Olfactory discrimination of nicotine-enantiomers by smokers and non-smokers   总被引:3,自引:2,他引:1  
This study reports an investigation of smokers and non-smokersperformed in order to determine differences in the olfactoryperception of the stereoisomers of nicotine; 38 subjects participated(20 smokers, 18 non-smokers). The investigated parameters were:hedonic ratings and intensity estimates, discrimination betweenenantiomers, estimates of detection thresholds and the odorousquality of nicotine enantiomers. Subjects were able to discriminatebetween the two stereoisomers of nicotine. Whereas both groupsreported the R(+) isomere to cause an unpleasant sensation,the S(–) isomere was perceived as pleasant by smokers,but not by non-smokers. The differences in the hedonic ratingsof S(–) nicotine between smokers and non-smokers mightbe due to the smokers' experience of the pharmacological actionof S(–) nicotine, which is the main isomere in cigarettesmoke.  相似文献

The effect of three gibberellin synergists on lettuce hypocotylelongation was studied. Dihydroconiferyl alcohol isolated fromlettuce plants and (–)-(6S, 1'S)-pestalotin isolated fromPestalotia cryptomeriaecola enhanced the promoting effect ofgibberellic acid on hypocotyl elongation of lettuce seedlingswith and without the cotyledons. On the other hand, TA, a triazinonecompound, did not enhance the gibberellin effect. The actionof (–)-(6S, 1'S)-pestalotin was strongly inhibited bycompetitive inhibitors of dihydroconiferyl alcohol such as caffeic,ferulic and trans-cinnamic acids. Of the two stereoisomers ofpestalotin, (+)-(6R, 1'R)-pestalotin enhanced the gibberellineffect but (+)-(6R, 1'S)-epipestalotin did not. (+)-(6R, 1'S)-Epipestalotinstrongly inhibited the action of (–)-(6S, 1'S)-pestalotinand dihydroconiferyl alcohol. TA did not affect the action ofdihydroconiferyl alcohol. Stress-relaxation analysis of the mechanical properties of thelettuce hypocotyl cell wall demonstrated that gibberellic acidcaused cell wall loosening and dihydroconiferyl alcohol andpestalotin did not influence this gibberellin effect. A comparison of the binding of (–)- and (+)-[3H]nicotine to rat brain membranes revealed that only the (–)-enantiomer showed high affinity binding; while the (+)-enantiomer was at least 1/10 as effective as the (–)-enantiomer when in competition with (–)-[3H]nicotine as the ligand. Positive cooperativity, which is observed with (–)-[3H]nicotine as the presence of low concentrations of (+)-nicotine, may account for the seeming paradox.  相似文献

