Four mature lactating Holstein cows fitted with permanent ruminal, duodenal and ileal cannulae were used to study the effect of extrusion at 195 degrees C of beans (Vicia faba cv Castel) on organic matter (OM), nitrogen (N) and starch degradation in the rumen and their flow to and absorption from the small intestine. The test protein sources, raw beans (RB) and extruded beans (EB), provided about 45% of the dietary protein. The diets were composed of 23.1% beans, 56.2% corn silage, 10.1% corn grain and 10.7% Italian rye-grass hay on a DM basis; the diets were isocaloric (4.5 Mcal/kg of DM) and isonitrogenous (14% of DM). Cr-EDTA, YbCl3 and purines were used as liquid, particulate and bacterial markers respectively. Extruding the beans did not influence intraruminal pH (6.6), ammonia-N (99 mg/l) and volatile fatty acids (97 mM/l) concentrations. Apparent digestibility in total tract of energy, OM, N and starch were not affected with inclusion of EB instead of RB, the corresponding mean values were: 66, 68, 64 and 95% of intake. Apparent ruminal digestion of OM, starch and N in the forestomach were 39, 58 and 52% for diets containing RB and 38, 72 and 45% for EB diets; efficiency of bacterial protein synthesis (gN/kg organic matter truly digested in the stomach) was higher for EN diets compared with RN diets (25 vs 22). Eating diets including EB increased non-ammonia-N, bacterial-N and dietary-N flows (g/d) to the duodenum compared with diets containing RB: 409 vs 366, 216 vs 194 and 193 vs 172 respectively; while starch flow was reduced (1.5 vs 2.2 kg/d) and OM flow was unchanged (9.8 kg/d). Apparent digestion from the small intestine (g/d) of nonammonia-N, bacterial-N and dietary-N were higher for EB diets: 268 vs 229, 181 vs 160 and 87 vs 69 respectively; meanwhile, starch digestion decreased (975 vs 1,612). The PDIA, PDIN and PDIE contents (g/kg of DM) of the RB were 33, 175 and 127 respectively; the corresponding values after extrusion were: 58, 147 and 178.  相似文献

Four rumen and proximal duodenum fistulated non-lactating Holstein cows were used to determine the effect of extrusion at 120 degrees C of whole horse beans (Vicia faba cv Talo) on in vitro nitrogen (N) solubility and in situ degradation of dry matter (DM) and crude protein (CP) in the rumen and intestine. Cows were fed a ration of 30% whole horse beans (WHB) and 70% Italian rye-grass hay. The degradation of DM and CP was estimated using nylon bags suspended in the rumen for 2, 4, 7, 16, 24 and 48 h; the effective ruminal degradability of DM and CP was evaluated assuming a ruminal outflow rate of 0.06/h. Bags incubated in the rumen for 16 h were introduced into the small intestine through the duodenal cannula and subsequently recovered in the feces. Extrusion of WHB reduced N-solubility in buffer solution (21.1 vs 74.9%). Processing diminished the effective rumen degradability of DM (74.6 vs 80.4%) and CP (70.2 vs 89.2%). Meanwhile, the amounts of DM and CP digested in the intestine increased: 9.6 vs 1.4% and 25.2 vs 3.0% respectively. Therefore, feeds containing extruded WHB increase the availability of dietary proteins in the intestine compared with diets containing raw WHB.  相似文献

The effect of whole lupin seeds (Lupinus albus cv Lublanc) at 120, 150 and 195 degrees C on in situ nitrogen degradability (Dg.N) was measured by the nylon bag technique using fistulated non-lactating Holstein cows. The N degradation was evaluated in nylon bags suspended in the rumen; heating the seeds at 120, 150 and 195 degrees C decreased the Dg.N value: 83.9, 72.9 and 53.0 respectively vs 95.3% (rumen outflow rate of 0.06/h). To estimate the total N disappearing in the digestive tract, bags were incubated in the rumen for 16 h, then in a pepsin bath for 2 h and then introduced into the duodenum for subsequently recovery in feces. The whole tract degradability of N was always high, approximately 98.3%. The amounts of N which disappeared in the intestine increased from 3.1 (untreated seeds) to 15.1, 26.3 and 44.7% as the temperature rose to 120, 150 and 195 degrees C respectively. The PDIN and PDIE contents (g/kg of DM) of the raw whole lupin seeds were 224 and 84 respectively; extrusion elevated these values by 10-32% for PDIN and 57-194% for PDIE. The augmentation in the supply of dietary proteins to the postruminal parts as a result of extrusion could rapidly benefit high yielding cows.  相似文献

