Crosses were produced between two lines of White Plymouth Rock chickens, one of which had been selected for low 8-week body weight for 31 generations (L) and the other of which was a bantam population (B). The parental lines, reciprocal F1s, reciprocal F2s and all possible back-crosses to each parental line (total of 16 populations) were available for study. Blood was obtained from 10 females within each population. DNA was extracted from blood mixes (equal amounts of blood from each individual) for each population, and from blood samples of each individual in the two parental lines. Fourteen line-specific DNA fingerprint (DFP) bands (those bands present in one parental population, but not in the other parental population) were analysed (eight from line L and six from line B). Regression analyses were conducted to compare the known proportion of genomic contribution from each parental population with values based on relative band intensity obtained with a scanning densitometer. The resulting regression coefficient of 1.004 demonstrated that DFP analysis of relative band intensity is an effective method of estimating the relative proportion of genome contributed by parental populations.  相似文献

Thirteen highly inbred chicken lines were analysed at the DNA level by DNA fingerprinting (DFP) and by polymerase chain reaction (PCR) using random primers. In general, the DFP patterns of individuals within a line were identical. The DFP band-sharing (BS) values among lines from different breeds (Leghorn, Fayoumi, Spanish) ranged from 0.10 to 0.20. The DFP BS values among Leghorn lines from different genetic backgrounds ranged from 0.42 to 0.79. The DFP BS values among lines selected for different major histocompatibility complex serotypes from a common genetic background ranged from 0.70 to 0.95. Some randomly amplified polymorphic DNA (RAPD) PCR products were specific to a single line, some to all lines from the same genetic base, and some to all lines from the same breed. The RAPD-PCR band-sharing values ranged from 0.66 to 0.99 for all between-line comparisons. Thus, the ability to detect biodiversity at the DNA level was greater in this study for DFP than for RAPD-PCR. The possible origin of line-specific bands, relative advantages of detecting biodiversity by using different molecular screening techniques and uses of highly inbred chicken lines in molecular analysis are discussed.  相似文献

DNA fingerprints obtained by multi-locus probes have been shown to be convenient tools to assess genetic variability and genetic distances in animal populations. This method has been applied in the present study to two Rhode Island Red lines of chickens divergently selected for residual food consumption (RFC) for 16 generations (R-: low RFC; R+: high RFC). Within each line, individuals were sampled from each tail of the phenotypic distribution of RFC for males and for females. The three probes used were two minisatellite probes, R18-1 and 33-6, and the endogenous avian retroviral element probe. Altogether, 101 bands were scored. Band-sharing levels obtained from analysis of pooled DNA were extremely high within-Line (> 0.9) and still high between the lines (0.82 to 0.86). When band frequencies were compared, correlation coefficients were lower between lines than between extreme groups within lines regardless of the performance level. Although the selected lines differed widely in the selected trait RFC and a few correlated traits, it was concluded that they are probably different by only a low proportion of their genome.  相似文献

Some muscle-bone relationships were studied in terms of gastrocnemius muscle weight, femur and tibia length and femur and tibia weight in four lines of mice (CBi-, CBi+, CBi/L and CBi/C) artificially selected for different body conformations and in the unselected control line (CBi). CBi- (low body weight--short tail) and CBi+ (high body weight--long tail) lines were divergently selected following the positive genetic correlation between body weight and tail (skeleton) length (agonistic selection). In contrast, CBi/L (low body weight--long tail) and CBi/C (high body weight--short tail) were also divergently selected but against the aforementioned correlation (antagonistic selection). The relationship between bone length and muscle weight was interpreted based on the assumption that the increased tension generated by the longitudinal growth of a bone, brings about an increase in the mass of the muscles attached to it. All CBi+, CBi/C and CBi/L mice showed enlarged femurs and tibias, but only those genotypes simultaneously selected for high body weight (CBi+ and CBi/C) showed heavier muscles than controls. The CBi+ and CBi- genotypes with agonistic selection differ in bone length and muscle weight, as it would be expected of the allometric modification of their body conformation, showing the associated longitudinal bone growth-muscle growth. CBi/C and CBi/L mice, with a non-allometric modification of body conformation, exhibited the same bone length but different muscle weight. Consequently, the antagonistic criterion allowed to confirm that the genetic influence on of the proposed muscle-bone relationships could be modified, thus making it possible to lengthen the bone through selection of a long skeleton and to avoid the correlated effect on muscle mass, by selecting for a low body weight, bringing forth presumptive evidence that both processes were genetically independent.  相似文献