The interaction of the nicotinic agonist (R,S)-3-pyridyl-1-methyl-2-(3-pyridyl)-azetidine (MPA) with different nicotinic acetylcholine receptor (nAChR) subtypes was studied in cell lines and rat cortex. MPA showed an affinity (Ki = 1.21 nM) which was higher than anatoxin-a > (−)-nicotine > (+)-[R]nornicotine > (−)-[S]nornicotine > and (+)-nicotine, but lower than cytisine (Ki = 0.46 nM) in competing for (−)-[3H]nicotine binding in M10 cells, which stably express the recombinant 4β2 nAChR subtype. A one-binding site model was observed in all competing experiments between (−)-[3H]nicotine binding and each of the agonists studied in M10 cells. MPA showed a 13-fold higher affinity for (−)-[3H]nicotine binding sites compared to the [3H]epibatidine binding sites in rat cortical membranes. In human neuroblastoma SH-SY5Y cells, which predominantly express the 3 nAChR subunit mRNA, MPA displaced [3H]epibatidine binding from a single population of the binding sites with an affinity in the same nM range as that observed MPA in displacing [3H]epibatidine binding in rat cortical membranes. Chronic treatment of M10 cells with MPA significantly up-regulated the number of (−)-[3H]nicotine binding sites in a concentration dependent manner. Thus MPA appears to have higher affinity to 4-subunit containing receptor subtype than 3-subunit containing receptor subtype of nAChRs. Furthermore MPA binds to 4β2 receptor subtype with higher affinity than (−)-nicotine and behaves, opposite to cytisine, as a full agonist in up-regulating the number of nAChRs. © 1998 Elsevier Science Ltd. R-(+)-2-(2-naphthoxy)propionic acid [(+)-NOP], but not (–)-NOP,was active in inducing calli from peel tissues of cucumber fruits.The callus-inducing activity of (+)-NOP was comparable to thatof 1-naphthylacetic acid. The light-catalysed isomerization of (+)-abscisic acid (ABA)to its trans isomer during isolation from leaves was monitoredby the addition of (±)-[2-14C]ABA to the extraction medium.(+)Trans-abscisic acid (t-ABA) was found to occur naturallyin rose (Rosa arvensis) leaves at 20µg/kg fresh weight;(+)-ABA was present at 594µg/kg. (±)-[2-14D]Trans-abscisicacid was not isomerized enzymically to ABA in tomato shoots. (±)-Abscisic acid was converted by tomato shoots to awater-soluble neutral product, ‘Metabolite B’, whichwas identified as abscisyl-ß-D-glucopyranoside. When(±)-[2-14C]trans-abscisic acid in an equimolar mixturewith (±)-[2-14C}ABA was fed to tomato shoots it was convertedto its glucose ester 10 times faster than was ABA. Trans-abscisyl-ß-D-glucopyrano8ide only was formedfrom (±)-[2-14C]t-ABA in experiments lasting up to 30h. Glucosyl abscisate was formed slowly from ABA and the freeacid fraction contained an excess of the unnatural (–).ABAas did the ABA released from abscisyl-ß-D-glucopyranosideby alkaline hydrolysis. The (+).ABA appeared to be the solesource of the acidic ‘Metabolite C" previously noted. The concentrations of endogenous (+)-, (+)-[2-14C]-, and (–)-[2-14C]ABAremaining as free acid, and also in the hydrolysate of abscisyl-ß-D-glucopyranoside,were measured by the ORD, UV absorption, and scintillation spectrometryof highly purified extracts of ABA from tomato shoots whichhad been supplied with racemic [2-l4C]ABA.  相似文献

Some previous work on zygotropism in Mucor is briefly reviewed,and further observations confirming earlier studies presented. In addition to positive zygotropism between (+) and (–)zygophores, there is claimed to be a corresponding ‘repulsion’between zygophores of the same mating type, (+) from (+) and(–) from (–). The stimulus appears to be transmittedby the diffusion of a volatile substance from one zygophoreto another. A tentative explanation of the phenomena describedis advanced, based on two postulated volatile hormones, eachevolved by zygophores of one mating type only, stimulating growthin zygophores of its own mating type and retarding it in thoseof the opposite type.  相似文献

测定了青杨脊虎天牛Xylotrechus rusticus (L.)雌、雄成虫对其寄主杨树中的水杨醛(0.95 μmol/μL)和非寄主植物中0.3 μmol/μL的叶绿醇、0.4 μmol/μL的水芹烯和0.6 μmol/μL的 R 型α-蒎烯、S 型α-蒎烯、S型β-蒎烯、3-蒈烯、罗勒烯、香草烯和松节油等10种植物挥发性气味物质的触角电位(EAG)反应。结果表明,与对照相比,这10种植物挥发物多能引起成虫明显的EAG反应( P<0.05,P<0.01 ),其中雌虫对松节油、水杨醛、R 型α-蒎烯和 S 型α-蒎烯的EAG反应较强; 雄虫对 R 型α-蒎烯的EAG反应最强,松节油次之。根据雌虫对这10种挥发物EAG反应的强弱,进一步测定了雌虫对0.00006、0.0006、0.006、0.06、0.6、0.12 μmol/μL的松节油、R 型α-蒎烯、S 型α-蒎烯以及0.000095、0.00095、0.0095、0.095、0.95、0.19 μmol/μL的水杨醛的EAG和行为反应。结果表明,雌虫对松节油、水杨醛和 R 型α-蒎烯的EAG反应随气味物质浓度的增加而增加,水杨醛浓度增加到0.95 μmol/μL、松节油和 R 型α-蒎烯浓度增加到0.6 μmol/μL以后,EAG反应值趋于平稳;对 S 型α-蒎烯的反应随浓度的增加而呈线性增加。水杨醛浓度低于0.095时,对雌虫没有明显的定向作用( P>0.05 ),高于此浓度时表现为驱避作用( P<0.05 ); 松节油在浓度低于或等于0.6 μmol/μL时对雌虫表现为驱避作用,浓度为0.6时驱避效果最佳( P<0.01 )。雌虫对 R 型α-蒎烯和 S 型α-蒎烯没有明显的定向行为反应。  相似文献