Effects of fatty acids of linseed in different forms, on ruminal fermentation and digestibility were studied in dry cows fitted with ruminal and duodenal cannulas. Four diets based on maize silage, lucerne hay and concentrates (65/10/25 dry matter (DM)) were compared in a 4 × 4 Latin square design experiment where the diets were: control diet (C), diet RL supplied 75 g/kg DM rolled linseeds, diet EL supplied 75 g/kg DM extruded linseeds, and diet LO supplied 26 g/kg DM linseed oil and 49 g/kg DM linseed meal. The diets did not differ in total organic matter (OM) and fibre digestibility, in forestomach and intestinal OM digestibility, and in duodenal N flow. Microbial N duodenal flow tended to be lower for RL versus C diet (P<0.1). Extrusion did not reduce ruminal crude protein (CP) degradation in vivo and in situ. Volatile fatty acid concentration and pattern, and protozoa concentration in the rumen, did not vary among diets. Results confirm the absence of a negative effect of a moderate supply of linseed on rumen function, as well as no effect of extrusion on its ruminal CP degradability.  相似文献

Wether lambs (n = 27, average BW = 40 kg) were used to test response to forage treated with Tasco-Forage (an extract of the brown kelp Ascophyllum nodosum) prior to conserving, or to direct feeding of the extract (Tasco-EX). Hays made from endophyte (Neoyphodium coenophialum)-infested tall fescue (Festuca arundinacea)-based pasture received 0 or 3 kg of Tasco/ha prior to harvest. Lambs, blocked by weight, were randomly allotted to three diets: (1) control hay, (2) treated hay, and (3) #1 + Tasco-EX fed at 1% of the diet. Hays were low in CP (<7%) so all lambs were fed soybean meal (12% of the diet) in addition to trace mineralized salt. Diets were fed at 1.5% BW to prevent refusals. Total collections (7 d) were made during periods without or with applied heat stress. After each period, rumen contents were obtained to determine pH, NH3 and VFA. Lambs were sacrificed post-trial. A subset was used to evaluate sensory traits and muscle fatty acids. Lambs were in negative N balance during the study and Tasco treatments did not affect N metabolism. Fecal N tended (P < 0.10) to increase with short duration heat stress causing a concomitant decrease (P < 0.05) in apparent N digestibility (58.6 versus 56.1%; S.E. = 0.7). Urinary N loss decreased (P < 0.001) with heat stress (8.0 versus 5.9 g/d; S.E. = 0.2), resulting in increased (P < 0.001) N retention (−2.1 versus −0.3 g/d; S.E. = 0.2). Apparent OM digestibility was not affected by heat stress but was greater (P < 0.05) for lambs fed Tasco-EX treatment than those fed treated hay. Treatment diets decreased (P < 0.05) ruminal butyrate. Heat stress increased (P < 0.05) acetate and total VFA and decreased (P < 0.01) ruminal pH. A tendency (P < 0.11) of increased 14:1ω5, decreased (P < 0.05) 18:0 and total saturated fatty acids in muscle was observed with Tasco diets. Meat sensory characteristics were not affected by treatment. Tasco may alter some aspects of rumen or lipid metabolism but has no effect on N metabolism or meat sensory characteristics of sheep fed restricted, low-quality diets.  相似文献