Chicken lines produced by divergent selection for the functional adrenal reserves showed significant between-line differences in the content of corticosterone and other hormones (thyroxin, progesterone), as well as in body weight, early maturation, and egg yield. DNA fingerprinting with the pGB725 probe revealed molecular changes in genomic DNA of the chicken lines subjected to plus and minus selection. The genetic distances between the original population and the selected chicken lines, which were estimated from the molecular hybridization patterns, reflected the history of breeding. Analysis of mixed DNA from several individuals of each line revealed specific hybridization bands that could serve as DNA markers during selection for the high and low corticosterone levels in blood.  相似文献

RAPD markers often primers were used to assess the polymorphism among pooled DNA of eight goose lines. The number of bands amplified by each primer ranged from 1 to 8, within a mean of 2.86. Some bands appeared specific for the line or genetic background. RAPD technique is an effective method for generating the polymorphic DNA marker in the goose. RAPD patterns from mixed DNA samples can reflect the genetic information of populations. The present study indicated that 10 generations selected for egg production and body weight under various pressure, resulted in genetic variation among goose lines as detected by RAPD. Selection for meat traits caused greater genetic diversity than selection for egg production.  相似文献

To assess the value of DNA fingerprints for the prediction of heterosis in chickens, retrospective analyses of data from three crossbreeding experiments and DNA fingerprints (DFP) of parental strains were conducted using two minisatellite and one middle-repetitive DNA probes. DFP bands were assessed on pooled DNA samples of 10-15 individuals per parental genetic group. The number of DFP bands evaluated in the experiments ranged from 81 to 139. The probes varied in their predictive value, but predictability of heterosis generally increased with multiple probes. Highly significant correlations (0.68-0.87) between band sharing ratios (SH) and heterosis were found in 25 crosses of White Leghorns in the first egg production cycle for age at sexual maturity, egg production, and mature body weight: traits with heterosis of 10% or more of the means. Regressions of SH explained 78.4% of the variation in heterosis in age at sexual maturity, 60.2% in egg production and 46.4% in mature body weight. For ``broiler' traits with heterosis of <1%, none of the correlations, based on 13 crosses, were significant. It was concluded that multilocus probe DFP of pooled DNA samples show promise as predictors of heterosis.  相似文献

Restriction fragment length polymorphisms for somatotropic genes were tested for associations with body weight and postweaning growth rate in mice. Polymorphisms for growth hormone (GH) and insulin-like growth factor 2 (IGF-2) genes were identified in stock population lines which had been subjected to long-term selection for high 42-day body weight (H lines) or randomly mated (FP and C lines). Two F2 populations of mice (5F2 and MF2) were generated from crosses between a single H line of mice and two unselected control lines and subsequently, two divergently weight selected sublines were generated from each F2 population. The GHh allele which had originally been fixed in three of four H lines and absent from all FP and C lines was found to have a significant (P less than 0.01) effect on 42-day weight and postweaning growth rate in the F2 populations. However, GHh was associated with lower 42-day weight in the F2 populations, suggesting that the positive association between GHh and weight in the stock population was unique to the high weight selected genetic background of those lines. In agreement with this, the frequency of GHh increased in sublines selected for high 42-day weight and decreased in sublines selected for low 42-day weight. The IGF-2H5 allele was associated with higher weights in a sex-dependent manner in 5F2. In the high selected subline derived from 5F2, a significant increase in the frequency of IGF-2H5 was observed. Therefore this allele, in contrast to GHh, appears to be a positive indicator of growth irrespective of genetic background.  相似文献

Quantitative trait loci (QTL) influencing body weight were mapped by linkage analysis in crosses between a high body weight selected line (DU6) and a control line (DUKs). The two mouse lines differ in body weight by 106% and in abdominal fat weight by 100% at 42 days. They were generated from the same base population and maintained as outbred colonies. Determination of line-specific allele frequencies at microsatellite markers spanning the genome indicated significant changes between the lines on 15 autosomes and the X chromosome. To confirm these effects, a QTL analysis was performed using structured F2 pedigrees derived from crosses of a single male from DU6 with a female from DUKs. QTL significant at the genome-wide level were mapped for body weight on chromosome 11; for abdominal fat weight on chromosomes 4, 11, and 13; for abdominal fat percentage on chromosomes 3 and 4; and for the weights of liver on chromosomes 4 and 11, of kidney on chromosomes 2 and 9, and of spleen on chromosome 11. The strong effect on body weight of the QTL on chromosome 11 was confirmed in three independent pedigrees. The effect was additive and independent of sex, accounting for 21-35% of the phenotypic variance of body weight within the corresponding F2 populations. The test for multiple QTL on chromosome 11 with combined data from all pedigrees indicated the segregation of two loci separated by 36 cM influencing body weight.  相似文献

