Summary Concentrations of phosphoadenylate nucleotides and the adenylate energy charge ((ATP+1/2ADP)/(ATP+ADP+AMP)) have been suggested as sensitive integrating measures of the energy state of organisms. This synoptic study investigated the seasonal and spatial variation of phosphoadenylate concentrations and AEC in two freshwater bivalve molluscs, the paper-shell clam, Anodonta imbecillis and the asian clam, Corbicula fluminea. Concentrations of all three adenylates, as well as the total adenylate concentration and adenylate energy charge of both species varied seasonally. These fluctuations were closely related to reproductive periods in both species. Total adenylate concentrations and ATP concentrations were slightly negatively correlated with shell length in A. imbecillis but the ADP and AMP concentrations and AEC were not significantly correlated with shell length. In C. fluminea the AEC was negatively correlated were positively correlated with shell length. Neither species exhibited significant differences in AEC between two collection locations. When C. fluminea collected from the Savannah River were acclimated and fed in the laboratory their AEC increased significantly.  相似文献

Adenylates (ATP, ADP, and AMP) may play a central role in the regulation of the O2-limited C and N metabolism of soybean nodules. To be able to interpret measurements of adenylate levels in whole nodules and to appreciate the significance of observed changes in adenylates associated with changes in O2-limited metabolism, methods were developed for measuring in vivo levels of adenylate pools in the cortex, plant central zone, and bacteroid fractions of soybean (Glycine max L. Merr cv Maple Arrow x Bradyrhizobium japonicum strain USDA 16) nodules. Intact nodulated roots were either frozen in situ by flushing with prechilled Freon-113(-156[deg]C) or by rapidly (<1 s) uprooting plants and plunging them into liquid N2. The adenylate energy charge (AEC = [ATP + 0.5 x ADP]/[ATP + ADP + AMP]) of whole-nodule tissue (0.65 [plus or minus] 0.01, n = 4) was low compared to that of subtending roots (0.80 [plus or minus] 0.03, n = 4), a finding indicative of hypoxic metabolism in nodules. The cortex and central zone tissues were dissected apart in lyophilized nodules, and AEC values were 0.84 [plus or minus] 0.04 and 0.61 [plus or minus] 0.03, respectively. Although the total adenylate pool in the lyophilized nodules was only 41% of that measured in hydrated tissues, the AEC values were similar, and the lyophilized nodules were assumed to provide useful material for assessing adenylate distribution. The nodule cortex contained 4.4% of whole-nodule adenylates, with 95.6% being located in the central zone. Aqueous fractionation of bacteroids from the plant fraction of whole nodules and the use of marker enzymes or compounds to correct for recovery of bacteroids and cross-contamination of the bacteroid and plant fractions resulted in estimates that 36.2% of the total adenylate pool was in bacteroids, and 59.4% was in the plant fraction of the central zone. These are the first quantitative assessments of adenylate distribution in the plant and bacteroid fractions of legume nodules. These estimates were combined with theoretical calculations of rates of ATP consumption in the cortex (9.5 nmol g-1 fresh weight of nodule s-1), plant central zone (38 nmol g-1 fresh weight of nodule s-1), and bacteroids (62 nmol g-1 fresh weight of nodule s-1) of soybean nodules to estimate the time constants for turnover of the total adenylate pool and the ATP pool within each nodule fraction. The low values for time constant (1.6-5.8 s for total adenylate, 0.9-2.5 s for ATP only) in each fraction reflect the high metabolic activity of soybean nodules and provide a background for further studies of the role of adenylates in O2-limited nodule metabolism.  相似文献

An assessment of energetic costs associated with P-glycoprotein (P-gp)-mediated xenobiotic efflux is important in understanding the energy budgets, tradeoffs, and fitness of organisms inhabiting contaminated environments. Here, a functional characterization and determination of the energetic costs associated with doxorubicin (DOX) efflux was examined in isolated hepatocytes of rainbow trout. The accumulation and efflux of DOX were both concentration dependent. The efflux of DOX over a 3 h incubation period resulted in a significant decrease in intracellular ATP concentrations (maximum decrease 25%) compared to control baseline levels, while significant increases in concentrations of ADP (max. 26%), AMP (max. 36%) and inorganic phosphate (max. 11%). were observed. In addition, significant reductions in the adenylate energy charge ([AEC]: max 11%), and phosphorylation potential ([PP]: max. 53%) were shown in cells incubated with DOX compared to control cells. Inhibition of DOX efflux (max. 61%) by the non-competitive P-gp inhibitor tariquidar (XR9576), demonstrated that changes in ATP, ADP, AMP, inorganic phosphate concentrations, AEC and PP in DOX-exposed hepatocytes were mainly due to P-gp activity. Overall, these results indicate that the exposure of trout hepatocytes to DOX increases energetic and metabolic costs that are associated specifically with P-gp efflux activity.  相似文献

