Papilla palatina, nasopalatine duct and taste buds of young and adult rats   总被引:1,自引:0,他引:1  
The morphology of the papilla palatina, the nasopalatine ducts and the taste buds situated within these ducts was studied in pups and adult rats using light and electron microscopy. During development, the papilla palatina grew in width and depth, becoming a protuberance in weanlings and adults. The nasopalatine ducts enlarged and two folds of the lateral walls of the ducts differentiated, reducing the width of the tubes in the region of the oral openings. Taste buds appeared postnatally. Light, dark, and perigemmal cells were found in all stages studied, but light cells were scarce up to 8 days of age. The taste pore appeared between 11 and 13 days of age; it lacked electron-dense material of cellular origin. Synaptic-like images could be found only in relation to dark cells. The papilla palatina, the nasopalatine ducts and the taste buds were fully developed by the 3rd week of life.  相似文献

The anatomy of the vomeronasal complex and, in connection with this, the structures of the rostral palate were studied in different species of mammals, namely members of the order Marsupialia, Scandentia, Insectivora, Primates, Rodentia, and Lagomorpha. The following results were obtained: The organs of Jacobson of all forms studied are well-developed. The organ of Jacobson is situated at the base of the nasal septum and opens rostrally, always closely connected to the nasopalatine duct. Even in rodents, lagomorphs and Solenodon, where the openings of the organs are positioned rostral to the ductus, both systems are nevertheless connected by means of special furrows. Accordingly the organs of Jacobson are functionally much more closely related to the oral cavity than to the nasal cavity, which they actually belong to. This can be emphasized by the peculiar structures of the rostral palate inclosing the papilla palatina and with it the oral openings of the nasopalatine ducts. In all species studied, the anterior part of the upper jaw presents a very interesting situation because the median furrow of the rhinarium communicates directly or indirectly with the sulcus papillae palatinae, thus forming a very distinct system of grooves which preserves a connection between the nasopalatine ducts and the preoral surroundings. In rodents, lagomorphs, and Solenodon, we find in this part of the palate a special situation because of their unusually arranged incisors, which are not separated by a diastema. However, also in these cases, there are distinct connecting passages between the papilla palatina and the extraoral surroundings. The conditions found in Ratufa bicolor and in early stages of the rat demonstrate that the extraordinary topography of the rostral palate in rodents is a secondary formation by means of ontogeny and phylogeny. Cebus apella, a platyrrhine simian, shows already a clear reduction of palatal structures compared to those found in prosimians. In Setifer setosus and Echinops telfairi, we find the papilla palatina and with it the oral openings of the nasopalatine ducts overgrown by a bipartite caudal branch of the rhinarium. The neonate Setifer allows us to reconstruct the mechanism of this overgrowing procedure. We find a similar situation in Erinaceus, where the papilla palatina remains uncovered, however. Because of contradictory bibliographical data, some elements of the vomeronasal complex in mammals needed to be carefully analysed in regard to structure and nomenclature: in many species the paraseptal cartilage bifurcates rostrally into a dorsal and a ventral branch.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Summary Tonsils of human fetuses at the 8th to the 28th gestational week (g.w.) were investigated by electron microscopy, enzyme histochemistry, and immunohistochemistry on cryostat sections. The development of the tonsilla palatina starts during the 14th g.w. when the mesenchyme underlying the mucous membrane of the tonsillar cavity becomes invaded by mononuclear wandering cells. In fetuses of about the 16th g.w. epithelial crypts grow down into the connective tissue and are infiltrated by T-lymphocytes. At the same time, precursors of interdigitating cells (IDC) can be identified among the epithelial cells. Frequently, lymphocytes and IDC-like cells are in close contact. From these findings it is concluded that the infiltrated crypt epithelium in the human tonsilla palatina represents a T-cell region. Primary follicles develop in earlier fetal stages than in all other secondary lymphoid organs. They contain precursors of dendritic reticulum cells and lymphoid cells that belong to the B-cell line. These primary follicles may be considered as the first assemblage of B-cell regions in human fetal lymphoid tissue. Summary In the present study attention was focussed on several lymphoid subpopulations and specific stationary cells of the human tonsilla palatina. They were labeled at the light- and electron-microscopic levels by means of monoclonal antibodies to cell surface antigens. Cells resembling interdigitating cells (IDC-like cells) within the crypt epithelium and the interdigitating cells in the parafollicular T-cell region express the HLA-DR antigen. This fact suggests a relationship between these two populations of cells. Both cell types were frequently found in close contact to T-helper cells labeled with Anti-Leu 3a. This fact is discussed as a confirmation of earlier suggestions that the tonsillar crypt epithelium serves as T-cell region. Cytotoxic/ suppressor-T cells (OKT 8 +) and Leu 7-positive cells do not appear to contact interdigitating cells. Anti-Leu 7 is a monoclonal antibody, that defines a differentiation antigen shown to be selectively expressed on human natural killer cells (NK-cells). With the use of the immuno-electron-microscopic labeling method it was possible to analyze the ultrastructure of this lymphoid subpopulation. Two morphologically distinguishable subtypes of Leu 7-positive cells populate different microenvironments: The Leu 7-positive large-granular lymphocyte was predominantly found in the crypt epithelium, while numerous Leu 7-positive cells located in the germinal centers had the appearance of small lymphocytes. This finding is discussed in favour of distinct phenotypes representing different stages in a differentiation pathway of the maturing NK-cell: Small Leu 7-positive lymphocytes in the germinal centers are supposed to be functionally inactive precursors, and only the Leu 7-positive large granulated lymphocytes in the crypt epithelium may represent differentiated active NK-cells. The structures of the rostral palate in regard to the vomeronasal complex of different species of mammals were studied. In all cases, we find a very interesting system of furrows which preserves a connection between the nasopalatine ducts and the preoral surroundings. For rodents, lagomorphs, Solenodon, Setifer, and Echinops, we find a special situation in this part of the palate. Here the incisors are not separated by a diastema nor the oral openings of the nasopalatine ducts are overgrown by a bipartite caudal branch of the rhinarium. The results of the anatomic studies of the vomeronasal complex and the rostral palate of the mammals investigated are discussed: First of all, some elements of the vomeronasal complex needed to be analysed in regard to structure and nomenclature, specifically the cartilago paraseptalis with its outer bar, the cartilago ductus nasopalatini and the cartilago palatina. Because of 2 criterions, the vomeronasal complex could be classified as either primitive or progressive. We find a primitive one in Didelphis, Tupaia, Solenodon, Oryctolagus, and all rodents. In contrast, the other insectivores studied and all primates show progressive structures at their vomeronasal complex. Finally, conclusions in regard to the function of the organs of Jacobson are derived from these studies. The significance of the "flehmen" mechanism for the functioning of the organs is questioned.  相似文献

