A number of data are consistent with the hypothesis that increases in intracellular Na+ concentration (Na+ i) during ischemia and early reperfusion lead to calcium overload and exacerbation of myocardial injury. However, the mechanisms underlying the increased Na+ i remain unclear. 23Na nuclear magnetic resonance spectroscopy was used to monitor Na+ i in isolated rat hearts perfused with a high concentration of fatty acid as can occur under some pathological conditions. Whole-cell patch-clamp experiments were also performed on isolated cardiomyocytes in order to investigate the role of voltage-gated sodium channels. Na+ i increased to substantially above control levels during no-flow ischemia. The results show that a pharmacological reduction of Na+ i increase by cariporide (1 mol/L, a Na+/H+ exchange blocker) is not the only protection against ischemia-reperfusion damage, but that such protection may also be brought about by metabolic action aimed at reducing fatty acid utilization by myocardial cells. This action was obtained in the presence of etomoxir (0.1 mol/L), an inhibitor of carnitine palmitoyltransferase-1 (the key enzyme involved in fatty acid uptake by the mitochondria) which also decreases long-chain acyl carnitine accumulation. The possibility of Na+ channels participating in Na+ i increase as a consequence of alterations in cardiac metabolism was studied in isolated cells. Sustained INa was stimulated by the presence of lysophosphatidylcholine (LPC, 10 mol/L) whose accumulation during ischemia is, at least partly, dependent on increased long-chain acyl carnitine. Current activation was particularly significant in the range of potentials between –60 and –20 mV. This may have particular relevance in ischemia. The quantity of charge carried by sustained INa was reduced by 24% in the presence of 1 mol/L cariporide. Therefore, limitation of long-chain fatty acid metabolism, and consequent limitation of ischemia-induced long-chain acyl carnitine accumulation, may contribute to reducing intracellular Na+ increase during ischemia-reperfusion.  相似文献

This paper describes and analyzes a type of Toraja (South Sulawesi, Indonesia) suicide in which a person kills him or herself after having been slighted or offended, usually by a close family member. Comparing and contrasting such suicides to similar types found elsewhere in Austronesia-speaking Oceania, the paper argues that self-inflicted deaths of this nature are not so much anomic, as suggested by some analysts, as indignant; they are committed by persons who feel that they have been severely mistreated or abused according to traditional notions of reciprocity, mutual aid, and the dangers of frustrated desire. The paper concludes by suggesting that Durkheim may have underestimated the frequency and importance of indignant suicide in traditional societies and that, conversely, contemporary analysts may be underestimating the importance of traditional norms and values in accounting for the high rates of suicide found in many parts of the Pacific today.  相似文献

In this paper I explore the interactions between colonial law and native customary law in the formation of contemporary property regimes in a rural village in Sabah, Malaysia, that I call Govuton. Govuton was one of the few known villages in Sabah that rejected colonial policies of land settlement that focused on settling private, individual property claims. Instead, village leaders negotiated with colonial officials for their village lands to be legally designated as corporately-held village property under the title of Native Reserve. While the Native Reserve served to protect village access to jointly-held property in the colonial period, in the contemporary period new land disputes are arising as different images of community and tradition are strategically deployed by villagers in order to win struggles over rights of ownership and access to resources in the current political economy. By adopting such an historical and site-specific view of the transformation of property rights several broader themes regarding the relationship between state and society and natural resource management emerge. First, this case study challenges the idea the colonial governments were a monolithic force imposing laws on an unresisting native population. Second, the notion that the community is an appropriate unit for natural resource management is questioned. And finally, this case study raises the possibility that the current trend toward strengthening or reinvigorating native customary law is not always in the interests of native peoples with diverse interests in natural resource management.  相似文献