The experiment was conducted on double-muscled Belgian Blue bulls, fitted with rumen, duodenal and ileal cannulas, according to a 5 × 5 Latin square design with one missing animal. All diets contained 320 g/kg of lupin seed (Lupinus albus, var. Lublanc) on a dry matter (DM) basis. Four grinding treatments of raw lupins (RAW) were used to obtain median particle sizes of approximately 0.5, 2.0, 4.2 or 6.0 mm. One extrusion treatment (EXTR) was also used and consisted of heating to 180 °C for 30 s. The pH and ammonia nitrogen (NH3–N) concentration kinetics in the rumen liquid differed between RAW and EXTR. The latter induced larger variation in these parameters after the meal. Quadratic effects of the grinding level on rumen fermentation parameters were observed at several sampling times for RAW. The grinding level had also quadratic effects on the intestinal digestibility of DM, organic matter (OM) and N, reaching a maximum with treatment 4.2 mm. The treatment 0.5 mm tended to reduce the microbial N flow at the duodenum whereas EXTR tended to increase the non NH3–N flow. The total digestible flow of N tended to increase with the intermediate grinding levels and EXTR. Lupin protein degradability was calculated at 0.78, 0.74, 0.65, 0.82 and 0.70 for the 0.5, 2.0, 4.2, 6.0 mm treatments and EXTR, respectively. Treatments 2.0 and 4.2 mm increased the digestible protein in the small intestine (DVE) content by more than 40% compared with treatment 0.5 mm. The balance between rumen fermentable N and energy (OEB) was reduced by 15% with the 4.2 mm and EXTR. Compared with treatment 4.2 mm, EXTR did not greatly improve the nutritional value of lupin seed. The results suggested that lupin seed should be coarsely ground or flattened to obtain a mean particle size between 2.0 and 4.2 mm for cattle feed, but that an insufficient grinding level (treatment 6.0 mm) induced a higher degradability of lupin protein, probably due to more intense rumination. Under our experimental conditions, the extrusion did not sufficiently improve the nutritional value of the seed to be economically viable. These results show that feeding standards should consider the influence of the grinding level of legume seeds in order to assess their nutritional value accurately.  相似文献

Abstract Concentrations of oxalate-degrading anaerobes in ruminal contents of sheep were determined from counts of colonies producing clear zones on a calcium oxalate medium (D agar with 7 mM CaCl2). Viable counts of oxalate degraders from a 55-kg sheep fed a diet containing 32% halogeton (4.6% oxalate) averaged 2.6 × 106/ g (dry weight). When the halogeton concentration in the diet was reduced to 16%, counts of oxalate degraders decreased nearly 300-fold. Oxalate-degrading isolates from this sheep were similar to OxB, the type strain of Oxalobacter formigenes . When a 45-kg sheep was fed diets containing 2.2, 1.5, and 0.8% oxalate, viable counts of oxalate degraders (enumerated on D agar with 14 mM CaCl2 and 20% filter-sterilized ruminal fluid) represented 0.85, 0.52, and 0.06% of the total viable population, respectively; total viable counts were essentially unchanges by these concentrations of dietary oxalate. Similar percentages of oxalate degraders were also observed when a 23-kg sheep was fed diets containing 1.5 or 0.8% oxalate. This report presents the first direct measurements of the concentrations of oxalate-degrading bacteria in the rumen and supports the concept that the availability of oxalate in the diet influences the proportion of oxalate-degrading bacteria in the rumen  相似文献

Feedlot beef production in tropics and subtropics is being limited by shortage of feed ingredients due to prolonged droughts resulting in high prices of cereal grains and reduced profitability. To alleviate feed shortages and increase profitability for the feedlot industry, it is important to explore underutilized non-conventional resources such as fruit by-products. The current study compared the nutrient intake, digestibility and utilization effects of feeding either 150 g/kg of dried citrus pulp (DCP) or grape pomace (DGP) as alternative dietary fibre sources to 7 months-old Angus steers (281 ± 15.7 kg) under feedlot conditions. Twenty-four steers were assigned to three dietary treatments (8 steers/treatment) in a completely randomised design. Each steer was treated as an experimental unit. Steers were adapted to diets for 21 d followed by 7 d of sample collection during which nutrient intake, digestibility, rumen fermentation parameters, nitrogen retention and microbial N supply were determined. All the data were analysed using the GLIMMIX procedures. Overall, steers fed DGP had the greatest dry matter intake (DM), organic matter (OM) crude protein (CP), ash free neutral detergent fibre (aNDFom), ether extract (EE) and starch intake followed by DCP and control (CON) diets (P ≤ .05). Apparent digestibilities of DM, OM and aNDFom were higher (P ≤ .05) for DCP diet compared to DGP and CON diets. Feeding DCP and DGP diets increased ruminal concentrations of total volatile fatty acids, acetate and isovalerate, and acetate to propionate ratio, and reduced propionate concentrations compared to the CON diet (P ≤ .05). The steers fed the CON diet had the greatest urinary excretions of allantoin, uric acid and total purine derivatives followed by those fed the DCP and DGP diets (P ≤ .05). Nitrogen intake, faecal nitrogen (N), N retention and N efficiency utilization were in the order of DGP > DCP > CON diets (P ≤ .05). Overall, feeding DGP as alternative fibre source to wheat bran improved nutrient intake, retention and efficiency of N utilization but reduced apparent nutrient digestibility compared to DCP. Current finding suggests that DGP may be a better fibre substitute for wheat bran in beef diets than DCP.  相似文献