1. Measurements of ATP, ADP and AMP concentrations in livers of rats that had been delivered by Caesarian section indicate a rapid shift from a low to a high [ATP]/[AMP] ratio. This change is consistent with the cessation of glycolysis and the initiation of gluconeogenesis at birth. 2. When newborn animals are exposed to a 100% nitrogen atmosphere the hepatic ATP concentration falls and AMP increases. 3. Calculations of the [ATP][AMP]/[ADP](2) ratio give values that are close to the equilibrium constant of adenylate kinase except when the ATP concentration is high. 4. This difference cannot be accounted for by the preferential binding of available Mg(2+) to ATP(4-) rather than ADP(3-). It is concluded that the relative proportions of adenine nucleotides at any level of phosphorylation are only partly regulated by adenylate kinase.  相似文献

Summary Pool sizes of ATP and ADP were analysed in freeze-stopped, lyophilised homogenates of needles from Norway spruce [Picea abies (L.) Karst.]. Control experiments in which possible changes in adenylate pools during sample acquisition were investigated did not reveal significant differences between needles taken from branches in situ or within a 30-min period after cutting off a branch. In addition, pool sizes of ADP and ATP were not affected by changes in light intensity (between 60 and 1500 E*-m-2*s-1), which inevitably occur when samples have to be taken from the upper region of older trees. Levels of ATP and ADP showed considerable seasonal changes (May through October) with the highest ratios of ATP/ADP in developing needles. In general, there was a tendency towards increased ratios of ATP/ADP with increasing needle age. This observation was corroborated by analyses of needles from spruce trees of different age and growing under different conditions. Needles from declining trees or from trees specifically fumigated with low concentrations of ozone and sulphur dioxide had significantly increased ratios of ATP/ADP compared to controls. The results are discussed with respect to physiological responses connected with natural senescence and induced ageing.  相似文献

Internal transport of O2 from the aerial tissues along the adventitious roots of intact maize plants was estimated by measuring the concentrations of adenine nucleotides in various zones along the root under an oxygen-free atmosphere. Young maize plants were grown in nutrient solution under conditions that either stimulated or prevented the formation of a lysigenous aerenchyma, and the roots (up to 210 mm long) were then exposed to an anaerobic (oxygen-free) nutrient solution. Aerenchymatous roots showed higher values than non-aerenchymatous ones for ATP content, adenylate energy charge and ATP/ADP ratios. We conclude that the lysigenous cortical gas spaces help maintain a high respiration rate in the tissues along the root, and in the apical zone, by improving internal transport of oxygen over distances of at least 210 mm. Changes in calcium and phosphorus concentrations, adenylate (AMP, ADP and ATP) levels, and ratios and ATPase activities of Macrobrachium nipponense were investigated during the moult cycle. Ca level in the exoskeleton was lowest in early postmoult (stage A), increasing at stages B and through intermoult (stage C) and peaking in premoult (stage D1 and D2). The P concentrations in the exoskeleton and muscle in late premoult and early postmoult stages were higher than those at other moult stages, and were lowest in the intermoult. Muscle adenylate energy charge (AEC) changed with moult stages, and was in agreement with the change in inorganic P level in the muscle. AEC may be a direct indicator of energy metabolic activity during the moult cycle. ATP/ADP and ATP/AMP ratios in premoult and postmoult stages were higher than that in intermoult stage. Na(+)-K(+)-ATPase activities of gills, muscles and hepatopancreatic of prawns were higher in early postmoult and late premoult animals, whereas they were lower in late postmoult, intermoult and early premoult animals. Gill residual ATPase activity was significantly higher in postmoult animals, while the peak value of hepatopancreatic residual ATPase activity appeared in intermoult stage.  相似文献

