Summary In order to minimize the adverse effect of CO2 gas in a packed bed immobilized yeast reactor, a fluidized bed reactor was used for the continuous production of ethanol from glucose. Immobilized yeast was prepared by entrapping whole cells of Saccharomyces cerevisiae within a Caalginate matrix. It was found that the efficiency of the ethanol production in a fluidized bed reactor was 100% better than that for a packed bed reactor system. The alcohol productivity obtained was 21 g/l/hr in a fluidized bed reactor at 94% of conversion level.  相似文献

Summary Methane was produced from H2 and CO2 byMethanobacterium thermoautotrophicum cells fixed on the surface of hollow fibers. The mineral solution permeated through the inside of fibers was consumed by the cells, while the gaseous substrate flowing outside the fibers was directly metabolized to methane. Methane production was proportional to hollow fiber length i.e., contact area between cell layer and gas phase. In repeated batch cultures, the production rates of methane and cell mass were 33.1 L/L reactor/day and 1.75 g cells/L reactor/day, respectively with 90% conversion rate.  相似文献

Function of Y in codon-anticodon interaction of tRNA Phe   总被引:7,自引:0,他引:7  
Molar association constants of binding oligonucleotides to the anticodon loops of (yeast) tRNAPhe, (yeast) tRNAHClPhe and (E. coli) tRNAFMet have been determined by equilibrium dialysis. From the temperature dependence of the molar association constants, ΔF, ΔH and ΔS of oligomer-anticodon loop interaction have been determined. The data indicate that the free energy change of codon-anticodon interaction is highly influenced by the presence of a modified purine (tRNAPhe), of an unmodified purine (tRNAFMet) or its absence (tRNAHClPhe). Excision of the modified purine Y in the anticodon loop of tRNAPhe results in a conformational change of the anticodon loop, which is discussed on the basis of the corresponding changes in ΔF, ΔH and ΔS.  相似文献

Summary The transformation of hydrogen sulfide into elementary sulfur and sulfate was investigated in a photo-bioreactor using autotropic bacteriaChlorobium thiosulfatophilum. The accumulations of sulfur and sulfate in the reactor were found to be dependent on the light energy and the feed rate of H2S. The optimum operation lines were established to limit sulfide or sulfate. Immobilization of the whole cells in strontium-alginate matrix enhanced the conversion more than with the free cells.  相似文献

We prepared hairpins that differ in the connectivity of phosphodiester linkages in the loop (RNA vs 2′, 5′-RNA). We find that the stability of the extra stable RNA hairpin 5′-rGGAC(UUCG)GUCC-3′ is the same as that observed for the hairpin containing a 2′,5′RNA loop, i.e. 5′-rGGAC(UUCG)GUCC-3′ (where UUCG = U2′p5′U2′p5′ C2′p5′G2′p5′). Also significant is the finding that when the stem is duplex DNA, duplex 2′,5′-RNA, or DNA:2′,5′-RNA, hairpins with the UUCG loop are more stable than those with UUCG loop.  相似文献

Summary The plasmid pRH271, harboring a xylanase gene cioned fromBacilius subtilis, has been transferred into a mutant ofE. coli SK2284 which allowed the release of part of the xylanase in the culture supernatant. Kinetic parameters of this recombinantE. coll strain were determined in microscale batch culture with and without the selective pressure of antibiotics. No significant difference in µmax was observed for the nontransformedE. coli strain when compared to the recombinant strain. However, K5 values for glucose were two times higher in the case of the recombinant strain. Preliminary study of xylanase production in a large batch farmenter was also described.  相似文献

Summary Thermophilic degradation of sugar beet pulp was studied in batch cultures at 55°C by different associations of bacteria, includingClostridium thermocellum,Methanobacterium sp. andMethanosarcina MP.C. thermocellum produced acetate, succinate, methanol, ethanol, H2 and CO2. The coculture ofC. thermocellum andMethanobacterium sp. produced trace amounts of ethanol and succinate; acetate concentration was about three times higher than in theC. thermocellum monoculture. The association of this coculture withMethanosarcina MP produced 5.5 mmol CH4/g dry weight sugar beet pulp.  相似文献