A potential limitation of the use of a dominant molecular marker system such as DNA fingerprinting (DFP) is the inability to distinguish homozygous from heterozygous allele state in an individual, and a resulting inaccuracy in estimating effects of the marker alleles. The objective of this study was to accurately estimate the effect of DFP markers on egg-production traits. A BC1 population was produced from two distinct layer lines. Four DFP bands, each originating predominantly in one of the two parental lines, were evaluated for linkage with egg-production quantitative trait loci in the BC1 population. The egg-production traits consisted of eight early period and seven late period measurements. Eight marker-trait linkages were identified out of 60 total statistical tests. By utilizing information on frequency of DFP bands in two parental lines, selecting F1 sires with DFP bands present, and backcrossing to the line lacking these bands, the population design allowed definitive identification of the DFP zygosity in the BC1 resource population hens. In this manner, accurate estimates of marker allele effects on egg-production traits were obtained from the dominant marker system of DNA fingerprinting.  相似文献

The variability of DNA content in turkey spermatozoa of four different lines and its correlation with body weight and sperm concentration were studied. In lines selected for lower body weight the DNA content was 2.034 and 2.036 pg per spermatozoon. In lines selected for higher body weight the DNA content was 2.267 and 2.370 pg per spermatozoon. Sperm concentration in 1 mm3 of semen, however, was higher in lines with a lower body weight (6.08–6.21 million) in comparison with lines selected for higher body weight (5.46–5.67 million). The correlations between the DNA content and sperm concentration were negative (r ranged from ?.457 to ?.860).  相似文献

Thirty-one bucks from two lines divergently selected for 63-d body weight (low, L and high, H) were solicited every week (twice at a 15min interval) during 18 weeks resulting in 482 ejaculates. While differing markedly on adult body weight (L: 4650g versus H: 5925g), both lines had the same testis weight. Libido did not differ between the lines. The proportion of ejaculates suitable for insemination was markedly higher in the L line (66.5% versus 44.2%). Mass motility and the volume of the ejaculates were higher in the L line while the sperm concentration was higher in the H line. Overall, the total number of spermatozoa per ejaculate was similar in both lines but the efficient number of spermatozoa per ejaculate, a synthetic criterion taking into account the ability of the ejaculate for insemination was higher in the L line (229 versus 170x10(6)). The L line had higher values of average path velocity, linearity and curvilinear velocity but a lower value of beat cross frequency. In the L line, both ejaculates had the same concentration, while in the H line, the first ejaculate was more concentrated than the second one. Both pleiotropy and linkage were examined as possible explanations for the fixation of the Hbb3 allele in the six Large lines of a population of mice divergently selected for six-week body weight (six replicates in each direction and six controls). A survey of over 1200 individuals in the lines still segregating at the Hbb locus excluded pleiotropy as a possible explanation. The results showed a nonsignificant effect of haemoglobin genotype on body weight. The linkage relationship of the Hbb locus was examined using a backcross mating system. The Hbb3 c+ region (chromosome 7) of a Large line was backcrossed into its corresponding Small line (Hbb4 c). The resultant difference in body weight between the two segregants (Hbb8 c+ [Hbb4 c; Hbb4 c/Hbb4 c) was measured. The results suggested linkage as the most plausible explanation for the fixation of the Hbb4 allele in the six Large lines.  相似文献

Genetic similarity and diversity of cultured catfishSilurus asotus populations collected from two areas in western Korea were examined using randomly amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR). Out of 20 random primers tested, 5 produced 1344 RAPD bands ranging from 8.2 to 13.6 polymorphic bands per primer. The polymorphic bands in these populations ranged from 56.4% to 59.6%. Polymorphic bands per lane within populations ranged from 4.9% to 5.3%. The similarity within the Kunsan population varied from 0.39 to 0.82 with a mean (± SD) of 0.56 ± 0.08. The level of bandsharing values was 0.59 ± 007 within the catfish population from Yesan. The genetic similarity in cultured catfish populations may have been caused because individuals from two populations were reared in the same environmental conditions or by inbreeding during several generations. However, in view of bandsharing values, polymorphic bands and also the specific major bands that were inter-population-specific, significant genetic differentiation between these populations were present even if bandsharing (BS) values were somewhat numerically different. Therefore, the number of RAPD polymorphisms identified in this study may be sufficient to permit estimating genetic similarity and diversity. However, in future, additional populations, sampling sites and individuals will be necessary to make up for these weak points.  相似文献