Nodulated soybeans (Glycine max L. Merr, cv. Maple Arrow) were exposed to various physiological and environmental treatments to determine the relationship between nodule adenylate pools and the degree of O2 limitation of nitrogenase. Adenylate energy charge (AEC = [ATP + 0.5 ADP]/[ATP + ADP + AMP]) and ATP/ADP ratios declined under conditions of decreased (10%) external pO2 but increased in nodules exposed to elevated (30%) external pO2. Nitrogenase activity was inhibited by both pO2 treatments, but recovered towards initial levels within 45 min. AEC also returned to initial levels during this period. To account for these and related data in the literature, it was hypothesized that 1) legume nodules regulate infected cell O2 concentration (Oi) to maintain adenylate pools at levels which limit respiratory metabolism: 2) treatments which decrease Oi alter the adenylate pools and further limit nodule metabolism; 3) treatments which increase Oi to levels in excess of a narrow range alter the adenylate pools and activate biochemical pathways which are not conducive to nitrogenase activity. In a preliminary test of these hypotheses, changes in AEC and ATP/ADP ratio were studied in nodules in which nitrogenase activity was inhibited by stem girdling, nitrate fertilization and exposure to an Ar:O2 atmosphere. All three treatments caused an increased O2 limitation of nodule respiration and nitrogenase activity. However, decreases in AEC were observed only in the stem girdling and nitrate fertilization treatment: Ar:O2 exposure had no effect on whole nodule AEC. While this result challenged the hypotheses suggesting a central role for adenylates in the regulation of O2-limited metabolism, it was noted that the Ar:O2 treatment would differ from the other treatments in that it would have a specific effect on the ATP demands for NH3 assimilation in the plant fraction. Since AEC and ATP/ADP ratio would be affected by both the rate of ATP synthesis (potentially an O2-limited process) and the demand for ATP, changes in these parameters in the whole nodule may not be a reliable indicator of adenylate-mediated O2 limitation. Futher studies are needed to examine in vivo changes in adenylate pools in the plant and bacteroid fractions in nodules which vary in their degree of O2-limited metabolism.  相似文献

The effects of different doses (0.01-0.1-1.0-10.0/mg/kg-1) of beta-carotene were studied on gastric secretory responses of 4 hr pylorus-ligated rats: development of gastric mucosal damage (as assessed by number and severity of lesions) produced by intragastric administration of 0.6 M HCl; tissue level of adenosine triphosphate (ATP), adenosine diphosphate (ADP), adenosine monophosphate (AMP), adenylate pool (ATP + ADP + AMP), ratio of ATP X ADP-1, "energy charge" (ATP + 0.5 ADP X X (ATP + ADP + AMP)-1) (during the development of gastric mucosal damage by 0.6 M HCl and of gastric cytoprotection by beta-carotene. It was found that beta-carotene did not decrease the gastric secretory responses of 4 hr pylorus-ligated rats; The development of gastric mucosal damage could be decreased dose-dependently by the administration of beta-carotene; the ATP transformation could be decreased by beta-carotene; the tissue levels of cAMP and AMP could be increased significantly and dose-dependently by beta-carotene; the ratio of ATP X ADP-1 could be increased significantly and dose-dependently by beta-carotene; the values of adenylate pool and "energy charge" remained unchanged.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

To assess the role of O2 in the regulation of nodule metabolism following a decrease or an increase in temperature, the fractional oxygenation of leghemoglobin (FOL) was measured in soybean (Glycine max L. Merr.) nodules during rapid and gradual changes in temperature from 20[deg]C to either 15 or 25[deg]C. The affinity of leghemoglobin for O2 was also measured at each temperature and the values were used to calculate the infected cell O2 concentration (Oi). After nodules were transferred to 15[deg]C, FOL and Oi increased and adenylate energy charge (AEC = [ATP + 0.5ADP]/[ATP + ADP + AMP]) increased from 0.70 to 0.78. The temperature increase was associated with a decrease in FOL and Oi. We concluded that changes in nodule temperature alter the respiratory demand of the nodules for O2, resulting in a change in Oi and a shift in the balance between ATP consumption and ATP production within the nodule tissue.  相似文献

To escape from starfish predators, giant scallops, Placopecten magellanicus, swim using series of strong phasic contractions interrupted by tonic contractions. To investigate whether these tonic contractions allow metabolic recuperation of the adductor muscle, we sampled scallops at rest (Control), after an initial series of phasic contractions (Phasic) and after 1 min of tonic contraction following their initial phasic contractions (Phasic + Tonic) and compared muscle levels of phosphoarginine, adenylate nucleotides (ATP, ADP and AMP) and adenylate energy charge (AEC). Scallops in the two active groups did not differ in the numbers of phasic contractions or the mean phasic force production. Phosphoarginine concentrations in the adductor muscle decreased with phasic activity and remained low after 1 min of tonic contraction. ATP and ADP and total adenylate levels did not differ between the three groups, but AMP levels were higher in the scallops sampled after phasic contractions than in control scallops. The AEC was reduced by phasic contractions but returned to control levels after 1 min of tonic contraction. A significant negative correlation between AEC and the number of claps in the Phasic group disappeared in the Phasic + Tonic group. Thus, tonic contractions following phasic contractions allow partial metabolic recovery of the adductor muscle by returning AEC to control levels. However, phosphoarginine levels did not recover during tonic contractions, and a negative correlation between the number of claps and phosphoarginine levels remained in the Phasic + Tonic group. By interspersing tonic contractions between series of phasic contractions, scallops improved muscle energetic status, which should help maintain phasic force production during the remainder of the escape response.  相似文献