Changes in the oral cavity resulting from the loss of teeth and the ensuing reconstruction of a set of teeth by dentures (partial or complete) may cause changes in the speech and voice of the patient. The aim of the present investigation was to study the changes in speech and voice in patients suffering from teeth loss and the degree of speech improvement using dentures. Voice and speech parameters of a set of tested syllables were analysed in 10 patients at the 2nd Clinic of Stomatology. The analysis was carried out by means of an FFT, SoundForge 5.0 programme. Differently expressed acoustic changes in both consonants and vowels were ascertained in a percentage of the patients under examination. These concerned especially the sibilant ("s", "(see text)"), labiodental ("f", "v") and vibrating ("r", "(see text)") consonants. Changes in the FFT spectrum and air leakage in constrictive consonants were also found. In some patients the vowels, especially the closed ones ("i", "u"), may change their fundamental frequency and show noise admixture manifested as a blurred delimitation of the formants. A denture should, inter alia, render it possible for the patient to produce the same articulation to which he/she had been accustomed before the loss of teeth. For the construction of dentures the most important factors from a phonetic point of view appear to be the following: overbite, overjet, the height of the plate, the thickness of the palatal material, the incisor position, and the modelling of the ruga palatina on the hard palate. In case of wrong denture construction the acoustic changes may continue, resulting in the patient's stress load dependent upon sex, age, psychic condition and seriousness of the problem.  相似文献