Summary A stock of Blattella germanica bearing the interchange T(3; 12)/3;12 was subjected to close inbreeding with selection for random disjunction at metaphase I. After 3–4 generations of selection, interchange quadrivalent chiasma frequency decreased, variability in free bivalent chiasma frequency increased sharply, and individuals with either random or directed disjunction were present in the stock. Random disjunction was modified from a ratio of 2112 (adj.-1; alt.-1; adj.-2; alt.-2) to a ratio of 1111. After 7–8 generations of selection, chiasma frequency appeared to stabilize at lower than normal levels and variability decreased for both quadrivalents and free bivalents. Directed disjunction was modified from a ratio of 2114 to 1112, and no individuals with the original high level of directed disjunction were detected. Chains-of-four tended to orient randomly, especially in individuals where the ring quadrivalents showed directed disjunction. Relaxation of inbreeding, but not selection, produced an increase in chiasma frequency and variability in both free bivalents and quadrivalents, but the modified ratios for both random and directed disjunction were retained. These results are discussed with respect to inbreeding effects and genetic control of chiasma frequency and metaphase I disjunction in interchange quadrivalents.  相似文献

Between 1984 and 1996, public health authorities in Israel maintained a secret policy of discarding blood donations made by Ethiopian-Israeli citizens and immigrants. Officials later attempted to justify this policy on the grounds that immigrants from Ethiopia were subject to high rates of infectious disease (especially HIV). In 1996, this led to an explosive and violent confrontation between Ethiopian-Israeli protestors and agents of the state, including police and public health authorities. This essay explores the cultural and political context of that confrontation, including the discourse of political violence which it occasioned. The conflict between Ethiopian-Israelis and the state was located within a wider set of political contexts, including the Israeli-Palestinian conflict, which was linked to it through a shared trope of spilled blood common to both. Cultural analyses which ignore this dynamic political context are in danger of seriously misrepresenting the meaning of the Blood Affair to its participants. At the same time, this essay also engages a critical analysis of the public health policies which led to the crisis. Public health and nationalist discourse reinforced one another at the expense of Ethiopian immigrants in general, and so-called Feres Mura Ethiopians in particular.  相似文献

Conclusions It follows from the discussion of the question of women and political violence in the contemporary Peru which I attempted to explore in its historical perspective in this essay, that the peculiarity of the division of labor in the Andean Sierra, based on the gender ideology which recognizes women's work as a complementary to men's work as relatively capable of resisting the extra-economic pressures of underdeveloped capitalism, has facilitated women's incorporation into an armed struggle led by the Sendero Luminoso. For Vicky Peláez, a leading Peruvian journalist, in the conditions of the Andean Sierra the movement could not survive and expand without the participation of women whose historical heritage of struggle has been multiplied by socio-economic and political crisis of the Peruvian state. On the other hand, the organizational framework of the PCP-SL, which recognizes and applies in practice the principle of equality between men and women, has stimulated female political activity. The inclusion of issues important to women in Sendero Luminoso's agenda has given additional impetus to the incorporation of women into class struggle not only as supporters or militants, but also as political and military leaders. Increasing participation of women in political and military actions creates additional conditions for the survival and expansion of collective action started by the PCP-SL years ago, using political violence as the main instrument of a new democratic revolution in Peru.

The fundamental statistical aspects of population coding of the movement direction in the motor cortex are studied numerically. The activity of neurons in a population is simulated using pseudorandom numbers so that the directional selectivity of the neurons is similar to that observed experimentally. Accuracy of the coding, which is evaluated by the root-mean-square (rms) error, is analyzed for various population sizes, degrees of variability of neuronal activity, and degrees of nonuniformity of distribution of the preferred directions. The dependence of the rms error on the population size shows a good fit to the inverse square-root law, from which it is estimated that a single population must contain around 10 000 neurons in order to attain the accuracy that allows 1 deg rms error, for example. The coding is studied further for populations with different types of tuning function. The results support the hypothesis proposed by Georgopoulos et al. (1988) except that the tuning function must be tuned in the sense that the average value of the function for movements with components in the preferred direction is larger than for movements away from the preferred direction.  相似文献

