.) Spreng, Myrica faya Ait., Erica arborea L. and two species of Ilex (I. platyphylla Webb & Berth. and I. canariensis Poivet.). The experimental stand had a density of 1693 trees ha – 1, a basal area of 33.7 m2ha – 1, and a cumulated volume of above-ground parts of trees of 231 m3 ha – 1 with a corresponding dry mass of 204 ton ha – 1. Diameters at breast height ranged from 6 to 46 cm. Mean concentration of plant dry mass per volume was 1.17 kg m – 3. The vertical pattern of leaf area distribution in individual trees for all tree species was characterized by a Gaussian-like curve. Stand leaf area index was 7.8. These evergreen, broad-leaved (laurisilva or lucidophyllous) forests represent a relic forest that was widespread in the Mediterranean region some 20 million years ago. Paralarvae of the family Gonatidae were sampled in the Gulfof Alaska during spring 2001–2003. Taxonomic characterswere determined to allow identification of the specimens tospecies. The dorsal head chromatophore pattern (DHCP) was themost robust character and allowed identification to speciesfor the first time without requiring the removal and examinationof the radula. Six different DHCPs were found among the sixspecies in the study area. The 1140 specimens collected consistedof the following six species: Berryteuthis anonychus (759),Berryteuthis magister (71), Gonatopsis borealis (155), Gonatuskamtschaticus (1), Gonatus madokai (4) and Gonatus onyx (143).The specimens had a size range of 3.0–20.63 mm dorsalmantle length with the majority of specimens smaller than 10 mm.All species showed an increasing trend in abundance from theshelf (0–200 m) to the slope (200–1000 m)to the basin (>1000 m) except G. onyx in 2001 and 2002.Wide variation in distribution and abundance was found for thefour most abundant species; however, in general, B. anonychuswas most abundant and widely distributed, followed by Gonatopisborealis, Gonatus onyx and B. magister. Alginate concentrations between 2 and 4% had little effect on the degradation rate of phenol by alginate-immobilized Pseudomonas putida. Ten-degree shifts from 25°C resulted in approximately 30% slower degradation. Maximal degradation rates were favored at pH 5.5–6.0. The response of degradation rate to increased air flow in the bubble column used was almost linear and an optimal higher than 16 vol vol−1 was indicated, although free cells appeared in the reaction medium above 12 vol vol−1. When the initial phenol concentration was raised, degradation rate was not significantly affected until levels higher than 1200 mg ml−1 where performance was markedly reduced. Increasing the ratio of total bead volume to medium volume gave progressively smaller increases in degradation rate. The direct shoot organogenesis was achieved from leaf explant of two commercially important clones of Populus deltoides on MS medium enriched with 15 mg/l adenine sulphate, 5 mg/l Ascorbic acid, 250 mg/l (NH4)2SO4 (referred to as PD1 medium) supplemented with 2.5 µM each of 6-benzylaminopurine and indole-3-acetic acid. Higher shoot organogenic potential was recorded from the explants of clone ‘G48’ as compared to clone ‘L34’. The age of leaf explant also affected the shoot organogenic potential, and maximum shoot organogenesis was recorded in case of 5th leaf from the top of microshoot. Histological studies revealed altered cell division resulting in the formation of meristematic pockets after 5 days of culture, these meristematic pockets grew into dome protuberances by 10th day. Organized shoots were visible after 15 days of culture. A clear three phases of shoot organogenesis viz induction (0–4 days), initiation and organization (4–10 days) and growth (11–16 days onwards) were observed. Marked variation in the activity of enzymes such as catalase, peroxidase, polyphenol oxidase and acid phosphatase was observed during these phases. The activity of these enzymes was found to increase in cultures grown on the medium resulting in shoot organogenesis during shoot development (after 7 days of culture).  相似文献

Dunaliella is a genus of green unicellular algae distributedin all the oceans and saline bodies of water throughout theworld and distinguished by unusual tolerance to salt. Sincethe cells of this genus do not possess a rigid cell-wall, theyrespond to changes in salt concentration by rapid alterationsin cell volume and then return to their original volume as aresult of adjustments in the amounts of intracellular ions andglycerol, this latter being the major organic osmoticum. Thepaper describes the behaviour of a mutant of D. parva 19/9 withreduced capabilities of growth above 0.5 kmol m–3 NaCl.The mutant is unusual in that its abilities to synthesize glyceroland pump out Na+ and Cl do not appear to be impaired;volume changes in the hyperosmotic range also appear to be roughlythe same as in D. parva. The average cell volume of mutant cellsis reduced (206µm3 as opposed to 255 µm3 in D. parva)and their rate of change of cell volume after an increase insalt concentration is lower; it took about 10 min for mutantcells in the light to reach a new cell volume whereas D. parvacells reached their new volume in less than 1 min. Both factorsmay be dependent on components of the cytoskeleton. The mutantthrows light on adaptations necessary to allow Dunaliella cellsto grow at high salt concentrations and demonstrates that halotoleranceincludes, but is not equivalent to, osmoregulation.