Nycticebus coucang is typically strepsirrhine. This condition is basically the same in all prosimians except Tarsius. The rhinarium shows a labial part; the marked furrow in its median plane clefts the margin of the upper lip. The latter is attached to the gum between the median incisors by means of paired folds (not corresponding to a frenulum labii superioris). The labial cleft is continued in the sulcus papillae palatinae. The ductus vomeronasalis opens into the ductus nasoplatinus which, in turn, enters the aboral part of the sulcus palatinus. Thus strepsirrhinism provides, occasionally, an open communication of the olfactory systems (nose, vomeronasal organ) with the environment in front of the rhinarium. Strepsirrhine primates possess an internarium very similar to the one of platyrrhine monkeys. The shape of the nostrils is not characteristics for strepsirrhinism. A split-lip condition is likely ancestral to the complete lip condition, since it is found in several stems of mammals, especially in archaic forms. Strepsirrhinism, such as in lemuroid prosimians, is probably just one case of the primitive split-lip condition. Therefore, a median furrow in the external hairy skin of the upper lip, found in some platyrrhine monkeys, could be a rudiment of a cleft, indicating that a kind of a split lip condition was ancestral to the undivided lips of platyrrhine monkeys. Taste buds occur in the epithelium of the lateral surface of the papilla palatina of Nycticebus. Other lorisid prosimians have not been studied in this respect. The taste buds test water soluble substances entering the sulcus either way. These substances may come from the environment entering the sulcus through the labial cleft.  相似文献

An intramembranous ossification at the anterior end of the cartilaginous nasal capsule is described for the first time in prenatal specimens of the anteaterTamandua and the slothCholoepus and redescribed in prenatal specimens of the armadillosDasypus andZaedyus. From comparisons of this bone with the septomaxilla of monotremes and various Mesozoic mammals, it is concluded that (1) the bone inTamandua andCholoepus is homologous with the central part (processus ascendens) of the bone inDasypus, Zaedyus, and other armadillos and (2) the xenarthran processus ascendens, in turn, is homologous with the central part of the septomaxilla of monotremes and various Mesozoic mammals. Therefore, the bone in question in xenarthrans is a true septomaxilla. It is further concluded that the armadillo septomaxilla has two neomorphic components: a lamina palatina beneath the cartilaginous nasal floor and a processus intrafenestralis extending rostrally into the nasal fossa.  相似文献

Partially purified thymidylate synthetase from Plasmodium berghei and mouse reticulocytes was characterized. The mol. wt of the enzyme from P. berghei was about twice that from mouse reticulocytes. The optimum pH of the enzyme from P. berghei was found to be 6.5-7.5 while that from the host was 7.0-8.0. The enzyme from P. berghei was more susceptible to pH denaturation than the enzyme from reticulocytes. The enzyme from both sources differed in their Km values for substrates. The enzyme from reticulocytes was less sensitive to inhibition by substrate analogs than that from P. berghei.  相似文献

Arginase from rat fibrosarcoma was purified about 1900-fold and its properties were compared with those of the enzyme from liver and kidney. Arginase from fibrosarcoma was a neutral protein of molecular weight 120,000 with a Km value of 11 mM for arginine. The activation energy was 7.2 kcal/mol and the pH optimum was 10. The fibrosarcoma enzyme was immunologically different from that of the liver. The arginase from fibrosarcoma closely resembled the arginase from the kidney in its electrophoretic, kinetic and immunological properties.  相似文献

The cDNA for prostaglandin endoperoxide synthase (cyclooxygenase) was cloned from human platelets by the polymerase chain reaction amplification method, and the primary structure of the enzyme was deduced from the nucleotide sequence. The enzyme was composed of 599 amino acids including 23-amino acid signal sequence, and the calculated molecular weight of the mature protein was 65,995. The enzyme was immunoaffinity-purified from human platelets. The N-terminal amino acid sequence determined by Edman degradation was Ala-Asp-Pro-Gly-Ala-Pro-Thr-Pro-, and the result confirmed the primary structure of the enzyme, which was deduced from the cDNA sequence.  相似文献