The potassium sensitive magnesium absorption through the rumen wall may be influenced by additional dietary properties, such as diet type, forage type or forage to concentrate ratio. These properties are likely associated to rumen passage kinetics modified by dietary fibre content. The study aimed to assess the effects of rumen passage kinetics on apparent Mg absorption and retention in lactating dairy cows fed modified levels of fibre. Six lactating Red-Holstein and Holstein cows, including four fitted with ruminal cannulas were randomly assigned to a 3 × 3 cross-over design. The experimental diets consisted of early harvested low NDF (341 g NDF/kg DM) and late harvested high NDF (572 g NDF/kg DM) grass silage (80% DM) and of concentrates (20% of DM). As the low-fibre diet was excessive in protein, a third high-fibre diet was formulated to be balanced in digestible protein with the low-fibre diet to avoid any eventual confounding effects of NDF and protein excess. All diets were formulated to contain iso-Ca, -P, -Mg, -K and -Na. Passage kinetics of solid and liquid phase of rumen digesta were evaluated using ruminal marker disappearance profiles. Cows fed the low-fibre diet had compared to the other diets, an up to 40% lower solid and 26% lower liquid phase volume of rumen digesta and a 10% numerically higher fractional rumen liquid passage rate. Rumen pH lost 0.6 units and Mg concentration in the rumen liquid phase tripled when cows were fed the low-fibre diet. Faecal Mg excretion was up to 14% higher in cows fed the low-fibre diet and Mg absorbability was 12% compared to up to 19% in other diets. Urinary Mg excretion in cows fed the low-fibre diet was half of the ones in the other treatments, but Mg retention was not affected. Dietary protein excess neither affected rumen passage kinetics nor Mg absorption and retention. Absorption of Mg was correlated with rumen liquid volume which both decreased with decreasing daily NDF intake (NDFi, 11.8 ± 2.4 l/kg NDFi). Consequently, daily Mg absorption decreased by 1.32 ± 0.28 g/kg decreasing NDFi. To conclude, in addition to the known antagonistic effect of dietary K, the present data indicate that Mg absorption was dependent from NDFi which modified rumen liquid volume, but was independent of dietary protein excess likely associated to low NDF herbages.  相似文献

In a 4 × 4 Latin-square experiment with a 2 × 2 factorial arrangement of treatments, 4 cattle fitted with a rumen and duodenal cannula were given four grass-containing diets [480 g kg−1 of the total dry matter (DM) intake] and barley (BU), barley + molasses (2:1) (BM), sugar-beet pulp (SU) or sugar-beet pulp + molasses (SM). Duodenal flow was estimated using Cr-mordanted straw and CoEDTA as markers, and microbial nitrogen entering the small intestine using purine bases of nucleic acids.

Molasses-containing diets had a higher (P < 0.01) organic matter (OM) digestibility. The proportion of digestible OM apparently disappearing in the rumen averaged 0.72 and was not significantly affected by the diet. When cattle received molasses, the quantity of microbial N entering the small intestine was higher (P < 0.05) and there was a trend towards a higher efficiency of microbial N synthesis (28.8 vs. 25.6 g N kg−1 OM apparently digested in the rumen). When S diets were consumed, total non-ammonia N flow at the duodenum exceeded N intake by 7.0 g day−1 and when B diets were consumed, it was 0.7 g day−1 less than N intake. Feed N degradability in the rumen and apparent N digestibility of S diets were lower (P < 0.05; P < 0.001) than those of B diets.