The underlying cause of Alzheimer's disease is thought to be the aggregation of monomeric beta-amyloid (Abeta), through a series of toxic oligomers, which forms the mature amyloid fibrils that accumulate at the center of senile plaques. It has been reported that L-(-)-nicotine prevents Abeta aggregation and toxicity, and inhibits senile plaque formation. Previous NMR studies have suggested that this could be due to the specific binding of L-(-)-nicotine to histidine residues (His6, His13, and His14) in the peptide. Here, we have looked at the effects of both of the L-(-) and D-(+) optical enantiomers of nicotine on the aggregation and cytotoxicity of Abeta(1-40). Surprisingly, both enantiomers inhibited aggregation of the peptide and reduced the toxic effects of the peptide on cells. In NMR studies with Abeta(1-40), both enantiomers of nicotine were seen to interact with the three histidine residues. Overall, our data indicate that nicotine can delay Abeta fibril formation and maintain a population of less toxic Abeta species. This effect cannot be due to a highly specific binding interaction between nicotine and Abeta, as previously thought, but could be due instead to weaker, relatively nonspecific binding, or to the antioxidant or metal chelating properties of nicotine. D-(+)-nicotine, being biologically much less active than L-(-)-nicotine, might be a useful therapeutic agent.  相似文献

(-)-[3H]Nicotine was found to bind specifically to membranes of human brains obtained at autopsy. The binding was stereospecific, (-)-nicotine being 40 times more potent than (+)-nicotine in displacing labeled (-)-nicotine. Saturation binding studies revealed the presence of two binding sites with dissociation constant (KD) values of 8.1 and 86 nM, and maximum binding capacity (Bmax) values of 36 and 90 fmol/mg protein, respectively. In competition studies, nicotinic agonists were 1,000 times more potent than ganglionic, neuromuscular, and muscarinic blocking drugs in displacing labeled (-)-nicotine. IC50 values for cholinergic drugs of (-)-[3H]nicotine binding were as follows: (-)-nicotine, 0.51 nM; acetylcholine, 12.6 nM; (+)-nicotine, 19.9 nM; cytisine, 27.3 nM; and carbachol, 527 nM. IC50 values of alpha-bungarotoxin, hexamethonium, d-tubocurarine, and atropine were larger than 50 microM. (-)-[3H]Nicotine binding was highest in the nucleus basalis of Meynert and thalamus and lowest in the cerebral cortex and caudate in the brain regions tested. These results suggest that nicotinic cholinergic receptors are present in human brain and that there are regional differences in the density of these receptors.  相似文献

Rabbit nasal olfactory and respiratory microsomes demonstrate high activity toward [3H]-(S)-nicotine, with specific activities of 22.2 and 6.5 nmol/min/mg protein, respectively. The major metabolite produced is (S)-nicotine delta 1'; 5'-iminium ion, with lesser amounts of nornicotine and the N'-oxide. Reconstitution of the rabbit nasal microsomal system with cytochromes P-450 NMa and NMb indicated that only P-450 NMa has significant activity toward nicotine, and the metabolite profile and turnover are similar to that observed with nasal microsomes. The low Km (35 microMs) and high Vmax (28 min-1) suggest that a significant portion of inhaled nicotine is metabolized by nasal tissues in the rabbit.  相似文献