The pH optimum of the ATPase activity in plasma membranes from Saccharomyces cerevisiae NCYC 431 from 8 h cultures was around 6.5 and that in membranes from organisms from 16 h cultures near 6.0. The Km[ATP] of the enzyme was virtually unaffected by the age of the culture from which organisms were harvested, although the Vmax of the enzyme in membranes from organisms from 8 h cultures was higher than that for organisms from 16 h cultures. Ethanol non-competitively inhibited ATPase activity in membranes, although the inhibition constant for the enzyme from organisms from 8 h cultures was lower than that from organisms from 16 h cultures. Glycine accumulation by the general amino acid permease was non-competitively inhibited by ethanol. Inhibition constants were virtually the same for glycine uptake by deenergized organisms from 8 h and 16 h cultures, but under energized conditions the value was greater for organisms from 16 h rather than 8 h cultures. The data indicate that inhibition of plasma-membrane ATPase activity by ethanol could account, at least in part, for inhibition of glycine accumulation by ethanol.  相似文献

The spontaneous and potassium-stimulated release of endogenous taurine and gamma-aminobutyric acid (GABA) from cerebral cortex and cerebellum slices from adult and developing mice was studied in a superfusion system. The spontaneous release of GABA was of the same magnitude in slices from adult and developing mice, but the spontaneous release of taurine was considerably greater in the adults. The potassium-stimulated release of GABA from cerebral cortex slices was about five times greater in adult than in 3-day-old mice, but the potassium-stimulated release of taurine was more than six times greater in 3-day-old than in adult mice. In cerebellar slices from 7-day-old mice, potassium stimulation also evoked a massive release of taurine, whereas the evoked release from slices from adult mice was rather negligible. Also in cerebellar slices the potassium-stimulated release of GABA exhibited the opposite quantitative pattern. The stimulated release of both GABA and taurine was partially calcium dependent. The results suggest that taurine may be an important regulator of excitability in the developing brain.  相似文献

采用随机完全区组设计对5年生麻疯树Jatropha curcas种源林进行结实性调查。结果表明,不同种源麻疯树单株产种量有不同程度的差异,云南元阳种源最高,达218.96 g,四川盐边种源最低,仅为48.42 g。不同种源麻疯树种子平均含油率也有所不同,广西隆林种源最高,达42.80%,贵州望谟种源最低,为37.23%。单株产种量与种源地纬度、年均气温显著相关,而与种源地无霜期极显著相关,其中与纬度为负相关,与年均气温、无霜期为正相关,说明纬度较低、气温较高和无霜期较长的种源在福建平和县的单株产种量高。而种子平均含油率与各地理气象因子之间没有显著相关性。根据单株产种量和种子平均含油率聚类分析,分别将9个种源分成优、良、中、差4个不同的类别,综合来看,云南东南部和广西西北部可能是麻疯树的优良种源区,该区域内的5个种源,单株产种量达147.43 g,种子平均含油率达41.07%。  相似文献

The gene-encoding HMT2-like sulfide dehydrogenase from Bacillus stearothermophilus JCM2501 was amplified and expressed in Escherichia coli and the enzymatic features were examined. The enzyme was detected mainly in the membrane fraction. It catalyzed the sulfide-dependent menaquinone (MK) reduction showing special enzymatic features distinct from other sulfide-quinone oxidoreductases (SQRs) from autotrophic bacteria. The purified protein from E. coli brought about the sulfide-dependent 2,3-dimethyl-1,4-naphthoquinone (DMN) reduction in vitro. The reduction was accelerated in the presence of either cyanide or 2-mercaptoethanol and phospholipids. The high reduction was followed by a change in Km values for sulfide and DMN. The purified enzyme utilized MK as an electron acceptor in the membrane fraction from E. coli. Under anaerobic conditions, sulfide was oxidized with reduction of fumarate or nitrate via the MK pool. The dehydrogenase was different from SQR in autotrophic bacteria in terms of the low affinity for sulfide and the activity enhancement in the presence of cyanide or 2-mercaptoethanol. The sulfide oxidation via MK in the cellular membrane of Gram-positive bacteria was certified.  相似文献