Abstract

We report on the three dimensional structure of an RNA hairpin containing a 2′,5′-linked tetraribonucleotide loop, namely, 5′-rGGAC(UUCG)GUCC-3′ (where UUCG = U2′p5′U2′p5′C2′p5′G2′p5′). We show that the 2′,5′-linked RNA loop adopts a conformation that is quite different from that previously observed for the native 3′,5′-linked RNA loop. The 2′,5′- RNA loop is stabilized by (a) U:G wobble base pairing, with both bases in the anti conformation, (b) extensive base stacking, and (c) sugar–base contacts, all of which contribute to the extra stability of this hairpin structure.  相似文献

Jet aerated loop reactors (JLRs) provide high mass transfer coefficients (kLa) and can be used for the intensification of mass transfer limited reactions. The jet loop reactor achieves higher kLa values than a stirred tank reactor (STR). The improvement relies on significantly higher local power inputs (~104) than those obtainable with the STR. Operation at high local turnover rates requires efficient macromixing, otherwise reactor inhomogeneities might occur. If sufficient homogenization is not achieved, the selectivity of the reaction and the respective yields are decreased. Therefore, the balance between mixing and mass transfer in jet loop reactors is a critical design aspect. Monitoring the dissolved oxygen levels during the turnover of a steady sodium sulfite feed implied the abundance of gradients in the JLR. Prolonged mixing times at identical power input and aeration rates (~100%) were identified for the JLR in comparison to the STR. The insertion of a draft tube to the JLR led to a more homogenous dissolved oxygen distribution, but unfortunately a reduction of mixing time was not achieved. In case of increased medium viscosities as they may arise in high cell density cultivations, no gradient formation was detected. However, differences in medium viscosity significantly altered the mass transfer and mixing performance of the JLR.  相似文献

Summary Continuous ethanol production byS. uvarum immobilized in a low-gelling temperature agarose namely SeaPlaque agarose was studied in a packed bed reactor at 30°C using sugarcane molasses containing 13.5% fermentable sugars as feed. The productivity at 95% conversion was 23 g/l.h (on reactor volume basis). The bioreactor was run continuously at a fixed dilution rate and it retained 60% of its initial activity upto 80 days.  相似文献

A 22 m long. 20 liter tubular loop fermentor (TLF) has been tested for oxygen transfer characteristics and as a reactor for mycelial growth. Model calculations show that the flow pressure drop has an important influence on the axial oxygen profiles. A design model that accounts for this influence is presented. Using the model, KL a values are calculated from the results of sulfite oxidation experiments. These are correlated with power consumption and aeration rates. The KL a dependence on aeration rate was found to be less than found with tank reactors. The growth kinetics of three metabolite-producing mycelial organisms in the TLF are presented: a Streptomyces, a Fusarium, and a Acrophialophora. In order to determine the influence of reactor type on the growth and product formation, these cultures have been grown in tanks and shake flasks. The antibiotic, product spectrum of Streptomyces is compared on the basis of inhibition tests and it is shown that the distribution of products is reactor dependent. The Fusarium culture produced a previously unknown metabolite, whose concentration in the loop fermentor was four times higher than in a shake flask. The Acrophialophora culture grew twice as fast in the loop fermentor, but produced essentially none of the specific product. Power Consumptions of up to 8 kW/m3 in the tubular fermentor did not appear to harm the mycelia.  相似文献