The seasonal variability of phytoplankton in the EquatorialAtlantic was analysed using Sea-viewing Wide Field-of-view Sensor(SeaWiFS)-derived chlorophyll a (Chl a) concentration data from1998 to 2001, together with in situ Chl a and primary productiondata obtained during seven cruises carried out between 1995and 2000. Monthly averaged SeaWiFS Chl a distributions werein agreement with previous observations in the Equatorial Atlantic,showing marked differences between 10° W in the EasternTropical Atlantic (ETRA) and 25° W in the Western TropicalAtlantic (WTRA) provinces (Longhurst et al. 1995. J. PlanktonRes., 17, 1245–1271). The seasonal cycle of SeaWiFS-derivedChl a concentration calculated for 0–10° S, 0–20°W (ETRA) is consistent with in situ Chl a measurements, withvalues ranging from 0.16 mg m–3, from February to April,to 0.52 mg m–3 in August. Lower variability was observedin 10° N–10° S, 20–30° W (WTRA) whereminimum and maximum concentrations occurred in April (0.15 mgm–3) and in August (0.24 mg m–3), respectively.A significant empirical relationship between depth-integratedprimary production and in situ measured sea surface Chl a wasfound for ETRA, allowing us to estimate the seasonal cycle ofdepth-integrated primary production from SeaWiFS-derived Chla. As for Chl a, this model was verified in a small area ofthe Eastern Equatorial Atlantic (0–10° S, 0–20°W), although in this instance it was not completely able todescribe the magnitude and temporal variability of in situ primaryproduction measurements. The annual euphotic depth-integratedprimary production rate estimated for ETRA by our empiricalmodel was 1.4 Gt C year–1, which represents 16% of theopen ocean primary production estimated for the whole AtlanticOcean.  相似文献

Freezing of healthy and rust (Puccinia lagenophorae) infectedleaves of Senecio vulgaris was compared calorimetrically bythermal analysis. In fully expanded leaves the threshold freezingtemperature was in the range –6.8 to –8.4 °Cin controls but –3.0 to –5.1 °C in leaves withsporulating rust sori. Comparable values in expanding leaveswere –5.0 to –8.9 °C and –3.9 to –6.7°C for healthy and rusted tissues, respectively. The bulktissue freezing point was between –1.0 and –4.0°C in both fully expanded and expanding healthy leaves,and was increased by infection by between +0.2 and 2.5 °C.Whereas healthy leaves supercooled by 3.1–5.8 °C,rusted leaves supercooled by only 1.8–4.9 °C Supercoolingof control leaves was reduced by dusting with aeciospores, particularlywhen leaves were wounded to simulate the rupture of the surfacecaused by sporulation, but wounding alone had no significanteffect. Supercooling of distilled water was also significantlyreduced by aeciospores, suspended at a concentration of 105spores ml–1. It is concluded that rust-induced changes in leaf freezing inS. vulgaris grown in controlled environments were due to anincrease in the number of sites for ice nucleation, caused bythe presence of the aeciospores, and increased penetration ofice into internal tissues, resulting from damage to the cuticleand epidermis. Although data for frost resistance obtained inthe growth-room are similar to previous field observations,the role of the above mechanisms under field conditions remainsunproven. Carbon dioxide and water vapour exchanges for single attachedleaves of the temperate C4 grass Spartina townsendii were measuredunder controlled environment conditions in an open gas-exchangesystem. The responses of net photosynthesis, stomatal resistance,and residual resistance to leaf temperature and photon fluxdensity are described. The light and temperature responses ofnet photosynthesis in S. townsendii are compared to informationon these responses in both temperate C3 grasses and sub-tropicalC4 grasses. Adaptation of photosynthesis in this C4 speciesto a cool temperate climate is indicated both by the light andtemperature responses of net photo-synthesis. Unlike the C4grasses examined previously, significant rates of net photosynthesiscan be detected at leaf temperatures below 10?C. Rates of netphotosynthesis equal or exceed those reported for temperateC3 grasses at all of the temperature (5–40?C) and photonflax density (13–2500µmol m–2 s–1) conditionsexamined. Maximum rates of net photosynthesis in S. townsendiiare almost double those reported for C3 herbage grasses. Unliketemperate C3 grasses, the major limitation to net photosynthesisat low leaf temperatures (10?C and below) is the stomatal resistance,showing that the low residual resistance characteristic of C4species is maintained in S. townsendii even at low leaf temperatures.  相似文献