The contents of ATP, ADP and AMP were determined by HPLC and adenylic energy charge (AEC) was estimated during different stages of the embryonic development of Bufo arenarum up to the tailbud stage. All the developmental stages studied showed a high ATP content (about 1.04-1.48 nmol/emb.). The concentration of ADP was low (0.025-0.041 nmol/emb.) but rose slightly at the neural tube stage. AMP was undetectable before the tailbud stage. AEC values were almost constant (about 0.987-0.992) throughout the period studied. Only a fall at the tailbud stage could be detected which can be related to this more advanced cellular differentiation stage.  相似文献

The ATP content in human erythrocytes depleted without glucose falls down to half of the initial value within 2-3 hours and reaches practically zero within more than 10 hours. The ADP content increases 2-3-fold during the 1st hour after depletion and then slowly decreases. The AMP content increases 10-fold during several hours, but the rate of this process constantly decreases. The adenylate pool decreases at a constant rate ranging from 0.13 to 0.25 mmol/l cell. h; this is accompanied by accumulation of IMP. Addition of glucose to depleted erythrocytes results in partial recovery of the ATP level within 1-2 hours. The sooner glucose addition after the depletion, the greater the recovery. Simultaneously the ADP and AMP levels drastically decrease to new constant values. The decline of the adenylate pool ceases and the rate of IMP accumulation increases. Normally, the [ATP]/adenylate pool ratio lies within the small interval 0.85-0.94 irrespective of significant individual differences in the absolute values of [ATP]. This ratio is decreased during depletion and restored to the initial value after glucose addition. The mass-action ratio of the adenylate kinase reaction changes greatly during depletion and restoration of erythrocyte ATP.  相似文献

The cytosolic and mitochondrial contents in ATP, ADP and AMP were measured in islets incubated for 45 min at increasing concentrations of D-glucose and then exposed for 20 s to digitonin. The latter treatment failed to affect the total islet ATP/ADP ratio and adenylate charge. D-Glucose caused a much greater increase in cytosolic than mitochondrial ATP/ADP ratio. In the cytosol, a sigmoidal pattern characterized the changes in ATP/ADP ratio at increasing concentrations of D-glucose. These findings are compatible with the view that cytosolic ATP participates in the coupling of metabolic to ionic events in the process of nutrient-induced insulin release.  相似文献

The Site of Oxygen Limitation in Soybean Nodules   总被引:4,自引:3,他引:1  
In legume nodules the [O2] in the infected cells limits respiration and nitrogenase activity, becoming more severe if nodules are exposed to subambient O2 levels. To identify the site of O2 limitation, adenylate pools were measured in soybean (Glycine max) nodules that were frozen in liquid N2 before being ground, lyophilized, sonicated, and separated on density gradients of nonaqueous solvents (heptane/tetrachloroethylene) to yield fractions enriched in bacteroid or plant components. In nodules maintained in air, the adenylate energy charge (AEC = [ATP + 0.5 ADP]/[ATP + ADP + AMP]) was lower in the plant compartment (0.65 ± 0.04) than in the bacteroids (0.76 ± 0.095), but did not change when the nodulated root system was exposed to 10% O2. In contrast, 10% O2 decreased the bacteroid AEC to 0.56 ± 0.06, leading to the conclusion that they are the primary site of O2 limitation in nodules. To account for the low but unchanged AEC in the plant compartment and for the evidence that mitochondria are localized in O2-enriched microenvironments adjacent to intercellular spaces, we propose that steep adenylate gradients may exist between the site of ATP synthesis (and ADP use) in the mitochondria and the extra-mitochondrial sites of ATP use (and ADP production) throughout the large, infected cells.  相似文献

Adenylate kinase (ATP:AMP phosphotransferase, EC 2.7.4.3) isolated from porcine skeletal and heart muscle and from rabbit muscle are inactivated when a single arginine residue is modified. In adenylate kinase from pig the modified residue was identified as Arg-97 by peptide-mapping. In native adenylate kinase Arg-97 is located at the bottom of the active site cleft. The protein fluorescence of modified adenylate kinase is reduced. Whereas the addition of AMP, ADP and MgATP quench the fluorescence of native adenylate kinase, the fluorescence of phenylglyoxal-modified adenylate kinase is only affected by ADP and MgATP. This finding is discussed in connection with the structural isomerization observed in native adenylate kinase by X-ray diffraction analysis.  相似文献

