ph1b基因在Aegilops有益基因直接遗传转移中利用的可能性   总被引:1,自引:1,他引:0  
第一次用中国春和中国春ph1b突变体对(中国春phlb突变体×Ae.uariabilis)F_1和(中国春ph1b突变体×Ae.turcomenica)F_1回交获得了成功,并通过连续回交,把Ae.turcomenica的抗白粉基因转移到了普通小麦中。证实了利用ph1b基因从山羊草属的一些种“直接遗传转移”有益基因到普通小麦中的可能性。  相似文献

在普通小麦地方品种自然群体中天然存在促进小麦-外源杂种部分同源染色体配对的基因phKL。本研究比较了phKL基因与人工Ph基因突变系诱导小麦-Aegilops variabilis及小麦-黑麦杂种部分同源染色体配对的作用大小。研究结果表明,诱导小麦- Ae. variabilis(或黑麦)部分同源染色体配对作用的顺序是ph1b > phKL > ph2b > ph2a,即phKL基因的作用介于Ph1与Ph2突变体之间。  相似文献

蓝粒小麦与太谷核不育小麦间染色体片段转移的研究   总被引:3,自引:1,他引:2  
本文研究的目的是对太谷核不育小麦籽粒进行颜色标记,以便进一步利用太谷核不育小麦。以蓝粒小麦4D-4E二体异代换系作为际记性状供体,利用中国春小麦ph突变体诱导部分同源染色体联会,增加同组染色体间的交换,使4D染色体上的Tai基因与4E染色体上的蓝胚乳基因连锁。在本实验杂交组合[(Taitai×phph)×蓝粒小麦]×phph~2 F_3中得到一个蓝粒高不育穗行,经分析与继代观察,初步认为这一穗行为4E带有蓝胚乳基因的染色体片段转侈到4D染色体上,使蓝胚乳基因与Tai基因发生不完全连锁。同时对ph突变体诱导部分同源染色体联会的作用等问题进行了探讨。  相似文献

用中国春ph2b突变体(Triticum aestivum L.cv.Chinese Spring)与华山新麦草(Psathyrostachys huashanica Keng)杂种F1的部分进行自交,而另一部分与3个不同的普通小麦品种以及中国春ph2b突变体进行回交,并对自交和回交一代的形态学和细胞学进行研究.结果表明:(1)含有AABBDDNs的自交F2的花粉母细胞减数分裂中期Ⅰ平均单价体数都在7以上,说明来自华山新麦草的Ns基因组可引起普通小麦ABD基因组中某一个(几个)同源组发生不联会或联会消失;(2)在回交一代(F1×CS、F1×CSph2b)中,平均单价体数都在7以下,说明其中的ph2b基因起到了促进染色体配对的作用;而在回交一代(F1×J-11、F1×郑麦-9023)中,则表现正常,与理论一致;(3)在所有自交和回交一代中,减数分裂期间均出现染色体桥、落后染色体、三分孢子体、多分孢子体以及微核等各种染色体异常行为,说明Ns基因组引起了所得材料细胞学上的不稳定性.从总体上讲,回交比自交能更快地恢复ph2b基因促进部分同源染色体配对的作用,为染色体重组提供更多的机会,期望在后代中获得新种质.  相似文献

本文利用普通小麦品系"中国春"(对照)、中国春ph1b突变体分别与八倍体小黑麦、六倍体小黑麦杂交,杂种F1的减数分裂前期Ⅰ染色体行为表现异常,中期Ⅰ出现较多的单价体、棒状二价体和多价体,在后期和末期出现落后染色体、染色体片断和微核。原因是ph1b基因的存在造成染色体联会机制紊乱,致使一些部分同源染色体配对并发生互换,有可能在以后的世代产生染色体易位与基因重组。  相似文献

本文利用17种异细胞质“中国春”小麦与黑麦,小黑麦杂交,回交,研究其性状与减数分裂行为的表现。首次观察到D2型Ae.crassa.4x细胞质对同源染色体配对有抑制作用,对部分同源染色体配对有促进作用,SV型Ae.kotschyi细胞质对同源染色体,部分同源配对均有抑制作用,S1型Ae.sharonesis细胞质对部分同源染色体配对有促进,还观察到G型细胞质T.timopheevi,T.zhukovskyi,D2型细胞质,Ae.crassa 可提高产生有功能雌配子数,首次合成G型,SV型,D2型细胞质雄性不育的八倍体小黑麦,D2型细胞质八倍体小黑麦是光敏性雄性不育,在15小时以上的长光照条件下表现不育,这对进一步了解异细胞质的作用,小黑麦杂种优势的利用和小黑麦的改良均有重要意义。  相似文献