Enantiomers of the earthy smelling odorant ‘geosmin’(t-1, 10-dimethyl-t-9-decalol) were tested on 52 human subjectsfor odor threshold ratio (+)/(–) variability. Thresholdconcentrations of (+) and (–) and their (+)/(–)ratios appeared to be monomodally distributed. For most subjects,the ratio varied within a predictable narrow range comparableto that found previously for carvone enantiomers and contraryto the large variability found for -ionone enantiomers (Polaket al., 1989). Three marine phytoplankton species (Skeletonema costatum, Olisthodiscusluteus andGonyaulax tamarensis) were grown in batch culturesat 15°C and a 14:10 L:D cycle at irradiance levels rangingfrom 5 to 450 µEinst m–2 s–1. At each irradiance,during exponential growth, concurrent measurements were madeof cell division, carbon-specific growth rate, photosyntheticperformance (both O2 and POC production), dark respiration,and cellular composition in terms of C, N and chlorophyll a.The results indicate that the three species were similar withrespect to chemical composition, C:N (atomic) = 6.9 ±0.4, photo-synthetic quotient, 1.43 ± 0.09, and photosyntheticefficiency, 2.3 ±0.1 x 10–3 µmol O2 (µgChl a)–1 h–1 (µEinst m–2 s–1)–1.Differences in maximum growth rate varied as the –0.24power of cell carbon. Differences in growth efficiency, werebest explained by a power function of Chl a:C at µ = 0.Compensation intensities, ranged from 1.1 µEinst m–2s–1 for S. costatum to 35 forG. tamarensis and were foundto be a linear function of the maintenance respiration rate.The results indicate that interspecific differences in the µ–Irelationship can be adequately explained in terms of just threeparameters: cell carbon at maximum growth rate, the C:Chl aratio (at the limit as growth approaches zero) and the respirationrate at zero growth rate. Two approaches to quantifying relationships between nutrientsupply and plant growth were compared with respect to growth,partitioning, uptake and assimilation of NO3 by non-nodulatedpea (Pisum sativum L. cv. Marma). Plants grown in flowing solutionculture were supplied with NO3 at relative addition rates(RAR) of 0·03, 0·06, 0·12, and 0·18d–1, or constant external concentrations ([NO3)of 3, 10, 20, and 100 mmol m–3 over 19 d. Following acclimation,relative growth rates (RGR)approached the corresponding RARbetween 0·03–0.12 d-1, although growth was notlimited by N supply at RAR =0.18 d-1. Growth rates showed littlechange with [NO3–] between 10–100 mmol m–3(RGR=0·15 –0·16 d-1). The absence of growthlimitation over this range was suggested by high unit absorptionrates of NO3, accumulation of NO3 in tissues andprogressive increases in shoot: root ratio. Rates of net uptakeof NO3 from 1 mol m–3 solutions were assessed relativeto the growth-related requirement for NO3, showing thatthe relative uptake capacity increased with RGR between 0·03–0·06d–1 , but decreased thereafter to a theoretical minimumvalue at RGR  相似文献