Summary This report describes the production of N-carbamyl--alanine, by resting cells ofPseudomonas putida, in two types of continuous fermentation processes: alginate-immobilized cells in a continuous stirred tank reactor and a free cell recycle system using microporous membranes. The four stereoisomers of 25-hydroxyvitamin D3-26,23 lactone (25-OHD3-26,23 lactone) were tested against in vivo 25-OHD3-26,23 lactone to determine their relative competition in the radioligand binding assays for 25-OHD3 and 1,25-(OH)2D3. The 25R-OHD3-26,23S lactone and in vivo 25-OHD3-26,23 lactone behaved identically in the radioligand binding assay for 25-OHD3 and were ~5-fold more potent than 25-OHD3 at displacing 25-OH[3H]D3. The 25S-OHD3-26,23S lactone was the poorest competitor in this assay, requiring a 10-fold excess relative to 25-OHD3 to displace 50% of the 25-OH[3H]D3. The order of competition in the 25-OHD3 radioligand binding assay was 25R-OHD3-26,23S lactone = in vivo 25-OHD3-26,23 lactone ? 25S-OHD3-26,23R lactone > 25-OHD3 ? 25R-OHD3-26,23R lactone > 25S-OHD3-26,23S lactone. The order of competition in the 1,25-(OH)2D3 cytosol receptor assay was essentially reversed from the competition in the 25-OHD3 assay and was 25S-OHD3-26,23S lactone > 25-OHD3 ? 25S-OHD3-26,23R lactone > 25R-OHD3-26,23S lactone = in vivo 25-OHD3-26,23 lactone. When tested in a high-performance liquid chromatographic system which separates all four stereoisomers, the in vivo 25-OHD3-26,23 lactone comigrated with synthetic 25R-OHD3-26,23S lactone. These data firmly establish that the naturally-occurring 25-OHD3-26,23 lactone has the 25R, 23S stereochemistry. In addition, these data are the first to demonstrate that the four stereoisomers of 25-OHD3-26,23 lactone have different affinities for the plasma vitamin D binding protein and the 1,25-(OH)2D cytosol receptor.  相似文献

Somatic hybridization of Lycopersicon peruvianum and Petunia hybrida was carried out to transfer cytoplasmic male sterility from Petunia to Lycopersicon. Cytological, morphological and biochemical analyses were performed to characterize the regenerated plants. Two regenerated plants, R3 and R6, were male sterile. R3 possessed chromosomes morphologically similar to those of both parental types. Leaf morphologies of these two plants and a third plant, R7, were intermediate between the two parents. The stability of RUBPCase was verified during parental plant development and after in vitro culture. Plant R7 presented a new form of the large subunit of RUBPCase.  相似文献

Summary The thermophilic fungusTalaromyces emersonii CBS 814.70 was co-immobilized with cellulose in calcium alginate beads. Cellulose production by this system was compared with a conventional process. The immobilized system continued to produce enzyme in a batch-fed reactor system while the non-immobilized had ceased production; levels of enzyme produced by the immobilized system were almost twice the maximum produced by the non-immobilized system.  相似文献

Summary An investigation of plasmid stability in aSaccharomyces cerevisiae double mutant has been performed. The host was a double recombinantura3 furl mutant containing a plasmid bearing the yeast URA3+ allele and an expression cassette for human 1-antitrypsin. The mutant was grown in continuous culture employing a semi-defined medium containing added uracil to provide non selective growth conditions. After 150 generations of continuous growth, no cured cells had been detected: the specific expression level of 1-antitrypsin remained constant throughout the experiment.  相似文献

The purified, reconstitutively active, β-subunit of the Fo·F1-ATP synthase of Rhodospirillum rubrum chromatophores was found to bind both 4-chloro-7-nitrobenzofurazan (NBD-Cl) and dicyclohexylcarbodiimide (DCCD). The binding stoichiometry at saturation was 1 mol of either reagent per mol of β. The NBD-modified β-subunit did rebind to the β-less chromatophores and restored all their lost ATP-linked activities as efficiently as the untreated β, whereas the DCCD-modified β-subunit lost completely its capacity to rebind to the depleted chromatophores. These results suggest that the amino acid residue which is modified by NBD-Cl in the isolated β-subunit is not essential for binding and may be also not for activity.  相似文献

Summary Acrylamide was continuously produced from acrylonitrile usingBrevibacterium sp. CHl grown and immobilized in a dual hollow fiber bioreactor of 8.0 cm3. The biomass reached as high as 200 gm/L of the space available for the cell growth. The volumetric productivity of the reactor was 88 gm/L. h and the conversion of acrylonitrile varied with acrylonitrile concentration, pH and feed rate.  相似文献