Rumen (P < 0.05) and total (P < 0.001) digestibility of neutral detergent fibre (NDF) and acid detergent fibre (ADF) was higher when S diets were given. The proportion of digestible fibre disappearing in the rumen was not affected by the diet. The rate and extent of silage and hay DM degradation were not significantly affected by the diet. However, dietary inclusion of molasses decreased (P < 0.05) the lag time of both hay and silage DM degradation.

The rumen dilution rate of liquid averaged 0.097 and that of particles, 0.049; neither was significantly different for either B and S diets or U and M diets. Duodenal liquid flow was higher (P < 0.05) for M diets.

Average rumen pH was not affected by the diet, but the molasses diets increased (P < 0.05) the range in rumen pH. The BM diet was associated with higher (P < 0.01) rumen ammonia concentration than the other diets. Low rumen ammonia concentrations (< 2 mM) were observed for long periods between feeds. The molar proportion of butyrate was higher on B diets and there was a trend towards a higher proportion of acetate and propionate on S diets. Molasses tended to increase the molar proportion of propionate and butyrate.  相似文献

The rumen microbial ecosystem is a complex system where rumen fermentation processes involve interactions among microorganisms. There are important relationships between diet and the ruminal bacterial composition. Thus, we investigated the ruminal fermentation characteristics and compared ruminal bacterial communities using tag amplicon pyrosequencing analysis in Yanbian yellow steers, which were fed linseed oil (LO) and propionate precursors. We used eight ruminally cannulated Yanbian yellow steers (510 ± 5.8 kg) in a replicated 4 × 4 Latin square design with four dietary treatments. Steers were fed a basal diet that comprised 80% concentrate and 20% rice straw (DM basis, CON). The CON diet was supplemented with LO at 4%. The LO diet was also supplemented with 2% dl-malate or 2% fumarate as ruminal precursors of propionate. Dietary supplementation with LO and propionate precursors increased ruminal pH, total volatile fatty acid concentrations, and the molar proportion of propionate. The most abundant bacterial operational taxonomic units in the rumen were related to dietary treatments. Bacteroidetes dominated the ruminal bacterial community and the genus Prevotella was highly represented when steers were fed LO plus propionate precursors. However, with the CON and LO diet plus malate or fumarate, Firmicutes was the most abundant phylum and the genus Ruminococcus was predominant. In summary, supplementing the diets of ruminants with a moderate level of LO plus propionate precursors modified the ruminal fermentation pattern. The most positive responses to LO and propionate precursors supplementation were in the phyla Bacteriodetes and Firmicutes, and in the genus Ruminococcus and Prevotella. Thus, diets containing LO plus malate or fumarate have significant effects on the composition of the rumen microbial community.  相似文献

To determine whether non-fibrous carbohydrate (NFC) supplementation improves fiber digestibility and microbial protein synthesis, 18 Corriedale ewes with a fixed intake level (40 g dry matter (DM)/kg BW0.75) were assigned to three (n = 6) diets: F = 100% fresh temperate forage, FG = 70% forage + 30% barley grain and FGM = 70% forage + 15% barley grain + 15% molasses-based product (MBP, Kalori 3000). Two experimental periods were carried out, with late (P1) and early (P2) vegetative stage forage. For P2, ewes were fitted with ruminal catheters. Forage was distributed at 0900 h, 1300 h, 1800 h and 2300 h, and supplement added at 0900 h and 1800 h meals. Digestibility of the different components of the diets, retained N and rumen microbial protein synthesis were determined. At the end of P2, ruminal pH and N-NH3 concentration were determined hourly for 24 h. Supplementation increased digestibility of DM (P < 0.001) and organic matter (OM; P < 0.001) and reduced NDF digestibility (P = 0.043) in both periods, with greater values in P2 (P = 0.008) for the three diets. Daily mean ruminal pH differed (P < 0.05) among treatments: 6.33 (F), 6.15 (FG) and 6.51 (FGM). The high pH in FGM was attributed to Ca(OH)2 in MBP. Therefore, the decreased fiber digestibility in supplemented diets could not be attributed to pH changes. The mean ruminal concentration of N-NH3 was 18.0 mg/dl, without differences among treatments or sampling hours. Microbial protein synthesis was greater in P2 (8.0 g/day) than in P1 (6.1 g/day; P = 0.006), but treatments did not enhance this parameter. The efficiency of protein synthesis tended to be lower in supplemented groups (16.4, 13.9 and 13.4 in P1, and 20.8, 16.7 and 16.2 g N/kg digestible OM ingested in P2, for F, FG and FGM, respectively; P = 0.07) without differences between supplements. The same tendency was observed for retained N: 2.55, 1.38 and 1.98 in P1, and 2.28, 1.23 and 1.10 g/day in P2, for F, FG and FGM, respectively; P = 0.05). The efficiency of microbial protein synthesis was greater in P2 (P = 0.007). In conclusion, addition of feeds containing NFCs to fresh temperate forage reduced the digestibility of cell walls and did not improve microbial protein synthesis or its efficiency. An increase in these parameters was associated to the early phenological stage of the forage.  相似文献