The definition of sports medicine indicates the possible therapeutic and research importance of games in the work of social scientists and clinicians. A cross-cultural comparison of super stressful competitions provides one insight into the importance of games in human societies. Fourteen such comparisons are made between Indians in Mexico who run 250-mile foot races and Americans in Alaska who conduct 1049-mile dog sled races. It is concluded that sports can help understand how a society defines itself and regulates violence. Games may be an essential human-specific need which provide entertainment, retain survival skills, ameliorate existential anxiety and promote religious and/or secular socialization. In this study, humans seem willing, if not insistent, to deploy considerable resources of time and material to sustain competitions among their own species, as well as between and with other species. Summary Tetraploid Vaccinium corymbosum genotypes exhibit wide variability in seed set following self- and cross-pollinations. In this paper, a post-zygotic mechanism (seed abortion) under polygenic control is proposed as the basis for fertility differences in this species. A pollen chase experiment indicated that self-pollen tubes fertilize ovules, but are also outcompeted by foreign male gametes in pollen mixtures. Matings among cultivars derived from a pedigree showed a linear decrease in seed number per fruit, and increase in seed abortion, with increasing relatedness among parents. Selfed (S1) progeny from self-fertile parents were largely self-sterile. At zygotic levels of inbreeding of F>0.3 there was little or no fertility, suggesting that an inbreeding threshold regulates reproductive success in V. corymbosum matings. Individuals below the threshold are facultative selfers, while those above it are obligate outcrossers. Inbreeding also caused a decrease in pollen viability, and reduced female fertility more rapidly than male fertility. These phenomena are discussed in terms of two models of genetic load: (1) mutational load — homozygosity for recessive embryolethal or sub-lethal mutations and (2) segregational load — loss of allelic interactions essential for embryonic vigor. Self-infertility in highbush blueberries is placed in the context of late-acting self-incompatibility versus early-acting inbreeding depression in angiosperms.  相似文献

As preparation for a below ground food web study, the spatial variability of three soil properties (total N, total C and pH) and two stable isotopes (13C and 15N of whole soil) were quantified using geostatistical approaches in upland pastures under contrasting management regimes (grazed, fertilised and ungrazed, unfertilised) in Scotland. This is the first such study of upland, north maritime grasslands. The resulting patterns of variability suggest that to obtain statistically independent samples in this system, a sampling distance of 13.5 m is required. Additionally, temporal change (a decline of 1) was observed in whole soil 15N for the grazed, fertilised plot. This may have been caused by new inputs of symbiotically-fixed atmospheric N2.  相似文献

During asexual fission in the ciliate Euplotes eurystomus, the macronucleus divides amitotically. The macronucleus was found to divide unequally, yielding sister pairs having a mean difference in DNA content of 11.6%. DNA content was determined by the Feulgen reaction using a fluorescent Schiffs reagent, and measuring fluorescence by cytophotometry. Variability in macronuclear DNA content was also examined in randomly-paired non-sister cells, and found to be greater than in sister cells. This greater variability could be due to accumulation of differences over a number of divisions, or to interclonal differences in equality of division. Two categories of non-sister cells were examined: recently divided, and parents constructed by averaging the DNA contents of progeny. Both showed similar variability in quantity of macronuclear DNA. The fact that cells surviving to divide showed no less variability in amount of DNA than cells immediately after division suggests that extremes in amounts of DNA resulting from unequal division are not selected against.  相似文献