本实验取10只Wistar大鼠的睾丸和附睾,睾丸石蜡切片,附睾精子涂片后用苯胺蓝染色显示赖氨酸含量。结果是睾丸生精小管中精原细胞和精母细胞染色较深即赖氨酸含量较高,精子细胞和精子染色渐淡即赖氨酸含量降低,而附睾精子显示,在附睾头部的精子染色较深,附睾尾部的精子几乎不着色,应用显微分光光度计测定附睾精子,计算出头部的精子赖氨酸含量在1左右,尾部的精子赖氨酸含量接近于零。本实验还检测了10例正常人及10例不育者精子的赖氨酸,结果为正常人精子的赖氨酸含量较低,不育者精子赖氨酸含量高且畸形率也高。提示精子赖氨酸含量高是核蛋白转型异常的征象,可能是男性不育的一个重要原因。  相似文献

The outer membrane (OM) of Fibrobacter succinogenes was isolated by a combination of salt, sucrose, and water washes from whole cells grown on either glucose or cellulose. The cytoplasmic membrane (CM) was isolated from OM-depleted cells after disruption with a French press. The OM and membrane vesicles isolated from the extracellular culture fluid of cellulose-grown cells had a higher density, much lower succinate dehydrogenase activity, and sodium dodecyl sulfate-polyacrylamide gel electrophoresis protein profiles different from those of the CM. The OM from both glucose- and cellulose-grown cells and the extracellular membrane vesicles from cellulose-grown cultures exhibited higher endoglucanase, xylanase, and acetylesterase activities than the CM and other cell fractions. Endoglucanase 2 was absent from the isolated OM fractions of glucose- and cellulose-grown cells and from the extracellular membrane vesicles of cellulose-grown cells but was present in the CM and intracellular glycogen granule fractions, while endoglucanase 3 was enriched in the OM. Cellobiosidase was located primarily in the periplasm as previously reported, while cellobiase was mainly present in the glycogen granule fraction of glucose-grown cells and in a nongranular glycogen and CM complex in cellulose-grown cells. The cellobiase was not eluted from glycogen granules by cellobiose, maltose, and maltotriose nor from either the granules or the cell membranes by nondenaturing detergents but was eluted from both glycogen granules and cell membranes by high concentrations of salts. The eluted cellobiase rebound almost quantitatively when diluted and mixed with purified glycogen granules but exhibited a low affinity for Avicel cellulose. Thus, we have documented a method for isolation of OM from F. succinogenes, identified the OM origin of the extracellular membrane vesicles, and located glycanases and cellobiase in membrane and glycogen fractions.  相似文献

Malic enzyme was found in both bean rust and cat stem rust uredospores. In bean rust uredospores it was shown to catalyze the formation of pyruvic acid from l-malic acid and to synthesize malic acid from pyruvic acid and CO2. The malic enzyme from bean rust uredospores was specific for NADP and dependent on manganous ions for activity. The specific activity of the bean rust malic enzyme in crude extracts of ungerminated uredospores was approximately 6 times greater than that found in crude extracts obtained from germinated uredospores. The malic enzyme was also found in extracts obtained from healthy and rust-infected bean leaves. The specific activity of the enzyme was approximately 2 to 5 times greater in partially purified extracts obtained from the infected bean tissue at 6 days after inoculation. The specific activity of the malic enzyme in crude extracts obtained from oat stem rust uredospores was 2 times greater than the specific activity of this enzyme in crude extracts obtained from bean rust uredospores. Phosphoenolpyruvate carboxylase activity could not be demonstrated in crude extracts obtained from the ungerminated uredospores of the bean rust fungus.  相似文献