The purpose of this study was to evaluate the effects of various N sources in concentrates containing high levels of cassava chips, with rice straw as the basal forage, on rumen ecology, rumen microbial counts, microbial crude (CP) protein synthesis, and digestibility of nutrients. Four ruminally fistulated crossbred (Brahman × native) beef steers with initial body weight (BW) of 400 ± 40.2 kg were randomly assigned according to a 4 × 4 Latin square design. The dietary treatments were different sources of N in the concentrates and were: T1 = urea (control; urea); T2 = soybean meal (SBM); T3 = urea CaCl2 mixture (U-Cal); T4 = urea CaSO4 mixture (U-Cas). All steers were kept in individual pens and supplemented with concentrate at 5 g/kg of BW daily. The experiment was 4 periods, and each lasted 21 d. During the first 14 d, all steers were fed their respective diets ad libitum and for during the last 7 d, they were moved to metabolism crates for total urine and fecal collection. Dry matter intake ranged from 9.8 to 10.5 kg daily and was not altered by diet, while digestibility of NDF differed among treatments and was highest with U-Cas supplementation (P<0.05). Ruminal NH3 N and plasma urea N with U-Cal, U-Cas, and SBM diets were lower compared with the urea supplemented group (P<0.05). Ruminal volatile fatty acid concentrations were not altered by treatments. Total viable, and cellulolytic bacteria, differed among treatments and were highest with U-Cas (9.1 × 1011, and 4.0 × 109 cfu/mL, respectively). In addition, efficiency of rumen microbial CP synthesis based on organic matter (OM) truly digested in the rumen was increased by SBM or U-Cal supplementation, and was highest with U-Cas supplementation (18.2 g of N/kg of OM truly digested in the rumen). Supplementation of U-Cas to a concentrate containing a high level of cassava chips improved rumen ecology and microbial CP synthesis in beef cattle, suggesting that urea calcium mixtures can replace soybean meal or urea in beef cattle diets without adverse affects on rumen fermentation and other rumen parameters.  相似文献

There is increasing interest in using locally produced protein supplements in dairy cow feeding. The objective of this experiment was to compare rapeseed meal (RSM), faba beans (FBs) and blue lupin seeds (BL) at isonitrogenous amounts as supplements of grass silage and cereal based diets. A control diet (CON) without protein supplement was included in the experiment. Four lactating Nordic Red cows were used in a 4 × 4 Latin Square design with four 21 d periods. The milk production increased with protein supplementation but when expressed as energy corrected milk, the response disappeared due to substantially higher milk fat concentration with CON compared to protein supplemented diets. Milk protein output increased by 8.5, 4.4 and 2.7% when RSM, FB and BL were compared to CON. The main changes in rumen fermentation were the higher propionate and lower butyrate proportion of total rumen volatile fatty acids when the protein supplemented diets were compared to CON. Protein supplementation also clearly increased the ruminal ammonia N concentration. Protein supplementation improved diet organic matter and NDF digestibility but efficiency of microbial protein synthesis per kg organic matter truly digested was not affected. Flow of microbial N was greater when FB compared to BL was fed. All protein supplements decreased the efficiency of nitrogen use in milk production. The marginal efficiency (amount of additional feed protein captured in milk protein) was 0.110, 0.062 and 0.045 for RSM, FB and BL, respectively. The current study supports the evidence that RSM is a good protein supplement for dairy cows, and this effect was at least partly mediated by the lower rumen degradability of RSM protein compared to FB and BL. Total and cellulolytic bacterial and fungal numbers were determined in ruminal and cecal contents of 20 blue duikers (Cephalophus monticola). The animals were equally divided by sex and fed two diets, either high roughage or high concentrate. The mean concentration for total bacterial numbers in the rumen was 26.0 × 108/g of contents, with values ranging from 2 × 108/g to 93 × 108/g. Cellulolytic numbers averaged 6.0 × 108/g with a range of 1.5 × 108/g to 24.0 × 108/g. No differences related to sex or diet were found. In contrast, total bacterial numbers in the cecum differed between diets (P < 0.02), i.e., 1,046 × 106 bacteria per g for animals fed the high-forage diet compared with 166 × 106/g for those fed the high-concentrate diet. Cellulolytic bacterial counts in the cecal contents averaged 3.1 and 7.0% of the total counts for the high-forage and high-concentrate diets, respectively. Low concentrations of fungi were found in both ruminal and cecal contents of some, but not all, animals. Unexpectedly, concentrations of bacteria and fungi in the rumen and cecum were highly correlated with their total numbers (concentration multiplied by total weight of contents).  相似文献