Gastric ulcer was elicited in rats by reserpine (5 mg x kg-1 sc.) administration. Ulcer formation (number and severity) was measured 6, 12, 18 and 24 hr after reserpine administration. At the time of killing of the animals, tissue levels of adenosine triphosphate (ATP), adenosine diphosphate (ADP), adenosine monophosphate (AMP), cyclic adenosine monophosphate (cAMP) were measured enzymatically and by radioimmunoassay in the gastric fundal mucosa. The sum of ATP + ADP + AMP (adenylate pool) and the ratio of ATP x ADP-1 were calculated. It was found that (1) the tissue levels of ATP, AMP, cAMP, sum of ATP / ADP + AMP (adenylate pool) and ratio of ATP x ADP-1 increased significantly in the gastric fundal mucosa 6 hr after reserpine administration, thereafter these values decreased gradually and significantly; (2) the tissue level of ADP increased significantly in the gastric fundal mucosa 6 hr after reserpine administration, meanwhile its level increased significantly at 18 and 24 hr; (3) the value of energy charge (ATP + 0.5 ADP x ATP + ADP + AMP-1) remained unchanged; (4) the peaks of biochemical alterations in the gastric fundus mucosa preceded he appearance of ulcers. It was concluded that (1) reserpine ulcer appears after an active metabolic response in the rat gastric fundal mucosa; (2) hypoxaemic damage in the gastric fundal mucosa can be excluded as a possible underlying mechanism of ulcer formation produced by reserpine administration; (3) before the appearance of reserpine ulcer, significant changes in the feedback mechanism, system, i.e. between the ATP--membrane ATPase--ADP and the ATP--adenylate cyclase--cAMP energy systems, can be observed in the rat gastric fundal mucosa.  相似文献

Various physiological and biochemical methods have been proposed for assessing the effects of environmental perturbation on aquatic organisms. The success of these methods as diagnostic tools has, however, been limited. This paper proposes that adenylate energy charge overcomes some of these limitations. The adenylate energy charge (AEC) is calculated from concentrations of adenine nucleotides ([ATP+½ADP]/[ATP+ADP+AMP]), and is a reflection of metabolic potential available to an organism. Several features of this method are: correlation of specific values with physiological condition or growth state, a defined range of values, fast response times and high precision. Several examples from laboratory and field experiments are given to demonstrate these features. The test organisms used (mollusc species) were exposed to a variety of environmental perturbations, including salinity reduction, hydrocarbons and low doses of heavy metal. The studies performed indicate that the energy charge may be a useful measure in the assessment of environmental impact. Its use is restricted, however, as several limitations exist which need to be fully evaluated. Further work relating values to population characteristics of multicellular organisms needs to be completed before the method can become a predictive tool for management.  相似文献

We analysed changes in AMP, ADP, and ATP concentrations and adenylate energy charge in Norway maple (Acer platanoides L.) and European beech (Fagus sylvatica L.) seeds during dormancy breaking (at 3 °C) and in the control variant at 15 °C. Values of the studied indicators in stratified beech seeds were generally higher at 15 °C, at least until germination (+3 °C). By contrast, in maple seeds, the values recorded during dormancy breaking by cold stratification were much higher than at 15 °C. Three peaks (usually in weeks 3, 6, and 8) were observed in maple seeds at 3 °C, but not at 15 °C. Among adenine nucleotides, AMP reached the highest levels in both species in both variants of the experiment.  相似文献

Carnosine is a naturally occurring dipeptide (beta-alanyl-L-histidine) found in muscles, brain and other tissues. This study was designed to test the ability of carnosine to offset metabolic disturbances induced by Schistosoma mansoni parasitism. Results indicate that parasitic infection caused elevation of liver weight/body weight in S. mansoni-infected hamsters, induced lipid peroxidation and reduced glycogen levels. Moreover, adenylate energy charge (AEC) and ATP/ADP and ATP/AMP concentration ratios were markedly lower in infected hamsters. Administration of carnosine (10 mg/day) for 15 days concurrent with infection effectively reduced worm burden and egg count. Administration of carnosine 2 and 4 weeks post-exposure only partially ameliorated the S. mansoni effects on metabolism. Carnosine treatment also normalized most of the parameters measured, including glycogen repletion, the antioxidant status and AEC. These finding support the use of carnosine for possible intervention in schistosomiasis.  相似文献