采用酸性聚丙烯酰胺凝胶电泳(APAGE)法对11份A担心Aegilops kotschyi及其S^1染色体组供体种Ae.longissima2份和U染色体组供体种Ae.umbellulata6份进行了醇溶蛋白位点的研究。结果表明:11份Ae.kotschyi共分离出32条带,31条具有多态性,占96.88%,每份材料可以分离出10-17条谱带,其中仅1条(3.12%)是共有带;11份Ae.kotschyi的遗传距离的变异范围在0-0.704之间,平均为0.409;11份Ae.kotschyi分离出的多数醇溶蛋白谱带均与其染色体组供体种Ae.longissi-ma及Ae.umbellulata相同,但仍有8条谱带未在两供体种中找到;11份Ae.kotschyi的醇溶蛋白多态性(96.88%)明显高于Ae.longissima(52.94%)与Ae.umbellulata(88.89%)11份Ae.kotschyi中有4份表现出了一定的特征带,分析知可能在γ区发生了较大的变异。  相似文献

通过四倍体小麦与粗山羊草杂交,利用幼胚拯救技术获得了PS5-Y287双二倍体(AABBDD)。该双二倍体PMC MⅠ染色体构型基本稳定,对小麦白粉病表现免疫,对条锈、叶锈和秆锈病表现良好的田间抗性。并且该种质籽粒外观品质好,是一份有利用价值的小麦种质材料。  相似文献

C-banding patterns and polymorphisms were analyzed in several accessions of the diploidAegilops speciesAe. uniaristata, Ae. mutica, andAe. comosa subsp.comosa and subsp.heldreichii, and standard karyotypes of these species were established. Variation in C-band size and location was observed between different accessions, but did not prevent chromosome identification. One accession ofAe. uniaristata was homozygous for whole-arm translocations involving chromosomes 1N and 5N. The homoeologous relationships of these chromosomes were established by comparison of chromosome morphologies and C-banding patterns to other diploidAegilops species with known chromosome homoeology. In addition, in situ hybridization analysis with a 5S rDNA probe was used to identify homoeologous groups 1 and 5 chromosomes. The present analysis permitted the assignment of allAe. mutica, comosa subsp.comosa, andAe. comosa subsp.heldreichii chromosomes, and three of the sevenAe. uniaristata chromosomes according to their homoeologous groups. The data presented will be useful analyzing genome differentiation in polyploidAegilops species.  相似文献

直接杂交与幼胚培养方法结合获得了小麦与东方山羊草、尾状山羊草属间杂种。F1杂种细胞遗传学研究发现,东方山羊草的染色体组显著促进部分同源染色体配对,完全掩盖了小麦ph1b基因的作用,尾状山羊草的染色体组一定程度抑制部分同源染色体配对,尤其强烈抑制小麦ph1b基因的作用。表明东方山羊草的S染色体组上存在类似于小麦的ph1b基因,尾状山羊草的C染色体组上存在类似于小麦的Ph1基因  相似文献

以5个同质异核二角型小麦雄性不育系ms(bicor-8222,ms(bicor)-83(37)65,ms(bicor)-偃师9号,ms(bicor)-80(6)及ms(bicor)-90-110为基本材料,与一批优良小麦品种(系)及部分亲本材料为父本进行测交,获得211个组合,考察其F1育性,结果表明;(1)5个同质异核不育系,除二角型非1B/1R不育系ms(bicor)-90-110与二角型1B/1R不育系ms(bicor)-83(37)65,ms(bicor)-偃师9号之间平均恢复度差异显著外,4个1B/1R不育系之间平均结实率差异不显著;(2)对同一不育系而言,与不同恢复系测交,其恢复度呈现连续变异;(3)同型非1B/1R不育系较1B/1R不育系恢复度普遍高;(4)对同一恢复系而言,各不育系测交F1的恢复度差异显著。  相似文献