The aim of the present experiment was to investigate an experimental brown midrib (Bm) maize hybrid in comparison with a control (Con) non-Bm maize hybrid on ruminal and total tract digestibility, ruminal fermentation, ruminal ingesta kinetics, nitrogen (N) utilisation and microbial efficiency. A total of six ruminally and duodenally cannulated German Holstein cows were used. Animals were fed diets of either 11.5 kg dry matter (DM) of a Con or a Bm maize silage plus 4.1 kg DM of concentrate. Ruminal and total tract digestibility of organic matter, neutral detergent fibre and acid detergent fibre did not differ between hybrids. Short-chain fatty acid concentrations and pH in the rumen were not affected, but ruminal mean retention time was lower for Diet Bm (Con: 45.4 ± 2.39 h; Bm: 40.6 ± 2.39 h; least squares means ± standard error). Cows fed Diet Bm had greater efficiency of N utilisation (Con: 30.1 ± 1.37%; Bm: 33.1 ± 1.37%) and increased flow of microbial crude protein at the duodenum (MCPF) (Con: 7.0 ± 0.37 g/MJ metabolisable energy (ME); Bm: 8.1 ± 0.37 g/MJ ME). Thus, MCPF and utilisable crude protein at the duodenum (uCP) were greater for Diet Bm (MCPF – Con: 1117 ± 52.1 g/d; Bm: 1306 ± 52.1 g/d; uCP – Con: 1594 ± 57.9 g/d; Bm: 1807 ± 57.9 g/d) and ruminal N balance was lower for Diet Bm (Con: 98.7 ± 8.92 g/d; Bm: 65.6 ± 8.92 g/d). The present results show that the Bm maize hybrid might be advantageous for dairy cow nutrition with regard to N utilisation and MCPF. However, further research is necessary to draw more precise conclusions on the potential of Bm maize hybrids in general.  相似文献

Biotin is involved in many vital metabolic pathways and must be provided for an efficient fermentation in the rumen, as well as for the intermediary metabolism of the host animal. Factors influencing ruminal biotin metabolism and output are widely unknown at present. Therefore, dairy cows fitted with permanent cannulas in the dorsal rumen and in the proximal duodenum were fed differently composed diets, and the biotin flow at the proximal duodenum was measured. The diets (on DM basis) consisted of 8.9 kg grass hay (Diet 1), 8.9 kg corn silage plus 2.0 kg concentrate (Diet 2), or 7.3 and 7.4 kg grass silage plus 10.0kg concentrate (Diets 3 and 4). The concentrate in Diets 3 and 4 contained 87% wheat and corn grain, respectively. The cows were pre-fed the rations for 21 days. Thereafter duodenal digesta was sampled every two h for 5 days. Cr2O3 served as a flow marker and the microbial proportion of total nitrogen at the duodenum was estimated by near infrared spectroscopy (NIRS). The duodenal flow of biotin was not related to biotin intake, but to the amount of fermented organic matter (FOM) and the amount of microbial protein (Biotin [mg/d] = 0.518 kg FOM - 0.300; r=0.85 and biotin [mg/d] = 0.012 x g microbial protein + 1.478; r = 0.84), irrespective of the composition of the diet fed. Mean daily biotin flow was 0.48 +/- 0.11 mg/kg FOM without any systematic effect of diet composition. The ruminal biotin balance, calculated as the difference between biotin flow at the duodenum and biotin intake, was positive (1.4 - 2.0 mg/d) in cows fed the mixed roughage/concentrate diets and negative (-0.71 mg/d) when the pure hay diet was fed.  相似文献