为探讨鸡视网膜母细胞瘤基因1(Retinoblastoma1,RB1)多态性对体重性状的影响,文章以东北农业大学高、低脂双向选择品系肉鸡为实验材料,采用MALDI-TOF-MS、PCR-RFLP方法进行基因多态性检测和个体基因型分析,共获得27个SNP位点的基因型数据。采用滑动窗口法构建单倍型,进而利用单位点和单倍型分别与鸡体重性状进行关联分析。结合单位点和单倍型分析结果,确定了RB1基因上4个显著影响1周龄体重的SNP位点,2个显著影响1、3周龄体重的SNP位点。研究结果表明RB1基因是影响鸡早期体重性状的重要候选基因。  相似文献

DNA fingerprints of Japanese quail male and female pure line breeders were obtained with probes 33.6, 33.15, and R18.1 and they yielded a total of 59 scoreable bands. Bandsharing (0 < BS < 1) was calculated within and between six quail lines of two origins, and under reciprocal recurrent (AA and BB), within-line (DD and EE) or no (PP and FF) selection. Twenty one pair types were compared. BS was 0.30 higher within line than between lines. BS with the control line was smaller for reciprocal recurrent selection lines than for lines under individual selection. Bandsharing between the two reciprocal recurrent selection lines was 0.19 lower than between lines under individual selection. These results indicate that the two selection methods had different effects on the genetic constitution of the lines, in agreement with previous observations made from the analysis of biochemical polymorphisms with the same set of birds. Egg production and weight traits of pure and crossbred progeny from fingerprinted quail were obtained and compared, and a linear relationship with the measure of bandsharing was estimated. No significant regression coefficient of performance on BS was found over all progeny genetic types. Heterosis from individual matings could also be estimated under the two selection methods since the same birds were parents of both pure and crossbred performance-tested quail. The association of heterosis with the difference between BS of parents of the purebreds and BS of parents of their half-sib crossbreds was favourable and significant for early production traits in lines DD and EE, but no relationship was found in lines AA and BB. These results indicate that the high level of heterosis obtained through reciprocal recurrent selection, and the heterosis observed under within-line selection may have, partly at least, a different genetic determinism. Therefore, the relationship of heterosis with BS may also depend on the past history of selection in the lines.  相似文献

The effects of selection for growth rate on weights and qualitative carcass and muscle traits were assessed by comparing two lines selected for live body weight at 63 days of age and a cryopreserved control population raised contemporaneously with generation 5 selected rabbits. The animals were divergently selected for five generations for either a high (H line) or a low (L line) body weight, based on their BLUP breeding value. Heritability (h2) was 0.22 for 63-d body weight (N = 4754). Growth performance and quantitative carcass traits in the C group were intermediate between the H and L lines (N = 390). Perirenal fat proportion (h2 = 0.64) and dressing out percentage (h2 = 0.55) ranked in the order L < H = C (from high to low). The weight and cross-sectional area of the Semitendinosus muscle, and the mean diameter of the constitutive myofibres were reduced in the L line only (N = 140). In the Longissimus muscle (N = 180), the ultimate pH (h2 = 0.16) and the maximum shear force reached in the Warner-Braztler test (h2 = 0.57) were slightly modified by selection.  相似文献

Summary Two experiments were conducted simultaneously to determine (1) if fast-growing fingerlings of channel catfish, Ictalurus punctatus, could be identified by simple visual selection of body size and (2) if initial size advantages influenced subsequent growth and carcass traits of divergently selected channel catfish. Exp. 1 included large (L), medium (M), and small (S) fingerling sizes from each of the control (C), selected upward (+) and selected downward (–) lines for body weight. Exp. 2 included all fmgerlings of the same size (25±5 g) from the 3 lines. Catfish from the L size-class, within each full-sib family in each line, were consistently heavier and longer than M and S size-classes throughout the 53-week experimental period. Fingerlings from the M size-class were also superior in growth to those from the S size-class. Catfish from the + line exceeded those from the C and –lines in body weight and total length at the conclusion of Exp. 1 but not in Exp. 2. This was attributed to the selection of equal size fmgerlings in Exp. 2 which may have excluded fingerlings with the best growth potential from the + body weight line. Results of the two experiments combined indicated that one generation of divergent selection has created genetic differences among lines of channel catfish.Supported by State and Hatch funds allocated to the Georgia Agricultural Experiment Station  相似文献