Summary As clonal plants grow they move through space. The movement patterns that result can be complex and difficult to interpret without the aid of models. We developed a stochastic simulation model of clonal growth in the tall goldenrod, Solidago altissima. Our model was calibrated with field data on the clonal expansion of both seedlings and established clones, and model assumptions were verified by statistical analyses.When simulations were based on empirical distributions with long rhizome lengths, there was greater dispersal, less leaf overlap, and less spatial aggregation than when simulations were based on distributions with comparatively short rhizome lengths. For the field data that we utilized, variation in rhizome lengths had a greater effect than variation for either branching angles or rhizome initiation points (see text). We also found that observed patterns of clonal growth in S. altissima did not cause the formation of fairy rings. However, simulations with an artificial distribution of branching angles demonstrate that fairy rings can result solely from a plant's clonal morphology.Stochastic simulation models that incorporated variation in rhizome lengths, branching angles, and rhizome initiation points produced greater dispersal and less leaf overlap than deterministic models. Thus, variation for clonal growth parameters may increase the efficiency of substrate exploration by increasing the area covered and by decreasing the potential for intraclonal competition. We also demonstrated that ramet displacements were slightly, but consistently lower in stochastic simulation models than in random-walk models. This difference was due to the incorporation of details on rhizome bud initiation into stochastic simulation models, but not random-walk models. We discuss the advantages and disadvantages of deterministic, stochastic simulation, and random-walk models of clonal growth.  相似文献

Conclusions An even broader theoretical conclusion is in order. The term modernization has been enshrined in the value-free literature on development which the Western academy has created and supported. But the experience of Brasov, a local manifestation of Romanian development, makes it clear that modernization is by no means confined to Western assumptions, and that it always occurs in specific ways depending on the social situation of the society undergoing change, and on the political nature of the program being adopted. In short, socialist modernization in Romania is not parallel to modernization under capitalist auspices in analogous areas.  相似文献

The subject RNA models the binding site for the coat protein of the R17 virus, as well as the ribosome recognition sequence for the R17 replicase gene. With an RNA of this size, overlaps among the sugar protons complicate assignments of the 1H NMR spectrum. The cross peaks that overlap significantly in 2D-NOE spectra can frequently be resolved by introducing a third, in our approach the double-quantum, frequency axis. In particular the planes in a 3D-NOE/2QC spectrum perpendicular to the 2Q axis are extremely useful, showing a highly informative repeating NOE-2Q pattern. In this experiment substantial J-coupling confers special advantages. This always occurs for geminal pairs (H5/H5 for RNA plus H2/H2 for DNA), as well as for H5/H6, for H3/H4 in sugars with substantial populations of the N-pucker, for H1/H2 for S-puckered sugars, and usually for H2/H3. For the 24-mer RNA hairpin the additional information from the 3D-NOE/2QC spectrum allowed assignment of all of the non-exchangeable protons, eliminating the need for stable-isotope labeling.  相似文献

Various direct, indirect (kinetic and thermodynamic), and combined mechanisms have been proposed to explain the conversion of redox energy into a transmembrane protonmotive force (p) by enzymatic complexes of respiratory chains. The conceptual evolution of these models is examined. The characteristics of thermodynamic coupling between redox transitions of electron carriers and scalar proton transfer in cytochromec oxidase and its possible involvement in proton pumping is discussed. Other aspects dealt with in this paper are: (i) variability of H+/e stoichiometries, in cytochromec oxidase and cytochromec reductase and its mechanistic implications; (ii) possible models by which the reduction of dioxygen to water at the binuclear heme-copper center of protonmotive oxidases can be directly involved in proton pumping. Finally a unifying concept for proton pumping by the redox complexes of respiratory chain is presented.  相似文献

In policy processes, biodiversity has been broadened into a holistic notion uniting a variety of cultural, social and economic issues with the biological conservation issue. Therefore, the formation of biodiversity conservation into concrete policy goals appears rather difficult. In this paper, we aim to explore the political implications and consequences of the strong dependency on science of the biodiversity issue. Concentrating especially on environmental policy options, we also examine the globalization and localization processes in the case of biodiversity. In spite of several controversial features and of an evident lack of knowledge on biological diversity, the issue undoubtedly has significant political strengths. However, from the viewpoint of research on environmental policies, the role of scientific knowledge in decision making should be recognized as an open empirical question depending on the concrete contexts of the decision-making process. Further, it is concluded that the concepts relating to global environment are objects of a continuous localization. Thus, any moral-technocratic solutions emphasizing simply universal interests as the basis for global biodiversity management must be met with caution. In future research there is a need for concrete case studies in order to clarify the local conditions and opportunities of biodiversity policies.  相似文献