Isoenergetic maize silage diets, fed at maintenance to 24 suckling cows (Exp. 1) and to 3 (Exp. 2) or 4 (Exp. 3) rumen fistulated sheep, were compared with a urea and controlled release NPN diet (Exps. 1 and 2) and with a protein-N supplemented diet (Exp. 3). Supplementation increased blood urea concentrations (44.7 +/- 22.3 vs. 97.6 +/- 23.7 mg urea-N x L(-1)) (Exp. 1) and renal urea excretion (2.5 +/- 1.1 vs. 7.6 +/- 1.8 g urea-N x d(-1)) (Exps. 2 and 3), whereas blood allantoin concentrations (286.7 +/- 77.0 micromol x L(-1)) (Exp. 1) and renal excretion of purine derivatives (357.6 +/- 90.7 mg purine-N x d(-1)) (Exps. 2 and 3) were not affected, indicating additional N supplementation did not improve rumen microbial growth. However, some deficiency of rumen degradable N might occur in non supplemented diets as suggested by the reduced rumen NH3-N and RNA concentrations (868 +/- 270 vs. 1466 +/- 466 mg RNA x kg(-1) rumen contents).  相似文献

When added to the diet of sheep, 2 g/d, Aspergillus oryzae fermentation extract (AO) stimulated total and cellulolytic bacterial numbers in rumen fluid by 34 and 90% respectively. AO had no effect on the numbers of protozoa or fungal zoospores. AO did not affect hydrogen production by the rumen fungi Neocallimastix frontalis (RE1), N. patriciarum (CX) or Piromonas communis (P) in pure culture or protozoal activity in vitro , estimated from the rate of breakdown of [14C] leucine-labelled Selenomonas ruminantium. It was concluded that increases in ruminal fibre digestion observed previously in animals fed AO, were most likely due to a stimulation of bacteria rather than eukaryotes in the rumen microbial population.  相似文献

This study examined the effect of substituting dry wormwood (Artemisia sp.) for rice straw in sheep diets on intake and apparent digestibility in vivo, nitrogen (N) balance and ruminal fermentation characteristics. Four Corriedale×Polwarth sheep (51.7 ± 1.3 kg) were individually housed in metabolism cages and fed diets (ad. libitum) with a 70:30 forage to concentrate ratio (DM basis), in which the basal rice straw was substituted with 0 (Control), 30 (LW), 50 (MW) or 100 (HW) g/kg of dry wormwood. The experimental design was a 4×4 latin square design in which 10 days of dietary adaptation was followed by 6 days of total feces and urine collection in each period. Rumen fluid was collected from a stomach tube at −0.5, 0.5, 1, 2, 4, and 8 h after the morning feed on day 6 of each collection period and analyzed for volatile fatty acids (VFA). The intakes of dry matter (DM), organic matter intake (OM), crude protein (CP), ether extract (EE), neutral detergent fibre (NDF) and acid detergent fibre (ADF) were higher (P<0.05) in sheep fed diets containing wormwood, than those fed the Control diet. Compared to the Control diet, CP digestibility was higher (P<0.05) in sheep fed MW and N intake, retained N, EE digestibility, urinary purine derivatives and microbial N yield were higher (P<0.05) in sheep fed diets, containing wormwood. Rumen pH was unaffected by treatment. Rumen NH3-N and VFA concentrations were similar across treatments except that most values for diets containing wormwood were higher (P<0.05) than those for the control diet within the first 2 h of feeding. The non-glucogenic acid ratio was also similar across Control, LW and MW treatments, but it was generally lower in MW versus the Control treatment. In conclusion, substituting wormwood for rice straw in the sheep diets increased feed intake, rumen fermentation, in vivo digestibility, N retention and microbial N yield, particularly at the medium and high levels of wormwood inclusion.  相似文献