5个普通小麦品系的粗厚山羊草4X,6X种细胞质的7个核代换系在中国昆明,元谋,杨陵,互助,依安5个不同纬度地点种植的自交结实率,结合各点光温条件的分析表明,粗厚山羊草细胞质普通小麦核代换系的雄性育性随日长增加而下降,6X种细胞质核代换系在日长≥14.28h开始出现完全雄性不育,4X种细胞质核代换系在日长≥15.38h开始出现完全雄性不育;该细胞质核代换系的雄性育性在日长因子主控下,对温度变化也表现一定敏感性。粗厚山羊草细胞质背景下不同核基因型对光,温条件的敏感性有显著差异。核质互作效应是这种敏感性的遗传方式,粗厚山羊草4X、6X种细胞质的光,温敏感性存在差异。6X种细胞质的光,温敏感性高于4X种细胞质。  相似文献

Aegilops longissima Schw. et Musch. (2n= 2x=14, SlSl) and Aegilops sharonensis Eig. (2n=2x=14, SlSl) are diploid species belonging to the section Sitopsis in the tribe Triticeae and potential donors of useful genes for wheat breeding. A comparative genetic map was constructed of the Ae. longissima genome, using RFLP probes with known location in wheat. A high degree of conserved colinearity was observed between the wild diploid and basic wheat genome, represented by the D genome of cultivated wheat. Chromosomes 1Sl, 2Sl, 3Sl, 5Sl and 6Sl are colinear with wheat chromosomes 1D, 2D, 3D, 5D and 6D, respectively. The analysis confirmed that chromosomes 4Sl and 7Sl are translocated relative to wheat. The short arms and major part of the long arms are homoeologous to most of wheat chromosomes 4D and 7D respectively, but the region corresponding to the distal segment of 7D was translocated from 7SlL to the distal region of 4SlL. The map and RFLP markers were then used to analyse the genomes and added chromosomes in a set of ’Chinese Spring’ (CS)/Ae. longissima chromosome additions. The study confirmed the availability of disomic CS/Ae. longissima addition lines for chromosomes 1Sl, 2Sl, 3Sl, 4Sl and 5Sl. An as yet unpublished set of Ae. sharonensis chromosome addition lines were also available for analysis. Due to the gametocidal nature of Ae. sharonensis chromosomes 2Sl and 4Sl, additions 1Sl, 3Sl, 5Sl, 6Sl and 7Sl were produced in a (4D)4Sl background, and 2Sl and 4Sl in a euploid wheat background. The analysis also confirmed that the 4/7 translocation found in Ae. longissima was not present in Ae. sharonensis although the two wild relatives of wheat are considered to be closely related. Background and Aims Aegilops markgrafii (CC) and its natural hybrids Ae. triuncialis (UtUtCtCt) and Ae. cylindrica (DcDcCcCc) represent a rich reservoir of useful genes for improvement of bread wheat (Triticum aestivum), but the limited information available on their genome structure and the shortage of molecular (cyto-) genetic tools hamper the utilization of the extant genetic diversity. This study provides the complete karyotypes in the three species obtained after fluorescent in situ hybridization (FISH) with repetitive DNA probes, and evaluates the potential of flow cytometric chromosome sorting.Methods The flow karyotypes obtained after the analysis of 4'',6-diamidino-2-phenylindole (DAPI)-stained chromosomes were characterized and the chromosome content of the peaks on the flow karyotypes was determined by FISH. Twenty-nine conserved orthologous set (COS) markers covering all seven wheat homoeologous chromosome groups were used for PCR with DNA amplified from flow-sorted chromosomes and genomic DNA.Key Results FISH with repetitive DNA probes revealed that chromosomes 4C, 5C, 7Ct, T6UtS.6UtL-5CtL, 1Cc and 5Dc could be sorted with purities ranging from 66 to 91 %, while the remaining chromosomes could be sorted in groups of 2–5. This identified a partial wheat–C-genome homology for group 4 and 5 chromosomes. In addition, 1C chromosomes were homologous with group 1 of wheat; a small segment from group 2 indicated 1C–2C rearrangement. An extensively rearranged structure of chromosome 7C relative to wheat was also detected.Conclusions The possibility of purifying Aegilops chromosomes provides an attractive opportunity to investigate the structure and evolution of the Aegilops C genome and to develop molecular tools to facilitate the identification of alien chromatin and support alien introgression breeding in bread wheat.  相似文献