The modulation of [3H]ACh release by nicotinic compounds was studied in superfused rat hippocampal synaptosomes loaded with [3H]choline. (−)-Nicotine (0.1–10 μM) evoked a dose-dependent increase in [3H]ACh release; higher concentrations were less effective. Nicotine-evoked release was Ca2+-dependent, and blocked by the nicotinic antagonists dihydro-β-erythroidine, mecamylamine, and pempidine. The α7-selective antagonist methyllycaconitine did not inhibit nicotine-evoked release when tested at 1 μM, although at 10 μM some attenuation of the response was observed. Six agonists tested were equally efficacious in stimulating [3H]ACh release, as judged by the maximum responses, and gave the following EC50 values: (±)-epibatidine 0.12 μM; (+)-anatoxin-a 0.14 μM; (−)-nicotine 0.99 μM; (−)-cytisine 1.06 μM; ABT-418 2.6 μM; isoarecolone 43 μM. Each agonist generated a “bell-shaped” dose response curve, suggesting desensitisation at higher concentrations. This is supported by analysis of repetitive stimulation with (−)-nicotine and (−)-cytisine: S2/S1 ratios declined sharply with increasing concentration, whereas subsequent KCl-evoked release remained constant. Simultaneous observations on extension growth and respirationrate (oxygen consumption) of 2-mm. sections excised from theextension zone of roots of pea (Pisum sativum) growing in distilledwater and 0·5 per cent. sucrose have yielded resultsclosely similar to those of Brown and Sutcliffe (1950). Respirationrate is not obviously correlated with growth rate either inwater or in sucrose, but it is strongly correlated with sectionlength. Respiration rate per unit section length (¬per unitfresh weight) shows a marked downward drift during extensionand is affected little by growth conditions. Tentative suggestionsare advanced to account for the small differences between driftsin o·5 per cent. sucrose and those in distilled water. Medium agitation produces an immediate and sustained stimulationof growth but no stimulation of oxygen uptake until the latergrowth stages. Thus respiration per unit section length is unaffectedby agitation at any stage. A typical growth response to ß-indolylacetic acid(IAA) was obtained, with a maximum stimulation (of about 35per cent.) at 1 part in 1011 and inhibitions increasing progressivelywith concentration beyond the threshold of about i part in 109.Both percentage stimulation and percentage inhibition of growthwere independent of the presence of sucrose. Respiratory responses to ß-indolylacetic acid werecomplex. In water no immediate response could be detected witheither a growth-stimulatory (10–11) or a growth-inhibitory(10–-8) concentration, while in 0·5 per cent. sucrosethe inhibitory concentration prevented the small immediate respiratoryrise due to the sucrose, probably by impeding sugar entry. Duringthe subsequent period of rapid growth (up to 36 hours) the smallrespiratory responses observed closely followed the small growthresponses to both concentrations of IAA, suggesting that theformer are the direct result of the differences in section lengthinduced by the auxin. When growth ceases (at 48 hours) sectionswhich have grown considerably in sucrose show respiratory ratesstill closely correlated with section length, whereas in waterboth concentrations of auxin induce marked depressions in respirationrate. It is concluded that ß-indolylacetic acid in bothgrowth-stimulatory and growth- inhibitory concentrations hasno direct effect on the activity of the respiratory enzyme systemof growing root cells. The small respiratory responses are bestexplained as resulting from differential changes in sectionsize and correlated changes in the enzyme complements of thegrowing cell.  相似文献

Rotifers are a relatively well-studied component of lacustrinesystems but their role is only poorly understood in estuaries.Three species of the genus Synchaeta—S. baltica, S. triophthalmaand S.cecilia—dominate the cold-water assemblage of rotifersin Chesapeake Bay. Laboratory experiments were conducted todetermine the temperature dependence of egg development time(EDT) for each species; EDT varied over an approximate rangeof 90–9 h as temperature (T) varied from 2 to 22°C.The EDT/temperature relationships could be closely fitted bya simple polynomial equation of the form log(EDT) = a + b(logT) + c(log T)2 for each species. Natural populations of thesethree rotifers were sampled during a cruise in the Potomac River(7–11 March 1983). Estimates of specific reproductiverates (b) were calculated based on the previously defined EDT/temperaturerelationship and the observed ratio of eggs/rotifers for eachspecies. The two most abundant species, S.triophthalma and S.cecilia,showed a clear dependence of b on the observed chlorophyll aconcentrations. Maximum reproductive rates ({small tilde}0.015h–1) were attained only at relatively high phytoplanktondensities within a bloom of Heterocapsa triquetra where thechlorophyll a concentrations exceeded 10 µg l–1.Estimates of secondary production suggest that Synchaeta spp.may contribute to the trophic flow of carbon in this systemwith a significance at least similar to that of the planktoniccopepods.  相似文献