All models of self-motion from optical flow assume the instantaneous velocity field as input. We tested this assumption for human observers using random-dot displays that simulated translational and circular paths of movement by manipulating the lifetime and displacement of individual dots. For translational movement, observers were equally accurate in judging direction of heading from a velocity field with a two-frame dot life and a direction field in which the magnitudes of displacement were randomized while the radial pattern of directions was preserved, but at chance with a speed field in which the directions were randomized, preserving only magnitude. Accuracy declined with increasing noise in vector directions, but remained below 2.6° with a 90° noise envelope. Thus, the visual system uses the radial morphology of vector directions to determine translational heading and can tolerate large amounts of noise in this pattern. For circular movement, observers were equally accurate with a 2-frame velocity field, 3-frame acceleration displays, and 2-frame and 3-frame direction fields, consistent with the use of the pattern of vector directions to locate the center of rotation. The results indicate that successive independent velocity fields are sufficient for perception of translational and circular heading.  相似文献

The molecular basis of familial hypercholesterolemia (FH) in three families of Spanish descent from La Habana was investigated by the candidate gene approach. The Arg3500Gln mutation of apolipoprotein B-100 was not found. Identification of low density lipoprotein receptor (LDLR) gene haplotypes segregating with FH guided the characterisation of three point mutations by automated sequencing. One, a Val408Met missense mutation, a founder mutation in Afrikaner FH patients, was recurrent, being associated with a distinct DNA haplotype. The other two, Glu256Lys and Val776Met missense mutations, were novel and modified highly conserved residues. These mutations were absent in normolipidemic subjects and were associated in heterozygous carriers with twice the cholesterol levels observed in noncarriers. Noticeably, cardiovascular complications were rarely observed in older heterozygotes, even in those with the Afrikaner FH-2 mutation. These findings confirm the molecular heterogeneity of LDLR gene mutations causing FH and the variability of their expression across different populations.  相似文献

Great variability involving 36 quantitative and 31 qualitative characters, as well as the habitats occupied has been shown in 18 populations of Elymus caninus from Poland, based on the detailed population and statistical analyses of morphological characters. The variability of these characters proves that the intraspecific units which have been distinguished on the basis of these characters do not have systematic significance. The two morphotypes of E. caninus (pauciflorum and caninus) described in this study will provide the basis for further genetic studies aimed at establishing their significance for considerations in the systematics of the species.It is the first paper from the series: Biodiversity of wild Triticeae (Poaceae) in Poland.  相似文献

We consider four models of the lateral distribution of proteins in lipid bilayer membranes and study the fraction of lipids which are adjacent to at least one protein (adjacent lipids) and how this quantity depends upon protein concentration. The models are (i) hard hexagons free to move from one lattice site to another; (ii) hard disks moving on a continuum; (iii) a mixture of two sizes of nearly-hard disks moving on a continuum; (iv) a modification of (ii). The hexagons or disks represent proteins, while unocupied lattice sites or the remainder of the continuum represents lipids. In (iii) large disks represent proteins and small disks represent lipids. In (iv) some of the continuum between pairs of disks, where packing defects might occur, is not occupied by lipids. We find that an analytical expression for the adjacent lipids (Hoffmann et al. 1981), which is in excellent agreement with the results of the Hexagon model (i), breaks down at a packing density of f A0.805, and we show by considering the hexagon pair correlation function, that this indicates the onset of random close packing, and that a transition to ordered close packing occurs at f A=0.866. We thus obtain an operational definition for a random distribution of hexagons: distributions of packing densities0.805. We show that the Disk model (ii) gives results for adjacent lipids that are greater than the Hexagon model and compare these results to the Two Disk model (iii) which gives a result substantially less than the Hexagon model (Mountain et al. 1986). We show that the Modified Disk model (iv) gives results in essential agreement with the Hexagon model except for f A0.77. Finally we discuss the general appearance of the motion restricted ESR spectrum and conclude that, of these four models, the Modified Disk or the Hexagon models best account for the data.
